Though the relationship between dietary fiber and physical health has been investigated widely, the use of dietary fiber from marine plants has been investigated relatively rarely. The Saccharina japonica byproducts after the production of algin contain a large amount of insoluble polysaccharide, which will cause a waste of resources if ignored. Soluble dietary fiber (SDF)prepared from waste byproducts of Saccharina japonica by alkaline hydrolysis method for the first time had a wrinkled microscopic surface and low crystallinity, which not only significantly reduced liver index, serum levels of aspartate aminotransferase (AST) and alanine amiotransferase (ALT), and liver fat accumulation damage to the livers of obese diabetic mice, but also activated the PI3K/AKT signaling pathway to increase liver glycogen synthesis and glycolysis. By LC-MS/MS employing a Nexera UPLC tandem QE high-resolution mass spectrometer, the 6 potential biomarker metabolites were screened, namely glycerophosphocholine (GPC), phosphocholine (PCho), pantothenic acid, glutathione (GSH), oxidized glutathione (GSSG), and betaine; several pathways of these metabolites were associated with lipid metabolism, glycogen metabolism, and amino acid metabolism in the liver were observed. This study further provided a detailed insight into the mechanisms of SDF from Saccharina japonica byproducts in regulating the livers of obese mice with type 2 diabetes and laid a reliable foundation for the further development and utilization of Saccharina japonica.  相似文献

Geoditin A, an isomalabaricane triterpene isolated from the marine sponge Geodia japonica, has been demonstrated to dissipate mitochondrial membrane potential, activate caspase 3, decrease cytoplasmic proliferating cell nuclear antigen (PCNA), and induce apoptosis of leukemia cells, but the underlying mechanism remains unclear [1]. In this study, we found fragmentation of Golgi structure, suppression of transferrin receptor expression, production of oxidants, and DNA fragmentation in human colon cancer HT29 cells after treatment with geoditin A for 24 h. This apoptosis was not abrogated by chelation of intracellular iron with salicylaldehyde isonicotinoyl hydrazone (SIH), but suppressed by N-acetylcysteine (NAC), a thiol antioxidant and GSH precursor, indicating that the cytotoxic effect of geoditin A is likely mediated by a NAC-inhibitable oxidative stress. Our results provide a better understanding of the apoptotic properties and chemotherapeutical potential of this marine triterpene.  相似文献

In this study, we investigated whether fermented rice bran (FRB) can ameliorate the oxidative stress induced by high glucose and hydrogen peroxide (H2O2) in 3T3-L1 adipocytes by analyzing reactive oxygen species (ROS), oil red O staining, as well as the expression of mRNAs related to glucose homeostasis and adipogenesis. It was first confirmed that rice bran fermented by Issatchenkia orientalis MFST1 extract increased free phenolic content compared to non-fermented rice bran. The FRB extract strongly inhibited ROS generation and upregulated the expression of PPAR-γ and adiponectin. Moreover, FRB upregulated GLUT4 related to glucose transportation and insulin sensitivity. Taken together, FRB extract ameliorated oxidative stress-induced insulin resistance by neutralizing free radicals and upregulating adiponectin in adipocytes. Our results provide information toward understanding the beneficial effects of FRB on oxidative stress.  相似文献

The effect of redox reactions catalysed by lactobacilli on properties of wheat gluten was followed during sourdough fermentations. Thiol levels in doughs remained high in doughs fermented with Lactobacillus sanfranciscensis or in chemically acidified doughs to which 10 μmol g–1 glutathione were added. In chemically acidified doughs or sourdoughs fermented with L. sakei or L. perolens, the thiol levels decreased during fermentation. Thiol-groups in gluten proteins were estimated by RP-HPLC separation of DAC-maleimide-labeled, propanol-soluble proteins extracted from wheat doughs. An increase of the SH groups in gluten proteins was observed in protein fractions from sourdoughs fermented with L. sanfranciscensis but not from chemically acidified doughs. A glutathione-reductase was expressed in L. sanfranciscensis and during its growth in sourdough resulted in the reduction of extracellular GSSG to GSH. It was concluded that in addition to the pH-dependent activity of cereal proteases, redox reactions catalysed by lactobacilli determine gluten quality during sourdough fermentations. The formation of thiols by L. sanfranciscensis interferes with gluten polymerisation. Due to differences in their central carbon metabolism, homo- or facultative hetero-fermentative lactobacilli have opposite effects on redox-reactions in wheat doughs.  相似文献

Breeding program strategies to develop novel short grain white rice varieties such as japonica (short grain) that introgress biotic stress resistance and high grain quality have been developed using indica rice (Pin Kaset + 4 and Riceberry) for applications in japonica rice (Koshihikari) improvement. Four breeding lines showing promising agronomic performance with short grain and low amylose content (< 20%) were obtained. In addition, sensory testing of these breeding lines showed high scores that similar to Koshihikari. Two promising lines, KP48-1-5 and KP48-1-9, which possessed a combination of four genes resistance to different biotic stresses (Bph3 + TPS + Xa21 + Pi-ta) and four genes for grain quality (GS3 + SSIIa + wxb + badh2), were developed using marker-assisted selection (MAS) with the pedigree method. The current study clearly illustrated the successful use of MAS in combining resistance to multiple biotic stresses while maintaining a high yield potential and preferred grain quality. Moreover, the results indicated that this breeding program, which includes crossing temperate japonica with indica, can create novel short grain rice varieties adapted to a tropical environment, like the japonica type.  相似文献

Traditionally, upland rice is grown in Asia in low-input, subsistence systems. More productive upland systems, using more fertilizer and improved varieties, are emerging in China and Philippines, and could contribute to productivity increases in rainfed environments in other countries. Here, we evaluate, on-station and on-farm, the yield under upland management of improved indica upland cultivars selected for yield under high-fertility conditions. These cultivars are compared with traditional and improved tropical japonica upland varieties, and with elite indica high-yielding varieties (HYV) developed for irrigated lowland production, to characterize the features of varieties that produce high yields in favorable upland environments. Forty-four improved and traditional varieties and experimental lines were evaluated in irrigated lowland, non-stressed upland, moderately stressed upland, severely water-stressed upland, and low-fertility upland environments in southern Luzon, Philippines. Correlations between yields in non-stress and mild-stress environments were low but positive. Some cultivars, like IR55423-01, were among the highest yielding under both conditions, indicating that high yield and moderate water-stress tolerance can be combined. Upland-selected indica varieties yielded 3.56 t ha−1 in favorable upland environments on-station in southern Luzon, outperforming improved tropical japonica and irrigated varieties by 23 and 69%, respectively. They were also the highest-yielding class in infertile, acid soils. The improved upland indica cultivars are about 110 cm tall under favorable upland conditions and maintain a harvest index of nearly 0.4, or about one-third higher than other cultivar types. The best upland-adapted rice varieties produced average yields on-farm of 3.3 and 4.1 t ha−1 in southern Luzon and Yunnan, respectively, outyielding traditional checks by 30–50% with moderate N application. Screening under both high-fertility, non-stress conditions and moderate reproductive-stage stress appears to be needed to develop cultivars combining high-yield potential with drought tolerance. Upland-adapted indica cultivars offer a new approach to increasing productivity and reducing risk in Asian rainfed rice systems.  相似文献

Asian cultivated rice shows allelic variation in sodium transporter, OsHKT1;5, correlating with shoot sodium exclusion (salinity tolerance). These changes map to intra/extracellularly-oriented loops that occur between four transmembrane-P loop-transmembrane (MPM) motifs in OsHKT1;5. HKT1;5 sequences from more recently evolved Oryza species (O. sativa/O. officinalis complex species) contain two expansions that involve two intracellularly oriented loops/helical regions between MPM domains, potentially governing transport characteristics, while more ancestral HKT1;5 sequences have shorter intracellular loops. We compared homology models for homoeologous OcHKT1;5-K and OcHKT1;5-L from halophytic O. coarctata to identify complementary amino acid residues in OcHKT1;5-L that potentially enhance affinity for Na+. Using haplotyping, we showed that Asian cultivated rice accessions only have a fraction of HKT1;5 diversity available in progenitor wild rice species (O. nivara and O. rufipogon). Progenitor HKT1;5 haplotypes can thus be used as novel potential donors for enhancing cultivated rice salinity tolerance. Within Asian rice accessions, 10 non-synonymous HKT1;5 haplotypic groups occur. More HKT1;5 haplotypic diversities occur in cultivated indica gene pool compared to japonica. Predominant Haplotypes 2 and 10 occur in mutually exclusive japonica and indica groups, corresponding to haplotypes in O. sativa salt-sensitive and salt-tolerant landraces, respectively. This distinct haplotype partitioning may have originated in separate ancestral gene pools of indica and japonica, or from different haplotypes selected during domestication. Predominance of specific HKT1;5 haplotypes within the 3 000 rice dataset may relate to eco-physiological fitness in specific geo-climatic and/or edaphic contexts.  相似文献

In this work, water-extraction of Vitis Vinifera L. (Black Grenache) leaves and analysis of aqueous extracts for anthocyanins were investigated. Vegetable leaves were cultivated in North of Tunisia at different dates. Anthocyanins were identified as main constituents in these natural dyeing materials using high performance liquid chromatography (HPCL). Dyeability of wool and its fastness properties using Vitis Vinifera L. leaves extract were also determined. Aqueous extract of Vitis Vinifera L. leaves has markedly yielded shades with good fastness properties. Thus, affordability is a point favorable for Vitis Vinifera L. leaves. Effects of dye bath pH and temperature on dyeability and fastness proprieties of wool fabrics by aqueous extract of Vitis Vinifera L. leaves were evaluated. Experimental results showed that fastness properties of dyed fabrics ranged from average to very good. Vitis Vinifera L. leaves extracts showed potential to dyeing wool fibers under highly acidic conditions (pH ≤2). The results obtained here revealed that natural dyes extracted from Vitis Vinifera L. leaves can be cost-effective for dyeing wool fabrics.  相似文献

The essential oils obtained by hydrodistillation from leaves of Chinese native Cryptomeria japonica and Cryptomeria fortunei were analyzed by GC and GC-MS. Fifty-seven compounds were identified in the essential oils of C. japonica with α-elemol (20.12%), kaur-16-ene (14.84%), β-phellandrene (5.97%), β-elemene (5.87%), α-eudesmol (5.62%) and β-eudesmol (5.03%) as main constituents. Forty components were identified in the oil of C. fortunei with kaurene (34.04%), α-elemol (13.34%), γ-eudesmol (10.80%), β-eudesmol (10.16%), α-pinene (2.75%) and γ-cadinene (1.92%) as the most abundant components. This study demonstrated the occurrence of α-elemol chemotype in C. japonica and kaurene chemotype in C. fortunei from China. The essential oil compositions of two Cryptomeria samples were shown that they can be used for green plant protection, pharmaceutical, perfume and food industries.  相似文献

Although the genetic diversity of rice germplasm has been well characterized globally, few studies have taken an in-depth view of a large number of rice landraces on a local scale. To better understand the relationships between rice genetic diversity and associated geographic and cultural factors, we collected and characterized 183 rice landraces from 18 villages along the Bahau and Kayan rivers in the Indonesian province of East Kalimantan on the island of Borneo. A genetic diversity analysis using 30 microsatellite markers detected a clear distinction between the indica and japonica varietal groups (F st?=?0.59), with 80% of the landraces identified as tropical japonica and 20% indica, which largely correlated with the field-level ecotypes: upland japonica and lowland indica. Indigenous knowledge from local farmers was gathered about the names, origins, and uses of the landraces, which provides a rich background to compare with the genetic relationships of these traditional varieties.  相似文献

Atopic dermatitis (AD) is a chronic inflammatory skin disease. Juniperus Chinensis heartwood has been known to have an excellent antibacterial and anti-inflammatory effect. This study examined the potential of the methanol extract of the Juniperus Chinensis heartwood as a natural dye with an efficacy for the treatment of atopic dermatitis. In an attempt to examine the effect of the Juniperus Chinensis heartwood extract, a skin lesion such as that of AD was induced in the NC/Nga AD mice model by topical administration. DNCB (2.4-dinitrochlorobenzene) was used to induce such AD-like skin lesions. The efficacy of the Juniperus Chinensis heartwood extract in the NC/Nga mice model was assessed by severity of skin lesion (NC mouse score), change in organ weight, serum IgE level, change in epidermal thickness, and mast cell count. The Juniperus Chinensis heartwood extract improved such skin lesions in these NC/Nga mice, and decreased spleen weight and serum IgE level. The results of histopathologic examination revealed that the treatment with the Juniperus Chinensis heartwood extract decreased epidermal thickness and mast cell count. In conclusion, the suppressive effect of local administration of the Juniperus Chinensis heartwood extract verified its potential for application as a natural dye with an efficacy of treatment for atopic dermatitis.  相似文献

Background: Cicer arietinum (Chickpea) is one of the most important harvests in the world with high nutritional value. Lack of essential oils in the seeds of Chickpea is an advantage in search for drug-like molecules with less toxicity. We evaluated anticonvulsant effect of C. arietinum in common animal models of epilepsy. Methods: Dichloromethane extract was obtained from C. arietinum seeds by percolation. Acute toxicity of the extract was assessed in mice. Protective effect of the extract was examined against tonic seizures induced by maximal electroshock (MES; 50 mA, 50 Hz, 1 s) in mice, clonic seizures induced by pentylenetetrazole (PTZ; 60 mg/kg, i.p.) in mice, and electrical kindling model of complex partial seizures in rats. The extract was fractionated by n-hexane to f1 and f2 fractions. The extract and fractions underwent phytochemical analysis by thin layer chromatography. The active anticonvulsant fraction, f1, was subjected to matrix assisted laser desorption/ionization (MALDI) mass analysis. Results: The crude extract had neither toxicity up to 7 g/kg nor protective activity in MES and kindling models. However, it significantly inhibited clonic seizures induced by PTZ. f1 fraction mimicked protective effect of the extract. Phytochemical screening revealed the presence of considerable amount of alkaloids in the extract and fractions. Moreover, a novel structural class was detected in f1 fraction. Conclusion: Finding an anticonvulsant molecule pertaining to a new structural class in the seeds of C. arietinum promises an effective and inexpensive source of antiepileptic medication. Background

Cadmium (Cd) accumulation in rice followed by transfer to the food chain causes severe health problems in humans. Breeding of low Cd accumulation varieties is one of the most economical ways to solve the problem. However, information on the identity of rice germplasm with low Cd accumulation is limited, particularly in indica, and the genetic basis of Cd accumulation in rice is not well understood.

Results

Screening of 312 diverse rice accessions revealed that the grain Cd concentrations of these rice accessions ranged from 0.12 to 1.23?mg/kg, with 24 accessions less than 0.20?mg/kg. Three of the 24 accessions belong to indica. Japonica accumulated significantly less Cd than indica (p < 0.001), while tropical japonica accumulated significantly less Cd than temperate japonica (p < 0.01). GWAS in all accessions identified 14 QTLs for Cd accumulation, with 7 identified in indica and 7 identified in japonica subpopulations. No common QTL was identified between indica and japonica. The previously identified genes (OsHMA3, OsNRAMP1, and OsNRAMP5) from japonica were colocalized with QTLs identified in japonica instead of indica. Expression analysis of OsNRAMP2, the candidate gene of the novel QTL (qCd3–2) identified in the present study, demonstrated that OsNRAMP2 was mainly induced in the shoots of high Cd accumulation accessions after Cd treatment. Four amino acid differences were found in the open reading frame of OsNRAMP2 between high and low Cd accumulation accessions. The allele from low Cd accumulation accessions significantly increased the Cd sensitivity and accumulation in yeast. Subcellular localization analysis demonstrated OsNRAMP2 expressed in the tonoplast of rice protoplast.

Conclusion

The results suggest that grain Cd concentrations are significantly different among subgroups, with Cd concentrations decreasing from indica to temperate japonica to tropical japonica. However, considerable variations exist within subgroups. The fact that no common QTL was identified between indica and japonica implies that there is a different genetic basis for determining Cd accumulation between indica and japonica, or that some QTLs for Cd accumulation in rice are subspecies-specific. Through further integrated analysis, it is speculated that OsNRAMP2 could be a novel functional gene associated with Cd accumulation in rice.

A method for batch preparation of fucoxanthin from brown algae was established, which possessed the advantages of high yield and high purity. The ultrasonic-assisted extraction method was used to obtain a crude extract from Sargassum fusiforme as the separation sample. Then the crude extract was separated by elution-extrusion countercurrent chromatography. The optimum preparation conditions of fucoxanthin were determined as follows: n-hexane-ethanol-water (20:9:11, v:v:v) as a two-phase solvent system, the mobile phase flow rate was 5 mL min−1, the revolution speed was 800 r min−1, the loading capacity was 60 mg 10 mL−1 and the temperature was 25 °C. By this method, 12.8 mg fucoxanthin with a purity of 94.72% was obtained from the crude extract of Sargassum fusiforme. In addition, when the loading capacity was 50 mg 10 mL−1, the purity of fucoxanthin reached 96.01%. Two types of by-products, chlorophyll and pheophytin, could also be obtained during the process of separation. This optimal method was further applied to separate fucoxanthin from Laminaria japonica and Undaria pinnatifida, and 6.0 mg and 9.7 mg fucoxanthin with a purity of 96.24% and 92.62% were acquired, respectively. Therefore, it was demonstrated that the preparation method of fucoxanthin established in this study had an applicability to brown algae, which improved the utilization value of raw materials.  相似文献

We analyzed guava fruits (Psidium guajava L. cv. Red Suprema) from Cuba to determine their chemical composition, total antioxidant capacity, as well as their protective effect against oxidative damage using an in vitro model of human dermal fibroblasts. The guava fruit is a natural source of bioactive compounds, such as polyphenols, vitamin C, folates and beta carotenes with proven health benefits. Human dermal fibroblasts were pre-incubated with different concentrations of guava crude extract and then subjected to oxidative stress using the AAPH stressor. The number of apoptotic and dead cells, as well as the markers of oxidative damage such as lipid and protein oxidation significantly decreased when cells were pre-incubated with guava crude extract and then exposed to the stressor. The activity of antioxidant enzymes also improved when cells were pre-incubated with guava crude extract in comparison to cells subjected to stress without prior pre-incubation with the guava extract. The results obtained in this study highlight the health benefits of guava regarding oxidative stress, proving it to be an important source of bioactive compounds associated with important biological properties.  相似文献

The present study evaluated the herbicidal potential of extracts from chicory (Cichorium intybus L.) roots on the germination of Echinochloa crusgalli L. Beauv and Amaranthus retroflexus L. Eight ultrasonic assisted-extraction (UAE) conditions were optimized, using an orthogonal matrix design. The extract concentrations that would yield the largest allelopathic effects on the plant species were estimated by a modeling analysis. Our results showed that an alcohol solvent extract of chicory root had significant herbicidal activity which depended on the extract concentrations and the target species. The half-inhibitory concentrations of crude extract of chicory root ranged from 0.5 g l−1 to 40.5 g l−1. At a frequency of 40 kHz, the optimum UAE conditions to produce an extract for use as herbicide against E. crusgalli L. included an ethanol content (Ec) of 50% (v/v), a solvent-to-solid ratio (SR) of 16:1, an ultrasound temperature (UT) of 35 °C, an impregnation time (Imt) of 24 h with two rounds of impregnation (Imr), a sonication period (St) of 120 min with two rounds of sonication (Sr) and an ultrasound input power (P) of 200 W. The optimum conditions to produce an extract for use against A. retroflexus L. included an Ec of 100% (v/v), a SR of 16:1, an UT of 20 °C, an Imt of 48 h with two Imr, a St of 30 min with one Sr and a P of 400 W. The extract had the largest inhibitory effects on the germination index and root growth of both E. crusgalli L. and A. retroflexus L. at concentrations ranging from 30.8 to 33.7 g l−1. At a concentration of 4.2 g l−1, the extract significantly enhanced the shoot growth of A. retroflexus L. Overall, chicory root extract has potential for use as a main ingredient in natural herbicides or for development as a novel plant-derived herbicide.  相似文献

Phlomis lychnitis L. (Lamiaceae) is consumed as a traditional herbal tea in Spain. The antioxidant-protective effects as well as its phytoconstituents have never been established. The ability of the methanolic extract to protect cells from oxidative stress was evaluated in rat pheochromocytoma cells (PC12) using hydrogen peroxide as toxic agent. The viability of PC12 cells pre-treated with the methanolic extract of Phlomis lychnitis, determined by the MTT and LDH assays, was significantly improved at the highest dose (p?<?0.01). The antioxidant activity was confirmed evaluating the capacity of the plant to scavenge ABTS, DPPH, O2 · ? radicals and to inhibit XO. Bioassay guided fractionation led to antioxidant compounds. Qualitative HPLC/DAD/ESI/MS analysis reported phenylpropanoids, verbascoside being the major antioxidant constituent.  相似文献

Heading date determines rice’s adaptation to its area and cropping season. We analyzed the molecular evolution of the Hd6 quantitative trait locus for photoperiod sensitivity in a total of 20 cultivated varieties and wild rice species and found 74 polymorphic sites within its coding region (1,002 bp), of which five were nonsynonymous substitutions. Thus, natural mutations and modifications of the coding region of Hd6 within the genus Oryza have been suppressed during its evolution; this is supported by low Ka (≤0.003) and Ka/Ks (≤0.576) values between species, indicating purifying selection for a protein-coding gene. A nonsynonymous substitution detected in the japonica variety “Nipponbare” (a premature stop codon and nonfunctional allele) was found within only some local Japanese japonica varieties, which suggests that this point mutation happened recently, probably after the introduction of Chinese rice to Japan, and is likely involved in rice adaptation to high latitudes. Phylogenetic analysis and genome divergence using the entire Hd6 genomic region confirmed the current taxonomic sections of Oryza and supported the hypothesis of independent domestication of indica and japonica rice.  相似文献

Genetic Control of Germination Ability under Cold Stress in Rice   总被引:2,自引:0,他引:2  
An F9 recombinant inbred lines (RIL) population, derived from a cross between IR28 (Oryza sativa L. spp. indica) and Daguandao (O. sativa L. spp. japonica), was used to construct a molecular linkage map and to identify germination ability including the traits of imbibition rate, germination rate, germination index, root length, shoot length and seed vigor at 14°C for 23 d. A composite interval mapping approach was applied to conduct genetic analysis for germination ability. The frequency distributions of the germination ability traits under the cold stress in the RIL population showed continuous segregation, suggesting they were quantitative traits controlled by several genes. A total of seven QTLs were identified on chromosomes 4, 6 and 9, including two for imbibition rate (qIR-6, qIR-9), one for germination rate (qGR-4), two for germination index (qGI-4-1, qGI-4-2) and two for root length (qRL-4-1, qRL-4-2). There were no detected QTLs controlling shoot length and seed vigor. The phenotypic variance explained by a single QTL ranged from 9.1% to 37.0%, and two major QTLs, qIR-6 and qGI-4-2, accounted for over 30% of the phenotypic variance. The expressions of QTLs were developmentally regulated and growth stage-specific. Most of the QTLs observed here were located in the regions similar to the QTLs for rice cold tolerance reported previously, indicating that these QTLs were reliable. However, qRL-4-2 is not reported before.  相似文献

Background:CE is a zoonotic parasitic infection caused by Echinococcus granulosus worldwide and is associated with economic losses among livestock animals. EG95 is an immunogenic antigen from the E. granulosus. Lactococcus lactis has been prested as a safe vehicle for antigen delivery. The goal of this study was to design a novel L. lactis strain displaying EG95 as a vaccine delivery system. Methods:The eg95 encoding gene fragment fused to the M6 anchoring protein was cloned into the pNZ7021 vector, and L. lactis NZ9000 displaying recombinant EG95 was constructed. The expression of an approximately 32-kDa EG95 protein was confirmed by Western blotting and immunofluorescence analysis. The immune responses were evaluated in BALB/c mice immunized orally and subcutaneously with the live and killed recombinant L. lactis, respectively. Results:Total IgG level in mice immunized with heat-killed recombinant L. lactis (pNZ7021-eg95) significantly increased compared to the control group. sIgA was significantly higher in mice received live recombinant L. lactis (pNZ7021-eg95) compared to the control mice. Splenic lymphocytes from immunized mice represented the high levels of IFN-γ and the low-levels of IL-4 and IL-10. Conclusion:Our results indicate that immunization with EG95-expressing L. lactis can induce both specific humoral and cellular immune responses in mice.
