An investigation of sheep flocks in the main sheep raising areas of New South Wales showed that the itchmite Psorergates ovis was frequently associated with fleece derangement. In 26 of the 41 flocks examined, P. ovis was the only ectoparasite detected. P. ovis and the sheep body louse Damalinia ovis, were found in 5 flocks. No external parasites were found on sheep examined from the 10 remaining flocks. The type of fleece derangement most frequently recorded was rubbing which in some cases was combined with areas of chewed fleece. Among flocks, there were positive relations between the prevalence of fleece derangement and prevalence of itchmite or scurf and between itchmite count and mean scurf score. Within flocks, itchmite infested sheep or sheep with scurf had higher prevalences of fleece derangement than sheep on which no mites or no scurf were found. Itchmite infested sheep had a higher prevalence of scurf than those with no detectable mite infestation. There were no significant differences in itchmite populations or fleece derangement between untreated flocks and flocks treated with synthetic pyrethroids, organophosphates or arsenic and rotenone.  相似文献

Naive and previously exposed young Hereford cattle were infested with Psoroptes ovis and maintained in stanchions for 7 weeks. Skin biopsy samples were examined at 1, 2, 5, and 7 weeks after infestation was induced, compared with those from noninfested control calves, and correlated with changes in the mite populations and development of dermatitis. Gross and microscopic appearance of lesions were qualitatively similar in both groups of infested calves. The lesions occurred early in the previously exposed calves, but progressed slowly, whereas lesions in the naive calves appeared later, but were rapidly progressive. The basic histopathologic pattern was chronic exudative superficial perivascular dermatitis compatible with allergy. The mite populations on the naive calves revealed exponential growth and high fecundity, and reached high population densities, whereas mite populations on previously exposed calves had low growth rates and low fecundity and reached population densities 100 to 1,000 times less than those on naive calves. Seemingly, there was a hypersensitivity to mite antigens. The hypersensitivity response may have been a mechanism by which humoral immunity was delivered to the mite microenvironment and may have contributed to acquired resistance by controlling mite populations. The mechanism of population control seems to be related partially to decreased fecundity of female P ovis.  相似文献

Lipid ingestion from sheep epidermis by Psoroptes ovis (Acari: Psoroptidae)   总被引:1,自引:0,他引:1  
Skin biopsies from three groups of sheep infested with Psoroptes ovis were cryofixed in liquid nitrogen to preserve outer epidermis with its lipid. From one group of five sheep (Group A), biopsies were taken from relatively healthy skin near the edge of scab lesions where mites had congregated. Frozen vertical sections from these biopsies were stained with Oil Red O and haematoxylin before mounting. Red lipid globules were plentiful within the body cavity of sectioned mites, but ingested lipid could not be distinguished from endogenous mite body stores by this technique. In a second group of five sheep (Group B), enclosed lumbar skin areas were coloured red with the lipophilic stains Oil Red O or Sudan IV. These enclosed coloured skin areas were inoculated with P. ovis and sampled by skin biopsy 1 or 4 days later. Cryofixed biopsies were cut into vertical frozen sections and mounted without staining for examination. Red-coloured lipid within mites, matching red-coloured lipid on outer epidermis, was evidence for the epidermal origin of P. ovis ingesta in the early stages of an infestation. From two other sheep (Group C), cryofixed biopsies were examined by scanning electron microscope and mites were seen with mouthparts embedded in abraded outer epidermis, but the precise depth of epidermal penetration was not determined. A light microscope survey of 3198 frozen vertical skin sections from the first 10 sheep (Groups A and B) showed that inner stratum corneum of the epidermis was the deepest penetration recorded for gnathosomes of P. ovis cryofixed in situ by liquid nitrogen. No structure of P. ovis was identified in dermal tissues.  相似文献

Psoroptes ovis of sheep origin, and Psoroptes cuniculi of rabbit origin were used in experimental infestations. In experiment I, groups of four rabbits and four sheep were infested with 50-100 mites of each isolate on the skin of the back (skin infestation, SI) or in the external auditory canal (aural infestation, AI). In rabbits, SI and AI with P. cuniculi and AI with P. ovis induced in all animals typical ear lesions and pronounced antibody reactions to P. cuniculi antigens in ELISA. After SI of rabbits with P. ovis no clinical signs were detected, no mites could be reisolated and no specific antibodies were detected. In sheep, P. ovis SI induced mange whereas AI did not induce typical clinical signs and mites could not be reisolated. In both these animal groups, ELISA revealed pronounced and comparable specific antibody reactions. After SI and AI with P. cuniculi no clinical symptoms were observed and no mites could be reisolated. Nevertheless, low levels of specific antibody were detected. In experiment II, clinical progression and antibody reactions to P. ovis SI in naive sheep were compared with sheep previously exposed to P. ovis or P. cuniculi. In both pre-exposed groups of animals, clinical signs appeared within 2 days after challenge infestation and three days earlier than in primarily infested sheep. Subsequently, no obvious difference in the clinical progression was observed between the three groups of animals. The results of this study document antigenetic crossreactivity of the two morphologically and genetically distinguishable Psoroptes species but differences in their biological behaviour and virulence which both are of epidemiological and taxonomic relevance.  相似文献

A single Merino sheep, artificially infested with the sheep scab mite, Psoroptes ovis, and a similarly infested Dorper sheep were placed with 9 uninfested Merino or 9 uninfested Dorper sheep respectively during winter and the rate of spread of infestation on the uninfested sheep observed. The same procedure was repeated in summer. It took 14 and 8 weeks respectively in winter before all sheep in the 2 groups displayed lesions of sheep scab, whereas in summer it took 10 and 12 weeks before all sheep had lesions.  相似文献

These studies were conducted to investigate the possible role of certain aspects of the on-host ecology and off-host survival of the sheep scab mite, Psoroptes ovis, in the dissemination of infestation. All developmental stages of the mite occurred in the fleece both proximally or distal to the skin of infested Merino and Dorper sheep. A larger proportion of mites was present in the fleece of Dorper sheep distal to the skin in the late afternoon and early morning than at other times during the day. Immature and adult mites readily transferred to tufts of wool or hair placed on infested sheep of both breeds. No mites could be found on wool or hair rubbed off onto tree trunks or branches or other structures in enclosures housing heavily infested sheep, nor could any mites be collected from the soil of these enclosures, whereas more than 80% of mites artificially seeded onto soil samples were recovered. The longest mean off-host survival times for larvae, nymphs, and male and ovigerous female mites were recorded at 10 degrees C, and were 9.25 days (RH = 90%), 15 days (RH = 33% and 75%), 10.5 days (RH = 75% and 90%) and 11.25 days (RH = 90%) respectively. Under natural climatic conditions ovigerous females in glass vials containing Merino wool survived for 17 days compared to 15 days for females in vials without wool; this difference was, however, not significant. The mean off-host pre-hatch period for eggs varied between 5.9 days (T = 25 degrees C and RH = 33%) and 22.1 days (T = 10 degrees C and RH = 75%), while the longest time individual eggs took to hatch at the latter temperature and RH was 31 days.  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed for the detection of specific antibodies against crude Psoroptes antigen. The diagnostic sensitivity was 93.7% in 191 sheep with clinical signs associated with mange. These animals originated from 29 flocks in which psoroptic mites were detected. All of 59 sheep infested with Psoroptes ovis were seropositive. Additionally, in 49% of 70 clinically unaffected sheep originating from P. ovis-infested flocks, specific antibodies could be detected, suggesting that asymptomatic infestations can be diagnosed by serology. The specificity of the ELISA was 96.5% as determined with 254 sheep originating from 44 flocks without clinical mange. Cross-reactivity in a low range was detected with selected sera of sheep with clinical chorioptic or forage mite infestations. Four sheep seroconverted 2 weeks after experimental P. ovis infestation, i.e. 2 weeks before clinical signs became obvious. After successful doramectin treatment of 14 sheep with naturally acquired P. ovis infestation, the ELISA values declined slowly but remained positive in seven cases beyond 17 weeks.  相似文献

Sheep scab mites, Psoroptes ovis, collected from a Merino donor sheep, were used to infest Merino and Dorper sheep, and Angora and Boer goats. Mites were placed on the sheep on 1 or 2 occasions and on 5 occasions on the goats. All the animals were examined at regular intervals for the presence of scab lesions and living mites. Both sheep breeds developed lesions, but those on the Merino sheep were always larger than those on the Dorper sheep at the same intervals after infestation. None of the goats developed lesions or showed signs of irritation, or harboured any mites.  相似文献

Skin biopsies from two Merino sheep heavily infested with Psorergates ovis were immersed in liquid nitrogen and cut into vertical frozen sections stained with lipophilic Sudan IV or Oil Red O and haematoxylin. A survey of the lateral distribution of mite sections showed a majority (ca 80%) were in or within 0.2 mm of the follicle mouth. A survey of vertical distribution showed no mite penetration deeper than inner stratum corneum where 57% of mite sections were seen; 30% were within outer stratum corneum or scurf; 13% were on the outer surface and less than 1% were detached. Lipid was the only material seen within stained mites at a location considered to be gut. This was supported by dosing sheep with quinacrine, taking biopsies at Day 6 and Day 14 and examining frozen sections under blue light. Fluorescent lipid was seen at a location considered to be mite gut. From these results it was recommended that acaricide treatments against P. ovis be lipophilic and administered transepidermally because less than 15% of mites were at a superficial location likely to be reached by topical application.  相似文献

Bovine scabies in a clinical problem during the cool autumn and cold winter months. In south Texas it is necessary to stanchion animals in order to elicit patent infestation during summer months. Environmental factors and grooming have been purported to account for summer resistance to infestation. However, stanchioned animals exhibit varied levels of susceptibility to infestation, suggesting that the noted variability may be influenced by the host immune response. In this study, animals were infested with Psoroptes ovis while in stanchions. The development of anti-P. ovis antibody activity was measured by the enzyme-linked immunosorbent assay and cellular immune function was monitored with mitogens and P. ovis antigens. A correlation was noted between the development of anti-P. ovis antibody activity and the increase in mite numbers and dermatitis. Coincident with the developing infestation was a suppression of T-cell function that appeared to be stress-related as a result of stanchioning. A working hypothesis is presented that attempts to correlate the developing humoral response and the depressed T-cell response with both an increasing dermatitis and mite population.  相似文献

The field efficacy of pour-on formulation of flumethrin 1 per cent was assessed in cattle naturally infested with Psoroptes ovis. In a finishing unit, 18 infested animals were selected, left untreated for one month and examined twice to monitor the progress of the infestation. They were treated twice 10 days apart with a pour-on formulation of flumethrin 1 per cent (2 mg/kg bodyweight). Skin samples were collected from the animals on days 0, 7, 14, 28 and 42 and examined for the presence of live mites. The clinical condition of the cattle was evaluated at the beginning and at the end of the trial using a standardised chart. Bodyweights were recorded on days -32, 0, 28 and 50. In addition the 65 cows in a calving unit were given the same treatment regimen and its efficacy was evaluated on days 0, 7, 14 and 28 in 11 P ovis infested animals. On the day of treatment most of the infested animals had a severe pyodermatitis. From day 7 onwards no live mites were found in the treated animals and there was a marked clinical improvement. The percentage efficacy on day 28 was 100 per cent in both units. In the finishing animals treatment was associated with a marked improvement in daily weight gains between day 0 and day 28.  相似文献

Five Merino and five Dorper sheep were artificially infested with the sheep scab mite Psoroptes ovis and the effect of infestation on their haematology, serum protein levels and live mass recorded for a period of 14 weeks. The reaction of the Merino sheep to infestation was more severe than that of the Dorper sheep. Haematological values fluctuated within the normal range during the assessment period. The mean haemoglobin concentration of the Merino sheep declined until antiparastic treatment was administered 10 weeks after infestation, after which it gradually increased. The lymphocyte counts of both breeds of sheep declined from 2 weeks to 10 weeks post-infestation, but increased after treatment, while the highest eosinophil counts were recorded in the Merino sheep at the height of the acute disease 8-10 weeks post-infestation. Serum albumin values for both breeds and serum globulin values for the Merino sheep were higher than normal during the entire 14-week observation period. A decrease in serum albumin and an increase in serum globulin concentration occurred at the height of infestation in both breeds. The mean live mass of a second group of five infested Merino sheep decreased by 6.4 kg over a 16-week period compared to a gain of 4.56 kg for five infested Dorper sheep.  相似文献

A sheep reared outdoors in Victoria was found to be heavily infested with a stored product mite, Sancassania berlesei (Acaridae), apparently subsequent to an earlier flystrike. Laboratory observation revealed the infestation to be self-sustaining. Shearing rapidly resolved the infestation but treatment with diazinon was ineffective. The infestation could be transferred to other sheep only in the presence of moisture; once established it caused an extensive skin lesion with considerable fluid loss, apparently contributing to the death of one animal. This lesion was extremely attractive to blowflies. Animals infested with mites showed few signs of irritation.  相似文献

A study was conducted in cattle experimentally infested with Psoroptes ovis to compare the prophylactic control against P. ovis provided by a long-acting injectable formulation of ivermectin to that of a commercially available injectable formulation of doramectin. Thirty Holstein steers were used. Animals were allocated by restricted randomization based on Day 0 body weight, forming six replicates of five animals each. Within each replicate, one animal was randomly allocated to one of the following treatment groups, with ivermectin and doramectin administered subcutaneously where indicated: (1) untreated controls; (2) ivermectin long-acting injectable (LAI) 630 mcg/kg, 56 days before challenge; (3) ivermectin LAI 630 mcg/kg, 42 days before challenge; (4) ivermectin LAI 630 mcg/kg, 35 days before challenge; or (5) doramectin 200 mcg/kg, 35 days before challenge. Animals were housed in individual pens 1 week prior to treatment. All animals were experimentally infested with P. ovis mites in the area between the shoulders, on the same day. Live mites were counted in scrapings from mange lesions at 2 sites on each animal 14, 21 and 28 days after challenge. Live mites were found in 33, 67 and 83% of the untreated controls on each respective evaluation. No P. ovis mites were found in steers treated with ivermectin LAI. Those animals showed lower (P < 0.05) mite counts than untreated controls on evaluations conducted 21 and 28 days after challenge. These results indicate that the ivermectin long-acting injectable formulation prevents induced infestations by P. ovis for at least 56 days after treatment. Doramectin injectable formulation, used at 200 mcg/kg, did not have a prophylactic effect 35 days after treatment.  相似文献

Ivermectin at 200 micrograms/kg bodyweight given either as a single subcutaneous injection or as an oral drench failed to eradicate Psoroptes ovis from artificially infested sheep. The oral drench reduced the mite populations by 43 per cent within 24 hours but no further significant decline was recorded over 38 days. The subcutaneous injection reduced the mite populations by 90 per cent after 10 days but live P ovis were present on all the treated animals 84 days after treatment. The efficacy of treatment was less the higher the initial mite burden. The injection had no effect on clinical sheep scab, and the disease continued to progress despite the mite mortality.  相似文献

The sampling distribution of Bovicola ovis (Schrank) on sheep was examined in two flocks, one with a light and one with a heavy infestation of lice. The derived distributions were used to calculate the sensitivity of detecting lice on individual sheep and in flocks by fleece parting regimes that varied in number of parts per animal and number of sheep per flock, different scenarios of flock sizes, proportion infested and louse density were examined. Lice were aggregated among fleece partings in the heavily infested flock and described by a negative binomial distribution with k values between 0.3 and 1.92. The distribution was indistinguishable from Poisson in the lightly infested flock. The assumed distribution had little effect on sensitivity, except when only one fleece part per animal was examined. On individual sheep where louse density was 0.5 per 10 cm part or greater, there were only marginal gains from inspecting more than 10 parts per animal. Increasing the number of sheep inspected always increased sensitivity more than increasing number of parts per sheep by an equivalent amount. This advantage was greatest in situations where a low proportion of sheep in the flock were infested with a high density of lice, and less where a low proportion of sheep were infested with a low density of lice, or a high proportion of sheep were infested with a high density of lice.  相似文献

OBJECTIVE: To evaluate efficacy of 2 deltamethrin emulsifiable concentrates that differed on the basis of vehicle (methyl glycol acetate [AMG] or 2-propylene glycol 1-methyl ether acetate [AMP]) for the treatment of sheep with mange. ANIMALS: 30 ewes between 11 months and 7 years old that weighed 16 to 71 kg and were naturally infested with Psoroptes ovis. PROCEDURE: Sheep were randomly allocated into 3 groups (13 sheep in group AMP, 13 sheep in group AMG, and 4 negative-control sheep). Each sheep was dipped twice (10-day interval between dippings) in the assigned formulation. Assessment of efficacy was performed on days 3, 7, 14, 21, 28, 35, 42, 49, 56, and 63 after the first dipping. Efficacy was assessed by determining the number of eggs or live mites on those days, as well as regrowth of wool at the end of the study. RESULTS: Psoroptic mange infestation was maintained in the 4 control sheep throughout the study. We did not detect live Psoroptes mites in scrapings after day 7 (AMP group) or after day 14 (AMG group). No parasites were seen after day 14 in either treatment group. Therefore, efficacy was 100% for both treatment groups from days 14 to 63. CONCLUSIONS AND CLINICAL RELEVANCE: The 2 formulations of deltamethrin were equally able to eradicate Psoroptes infestation of sheep after 2 dippings performed in accordance with the label recommendations.  相似文献

Calves infested with Psoroptes ovis (Hering, 1838) for the first time (naive) or previously infested calves were examined for serum antibody activity to P. ovis (obtained from rabbits) antigens by enzyme-linked immunosorbent assay (ELISA) to determine the temporal appearance of specific serum antibody activity. The development of the serum antibody activity to P. ovis antigens was then correlated with the development of lesions (% scab) and with changes in the number of P. ovis. The serum antibody activity [ELISA OD414 value (EODV) greater than 0.290] to P. ovis antigens in naive calves, in most cases, is first detected at the same time or slightly after the detection of mites and mite-caused lesions; and the development of specific serum antibody activity paralleled the increase in the P. ovis population and the percentage scab until 7 weeks post-infestation (PI). In previously infested calves, if serum antibody activity to P. ovis antigens was not already present from the previous infestation, specific serum antibody activity was detected at the same time and developed in a similar manner as in the naive calves. The serum antibody activity to P. ovis antigens could be detected after calves were relieved of their P. ovis burdens by pesticide treatment or after the P. ovis population began to decline when the calves were allowed to groom themselves. Serum antibody activity to P. ovis antigens was not detected in any of the control calves during the test period.  相似文献

The availability of large numbers of undamaged sheep scab mites, Psoroptes ovis, would be beneficial for discovery screening and development trials. There are several reported procedures for removing scab mites from sheep but they have limitations. To overcome this, a simple but versatile method employing the use of pumped saline was developed to remove all stages of the P. ovis mite from sheep. The method takes no more than 2 min to remove mites from the selected site with relative ease and is not affected by the condition of the fleece or lesion. The number of mites removed with the new method was 5-10 times more than detected by visual examination. These mites were undamaged and survived off-host for up to 16 days. The robust, portable equipment is easy to use under field conditions, making this method suitable for use as a diagnostic tool for early detection and monitoring of scab mites thus providing opportunities for development of novel alternative control strategies.  相似文献

In sheep, lesions caused by Psoroptes ovis, the sheep scab mite, may become colonized by Staphylococcus aureus. The present study compares clinical signs, lesional area and the immune response to P. ovis and S. aureus in P. ovis-infested sheep with and without secondary S. aureus infection. No differences were detected in the clinical signs or lesional areas in the S. aureus-positive and -negative sheep. However, 6 weeks after infestation an IgG but not IgE isotype antibody response to S. aureus was detected in the S. aureus-positive but not the S. aureus-negative group of sheep. This response targeted S. aureus antigens with molecular weights of approximately 36, 38, 50 and 65 kDa. In addition, 6 weeks after infestation an IgE response to P. ovis was detected in the S. aureus-positive but not the S. aureus-negative group of sheep.  相似文献
