Genetic relationships among 13 grasspea (Lathyrus sativus L.) landraces mainly collected in Southern Italy were assessed using agronomic traits, biochemical and molecular markers. Field trials were carried out in two locations and revealed a high influence of field locations on yield, but a low genotype × environment interaction. Despite this, the agronomic data obtained provided useful information for the choice of the best grasspea landraces for southern Italian marginal areas. Seed storage proteins utilised as biochemical markers were not able to detect polymorphisms, on the contrary both classes of molecular markers used i.e. AFLP and SSR, provided useful information on genetic variation and relationships among landraces. Even though the number of polymorphic fragments detected by AFLP technique was low, it was sufficient to discriminate all the accessions. The use of SSR to detect polymorphic sites in grasspea showed that most landraces were clearly grouped in two sub-clusters. One comprised two landraces from most northern localities, while all the other landraces were clustered together at a very narrow genetic distance.  相似文献

We investigated the genetic variation and relationships among 35 melon landraces collected from the Xinjiang Uygur Autonomous Region in northwestern China by using 19 polymorphic simple sequence repeat markers (SSRs). A total of 55 polymorphic alleles were amplified. The number of alleles per SSR locus ranged from 2 to 5 with an average of 2.89 alleles per locus. The average gene diversity (GD) was 0.42 with a range of 0.06–0.71, and the average observed heterozygosity was 0.22 with a range of 0.06–0.97, indicating that the genetic diversity among the Xinjiang melon landraces was abundant. Genetic variation was also detected between the landrace populations in different regions in Xinjiang. The most abundant genetic diversity was observed among the landraces in Eastern Xinjiang, with the highest GD of 0.45 and PIC of 0.39. Eleven alleles (20 %) were found exclusively in the landraces from Eastern Xinjiang, and two alleles (3.6 %) were unique to the landraces from Southern and Northern Xinjiang. The genetic similarity matrix was defined on the basis of Jaccard coefficient to determine the genetic relationships among Xinjiang landraces. Cluster analysis was performed using the unweighted pair group method with arithmetic means, showing that the ‘wild Hami’ (XJ-34) landrace was distinct from the 34 other landraces that were divided into three clusters. Therefore, the genetic background of XJ-34 differed from that of the other landraces. The landraces were not precisely separated on the basis of their geographic origins, although most of these landraces were likely grouped near one another, as visualized through principal coordinate analysis. Thus, western China is one of the primary or secondary centers of melon diversity because of the relatively higher genetic variation detected among Xinjiang landraces. Except the ‘wild Hami’ landrace, Xinjiang melon landraces could be classified into two botanical varieties, namely, var. inodorus and var. cantalupensis. The “Sedano Nero di Trevi” (Black Celery from Trevi) is a landrace grown in Umbria (Italy) at risk of extinction. A morpho-physiological and molecular characterisation of the landrace and a detailed investigation of its cultural and cultivation context were carried out with the aim of facilitating a request for a quality mark, implement in situ (on farm) conservation schemes that are being developed in Italy and register the landrace in the European common catalogue of ‘conservation varieties’. These actions can help the landrace survive. Six farmer populations of the landrace and four élite varieties were characterised for thirteen morpho-physiological traits and nine AFLP primer combinations. The “Sedano Nero” farmer populations were better performers than élite varieties for important agronomic traits, such as yield, under the local agronomic conditions. Both morpho-physiological traits and molecular markers clearly distinguished the landrace from the élite cultivars. In addition, a structure was detected in the landrace with farmer populations being characterised by specific alleles. These results are discussed in connection with the local seed system and possible in situ conservation strategies for landraces.  相似文献

Chinese cherry (Prunus pseudocerasus Lindl.) is an indigenous fruit crop with high nutritional and ornamental value. It is widely cultivated in China, and many landraces have accumulated in large distribution regions. Genetic diversity analysis of Chinese cherry landraces is useful for breeding programs, as it helps to select genetic material to be used for further crossing. In this study, 110 Chinese cherry accessions from nine populations were assessed using fifteen morphological traits and seventeen ISSR markers. High average coefficient of variation (CV?=?17.43%) was detected based on morphological analysis. For molecular characterization, the global gene diversity (h?=?0.2816) and Shannon information index (I?=?0.4253) suggested a moderate high level of genetic diversity. Model-based STRUCTURE and principal coordinate analysis revealed three and two major gene pools based on morphologic and molecular data, respectively. Distinct distribution patterns of genetic variation between samples from southwest China and north China indicated two potential original domestication locations. We inferred that botanical features such as predominantly selfing characteristics, long lifespan, and insects-pollinated trait lead to the survival of abundant genetic variation and special genetic structure. This study provided morphological and molecular evidences for understanding genetic diversity and new insights into genetic consequence of populations for Chinese cherry landraces. Important implications for breeding programs and resources conservation were also provided.  相似文献

Characterization of 15 chickpea (Cicer arietinum L.) accessions (6 cultivars, 3 selected lines and 6 landraces) was performed using morphological, agronomic, and technological traits, together with biochemical (seed storage proteins) and molecular markers. Field trials, performed in two different geographical areas of Southern Italy, one in a plain area near Tyrrhenian sea and one in an hilly internal area, revealed a high influence of field location on yield. A wide variation was also registered for technological traits and molecular markers. Microsatellite (SSR) markers grouped chickpea accessions in different clusters, providing useful information on genetic variation and relationships among them, with polymorphic fragments useful to discriminate among all accessions. On the contrary, seed storage protein pattern showed scarce variation resulting very similar among all the accessions considered.  相似文献

‘Microcenters of diversity’ are restricted geographical areas within which a significant diversity of genetic resources is accumulated. Maize genetic variability, one of the largest among cultivated species, particularly in far western Santa Catarina, southern Brazil, has shown a rich store of landraces, conserved on farm by small-scale farmers. Thus, this study aimed to characterize the diversity of Zea mays L. landraces in two municipalities of this micro-region in Santa Catarina, as well as its geographic distribution, developing the census of diversity as a methodological proposal. Diagnosis was made according to the farmers’ knowledge. The field survey was conducted in 70 rural communities involving 2049 farms. The methodology allowed identifying 136 populations of wild relatives, of which some belong to the species Zea luxurians (Durieu and Ascherson) Bird, and 1513 populations of maize landraces, comprising 1078 of popcorn, 337 of common maize, 61 of sweet maize and 37 of flour maize. The identification of 59 morphological groups, the high use, adaptive and agronomic values, and the Shannon index (H′), estimated according to the grain shape (0.79), endosperm type (0.73), size (0.87), color (1.40), and morphological group (3.16), showed at set a considerable diversity distribution conserved on farm in this micro-region. Landrace richness and the presence of wild relative species, associated with the local human activity and sociocultural aspects, allowed to characterize the far western Santa Catarina as a ‘microcenter of diversity’ of Zea mays L.  相似文献

The eleven most known landraces from central and southern Italy were analysed using ISSR markers on 15 randomly chosen individuals for each landrace, with the aim of assessing genetic variation within and among landraces and possibly ascertaining their origin and genetic relationships. A total of 164 loci were observed, 128 of which (78.05%) were polymorphic. Gene diversity over all landraces was I = 0.3759. The highest within-landrace diversity was observed in samples from the Apennine ridge and for one Sicilian landrace; on the other hand, samples from the small Sicily islands were less variable. Principal Component Analysis and AMOVA allowed the discrimination of groups of landraces with higher similarity. Analyses indicate that the small Sicily islands landraces are very closely related to one another and seem to be derived from the peninsular material; moreover, they help disclose relationships among geographically close landraces. The knowledge so acquired can, therefore, contribute to elaborate strategies for increasing the economical value of élite landraces and to protect producers from frauds.  相似文献

Ninety-five common bean (Phaseolus vulgaris L.) landraces from Spain were evaluated in three different environments in northern Spain for their agronomic performance and seed quality. Significant differences among landraces were found for 14 quantitative traits related to phenology, yield and its components, and seed quality traits. Environmental effects were significant for all traits evaluated except for seeds pod?1, seed width/thickness, seed weight, and seed water absorption. Landrace by environment interactions were significant for all traits except for seeds pod?1 and seed water absorption. Selection of new breeding lines for agronomic performance and seed quality within landraces should be reliable because many of them are mixture of lines. Some heirloom varieties belonging to the types faba, caparrón,riñón,ganxet and tolosana had the best performances regarding to seed quality and yield. Principal component analysis revealed differences among environments affecting the performance of the bean landraces. Variation in the landraces seems to be organized in a different way in each one of the test environments, therefore, low plasticity and specific adaptation of Spanish bean landraces to different environments is derived from this study. Some landraces, especially those large and white seeded should be an useful resource for sustainable farming systems in different biogeographical areas and a worthy germplasm for the genetic improvement of agronomic value and seed quality.  相似文献

Eight species of the genus Asparagus, members of the group of European species closely related to A. officinalis, were analysed using internal transcribed spacer (ITS), and expressed sequence tag-derived simple sequence repeat (EST-SSR) markers, as well as cytological observations of their hybrids, to study their phylogenetic relationships and the possibility of broadening the narrow genetic base of cultivated varieties. Phylogenetic analysis using ITS data revealed two major clades: clade I consisting of A. acutifolius and clade II (referred to in this study as the ‘officinalis group’) comprised of sequences derived from species closely related to A. officinalis; but the different species within the ‘officinalis group’ could not be clearly separated. In contrast, cluster analysis of EST-SSR marker data showed six major clades and clearly separated each population, grouping most of the genotypes from each population together. That is, EST-SSR markers were found to be more informative than ITS markers about the relationships within the ‘officinalis group’, indicating that EST-SSR markers are more useful than ITS sequences for establishing phylogenetic relationships at the intrageneric level. All the crosses carried out at the same ploidy level were successful. The high crossability, together with the regular meiotic behaviour and high pollen and seed fertility observed in the interspecific hybrids analysed, suggest relatively close relationships between the species studied. We conclude that the group of species classified in the ‘officinalis group’ are in the primary gene pool, indicating that these species could be used to increase the genetic diversity of the cultivated species. In addition, the tetraploid landrace “Morado de Huétor” could be employed as a bridge to generate new cultivated germplasm.  相似文献

The recent emergence of wheat stem rust race Ug99 (TTKSK) and related strains threaten Ethiopian as well as world wheat production because they overcome widely used resistance genes that had been effective for many years. The major cause which aggravates the ineffectiveness of Ethiopian wheat varieties against stem rust is the narrow genetic base on which the breeding for resistance has been founded, however, little is known about the resistance genotypes of Ethiopian durum wheat varieties and tetraploid wheat landraces. The objective of the study was to identify stem rust resistance genes that are present in the Ethiopian tetraploid wheat landraces and improved durum wheat varieties using molecular markers and assess which genes are effective for current Ethiopian stem rust races of Puccinia graminis f. sp. tritici including Ug99. The investigated 58 tetraploid wheat accessions consisted of 32 (Triticum durum s.l. incl. Triticum aethiopicum Jakubz., Triticum polonicum) landraces and 22 registered T. durum varieties released in Ethiopia between 1966 and 2009 and four T. durum varieties from ICARDA. A total of 17 molecular markers (SSR, EST and InDel) linked or diagnostic for stem rust resistance genes Sr2, Sr13, Sr22 and Sr35 were used for genotyping. Haplotype analysis indicated that only few of the Ethiopian durum wheat varieties carried Sr13. The resistant variety ‘Sebatel’ showed a haplotype for Sr2 and Sr22 and variety ‘Boohai’ for Sr22, however further evaluation is needed for the diagnostic value of these haplotypes. This study is the first report on the presence of stem rust resistance (Sr) genes in Ethiopian durum wheat varieties and tetraploid wheat landraces based on linked or associated molecular markers. Thus it might help in the identification of varieties carrying resistant alleles that provide valuable genetic material for the development of new improved varieties in further breeding programmes.  相似文献

This study was carried out to estimate the level of diversity existing within some common bean landraces still cultivated in Nebrodi mountains, North-western area of Sicily. The multidisciplinary approach adopted to reach this goal involved the characterisation of collected material through morphological, biochemical and molecular marker analyses. The nutritional quality of seeds was also investigated in view of the proposition of the best landraces as niche products. Results showed that those bean landraces retain a considerable level of heterogeneity. The use of both biochemical and molecular markers showed that all landraces clustered into two main groups, corresponding to the Andean and Mesoamerican gene pools. Our results suggest that the best strategy for preserving the diversity of common bean from a restricted area such as Nebrodi mountains, necessitates of a deep knowledge of germplasm to avoid the loss of precious genetic resources or, on the contrary, the safeguard of populations genetically redundant.  相似文献

The increasing consumers’ request for new speciality wheat derivatives and the possible widening of marketing opportunities is giving to both landraces and old varieties, new chance of on farm survival providing a low cost way to safeguard these important genetic resources. A farmer’s consortium acting in north-western Italy, is attempting the introduction of some old bread wheat landraces in marginal environments of Piedmont and Liguria regions. The reason of this attempt is the local market demand of old bread varieties suitable for the preparation of traditional cakes and biscuits. Three Italian bread wheat landraces, a mixture of durum and bread wheat landraces and three modern varieties were evaluated. The entries were cropped in Val Borbera and Val d’Aveto (Piedmont and Liguria region, respectively) in the same growing season (2009–2010). The high variation of gliadin profile detected within the landraces indicates that all have retained the genetic heterogeneity typical of the old wheat landraces. In consequence of the health-promoting effects of whole grain consumption, eleven nutritional and technological traits of whole flours were investigated. The analysis of collected data revealed appreciable differences among the flours obtained from the landraces and the modern varieties. These differences can be mainly attributed to the intensive breeding carried out on bread wheat in the last century. The results of this study suggest that the old bread wheat landraces could have good chance of survive on farm when their characteristics fulfil the requirements of local communities.  相似文献

Temporal variation of diversity in Italian durum wheat germplasm   总被引:1,自引:0,他引:1  
The aim of this work is to analyse the temporal change of genetic diversity in Italian durum wheat germplasm. The germplasm deployed in this study (158 accessions), belonging to 5 different historical classes, was characterised for its microsatellite and gliadin markers. The level of genetic diversity (He), based on gliadin and SSR markers results – on average – greater in indigenous landraces present in Italy before 1915, with the exception of pure line material which had been selected from landraces (showing highest level of heterozigosity for gliadin markers). Genotypes obtained from crosses or mutagenesis (referring to the 1950–1960 period) along with those resulting from crosses between CIMMYT lines and old materials (1970s and beyond) were also genetically more diverse. Forty-nine percent of indigenous landraces were genetically heterogeneous. Nine out of 53 landrace accessions were able to capture 4 different SSR private alleles. It is speculated that the reduction of allele richness is an indicator of the genetic erosion of the pre-breeding germplasm and it is pointed out that the implementation of appropriate methods of genetic conservation of this germplasm is a priority for breeding and food safety.  相似文献

Smallanthus sonchifolius is a periennal herb originally cultivated in South America and now grown in several other countries. Recently, greater attention has been focused on this plant due to its agronomical, nutritional and pharmacological characteristics. In this paper the application of RAPDs and AFLPs for the analysis of genetic diversity in a group of 5 Smallanthus sonchifolius landraces is presented. Both methods proceed through the direct analysis of DNA, and their results were compared with the total phenolic content of each landrace and its morphological traits. Using 61 RAPD primers, 85 informative bands were identified, corresponding to 28.7% of polymorphism. In comparison, only six selected AFLP primer pairs produced 84 informative bands, with a similar percentage of polymorphism (23.4%). RAPD and AFLP markers were analyzed separately. Total phenolic content varied twofold among the five landraces analysed, ranging from 3,494 to 6,849 mg/g. Each type of molecular marker resolved two main groups that included the same genotypes, but with different within-group relationships among genotypes. The two groups are consistent with some phenotypic characters but they do not reflect faithfully their geographical origin. Most notably, the two groups comprise landraces with higher and lower total phenolic content, respectively. Dendrograms based on the two molecular data sets graphically depicted the ability of both methods to differentiate all the cultivars studied. Data obtained suggest that the two molecular markers applied are useful to investigate intra-specific genetic variability in Smallanthus sonchifolius, and predict well the total phenolic content of each landrace.  相似文献

Randomly amplified polymorphic DNA (RAPD), inter-simple sequence repeat (ISSR) and a semi-random PCR system were used to analyze the genetic diversity of 16 Italian common bean landraces and their relationship to four commercial cultivars. Of the primers tested, 8 ISSR, 6 RAPD and 7 semi-random primers produced polymorphic and reproducible DNA fragments. A higher proportion of polymorphic bands were observed using ISSR (85%) and semi-random (90%) primers than RAPD (69%) method. The combination of any two semi-random markers allowed the identification of all 20 bean genotypes. In contrast ISSR (except for primer (CAC)3GC) and RAPD markers appeared to be less informative as more than two markers were necessary to achieve the same diagnostic level. Moreover, 7 ISSR, 2 RAPD and 8 semi-random exclusive bands were identified as putative population-specific markers. Semi-random and ISSR derived dendrograms showed similar tendencies in terms of genetic relatedness, whereas clustering of genotypes within groups was not similar when compared with the RAPD technique. Despite the different ability to resolve genetic variation among the investigated landraces, two major clusters with less than 60% (ISSR) and 40% (RAPD and semi-random) genetic similarity were formed with all three marker systems. The two groups were correlated with the phaseolin patterns and seed size of the landraces. The analysis showed that the cultivar ȁ8Lingua di Fuocoȁ9 and most of the landraces (13 out of 16) collected in Italy belong to the Andean gene pool, whereas only the three populations from Pratomagno belong to the Middle American gene pool.  相似文献

Lima bean is a species cultivated broadly in the Americas and has been cultivated in the Caribbean for at least 500 years. In order to determine the genetic structure and diversity of Lima bean from the Caribbean, 50 landraces from Haiti, the Dominican Republic and Puerto Rico were collected and analyzed using 24 SSR markers. All landraces in this Caribbean collection were found to be of Middle-American descent. The genetic diversity (HE) was highest in the landraces from Puerto Rico and lowest in Haitian landraces. The observed proportion of heterozygotes (HO) was higher in the Haitian landraces and lowest in Puerto Rican landraces. Unweighted pair-group method with arithmetic averaging analysis showed that the landraces clustered into 3 clusters with all Haitian landraces grouping in one cluster. We also assessed the agro-morphological characteristics of the collection as well as the content of cyanogenic glucoside, linamarin, in leaves and dry seeds. Lima bean, which is a model crop for indirect plant defenses against herbivory, also possess linamarin as a source of direct plant defense. Upon tissue damage, linamarin is converted to toxic hydrogen cyanide. In our collection 44.6 % of the landraces had average seed HCN content ≤200 ppm which is the permitted level for Lima bean seed in the US. Our results also identified the landraces in this collection which have high linamarin levels in the leaves while having low levels in the seeds. Such landraces have the desirable combination of traits and will be the focus of our future plans for agronomic trait improvement though breeding.  相似文献

The genetic structure and diversity of eight pairs of rice landraces from in situ (collected in 2007) and ex situ (collected in 1980) conservation programs were studied using 20 pairs of microsatellite markers with high polymorphism. Each pair of rice landraces shares a name and origin and has similar seed and plant traits. The number of alleles detected in the populations from in situ conservation ranged from 43 to 88 with the mean number of alleles per locus ranging from 2.15 to 4.40, while the number of alleles detected in the populations from ex situ conservation ranged from 33 to 65, and the mean of alleles per locus ranged from 1.65 to 3.25. Compared to the ex situ populations, the number of alleles, the number of specific alleles and the genetic diversity index showed a significant increase in the in situ populations. Further, the numbers of specific alleles from in situ populations were 2.1–5.0 times greater than in ex situ populations except for rice landrace ‘Qitougu’. An AMOVA showed that the within-landrace genetic structure differed significantly between in situ and ex situ conservation treatments with differences exceeding 20%. The analysis of genetic similarity reached similar conclusions to those of the AMOVA. Compared with ex situ conservation programs, the rice landraces under in situ conservation programs had more alleles and higher genetic diversity in Yunnan of China.  相似文献

In the present study, three pea Greek landraces Schinousa (AUASCHIN001), Andros (AUAANDRO001), and Amorgos (AUAAMORG001), and the commercial variety Onward were grown following either conventional or organic farming practices. The main objective of the study was to assess the suitability of these three landraces for organic cultivation as compared with a standard commercial variety. The total fresh pod yield produced by AUASCHIN001 and AUAAMORG001 was significantly lower than that obtained from ‘Onward’ and AUAANDRO001. Furthermore, the three landraces exhibited a higher competition to weeds than the commercial variety. Under organic farming conditions, AUASCHIN001 and AUAAMORG001 produced appreciably less shoot biomass than ‘Onward’ and this resulted in commensurate decreases in the total amount of BNF (biological nitrogen fixation) in their plant tissues, although the percentage of nitrogen (N) fixed from the atmosphere was significantly lower in ‘Onward’. AUAANDRO001 rendered the highest amounts of BNF in the organic farming systems, which was similar to that produced by ‘Onward’ in the conventional system. These results indicate that the tested landraces and especially AUAANDRO001 are more adaptable to low soil N levels and high weed competition, and thus they are more suitable for organic cultivation than ‘Onward’ which performs best in conventional cropping systems.  相似文献
