Olive fruits are seriously deteriorated by pre and postharvest damage due to the attack of insects, such as Bactrocera olaea, which strongly alters the quality of olives. Defence response in olive fruits injured both by pathogens and by mechanical damages has been associated with the enzyme β-glucosidase, which specifically hydrolyses oleuropein, producing highly reactive aldehyde molecules. In situ detection of β-glucosidase activity in olive fruit tissues following injury, which simulates Bactrocera oleae punctures, is reported. The assay was performed in two cultivars showing different degrees of susceptibilities to fly infestation. In both cultivars, the histochemical assay for β-glucosidase showed that within 20 min after the injury, a strong β-glucosidase activity could be observed in the damaged tissues. Thereafter a progressive enzyme inactivation occurred starting from tissues around the boundary of the injury with decrease of the enzyme activity and stopped after 3h. Whereas the mass of active celsreached a distance of (300±50)μm from the edge of the injury. Biochemical analyses showed that in extracts of the injured fruit, β-glucosidase activity rapidly increased within 20 min from injury, thereafter decreasing and reaching values comparable with those in intact fruits. Following puncture, the oleuropein contents did not change significantly in the high susceptibility cultivar, whereas it rapidly decreased in the cultivar showing low susceptibility. The results strongly suggest that olive fruits susceptible towards fly infestation could be related to the ability of the oleuropein-degrading-β-glucosidase to produce the highly reactive molecules in the damaged tissues. As a consequence of puncture, high level of peroxidase activity was detected. This feature also suggested that this enzyme could play a key role in the defence response against insect injuries.  相似文献

Attract and kill (AK) technology was evaluated against the Mediterranean fruit fly (Med-fly), Ceratitis capitata W.. Treatment was carried out in two peach orchards of four hectares each; first one located in Borj-Touil and the second in El-Kssibi Mornag, Northern Tunisia during the 2012 field season. AK is a specialized system based on hydrolyzed proteins and alpha-cypermethrin. During this study, 400 AK bait stations were placed in every hectare, four weeks before fruits color changes. The tested AK system was found effective in reducing the number of C. capitata population to 70% comparing to the untreated orchards. Fruit damages assessment showed significant differences between treatments in respect of decreasing infestation onto fruits Study results indicated that AK baited treatments were able to reduce dropped and soften fruits infestation seven times than that of control plots. Fruit damages were restricted to 4%-5% in treated areas with AK system in compare to 31%-35% recorded in control plots. This novel AK system showed good performance to combat Med-fly and can successfully used to combat various fruit fly species when applied for an area wide application program.  相似文献

Analysis of Major Carotenoid Composition and Its Content of Citrus Fruit   总被引:1,自引:0,他引:1  
a-carotene, β-carotene, lycopene, β-cryptoxanthin, zeaxanthin and lutein content of fruits in53 citrus cultivars were determined using HPLC. In both peel and pulp of citrus fruit, the major carotenoidswere lutein, zeaxanthin and β-cryptoxanthin. β-carotene content was relatively low and extremely low was theamount of or-carotene. Among the 53 cultivars tested, lycopene was detected only in pulp of Cara Cara navelorange. Carotenoid content in both peel and pulp of citrus fruit was the highest in Citrus reticulata Blanco andlowest in Citrus grandis Osbeck. Consequently, as far as the health protection value is considered, fruit ofCitrus reticulata Blanco ranks probably higher than other citrus fruits. In fruit of most Citrus retieulatavarities, β-cryptoxanthin was the main carotenoid component in pulp and its amount approximated that of lu-tein in peel. Content of lutein, zeaxanthin and β-cryptoxanthin in peel was about 2.5 - 15 times that in pulp onthe basis of fresh weight. Thus peel was inferred to be the principal location for the carotenoid stock in citrus fruit.  相似文献

Citrus fruits are rich in carotenoids.In the carotenoid biosynthetic pathway,lycopene β-cyclase(LCYb,EC:1.14.-.-) is a key regulatory enzyme in the catalysis of lycopene to β-carotene,an important dietary precursor of vitamin A for human nutrition.Two closely related lycopene β-cyclase cDNAs,designated CsLCYb1 and CsLCYb2,were isolated from the pulp of orange fruits(Citrus sinensis).The expression level of CsLCYb genes is lower in the flavedo and juice sacs of a lycopeneaccumulating genotype Cara Cara than that in common genotype Washington,and this might be correlated with lycopene accumulation in Cara Cara fruit.The CsLCYb1 efficiently converted lycopene into the bicyclic β-carotene in an Escherichia coli expression system,but the CsLCYb2 exhibited a lower enzyme activity and converted lycopene into the β-carotene and the monocyclic γ-carotene.In tomato transformation studies,expression of CsLCYb1 under the control of the cauliflower mosaic virus(CaMV) 35S constitutive promoter resulted in a virtually complete conversion of lycopene into β-carotene,and the ripe fruits displayed a bright orange colour.However,the CsLCYb2 transgenic tomato plants did not show an altered fruit colour during development and maturation.In fruits of the CsLCYb1 transgenic plants,most of the lycopene was converted into β-carotene with provitamin A levels reaching about 700 μg g-1DW.Unexpectedly,most transgenic tomatoes showed a reduction in total carotenoid accumulation,and this is consistent with the decrease in expression of endogenous carotenogenic genes in transgenic fruits.Collectively,these results suggested that the cloned CsLCYb1 and CsLCYb2 genes encoded two functional lycopene β-cyclases with different catalytic efficiency,and they may have potential for metabolite engineering toward altering pigmentation and enhancing nutritional value of food crops.  相似文献

Dynamic changes in calcium content were investigated in eight apple cultivars. The results showed that the calcium concentration in leaves and shoots increased with fruit development. The cultivars displayed only a small difference in the calcium concentration during the early stage of development, the difference became very significant at the late stage of development, especially in shoots. In shoots, for example, calcium content was highest in Starkrimson (19 638.6 mg kg^-1) and lowest in Fuji (8 751.3 mg kg^-1). Calcium concentration was highest in young fruits and was found to decrease with the growth of fruit, and was characterized by a dramatic drop at the rapid expansion stage. There was a significant difference among cultivars. Young Starkrimson fruits contained the highest calcium concentration of 506.52 mg kg^-1 among cultivars tested, followed by Pink Lady and Fuji. The calcium concentration in mature fruits from high to low is as follows: Starkrimson, Sansa, Pink Lady, Senshu, Gala, Fuji, Red General and New Century. In this study, it was found that eight cultivars continuously assimilated calcium during the whole growing season, especially at the young stage when fruit took up 35-46% of total calcium. The calcium content in fruitlets was low in all cultivars; in the expansion stage, there was rapid absorption of about 30% of total calcium, whereas in the ripening fruit, content of calcium was reduced. The calcium accumulation increased with fruit growth in stalk, similar to that in fruit.  相似文献

The test analyzed the regularity of biosynthesis and degradation of the main functional components, such as zeaxanthin, β-carotene, and esterified carotenoids in the fruit of Lycium barbarum L. in order to provide theoretical basis for improvement of processing condition, appearance quality, and preservation of carotenoids. RP-HPLC was adapted to assay the changes of the main carotenoids of the different harvested stage fruit during the drying processing. Quantification was realized using external standard with gradient elution. The results showed that zeaxanthin and β-carotene contents in fruits increased dramatically, 2-22 times that of fresh fruits at the beginning of the drying period. In the middle of drying period, degradation occurred to a some extent, and the fall fruit degraded to a large extent. At the end of drying period, zeaxanthin and β-carotene contents increased to a little extent until a balanced state is obtained. Zeaxanthin dipalmitate content had a total degradation to more than 40% at the beginning of the drying period, and increased a little at the middle period, then reached a balanced state finally. The total carotenoid content analysis showed that the summer fruit had higher carotenoid content than the fall fruit. The experiments demonstrated zeaxanthin and β-carotene contents in fruits increased and zeaxanthin dipalmitate decreased during the drying process, which had an effect on the production appearance.  相似文献

Effects of low temperature and INA bacteria on the change of chlorophyll a fluorescence inyoung fruit from two apricot cultivars were investigated. Low temperature decreased the potential activity(Fv/Fo) ,conversion efficiency of primary light energy (Fv/Fm)of PS Ⅱ and photochemical quenching (qP) inyoung fruit of two apricot cultivars. Low temperature enhanced non-photochemical quenching qN, decreasingthe quantum yield of photosynthetic electron transfer. The presence of ice nucleating active (INA) bacteria in-tensified the effects of low temperature, raised the injury temperature threshold from - 4℃ to - 2 - - 3℃.INA bacteria can be a factor to induce frost susceptibility of apricot fruit. The amount of damaged PS Ⅱ activi-ty center was related to apricot fruit size and cultivar.  相似文献

To elucidate the response of oxidative metabolism, triggered by elevated ammonium(NH_4~+) concentrations, on root growth of wheat seedlings, Yumai 49(NH_4~+-tolerant) and Lumai 15(NH_4~+-sensitive) cultivars were supplied with either 5.0 mmol L~(–1) NH_4~+-N(EAC) or 5.0 mmol L–1 NO_3–-N(CON) under hydroponic conditions. Root growth in both cultivars was significantly reduced under EAC, and the negative effect was greater in Lumai 15. EAC enhanced the activities of monodehydroascorbate reductase and dehydroascorbate reductase in the roots of both cultivars, while it decreased ascorbic acid(ASA) content and GDP-mannose pyrophosphorylase(GMPase) activity at the 12 th day after treatment in Lumai 15 by 62.0 and 71.4%; and in Yumai 49 by 38.8 and 62.2%, respectively, indicating that the regeneration of ASA was increased, but the biosynthesis of ASA was reduced under EAC treatment. Moreover, EAC increased DHA/ASA, reactive oxygen species(ROS), and malondialdehyde contents, as well as antioxidant enzyme activities in the roots of both cultivars. Relatively greater increases in ROS and soluble sugar, and lower antioxidant enzyme activities in Lumai 15 indicate severe disruption of oxidative metabolism when compared to Yumai 49. Results reveal that the reduction of ASA biosynthesis via decreased GMPase activity under the EAC condition probably acts as a trigger for accumulated ROS and imbalanced redox status, resulting in root growth inhibition during wheat seedling growth stage. Yumai 49, being an NH_4~+-tolerant cultivar, had the stronger capacity to protect itself from oxidative stress, which allowed it to retain a lower DHA to ASA ratio by maintaining a better redox homeostasis than could be maintained in the NH_4~+-sensitive cultivar Lumai 15.  相似文献

In a field experiment, rice cultivar Shanyou 63 was used as a check cultivar to investigate the changes in activity of peroxidase and contents of malondialdehyde and soluble protein after full heading stage in flag leaves of two super high-yielding inbred rice, including Shengtai 1 and Yuxiangyouzhan. The results showed that the peroxidase activities and soluble protein contents in the leaves of the three cultivars tended to increase gradually to peak values and thereafter descended during stage after full heading. Moreover, both the peroxidase activities and soluble protein contents of Shengtai 1 and Yuxiangyouzhan peaked at 8 days after full heading, which was compared with that of Shanyou 63 at 4 days later. Meanwhile, the malondialdehyde contents in the leaves of the three cultivars increased gradually after full heading stage. In addition, activities of peroxidase and contents of malondialdehyde and soluble protein in different cultivars were various at the same time. In general, for Shengtai 1 and Yuxiangyouzhan, the peroxidase activities and soluble protein contents were higher than those of Shanyou 63, whereas the malondialdehyde contents were lower compared with that of Shanyou 63. And these features could make the senescence of leaf become later and slower and photosynthetic product was more in Shengtai 1 and Yuxiangyouzhan than that in Shanyou 63.  相似文献

Strain of Pseudomonas Lip35 producing lipase was isolated in a refrigerator. Lipase production and characterization of this strain were investigated under different conditions. The Pseudomonas was cultivated in shaking flasks in a fermentation medium in various nutritional and physical environments. Lipase production has been influenced by the presence of yeast-extract, soybean powder, NaCI, and Tween-80. Maximum lipase productivity was obtained when the physical environment of the fermentation medium was optimal for 67 h. The production of lipase reached 58.9 U·mL^-1. The lipase of Pseudomonas Lip35 can be considered to be inducible, but the inducer had little influence on the production of lipase. The lipase was characterized and showed high lipolytic activity from pH 7.5-8.0. The optimum temperature was observed at 20℃ and the thermal inactivation of lipase was obvious at 60℃. The lipase activity was inhibited by K+, stimulated by Ca^2+, and thermostability decreased in the presence of Ca^2+, therefore the lipase was Ca^2+ -dependent cold-adapted enzyme.  相似文献

This paper presents a method based on computer vision systems (CVS) to estimate shrimp dehydration level by analyzing color during drying process. Since the most commonly used color space in food industry is L*a*b, transformation of RGB digital images to L*a*b units was carried out using direct two steps model with γ factor. Experimental data obtained from images captured at different drying temperatures (100–130 °C) and several time intervals (15–180 min) were analyzed with a complete randomized block design (CRBD), and the means were compared with Duncan's multi-range test. Multiple linear regression (MLR) and artificial neural networks (ANN) were applied for correlating the color features to moisture content of dried shrimp determined chemically. Results obtained with these two models lead to 0.80 and 0.86 correlation coefficients in MLR and ANN models, respectively. While there is no statistical difference at p < 0.05 between the two modeling approaches, both approaches indicate successful prediction of shrimp dehydration with high correlation to those found by the more expensive and intrusive chemical method. The automated vision based system, therefore, has the advantage over conventional subjective methods and instrumental ones for being objective, fast, non-invasive, inexpensive and precise.  相似文献

In this study, the oxidative damage of α-terthienyl (α-T) to the Spodoptera litura (SL) cell and its mechanism were investigated. MTT was used to compare the toxicity of α-T and rotenone to the SL cell. The output of malondialdehyde and relative content of glutathione were determined with 2-thiobarbituric acid (TBA) and 5, 5'-dithio-bis (2-nitrobenzoic acid) (DTNB), respectively. Transmission electron microscope (TEM) was employed to observe the influence of α-T on the membrane and organelle of the SL cell. The result showed that the IC50 value of α-T to the SL cell was 0.21 μg mL^-1, whereas the corresponding dose of rotenone was 12.25 μg mL^-1. The output of MDA had the same changing tendency with the concentration of α-T, whereas the content of GSH had the negative correlation with it. According to TEM, the cell membrane and karyotheca swelled and couldn't retain integrity, the intracellular substances leaked out, unidentified granules appeared in the SL cell. The mitochondria expanded, and the membrane and subcellular organelle were damaged severely. In this study, it was found that after oxidative damage induced by α-T, the output of MDA increased notably, whereas the relative content of GSH decreased. This indicated that the antioxidant ability of cell weakened. The result of TEM implied that the SL cell suffered from oxidative damage under the appointed dose.  相似文献

Haynaldia villosa (2n=2X= 14, VV), a relative of wheat, plays important roles in wheat improvement mainly owing to its disease resistance. Powdery mildew resistance gene Pm21 has been successfully transferred into wheat by Cytogenetic Institute, Nanjing Agricultural University, China, and is widely used in the current wheat breeding programs. In this research, our objective is to further transfer and utilize the beneficial genes such as eye-spot resistance, yellow rust resistance, and gene of the tufted bristles on the glume ridge (a remarkable morphology) mapped on 2V of Haynaldia villosa. A disomic addition line with gametocidal chromosome 3C ofAegilops triuncialis added in Norin-26 was crossed to the wheat-H, villosa disomic substitution 2V(2D) and the hybrid F1 was then self-crossed. Chromosome C-banding, genomic in situ hybridization (GISH), and meiotic analysis in combination with molecular markers were applied to detect the chromosome variations derived from hybrids Fz and F3. To date, four translocations including one small segmental translocation T6BS·6BL-2VS, two whole arm translocations (preliminarily designed as T3DS·2VL and T2VS.7DL) and one intercalary translocation T2VS·2VL-W-2VL, one deletion Del. 2VS·2VL-, one monotelosomic Mt2VS, and one isochromosome 2VS·2VS line have been developed and characterized. One wheat SSR marker Xwmc25.120 tagging 2VS and one wheat STS marker NAU/STSBCD135-1 (2BL) tagging 2VL were successfully used to confirm the alien chromosome segments involved in the seven lines. The tufted bristles on the glume ridge appeared in lines T2VS-7DL, Mt2VS, 2VS-2VS as well as the parent DS2V(2D), whereas in T3DS·2VL, this trait did not appear. The gene controlling the tufted bristles was located on 2VS. Gametocidal chromosome 3C ofAegilops triuncialis could successfully induce chromosome 2V structural changes.  相似文献

【目的】近年来受全球气候变暖等环境影响,我国亚热带区域森林虫害呈加剧态势,开展叶片生理性状与虫害程度关系研究具有重要意义。【方法】采用典型样地调查,对刨花楠Machilus pauhoi和闽楠Phoebe bournei人工混交林受害植株健康叶片的非结构性物质含量、酶活性等主要生理性状与虫害等级关系进行研究。【结果】虫害导致刨花楠和闽楠受害植株健康叶片的相对含水量显著上升,但对叶片干物质质量分数的影响并不明显。刨花楠和闽楠健康叶片可溶性糖质量分数在中度虫害时最低。刨花楠健康叶片可溶性蛋白质量分数随虫害等级的提高而降低,闽楠健康叶片可溶性蛋白质量分数在轻度虫害时最高。健康叶片总氨基酸质量分数最高出现在刨花楠轻度虫害和闽楠中度虫害时。二者可溶性蛋白和可溶性糖质量分数的比值均在中度虫害时达到最大值,但二者可溶性蛋白和可溶性糖质量分数比值的最小值出现在不同虫害等级。轻度和中度虫害的闽楠健康叶片可溶性蛋白与可溶性糖质量分数比值差异显著。2种楠树健康叶片的过氧化物酶(POD)活性总体呈先降后升的趋势;超氧化物歧化酶(SOD)活性则均以中度虫害时最高,重度虫害时最低,各等级间差异不显著。【结论】2种楠树受相同虫害等级的影响并不总是具有一致性;2种楠树叶片酶活性对同等虫害的响应具有一致性。2种楠树的受害叶片和健康叶片具有相互补偿和自我调节作用,且2种楠树均具有自我愈伤能力和防御机制。研究结果可为进一步探讨未来环境变化下植物叶片对虫害的响应机制,开展人工林虫害防治技术研究等提供理论依据。  相似文献

Eggplant is one of the most common vegetable crops grown in India and other parts of the world. Leucinodes orbonalis is the key pest infesting eggplant, with international importance as it causes heavy damage in all eggplant growing areas. It is very difficult to control this pest since it feeds inside the shoot and fruit, and there is every possibility of toxic residues remaining in the fruits as they are harvested at frequent intervals and are consumed after little cooking. In the sub-himalayan region of West Bengal, India the pest was found to be most active during summer and the rainy season, particularly during May–August, and caused 49.5–81.0% damage to fruits. Peak infestation (81.0% fruit damage) was noticed in the first week of June (22nd standard week), when the mean temperature, mean relative humidity and weekly rainfall were 27.8°C, 79.2% and 81.2 mm respectively. The pest became less active during winter months particularly in December–January. Borer infestation showed a significant positive correlation (P = 0.05) with maximum and mean temperature, minimum and mean relative humidity and rainfall, whereas with maximum relative humidity the correlation was negative but non-significant. The insecticides evaluated in the field for L. orbonalis control on eggplant revealed that avermectin (Vertimec 1.9 EC; 0.5 ml/l) was the most effective in suppressing dead heart caused by the pest, closely followed by Beauveria bassiana (Biorin 107 conidia/ml; 1 ml/l) and Bacillus thuringiensis Berliner (Biolep 5 × 107 spores/ml; 1 g/l). Significantly lower levels of fruit damage were recorded after avermectin treatment, closely followed by DDVP (0.05%) (Nuvan 76 SL; 2 ml/3 l) when compared with the untreated control. The other insecticides tested, including a neem formulation (Neemactin 0.15 EC; 2.5 ml/l) and malathion (0.05%) (Malathion 50 EC; 1.0 ml/l), were less effective. None of the insecticides evaluated produced satisfactory results against L. orbonalis. However, avermectin, besides being environmentally safe, was effective for a longer duration and could thus be recommended for Integrated Pest Management programmes on eggplant.  相似文献

通过对浮萍表观生长状态的观察记录以及对其抗氧化酶、丙二醛含量的测定,研究泽漆水浸液对浮萍(Lemna minor L.)的抑制活性,并探索利用茶皂素作为浸提辅助剂,检测是否提高泽漆对浮萍的抑制作用。结果表明:低浓度泽漆水浸液(0.001、0.003、0.005 g/mL)胁迫下,浮萍部分抗氧化酶系统遭受破坏,过氧化氢酶、过氧化物酶活性下降,超氧化物歧化酶活性先升高后降低,未出现明显表观受伤症状;高浓度泽漆水浸液(0.01、0.015、0.02 g/mL)胁迫36 h,90%~100%个体受伤严重甚至死亡,受伤个体、程度与浓度、时间成正比,茶皂素助浸液胁迫36 h时,出现明显受伤症状的个体数≤10%。茶皂素作为浸提辅助剂所得泽漆浸提液没有加强对浮萍的抑制作用,反而减弱了这种作用。泽漆水浸液对浮萍有明显的抑制作用,今后可进一步分析泽漆水浸液和其茶皂素助浸液之间化学成分种类和含量的差异,这有助于抑制浮萍有效成分的鉴定。  相似文献

选用60日龄的盆栽早小洋菊和晚小洋菊菊苗接种菊小长管蚜无翅成蚜,15 d后发现前者的蚜数和蚜虫繁殖倍率皆显著地大于后者。检测了2个品种株高、分枝数、叶片长和宽、叶片长宽比、叶裂数等株形特征,测定二者叶片上、下表皮的亮度L*、色度坐标a*b*、饱和度C、总色值E*等色度特征,并分析了各种特征的数值与蚜数相关性。结果显示,叶片长宽比、叶片上表皮L*、a*、b*皆分别与蚜数正相关 (P<0.01);早小洋菊叶片长宽比显著大于晚小洋菊 (P<0.05);早小洋菊上表皮L*、a*、b*和C分别大于晚小洋菊(P<0.05)。结论认为,该蚜嗜好叶片形状狭长、叶色更黄绿更加鲜艳明亮的早小洋菊。  相似文献
