Lettuce, spinach and chicory are generally considered the main leafy vegetables, while a fourth group denoted by ‘minor leafy vegetables’ includes, amongst others, rocket salad, lamb’s lettuce, asparagus, artichoke and rhubarb. Except in the case of lettuce, central crop databases of leafy vegetables were lacking until recently. Here we report on the update of the international Lactuca database and the development of three new central crop databases for each of the other leafy vegetable crop groups. Requests for passport data of accessions available to the user community were addressed to all known European collection holders and to the main collection holders located outside Europe. Altogether, passport data of 17,530 accessions from a total of 129 collections were collected. The four separate databases were made available on line via a common entry page accessible at http://documents.plant.wur.nl/cgn/pgr/LVintro/. Based on a literature study, an analysis of the gene pool structure of the crops was performed and an inventory was made of the distribution areas of the species involved. The results of these surveys were related to the contents of the newly established databases in order to identify the main collection gaps. Priorities are presented for future germplasm acquisition aimed at improving the coverage of the crop gene pools in ex situ collections.  相似文献

Series of 10,348 accessions belonging to 21 species (hexaploid, tetraploid, diploid) of the genus Triticum and 489 accessions belonging to 20 species of the genus Aegilops were scored for disease resistance during a period of 60 years. Tests were performed at the seedling stage for powdery mildew (Erysiphe graminis DC. f. sp. tritici March.), leaf rust (Puccinia recondita Rob. ex Desm. f. sp. tritici Erikss.), stripe rust (Puccinia striiformis West. f. sp. tritici Erikss.) and eyespot (Pseudocercosporella herpotrichoides (Fron.) Deight.) but also at the adult plant stage considering powdery mildew, leaf rust, stripe rust, eyespot and glume blotch (Septoria nodorum Berk.). About 150,000 disease scores recorded on index cards using different scoring scales were transferred to the computer, converted into a 1–9 scale and used to summarise the results. Within the genus Triticum 20% of the material analysed was classified as heterogeneous. For the accessions without detectable segregation a large variability for resistance/susceptibility was detected. At the adult plant stage resistant accessions without visible infections were identified for all diseases. The percentages of resistant accessions at that growth stage were always higher than the ones found in the material tested at the seedling stage. The probability for finding resistant material was shown to be highest in the diploid species ( > 50%) but decreased with increasing ploidy level to about 10% in the hexaploids. For Aegilops it was shown that most of the accessions were homogeneous and highly resistant against powdery mildew (seedling and adult plant stage), leaf rust (adult plant stage) and eyespot (seedling and adult plant stage/natural infection). The genetic resources of Allium in India are potential source of genes for widening the crop genetic base. Despite their high economic value, limited number of germplasm accessions of wild species have been collected and conserved mainly due to difficult access to areas of occurrence. The present work mainly highlights information on species diversity distribution and utilization of wild Allium species to assess the value of Indian germplasm and prioritization for future collection and conservation programmes.  相似文献

Results are presented of a 1994 postal survey of holdings of local onion (Allium cepa L.) germplasm by institutions in West Africa. Data obtained from respondents included the number of accessions of onion or shallot held and the conditions in which onion seed was stored. Twenty-four replies were received and eighteen sites where collections were held were identified. The numbers of local A. cepa accessions in local collections varied in number from 38 down to nil, but many local collections were of less than five accessions. The largest number of national onion accessions was held at CERRA, Maradi, in Niger (16), and the largest regional collection was at the Station de Farako-bâ, Burkina Faso (38). Two sites held substantial shallot collections: CRA Bareng in Guinée and IDESSA, Bouaké, Côte d'Ivoire. Conditions under which onion seed was stored varied from low temperature, controlled storage in moisture-proof packs with desiccants, to storage in plastic envelopes under ambient laboratory conditions.  相似文献

The shallot is considered an important genetic resource for Allium breeding programs because, compared to the bulb onion, the shallot contains higher levels of several functional chemical compounds. However, there may be differences in content, composition and beneficial activity among shallot landraces. This study was carried out to characterize the differences in phenolic, quercetin, sugar, S-alk(en)yl-l-cystein sulfoxide (ACSO), and saponin contents and antioxidant capacities of a shallot germplasm including 31 strains derived from different regions of Vietnam and six other countries. A wide variation was observed in the quantitative analyses of the chemical contents. Shallots with high contents of polyphenols, saponins, and quercetins were found in the south of Vietnam and other low-latitude countries. Meanwhile, those possessing fairly high ACSO and sugar contents were observed in the north of Vietnam. Qualitative analysis of saponins via thin layer chromatography did not show clear variation among shallot strains, but polymorphism was observed between the shallot and other Allium species, such as A. roylei. The principal component analysis could clearly discriminate shallot strains by their geographical origins. All shallot strains showed potent antioxidant activities in a DPPH assay. The highest antioxidant capacity was in the strains possessing relatively high contents of polyphenol, quercetin, and saponin. Significant correlations were found between antioxidant capacity (IC 50 ?1 ) and four groups of chemical compounds (polyphenols, quercetins, saponins, and ACSOs) (r = 0.40–0.59). A strong correlation was observed between IC 50 ?1 and quercetin contents (r = 0.59, p < 0.01). The six Fusarium-inoculated shallot strains seemed to be adequately resistant against disease, and the levels of resistance may be related to the saponin content in the bulb tissues.  相似文献

An exploration and collection mission for wild populations of Capsicum was carried out in the fall of 2006 and 2007, in 13 Mexican states and in the U.S. states of Arizona and Texas. The aim of this collection was to expand the number of accessions of wild chile pepper (Capsicum annuum var. glabriusculum and Capsicum frutescens) that are publicly available for research in plant improvement and for subsequent use in an inquiry into the domestication of C. annuum. While Mexico and the United States National Plant Germplasm System both have germplasm repositories INIFAP—Instituto Nacional de Investigaciones Forestales—Agrícolas y Pecuarias and USDA GRIN—United States Department of Agriculture’s Genetic Resources Information Network) with accessions of C. annuum var. glabriusculum, the very limited number available, their age, and/or validity of the information attached to many accessions do not allow for extensive research. Four hundred and sixty-six plants were sampled over two field seasons, of which copies of the collection reside in both UAA and at UC Davis. Given the current environment with the intellectual property of varieties of crop plants and, particularly, the extreme restrictions affecting explorations and the official procuring and sharing of germplasm across national borders, this U.S.—Mexico collaboration is one of the few examples of joint U.S.—Mexico germplasm collection efforts.  相似文献

Cacao (Theobroma cacao L.) is a significant agricultural commodity in the Dominican Republic, which ranks 11th in the world for cacao exports. To estimate genetic diversity, determine genetic identity, and identify any labeling errors, 14 SSR markers were employed to fingerprint 955 trees among cacao germplasm accessions and local farmer selections (LFS). Comparisons of homonymous plants across plots revealed a significant misidentification rate estimated to be 40.9 % for germplasm accessions and 17.4 % for LFS. The 14 SSRs amplified a total of 117 alleles with a mean allelic richness of 8.36 alleles per locus and average polymorphism information content (PIC) value of 0.67 for the germplasm collection. Similar levels of variation were detected among the LFS where a total of 113 alleles were amplified with a mean of 8.07 alleles per locus and PIC of 0.57. The observed heterozygosity (Hobs) was 0.67 for the germplasm collection and 0.60 for LFS. Based on population structure analysis 43.9 % of the germplasm accessions and 72.1 % of the LFS are predominantly of the Amelonado ancestry. Among these Amelonado, 51.7 % for the germplasm collection and 50.6 % for LFS corresponded to Trinitario hybrid lineage. Criollo ancestry was found in 7.6 and 9.5 % of the germplasm accessions and LFS, respectively. The Contamana, Nacional, and Iquitos backgrounds were also observed in both populations, but the Curaray background was only detected in the germplasm accessions. No Purús or Guiana ancestry was found in either of the populations. Overall, significant genetic diversity, which could be exploited in the Dominican Republic breeding and selection programs, was identified among the germplasm accessions and LFS.  相似文献

We present here the first comprehensive genetic characterization of wild melon accessions from northern India. The genetic diversity among 43 wild melon accessions collected from the six agro-ecological regions of the Punjab State of India was assessed by measuring variation at 16 Simple Sequence Repeat (SSR) loci, morphological traits of plant habit and fruit morphological traits, two yield-associated traits, root nematode resistance and biochemical composition (ascorbic acid, carotenoids, titrable acidity). Variation among accessions was observed in plant habit and fruit traits and wild melon germplasm with high acidity and elevated carotenoid content and possessing resistance to Meloidogyne incognita was identified in the collection. A high level of genetic variability in wild melon germplasm was suggested by SSR analysis. Comparative analysis using SSRs of the genetic variability between wild melons from the north and other melons from the south and east regions of India and also reference accessions of cultivated melon from Spain, Japan, Korea, Maldives, Iraq and Israel, showed regional differentiation among Indian melon accessions and that Indian germplasm was not closely related to melon accessions from other parts of the world. Lupinus angustifolius L. is a Mediterranean species, domesticated in the 20th century, representing an important grain legume crop in Australia and other countries. This work is focused on the collection of wild germplasm and on the characterisation of morphological and molecular diversity of germplasm accessions. It reports the collection of 81 wild L. angustifolius accessions from the South and Centre of Portugal, available at the ‘Instituto Superior de Agronomia Gene Bank’, with subsequent morphological and molecular characterisation of a selection of these and other accessions. A multivariate analysis of morphological traits on 88 L. angustifolius accessions (including 59 wild Portuguese accessions, 15 cultivars and 14 breeding lines) showed a cline of variation on wild germplasm, with plants from Southern Portugal characterised by earlier flowering, higher vegetative development and larger seeds. AFLP and ISSR molecular markers grouped modern cultivars as sub-clusters within the wider diversity of wild germplasm, revealing the narrow pool of genetic diversity on which domesticated accessions are based. The importance of preserving, characterising and using wild genetic resources for L. angustifolius crop improvement is outlined by the results obtained.  相似文献

At the genebank of IPK (Leibniz Institute of Plant Genetics and Crop Plant Research) in Gatersleben, Germany, three long-term conservation methods (field genebank, in vitro slow-growth and cryopreservation) are used for mint germplasm. The plant material of the field genebank was the source to establish the slow-growth in vitro culture collection, from which the cryopreserved collection was set up, using a droplet-vitrification protocol. The genetic identity of 161 samples of three mint accessions (MEN 198, MEN 166 and MEN 186), stored for several years using those three methods, was studied using RAPD markers. Accession ‘MEN 198’ was the only one with a unique RAPD fragment pattern for all its samples, independently of the conservation procedure employed. The field collections of accessions ‘MEN 166’ and ‘MEN 186’ were made up of different genotypes. None of the genotypes detected in the plots of these accessions was represented in the in vitro and cryopreserved samples analyzed, which showed a unique genotype for each accession. From this work we conclude that, for an appropriate management of a germplasm collection of vegetatively propagated species, the determination of the genetic composition of the donor material and a periodical assessment of the preserved material should be carried out.  相似文献

Tomàtiga de Ramellet (Ramellet) is a traditional long-storage tomato from the Mediterranean Islands of Mallorca, Menorca, Eivissa (Ibiza), and Formentera (Balearic Islands). To investigate the observed phenotypic and shelf life variability within local varieties, we studied 142 accessions characterized as Ramellet varieties, and 29 accessions representing other local varieties. The 171 accessions constitute a new germplasm collection at the University of Balearic Islands (UIB). The evaluation of the collection demonstrated wide variability in most agronomic, morphological and quality traits of Ramellet accessions and clearly differentiates them from other local varieties with respect to fruit shape, size, fruit composition, and fruit storage. All accessions of Ramellet evaluated have the ripening mutation alcobaça (alc), suggesting an important role for this variant in controlling shelf life. Results of evaluation also showed wide variability between the Ramellet accessions for storage time, suggesting that other traits like fruit size and composition may play a role in fruit conservation. Observed variation within the Ramellet accessions is consistent with a genetically variable population or landrace group. The establishment of this collection will help prevent genetic erosion of this local landrace group and constitute a resource for future investigation and breeding.  相似文献

The tall fescue (Festuca arundinacea Schreb.) seed collection of the Centro de Investigaciones Agrarias de Mabegondo was surveyed to determine Acremonium infection rates. One hundred seeds from each of 19 accessions were microscopically examined for endophyte. Seventeen accessions (90%) contained endophyte with a range of 7 to 100%. Leaf sheaths of seedlings from the infected accessions were examined to determine the percentage of infection with viable endophyte. Only seven (41%) of the seventeen infected accessions collected and stored in 1995 at 4°C and 45–50% relative humidity contained viable endophyte with a range of 70 to 100%. While existing germplasm collections of Festuca arundinacea can provide only a small basis of Acremonium germplasm for study, newly collected Festuca arundinacea accessions will offer a greater diversity of Acremonium genotypes.  相似文献

Information on the variation available for different plant attributes has enabled germplasm collections to be effectively utilised in plant breeding. A world sourced collection of white clover germplasm has been developed at the White Clover Resource Centre at Glen Innes, New South Wales. This collection of 439 accessions was characterised under field conditions as a preliminary study of the genotypic variation for morphological attributes; stolon density, stolon branching, number of nodes, number of rooted nodes, stolon thickness, internode length, leaf length, plant height and plant spread, together with seasonal herbage yield. Characterisation was conducted on different batches of germplasm (subsets of accessions taken from the complete collection) over a period of five years. Inclusion of two check cultivars, Haifa and Huia, in each batch enabled adjustment of the characterisation data for year effects and attribute-by-year interaction effects. The component of variance for seasonal herbage yield among batches was large relative to that for accessions. Accession-by-experiment and accession-by-season interactions for herbage yield were not detected. Accession mean repeatability for herbage yield across seasons was intermediate (0.453). The components of genotypic variance among accessions for all attributes, except plant height, were larger than their respective standard errors. The estimates of accession mean repeatability for the attributes ranged from low (0.277 for plant height) to intermediate (0.544 for internode length).Multivariate techniques of clustering and ordination were used to investigate the diversity present among the accessions in the collection. Both cluster analysis and principal component analysis suggested that seven groups of accessions existed. It was also proposed from the pattern analysis results that accessions from a group characterised by large leaves, tall plants and thick stolons could be crossed with accessions from a group that had above average stolon density and stolon branching. This material could produce breeding populations to be used in recurrent selection for the development of white clover cultivars for dryland summer moisture stress environments in Australia. The germplasm collection was also found to be deficient in genotypes with high stolon density, high number of branches, high number of rooted nodes and large leaves. This warrants addition of new germplasm accessions possessing these characteristics to the present germplasm collection.  相似文献

In this paper, we report on morphological diversity, ecology and flowering phenological records of an endangered and an endemic species of Morocco Pyrus mamorensis Trab. For this study, a survey was undertaken in North-Western Morocco (Mamora forest and Zaër region) whereby 67 accessions of P. mamorensis were sampled. A set of 33 genotypes of Pyrus spp. from the international collection INRA-Angers (France) of the genus Pyrus were also analyzed. The variability of the total of 100 accessions was assessed by twenty-six characters including fourteen qualitative and twelve quantitative traits. The results demonstrated high levels of morphological variation in the sampled accessions of P. mamorensis and established the relationships among the local germplasm using the UPGMA cluster analysis and the factorial discriminant analysis. Furthermore, we were able to distinguish three types of flowering in the Mamora forest: early, intermediate and late flowering type. Otherwise, the UPGMA classification comparing local accessions of P. mamorensis to the international collection led to the formation of four major groups showing the closest correspondence to their geographical origins: (1) The North African species represented by P. mamorensis, (2) The East Asian pea species, (3) The West Asian species and (4) The European species. These findings suggest the importance of implementing a conservation strategy preserving the genetic resources of wild pear in North-Western Morocco.  相似文献

Thirty-seven chloroplast molecular markers were used to evaluate the genetic diversity and infer the phylogenetic relationship of 24 Algerian Citrus accessions from the Institut Technique de l′Arboriculture Fruitière et de la Vigne germplasm bank. The reliability and consistence of the clustering distribution was further asserted including 5 Spanish accessions from the Instituto Valenciano de Investigaciones Agrarias. The accessions were positioned on a phylogenetic tree of the genus Citrus based on previous analyses of the whole sequence of citrus chloroplast. Algerian accessions clustered into two main clades mostly differentiated by the occurrence of either mandarin or pummelo chloroplast types. All 7 mandarins analyzed were grouped in the same clade while the other cluster subdivided in 4 groups, included 1 lumia, 3 lemons, 2 grapefruits and 11 sweet oranges. Algerian grapefruit accessions were grouped together with the pummelos in a single cluster while all sweet oranges formed an independent and homogenous clade. Interestingly, the lemons studied were clustered in 3 different subclusters while Citrus lumia genotype was isolated in a different group. These results suggest that in contrast to the studied Algerian mandarins or sweet oranges, that share all the same mandarin or sweet orange chloroplast haplotype, the high diversity of current lemon accessions is at least partially correlated with the identity of different pummelo progenitors which evolved from a common ancestor. In addition, the data indicate that Citrus lumia is a new type of citrus chloroplast that appears to be phylogenetically related to the chloroplasts of the pummelo and micrantha group.  相似文献

The molecular diversity and population structure present in the Ethiopian sorghum collection maintained at the USDA–ARS National Plant Germplasm System (NPGS) has not been studied. In addition, 83 % of the accessions in the Ethiopian collection lack passport information which has constrained their evaluation and utility. Therefore, 137 Ethiopian accessions from NPGS were randomly selected and characterized with 20 strategically selected simple sequence repeat markers. These markers indentified 289 alleles with average polymorphic information content of 0.78. The allele frequency distribution reflects that 62 % of the alleles were rare (<0.05), 17 % range from 0.05 to 0.10, and 22 % were higher than 0.10. Expected and observed heterozygosity were estimated at 0.78 and 0.23, respectively, demonstrating Ethiopia has high sorghum genetic diversity germplasm. Population structure analysis indentified two subpopulations of 77 and 41 accessions, respectively, while a third group was constituted by 19 accessions whose classifications were not defined (i.e. hybrids). Analysis of molecular variances determined variation within subpopulations as the major source of variation. Likewise, genetic differentiation between subpopulations was moderate (Fst = 0.10). These results indicated that a continuous exchange of genes between subpopulations of sorghum exists in Ethiopia. The absence of a well defined population structure positioned this germplasm as an important resource for the study and dissection of agricultural traits by association mapping. However, genetic redundancy analysis indicated the presence of highly related accessions, therefore, strategic selected accessions must be consider prior to phenotype evaluation of larger number of accessions. The Ethiopian collection is composed of highly genetically diverse germplasm, and the genetic information presented herein is valuable to ex situ and in situ conservation programs to promote the use of this germplasm for breeding programs.  相似文献

Momordica cochinchinensis (Cucurbitaceae) is native to South East Asia and is important for its nutritional and medicinal properties. There is little information available on morphological and genetic diversity, crop selection and strategic germplasm collection for M. cochinchinensis. In this study, 42 accessions of M. cochinchinensis collected from Vietnam, Thailand and Australia were analysed for diversity study both at a morphological and molecular level. Both morphological and molecular analyses were in agreement and clustered the accessions based on the country of origin. The markers investigated in this study could be used for future applications with samples from unknown origins or for gene tracking for important cultivars. The Momordica accessions were highly polymorphic based on the markers suggesting high genetic diversity. Therefore the results from this study will be important in future strategic germplasm collection, conservation, horticultural development and commercial propagation of M. cochinchinensis.  相似文献

Cornus officinalis is plant species that is widely used in traditional Chinese medicine. To help the efficient utilization and conservation of this species, the genetic diversity of 73 germplasm resources collected from Zhejiang, Henan and Shaanxi provinces in China were used in a primary core collection. The germplasm resources were classified nine types and the analysis of morphological traits and chemical components showed wide variations among the germplasm types. The primary core collection of C. officinalis was evaluated by using 18 inter-simple sequence repeat (ISSR) markers and a mini core collection was developed based on three strategies on construction of a core germplasm resources. The ISSR results showed that a total of 233 alleles were identified in these 73 accessions, the genetic diversity (P?=?91.02%, H?=?0.293, I?=?0.446) revealed with an average of 2 alleles per locus. The genetic diversity index analysis suggested that there is a great richness and uniqueness of genetic variation in this primary core collection. Cluster analysis by UPGMA and STRUCTURE placed the 73-germplasm resources into four linkage clusters. Almost all samples from one sampling location were clustered together. Compared to the RS and SW strategy, the LDSS strategy proved to be more representative in the core collection of C. officinalis. Finally, a mini core collections by LDSS, composed of 18 accessions (24.66% of the primary core collection), let more than 94.85% of polymorphic loci remained, 97.55% of the observed number of alleles and 99.28% of the effective number of alleles of the mini core collection, respectively. The Nei’s gene diversity and Shannon’s Information index of core collection were 0.294 and 0.443, respectively. Taken all together, the results indicated that mini core collections constructed by the LDSS strategy represented their primary core collections and provided a rational framework for intensive surveys of natural variation in C. officinalis genetic resources.  相似文献
