Eighteen genotypes of Brassica napus were crossed to a cytoplasmic male sterile (CMS) line of B. napus BO 15 carrying B. tournefortii cytoplasm (‘tour’ cytoplasm). Fourteen genotypes were found to be stable maintainers of the ‘tour’ CMS. Of the remaining four genotypes, GSL-1 and ‘Asahi-natane’ were found to be heterozygous and ‘Mangun’ and ‘Yudal’ were homozygous for the restorer gene. Analysis of the F1 and F2 progenies of (CMS) BO 15 בMangun’ and (CMS) BO 15 בYudal’ showed that fertility restoration is controlled by a single dominant gene. The availability of a number of stable maintainer lines and the simple inheritance pattern of fertility restorer gene makes ‘tour’ CMS a useful system for hybrid seed production in rapeseed.  相似文献

‘Polima’ cytoplasmic male sterility (CMS) was transferred from ‘Polima’ Brassica napus ‘ISN 706’to five different cultivars of Brassica campestris (‘Pusa kalyani’, ‘Pant toria’, ‘Candle’, ‘Tobin’ and ‘ATC 94211′) by repeated backcrossing. It was observed that, while ‘Polima’ CMS manifested complete and stable male sterility in the nuclear backgrounds of ‘Pusa kalyani’, ‘Pant toria’, and ‘Tobin’, the cultivars ‘Candle’ and ‘ATC 94211’possessed the restorer gene for this CMS in the heterozygous condition. An analysis of F1 and F2 generations of ‘Polima’‘Pusa kalyani’בCandle’ and ‘Polima’‘Pusa kalyani’בATC 94211’ revealed that restoration is controlled by a single dominant gene. Identification of stable maintainers and restorers of ‘Polima’ CMS could facilitate the development of hybrid varieties in B. campestris.  相似文献

A new cytoplasmic male‐sterile (CMS) system for hybrid wheat breeding, YA‐type CMS line with the cytoplasmic mutant from the common wheat variety ‘CA8057’, was developed by the Institute of Genetics and Developmental Biology, Chinese Academy of Sciences. The pollen sterility of YA‐type CMS line was easily maintained but difficult to restore. Some sterile lines with desirable agronomic performance, such as msYA‐‘CA8057’ (BC17), msYA‐‘Yuandong 6’ (BC9), msYA‐‘Jin 411’ (BC9), msYA‐‘WL1’ (BC10), msYA‐‘Yanshi 9’ (BC10), msYA‐‘BPm16’ (BC9), msYA‐‘Jindong 8’ (BC9) and msYA‐‘Jinmai 33’ (BC9), were bred and a restorer line GR1 was screened with 26 new restorer lines being developed by transferring restorer genes from GR1. It was found that abnormal phenomena occurred at the uninucleate‐pollen stage and the abortive pollen was poor in starch content and other components. The variance analysis of agronomic traits in eight sterile lines indicated that there was no general negative effect of cytoplasm. The genetic analysis for fertility restoration showed that two pairs of independent major genes (designated YARf1YARf1YArf2YArf2) and some minor genes could be involved in the fertility restoration in restorer line GR1, and YARf1 was epistatic over YARf2 for the genetic effect of fertility restoration. As a new CMS system, the YA‐type CMS line was of potential value for hybrid wheat breeding and should be further studied.  相似文献

Total DNA from WA type CMS lines: Zhenshan 97 A, Longtepu A and theirmaintainers Zhenshan 97 B, Longtepu B wasextracted by CTAB method. One hundredprimers were used for screening RAPDmarkers to distinguish CMS line (A) andmaintainer (B) plants at seedling stage.Results showed that under the conditions of37 °C annealing temperature and1.5 mM MgCl2 concentration, in Zhenshan97 A, Longtepu A there was a 1600 bp DNAfragment in product amplified by primerOPA12, while in Zhenshan 97 B, and LongtepuB no 1600 bp fragment was found. The 1600 bpfragment was also found in DA type CMS lineXieqingzao A, but was absent in XieqingzaoB. Also in the restorer line, Minghui 63the 1600 bp fragment was absent. In F1and F2 generation of Zhenshan97A/Minghui 63, all plants investigated hadthe 1600 bp fragment. When mitochondrial DNA(mtDNA) was isolated from the three CMS (A)and their B lines and amplified by OPA12,results showed that the 1600 fragment wasfound in all the three A lines and wasabsent in the three B lines. In DA typeXieqingzao A, two other fragments (700 bp,1000 bp) were also found except the 1600 bp.These results indicate that the 1600 bpfragment derived from CMS mitochondrial DNAcan be used as a RAPD marker to distinguishA and B plants at seedling stage, and thefragments (700 bp, 1000 bp) can be used todistinguish WA and DA cytoplasms.  相似文献

Cytoplasmic male sterility (CMS) system based on the cytoplasm from Moricandia arvensis (mori) was investigated for fertility restoration and agronomic potential. Fertility restorer gene for mori CMS was introgressed from cytoplasm donor species as all the evaluated Brassica juncea genotypes (155) acted as sterility maintainers. The allosyndetic pairing between Ma and the A/B genome chromosomes in the monosomic addition plants (2n= 18II + 1Ma) facilitated the gene introgression. Partial fertility restoration (43–52% pollen grain stainability) in F1 hybrids and absence of segregation for male sterility in F2 progenies suggested gametophytic control of fertility restoration. The pollen fertility in the F1 hybrids was, however, sufficient to ensure complete seed set upon bag selfing. Introgression from M. arvensis also helped in correction of chlorosis associated with mori cytoplasm in CMS and fertile alloplasmic B. juncea plants. Yield evaluation of thirty F1 hybrids having the same nuclear genotype but varied male sterilizing cytoplasms (mori, oxy, lyr, refined ogu), in comparison to respective euplasmic hand bred control hybrids, allowed an estimate of yield penalty associated with different CMS systems. It ranged from 1.8% to 61.6%. Hybrids based on cytoplasmically refined ogu were most productive followed by those based on cytoplasmically refined mori CMS. The male sterility systems emanating from somatic hybridization were found superior than those developed from sexual hybridization.  相似文献

Summary A male fertile revertant was isolated from M1 of a cytoplasmic male sterile indica rice line II-32A, the dry seeds of which were treated with 60Co- rays at a dose of 290 Gy. The acquired revertant T24 was morphologically and agronomically similar to II-32B, the maintainer of II-32A. Testcrosses of the revertant with II-32A and Zhenshan 97A showed that the revertant was able to restore the fertility of CMS lines. Genetic analysis of the progenies between T24 and II-32A, Zhenshan 97A XieqingzaoA and DZhenshan 97A, which have different cytoplasms, showed that the fertility restoration of four CMS lines by T24 involved one nuclear gene, indicating that T24 was a result of the mutation of one nuclear gene. The mechanism of the restoration of CMS line by T24 was obviously different from other restorers such as Minghui 63 and 20964, which were shown to behave in two gene mode in fertility restoration. The discovery of the revertant T24 contributes to the understanding of CMS and fertility restoration of CMS in rice. The T24 and its parent II-32A may constitute a pair of near isogenic lines for the restoring gene, which should be valuable materials for molecular genetic analysis of CMS.  相似文献

Genetic background has a significant effect on the expression of cytoplasmic-nuclear male sterility (CMS) in pearl millet (Pennisetum glaucum (L.) R. Br.). Therefore, a reliable characterization of CMS sources requires the use of near-isonuclear lines and their hybrids. We used this approach to characterize five CMS sources (A1, A2, A3, Av, and A4). Male sterility of 81A4 was the most stable, followed by 81A1 and 81Av, indicating the relative merits of these CMS sources in breeding stable male-sterile lines. Male-sterile lines 81A2 and 81A3 were highly unstable, indicating their minimal value. Differential male fertility restoration patterns of hybrids made on 81AV and 81A4 suggest that the Av and A4 cytoplasms represent CMS systems that are different from each other and from A1, A2 and A3. An evaluation of topcross hybrids of 15 diverse populations made on 81A1 and 81A4 showed that each population had restorers and maintainers of both cytoplasms and that the frequency of maintainers of A4 was as high as, or higher than, that of the A1 cytoplasm. Thus, use of the A4 cytoplasm can substantially increase the probability of breeding stable male-sterile lines based on inbreds developed from diverse genetic backgrounds, and also provide the opportunity for breeding restorers from each of these diverse genetic sources.  相似文献

Most of the cytoplasmic-nuclear male-sterile (CMS) lines of soybean were developed only from a limited cytoplasm sources and performed not as good as required in hybrid seed production, therefore, to explore new male-sterile cytoplasm sources should be one of the effective ways to improve the pollination and hybridization for a better pod-set in utilization of heterosis of soybeans. In the present study, total 80 crosses between 70 cultivated and annual wild soybean accessions and three maintainers (N2899, N21249, and N23998) of NJCMS1A were made for detecting potential new sources with male-sterile cytoplasm. The results showed that in addition to the crosses with N8855.1 (the cytoplasm donor parent of NJCMS1A) and its derived line NG99-893 as cytoplasm parent, there appeared three crosses, including N21566 × N21249 and N23168 × N21249, with male-sterile plants in their progenies. According to the male fertility performance of backcrosses and reciprocal crosses with the tester N21249, the landrace N21566 and annual wild soybean accession N23168 were further confirmed to have male-sterile cytoplasm. Accordingly, it was understood that the source with male-sterile cytoplasm in soybean gene pool might be not occasional. The results also showed that the genetic system of male sterility of the newly found cytoplasm source N21566 was different from the old cytoplasm source N8855.1, while N23168 was to be further studied. Based on the above results, the derived male-sterile plants from [(N21566 × N21249) F1 × N21249] BC1F1 were back-crossed with the recurrent parent N21249 for five successive times, and a new CMS line and its maintainer line, designated as NJCMS3A and NJCMS3B, respectively, were obtained. NJCMS3A had normal female fertility and stable male sterility. Its microspore abortion was mainly at middle uninucleate stage, earlier than that of NJCMS1A and NJCMS2A. The male fertility of F1s between NJCMS3A and 20 pollen parents showed that 7 accessions could restore its male fertility and other 13 could maintain its male sterility. The male sterility of NJCMS3A and its restoration were controlled by one pair of gametophyte male-sterile gene according to male fertility segregation of crosses between NJCMS3A and three restorers. The nuclear gene(s) of male sterility in NJCMS3A appeared different from the previously reported CMS lines, NJCMS1A and NJCMS2A. The development of NJCMS3A demonstrated the feasibility to discover new CMS system through choosing maintainers with suitable nuclear background.  相似文献

Anther culture of two wide hybrids (Diplotaxis erucoides × Brassica campestris) × B. juncea and (D. berthautii × B. campestris) × B. juncea, their CMS lines and the parent species elicited a range of responses highlighting the importance of the genotype. Androgenesis was expressed in cultured anthers of CMS (D. erucoides) B. juncea (22.8%), in restored pollen fertile plants of this CMS line (1.66%), and in the parent, B. juncea cv Pusa Bold (13.02%). AgNO3 was essential for androgenic response in the CMS lines, and it markedly increased the frequency of androgenesis in the cultivated species. Multiple crops of microspore embryos were obtained from responsive anthers of CMS plants in anther recultures. As high as92% microspore embryos of the CMS line germinated on basal B5medium and formed normal plantlets. A germplasm collection of 152 diverse rapeseed accessions from Canada, China, France, India, Poland and South Korea was assayed for identifying new fertility restorers and sterility maintainers for a Tournefortii (tour) cytoplasmic male sterility (CMS) system in rape‐seed. Only 16 (10.5%) genotypes showed complete fertility restoration following hybridization with tour CMS line NE 409A. Notable among these were GSL 8851, GSL 8953, Mokpo # 9, Mali, Buk‐wuk‐13, Kuju‐27 and Mokpo # 84. As many as 78 (51.3%) genotypes were perfect maintainers of sterility, the remaining 58 (38.2%) genotypes were classified as partial maintainers. To study the inheritance of fertility restoration, 20 CMS (tour) rapeseed lines were crossed with the four best fertility restorers, namely GSL 8851, GSL 8953, Kuju‐27 and Mokpo # 9, to obtain F2 and test cross populations. Segregation data indicated that fertility restoration for tour CMS was governed by two genes, of which, one is stronger than the other (χ212:3:1). Differences in gene interactions were also observed (χ29:3:4) which could be explained on the basis of influence of female parent genotypes/or modified expression of the restorer gene(s) in different genetic backgrounds. Tests of allelism indicated that the restorer genes present in the four restorers evaluated were allelic.  相似文献

‘Polima’ cytoplasmic male sterility (CMS) was transferred to three different genotypes of B. napus i.e. GSL-1, ISN706 and HNS-8 by repeated backcrossing and was found to be completely stable in the nuclear background of synthetic B. napus genotype, ISN706, derived from a cross B. campestris ssp. oleifera var. brown sarson ×B. oleracea var. botrytis cv. Tusa Katki'. The BC5 and BC6 generation lines of ISN706, GSL-1 and HNS-8 with ‘Polima’ CMS were grown under a range of temperature and photo-period conditions to test their stability. No breakdown in sterility was observed in ‘Polima’ ISN706 and, therefore, this genotype can be used as a maintainer for hybrid seed production.  相似文献

Cytoplasmic male sterility (CMS) is considered an efficient genetic tool in pearl millet hybrid breeding. Of the several CMS sources available in pearl millet, A1 is the only CMS widely exploited to produce commercial hybrids in India. To explore the possibility of using alternate CMS sources, we studied the cytoplasmic effects of different CMS sources on agronomic characters in pearl millet. Five CMS (A) lines representing A1, A2, A3, A4 and A5 cytoplasms, their respective maintainer (B) lines and eight restorer (R) lines were used to generate 40 A × R and B × R experimental crosses. The experimental material was evaluated at two different locations in India. Analysis of combining ability and heterosis revealed that A4 and A5 cytoplasms had desirable effects for earliness. The A5 CMS was found to be particularly promising, as compared to other CMS sources for improving grain yield. The study also indicated that the cytoplasmic effects on general combining ability (GCA) for various agronomic characters were largely non-significant. However, cytoplasmic effects on specific combining ability and heterosis were found to be modulated by cytoplasmic-nuclear interactions and influenced by the environmental conditions. The study also demonstrated the advantage of utilizing diverse male-sterile and restorer combinations in maximizing the productivity as well as for genetic and cytoplasmic diversification of hybrids in pearl millet.  相似文献

Two new cytoplasmic male-sterile (CMS) lines have been developed in Brassica juncea using the bridgecross hybrids (Diplotaxis erucoides×Brassica campestrisB. juncea and (Diplotaxis berthautii×B. campestrisB. juncea. These were backcrossed ×ve times with pollen of B. juncea. The CMS line (D. erucoides) B. juncea segregated into tall and short true breeding types; both resembled the cultivar B. juncea in vegetative and floral morphology and in cytology, except for a greater number of secondary branches and smaller anthers with empty sterile pollen in the CMS line. Female fertility was as good in the CMS line as in the cultivar. The other CMS line (D. berthautii) B. juncea resembled the cultivar in vegetative morphology and cytology. Four true breeding floral types were isolated as follows: (1) smaller and indehiscent anthers containing empty, sterile pollen, (2) all six stamens petaloid, (3) one petaloid stamen and five stamens antherless, and (4) apetalous flowers with all six stamens antherless.  相似文献

Allelism to the highly polymorphic Mia locus was demonstrated for the powdery-mildew resistance of two Hordeum spontaneum derived winter–barley lines, ‘110-4 × Sonja’ and ‘142–29 × Dura’, by testing the F2 progeny of crosses between these lines and the winter-barley cv. ‘Triton’ (Mlal3) with two appropriate isolates. The results were confirmed by RFLP analysis, using the probe MWG 1H036, which is very closely linked to the Mia locus. The designations Mla29 and Mla32 are proposed for the genes identified in the two lines.  相似文献

Cytoplasmic male‐sterile (CMS) lines are being used to produce hybrid seeds. Thus far, four CMS sources in soybean [Glycine max (L.) Merr.] have been reported in China. However, they are not sufficient or efficient in meeting the requirements of commercial soybean hybrid seed production. In this study, 33 varieties were tested for CMS using 45 crosses among 37 landraces and 17 annual wild soybean accessions (Glycine soja Sieb. et Zucc.). The cross of N23661 × N23658 showed partial to complete male sterility in backcross generations, while the corresponding reciprocal cross showed normal male fertility. Thus, the cytoplasm of N23661 is male‐sterile, the continuously backcrossed line is a male‐sterile line (designated NJCMS4A), and N23658 is its maintainer (designated NJCM4B). The male fertility of NJCMS4A was restored by another accession, Nansheng9403. Accordingly, NJCMS4A along with its maintainer and restorer composes a complete set of three lines for producing hybrid soybean. Using mitochondrial markers and sequence analyses, NJCMS4A is a CMS line with its cytoplasm not identical to the four previously reported CMS sources in soybean.  相似文献

Search for male sterility-inducing cytoplasm in wild species of the genus Oryza was attempted with a view to diversify the base of the cytoplasmic genetic male sterility system currently used in the development of commercial rice hybrids. A total of 132 interspecific crosses were made involving accessions of four wild and two cultivated species, all belonging to the A genome. Wild accessions possessing sterility-inducing cytoplasms were identified following reciprocal and sterile F2 backcross methods. Sterile segregants were pursued through substitution backcrosses to develop cytoplasmic male-sterile (CMS) lines. CMS lines were developed with the cytoplasm of either O. rufipogon (VNI) or O. nivara (DRW 21018, DRW 21001, DRW 21039, DRW 21030 and RPW 21111). Based on shape, staining, and abortive pattern of pollen and also on type of interaction with a set of restorers and maintainers for known cytoplasmic male steriles of WA source (V 20A), the newly-developed CMS lines were grouped into four classes. Of these, RPMS 1 and RPMS 2 showed gametophytic male sterility with a restorer reaction different from WA cytoplasmic male-sterile stocks.  相似文献

Random amplified polymorphic DNAs and atp9‐related sequences were amplified in cytoplasmic male sterile (CMS) and maintainer lines drawn from a backcross programme to represent all five known cytoplasm types in chives. From these PCR amplifications, markers associated with CMS‐inducing cytoplasm types, (S1) and (S2), and for two of the three known normal cytoplasm types, (N2) and (N3), were developed. These newly developed PCR markers were used to determine the cytoplasm types in 126 plants representing 12 German chive varieties. The dependability of these PCR markers was confirmed by analysis with previously described and marker‐trait linked restriction fragment length polymorphisms. Two to five cytoplasm types were found in each of the 12 German chive varieties investigated. While the (S1) cytoplasm occurred, on average, at a frequency of 5% and the (S2) cytoplasm at 12%, the three normal cytoplasms (N1), (N2) and (N3) were present at 30, 29 and 24%, respectively. Thus, the prospects of finding maintainers for both CMS systems are relatively high in this population, if the frequency of non‐restoring alleles for the nuclear genes involved is also high enough.  相似文献

Substituting the nuclear genome of Brassica rapa into the cytoplasmic background of Enarthrocarpus lyratus through backcross substitution helped in developing cytoplasmic male sterility (CMS). Alloplasmic male sterile plants had pale green leaves, small flowers with narrow petals and rudimentary anthers. Female fertility, low initially, improved considerably with advanced backcross generations. Male sterility expression was stable throughout the growing season. Except for EC 339014, all B. rapa accessions (38) evaluated were partial maintainers of the male sterility. Introgression of gene(s) for fertility restoration from the cytoplasm donor species was facilitated by homoeologous pairing between B. rapa and E. lyratus genomes, as was apparent from the very frequent occurrence of a trivalent in the monosomic addition plants (2n = 10 II + 1 I). Backcrossing of fertile monosomic addition plants with B. rapa led to the recovery of male fertile plants possessing the stable euploid chromosome number (2n = 20). These plants restored male fertility in crosses with different (lyr) CMS B. rapa genotypes, confirming the introgression of fertility restorer gene(s) from E. lyratus, the cytoplasm donor species.  相似文献
