Altogether 496 samples of meat, lymph nodes, process water and swabs from different places in the abattoir were examined for the presence of Listeria spp. L. monocytogenes was isolated from 31 (6%) and other Listeria spp. from 65 (13%) samples. L. monocytogenes was isolated from 2 of 10 beef meat samples, 4 of 50 pig meat samples and 1 of 21 lymph nodes of pigs. No Listeria bacteria were isolated from lymph nodes of cattle. The highest percentage of Listeria was recovered from the unclean sections (cattle 22% and pigs 27% ) and the highest frequency was observed during the winter months.  相似文献

The objective of the study was to investigate reservoirs and transmission of S. aureus in ewes and lambs in 3 meat sheep flocks. Repeated sampling of milk, teat skin, nasal- and vaginal mucous membranes was performed and samples were analysed for S. aureus. For comparison, samples were also collected from cows and young heifers in 3 dairy cattle herds. Selected isolates were compared by pulsed-field gel electrophoresis (PFGE). S. aureus was detected in 8 (1.5%) of 520 milk samples from ewes and in 38 (6.4%) of 588 milk samples from cows. From body site swabs, S. aureus was found in 394 (32.6%) of 1208 samples from sheep and in 67 (16.0%) of 420 samples from cattle. The proportion of S. aureus-positive nasal swabs from ewes and cows were 56.7% and 13.9%, respectively. From lambs, 58.2% of the nasal swabs were S. aureus-positive. In each flock, one S. aureus pulsotype predominated. Identical S. aureus pulsotypes were found in milk and from body sites. Paired S. aureus isolates from the nasal cavity of (i) ewes and their lambs, (ii) twins and (iii) from repeated swabs of individual ewes were compared by PFGE, and in the majority of cases the two isolates were identical. The results contribute new knowledge indicating frequent transmission of S. aureus between the dam and her lambs and within animals in a flock. In contrast to cattle, S. aureus is frequently present in the nose of sheep which may represent the primary reservoir of S. aureus in sheep flocks.  相似文献

The aim of this study was to compare the prevalence of Salmonella serotypes at two different sites on pig carcasses from herds classified as high-risk or low-risk and to elucidate the relationship between carcass contamination levels and serological status. Caecal samples and carcass surface swabs were cultured for Salmonella from a total of 210 pigs from low risk herds (< 19% of pigs in herd Salmonella seropositive) and 209 pigs from high risk herds (> 32% of pigs in herd Salmonella seropositive) in three abattoirs. Meat juice samples were collected for analysis by ELISA. The prevalence of Salmonella in the caecal contents of "low-risk" pigs was 10%, which was significantly lower than the 19% prevalence in "high-risk" pigs (p < 0.01). The corresponding figures for skin samples collected immediately post-evisceration were 2% and 12%. The predominant Salmonella serotype in the caecal contents of both the low-risk and high-risk pigs was Salmonella Typhimurium. Salmonella Kentucky and Salmonella Derby were the most frequent isolates from the carcass surface swabs of low- and high-risk pigs respectively. There was a positive association between seropositivity of pigs from high-risk herds and caecal carriage (p < 0.05). Results showed that herd categorisation based on serological results was useful in predicting Salmonella isolation rates from caecal samples and surface swabs of slaughtered pigs.  相似文献

Fecal samples collected from cattle at processing during a 1-year period were tested for verotoxins (VT1, VT2), Escherichia coli O157:H7, and Salmonella. Verotoxins were detected in 42.6% (95% CI, 39.8% to 45.4%), E. coli O157:H7 in 7.5% (95% CI, 6.1% to 9.1%), and Salmonella in 0.08% (95% CI, 0.004% to 0.5%) of the fecal samples. In yearling cattle, the median within-lot prevalence (percentage of positive samples within a lot) was 40% (range, 0% to 100%) for verotoxins and 0% for E. coli O157:H7 (range, 0% to 100%) and Salmonella (range, 0% to 17%). One or more fecal samples were positive for verotoxins in 80.4% (95% CI, 72.8% to 86.4%) of the lots of yearling cattle, whereas E. coli O157:H7 were detected in 33.6% (95% CI, 26.0% to 42.0%) of the lots. In cull cows, the median within-lot prevalence was 50% (range, 0% to 100%) for verotoxins and 0% (range, 0% to 100%) for E. coli O157:H7 and Salmonella (range, 0% to 0%). Verotoxins were detected in one or more fecal samples from 78.0% (95% CI, 70.4% to 84.2%) of the lots of cull cows, whereas E. coli O157:H7 were detected in only 6.0% (95% CI, 3.0% to 11.4%) of the lots of cull cows. The prevalence of verotoxins in fecal samples was lower in yearling cattle than in cull cows, whereas the prevalence of E. coli O157:H7 in fecal samples was higher in yearling cattle than in cull cows. The prevalence of E. coli O157:H7 in fecal samples was highest in the summer months. Rumen fill, body condition score, sex, type of cattle (dairy, beef), and distance travelled to the plant were not associated with the fecal prevalence of verotoxins or E. coli O157:H7. The prevalence of verotoxins in fecal samples of cull cows was associated with the source of the cattle. It was highest in cows from the auction market (52%) and farm/ranch (47%) and lowest in cows from the feedlot (31%). In rumen samples, the prevalence of verotoxins was 6.4% (95% CI, 4.2% to 9.4%), and it was 0.8% (95% CI, 0.2% to 2.3%) for E. coli O157:H7, and 0.3% (95% CI, 0.007% to 1.5%) for Salmonella.  相似文献

Thirty-one farrow-to-finish pig units with a Zoonoses National Control Programme (ZNCP) for Salmonella above the UK target of 10 per cent during the previous 12 months were selected for the study. Pooled faecal samples were collected from different groups of pigs. Furthermore, mice, rat and bird faeces and carcases were collected, if found on the unit. In total, 937 samples were collected on-farm and analysed for Salmonella. The four carcases selected monthly per producer by the slaughterhouse for the British Pig Executive ZNCP Salmonella Programme were tested for antibodies to groups B and C with a mix-ELISA test. The same four carcases were swabbed externally and internally, mesenteric lymph nodes (MLNs) were collected and colonic contents were swabbed. A wide variety of Salmonella serovars was isolated from the samples. Most of the isolates were detected in the rearing herd (on-farm) and in the MLNs (slaughterhouse). There was no correlation between ELISA results from the meat juice and bacteriological isolations using Spearman correlation analyses. Furthermore, no significant association was found between positive results of ELISA and positive results from the bacteriological samples taken.  相似文献

Summary The cervical lymph nodes of pigs, the retropharyngeal and submandibular lymph nodes of cattle and faecal samples from both animal species were examined for the presence of Corynebacterium equi. The organism was recovered from 19 (35 per cent) of 54 porcine cervical lymph nodes and from 0 of 54 bovine retropharyngeal and submandibular lymph nodes. Fifteen (50 per cent) of 30 bovine faecal and 11 (35 per cent) of 31 porcine faecal samples yielded C. equi.  相似文献

An outbreak of tuberculosis in pigs and cattle caused by Mycobacterium africanum produced lesions in the pigs similar to those caused by M tuberculosis, M bovis and M avium, with caseation in the lymph nodes of the head and in the jejunal lymph nodes. Bacteriological examination of the dysgonic mycobacteria isolated showed that they were M africanum I. The intradermal tuberculin test was very reliable in pigs, differentiating between mammalian and avian reactions, and the results of the test were in accordance with the lesions found at meat inspection. No clinical signs were observed during the outbreak, and the infection was neither serious nor progressive. There were no lesions in the tuberculin-positive cattle. The source of the infection remains unknown.  相似文献

OBJECTIVE: To compare the frequency of isolation, genotypes, and in vivo production of major lethal toxins of Clostridium perfringens in adult dairy cows affected with hemorrhagic bowel syndrome (HBS) versus left-displaced abomasum (LDA). DESIGN: Case-control study. ANIMALS: 10 adult dairy cattle with HBS (cases) and 10 adult dairy cattle with LDA matched with cases by herd of origin (controls). PROCEDURE: Samples of gastrointestinal contents were obtained from multiple sites during surgery or necropsy examination. Each sample underwent testing for anaerobic bacteria by use of 3 culture methods. The genotype of isolates of C. perfringens was determined via multiplex polymerase chain reaction assay. Major lethal toxins were detected by use of an ELISA. Data were analyzed with multivariable logistic regression and chi2 analysis. RESULTS: C. perfringens type A and type A with the beta2 gene (A + beta2) were the only genotypes isolated. Isolation of C. perfringens type A and type A + beta2 was 6.56 and 3.3 times as likely, respectively, to occur in samples from cattle with HBS than in cattle with LDA. Alpha toxin was detected in 7 of 36 samples from cases and in 0 of 32 samples from controls. Beta2 toxin was detected in 9 of 36 samples from cases and 0 of 36 samples from controls. CONCLUSIONS AND CLINICAL RELEVANCE: C. perfringens type A and type A + beta2 can be isolated from the gastrointestinal tract with significantly greater odds in cattle with HBS than in herdmates with LDA. Alpha and beta2 toxins were detected in samples from cows with HBS but not from cows with LDA.  相似文献

In a field study with fattening pigs, effects of feed particle size at the dietary presence of organic acids on Salmonella prevalence were measured. On two farms (f1/f2), each holding ~800 pigs, diets based on finely ground (control) or coarsely ground ingredients (experiment) were fed as crumbs. On f1 both control and experimental grower and finisher diets contained identical concentrations of formic and propionic acid (0.4% and 0.2% respectively). On f2 only finisher diet of the experimental group contained 1.2% potassium diformate. At the start of the fattening period no statistical differences were measured between Salmonella prevalence in animals fed control and experimental diets on both farms. At slaughter Salmonella prevalence in caecal contents was lower (p < 0.05) on f1 in animals fed the experimental diet. Furthermore, the number of seronegative meat juice samples taken from these animals [optical density (OD) <10] was higher (p < 0.001); seropositive as well as distinct seropositive samples (OD ≥20 and ≥40 respectively) were less frequent (p < 0.01) compared to samples from animals fed the control diet. Feeding the experimental diet on f2 resulted in a lower Salmonella detection rate in faeces before slaughter (p < 0.01). Salmonella prevalence was lower in caecal content at slaughter for pigs fed the experimental diet compared to those fed control diet (p < 0.0001). The number of distinct seropositive meat juice samples (OD ≥40) was lower (p < 0.01) for pigs fed the experimental than for those fed the control diet. In comparison to pigs in the control group, starch concentrations in the caecal content from pigs in the experimental groups on both farms were higher (p < 0.05) and the pH values lower (p < 0.05). Propionate (p < 0.0001) and butyrate (p < 0.01) concentrations were higher in the caecal content taken from pigs in the experimental group on f2.  相似文献

Meat samples from diaphragm pillars were randomly taken from 3,048 pigs of 52 Bavarian herds after slaughtery. Meat-juice was collected and tested for salmonella antibodies in an indirect ELISA. The number of samples was calculated according to the annual production of slaughter pigs of a farm outlined in the "Leitlinien für ein Programm zur Reduzierung des Eintrags von Salmonellen durch Schlachtschweine in die Fleischgewinnung" from February 05th, 1998 (< 100 slaughter pigs: 45 samples, 100-200 slaughter pigs: 50 samples, > 200 slaughter pigs: 60 samples per year). Salmonella antibodies were detected in 48 carcasses (1.6%) of 12 farms (23.1%). However, 33 (68.8%) of these carcasses were originated from a single farm which had to be classified into category III (prevalence of > 40% in the samples). No bacteria could be isolated from this farm in a follow up examination. The 51 other farms (98%) were classified into category I (prevalence of < 20% in the samples). Farms with in/out-management showed a higher degree of reagents (2.1T%) than farms with continuous stabling (0.8T%). In a pig experimentally immunized with LPS-antigen preparations of Salmonella typhimurium it was shown that antibodies induced were nearly at the same level in all meat samples and even in selected organs (liver, kidney, parotis, mesenteric lymph nodes).  相似文献

Bile samples were collected from 477 Iowa dairy cows and were cultured for thermophilic campylobacters. The prevalence of thermophilic campylobacters in the bile was 15.5%. Campylobacter jejuni and C coli from dairy cattle, chickens, pigs, sheep, and human beings were serotyped to develop host-species profiles. Human and cattle serologic profiles were the most similar, and human and chicken profiles shared several similarities. Epidemiologic data from 168 human cases of campylobacteriosis indicated that 23% of the cases were associated with consumption of unpasteurized milk.  相似文献

Arginine-utilising mycoplasmas isolated from the pneumonic lungs of 4 pigs were identified as M. hyosynoviae by their growth characteristics, biochemical activity and serological similarity to each other and to 2 type strains of M. hyosynoviae in growth inhibition tests. A purified culture of one M. hyosynoviae isolate was inoculated intravenously into 3 pigs which were killed 9 days later. Although arthritis was not observed clinically, M. hyosynoviae was isolated from 8 of 9 joints cultured, the tonsils and retropharyngeal lymph nodes of all 3 pigs and the lungs of 2 pigs. No histopathological changes were observed in synovial membranes of the joints cultured or lungs but severe depletion of lymphocytes in the cortex of lymphoid follicles in lymph nodes and tonsils was observed.  相似文献

Most Salmonella control programmes are based on serological testing in the slaughterhouse. However, from a point of view of carcass contamination, it is rather the presence of Salmonella spp. in the animal at the time of slaughter that is important. The aim of this cross-sectional study was to investigate the possible discrepancies between the isolation of Salmonella spp. in the mesenteric lymph nodes and the results of serological screening. In total, 1821 fattening pigs originating from 60 Belgian farrow-to-finish herds were sampled in the slaughterhouse. The serum samples were analysed using an indirect mix-ELISA for the presence of Salmonella antibodies and evaluated at 3 cut-off values namely 10, 20, and 40% Optical Density (OD). All mesenteric lymph node samples were submitted to qualitative Salmonella isolation and a representative number of isolates was serotyped. From each herd, 30 animals were screened both serologically and bacteriologically and the herd was considered as positive when at least one sample was positive. At the herd level, 83.6% (cut-off OD 40%) to 100.0% (cut-off OD 10%) of the herds from which Salmonella had been isolated were evaluated as seropositive. At the individual level, only 34.5% (cut-off OD 40%) to 82.8% (cut-off OD 10%) of the animals from which Salmonella had been isolated were seropositive. Overall, a weak agreement was found between bacteriology and serology for Salmonella diagnosis. If pig herds are categorised using serological tests in the slaughterhouse, one should be aware of the fact that slaughter pigs can still harbour Salmonella spp. in the mesenteric lymph nodes, without being detected in serological tests. The cut-off value used to evaluate a sample as serologically positive and the number of samples per herd are of major importance to classify herds correctly in order to protect human health.  相似文献

We examined patterns of lymphocyte localization in female dairy cattle following infusion of 51Cr-labeled autologous lymphocytes prepared from surgically excised mammary or ileal mesenteric lymph nodes. Labeled lymphocytes prepared from mammary lymph nodes were recovered in proportionally high numbers from mammary and prescapular lymph nodes, and in low numbers from intestinal mesenteric nodes. This pattern was observed in both heifers and lactating cows. In contrast, labeled lymphocytes prepared from ileal mesenteric lymph nodes of lactating cows were recovered in proportionally high numbers from intestinal mesenteric nodes, and in low numbers from mammary and prescapular nodes. These findings, when compared with previous results in sheep and swine, support the hypothesis that lymphocytes do not migrate efficiently between the gut and mammary gland of ruminants.  相似文献

Each of 12 cattle was inoculated either subcutaneously and intradermally or into a mesenteric lymph node with 1 of 8 species of live atypical mycobacteria isolated from cattle, cattle trough water and feral pigs. Seventy-eight days after inoculation the cattle were tuberculin tested with bovine PPD, avian PPD and homologous heat-concentrated syntheic medium tuberculins. They were killed 85 days after inoculation. Organisms were cultured from caseous granu-lomas at all sites in cattle inoculated with M. avium serotype 2. M. simiae was recovered from a granuloma at the subcutaneous site. Acid-fast bacilli were isolated from the mesenteric lymph node inoculated with trough water organisms. At 72 h, all the cattle had produced skin reactions of 4 mm or more to the homologous tuberculins and all except 1 produced a similar response to avian PPD. Only isolates of bovine origin sensitised cattle to bovine PPD to this degree, and these reactions were less than the corresponding response to avian PPD.  相似文献

The goal of this study was to estimate the accuracy of the invA-gene-based polymerase chain reaction (PCR) and a culture technique based on pre-enrichment with buffered peptone water, three selective enrichment media (selenite, tetrathionate and Rappaport-Vassiliadis broths) and four selective, solid media (Xylose-Lysine-Tergitol-4, Salmonella/Shigella, Hekton-Enteric and MacConkey), for the detection of Salmonella organisms from caecal samples from slaughter pigs. For this purpose a latent-class (Bayesian) approach was used. Two hundred and three slaughtered pigs were used after grouping them into two groups of 96 and 107 animals. Sensitivity (Se) was estimated to be 56% (95% probability interval 40, 76) for culture and 91% (81, 97) for PCR. The specificity (Sp) of the PCR was 88% (80, 95) while the Sp of the culture had been considered 100% in the statistical analysis as all culture-positive samples were confirmed by serotyping. PCR Se was not affected by the Salmonella serotypes present in the samples analysed. Accordingly, a minimum of 25.5% of the pigs was estimated to harbour Salmonella organisms in their faeces. It was concluded that bacteriology on caecal samples alone was a poor diagnostic method, and that the PCR method could be considered a cost-effective alternative to culture in Salmonella monitoring programmes. However, given the moderate Sp of this molecular technique, PCR-positive samples should be further confirmed through bacteriology.  相似文献

Twelve pigs were inoculated orally with Mycobacterium avium. The doses used were 0.5, 2 or 10 mg daily for 5 days, or 10, 50 or 180 mg once (1 mg = 37 × 106 viable units). Two pigs were used per dose, 1 of which was sacrificed 3 days, the other 28/31 days after the last inoculation (Table 1).Three days after inoculation, M. avium was found in the tonsils and in the intestinal mucosa of all 6 pigs, and in the mesenteric lymph nodes of 4. Viable unit counts for tonsils and intestinal mucosa were highest in pigs inoculated with 180 mg×1 and 10 mg×5. Histopathologically these pigs showed activation of the lymphoid tissue in the tonsils, Peyer patches and mesenteric lymph nodes. Twenty-eight/31 days after inoculation a spreading of the infection had taken place in all pigs, most often to the liver, less frequently to the spleen and the lungs. The kidneys and the musculature were not infected (Table 4). A correlation was apparent between the size of dose and the number of viable organisms in the tissues. Divided doses gave about 10 times higher viable counts than a single dose with the same total number of organisms (Table 5).No gross lesions were found 28/31 days after inoculation. Microscopic granulomatous lesions were found in the tonsils of 6 pigs, in the intestinal mucosa of 4 pigs, in the mandibular and mesenteric lymph nodes of 6 pigs, in the retropharyngeal lymph nodes of 3 pigs, and less frequently in the parotid and hepatic lymph nodes (Table 3).Five of 6 pigs were weakly sensitive to avian tuberculin PPD, 1000 t.u. per dose, when tested 22/25 days after inoculation; 1 of these pigs cross-reacted to human tuberculin (Table 2).  相似文献
