ABSTRACT:   Using full-cycle cultured (FC) Pacific bluefin tuna (body length [BL], 42.6–66.4 cm; body weight [BW], 1.66–7.40 kg, n  = 15), the changes in chemical compositions and histological structure of the cephalal parts of the dorsal ordinary muscles (DOM) occurring with growth were investigated. A positive correlation ( r  = 0.9644, P  < 0.05) was observed between BL and BW with growth. The protein, lipid and ash contents of DOM and condition factor did not change with growth. However, the glycogen content of DOM increased ( P  < 0.05) from approximately 55 cm (BL) in this study. Using optical microscopic photographs, the various shapes of muscle fibers were observed and it was noted that the muscle fiber diameter increased ( P  < 0.05) with growth. Using transmission electron microscopic observation, many glycogen granules were observed in muscle fibers (especially, side of connective tissue) of DOM throughout the growth stage in this study. These results indicate that the glycogen content of DOM of FC Pacific bluefin tuna increases before the lipid with growth.  相似文献

We investigated the changes in myoglobin (Mb) content and proximate compositions of the cephalad parts of the dorsal ordinary muscles (Ce‐DOM) of full‐cycle cultured (FC) Pacific bluefin tuna [body length: 42.6–115.4 cm, body weight (BW): 1.7–38.4 kg, killed from July 2004 to October 2005]. The Mb content of Ce‐DOM increased (P<0.01) until a BW of 7.0 kg was reached. However, the Mb content of Ce‐DOM had a wide variety above a BW of 7.0 kg. On the other hand, the moisture, protein and ash contents of Ce‐DOM of FC tuna decreased (P<0.05 and 0.01) with an increase in BW. However, the lipid content of Ce‐DOM increased gradually above about a BW of 20.0 kg. Furthermore, the Mb content in protein of the Ce‐DOM of FC tuna increased with an increase in BW. These results indicated that the increase in Mb content of Ce‐DOM of FC tuna was not related to changes in proximate composition between a BW of 1.7 and 38.4 kg.  相似文献

ABSTRACT:   The bluefin tuna tested were reared for 22 months from eggs before the beginning of the experiment, and sampling was performed every 3 months over the following year. The experimental results showed that the mercury concentration in the muscle ranged from 0.32 to 0.85 μg/g, which is lower than that found in wild bluefin tuna of a similar size. Increase in the mercury concentration corresponding to the increase in body weight was not shown, and it was quite different with wild bluefin tuna. Furthermore, no significant relationship was found between the lipid concentration and the mercury concentration in muscle. Among the internal organs of cultured bluefin tuna, the heart (0.32–0.66 μg/g), liver (0.43–0.99 μg/g) and spleen (0.59–1.0 μg/g) contained higher concentrations of mercury. It was estimated that the full-cycle cultured bluefin tuna had been fed small fish containing lower concentrations of mercury, and that the mercury concentration of tuna would be almost equal throughout the year because the effect of mercury accumulation would be weakened by body growth. Therefore, it was concluded that selecting diet fish species might decrease mercury contents in cultured bluefin tuna.  相似文献

ABSTRACT:   In the present study, the cDNA encoding myoglobin (Mb) of bigeye tuna Thunnus obesus was cloned and its amino acid sequence deduced in order to investigate the relationship between the primary structure and thermostability of scombridae fish Mb. An open reading frame of bigeye tuna Mb cDNA contained 444 nucleotides encoding 147 amino acids. The primary structure of bigeye tuna Mb was highly conserved when compared with those of bluefin tuna and yellowfin tuna Mb, the sequence identity being 95.2–100.0%. It also showed relatively high identity (82.3–89.1%) with the counterparts of scombridae fish. Myoglobin was then isolated from the dark muscle of four scombridae fish including bigeye tuna. Differential scanning calorimetry and circular dichroism measurements on these Mb revealed that the thermostability of bigeye tuna Mb was lowest and that of skipjack Katsuwonus pelamis Mb highest among the scombridae fish Mb examined. The α-helical contents of scombridae fish Mb at 10°C were in the range of 39.8–44.8%, clearly lower than that of horse Mb (55.3%), suggesting instability of fish Mb. The melting temperatures of these Mb fell in the range of 75.7–79.9°C, lower than that of horse Mb (84.2°C). These results strongly suggest the instability of fish Mb.  相似文献

ABSTRACT:   Using the skeletal muscle of full-cycle cultured Pacific bluefin tuna (body weight: 13.1 ± 2.6 kg, cultured for about 21 months), the proximate compositions, breaking strength and histological structure of the front and rear parts of the dorsal ordinary muscles (FD-OM and RD-OM) and the ventral ordinary muscles (FV-OM and RV-OM) were compared. The FV-OM showed low moisture, protein, ash and high fat contents ( P  < 0.05, respectively) for the other three positions. The breaking strength of FD-OM, RD-OM and RV-OM increased up to 15–18 h and decreased later. However, the breaking strength of FV-OM was maintained during chilled storage. The pH of all muscles decreased up to 15 h, and then stayed at pH 5.7–5.8. However, the pH of FV-OM stayed at a higher level (pH 5.9). The histological structure observed by optical microscopy showed a space extension among muscle cells after 24 h in all positions, and these results were also supported by image analysis.  相似文献

Using the full-cycle cultured (FC) Pacific bluefin tuna [body weight 16.3±1.9 kg (pre-fasting group, pre-FG), 14.2±0.9 kg (post-fasting group, post-FG)], changes in the physical/chemical properties of the cephalal parts of dorsal (D) and ventral (V) ordinary muscles (OM) by fasting (6 days) during chilled storage (4°C) were investigated. Condition factors were 26.7 (pre-FG) and 20.3 (post-FG, P < 0.05). Fasting changed the liver color to green. Fasting also decreased the amount of protein and lipid contents of the DOM and VOM of FC tuna. The breaking strength and pH of the DOM and VOM of post-FG tuna were higher (P <0.05) than for pre-FG tuna during storage. In contrast, the glycogen contents of DOM and VOM of post-FG tuna were lower than for pre-FG tuna. The color values (L*, a* and b*) of DOM of post-FG tuna were lower than for pre-FG tuna throughout the storage period. In addition, the metmyoglobin (metMb) content of DOM of post-FG tuna was lower (P <0.05) than that of pre-FG tuna, and the metMb content of VOM of post-FG tuna remained low after fasting. These results indicate that fasting suppresses deterioration (especially meat color) of FC tuna muscles during chilled storage.  相似文献

ABSTRACT:   To examine the effects of insulin-like growth factor (IGF)-I on the growth performance of fish, juvenile red snapper Lutjanus erythropterus with an initial body weight of approximately 12 g were fed diets containing different amounts of IGF-I containing Escherichia coli BL21 powder (BL21-tIGF-I). The growth of fish was enhanced by lower doses (0.25, 0.5 and 1%), but deteriorated at higher amounts (2.5 and 5%). The best results were obtained with 0.5% BL21-tIGF-I supplementation, which resulted in 154 and 85% weight gain and feed efficiency, respectively. In addition, muscle proteins appeared to be elevated, but muscle lipids were reduced except for in the 1% BL21-tIGF-I group. In contrast, liver lipids were reduced at 0.25, 0.5 and 1%, but increased at 2.5 and 5% BL21-tIGF-I. Furthermore, to examine the effects of BL21-tIGF-I on later-stage red snapper, we selected fish of approximately 23–24 g weight and fed them a diet either with or without 0.5% BL21-tIGF-I for 12 weeks. Results showed that the growth performance of later-stage fish was better with BL21-tIGF-I than those without supplementation after 6 weeks of feeding. The average body weight of fish that did not receive BL21-tIGF-I during the first 6 weeks, but were fed BL21-tIGF-I in weeks 7–12, was significantly higher than those fed without BL21-tIGF-I for the entire experiment. These results suggest that a continuous supply of BL21-tIGF-I may be beneficial for the growth performance of red snapper.  相似文献

ABSTRACT:   Ten newly hatched phyllosoma of Panulirus japonicus were cultured individually to monitor body length ( BL ) and intermolt period, and 2000 were cultured in groups to sample specimens for measurement of body weight. Phyllosoma were fed with Artemia and mussel gonad; the culture seawater temperature was 24–26°C. The individually cultured phyllosoma showed an increment in body length by the first molt of approximately 0.5 mm, and the molt increment increased to approximately 1 mm at 5 mm BL ; it was constant to 15 mm BL . Thereafter, the molt increment increased exponentially. The duration of the first instar was 6–7 days. Instar duration increased with development up to approximately 2 weeks at the 20th instar (∼16 mm BL ) and then became constant. Of the 10 larvae reared individually, five metamorphosed to the puerulus stage. The entire phyllosoma life ranged from 245–326 days (mean 289.0 days), and the number of instars ranged from 22–29 (mean 26.2). Body length in the final instar ranged from 28.50–33.10 mm (mean 30.280 mm). For the phyllosoma cultured in groups, relationships between BL and wet/dry body weights ( WW / DW , mg) were expressed as exponential equations: WW  = 0.0686 BL 2.2023 and DW  = 0.0209 BL 2.1905.  相似文献

A feeding trial was conducted to investigate the effects of dietary lipid level on growth and body composition of juvenile river chub, Zacco barbata (Regan). Juvenile river chub with 0.26 g body weight were given five isoenergetic and isonitrogenous purified diets containing 0–20% lipids in 5% increments for 8 weeks. The dietary lipid source was a 1:1 (w/w) mixture of menhaden fish oil and soybean oil. There was no significant difference on weight gain, feed conversion ratio, or protein efficiency ratio, among fish fed different experimental diets ( P  > 0.05). Body fat contents of both initial fish and fish fed the diet containing 20% dietary lipid were significantly greater than those fed other diets ( P  < 0.05). The percentage of total n -3 fatty acids in the body lipids of fish at the beginning of the trial was 12.34%. This value decreased to 3.83% by the end of the trial for fish fed a control diet with no supplemented lipid. Similar trends were observed in linoleic acid contents that were 6.11% and 3.09%, respectively, for initial fish and the fish fed the control diet for 8 weeks. However, the absolute amount of these fatty acids stayed the same in the initial and in the control group. Meanwhile, as dietary lipid level increased, the percentages of these fatty acids in body lipids of fish at the end of the trial also increased.  相似文献

ABSTRACT:   Two test diets with and without 5% Porphyra spheroplasts (PS) were formulated using white fishmeal as the main protein source. Red sea bream Pagrus major (mean body weight 15.4 ± 0.1 g) were maintained in a flow-through system (100 L) of thermo-controlled sea water (salinity 32–34, 25°C, 8 L/min) with ordinary aeration (400–600 mL/min) under laboratory light conditions (light–dark 12 h:12 h). Fish were fed three times a day at 10:00, 14:00 and 18:00 hours by hand for 42 days at 6% body weight on each experimental diet. Studies revealed that growth performance, survival and nutrient retention were significantly ( P  < 0.05) higher in the groups fed a diet containing spheroplasts (PS diet). Further, the fish fed the PS diet showed significantly ( P  < 0.05) lower feed conversion rates. Both groups of the PS and control diets had similar levels of body nutritional profile in terms of proximate compositions and fatty acids without compromising blood serum related parameters. From these experimental results, thus, it is comprehensible that a supplementary diet containing Porphyra spheroplasts can be used for maximizing not only growth of P. major but also for utilization of the feed ingredients.  相似文献

The present experiment was conducted to investigate the effects of dietary protein to carbohydrate ratios on growth and body composition of juvenile yellow catfish, Pelteobagrus fulvidraco . Nine diets were formulated to contain three protein levels (30%, 36% and 42%), each with three carbohydrate levels (24%, 30% and 36%). Each diet was randomly assigned to triplicate groups of 20 fish (initial mean body weight: 8.24±0.20 g) in indoor flow – through fibreglass tanks. The experiment continued for 8 weeks. Weight gain and specific growth rate were similar for the fish fed the 36% and 42% protein diets but higher than that fed the 30% protein diet. At the 36% protein level, carbohydrate contents varying from 24% to 36% ( P / E ratio of 24.0–28.2 mg protein kJ−1) had no significant effects on growth performance and feed utilization ( P >0.05). Protein efficiency ratio tended to increase with dietary carbohydrate level at the same protein level. Dietary treatments significantly influenced body composition ( P <0.05), but not the condition factor, viscerosomatic index, hepatosomatic index and intraperitoneal fat ratio ( P >0.05). Based on these observations, 36% protein and 24–36% carbohydrate with the P / E ratio of 24.0–28.2 mg protein kJ−1 seemed suitable for optimal growth and feed utilization, and carbohydrate could cause protein-sparing effect in diets for juvenile yellow catfish.  相似文献

ABSTRACT:   A 56-day feeding trial was conducted to evaluate the effects of Porphyra spheroplasts (PS) as a feed additive on growth, carcass composition and feed utilization of black sea bream. Four experimental diets with or without PS (0, 1, 3, 5% inclusion level) were formulated to determine the proper incorporation level for best growth and utilization using white fishmeal as the principal protein source. Three replicate groups of fish averaging 1.28 ± 0.05 g (mean ± SD) were maintained in flow-through thermo-controlled sea water (32–34 psu, 25°C) aquaria (100 L) under the laboratory conditions (L12 : D12) and fed three times a day at 09:00, 13:00 and 17:00 hours by hand at approximately 3% body weight on each diet. The rearing trial revealed that growth performance (in terms of weight gain, specific growth rate) are almost the same level in all the dietary groups, nevertheless the survival, nutrient utilizations and retentions (feed efficiency, protein efficiency ratio, protein retention rate, lipid retention rate) were significantly higher ( P  < 0.05) in a group fed on the diet containing 3% spheroplasts. Further, the fish fed on the diet containing PS had significantly higher ( P  < 0.05) crude lipid level in their dorsal muscles and viscera. The fish fed a PS-based diet showed higher level of eicosapentaenoic acid and docosahexaenoic acid (DHA) in viscera and DHA in dorsal muscles. The results obtained in this feeding trial suggest that the optimum dietary PS supplementation level up to 3% for black sea bream can be considered as improved consequence on growth performance, nutrient utilization or body composition were noticed.  相似文献

ABSTRACT:   Studies were conducted on growth and reproduction of the red frog crab, Ranina ranina (Linnaeus, 1758), in the Andaman Sea off Thailand from 1998 to 1999. Samples were collected from Ko Similan, Ko Surin and Thai-Myanmar boundary waters by gill net. Results showed that the relationship between carapace length (CL) and body weight (BW) for males (CL, 5.84–14.10 cm) was BW = 0.2598 CL3.0931, and for females (CL, 5.35–10.97 cm) was BW = 0.4280 CL2.8656. Growth in each gender showed allometric growth. The average CL of males was significantly larger than that of females. Average male and female crab sizes in 1999 were smaller than in 1998. The spawning season was found to be from November to February; maturing male crabs were abundant from September to December. Average sizes of CL at first maturity for males and females were 7.44 and 7.22 cm, respectively. Fecundity ranges of ovigerous female crabs were 74 600–167 900 eggs with an average egg diameter of 0.62 mm. The monthly sex ratios (male : female) varied between 1:0.56 and 1:2.77.  相似文献

ABSTRACT:   The diet shift of larval and juvenile Ryukyu-ayu Plecoglossus altivelis ryukyuensis was investigated for the landlocked population stocked in the Fukuji Reservoir, Okinawa Island. Gut contents of 274 specimens (5.1–31.4 mm in body length) including 31 yolk-sac larvae (5.7–8.0 mm) were examined. The diet of larval and juvenile landlocked P. a. ryukyuensis was mainly composed of zooplankton such as dinoflagellates, rotifers, copepods, and cladocerans. Feeding incidence was 48% in total and it gradually increased with growth. Diet analysis clearly demonstrated a diet shift during the larval and juvenile periods. For yolk-sac larvae and 5.0–19.9 mm fish, the diet was dominated by dinoflagellates Gymnodinium sp., and Peridinium sp., both numerically and by frequency, followed by rotifers that mainly consisted of Polyarthra spp. Diet breadth (Levin's standardized index BA) diversified with growth, and feeding on copepods and cladocerans gradually increased in 10.0–14.9 mm fish. Diet overlap (Morisita's index: C) showed that diet drastically changed in 20.0–24.9 mm fish with a specialization on cladocerans Bosminopsis deitersi and Diaphanosoma brachyurum . This diet shift probably corresponded with an increase of feeding ability resulting from an enlargement of the mouth, increased swimming ability, and the development of sense organs.  相似文献

ABSTRACT:   In order to develop an artificial diet, the dietary utility of enzyme-treated fish meal was investigated for juvenile Pacific bluefin tuna Thunnus orientalis (PBT). Diets containing each 63% of Chilean fish meal (FM), enzyme-treated Chilean fish meal (EC) and enzyme-treated Peruvian fish meal (EP), with 10% bonito oil and raw sand lance Ammodytes personatus (SL) were fed to juvenile tuna six times per day for one week. In a different trial, diets EC and SL were fed to tuna six times per day for 2 weeks. Only diet EC sustained similar growth or caused lower survival and higher feed efficiency, hepato- and enterosomatic indices and final carcass lipid content as compared to those of SL. Diets FM and EP led to lower specific growth rate (SGR) but similar feed efficiency, survival and hepatosomatic index, yet higher enterosomatic index. Moreover, PBT fed diet EC for 2 weeks led to similar growth performance but higher final carcass and hepatic lipid contents, and plasma cholesterol and phospholipid levels than those fed SL. Carcass fatty acid composition of diet EC group had lower 20:5  n -3 and 22:6  n -3 levels than the SL group. These results revealed that EC, as a suitable dietary protein source, could sustain growth of PBT, while dietary bonito oil led to higher carcass lipid but lower accumulation of n -3 highly unsaturated fatty acids.  相似文献

ABSTRACT:   The development of embryos, larvae and juveniles of the Ryukyu-ayu, Plecoglossus altivelisryukyuensis are described based on laboratory-reared specimens.The eggs were spherical, 0.98–1.18 mm (mean 1.06 mm)in diameter with an adherent membrane. The incubation period after fertilizationwas approximately 155 h at a water temperature of 19.7–22.0°C(mean 20.7°C). Newly hatched larvae were 5.0–5.9 mm(mean 5.5 mm) in body length (BL) with 59–62 myotomes.Within 5 days after hatching, the larvae had attained 6.4–8.2 mm(mean 7.8 mm) BL and had completely consumed their yolk.Notochord flexion began at 13.7 mm BL and was completedby 16.3 mm BL. The rudimental dorsal, anal, pelvic andadipose fins appeared at 14.7, 16.3, 21.8 and 21.8 mm BL, respectively.All fin rays reached the same fixed number of adult fish at about28 mm BL. The comb-like teeth began to form at approximately30 mm BL and were fully developed at about 40–50 mm BL.The proportions of P. a. ryukyuensis specimens,which were consistent with adult fish, occurred at approximately40 mm BL.  相似文献

ABSTRACT:   Reproductive biology of the common ponyfish Leiognathus equulus was described based on 958 specimens caught by otter trawlers in the south-western waters off Taiwan and landed at Tungkang and Linuan fishing ports from March 2000 to February 2001. Macroscopic appearance of the ovary, gonad-somatic index, oocyte diameter frequency distribution, and histological examination suggested that the spawning season of the common ponyfish in south-western Taiwan waters is from May to August. Eight stages of oocyte development were determined based on histological examination, and ovarian development can be divided as immature, early maturing, late maturing, mature and spent stages. The sex ratio of 0.52 was not significantly different from 0.5 using χ2 test, but females were overwhelmingly more when fork length (FL) was greater than 170 mm. The relations between fecundity (F), batch fecundity (BF), and body weight (BW) were estimated to be F =−5328 + 1.2946 BW ( n  = 132, P  < 0.05) and BF = −11.26 + 0.2748 BW ( n  = 25, P  < 0.05), respectively. Mean fecundity and mean batch fecundity (± standard error) were estimated to be 129 955 ± 79 343 and 28 160 ± 14 698, respectively. Sizes at 50% maturity estimated from the Logistic model were 162 mm FL and 158 mm FL for females and males, respectively.  相似文献

ABSTRACT:   The effects of silvering state of wild female Japanese eels Anguilla japonica on the success of induced maturation and the following spawning were examined. Thirty-eight females, collected in Mikawa Bay, were divided into four stages based on their silvering state: yellow (Y1), late-yellow (Y2), silver (S1) and late silver eels (S2). Despite injections of salmon pituitary extract (SPE) through the standard technique, Y1 and Y2 eels did not respond to the treatment with undeveloped gonad (gonad-somatic index [GSI]: 0.3–0.9), and all these females died by 5 weeks, probably due to an abnormal physiological condition. Most S1 (81%) and S2 eels (100%) matured completely (GSI: 17.8–51.4), and finally spawned successfully (69% for S1, 89% for S2). S2 eels fully matured with oocytes of over 750 μm in diameter by significantly smaller number of injections of SPE (5–6 times) than the case of S1 eels (6–8 times). The amount of eggs released by S2 eels (0.65 ± 0.11 g/fish per body weight [BW]) was significantly larger than those by S1 eels (0.54 ± 0.09 g/fish per BW). There was no difference in fertilization and hatching rates between eggs released by S1 eels and those of S2 eels. These results indicate that the success of induced maturation and spawning in wild female Japanese eels depends on their silvering state, and matured eggs can be obtained efficiently through the use of S2 eels rather than other stages.  相似文献

Growth performance, carcass quality, survival and haematological responses were determined when Huso huso juvenile (41.7±1.8 g) fed diets containing Ergosan (an algal product) at 0, 2.0, 4.0 and 6.0 g kg−1 for 60 days. Each diet was fed to triplicate groups of fish at 10-day intervals (1–10, 20–30 and 40–50 with non-supplemented diets and 10–20, 30–40 and 50–60 with supplemented diets). Results showed that fish fed diets containing Ergosan had significantly higher growth than the control group ( P <0.05). Survival was not different among all dietary treatments ( P >0.05). Food conversion ratio in the fish fed a diet containing 4.0 and 6.0 g kg−1 Ergosan was significantly better than the other treatments ( P <0.05), whereas protein efficiency ratio was not different between experimental diets. Lymphocyte count in the fish fed diets containing Ergosan was higher than the other treatments. Haematocrit, haemoglobin (Hb) concentration, number of erythrocytes, total leucocytes, monocyte, eosinophil, myelocyte, mean corpuscular volume, mean corpuscular haemoglobin and mean corpuscular haemoglobin concentration were not different between treatments. Neutrophil count in the control group was higher than the fish fed supplemented diets. Furthermore, whole body lipid, moisture and fibre were not different among dietary treatments ( P >0.05) but body protein in the fish fed a diet containing Ergosan at the level of 2.0 and 4.0 g kg−1 was higher than the other treatments. Whole body ash content was higher in the control group. It was concluded that dietary administration of Ergosan can influence some growth and haematological parameters in great sturgeon, H. huso juveniles.  相似文献

Triplicate groups of Mystus nemurus (Cuvier & Valenciennes) were fed isoenergetic semipurified diets containing seven dietary protein levels from 200 to 500 g kg–1 diet for 10 weeks. Dietary protein was supplied by graded amounts of a protein mixture (tuna muscle meal:casein:gelatine) at a fixed ratio of 50:37.5:12.5. Mystus nemurus fingerlings of initial weight 7.6 ± 0.2 g were fed close to apparent satiation at 2.5% of their body weight per day in two equal feedings. Growth performance and feed utilization efficiency increased linearly with dietary protein level from 202 to 410 g kg–1 diet and declined with protein levels of 471 g kg–1 diet or above. Protein efficiency ratio and apparent net protein utilization started to decline when the fish were fed with dietary protein levels exceeding 471 g kg–1 diet. Fish fed with lower protein diets (202–295 g kg–1 diet) had significantly ( P  < 0.05) higher carcass lipid content compared with fish fed with higher protein diets. Carcass lipid contents were inversely related to moisture content. Dietary protein did not significantly affect fish carcass protein and ash content. Using two-slope broken-line analysis, the dietary protein requirement for M. nemurus based on percentage weight gain was estimated to be 440 g kg–1 diet with a protein to energy ratio of 20 mg protein kJ–1 gross energy. This level of protein in the diet is recommended for maximum growth of M. nemurus fingerlings weighing between 7 and 18 g under the experimental conditions used in this study.  相似文献
