This study evaluated the supplementation effects of three herbal plants (thyme [THY], eucalyptus [EUC] and celery [CEL]), a commercial phytogenic additive ?containing essential oils (PFA‐EO, Digestarom® P.E.P.) and monensin (MON) in calf starter on performance, rumen fermentation and blood metabolites during pre‐ (days 3–55) and post‐weaning (days 56–70). Sixty‐six Holstein dairy calves (3 days of age, 41.2 ± 3 kg of BW) were allocated to one of six starters supplemented with: (i) no additives (CON), (ii) MON (30 mg/kg), (iii) THY (23 g/kg), (iv) CEL (23 g/kg), (v) EUC (23 g/kg) and (vi) PFA‐EO (3 g/kg). All the calves were offered starters ad libitum plus 6 L of whole milk daily. Starter intake tended to be the highest in calves fed PFA‐EO and THY; intermediate in calves fed CON, MON and EUC; and the lowest in those fed CEL. Average daily gain (ADG) and feed efficiency (FE) remained unaffected by dietary treatments during the pre‐weaning. During the post‐weaning period, ADG and FE ?were greatest in calves fed EUC followed by those fed CON, MON, PFA‐EO and THY, and then in those fed CEL. No differences were observed among the treatments in skeletal growth, faecal score, rumen pH or ammonia‐N concentration. Compared to calves fed CON and MON, those fed the herbal plants or PFA‐EO tended to recorded higher molar proportions of acetate and butyrate, and the acetate: propionate ratio. Blood malondialdehyde level did not differ among treatments, but calves on CON had the highest glucose concentration, and those fed PFA‐EO recorded the highest value for β‐hydroxyl butyrate on day 70. In conclusion, the results indicate that the three herbs and PFA‐EO are capable of modulating some of the rumen fermentation parameters and blood metabolites as well as eucalyptus could potentially be a better alternative to monensin for improving post‐weaning performance.

The potential effect of dietary forage supplementation on the performance and rumen development in dairy calves is well established. However, limited research has been directed to the comparative effects of forage offering methods on calf performance. The objective of the present study was to determine the effects of forage provision methods (total mixed ration or free choice) on the performance, nutrient digestibility, rumen fermentation and nutritional behaviour in newborn calves. Forty‐five Holstein dairy calves (3 days of age and 41 ± 2 kg of body weight) were assigned to the following three groups (n = 15): (i) starter without forage provision (CON), (ii) starter supplemented with 10% alfalfa hay (AH) as a total mixed ration (AH‐TMR) and (iii) starter and AH as a free‐choice provision (AH‐FC) for a period of 70 days. All the calves were offered 5 l of milk/day from day 3 to 50, and 2.5 l/day from day 50 until weaning on day 56. Dry matter intake (DMI) was greater (p < 0.01) in the AH‐TMR and AH‐FC treatments than in the CON during the pre‐ and post‐weaning periods. Calves fed the AH‐FC diet showed the highest post‐weaning DMI among the treatments. The calves receiving ad libitum forage tended (p = 0.08) to increase crude protein digestibility and overall volatile fatty acids (VFA) concentrations in the rumen. No differences were observed among the treatments at the time spent on standing, lying, eating and performing non‐nutritive oral behaviours. Compared to CON calves, animals in the AH‐TMR treatment spent more time (p < 0.05) ruminating. In conclusion, our data suggest that forage supplementation in both forage offering methods increased total DMI, ruminal pH and ruminating time in dairy calves. Hence, there is no benefit in the free‐choice provision of AH in dairy calves.

The effect of monensin on the growth and rumen metabolism of young calves (30 days old initially) was followed in 70-day experiment. Calves diet consisted of a milk substitute (4 1 per day), a concentrate mixture (13 g per 1 kg of live weight per day) and meadow hay ad libitum. Ten calves were fed 0.65 mg of monensin (SPOFA, Czechoslovakia) per 1 kg of live weight per day. Ten calves served as a control. The non-glucogenic/glucogenic ratio of VFA, mol-% acetate and butyrate were significantly lower and propionate higher in monensin-treated calves. Monensin-fed calves gained non significantly more (+ 7.2%) than control calves. A possible mode of action of monensin in young calves is discussed.

The objective of this experiment was to investigate the effects on performance, weaning age and rumen fermentation characteristics in Holstein calves when fennel powder was added to their starter diets. Thirty Holstein calves with a mean birth weight 40 kg (SD = 0.5) were allocated randomly to one of the following experimental diets: (i) control (starter diet without fennel powder), (ii) starter diet containing 0.4% of fennel powder and (iii) starter diet containing 0.8% of fennel powder (DM basis). The effect of treatments on mean dry matter intake was significant (p < 0.05) in the post‐weaning and total experimental periods. Average daily weight gain before (0.38, 0.49 and 0.47 kg/day) and after (0.6, 1.01 and 0.83 kg/day) weaning and during the entire study (0.45, 0.7 and 0.58 kg/day) was influenced by diets of 1, 2 and 3, respectively (p < 0.05). Maximum daily weight gain and the best feed conversion ratio were achieved with 0.4% fennel powder. Mean weaning age of the calves supplemented with fennel powder was lower (p < 0.05) than that of the control group. Ruminal fluid pH in calves offered starter containing 0.8% fennel powder was lower (p < 0.05) compared to calves fed the other diets. Ammonia nitrogen content increased (p < 0.05) in the third week of feeding fennel powder. The mean concentration of total short‐chain fatty acids (SCFA) and propionate molar percentage in the ruminal fluid of the calves fed with the fennel powder were higher (p < 0.05) at 6 weeks and 2 weeks after weaning than control group; however, acetate‐to‐propionate molar ratio was lower (p < 0.05). The results showed that adding 0.4% fennel powder to the starter increased the propionate molar percentage in the rumen and improved the calf performance, allowing the calves to be weaned at an earlier age.

The effects of sugar and forage inclusion in calves' starter and their interaction on animal performance and rumen fermentation parameters were investigated. Twenty‐eight neonatal Holstein male calves 3 days of age with average body weights of 42 ± 4 kg were allocated to four different treatments. All calves were fed a similar basal diet consisting of milk and concentrate. The experimental treatments were: (i) basal diet with no supplementation (Control, hereafter designated by C), (ii) basal diet plus 5% granular sugar cane (Sugar, designated by S), (iii) basal diet plus 5% forage (Forage, designated by F) and (iv) basal diet plus 5% forage with 5% granular sugar cane (F × S). Supplement ingredients were used on a dry matter (DM) basis. Rumen fluid parameters were measured twice on days 35 and 70 of the study period. The calves were weaned when they could consume 1 kg of starter for three consecutive days. The results show that starter intake was not affected by treatment; however, the lowest ADG was observed with calves in the sugar treatment. Weaning age was affected by treatments, and forage showed to reduce milk consumption period down to its shortest. Forage–sugar interaction was found to have no effects on animal performance. The structural body indices as well as the health status of the calves were similar in different treatments. Rumen pH did not differ among the treatment groups. Among the rumen parameters, total VFA concentration and molar proportions of butyrate and propionate did not exhibit any significant differences among the treatments. However, ruminal acetate concentration decreased in calves that fed sugar cane during the early weeks of the study period. Comparison of forage and sugar included in the starter diets revealed that forage reduced weaning age, while sugar cane had a negative effect on calves' performance.

An experiment was conducted to test the hypothesis that the supplementation of crude protein (CP) results in rumen acidosis and increased histamine concentrations in dairy cows. Six ruminally fistulated, non-pregnant dry cows were fed three experimental rations in a replicated 3 × 3 Latin square design. The CP contents in the low-CP, the high rumen undegradable protein (high-RUP) and the high rumen degradable protein (high-RDP) rations were 112, 259 and 266 g/kg dry matter (DM) respectively. The cows were fed 7.7 kg DM of the concentrates and 2.7 kg DM of rice straw. High levels of RDP in the ration significantly increased the ammonia, total volatile fatty acid (VFA) and histamine concentrations in the rumen fluid. However, supplemental CP, whether degradable or undegradable, did not significantly affect the pH of rumen fluid. Plasma urea nitrogen concentration was higher in both high-RDP and high-RUP in relation to cows fed the low-CP ration. The rise in ruminal histamine concentrations was physiologically non-relevant, most likely because rumen pH was not affected by supplemental CP at the installed level of DM intake. Therefore, it can be concluded that the issue of supplemental CP, rumen pH and ruminal histamine concentrations has not yet been settled. Further research is warranted to understand these relationships.

Two sheep with a ruminal fistula and an isolated small rumen were studied for the secretion of ammonia nitrogen, urea nitrogen, and amino nitrogen into the isolated rumen at different levels of volatile fatty acids (VFA) (50, 133-97, and 97-66 M Mol 1(-1)) in the rumen. The VFA level in the rumen was found to exert a great influence on the quantitative secretion of endogenous nitrogen from the blood through the rumen wall into rumen content. When the VFA level in the rumen was increased by administration of a single dose of acetic, propionic, and butyric acid, the secretion of ammonia nitrogen and amino nitrogen abruptly dropped and the secretion of urea into the isolated rumen slightly increased. The over-all amount of nitrogen (NH3-N + urea-N + amino-N) that had passed into the isolated rumen in the course of an hour showed a highly significant correlation with the passage of nitrogen in the form of ammonia and amino nitrogen and was greatest before the application of VFA to the rumen, i.e. at the level of 50 m mol 1-1. Of the metabolites under study, which were passing to the isolated rumen, amino nitrogen shared the greatest proportion (45.38-46.54%). When the VFA level in the rumen was raised, the proportion of ammonia secreted to the isolated rumen decreased and the proportion of urea in the total amount of nitrogen increased.

The contents of volatile fatty acids in the rumen fluid were studied in calves at an early (seven weeks of age) and traditional weaning term (nine weeks of age). Weaning at the age of seven weeks enabled to save 16.2 kg of milk replacer per calf; there was also a lower content of concentrates in the starter feed with a supplement of Amylastim. The health condition of the calves was good in both groups. The average daily weight gains for the period from the second to the ninth week of age were 0.550 kg in the early weaned calves and 0.690 kg in the calves weaned at a normal time. Early weaning had a positive influence on the development of rumen metabolism. Calves weaned at the age of seven weeks, compared with those weaned at the age of nine weeks, had much higher concentrations of VFA (p less than 0.05) in rumen fluid (at the age of seven weeks: 130.49 mmol per litre vs. 111.53 mmol per litre; at the age of eight weeks: 119.74 mmol per litre vs. 96.98 mmol per litre). Early weaned calves had the statistically significantly (p less than 0.05) higher proportions of propionic acid, butyric acid, i-butyric acid, and valeric acid. Later-weaned calves had the significantly higher (p less than 0.05) contents of acetic acid and i-valeric acid.

The objective of this study was to determine the effects of different forage sources on the growth performance, nutrient digestibility and blood metabolites of dairy calves. Individually housed calves (n = 40; body weight = 41.2 ± 3.5 kg) were randomly allocated (n = 10 calves per treatment: five males and five females) to one of the following four treatments: (i) starter without forage provision (CON), (ii) starter plus chopped alfalfa hay (AH), (iii) starter plus chopped wheat straw (WS) and 4) starter plus dried sugar beet pulp (BP) flakes. Calves fed AH diets had lowest (p < 0.05) starter intake than those fed other diets, and WS promoted a significant increase (p < 0.01) in starter intake during 43–80 days. Forage intake was greatest (p < 0.01) for calves fed AH than those fed WS and BP. Calves in the AH treatment consumed less (p < 0.01) total dry matter intake than those offered other forage treatments. Final body weight was greatest (p < 0.05), and age of weaning was lowest for calves fed BP than other treatments. Calves in the BP treatment had greater (p < 0.05) average daily gain (ADG) than CON and WS treatments, but similar to AH calves. Digestibility of NDF and ADF was greater (p < 0.05) in BP treatment than other treatments. Calves fed BP had greater (p < 0.05) digestibility of DM and OM than those fed CON diets and similar to those fed AH and WS diets. Calves in the AH treatment had greater (p < 0.05) CP digestibility than CON, but similar to WS and BP calves. Blood beta‐hydroxybutyrate concentration was lower in forage‐offered calves than CON one. Body measurements (with the exception of body barrel) did not differ across treatments. It was concluded that BP improves final body weight, ADG and nutrient digestibility of calves than starter without forage provision during weaning transition.

The aim of the present study was to investigate whether besides age and solid feed intake, monocarboxylic acid transporter type 1 (MCT1) expression in the rumen epithelium of calves is affected by liquid feed type [whole milk (WM) or milk replacer (MR)]. Thirty bull calves at the mean age of 5 days were randomly allocated to five experimental groups (six calves/group). Six calves were slaughtered immediately after allocation to the trial (5 days of life), eighteen calves were fed MR and slaughtered at week intervals (on 12, 19, 26 days of life respectively), and six calves were fed WM and slaughtered at the 26 days of life. MCT1 protein abundance and the MCT1 mRNA level were investigated in the dorsal and ventral sack of the rumen. Solid feed intake and short‐chain fatty acids (SCFA) concentration in the rumen fluid increased linearly with calves' age. The amount of the MCT1 protein and mRNA in the dorsal sac of rumen as well as the amount of MCT1 protein in the cranial ventral sac of rumen also increased linearly with calves' age. Calves fed WM had greater solid feed intake in the last week of the study as compared to calves fed MR, but SCFA concentration in the rumen fluid was not different. MCT1 mRNA expression in the cranial dorsal sac of rumen and protein MCT1 expression in both dorsal and ventral cranial sack of the rumen were higher in calves fed WM as compared to calves fed MR. This study confirmed age‐dependent changes of MCT1 expression in the rumen epithelium of newborn calves and showed that its expression might be affected by liquid feed type.

Two trials were conducted to determine the influence of realimentation diet energy, protein, B-vitamin (BV) and Lactobacillus acidophilus (LAC) content on recovery of rumen activity and feed consumption in beef steers. In trial 1, ruminal-fistulated steers were fasted and refed 1) prairie hay, 2) 10% protein (LCP), 3) 12.5% protein (MCP), 4) LCP + BV or 5) LCP + LAC. In trial 2, calves were fasted and refed 1) 60% cottonseed hulls-40% alfalfa dehy (high roughage), 2) LCP, 3) 15% protein (HCP), 4) LCP + BV or 5) LCP + LAC. Rumen fermentative capacity declined 74% (P less than .05) during feed and water deprivation, but returned to control levels by d 7 of realimentation. On d 3 of realimentation, steers fed the LCP and MCP diets had molar proportions of ruminal butyrate in excess of 35%. Steers fed the hay, LAC and BV diets did not have a high butyrate fermentation. In trial 2, calves lost about 15% of their body weight during feed and water deprivation. Calves fed the high roughage diet appeared to return to prefast feed and energy intakes more slowly than steers fed the medium roughage diets. Results of this study indicate that rumen fermentative capacity is a factor limiting feed intake in fasted calves for 7 to 14 after the reintroduction of feed and water.

This study aimed to evaluate the effects of supplementing Saccharomyces cerevisiae (SC) during the pre‐ and post‐weaning periods on growth, metabolic and hormonal responses, and rumen fermentation in calves. Three‐week‐old Holstein calves were assigned to either control (n = 12) or SC group (n = 12), the latter of which received 2 × 109 cfu/day of SC. The experiment was conducted over a period of 7 weeks around weaning. Daily gain (DG) in the SC group was higher (p < .05) than that in the control group. In the SC group, plasma glucose, insulin, and growth hormone (GH) concentrations were higher (p < .05) and concentrations of glucagon and insulin‐like growth factor 1 (IGF‐1) tended to be higher (p < .1) than in the control group. Proportion of rumen propionate and concentration of rumen ammonia nitrogen at 10 weeks of age were greater (p < .05) in the SC group than that in the control group. Supplementation of SC around weaning may improve dietary nutrient and energy availability and increase plasma GH and IGF‐1 concentrations. These changes observed in SC‐supplemented calves could be closely related to the improvement of DG.

The objective of this study was to investigate the effect of sundried pistachio by‐products (PBP) as a replacement of alfalfa hay (AH) on blood metabolites, rumen fermentation and populations of rumen bacteria involved in biohydrogenation (BH) in Baluchi sheep. Four adult male Baluchi sheep (41 ± 1.3 kg, BW) fitted with ruminal cannulae were randomly assigned to four experimental diets in a 4 × 4 Latin square design. The dietary treatments were as follows: (i) control, (ii) 12% PBP (0.33 of AH in basal diet replaced by PBP), (iii) 24% PBP (0.66 of AH in basal diet replaced by PBP) and (iv) 36% PBP (all of AH in basal diet replaced by PBP). The basal diet was 360 g/kg dry matter (DM) alfalfa hay, 160 g/kg DM wheat straw and 480 g/kg DM concentrate. The trial consisted of four periods, each composed of 16 days adaptation and 4 days data collection including measurement of blood metabolites, rumen fermentation and population of bacteria. No differences were observed in rumen pH among the treatments, while rumen ammonia‐N concentrations were decreased (p< 0.05) with increasing PBP by up to 36% DM of the diets. Using of 36% PBP in the diet reduced (p < 0.05) total volatile fatty acids (VFA) concentrations and the molar proportion of acetate, while the concentration of propionate, butyrate and acetate to propionate ratio were similar to all other treatments. The concentration of blood urea nitrogen (BUN) decreased (p < 0.01) with increasing PBP by up to 36% DM in the diets of sheep. However, other blood metabolites were not affected by the experimental diets. It was concluded that PBP in replacement of AH had no effects on the relative abundance of Butyrivibrio fibrisolvens and Butyrivibrio proteoclasticus in relation to the control diet.

We conducted two feeding experiments to evaluate the effects of supplementation with either cellooligosaccharide or kraft pulp on growth performance in grazing beef calves (Japanese Black) from 4 weeks pre‐weaning to 12 to 16 weeks post‐weaning. In Experiment 1 (20‐week duration), nine calves (2.9‐month‐old females) were assigned to either a control group (CON) or an experimental group (CEL) fed cellooligosaccharide at a rate of 10 g/day mixed with concentrate. Average daily weight gain tended to be greater in CEL than in CON, especially after 1 month of weaning. In Experiment 2 (16‐week duration), 10 calves (2.0‐month‐old females) were assigned to either a control group or an experimental group (KRA) fed kraft pulp at a rate of 10% replacement of total digestible nutrients with concentrate. The proportion of fibrolytic bacteria increased and that of methanogenic Archaea decreased in the rumen microbial community composition of KRA calves in Experiment 2, whereas the decrease in Fibrobacter and Archaea was observed in CEL calves at first 4 weeks in Experiment 1. We conclude that beta‐glucan prebiotic supplementation to grazing calves at pre‐weaning would affect rumen microbial composition and modified rumen fermentation characteristics, leading to a better rumen environment via different means.

This study aimed to identify the genes associated with the development of the rumen epithelium by screening for candidate genes by digital differential display (DDD) in silico. Using DDD in NCBI's UniGene database, expressed sequence tag (EST)‐based gene expression profiles were analyzed in rumen, reticulum, omasum, abomasum and other tissues in cattle. One hundred and ten candidate genes with high expression in the rumen were derived from a library of all tissues. The expression levels of 11 genes in all candidate genes were analyzed in the rumen, reticulum, omasum and abomasum of nine Japanese Black male calves (5‐week‐old pre‐weaning: n = 3; 15‐week‐old weaned calves: n = 6). Among the 11 genes, only 3‐hydroxy‐3‐methylglutaryl‐CoA synthase 2 (HMGCS2), aldo‐keto reductase family 1, member C1‐like (AKR1C1), and fatty acid binding protein 3 (FABP3) showed significant changes in the levels of gene expression in the rumen between the pre‐ and post‐weaning of calves. These results indicate that DDD analysis in silico can be useful for screening candidate genes related to rumen development, and that the changes in expression levels of three genes in the rumen may have been caused by weaning, aging or both. © 2015 Japanese Society of Animal Science
