Baking is the most representative manufacturing process applied to bread, involving thermal and moisture conditions that facilitate the Maillard reaction (MR) and, at the same time, the destruction-formation of natural-labile and thermally-induced antioxidant compounds respectively. In the present paper, the use of a new approach to measure the Global Antioxidant Response (GAR) of cereal derivatives is proposed: a combination of in vitro digestion – which enables measurement of the bioaccessible fraction – and the QUENCHER, which makes it possible to determine the antioxidant activity of the insoluble fraction, since it is a simple and direct procedure for determining the total antioxidant capacity of solid products. After digestion, the results obtained by the antioxidant assays are up to 20-fold higher than those reported using the standard extraction methods. The non-extractable residue displayed significant antioxidant activity that accounted for up to 17% of the total antioxidant activity. Moreover, the GAR obtained in some of the assays developed was 10–40% higher than the antioxidant activity registered by the QUENCHER procedure in wheat bread, and the difference was even higher in wheat bran bread. Therefore, the use of the GAR approach could avoid underestimation of the antioxidant activity of cereal derivatives.  相似文献

Antioxidant properties of amaranth extracts isolated sequentially by acetone and methanol/water from defatted plant leaves, flowers, stems and seeds were assessed by ABTS+?, DPPH?, ORAC and total phenols content (TPC) assays. In addition, antioxidant properties of solid plant material were evaluated by the direct QUENCHER method using the same assays. Leaves and flowers of amaranth as well as their extracts possessed the highest antioxidant activities. Radical scavenging capacity in ABTS+? assay for leaves, flowers, stems and seeds evaluated by QUENCHER method were 144.24?±?2.41, 112.33?±?7.45, 19.05?±?1.13 and 21.82?±?1.06 μmol trolox equivalents in 1 g of dry weight, respectively. On-line HPLC-DPPH? assay was used to determine the activity of separated compounds and it was observed that rutin was the main radical scavenger in amaranth extracts. Preliminary screening of extract composition was performed by UPLC/ESI-QTOF-MS and rutin, nicotiflorin, isoquercitrin, 4-hydroxybenzoic and p-coumaric acids were identified by measuring their accurate mass and retention time.  相似文献

Betalains are water-soluble plant pigments that are widely used as food colorants, and have a wide range of desirable biological activities, including antioxidant, anti-inflammatory, hepatoprotective, anti-cancer properties. They can be produced from various plants, notably beetroot, but betalain products obtained in this way also have some undesirable properties and are difficult to standardize. A potentially attractive alternative is to use hairy root cultures. In the study reported here, we found that betalain extracts obtained from hairy root cultures of the red beetroot B. vulgaris cv. Detroit Dark Red also had higher antioxidant activity than extracts obtained from mature beetroots: six-fold higher 2,2-dyphenyl-1-picrylhydrazyl radical scavenging ability (90.7% inhibition, EC50 = 0.11 mg, vs 14.2% inhibition, EC50 = 0.70 mg) and 3.28-fold higher oxygen radical absorbance capacity (4,100 μM TE/g dry extract, vs 1,250 μM TE/g dry extract). The high antioxidant activity of the hairy root extracts was associated with increased concentrations (more than 20-fold) of total phenolic concomitant compounds, which may have synergistic effects with betalains. The presence of 4-hydroxybenzoic acid, caffeic acid, catechin hydrate, and epicatechin were detected in both types of extract, but at different concentrations. Rutin was only present at high concentration (1.096 mg.g−1 dry extract) in betalain extracts from the hairy root cultures, whereas chlorogenic acid was only detected at measurable concentrations in extracts from intact plants.  相似文献

An analytical study was carried out on the presence of antioxidant constituents and the in vitro antioxidant capacity in the extracts of three species of Spanish red-skinned cactus pear fruits (Opuntia ficus-indica, Opuntia undulata and Opuntia stricta). The cactus pear fruit extracts were analyzed for determined constituents: ascorbic acid, flavonoids (quercetin, isorhamnetin, myricetin, kaempferol and luteolin), betalains, taurine, total carotenoids and total phenolics. The antioxidant capacity was assessed by means of two different methods: the 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (Trolox equivalent antioxidant capacity) method and the 2,2-diphenyl-1-picrylhydrazyl radical method. Opuntia ficus-indica fruit extract had the strongest antioxidant capacity and taurine content. O. stricta fruits were the richest in ascorbic acid and total phenolics, whereas O. undulata fruits showed the highest carotenoid content. Quercetin and isorhamnetin were the main flavonoids detected. This study provides basic information on the presence of bioactive compounds and antioxidant capacity in extracts of cactus pear fruits, in order to consider these extracts as ingredient for the production of health-promoting food.  相似文献

The present study evaluates variations among flaxseed varities in terms of fatty acid composition, phytochemical profile, and antioxidant activity determined by oxygen radical absorbance capacity (ORAC), 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and ferrous ion reducing antioxidant power (FRAP) assays. Significant variations in fatty acid composition, phenolic acids and lignan were observed in flaxseed varieties from different countries. Among them, unsaturated fatty acids accounted over 4/5 of total fatty acid content. The highest ratio of linolenic acid of total fatty acid was observed in USPEA, whereas the lowest one was found in Yexiao. USPEA showed the highest content of total phenolics, as well as flaxseed lignan. In general, total phenolics appeared to be the main contributors of antioxidant capacity of flaxseed, which presented significant positive correlation. Our study revealed that both cultivar and origin of seed significantly affected fatty acid composition, phenolic acids, lignans and subsequent antioxidant activity of flaxseed. These results provided new aspects of breeding resources of flaxseed cultivars by presenting their quality specification and possible commercial value.  相似文献

Substantial evidence exists to support the hypothesis that high fruit and vegetable consumption, rich in antioxidants, can reduce the incidence of several disease states. The aim of this study was to compare the results obtained by six spectrophotometric biochemical methods including the ferric reducing antioxidant power (FRAP), 2, 2-diphenyl-1-picryhydrazyl (DPPH?), 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS??), copper (II) reducing capacity (CUPRAC) and Cerium (IV) reducing antioxidant capacity (CERAC) assays as well as Folin-Ciocalteu method (FC) for the measurement of total antioxidant capacity (TAC) and total polyphenols (TP) in different commercially available vegetable juices. This study aimed to evaluate the effect of autohydrolysis temperature of corn residues in the antioxidant activity of the phenolic compounds extracted from the liquid phase. The treatments were carried out at 160, 180, 190 and 200 °C for 30 min in a pressurized batch reactor. Two different methods for phenolic compounds extraction from the autohydrolysis liquors were investigated. For that purpose, solvent extraction with ethyl acetate and acidic precipitation were performed for phenolic compounds recovery. These methods have been compared in terms of extraction yield, physicochemical properties of obtained polyphenols (characterization by Fourier Transform Infrared Spectroscopy, Thermogravimetric Analysis and Gel Permeation Chromatography), total phenolic content, total antioxidant capacity and Trolox equivalent antioxidant capacity (TEAC) values, measured in DPPH (2,2-diphenyl-1-picrylhydrazyl) test system. The maximum phenolic contents ranged from 6.04 mg GAE/100 mg extract in acidic precipitated samples to 16.45 mg GAE/100 mg extract in ethyl acetate soluble fractions. The results indicated that the ethyl acetate fractions possessed the highest antioxidant activity, reaching after 30-60 min the same capacity reported for the reference synthetic antioxidants (Trolox).  相似文献

Since ancient times, ginger (Zingiber officinale) has been widely used for culinary and medicinal purposes. This rhizome possesses several chemical constituents; most of them present antioxidant capacity due mainly to the presence of phenolic compounds. Thus, the physical conditions for the optimal extraction of antioxidant components of ginger were investigated by applying a Box-Behnken experimental design. Extracts of ginger were prepared using water as solvent in a conventional solid–liquid extraction. The analyzed variables were time (5, 15 and 25 min), temperature (20, 55 and 90 °C) and sample concentration (2, 6 and 10 %). The antioxidant activity was measured using the 2,2-diphenyl-1-picrylhydrazyl method and a modified ferric reducing antioxidant power assay while total phenolics were measured by Folin & Ciocalteu’s method. The suggested experimental design allowed the acquisition of aqueous extracts of ginger with diverse antioxidant activity (100–555 mg Trolox/100 g, 147–1237 mg Fe2+/100 g and 50–332 mg gallic acid/100 g). Temperature was the determining factor in the extraction of components with antioxidant activity, regardless of time and sample quantity. The optimal physical conditions that allowed the highest antioxidant activity were: 90 °C, 15 min and 2 % of the sample. The correlation value between the antioxidant activity by ferric reducing antioxidant power assay and the content of total phenolics was R2?=?0.83. The experimental design applied allowed the determination of the physical conditions under which ginger aqueous extracts liberate compounds with antioxidant activity. Most of them are of the phenolic type as it was demonstrated through the correlation established between different methods used to measure antioxidant capacity.  相似文献

Total phenolics, flavonoid contents and antioxidant capacity from a wide collection of rice germplasm were measured, and their relations to grain color, grain size and 100-grain weight were investigated. Highly significant genotypic differences were observed in total phenolics, flavonoid contents and 2,2-azino-bis-(3-ehylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS) radical cation antioxidant capacity. They displayed an increasing order in the white rice, red rice and black rice, yet several white rice had higher phenolics and flavonoids contents than the red rice. Significant positive pair-wise correlations were found among the phenolics, flavonoid contents and antioxidant capacity, and the coefficient between the phenolic contents and antioxidant capacity was extremely high (r = 0.96). Among all rice accessions, the grain color parameters had negative correlations with the phenolics, flavonoid contents and antioxidant capacity (p < 0.001). The negative correlation between a* and antioxidant capacity, and the positive correlation between H° and antioxidant capacity were consistent within the respective white rice and red rice groups. Flavonoid contents had positive correlation with grain length and length to width ratio, and had negative correlation with the 100-grain weight among all rice accessions. It was also found that 100-grain weight still had negative correlations with phenolics, flavonoid contents and antioxidant capacity within the white rice genotypes. These relationships may serve as indexes to indirectly select breeding lines high in the phenolics, flavonoids and antioxidant capacity. Principal component analysis including the information for phenolics, flavonoids, antioxidant capacity, grain color parameters, grain size and 100-grain weight extracted five principal components that explained 83.7% of the total variances. The results of this study may provide new opportunities for rice breeders and eventually commercial rice growers to promote the production of rice with enhanced nutritional quality.  相似文献

本研究以槟榔为对象,采用色差、气质、液相、紫外等分析手段,研究不同干燥方法对槟榔外观形态、活性成分和抗氧化能力的影响。结果表明:槟榔的干燥速率与温度呈正相关,最快到达干燥终点的是微波干燥;干燥后槟榔外皮亮度增加,绿色度下降,黄色度提高;槟榔干制品的厚度与干燥温度呈负相关,低温的冻干可基本保持槟榔的厚度;干燥过程中槟榔的硬度出现先下降再上升的趋势,在失水率50%时硬度最低,为后续粉碎工艺提供理论支持;干燥后槟榔的总黄酮、总酚含量均显著高于新鲜青果,抗氧化能力均显著强于新鲜样品;而槟榔碱含量干燥样品均显著低于新鲜样品,因此可推断槟榔的抗氧化能力可能与其总黄酮、总酚含量相关,与槟榔碱无明显相关。不同干燥方式对槟榔的影响明显不同,在生产中应根据实际需求选择不同的干燥方式或复合使用。  相似文献

A simple and rapid procedure was developed for the direct measurement of the antioxidant capacity of cereals. It entails grinding of cereals, mixing with 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) reagent, centrifugation and measure of the absorbance. The ABTS reagent was dissolved in a mixture of ethanol:water (50:50, v/v), instead of 100% ethanol, in order to overcome low solubility of water-soluble antioxidant compounds of some cereals. A reaction time of 30 min allowed plateau values to be reached during the antioxidant capacity measurement of cereal samples. The accuracy of the direct procedure was confirmed by measuring, in solid state, the antioxidant activity of pure phenolic compounds.The direct procedure gave results of total antioxidant capacities significantly higher than those determined by the traditional procedure (multiple extraction followed by alkaline hydrolysis) for most whole meal cereals, suggesting that such a procedure was not always sufficient to properly assess the antioxidant capacity of bound phenolic compounds in cereals. The proposed extraction-independent procedure for measuring antioxidant capacity of cereals will facilitate the inter-laboratory data comparison, the construction of reliable antioxidant capacity database and the screening of large sampling of cereals for their nutraceutical characteristics.  相似文献

The aim of present study was to evaluate antioxidant capacity and cardioprotective potential of leaves infusions and partially purified fractions of Quercus sideroxyla and Q. eduardii (red oaks) and Q. resinosa (white oak). Consumption of polyphenol-rich beverages derived from plants, such as oak may represent a beneficial diet in terms of cardiovascular protection. Infusions from Oak leaves were obtained and probed for total phenolics by Folin-Ciocalteu, DPPH and hydroxyl radicals scavenging by DPPH test and Deoxy-D-ribose method, the antioxidant capacity was evaluated by FRAP and ORAC tests, inhibitions of Low Density Lipoproteins (LDL) oxidation and Angiotensin Converting Enzyme (ACE) activity were measured. A HPLC analysis was performed by HPLC-MS. Bioactive polyphenols such as gallic and ellagic acids, catechin, quercetin and derivatives: naringenin and naringin were detected in Quercus infusions. A distinctive HPLC profile was observed among the red and white oak samples. Q. resinosa infusions have exhibited the highest antioxidant activity in comparison with the other species, although in the inhibition of LDL oxidation no differences were observed. In the inhibition of the ACE, Q. resinosa was more effective (IC50, 18 ppm) than Q. sideroxyla, showing same effect as the control Captopril. From the results it is possible to postulate that not only chelating activity is important in these infusions, especially in Q. resinosa.  相似文献

Sales of multivitamins have been growing rapidly and the concept of natural multivitamin, plant-based multivitamin, or both has been introduced in the market, leading consumers to anticipate additional health benefits from phytochemicals that accompany the vitamins. However, the lack of labeling requirements might lead to fraudulent claims. Therefore, the objective of this study was to develop a strategy to verify identity of plant-based multivitamins. Phytochemical fingerprinting was used to discriminate identities. In addition, multiple bioassays were performed to determine total antioxidant capacity. A statistical computation model was then used to measure contributions of phytochemicals and vitamins to antioxidant activities. Fifteen multivitamins were purchased from the local markets in Seoul, Korea and classified into three groups according to the number of plant ingredients. Pearson correlation analysis among antioxidant capacities, amount phenols, and number of plant ingredients revealed that ferric reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picryhydrazyl (DPPH) assay results had the highest correlation with total phenol content. This suggests that FRAP and DPPH assays are useful for characterizing plant-derived multivitamins. Furthermore, net effect linear regression analysis confirmed that the contribution of phytochemicals to total antioxidant capacities was always relatively higher than that of vitamins. Taken together, the results suggest that phytochemical fingerprinting in combination with multiple bioassays could be used as a strategy to determine whether plant-derived multivitamins could provide additional health benefits beyond their nutritional value.  相似文献

为比较不同品种荔枝果皮酚类物质含量、组成及其抗氧化活性差异,用80%甲醇分别提取6个品种荔枝果皮中的酚类物质,测定其总酚和总黄酮含量。采用铁离子还原能力(FRAP)和自由基离子清除能力(ABTS)等方法评价其抗氧化活性。通过高效液相色谱(HPLC)鉴定不同的单体酚并分析其在不同品种荔枝果皮之间的差异。结果表明,不同品种荔枝果皮的酚类物质含量为28.69~68.48 mg/g,不同品种荔枝果皮的酚类物质含量和抗氧化能力存在显著的差异(P<0.05)。酚类物质的含量、FRAP和ABTS抗氧化活性能力均以‘荔枝王’为最高,分别为68.48 mg/g546.31 μmol/g和511.25 μmol/g。液相色谱结果鉴定出‘糯米糍’中的8种单体酚,而含量相对较高的是A型原花青素三聚体和原花青素A2。研究结果可为荔枝加工废弃物荔枝果皮的开发利用提供理论依据。  相似文献

目的研究大叶蟹甲草不同萃取部位的抗氧化活性。方法采用ABTS、DPPH自由基及还原性反应体系,利用分光光度法测定大叶蟹甲草的总提液、乙酸乙酯层、正丁醇层和石油醚层在不同浓度下的抗氧化能力及其总酚含量。结果大叶蟹甲草的不同萃取部位对AB'IS、DPPH自由基均具有较强的清除能力及还原能力,其结果与浓度呈良好的量效关系。其中乙酸乙酯层抗氧化能力最强,在浓度为0.62mg/mL时,对ABTS自由基的最大清除率为91.57%,对DPPH·的最大清除率为94.87%,与同浓度VC的清除率接近,且其IC50分别为0.066mg/mL、0.004mg/mL,测得乙酸乙酯层总酚含量最高,达10.45%。大叶蟹甲草具有较好的抗氧化能力,其中乙酸乙酯层抗氧化能力最显著,与总酚含量有较大的相关性,同时该结果也客观地反映了DPPH法、ABTS-+法及还原性法测定大叶蟹甲草抗氧化活性的准确性。  相似文献

Damiana (Turnera diffusa Willd) is traditionally used in the elaboration of tea, liquors, in the production of cosmetic products, and as a medicinal plant. The aim of this study was to evaluate the antioxidant content and trace element concentrations in leaves and stems from damiana grown wild and under cultivated conditions in semi-arid zones. Plants were collected at three different sites, two where damiana grows wild and one where it is cultivated for experimental purposes. Significant differences in the concentration of trace elements, vitamin C and total phenolic compound content, as well as antioxidant enzyme activities, total antioxidant capacity, and lipid peroxidation levels were found between leaves and stems, as well as among sampling sites. Environmental conditions, in combination with genetic factors, apparently affect the trace element concentrations, antioxidant content, and total antioxidant capacity in damiana grown wild. Results suggest that nutrient enrichment, via the addition of fertilizers, contributed to increased antioxidant defenses in the cultivated damiana plants. Furthermore, we suggest that genetic selection, controlled environmental conditions, and specific agrotechnical practices can increase the antioxidant yield in leaves and stems of cultivated damiana.  相似文献

Knowing the quality of mill streams is very important for getting flours with improved quality. In the present work, 49 mill streams, obtained by milling common wheat with an industrial roller mill, were characterized in terms of phenolic content, antioxidant activity and iron content. The antioxidant activity was estimated using different methods: DPPH-radical scavenging, Trolox equivalent antioxidant capacity, and iron reducing antioxidant power. The break tailings and first tailings of the reduction passages had the largest contents of iron. The total phenolic contents (TPC) from flour streams ranged between 76.3 and 369 mg ferulic acid equivalent/100 g d.w. The highest TPC were obtained for the second fractions resulting from 1st and 2nd break rolls. The highest antioxidant activity was registered for flour streams collected from: the last two break rolls, the reduction rolls that process the tailings of the first three reduction passages and bran finisher. Trolox equivalent antioxidant capacity had the same trend as DPPH-radical scavenging activity. The highest iron reducing antioxidant power was found in the tailings of the reduction rolls. The knowledge provided by this study could be successfully applied for getting flour with improved antioxidant activity and iron content by mixing different types of mill streams.  相似文献

Fruit skin color, total phenolics, total anthocyanins, soluble solids content, titratable acidity and total antioxidant capacity in fresh fruits of a number of local and well-known fig (Ficus carica L.) genotypes and cultivars grown in northeastern Turkey were determined. TEAC (Trolox equivalent antioxidant capacity) and FRAP (ferric reducing antioxidant power) assays were used to determine total antioxidant capacity. Fruit skin color of genotypes were found to be very diverse, i.e., light green, light purple, purple, dark purple and black. The content of total phenolics, soluble solids content (SSC) and titratable acidity greatly varied in the range from 24 to 237?mg of gallic acid equivalent per 100?g fresh weight, 18.60 to 26.30?% and 0.16 to 0.47?% in local genotypes and studied cultivars. In general, total antioxidant capacities determined by two methods expressed higher values in the local fig genotypes compared with the cultivars. The results suggested that genotype is the main factor that determines difference in the composition of bioactive compounds in figs and provide information on putative health benefits locally grown genotypes.  相似文献

The consumption of fruits and vegetables with high antioxidant activities leads to best healthful results. Therefore, in the present investigation we tried to find the peak of the kiwifruits antioxidant activity during the first 10 days of ethylene treatment (100 ppm at 20 °C).In order to receive the most reliable data five different antioxidant assays were used: ferric-reducing/antioxidant power (FRAP); cupric reducing antioxidant capacity (CUPRAC); trolox equivalent antioxidant capacity (TEAC); 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH); and Folin-Ciocalteau. It was found by all applied methods that kiwifruit samples have the highest contents of polyphenols and flavonoids and the highest antioxidant activity on the 6-th day of the ethylene treatment. The correlation coefficients between polyphenols, flavonoids and antioxidant activities of kiwifruit methanol extracts with TEAC and CUPRAC, were as followed: 0.81 and 0.63, and 0.23 and 0.17, respectively, and showed that the free polyphenols correlation coefficients were higher than that of the flavonoids.In conclusion: during ethylene treatment the bioactivity of kiwifruit is increasing and reaches its maximum at the 6th day and therefore it is the optimum time for kiwifruit consumption; total polyphenols were the main contributor to the overall antioxidant activity of kiwifruit; the most sensitive test for antioxidant activities determination is FRAP.  相似文献
