Human T cell leukemia viruses use a receptor determined by human chromosome 17   总被引:33,自引:0,他引:33  
Human T cell leukemia viruses (HTLV-I and HTLV-II) can infect many cell types in vitro. HTLV-I and HTLV-II use the same cell surface receptor, as shown by interference with syncytium formation and with infection by vesicular stomatitis virus (VSV) pseudotypes bearing the HTLV envelope glycoproteins. Human-mouse somatic cell hybrids were used to determine which human chromosome was required to confer susceptibility to VSV(HTLV) infection. The only human chromosome common to all susceptible cell hybrids was chromosome 17, and the receptor gene was localized to 17cen-qter. Antibodies to surface antigens known to be determined by genes on 17q did not block the HTLV receptor.  相似文献

Analysis of the ability of heteroantisera, monoclonal antibodies, and antibodies to synthetic peptides to react with viral glycoproteins deglycosylated with endoglycosidase F revealed that the reactivities of most of the antibodies to these glycoprotein antigens were influenced by the attached carbohydrate moieties. All heteroantisera prepared in rabbits or goats to either fully glycosylated retroviruses or influenza virus were virtually unreactive toward the viral glycoproteins after carbohydrate removal. Analyses with a panel of monoclonal antibodies to purified Rauscher murine leukemia virus gp70 indicated that the reactivity of most of these antibodies improved while the reactivity of others decreased or remain unchanged after carbohydrate removal. Most of the antibodies to synthetic peptide sequences in the influenza virus hemagglutinin also improved in reactivity after carbohydrate removal. These data indicate that carbohydrate side chains on viral glycoproteins influence the immune response to these antigens, and the more native the glycoprotein immunogen, the more dramatic the carbohydrate influence. Thus the immune response to these glycoproteins is not simply a function of the immunogenicity of certain domains over others but rather is a direct measure of carbohydrate influences on the host's perception of the foreign antigen.  相似文献

继排除紫色卷叶病非细菌、真菌、线虫病引起外,围绕病毒、营养匮乏、药剂和害虫分泌的毒物与紫色卷叶病关系,开展剑麻植株体内点滴营养与药剂及非抗性麻苗接种已在南瓜上脱毒3~4代的粉蚧若虫,探讨对紫色卷叶病发生的影响。初步结果为营养和药剂对该病没作用或作用不大;该病与粉蚧(新菠萝灰粉蚧)害虫分泌的毒物有密切关系。  相似文献

In this study, two glycoproteins (gp160 and gp120) that are encoded by human T-cell lymphoma virus type III (HTLV-III) were the antigens most consistently recognized by antibodies found in patients with the acquired immune deficiency syndrome (AIDS) and with the AIDS-related complex (ARC) and in healthy homosexual males. The techniques used to detect the glycoproteins were radioimmunoprecipitation and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (RIP/SDS-PAGE). Although most antibody-positive samples from ARC patients and from healthy homosexual males also reacted with the virus core protein p24, less than half of the AIDS patients revealed a positive band with p24 under the same conditions. The ability to detect antibodies against a profile of both the major env gene encoded antigens and the gag gene encoded antigens suggests that the RIP/SDS-PAGE may be a valuable confirmatory assay for establishing the presence or absence of antibodies to HTLV-III in human serum samples.  相似文献

In an examination of the molecular basis of oral transmission of bunyaviruses by mosquitoes., La Crosse (LAC), snowshoe hare (SSH), and LAC-SSH reassortant viruses were compared in their ability to be transmitted to laboratory mice by the natural mosquito vector of LAC virus, Aedes triseriatus. Both LAC virus and the reassortment viruses containing the middle-sized (M) segment from the LAC parent were efficiently transmitted. In contrast, SSH virus and reassortment viruses containing the M RNA from the SSH parent were inefficiently transmitted. Thus, the M RNA segment, which codes for the virion glycoproteins, may be a major determinant of oral transmission of bunyaviruses by mosquitoes.  相似文献

The protein coding region of the herpes simplex virus type-1 glycoprotein D (gD) gene was mapped, and the nucleotide sequence was determined. The predicted amino acid sequence of the gD polypeptide was found to contain a number of features in common with other virus glycoproteins. Insertion of this protein coding region into a bacterial expressor plasmid enabled synthesis in Escherichia coli of an immunoreactive gD-related polypeptide. The potential of this system for preparation of a type-common herpes simplex virus vaccine is discussed.  相似文献

Antibodies from the serum of patients with the acquired immune deficiency syndrome (AIDS) or with the AIDS-related complex and from the serum of seropositive healthy homosexuals, recognize two major glycoproteins in cells infected with human T-cell lymphotropic virus type III (HTLV III). These glycoproteins, gp160 and gp120, are encoded by the 2.5-kilobase open reading frame located in the 3' end of the HTLV-III genome, as determined by amino terminus sequence analysis of the radiolabeled forms of these proteins. It is hypothesized that gp160 and gp120 represent the major species of virus-encoded envelope gene products for HTLV-III.  相似文献

Almost all of the key molecules involved in the innate and adaptive immune response are glycoproteins. In the cellular immune system, specific glycoforms are involved in the folding, quality control, and assembly of peptide-loaded major histocompatibility complex (MHC) antigens and the T cell receptor complex. Although some glycopeptide antigens are presented by the MHC, the generation of peptide antigens from glycoproteins may require enzymatic removal of sugars before the protein can be cleaved. Oligosaccharides attached to glycoproteins in the junction between T cells and antigen-presenting cells help to orient binding faces, provide protease protection, and restrict nonspecific lateral protein-protein interactions. In the humoral immune system, all of the immunoglobulins and most of the complement components are glycosylated. Although a major function for sugars is to contribute to the stability of the proteins to which they are attached, specific glycoforms are involved in recognition events. For example, in rheumatoid arthritis, an autoimmune disease, agalactosylated glycoforms of aggregated immunoglobulin G may induce association with the mannose-binding lectin and contribute to the pathology.  相似文献

A variety of molecular chaperones and folding enzymes assist the folding of newly synthesized proteins in the endoplasmic reticulum. Here we investigated why some glycoproteins interact with the molecular chaperone BiP, and others with the calnexin/calreticulin pathway. The folding of Semliki forest virus glycoproteins and influenza hemagglutinin was studied in living cells. The initial choice of chaperone depended on the location of N-linked glycans in the growing nascent chain. Direct interaction with calnexin and calreticulin without prior interaction with BiP occurred if glycans were present within about 50 residues of the protein's NH2-terminus.  相似文献

Reticuloendotheliosis virus (REV) causes the atrophy of immune organs and immuno-suppression in chickens, but the underlying molecular mechanism of the immune response after infection by REV is not well understood. Presently, the RNA-seq was used to analyze the regulation of immune response to REV in chicken lymphocytes from peripheral blood. Overall, 134 differentially expressed long non-coding RNAs (lncRNAs) between cells with REV infection or without in vitro were screened. Based on the differentially expressed protein-coding genes, the nucleotide-binding oligomerization domain (NOD)-like receptor pathway related to immune regulation was enriched. Two lncRNAs (L11530 and L09863) were predicted to target the NOD1 and tumor necrosis factor receptor-associated factor 5 (TRAF5) gene, respectively, which are involved in the NOD-like receptor pathway with cis-regulation way. The in vitro results revealed the significantly up-regulated (P<0.01) levels of lncRNA-L11530 and its target gene, NOD1, and the significantly down-regulated (P<0.05) levels of lncRNA-L09863 and its target gene, TRAF5, in lymphocytes after REV infection. These changes also occurred in vivo in blood lymphocytes of chickens infected with REV. Further, L09863 and L11530 were respectively interfered, the expression levels of their target genes NOD1 or TRAF5 were significantly down-regulated, accompanied by the change of IL-8 and IL-18 secretions in lymphocytes. The NOD-like receptor pathway appears to be important in the immune response to REV, LncRNA-11530 and IncRNA-09863 might involve in the immune regulation on REV infection by targeting NOD1 or TRAF5 in blood lymphocytes of chickens. Our findings reveal a new regulation of lncRNAs (L11530 and L09863) on immunity in chicken peripheral blood lymphocytes for REV infection by changing the expression of the target genes via the NOD-like receptor pathway.  相似文献

[目的]研究大鲵皮肤分泌物的气味成分及其除臭方法.[方法]采用静态顶空法从大鲵皮肤分泌物中收集气体,经气质联用色谱分析挥发性成分,并利用乙醇溶解、蛋白酶解、活性炭吸附的方法对其进行脱臭.[结果]大鲵皮肤分泌物气味成分共有9种挥发性物质,包括二甲基二硫醚(C2H6S2)和8种脂类化合物;大鲵分泌物中臭味成分为二甲基二硫醚.不同方法对大鲵皮肤分泌物的脱臭效果由高到低依次为乙醇>复合酶解>活性炭.大鲵皮肤分泌物经过乙醇脱臭和酶解脱臭后,无臭味,均未检出二甲基二硫醚.[结论]该研究可为大鲵资源的开发和利用奠定基础.  相似文献

Cloning and Sequence of Glycoprotein H Gene of Duck Plague Virus   总被引:9,自引:0,他引:9  
The glycoprotein H (gH) gene homologue of duck plague virus (DPV) was cloned by degenerate polymerase chain reaction (PCR) and sequenced. It was located immediately downstream from the thymidine kinase gene (TK). In addition, the 3'-end of the gene homologue to herpesvirus UL21 was located downstream from the gH gene. DPV gH gene open reading frame (ORF) was 2 505 bp in length and its primary translation product was a polypeptide of 834 amino acids long. It possessed several characteristics of membrane glycoproteins, including an N-terminal hydrophobic signal sequence, an external domain containing eight putative N-linked glycosylation sites, a C-terminal transmembrane domain, and a charged cytoplasmic tail. Comparison with other herpesvirus revealed identities of 20.2, 25.1, 23.0, 23.0, 26.5 and 26.0% with the gH counterparts of the human herpesvirus virus 1 (HSV1), equine herpesvirus 4 (EHV4), bovine herpesvirus 1 (BHV1), pseudorabies virus (PRV), gallid herpesvirus 2 (GHV2) and gallid herpesvirus 3 (GHV3), respectively.  相似文献

Evidence was presented earlier that a host-cell receptor for the highly neurotropic rabies virus might be the acetylcholine receptor. The amino acid sequence of the glycoprotein of rabies virus was compared by computer analysis with that of snake venom curaremimetic neurotoxins, potent ligands of the acetylcholine receptor. A statistically significant sequence relation was found between a segment of the rabies glycoprotein and the entire sequence of long neurotoxins. The greatest identity occurs with residues considered most important in neurotoxicity, including those interacting with the acetylcholine binding site of the acetylcholine receptor. Because of the similarity between the glycoprotein and the receptor-binding region of the neurotoxins, this region of the viral glycoprotein may function as a recognition site for the acetylcholine receptor. Direct binding of the rabies virus glycoprotein to the acetylcholine receptor could contribute to the neurotropism of this virus.  相似文献

West African HIV-2-related human retrovirus with attenuated cytopathicity   总被引:22,自引:0,他引:22  
Clinical and seroepidemiological studies in West Africa indicate that human immunodeficiency virus type 2 (HIV-2) is widespread and associated with immunodeficiency states of variable degree. In this study, an isolate of HIV-2 from a patient in Senegal was molecularly cloned and characterized. This isolate (HIV-2ST) was shown by hybridization and restriction enzyme analysis to be more related to the prototype HIV-2ROD than to other human or primate retroviruses. Cultures of HIV-2ST showed genotypic polymorphism, and clones of the virus had transmembrane envelope glycoproteins of 30 and 42 kilodaltons. Unlike other immunodeficiency viruses, HIV-2ST did not cause cell death or induce cell fusion in peripheral blood lymphocytes or in any of four CD4+ cell lines tested. Although HIV-2ST entered cells by a CD4-dependent mechanism and replicated actively, cell-free transmission of the virus was retarded at the level of cell entry. These findings suggest that immunodeficiency viruses prevalent in West African populations are members of the HIV-2 virus group and that certain strains of this virus have attenuated virulence.  相似文献

Glycoprotein B (gB) is the most conserved component of the complex cell-entry machinery of herpes viruses. A crystal structure of the gB ectodomain from herpes simplex virus type 1 reveals a multidomain trimer with unexpected homology to glycoprotein G from vesicular stomatitis virus (VSV G). An alpha-helical coiled-coil core relates gB to class I viral membrane fusion glycoproteins; two extended beta hairpins with hydrophobic tips, homologous to fusion peptides in VSV G, relate gB to class II fusion proteins. Members of both classes accomplish fusion through a large-scale conformational change, triggered by a signal from a receptor-binding component. The domain connectivity within a gB monomer would permit such a rearrangement, including long-range translocations linked to viral and cellular membranes.  相似文献

[目的]了解不同培养基和储存条件对家蝇幼虫分泌物中溶菌酶含量的影响。[方法]分别以猪粪和鸡粪为培养基饲养家蝇幼虫,分别为猪粪处理组和鸡粪处理组,用麦麸饲养幼虫为正常对照组,麦麸中添加大肠杆菌菌液饲养幼虫为诱导对照组,分别提取各组2龄和3龄家蝇幼虫的分泌物,使用试剂盒检测分泌物中溶菌酶含量;分别提取正常对照组和诱导对照组中3龄幼虫的分泌物,测定在室温、4℃和-20℃下不同时间溶菌酶含量的变化。[结果]猪粪处理组、鸡粪处理组、正常对照组和诱导对照组中分离的3龄幼虫分泌物中溶菌酶的平均含量分别为2.462、2.782、1.019、2.117μg/m L,均显著高于2龄幼虫(P0.05)。猪粪处理组、鸡粪处理组和诱导对照组2龄和3龄幼虫分泌物的溶菌酶含量均高于正常对照组(P0.05)。在室温下,正常对照组和诱导对照组中溶菌酶的活性均仅能维持14 d左右;在-20℃下,溶菌酶的活性均可维持70 d左右;在4℃下,正常对照组和诱导对照组中溶菌酶活性的维持时间介于室温和-20℃之间,且2组间溶菌酶含量随时间的变化规律有一定差异。[结论]3龄蝇蛆分泌物中溶菌酶含量高于2龄蝇蛆;鸡粪培养基和猪粪培养基能显著增强家蝇2龄和3龄幼虫分泌物中溶菌酶含量;在-20℃储存条件下,家蝇溶菌酶可在较长时间内保持较高的活性。  相似文献
