In response to pathogens, the higher vertebrate innate immune system activates pro‐inflammatory caspase‐1 which is responsible for the processing and secretion of several important cytokines involved in the host's defence against infection. To date, caspase‐1 has been described in few teleost fish, and its activity has been demonstrated through substrate cleavage and inhibition by pharmacological agents. In this study, the detection of the active form of caspase‐1 during the immune response in salmonid fish is described, where two antibodies were produced. These antibodies differentially recognize the structural epitopes of the inactive pro‐caspase‐1 and the processed active form of the caspase. Firstly, caspase‐1 activation was demonstrated in vitro by ELISA, Western blotting and immunocytochemistry in rainbow trout macrophages exposed to different pathogen‐associated molecular patterns plus the pathogen Aeromonas hydrophila. This activity was clearly abrogated by a caspase inhibitor and seems to be unrelated to IL‐1β secretion. Caspase‐1 activation was then validated in vivo in gill cells from fish challenged with Aeromonas salmonicida. These results represent the first demonstration of caspase‐1 activation in salmonids, and the first evidence of the putative regulatory role which this protease plays in inflammatory response in this fish group, as described for some other teleosts and mammals.  相似文献

This study presents an Ex‐Post Facto analysis of diseases of wild juvenile and adults of Litopenaeus setiferus collected from a field survey at the Natural Protected Area of Terminos lagoon, southern Mexico. The objective of the present approach was to determine if sampling site and/or shrimp age were contributing risk factors for disease between juvenile and adult shrimp; if there was a determined period of time in a year cycle when diseases were more critical, and if the analysis would help to decision‐ making considering what population would pose less risk of disease‐carrying when withdrawn for experimental purposes; all under an after‐the‐fact (ex‐post facto) approach. We identified that juvenile shrimp were at more risk of contracting some diseases in the estuarine environment and June, July and August months, were found to be a critical period when colonizing and parasitic diseases maintained a significant high prevalence in the shrimp population. These assumptions may help for decision‐making when wild shrimp have to be withdrawn from their natural environment for research purposes.  相似文献

Sea urchins are commercially harvested for their edible gonads, also known as roe or uni. Most species, particularly those from the Northern hemisphere are reported to exhibit limited variation in gonad colour, which is proposed to be due to carotenoid pigments. The commercially harvested species from New Zealand, Evechinus chloroticus, and from Australia, Heliocidaris erythrogramma, however, exhibit considerable gonad colour variation, ranging from a culinary desirable light yellow/orange to an undesirable brown. Gonads from E. chloroticus and H. erythrogramma were characterized spectrophotometrically and the carotenoid content extracted and analysed by reversed‐phase high‐performance liquid chromatography. The main carotenoid found in the gonads of both species was 9′‐cis‐echinenone, with lower levels of all‐trans‐echinenone, lutein and isozeaxanthin. Carotenoid composition contributed to the yellow/orange colour components of the gonads, but was not correlated with the darker roe coloration observed in some E. chloroticus individuals. Our results highlight important considerations relating sea urchin gonad coloration to carotenoid content and as such will provide useful information for the future aquaculture and processing of these two species.  相似文献

The identification of anthropogenic and environmental drivers on length‐at‐age of fish stocks is important to understanding ecosystem dynamics and harvest intensity. We evaluated coastwide annual growth of n = 187,115 Atlantic Menhaden (Brevoortia tyrannus) and n = 299,185 Gulf Menhaden (B. patronus), using samples collected from the North, Mid‐, and South Atlantic from 1961 to 2016 and across the Gulf of Mexico from 1977 to 2016. Using hierarchical models of age 1 growth and age 2 growth, we evaluated a suite of candidate predictors including fishery landings, easterly (U) and northerly (V) wind velocity, river discharge, juvenile abundance, and the Atlantic Multi‐decadal Oscillation (AMO). We found age 2 growth rates were smaller than age 1 growth rates for both species and that Atlantic Menhaden growth rates were 3–4 times greater than Gulf Menhaden. Age 1 growth rate of Atlantic Menhaden was positively affected by landings lagged by one year, indicating a density‐dependent mechanism. In addition, AMO (negative effect), and wind U (positive effect) and wind V (negative effect) in the North Atlantic region were significant factors influencing coastwide age 1 Menhaden growth. Wind V (negative effect) and AMO (positive effect) influenced age 1 Gulf Menhaden growth. No environmental factors were found to have an effect on age 2 Atlantic Menhaden growth, and AMO was the only significant predictor (weak negative effect) of age 2 Gulf Menhaden growth. Fishing pressure was the primary influence on age 1 Atlantic Menhaden growth, whereas age 1 Gulf Menhaden growth was primarily influenced by environmental conditions.  相似文献

The natural amorphous polymer poly‐β‐hydroxybutyrate (PHB‐A: lyophilized Ralstonia eutropha containing 75% PHB) was used as a biological agent to control bacterial pathogens of blue mussel (Mytilus edulis) larvae. The larvae were supplied with PHB‐A at a concentration of 1 or 10 mg/L for 6 or 24 hr, followed by exposure to either the rifampicin‐resistant pathogen Vibrio splendidus or Vibrio coralliilyticus at a concentration of 105 CFU/ml. Larvae pretreated 6 hr with PHB‐A (1 mg/L) survived a Vibrio challenge better relative to 24 hr pretreatment. After 96 hr of pathogen exposure, the survival of PHB‐A‐treated mussel larvae was 1.41‐ and 1.76‐fold higher than the non‐treated larvae when challenged with V. splendidus and V. coralliilyticus, respectively. Growth inhibition of the two pathogens at four concentrations of the monomer β‐HB (1, 5, 25 and 125 mM) was tested in vitro in LB35 medium, buffered at two different pH values (pH 7 and pH 8). The highest concentration of 125 mM significantly inhibited the pathogen growth in comparison to the lower levels. The effect of β‐HB on the production of virulence factors in the tested pathogenic Vibrios revealed a variable pattern of responses.  相似文献

The aim of this study was to evaluate the effects of dietary lipids on protein‐sparing and lipoprotein lipase (LPL) mRNA expression in culture using 360 juvenile soft‐shelled turtles (Pelodiscussinensis) (initial weight 4.26 ± 0.14 g). The turtles were allotted to six diets with three duplicates for 60 days. A control diet with 46% protein and 55% fishmeal (CD) and five isonitrogenous diets with 41.3% protein and 45% fishmeal (F, S, L1, L2 and L3) were used, containing the following three lipid types: fish oil, soybean oil and mixed oils (soybean oil: fish oil = 1:1). The results showed that the survival rate was not affected by dietary lipids (P > 0.05). The highest weight gain and lowest feed coefficient ratio were seen in the L3 diets (P < 0.05). Turtles fed with L2 and L3 diets had lower superoxide dismutase activities, higher alanine aminotransferase activities and higher cholesterol concentrations than those exposed to other diets (P < 0.05). Hepatic LPL activity and LPL mRNA expression were higher in the L3 diets than in the other diets (P < 0.05). Overall, there were obvious protein‐sparing effects of dietary lipids and LPL mRNA expression was stimulated by high dietary lipids in soft‐shelled turtles in this study.  相似文献

The present study was conducted to administer a commercial multi‐strain probiotic (MP), Yilibao (Bacillus velezensis:Bacillus cereus:Lactobacillus casei = 2:2:1), in Nile tilapia, Oreochromis niloticus. In terms of aerobic Bacillus spp. counts, we produced five diets containing 0, 0.34, 1.68, 3.36 and 6.72 g/kg of MP dry product (Control, T1, T2, T3, T4). Seven hundred and fifty tilapia juveniles (13.26 ± 0.01 g) distributed into 25 tanks in five replications were fed their diet for eight weeks. Results showed that fish fed T3 and T4 diets displayed significantly higher final body weight and weight gain (p < .05). Further, all MP‐treated fish exhibited remarkably decreased plasma lipid profiles (cholesterol, triglycerides) (p < .05) and fish fed T3 and T4 diets displayed significantly higher plasma myeloperoxidase activity and complement C3 content (p < .05). Additionally, fish fed T3 and T4 diets exhibited significantly promoted total antioxidant capacity, glutathione peroxidase activity, glutathione level and declined malondialdehyde content in the plasma and/or liver (p < .05). Moreover, significantly elevated chymotrypsin activity, villus height and intraepithelial lymphocytes counts were found in fish fed T3 and T4 diets (p < .05). As fish fed T3 and T4 diets surpassed the other treatments in growth, immune–antioxidative status and gut health, the recommended dose for tilapia is 3.36–6.72 g/kg.  相似文献

Almost all environmental management comes at an economic cost that may not be borne equitably by all stakeholders. Here, we investigate how heterogeneity in catch and profits among fishers influences the trade‐off among the triple‐bottom‐line objectives of recovering a fish population, maximizing its economic value and distributing restrictions equitably across fishers. As a case‐study, we examine management reform of an ecologically and economically important coral reef fishery operating within the Great Barrier Reef Marine Park. Using a simulation model, we find that total profitability of the fishing industry is 40% lower if recovery plans are equitable when compared to the most economically efficient plan. However, efficient recovery plans were typically highly inequitable because they required some fishers to cease fishing. Equity was defined according to different norms, and the efficiency loss was greatest when catch losses were shared equally across fishers rather than in proportion to their historical catch. We then varied key social, economic and biological parameters to identify cases when equity and efficient recovery would trade‐off most strongly. Recovery plans could be both efficient and equitable when heterogeneity in fisher's catches and individual catch efficiencies was lower. If fishers were homogenous then equitable plans could have maximal economic efficiency. These results emphasize the importance of considering heterogeneity in individual fishers when designing recovery plans. Recovery plans that are inequitable may often fail to gain stakeholder support, so in fisheries with high heterogeneity we should temper our expectations for marked increases in profits.  相似文献

Netminder®, a water‐based silicone barrier release coating, was highly effective in reducing biofouling of lantern nets during grow‐out/overwintering of bay scallops, Argopecten irradians irradians, in a large‐scale restoration programme in New York, USA. Compared with untreated nets, scallops held in Netminder®‐coated nets experienced reduced survival after 8 months, probably due to higher loads of epibionts on their shells. However, this was countered by higher reproductive and overall condition of scallops in treated vs. untreated nets; there were no consistent differences in shell growth in different net treatments. Although further testing is recommended under a wider range of conditions, we conclude that for the purposes of our scallop restoration work, coating nets with Netminder® would eliminate the need for a gear change in spring, thus reducing labour costs; the coating process should be refined so that Velcro closures of nets do not become sealed shut.  相似文献

The anadromous, or sea‐going, life history form of brown trout, or sea trout ( Salmo trutta), may lead to potential mixing of populations while foraging at sea. In this article, we assess the potential that multiple populations are using common semi‐enclosed estuaries and quantify the potential levels of straying (i.e. dispersal) of foreign‐produced individuals into populations by using otolith chemical signatures as natural ‘tags’. To do so, we created a database of juvenile fish otolith chemistry (a marker of freshwater production) from four rivers and compared the chemistry of harvested fish in two estuaries important to anglers, the Renews River and Chance Cove Brook, to the database. A discriminant function analysis revealed significant differences in the otolith chemistry of juvenile fish inhabiting the four rivers with a 97% cross‐validated accuracy when classifying individual juveniles to their natal river, indicating our baseline was robust. When assigning adults caught over 3 years (2007–2009) in the recreational fishery in the Renews River estuary, it was determined that over 95% of the fish caught each year originated from Renews River. In contrast, harvested fish in Chance Cove during 2009 were disproportionately comprised of fish produced in Renews River, suggesting the potential for source‐sink population dynamics in Newfoundland. Taken as a whole, these results indicate limited population mixing in nearshore estuaries of this region, but also highlight the potential for some populations to subsidise the harvest by anglers in different areas.  相似文献

A feeding trial was conducted to explore the effect of dietary Houttuynia cordata leaf extract (HCLE) and leaf meal (HCLM) on immunological responses and expression of interferon‐gamma (IFN‐γ) and tumour necrosis factor‐alpha (TNF‐α) gene in Labeo rohita fingerlings. Six isonitrogenous (350 g/kg CP) and isocaloric (17 MJ/kg DE) purified experimental diets were formulated with Houttuynia cordata leaf extract and leaf meal comprising control, C (0 g/kg HCLE and HCLM), E2.5 (2.5 g/kg HCLE), E5 (5 g/kg HCLE), E10 (10 g/kg HCLE), M10 (10 g/kg HCLM) and M20 (20 g/kg HCLM). Labeo rohita fingerlings (3.37 ± 0.23 g) were distributed in six experimental groups in triplicates following the complete random distribution. Fish were fed twice daily with respective experimental diets for a period of 60 days. A significantly (p < .05) lower lactate dehydrogenase, malate dehydrogenase, superoxide dismutase and catalase activities were registered in supplemented groups compared with control group, while respiratory burst and lysozyme activities were significantly (p < .05) higher in E10 group compared with other experimental groups. Haemoglobin, total leucocyte count, total erythrocyte count and haematocrit values were significantly (p < .05) higher in E10 group. The expression of IFN‐γ and TNF‐α in both the kidney and liver was significantly up‐regulated in leaf extract and meal supplemented groups with the highest expression in the fish of E10 group. Overall, these results suggest that the dietary supplementation of ethanolic extract of the Houttuynia cordata leaf at 10 g/kg level can enhance the immune response of L. rohita fingerlings.  相似文献

Shrimp farming is a fast‐expanding activity that has supported the growth in the supply of these crustaceans to consumers around the world. However, the end product is vulnerable to contamination at all stages of the process, including the rearing tanks, where current practices prioritize to raise stocking densities and the minimization of water renewal. It is thus important to evaluate the potential of these systems for the proliferation of undesirable microorganisms, which may render the product unfit for human consumption. In the present study, the presence of coagulase‐positive Staphylococcus, Salmonella spp., Vibrio spp., and total and thermotolerant coliforms was verified in biofloc tank and conventional pond systems used for the rearing of Litopenaeus vannamei in Pernambuco, Brazil, and the results were compared with the legislation regulating the marketing of fresh shrimp. Samples were collected from two biofloc tanks with a density of 375 shrimp m?2, and two conventional ponds with 12 shrimp m?2. None of the samples tested positive for either Salmonella spp. or coagulase‐positive Staphylococcus, which is consistent with the legislation. While no standards are defined legally for Vibrio spp. or coliforms, very low concentrations were recorded in both systems, in comparison with other studies. While some variation in bacterial contamination was observed over the rearing process, the end product of both systems was fit for human consumption. The results of the study indicate that, while the water is not renewed in the biofloc system, the development of undesirable microorganisms can be controlled, with no adverse effects for the end product.  相似文献

This study aimed at evaluating the effects of two starvation modes (fasting‐feeding mode and feeding‐fasting mode) on survival, growth and development of mysis larvae of Exopalaemon carinicauda. It showed that food restriction significantly affected the survival, growth and development in both starvation modes. With the extension of initial starvation period, a significantly reduced survival rate during metamorphosis was observed. In the fasting‐feeding mode, the duration of metamorphosis was extended and the body lengths of the individuals, which experienced successful metamorphosis to the post‐larval stage 1, were much shorter. In the feeding‐fasting mode, the extension of initial feeding period led to dramatically increased survival rate and body lengths, while there was no significant difference in duration of metamorphosis of those groups that reached to post‐larval stage. The 50% point‐of‐no‐return and the 50% point‐of‐reserve‐saturation were 3.85 days and 4.81 days respectively. The results of this study indicate that E. carinicauda mysis have the ability to withstand or recover from relatively extended starvation, but to ensure the success of commercial seedling, timely and adequate feeding is necessary. The information obtained from this study could provide a basis to optimize the feeding schedule of artificial seedling of E. carinicauda.  相似文献

In the present study, the effects of Artemia supplemented with 2‐β‐mercapto‐ethanol (β‐ME) treated yeast cell, Saccharomyces cerevisiae, on growth and reproductive performance, lysozyme activity, and disease resistance to Aeromonas hydrophila of freshwater ornamental species, Poecilia latipinna, were investigated. Within 60 days, molly fish were fed with three treatments including commercial food (T1), un‐supplemented Artemia (T2), and Artemia supplemented with β‐ME‐treated yeast cell (at concentration of 4 × 107 CFU/L of water) (T3). After the feeding period, the fish were exposed to 100 μl of a suspension (1.1 × 107 cells/ml) of A. hydrophila (BCCM5/LMG3279) and the cumulative mortality rates were recorded for 12 days. No significant difference was found between survival rate and growth performance of P. latipinna except for weight gain that was higher in fish fed through Artemia supplemented with β‐ME‐treated yeast cell compared to control group. Fecundity rate was significantly improved in fish fed using T3 with the maximum amount of 49.5 ± 2.29 per female (p < 0.05). Besides, lysozyme activity was significantly increased in group 3 (p < 0.05). Moreover, lowest fish mortality was significantly observed in this treatment (p < 0.05). In addition, the number of colonies formed by yeast cell in T3 (634 × 103 CFU/g intestine) showed significant difference with other treatments (p < 0.05). In sum, Artemia enriched with β‐ME‐treated yeast improved reproductive indices, immune responses, and resistance against A. hydrophila of P. latipinna.  相似文献
