Managing the poultry red mite, Dermanyssus gallinae (De Geer) by conventional means (i.e., synthetic acaricides) has become increasingly problematic. As a possible alternative, research has identified several plant essential oils that are toxic to D. gallinae. However, essential oils are highly volatile and any acaricidal effect they exert could be short-lived in practice.This study investigated the short-lived toxicity of six lavender essential oils to D. gallinae. In sealed Petri-dishes, mites were exposed to filter papers impregnated with essential oil at a concentration of 0.14 mg/cm3. When filter papers were used immediately after impregnation, 66–90% D. gallinae mortality was observed after 24 h, depending upon the essential oil used. If impregnated filter papers were left in a fume cupboard for 24 h prior to use, mortality rates of D.gallinae fell to 11% or less.  相似文献

1. This study was designed to evaluate the antitrichomonal effects of P. harmala alkaloid extract against T. gallinae, both in vitro and in vivo, as well as comparing it to that of metronidazole, conventional antitrichomonal medication and harmine and harmaline, the two alkaloids present in P. harmala.

2. T. gallinae were collected by the wet mount method from infected free-living pigeons. The in vitro assay was performed using multi-well plates containing test compounds in final concentrations of 5, 10, 15, 20, 30, 50 or 100 μg/ml. The in vivo assay was done on 60 experimentally infected pigeons dosed with metronidazole at 50 mg/kg body weight (BW) or alkaloids at 25 mg/kg BW.

3. The 24 h minimum inhibitory concentration (MIC) of alkaloid extract was 15 µg/ml while that of metronidazole was 50 µg/ml. Harmine and harmaline revealed 24 h MIC of 30 and 100 µg/ml, respectively. Treatment of infected pigeons with alkaloids led to a full recovery after 3 d but with metronidazole total eradication of trophozoites was not achieved.

4. In conclusion, data of the present study suggested P. harmala is a potent natural anti-trichomonal agent, effective against T. gallinae.  相似文献

1. The responses to genetic selection on yolk proportion as a technique for increasing egg dry matter content, an important criterion for the egg-product industry, was investigated in a pedigree flock of White Leghorn hens.

2. Parents were preselected on high and low yolk proportion from a base population. The absolute estimated breeding value for yolk proportion of both groups differed by 3%. The realised selection difference in dry matter content of eggs between groups was more than 1% in the analysed offspring population.

3. Heritability estimates were moderate and dry matter had a lower heritability (h2 = 0.39) than yolk proportion (h2 = 0.44).

4. The genetic correlation between yolk proportion and dry matter content was highly positive (rg = 0.91). Genetic correlations with egg weight were negative and would have to be compensated for in a breeding programme (rg = ?0.76 with yolk proportion and r= ?0.64 with dry matter content). The genetic correlation between the laying performance and yolk proportion was r= 0.28 and close to zero (r= ?0.05) for dry matter content.

5. Easy recording and lower undesirable correlations make yolk proportion more suitable for commercial selection compared with egg dry matter content in layer breeding.  相似文献

Though important, CO2 effluxes from non-agricultural ecosystems are poorly characterised. Given that thicket vegetation is prevalent in Eastern Cape, South Africa, we monitored soil CO2 effluxes from an intact thicket, degraded thicket (DT) and grassland (G) over 10 months, as affected by temperature, moisture and penetration resistance (PR). High soil moisture (16%) reduced PR (4?kg cm?2), which raised winter effluxes (1.2 µmol m?2 s?1), while low moisture (2%) resulted in hard dry soil (14?kg cm?2) that suppressed spring effluxes (0.2 µmol m?2 s?1). There was good interaction between PR and moisture (r = ?0.53), with seasonal effluxes increasing with increasing moisture (r = 0.9, p = 0.0001) and decreasing PR (r = ?0.66, p = 0.02). Temperature effects were significant under unlimited moisture supply. Thus high summer temperature (40 °C) gave lower effluxes in DT and G (<1 µmol m?2 s?1) due to limited moisture (<10%), whereas high autumn temperature (48 °C) and good moisture (16%) accelerated CO2 emissions (averaging 2 µmol m?2 s?1) from all covers. Although semi-arid ecosystems are limited by low moisture and sandy soils, they contribute to CO2 emissions under high moisture and increasing temperature.   1. Poultry fed on wheat-based diets regularly ingest wheat germ agglutinin (WGA) that has toxic effects in vitro on intestinal epithelial cells (IEC) obtained from 14-d-old broilers. Cytotoxicity and the potential role of 14 intestinal bacterial strains in the removal of bound lectins in epithelial cell cultures were investigated.

  2. Cytotoxicity was dependent on time and lectin concentration; the lethal dose (LD50) was 8.36 µg/ml for IEC exposed for 2 h to WGA.

  3. Complementary sugars to WGA were detected on the surface of one Enterococcus and 9 Lactobacillus strains isolated from poultry. These strains were evaluated as a lectin removal tool for cytotoxicity prevention.

  4. Incubation of lactic acid bacteria with WGA before IEC–lectin interaction caused a substantial reduction in the percentage of cell deaths. The protection was attributed to the amount of lectin bound to the bacterial surfaces and was strain-dependent. L. salivarius LET 201 and L. reuteri LET 210 were more efficient than the other lactic acid bacteria assayed.

  5. These results provide a basis for the development of probiotic supplements or cell-wall preparations of selected lactic acid bacteria intended to avoid harmful effects of a natural constituent of the grain in wheat-based diets.

1. Pharmacokinetics of acetylsalicylic acid (ASA) and sodium salicylate (SS) were assessed following single intravenous (i.v.) and oral administration at doses of 50 mg/kg body weight to chickens and turkeys. Plasma drug concentrations were determined using high-performance liquid chromatography with ultraviolet detection and pharmacokinetic variables were calculated using a non-compartmental model.

2. The mean residence time (MRT) of salicylate (SA) after i.v. administration of SS was 6.08 ± 0.59 and 3.32 ± 0.27 h and after oral administration was 6.95 ± 0.72 and 4.55 ± 0.71 h in chickens and turkeys, respectively. The elimination half-life (T 1/2 e) was shorter in turkeys compared with chickens. The value of body clearance (ClB) was higher in turkeys than in chickens, but the apparent volume of distribution (V ss) was similarly low in both species. The bioavailability of SS was complete and the maximal plasma concentration of SA (C max) after oral administration was 96.93 ± 8.06 and 91.76 ± 9.64 µg/ml, respectively, in chickens and turkeys.

3. The MRT of ASA after iv administration was 0.24 ± 0.08 and 0.24 ± 0.02 h and after oral administration was 0.78 ± 0.25 and 0.59 ± 0.13 h, respectively, in chickens and turkeys. In both species, T 1/2 e was very short, ClB and V ss were similar and markedly higher than those of salicylate. The bioavailability of unchanged ASA was low and C max after oral administration was 6.9 ± 3.6 µg/ml in chickens and 8.6 ± 1.3 µg/ml in turkeys.  相似文献

  1. Sufficient floor space is a fundamental precondition for poultry to perform normal behavioural patterns. To calculate and determine stocking densities, it is essential to know the absolute minimum surface area required by any given animal (body space). Additional space is required for characteristic behaviours (behavioural space) and for adequate inter-individual distances, group sizes and room to perform social interactions have to be taken into account.

  2. To calculate body space, planimetric measurements were carried out by the colour contrast method “KobaPlan” in various poultry species in standing and sitting positions and at a number of different ages. They included laying hens (Lohmann brown (LB), Lohmann selected Leghorn (LSL)), broiler breeders (Ross, both genders), broiler chickens (Ross 308, both genders), turkeys (BUT 6, males), Peking ducks (Cherry Valley, both genders) and Muscovy ducks (Canedins R51, males).

  3. Depending on live weight, age, plumage condition and body position, LB hens occupied an average area between 401 cm2 and 542 cm2, LSL hens between 353 cm2 and 445 cm2, broiler breeder females between 440 cm2 and 537 cm2, broiler breeder males 623 cm2 up to 945 cm2, broiler chickens up to 434 cm2, male fattening turkeys up to 1808 cm2, Muscovy drakes up to 873 cm2 and Peking ducks up to 627 cm2.

  4. The values can be regarded as necessary minimum spatial requirements for the measured poultry species and genotype.

  5. The current method offers the potential to record the area occupied by animals exhibiting species-specific behavioural patterns.

An experiment was conducted to evaluate the effects of UV-B radiation on immunity and carcass characteristics in quail. Three hundred Korean quail at 3 d old were weighed and randomly allocated into 5 groups (i.e., 30 males and 30 females per group), including 1 control group and 4 experiment groups (UV-1, UV-2, UV-3, and UV-4). Quail were subjected to UV-B irradiation with different doses: 0 µW/cm2 (control), 1.4 µW/cm2 (UV-1), 2.8 µW/cm2 (UV-2), 5.6 (UV-3), and 8.4 µW/cm2 (UV-4) each day. At 38 d of age, 15 males and 15 females in each group were randomly selected to measure immune index concentrations and carcass characteristics. The mean serum IgG, IgA, and alexin C4 concentrations of UV-1 quail were significantly improved, whereas UV-3 and UV-4 quail had better live weight and carcass characteristics. Taking into account immunity and carcass characteristics together, we conclude that the optimum dose of UV radiation for Korean quail is 1.4 µW/cm2 each day. Moreover, female quail had better carcass characteristics than males in most indices of carcass characteristics.  相似文献

Abstract

Tests were conducted to determine the concentrations of copper sulfate needed to kill Flavobacterium psychrophilum, the cause of bacterial coldwater disease, either in vitro or on Rainbow Trout Oncorhynchus mykiss eggs. For the in vitro test, a plastic strip dipped in a solution of F. psychrophilum was exposed for 15 min to copper sulfate solutions of 0, 1, 5, 10, 20, 35, 50, 75, or 100 mg/L. Bacteria were “too numerous to count” at concentrations ≤10 mg/L CuSO4; significant reductions in prevalence relative to untreated controls were noted for concentrations ≥35 mg/L. However, CFUs were still observed at 50 and 75 mg/L (20% of plates with tryptone yeast extract salts media). No yellow-pigmented CFUs typical of F. psychrophilum were observed at 100 mg/L CuSO4. For the in vivo test, eggs were exposed for 15 min to 100, 300, 500, and 700 mg/L CuSO4 or 100 mg/L iodine (control). Survival to hatch was significantly lower at 500 (44.3 ± 15.2%, mean ± SD) or 700 mg/L CuSO4 (1.7 ± 0.8%) than for controls treated with 100 mg/L iodine (93.6 ± 0.9%) or at copper sulfate concentrations ≤300 mg/L. The 15-min LD50 and LD10 for copper sulfate were 461 mg/L (95% confidence interval: 457–466 mg/L) and 259 mg/L (251–266 mg/L). The prevalence of yellow CFUs at 100 mg/L CuSO4 (40.0%) was significantly higher than in untreated controls. Significant reductions in yellow CFUs were achieved using 300, 500, or 700 mg/L CuSO4 (7.5, 2.5, or 0.0% of plates with CFUs, respectively) or 100 mg/L iodine (2.5%), relative to untreated control eggs. Overall, since the concentrations of copper sulfate required to eliminate F. psychrophilum were toxic to the eggs, copper sulfate is not recommended for coldwater disease control in Rainbow Trout eggs based on conditions and parameters in this study.

Oxygen permeabilities (KO2 ) of the shell and shell membranes of fertile and infertile chicken eggs were measured at 37.5 °C and a relative humidity of 0.60 throughout 14 d incubation, with turning. The KO2 of the shell and membranes of infertile eggs was around 1.0 × 10?7 cm3 O2STP sec?1 cm?2 Torr?1 (1 Torr = 133.322 Pa) throughout incubation. With fertile eggs, from which there was a linear loss of water during incubation, the KO2 of the shell and shell membranes was about 1.0 × 10?7 cm3 O2STP sec?1 cm?2 Torr?1 for the first four days of incubation. Thereafter the majority of shells and membranes had a Ko2 of about 1.0 × 10?6 cm3 O2STP sec?1 cm?2 Torr?1.

2. A diminution of the Na+ and K+ content of the shell membranes of fertile eggs was not associated with changes in the dimensions of the glyco‐protein mantle on the cores of the individual fibres of the membranes. There was, however, a progressive deterioration in the limiting membrane of fertile but not of infertile eggs.

3. It was concluded that changes in the O2 resistance of the integument of fertile eggs were not a product of change in either of the shell membranes but of damage caused to the limiting membrane by the chorioallantois.  相似文献

The objective of the present study was to investigate the morphometrical features of the cat cerebellum using design-based stereology. Cerebellar hemispheres from four male cats were examined. Isotropic, uniform random sections were obtained and processed for light microscopy. Cerebellar total volume (V), white (WM) and grey matter (GM) volume fractions, and the volumes of the molecular and granular layers were measured using the Cavalieri's estimator and the point counting system. Cerebellar surface area was estimated using test lines, and Purkinje cellular and nuclear volumes were analysed using the nucleator technique. The volume of the cat cerebellar hemispheres was 2.06 ± 0.29 cm3. The relative volume fractions of the GM and WM were 70.6 ± 2.6% and 29.3 ± 2.6%, respectively. The surface area of the cerebellar hemisphere was 68.2 ± 17.8 cm2. The volumes of the molecular and granular layers were estimated at 0.89 ± 0.16 cm3 and 0.56 ± 0.1 cm3, respectively. The Purkinje cell volumes were found to be ranging from 1,717 to 28,489 µm3, of which cells with a perikaryon volume of 6,994 µm3 had a higher incidence. The Purkinje nuclear volume was estimated at 440–3,561 µm3, and nuclei with a volume of 1,252 µm3 were the most frequently occurring ones. Our data might contribute to the veterinary comparative neuroanatomy knowledge, help develop experimental studies in this field, and possibly lead to advancement in the diagnosis and treatment of nervous diseases in the cat.  相似文献

1. Excessive deposition of body fat, especially abdominal fat, is detrimental in chickens and the prevention of excessive fat accumulation is an important problem. The aim of this study was to identify blood biochemical indicators that could be used as criteria to select lean Yellow-feathered chicken lines.

2. Levels of blood biochemical indicators in the fed and fasted states and the abdominal fat traits were measured in 332 Guangxi Yellow chickens. In the fed state, the genetic correlations (rg) of triglycerides and very low density lipoprotein levels were positive for the abdominal fat traits (0.47 ≤ rg ≤ 0.67), whereas total cholesterol, high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C) showed higher negative correlations with abdominal fat traits (–0.59 ≤ rg ≤ ?0.33). Heritabilities of these blood biochemical parameters were high, varying from 0.26 to 0.60.

3. In the fasted state, HDL-C:LDL-C level was positively correlated with abdominal fat traits (0.35 ≤ rg ≤ 0.38), but triglycerides, total cholesterol, HDL-C, LDL-C, total protein, albumin, aspartate transaminase, uric acid and creatinine levels were negatively correlated with abdominal fat traits (–0.79 ≤ rg ≤ ?0.35). The heritabilities of these 10 blood biochemical parameters were high (0.22 ≤ h2 ≤ 0.59).

4. In the fed state, optimal multiple regression models were constructed to predict abdominal fat traits by using triglycerides and LDL-C. In the fasted state, triglycerides, total cholesterol, HDL-C, LDL-C, total protein, albumin and uric acid could be used to predict abdominal fat content.

5. It was concluded that these models in both nutritional states could be used to predict abdominal fat content in Guangxi Yellow broiler chickens.  相似文献

  1. The aim of the current study was to investigate the interaction of the nitric oxide and cannabinoidergic systems on feeding behaviour in neonatal chicken.

  2. A total of 6 experiments were designed to evaluate the interaction between cannabinoidergic and nitrergic systems on food intake in 3-h food-deprived (FD3) neonatal chickens. In Experiment 1, chickens received intracerebroventricular (ICV) injections of saline, 2-arachidonoylglycerol (2-AG) (a CB1 receptor agonist, 2 µg), l-arginine (nitric oxide precursor, 200 nmol) and co-administration of 2-AG + l-arginine. In Experiment 2, ICV injection of saline, 2-AG (2 µg), l-NAME (a nitric oxide synthesis inhibitor, 100 nmol) and their combination (2-AG + l-NAME) were applied to the birds. In Experiment 3, injections were saline, CB65 (a CB2 receptor agonist, 1.25 µg), l-arginine (200 nmol) and CB65 + l-arginine. In Experiment 4, birds received ICV injection of saline, CB65 (1.25 µg), l-NAME (100 nmol) and CB65 + l-NAME. In Experiment 5, chickens were ICV injected with saline, l-arginine (800 nmol), SR141716A (a selective CB1 receptor antagonist, 6.25 µg) and l-arginine + SR141716A. In Experiment 6, birds were injected with saline, l-arginine (800 nmol), AM630 (a selective CB2 receptor antagonist, 5 µg) and l-arginine + AM630. Cumulative food intake was recorded until 2-h post injection.

  3. ICV injection of CB1 and CB2 receptor agonists increased food intake. Co-injection of 2-AG + l-NAME increased the hyperphagic effects of CB1 receptors. CB2 receptor-induced food intake was not affected by co-administration of CB65 + l-NAME. l-Arginine decreased food intake and this effect was amplified by co-injection of l-arginine + SR141716A. However; CB2 receptor antagonists had no effect on l-arginine-induced hypophagia.

  4. The results suggest that there is an interaction between endogenous nitric oxide and the cannabinoidergic system on feeding behaviour which is mediated via CB1 receptors in the neonatal chicken.

ABSTRACT

1. The potential of lecithin and lysolecithin to improve lipid digestion and growth performance was investigated in three experiments: 1. an in vitro model that mimics the intestinal conditions of the chick, 2. a digestibility trial with chicks (5–7 days of age), and 3. a performance trial until 21 days of age.

2. In experiment 1, palm oil (PO), palm oil with lecithin (PO+L), and palm oil with lysolecithin (PO+LY) were subjected to in vitro hydrolysis and applied to Caco-2 monolayers to assess lipid absorption.

3. The in vitro hydrolysis rate of triglycerides was higher in PO+LY (k = 11.76 × 103/min) than in either PO (k = 9.73 × 103/min) or PO+L (k = 8.41 × 103/min), and the absorption of monoglycerides and free fatty acids was highest (P < 0.01) for PO+LY. In experiment 2, 90 broilers were assigned to three dietary treatments: a basal diet with 4% palm oil, and the basal diet supplemented with either 250 ppm lecithin or lysolecithin.

4. ATTD of crude fat was higher in broilers supplemented with lysolecithin, but was lower in broilers supplemented with lecithin. DM digestibility and AMEn in birds supplemented with lysolecithin were significantly higher (3.03% and 0.47 MJ/kg, respectively).

5. In experiment 3, 480 broilers were randomly allocated to four dietary treatments: basal diet with soybean oil (2%), basal diet with lecithin (2%), soybean oil diet with 250 ppm lysolecithin, or lecithin oil diet with 250 ppm lysolecithin.

6. Lecithin diets significantly reduced weight at day 10 and 21 compared with soybean oil. However, the addition of lysolecithin to lecithin-containing diets significantly improved bird performance.

7. The results of these studies showed that, in contrast to lecithin, lysolecithin was able to significantly improve the digestibility and energy values of feed in young broilers.  相似文献

1. The pharmacokinetics of difloxacin were investigated in healthy and E. coli-infected broiler chickens following intravenous and oral administration of a single dose of 10 mg/kg bodyweight.

2. After intravenous injection of difloxacin, the serum concentration–time curves were best described by a two-compartment open model. The distribution and elimination half-lives (t0.5α) and (t0.5el), respectively, were 0.10 ± 0.016 h and 3.7 ± 0.08 h in healthy chickens compared with 0.05 ± 0.005 h and 6.42 ± 0.71 h in E. coli-infected birds. The volumes of distribution Vdss were 3.14 ± 0.11 and 9.25 ± 0.43 l/kg, with total body clearance (Cltot) of 0.65 ± 0.018 and 1.14 ± 0.1 ml/kg/h, respectively.

3. Following oral administration, difloxacin was absorbed with t0.5(ab) of 0.57 ± 0.06 and 0.77 ± 0.04 h and was eliminated with t0.5(el) of 4.7 ± 0.34 and 3.42 ± 0.19, respectively, in normal and infected chickens. The peak serum concentrations were 1.34 ± 0.09 and 1.05 ± 0.06 µg/ml and attained a Tmax of 2.27 ± 0.07 and 2.43 ± 0.06 h, respectively. The systemic bioavailability of difloxacin following oral administration was 86.2% in healthy chickens and 90.6% in E. coli-infected birds. The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of difloxacin against the field strain of E. coli O78 in vitro were 0.02 µg and 0.04 µg/ml, respectively.

4. These results show that administration of a therapeutic dose of difloxacin is effective in the treatment of E. coli infection in chickens. The serum concentration of the drug was much higher than the MIC of the E. coli O78 strain in both healthy and infected chickens.  相似文献

The developmental potential of nuclei from a bovine mammary epithelial cell line (BMEC) in nuclear transfer was investigated. For nuclear transfer donors, BMEC cells (passage 15) were cultured for 4–5 days after seeding at cell densities of 1.0 × 105 cells/cm2 (high‐density group) or 0.8 × 104 cells/cm2 (low‐density group). First, the effective electric stimulation for fusion of enucleated oocytes with BMEC cells was examined. Fusion rates reached maximum with two DC pulses of 30 V/150 µm for 20 µs in the high‐density group and with two DC pulses of 25 V/150 µm for 10 µs in the low‐density group. The fusion rate (37.5%) in the high‐density group was significantly (P < 0.005) lower than in the low‐density group (71.4%). Second, the in vitro developmental potential of nuclear transfer embryos derived from BMEC cells was examined. In the high‐density and low‐density groups, 18.8% and 24.1% of fused oocytes, respectively, developed to blastocyst stage. The results of this study indicate that nuclei from BMEC cells support the development of nuclear transfer embryos to the blastocyst stage and that the efficiency of oocyte–cell fusion is affected by the culture conditions of the donor BEMC cells before nuclear transfer.  相似文献

1. The molecular weight of delta‐globulin was estimated chromato‐graphically to be about 10,700 daltons.

2. Ultracentrifuge experiments at 2.7 × 105 g gave values of s 20, W = (1.49 + 0.16C) × 10?13 s for the sedimentation coefficient and D20 w = (1.12 + 0.19c) × 10?6 cm2 s?1 for the diffusion coefficient, c being the protein concentration (g/100 ml).

3. According to the Svedberg equation, these imply a molecular weight of 12,470 daltons, assuming [v_bar] = 0.74 ml/g.

4. Using refractometer measurements of protein concentration it was found that E 1% 1cm (278 nm) = 5.57 at pH 7.8.

5. From the ultraviolet absorption spectrum of the protein in 0.1 N NaOH it was concluded that the molecule probably contains four tyrosine residues and no tryptophan.

The secretion of acetylcholinesterase (AChE) by female and male Heligmosomoides polygyrus was studied in different in vitro culture media. AChE secretion was increased in the presence of fetal calf serum or bovine serum albumin (BSA). In the absence of crowding effects, specific AChE activity in excretion/secretion products was higher for male (2.41 ± 0.07 µmol min?1 l?1 mg?1) than for female (0.56 ± 0.04 µmol min?1 mg?1) worms but on a per nematode basis both sexes showed comparable rates of secretion. Acetylthiocholine iodide was the favoured substrate of the enzyme. When the nematodes were incubated in vitro with albendazole (ABZ), ricobendazole (RBZ), mebendazole (MBZ), levamisole (LVM), morantel (MRT) or ivermectin (IVM), at concentrations from 1 mM to 10 nM, in RPMI medium for 2 or 6 h and then transferred to a drug‐free medium (RPMI medium supplemented with 0.5 % BSA) for 24 h or continuously exposed to the drugs in supplement‐free medium (24 h), the concentration‐ and time‐dependent inhibitory effects on AChE secretion were observed. The continued exposure to the drugs for all incubation periods (with a single exception for LVM 1 mM) produced the highest levels of inhibition. Under these conditions, the concentrations inhibiting the secretion of AChE by 50 % (IC50) relative to drug‐free controls were estimated. The IC50 values ranged from 0.012 µM (IVM) to 2.96 µM (MRT). The potential of this bioassay for the selective primary evaluation of new compounds with broad‐spectrum anti‐nematodal activity is discussed.  相似文献
