This paper is the first report of the production of a dermonecrotic toxin by pasteurella strains that do not belong to the species Pasteurella multocida subspecies multocida. Four strains, isolated from cattle with atrophic rhinitis, were characterised phenotypically. The strains were related to pasteurellaceae, but their taxonomic position remained unclear. The strains produced a toxin that caused a haemorrhagic dermonecrosis in guinea pigs and was lethal to mice. Both effects were neutralised by an antiserum against the purified dermonecrotic toxin of P multocida subspecies multocida. Western blot analysis of culture filtrates of the bovine strains revealed a protein, with the same molecular weight as dermonecrotic toxin, which reacted with both polyclonal and monoclonal antibodies against the toxin. In an immunodiffusion test, anti-dermonecrotic toxin serum did not discriminate between the toxin of the bovine strains and the toxin of P multocida subspecies multocida. It is concluded that these atypical pasteurella strains produce a toxin that is closely related to the dermonecrotic toxin of P multocida subspecies multocida.  相似文献

In a commercial rabbitry nasal swabs were taken from 36 animals with enzootic upper respiratory disease resembling porcine atrophic rhinitis. 35 Pasteurella multocida strains were isolated from 17 rabbits. Among 30 strains tested for dermonecrotic toxin production 3, derived from 3 animals, were positive in the guinea pig skin test. 15 Bordetella bronchiseptica strains were recovered from 14 rabbits. No toxigenic strains were found among 6 isolates tested using the same method.  相似文献

This study was performed to isolate a vaccine strain of S. aureus from clinical or subclinical mastitis and to choose the most optimal adjuvant for immune response of alpha toxin and capsular polysaccharide (CPS) of field strain. Of thirty strains of S. aureus isolated from milk of clinical or subclinical mastitis, V112 strain isolated from milk of gangrenous mastitis was used in this vaccine. Twenty one of rabbits were allocated into 5 groups based on adjuvants and immunized twice every 2 weeks for 8 weeks. This vaccine was composed of alpha toxin (10 hemolytic units) and formalinized whole cells (1 x 10(11) cells/ml. Five rabbits received PBS solution as a control group. The highest antibody titers against alpha toxin and CPS were observed in dextran sulfate- and aluminium hydroxide-adjuvant group at 8 weeks after immunization, respectively. These results of the study showed that one adjuvant could not induce strong and long-term immune response of alpha toxin and CPS antigens. Therefore, the use of combined adjuvants in subunit vaccine may be useful and feasible.  相似文献

Since the ban on growth-promoting antibiotics in animal feed in the European Union, necrotic enteritis has become a major cause of mortality in broiler chickens. Despite the importance of the disease, the pathogenesis is still not completely understood. In the current study, Clostridium perfringens strains isolated from healthy flocks and isolates from outbreaks of necrotic enteritis were evaluated for the ability to cause gut necrosis in an intestinal loop model in laying hens and in an experimental infection model in broilers. High, intermediate and low alpha toxin producing strains were chosen from each isolation source. Only the isolates from field outbreaks induced necrotic gut lesions, independent of the amount of alpha toxin produced in vitro. It was also shown that alpha toxin producing isolates from calf hemorrhagic enteritis cases were not able to induce necrotic enteritis in poultry. These results suggest the presence of host specific virulence factors in C. perfringens strains, isolated from chickens with intestinal necrotic enteritis lesions.  相似文献

A toxin produced by strains of Pasteurella multocida isolated from pigs with atrophic rhinitis caused a cytopathic effect in cell cultures derived from embryonic bovine lung. The toxin was produced during the late logarithmic phase of bacterial growth and inactivated by heating for 30 minutes at 56 degrees C. The cell culture assay was reproducible and 10(3) to 10(4) times more sensitive than a lethal assay in BALB/c mice. There was complete agreement between results in the two tests with 76 isolates of P multocida. Neutralising activity was demonstrated in both assays with sera from infected gnotobiotic piglets. It was concluded that embryonic bovine lung cell cultures provided a sensitive in vitro test for the differentiation of toxigenic from non toxigenic isolates of P multocida. The assay could be used in diagnostic laboratories and for characterisation of the toxin.  相似文献

兔的梭菌性肠炎是由产气荚膜梭菌感染引起的严重危害养兔业的一种疾病,为了更好地控制此病,本研究调查了青岛地区规模化兔场爆发此病时产气荚膜梭菌的毒素型及遗传多样性。2010年11月-2012年5月期间,采集青岛地区规模化养兔场疑似产气荚膜梭菌感染兔的肝脏进行产气荚膜梭菌的分离鉴定,采用Multiple—PCR方法对分离菌株进行毒素型分析,应用ERIC-PCR方法分析分离菌株的遗传多样性。共分离到25株产气荚膜梭菌,其中A型24株(96%),C型1株(4%)。用ERIC—PCR方法将25株分离株分于9个聚类中,其中V型为主要流行型。结果表明:青岛地区规模化兔场中产气荚膜梭菌流行的毒素型主要为A型,且具有多种基因亚型,其中V型为主要流行型。此结果为该地区兔产气荚膜梭菌病的免疫和微生态防治提供了重要的参考依据。  相似文献

Temperature-sensitive mutants were isolated and characterised from wild strains of A, D, and A/D serotypes of Pasteurella multocida by mutagenic treatment, using N-methyl-N-nitro-n-nitrosoguanidine. The mutants differed from parent strains by growth intensity at 41 degrees C and 43 degrees C as well as by reduced virulence to mice and decreased toxin formation capacity. Mice were totally protected from intraperitoneal challenge infection, using the tenfold LD100, by two subcutaneous injections of 10(7) mutant germs.  相似文献

In vitro experiments were undertaken to study the adhesion and colonization to tracheal mucosa, lung and aorta explants from freshly killed rabbits of two different strains of Pasteurella multocida. Serotype A:3 (capsulated, fimbriae +, haemagglutination -, dermonecrotic toxin -) isolated from a rabbit with rhinitis, and serotype D:1 (non-capsulated, fimbriae +, haemagglutination +, dermonecrotic toxin +) isolated from a dead rabbit with septicaemia, were used. When the explants were observed under the scanning electron microscope, the type D strain was highly adherent to trachea and aorta explants compared to the type A strain. Adhesion to lung explants was best achieved by the type A strain after 45 min incubation, but after 2 h incubation no significant difference was observed between the strains. Our data indicate that the presence of fimbriae and the absence of capsule seem to enhance the adherence of P. multocida type D strain to tracheal tissue. The capsular material of P. multocida type A strain and the toxin of the type D strain seem to influence the adherence to lung tissue in rabbit. Adhesion of strain D to aorta may indicate the expression of receptors on the endothelium to that strain and may also explain the ability of certain strains to cause septicaemia.  相似文献

A total of 186 campylobacter strains from aborted calf and sheep fetuses, from scouring dogs, rabbits and man, and from retailed poultry were isolated and examined biochemically and serologically for heat stable antigens. Immune sera were produced in rabbits against Penner reference strains from 1 to 60, and against two field isolates. Out of 186 biochemically tested strains 179 (96.2%) proved C. jejuni and only 6 (3.2%) C. coli. One strain has been identified as C. laridis. In cattle and sheep 3.2 and 21.7% respectively of all campylobacter abortions were due to C. jejuni infection. The same agent caused 12.7% of diarrhoea of dogs. The campylobacter infection rate of freshly slaughtered and dressed chicken varied between 25 and 64.3%. Out of the serologically examined 140 C. jejuni strains 118 (84.3%) could be assigned to 16 Penner serogroups and 13 (9.3%) to 2 further serogroups. Serogroups 8 (31.4%), 1 (19.3%) and 2 (12.1%) occurred most frequently. The human isolates represented the widest serotype distribution, as 32 tested strains belonged to 12 serogroups. All those serogroups which caused abortion or diarrhoea in animals or were isolated from poultry carcases were isolated also from man with diarrhoea, but some serogroups were found only in man.  相似文献

为研究确定新疆北疆地区规模化奶牛场犊牛腹泻病原性大肠杆菌的优势血清型、致病性及毒力因子特征。采用细菌学、免疫学及分子生物学的方法对从新疆呼图壁、石河子、奎屯3个主要奶牛生产基地14个规模化奶牛场10日龄内腹泻犊牛直肠棉拭子样品进行了大肠杆菌的分离与鉴定、O血清群、黏附素及肠毒素的测定。结果从302份样品中分离并经生化鉴定获得180株呈β溶血的大肠埃希菌,其中94株对小鼠有致病性;对其中53株代表菌株的O血清型、黏附素及肠毒素测定,结果为8株携带K99菌毛及STa毒素基因,6株携带F41茵毛基因,6株产生LT毒素;28株分离株分布于16个O血清型,其中O101,O6,O114,O78为优势血清型,占被测菌株的50%。结果表明,新疆北疆主要奶牛养殖区致犊牛腹泻大肠杆菌是以携带K99、F41和产ST的溶血性大肠杆菌为主,其研究结果为犊牛大肠杆菌性腹泻的免疫防治提供病原学依据。  相似文献

1125 and 1146 E. coli strains isolated from suckling and weaned piglets with diarrhea, respectively, and 724 strains from healthy piglets were tested for the presence of fibriae and production of enterotoxins. The fimbriae were determined by hemagglutination and slide agglutination tests, enterotoxins—by the use of ileal loop test in piglets (LT and STb enterotoxins) and suckling mouse assay (STa enterotoxin). It was found that 72.8 and 53.0% strains, isolated from diseased suckling and weaned piglets, respectively, possessed specific fimbrial hemagglutinins, in most cases with K88 antigen. Additionally, 987P fimbriae were detected in 14.0 and 0.7% strains isolated from piglets with diarrhea. Only 5 strains (0.7%) recovered from healthy piglets had specific fimbriae, usually with undetermined antigenic structure. F1 fimbriae (called common or unspecific) were found in strains isolated both from diseased (15.2 and 16.3% strains, respectively) and healthy piglets (27.1% strains). It was noted that the strains isolated from suckling and weaned piglets with diarrhea in most cases were enterotoxigenic (90.5 and 69.1% strains, respectively) and most frequently produced heat-labile toxin LT alone or with STb. 18.5% of enterotoxigenic strains isolated from healthy piglets produced STa toxin.  相似文献

A widespread epizootic due to a granular variety of Trichophyton mentagrophytes occurred in a new stock of laboratory guinea pigs imported to Perth from one of the Eastern States of Australia. The infection spread quickly among these animals, and infection occurred in rabbits and mice at the breeding station and in 4 people. The human infections were contracted either directly through handling animals or indirectly by means of fomites. The strain of T. mentagrophytes causing the epizootic was distinctive from strains previously isolated in Western Australia.  相似文献

作者观察了从湖南6种动物分离出的隐孢子虫卵囊的光镜下形态,并以鸡源、牛源和猪源隐孢子虫卵囊人工感染实验动物作交叉感染性研究。结果鉴定出隐孢子虫3个种,即贝氏隐孢子虫(C.baileyi),寄生于鸡、鸭;微小隐孢子虫(C.Parvum),寄生于牛、山羊、猪、家兔和小鼠;鼠隐孢子虫(C.muris),寄生于牛。交叉感染结果表明,来自鸡的隐孢子虫可以感染雏鸡和雏鸭,而不能感染小鼠和家兔;来自牛的隐孢子虫可以感染小鼠和家兔而不能感染雏鸡,来自猪的隐孢子虫可以感染小鼠而对雏鸡无感染性。作者认为,哺乳类和鸟类的隐孢子虫可能在宿主纲的水平上具有宿主持异性。  相似文献

Chromosomal DNA from 13 different selected Pasteurella multocida spp. multocida strains of serotypes A and D were isolated and compared. All 10 toxigenic strains were recognized by a DNA probe which included the toxA gene coding for the Pasteurella multocida toxin (PMT). None of the three nontoxigenic strains reacted with the DNA probe. Toxin from the 10 toxigenic strains were isolated and compared. All were found to possess the biological characteristics previously described for the PMT isolated from P. multocida ssp. multocida NCTC 12178, including molecular mass of approx. 143 kDa and reactivity with a series of monoclonal antibodies. Toxin prepared from different toxigenic strains could not be differentiated immunologically by tandem crossed immunoelectrophoresis, Toxin, which was affinity purified from four of the strains and subsequently inactivated by formaldehyde, was cross-protective when used for vaccination of mice before challenge with PMT. It is concluded that the toxin from toxigenic strains of P. multocida ssp. multocida must be very similar, if not identical.  相似文献

Eleven Clostridium perfringens type C strains isolated from fatal cases of hemorrhagic enterotoxemia of Canadian calves, a piglet, and a foal were studied for the production of soluble antigens. All the isolates from calves and a foal failed to produce delta toxin, but were capable of producing large amounts of lethal beta toxin. A strain isolated from a piglet produced delta, but very little beta toxin. Other differences were relatively minor. The results indicated that young domestic animals may be susceptible to all subtypes of C. perfringens type C. A simple method of using blood agar plates coated with type A antiserum for demonstration of hemolytic patterns was found advantageous in differentiation of C. perfringens strains.  相似文献

Young male white Swiss mice were fed control diet or diet supplemented with 20 or 10 parts per million (ppm) of T-2 toxin for two or three weeks. These mice then were inoculated with herpes simplex virus type 1 (HSV-1) (9.6 x 10(6) plaque forming units) intraperitoneally. To compare the effects of T-2 toxin against a known immunosuppressive drug, cyclophosphamide was injected intraperitoneally at 150 mg/kg, 24 hours after treatment with HSV-1, into mice fed the control diet. Mice were necropsied and tissues were collected for microscopic and virologic examination. White Swiss mice which consumed a daily diet containing 20 ppm of T-2 toxin for two or three weeks were highly susceptible to HSV-1 infection and 27 of 36 (75%) died as a result of extensive hepatic and adrenal necrosis. Although HSV-1 was isolated from livers and brains of mice fed 20 ppm of T-2 toxin for two or three weeks, there was little or no inflammatory response found in the adrenals, livers, spinal cords, brains, or ganglia. The necrotizing encephalomyelitis observed in control mice was absent. High levels of dietary T-2 toxin appeared to be more immunosuppressive than cyclophosphamide because only one mouse died after treatment with HSV-1 and cyclophosphamide. Mice treated with cyclophosphamide had changes in brain, spinal cord, spleens, thymus, and bone marrow which were similar to those fed 20 ppm of T-2 toxin and infected with HSV-1, however, liver lesions were much less severe. HSV-1-infected mice on a diet with 10 ppm T-2 toxin had lesions of intermediate severity when compared with HSV-1-infected mice fed a diet with 20 ppm T-2 toxin and HSV-1-infected mice on control diets. Necrosis was less extensive in the livers and adrenals. The infrequent isolation of virus from liver and brain was consistent with the lack of intranuclear inclusion bodies and a more marked inflammatory response. Ten ppm of dietary T-2 toxin only depressed bone marrow and splenic red pulp to a mild or moderate degree. This may have enhanced the necrotizing encephalomyelitis observed in mice killed on days 6 and 8 after HSV-1 infection. Liver lesions were mild and those of the adrenals were moderate in mice fed control diet. The rare isolation of HSV-1 from the liver and brain and the findings of a moderate to severe necrotizing encephalomyelitis in these mice was consistent with an essentially functional immune system.  相似文献

Forty-seven Staphylococcus hyicus strains, one each from 47 of 81 cattle examined, were isolated from cutaneous lesions on different parts of the body. Nineteen of the strains were isolated in pure culture and the rest were isolated in association with either other staphylococci or microfilariae or mange mites. Three out of 10 representative strains produced cutaneous lesions when inoculated intradermally into rabbits.  相似文献

A型魏氏梭状芽孢杆菌XP5株分离自典型猝死症病猪,将其制成氢氧化铝灭活疫苗。疫苗0.6ml、2ml和4ml的剂量可以分别保护小鼠、兔和猪抵抗标准A型魏氏梭状芽孢杆菌毒素1MLD的攻击。在两个疫病流行区应用了疫苗,1215头接种疫苗猪中死于猝死症的有12头,死亡率0.99%,8152头未接种疫苗的对照猪中,死于猝死症的有284头,死亡率3.48%,二者显著差异(X2=12.23,P<0.01)。  相似文献

Epizootic rabbit enteropathy (ERE), a highly lethal (30-80% mortality) disease of broiler rabbits aged 6-14 weeks, first appeared in 1997 in French intensive enclosed rabbitries and is of unknown aetiology. Bacteriological, virological and parasitical examination of the intestinal contents of rabbits that had died either in spontaneous field cases or after experimental reproduction of ERE, were undertaken in an attempt to identify infectious agents that may play a role in the disease. Two bacterial strains, Clostridium perfringens and non-enteropathogenic Escherichia coli were repeatedly isolated at high faecal counts from naturally infected animals. In field cases, a correlation between typical gross lesions of epizootic enteropathy and the presence of the alpha toxin of Cl. perfringens was observed (P<0.0001; Chi-squared test). Although attempts to reproduce the disease by inoculation with different pools of cultivable bacterial strains failed, the disease was successfully reproduced by inoculation with one French and two Belgian samples of caecal contents.  相似文献
