Small fruits like bilberry, blackberry and mulberry are rich sources of anthocyanins and other phenols, compounds with a certified antioxidant activity and spectacular effects in some chronic diseases. Romanian bilberry, blackberry and mulberry extracts were tested as anti-hyperglycemic agents on diabetic rats. Anthocyanins extraction was carried out with 80 % acidified ethanol in ultrasonically conditions at 23?±?2 °C and 40 kHz. Monomeric anthocyanins content was determined by pH differential method and varied between 1200 and 2800 mg/L. The analyses of anthocyanins were achieved using high performance liquid chromatography and mass spectrometry. Phenolics content was determined by Folin-Ciocalteu procedure and values varied between 2320 and 4250 mg/L gallic acid. Antioxidant activities of extracts were estimated by DPPH scavenging method and the values varied between 8 and 16 miliequivalents Trolox. In order to evaluate the toxicology of the extracts, the heavy metals concentration and pesticides content were analyzed. The extracts were administrated to diabetic rats in drinking water for five weeks. The administration of bilberry extract offered no satisfactory results. Treatment with blackberry extract determined a significant decrease of glucose level from 360 to about 270 mg/dL (p?<?0.05). The mulberry extract administration determined a significant decrease of glucose level from 252 mg/dL at the start day to 155 mg/dL at the final of experiment (p?<?0.05).  相似文献

The purpose of this study was to analyze the extraction efficiency of antioxidants from mango peel by comparing two techniques: microwave-assisted (MAE) and traditional solvent (TE) extraction. The number of extraction steps, water content in the extractant, peel weight-to-solvent volume ratio in extractions and extraction time all had an influence on obtaining extracts with high antioxidant capacity, but the extraction technique and the water content in the extractant were the factors with the greatest effect. Using three steps, a water content of 50 % in the ethanol:water extractant, an extraction time of 60 min and a weight-to-volume ratio of 1:10 or 1:50 (w/v) led to the highest antioxidant activity and phytochemicals content in extracts. The extraction time needed to extract phytochemicals from mango peel was similar when MAE and TE were used. The reduction of particle size can facilitate the extraction of phytochemical compounds. In this study was evaluated the effect of rice bran particles size in the yield and characteristics of γ-oryzanol compound. The γ-oryzanol extraction was realized with hexane and isopropanol solvents and quantification by spectrophotometric method. The γ-oryzanol extracts were characterized in relation of theirs majority components in HPLC-UV and the antioxidant capacity verified by the free radical DPPH consumption. The γ-oryzanol yield varied of 0.10–1.54 mg/g of bran, and the highest yield was obtained in particles smaller than 0.39 mm. The γ-oryzanol majority components presence (cycloartenyl ferulate, 2,4-methylenecycloartanyl ferulate, campesteryl ferulate e β-sitosteryl ferulate) in the extracts was confirmed and verified differences in the profile of this components in function of different particles sizes. The γ-oryzanol extract obtained from particle sizes between 0.73 and 1.67 mm demonstrated most specific inhibition of DPPH radical (6.7%) and IC50 6.63 μg/mL. When the particle size is reduced, the access surface to the extraction solvent is increased resulting in more γ-oryzanol extraction, however the extract from larger particles was more efficient as antioxidant.  相似文献

Extrusion is an alternative pretreatment for oil extraction from cereal resource. The objective of this study was to assess the feasibility of extrusion pretreatment of oat bran for oil extraction. The effects of extrusion conditions including feed moisture content, screw speed and die temperature on the extraction, quality and antioxidant capacity of oat bran oil were investigated. The results showed that the extraction yield of oat bran oil varied from 3.93% to 8.43% under different extrusion conditions. A feed moisture content of 25% had an important effect on the extraction of oil from oat bran. Extrusion pretreatment could increase oil extraction efficiency by increasing expansion index and porosity of oat bran extrudates. Extrusion processing had no positive or adverse effect on the fatty acid compositions, and acid, peroxide and iodine values. Meanwhile, extrusion processing did not affect the phytochemical components and antioxidant capacity of oat bran oil. But refining process could result in decrease of phytochemical components and antioxidant capacity of oat bran oil. These results indicated that extrusion is a feasible and potential pretreatment procedure for oil extraction from oat bran.  相似文献

不同品种桑葚叶总酚含量及其抗氧化活性比较   总被引:1,自引:0,他引:1  
为了研究不同品种桑葚叶的总酚含量差异及应用价值,以收集引种的40个品种桑葚叶为研究对象,采用超声波辅助提取桑葚叶中酚类物质,以总酚提取率为指标,利用单因素试验和正交试验考察各因素对超声辅助提取桑葚叶总酚提取率的影响;采用DPPH自由基清除能力评价不同品种桑葚叶提取物的抗氧化能力,同时对不同品种桑葚叶中总酚含量及其抗氧化能力进行相关性分析。结果表明:超声辅助提取桑葚叶总酚的最佳工艺条件为:超声温度65 ℃、超声时间30 min、固液比1∶45(g/mL)、乙醇浓度60%,4个因素对桑葚叶总酚提取率影响大小顺序为:超声温度>超声时间>固液比>乙醇浓度。不同品种桑葚叶总酚含量差异较大,其中‘条桑五号’总酚含量最高,为(26.35 ± 0.29)mg/g,‘滇桑’总酚含量最低,为(20.44 ± 0.15)mg/g;不同品种桑葚叶抗氧化活性也存在差异,且趋势与总酚含量基本一致,‘条桑五号’桑葚叶抗氧化活性最强,清除DPPH自由基能力IC50为(77.64 ± 0.34)mg/L,总抗氧化能力(FRAP)TEAC值为(2.58 ± 0.11)mmol/g;‘滇桑’桑葚叶抗氧化活性最弱,清除DPPH自由基能力IC50为(210.30 ± 0.19)mg/L,总抗氧化能力(FRAP)TEAC值为(0.73 ± 0.04)mmol/g。桑葚叶总酚含量与其提取物抗氧化能力呈正相关,选择总酚含量高的桑葚品种栽培,可提高桑葚的综合附加值。  相似文献

In this research, some herbal teas and infusions traditionally used in the treatment of diabetes in Turkey, have been studied for their antidiabetic effects on in vitro glucose diffusion and phenolic contents and antioxidant activities. Ten aqueous herbal tea extracts were examined using an in vitro method to determine their effects on glucose movement across the gastrointestinal tract. Total phenol content of herbal teas was analyzed by Folin–Ciocalteu’s procedure. Antioxidant activities of herbal teas were evaluated by the effect of extracts on DPPH radical and hydrogen peroxide scavenging. Antioxidant activity was defined as the amount of the sample to decrease the initial DPPH· concentration by 50% as efficient concentration, EC50. Antiradical activity [AE] was calculated as 1/EC50. Values were evaluated statistically. Results support the view that none of the herbal teas showed antidiabetic effect on glucose diffusion using in vitro model glucose absorption. Teas were arranged in the order of green tea > peppermint > thyme > black tea > relax tea > absinthium > shrubby blackberry > sage > roselle > olive leaves according to their total phenol contents. Among ten herbal teas, green tea had the highest hydrogen-donating capacity against to DPPH radical. Ranking of the herbal teas with respect to their DPPH radical scavenging activity were green tea > peppermint > black tea > thyme > relax tea > absinthium > roselle > olive leaves > sage > shrubby blackberry. It was determined that adding flavoring substances such as lemon, bergamot, clove and cinnamon, which are commonly used in preparation of black tea in Turkey resulted to have synergistic effect on total antioxidant activities of black and peppermint teas. The highest hydrogen peroxide inhibition value (65.50%) was obtained for green tea at a 250 μl/ml concentration. The H2O2 scavenging activity of herbal teas decreased in the order green tea > peppermint > relax tea > black tea > thyme > olive leaves > sage > absinthium > shrubby blackberry > roselle. In particular, their phenolic compounds and antioxidant activities may be useful for meal planning in type 2 diabetes. They could contribute to sustain plasma antioxidant level because antioxidants present in plants and herbs prevent the development of vascular diseases seen in type 2 diabetes.  相似文献

Potato starch has been used in the protection of antioxidants, but its use is limited due to its physicochemical properties. The objective of this work was to evaluate the effect of physical modification of potato starch by gelatinization, spray-freezing on liquid nitrogen and freezing in liquid nitrogen on the physicochemical properties and stability of powders added with blackberry juice. Frozen potato starch with liquid nitrogen exhibited higher colour retention and anthocyanins during processing, as well as adequate physicochemical and flow properties. While the spray-frozen starch on liquid nitrogen had a higher retention of anthocyanins, polyphenols and antioxidant activity during storage under controlled conditions. Atomic force microscopy revealed the modification in the microstructure of the developed powders. These results demonstrated that the microstructure of the powders was modified by conferring changes in the water adsorption capacity affecting their physicochemical properties and their stability during storage.  相似文献

The aim of this study was to investigate the free individual phenolics and the in vitro antioxidant capacity of blackberry, acerola, yellow guava, guabiju, jambolan and jabuticaba fruits in two edible stages. Of the thirty-three phenolics investigated by liquid chromatography - tandem mass spectrometry (LC-MS/MS), twenty-five were quantified and the major ones were catechin, isoquercitrin, epicatechin and gallic acid. The highest values for the total phenolic content (in dry matter) were observed for acerola (83.6 to 97.7 mg gallic acid equivalents g?1 DM) and blackberry (18.9 to 28.3 mg gallic acid equivalents g?1 DM); however, acerola, jabuticaba, and blackberry showed the highest antioxidant capacities (134.6 to 1120.4 mg Trolox equivalents g?1 for 2,2-diphenyl-1-picrylhydrazyl and 43.6 to 501.8 μmol Trolox equivalents g?1 for ferric reducing antioxidant power). For most fruits, the antioxidant capacity decreased during the ripening, possibly due to a decrease in the concentration of most of the phenolics.  相似文献

The pressurized liquid extraction (PLE) technique was used to obtain protein extracts with antioxidant capacity from salmon muscle remains, heads, viscera, skin, and tailfins. A protein recovery percentage ≈28% was obtained for all samples except for viscera, which was ≈92%. These values represented an increase of 1.5–4.8-fold compared to stirring extraction (control). Different SDS-PAGE profiles in control and PLE extracts revealed that extraction conditions affected the protein molecular weight distribution of the obtained extracts. Both TEAC (Trolox equivalent antioxidant capacity) and ORAC (oxygen radical antioxidant capacity) assays showed an outstanding antioxidant activity for viscera PLE extract. Through liquid chromatography coupled with electrospray ionization triple time-of-flight (nanoESI qQTOF) mass spectrometry, 137 and 67 peptides were identified in control and PLE extracts from salmon viscera, respectively None of these peptides was found among the antioxidant peptides inputted in the BIOPEP-UMP database. However, bioinformatics analysis showed several antioxidant small peptides encrypted in amino acid sequences of viscera extracts, especially GPP (glycine-proline-proline) and GAA (glycine-alanine-alanine) for PLE extracts. Further research on the relationship between antioxidant activity and specific peptides from salmon viscera PLE extracts is required. In addition, the salmon side streams studied presented non-toxic levels of As, Hg, Cd, and Pb, as well as the absence of mycotoxins or related metabolites. Overall, these results confirm the feasible use of farmed salmon processing side streams as alternative sources of protein and bioactive compounds for human consumption.  相似文献

White, blue, red and purple corns (Zea mays L.) were lime-cooked to obtain masa for tortillas. The total phenolics and anthocyanins content, antioxidant activity expressed as total reducing power (TRP), peroxyl radical bleaching (PRAC), total antioxidant activity (TAA) and quinone reductase (QR) induction in the murine hepatoma (Hepa 1 c1c7 cell line) as a biological marker for phase II detoxification enzymes were investigated. Among the extracts prepared from raw corn varieties the highest concentration of total phenolics, anthocyanins, antioxidant index and induction of QR-inducing activity were found in the Veracruz 42 (Ver 42) genotype. The nixtamalization process (masa) reduced total phenolics, anthocyanins and antioxidant activities and the ability for QR induction when was compared to raw grain. Processing masa into tortillas also negatively affected total phenolics, anthocyanin concentration, antioxidant activities, and QR induction in the colored corn varieties. The blue variety and its corresponding masa and tortillas did not induce QR. The bioactivity of two kiwifruit’s cultivars growing under organic and conventional conditions were studied and compared. The bioactive compounds were extracted with water and ethanol using similar conditions which are applied in pharmaceutical applications and for daily fruit consumption such as tea drink. Antioxidant radical scavenging assays [ferric-reducing/antioxidant power (FRAP); cupric reducing antioxidant capacity (CUPRAC); 2, 2-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS)], fourier transform infrared (FT-IR) and ultraviolet spectroscopy, two (2D-FL) and three-dimensional (3D-FL) fluorometry were used for the detection of biologically active metabolites derived from kiwifruits (total phenols, flavonoids, chlorophylls, carotenoids and ascorbic acid). The correlation between the total phenol content (TPC) and other bioactive compounds, and their total antioxidant capacities (TAC) was calculated for studied kiwifruit’s extracts. The interaction between drugs and human serum albumin (HSA) plays an important role in the distribution and metabolism of drugs. The properties of kiwifruit’s phenol extracts showed their ability to quench HSA, forming the complexes similar to the ones between the proteins and pure flavonoids such as quercetin. The cultivar ‘Bidan’ exhibited significantly higher TAC than the classic ‘Hayward’. In conclusion, for the first time ‘Bidan’ organic kiwifruit was analyzed and compared with widely consumed ‘Hayward’, using its bioactive and fluorescence properties. The influence of physiologically active kiwifruit’s compounds on human health, through our investigations in vitro and scientifically proven information, was explained. Relatively high content of bioactive compounds, high antioxidant and fluorescence properties of kiwifruit justify its use as a source of valuable antioxidants.  相似文献

Fishery by-products are rich in biologically active substances and the use of green and efficient extraction methods to recover these high-added-value compounds is of particular importance. In this study, head, skin and viscera of rainbow trout and sole were used as the target matrices and accelerated solvent extraction (ASE) (45–55 °C, 15 min, pH 5.2–6.8, 103.4 bars) and pulsed electric fields (PEF) (1–3 kV/cm, 123–300 kJ/kg, 15–24 h) were applied as extraction technologies. The results showed that ASE and PEF significantly increased the protein extract efficiency of the fish by-products (p < 0.05) by up to 80%. SDS-PAGE results showed that ASE and PEF treatments changed the molecular size distribution of the protein in the extracts, which was specifically expressed as the change in the area or number of bands between 5 and 250 kDa. The antioxidant capacity of the extracts was evaluated by oxygen radical absorbance capacity (ORAC) and total antioxidant capacity (ABTS) assays. The results showed that both ASE and PEF treatments significantly increased the antioxidant capacity of rainbow trout and sole skin and head extracts (p < 0.05). ASE and PEF extraction processes can be used as new technologies to extract high-added-value compounds from fish by-products.  相似文献

以紫色茶树品种‘紫嫣’和‘紫娟’为材料,以绿色品种‘福鼎大白茶’为对照,探究不同季节紫色茶树新梢花青素及主要生化成分含量的变化。结果表明:春、夏、秋季‘紫嫣’和‘紫娟’花青素总量变化均表现为先升高再降低的趋势,且夏季含量最高,分别为129.83 mg/100 g和109.73 mg/100 g,花青素各组分的季节性变化规律与花青素总量基本一致。‘紫嫣’和‘紫娟’水浸出物、氨基酸和咖啡碱含量在春季最高,茶多酚含量在夏季最高,且不同季节间差异显著。与‘福鼎大白茶’相比,‘紫嫣’和‘紫娟’中茶多酚、儿茶素和花青素含量较高。‘紫嫣’和‘紫娟’氨基酸组分中,茶氨酸占总量比例最高,分别为52.13%和59.13%;儿茶素组分中,表没食子儿茶素没食子酸酯(EGCG)含量最高,且酯型儿茶素含量显著高于简单儿茶素。综合结果显示,‘紫嫣’和‘紫娟’春梢具有适制绿茶、夏梢适制红茶的物质基础。  相似文献

Interest in new sources of anti-inflammatory and antioxidant compounds has recently become a major research issue, with the cornelian cherry (Cornus mas L.) receiving particular attention for its significant amounts of phenolic compounds and vitamins, which exhibit a wide range of biological and pharmacological properties. This study was aimed at increasing knowledge regarding the cornelian cherry in Italy through the analysis of biologically active substances in the locally available genotype “Chieri”. Spectrophotometric methods were applied to evaluate antioxidant activity, total anthocyanin content and total polyphenolic content. Identification and quantification of the main phytochemical compounds (polyphenols, monoterpenes, organic acids and vitamin C) was performed via high performance liquid chromatography coupled to a diode array detector. C. mas extracts showed high levels of total soluble solids and low acidity. High amounts of phenolic secondary metabolites were observed, with particular reference to anthocyanins (134.71 mgC3G/100 gFW), which confer remarkable nutraceutical properties to the analysed samples. These results highlight the potential of C. mas fruits as a good source of natural antioxidants, suggesting their use as a functional food. Future studies should focus on identifying other specific phytochemical compounds and the genetic traits of local varieties in order to improve cornelian cherry cultivars for food and medicine production.  相似文献

This study was aimed at characterizing the anthocyanins and phenolics profile in different varieties of pigmented corn and wheat and in some of their milling fractions. Acid/ethanol extracts were used to assess total anthocyanins, overall antioxidant activity, the overall polyphenol profile, and for evaluating the inhibition of pancreatic α-amylase and of intestinal α-glucosidase. Both enzymes were inhibited in a dose-dependent manner by all extracts, but individual extracts had specific effects on each enzyme. Anti-inflammatory response was evaluated by using acid-free extracts and Caco-2 cells transiently transfected with a luciferase reporter gene responding to cytokine stimulation. The immune response of interleukin-stimulated cells decreased significantly in a dose-dependent manner in the presence of 20–50 μM/l anthocyanins from all grains extracts, again with a different efficiency. The inhibitory ability and the anti-inflammatory capability of these extracts are in most cases higher than in similar extracts from other sources, suggesting that activities in each extract may imply specific synergies between anthocyanins and other phenolics.

Astaxanthin, a powerful antioxidant, is a good candidate for the prevention of intracellular oxidative stress. The aim of the study was to compare the antioxidant activity of astaxanthin present in two natural extracts from Haematococcus pluvialis, a microalgae strain, with that of synthetic astaxanthin. Natural extracts were obtained either by solvent or supercritical extraction methods. UV, HPLC-DAD and (HPLC-(atmospheric pressure chemical ionization (APCI)+)/ion trap-MS) characterizations of both natural extracts showed similar compositions of carotenoids, but different percentages in free astaxanthin and its ester derivatives. The Trolox equivalent antioxidant capacity (TEAC) assay showed that natural extracts containing esters displayed stronger antioxidant activities than free astaxanthin. Their antioxidant capacities to inhibit intracellular oxidative stress were then evaluated on HUVEC cells. The intracellular antioxidant activity in natural extracts was approximately 90-times higher than synthetic astaxanthin (5 µM). No modification, neither in the morphology nor in the viability, of vascular human cells was observed by in vitro biocompatibility study up to 10 µM astaxanthin concentrations. Therefore, these results revealed the therapeutic potential of the natural extracts in vascular human cell protection against oxidative stress without toxicity, which could be exploited in prevention and/or treatment of cardiovascular diseases.  相似文献

The effects of three independent variables: solvent polarity, temperature and extraction time on the antioxidant capacity, total phenolic content and phenolic acid composition in extracts obtained from sunflower shells before and after enzymatic treatment were studied. Response surface methodology based on three-level, three-variable Box-Behnken design was used for optimization of extraction parameters and evaluation of their effect on antioxidant capacity and total phenolic content in shell extracts.The average antioxidant capacities of extracts from sunflower shells without enzymatic treatment (368.1-1574.4 μmol TE/100 g) were higher than those for cellulolytic and pectolytic enzymes-treated shells (222.7-1419.0 and 270.7-1570.7 μmol TE/100 g, respectively). The content of total phenolic compounds ranged between 58.2-341.2 mg CGA/100 g, 26.7-277.3 mg CGA/100 g and 51.4-301.5 mg CGA/100 g for extracts obtained from shells without enzyme and treated with cellulolytic and pectolytic enzymes, respectively. Total phenolic content (TPC) in the studied shell extracts correlated significantly (p < 0.0001) positively with their antioxidant capacity determined by the ferric reducing antioxidant power (FRAP) method (r = 0.9275). Results of FRAP, TPC and phenolic acid composition in the studied shell extracts depend on the extraction conditions (solvent polarity, temperature, time), but they are independent on the addition of enzyme solutions. The antioxidant capacity and total phenolic content in the resulting extracts increased with a line in extraction temperature and solvent polarity.  相似文献

This study aims to explore novel extraction technologies (ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE), ultrasound–microwave-assisted extraction (UMAE), hydrothermal-assisted extraction (HAE) and high-pressure-assisted extraction (HPAE)) and extraction time post-treatment (0 and 24 h) for the recovery of phytochemicals and associated antioxidant properties from Fucus vesiculosus and Pelvetia canaliculata. When using fixed extraction conditions (solvent: 50% ethanol; extraction time: 10 min; algae/solvent ratio: 1/10) for all the novel technologies, UAE generated extracts with the highest phytochemical contents from both macroalgae. The highest yields of compounds extracted from F. vesiculosus using UAE were: total phenolic content (445.0 ± 4.6 mg gallic acid equivalents/g), total phlorotannin content (362.9 ± 3.7 mg phloroglucinol equivalents/g), total flavonoid content (286.3 ± 7.8 mg quercetin equivalents/g) and total tannin content (189.1 ± 4.4 mg catechin equivalents/g). In the case of the antioxidant activities, the highest DPPH activities were achieved by UAE and UMAE from both macroalgae, while no clear pattern was recorded in the case of FRAP activities. The highest DPPH scavenging activities (112.5 ± 0.7 mg trolox equivalents/g) and FRAP activities (284.8 ± 2.2 mg trolox equivalents/g) were achieved from F. vesiculosus. Following the extraction treatment, an additional storage post-extraction (24 h) did not improve the yields of phytochemicals or antioxidant properties of the extracts.  相似文献

This study aimed to recover bioactive compounds by solid-liquid extraction from the agro-industrial residue obtained during juçara fruits processing into pulp. A preliminary study using different solvents (methanol, ethanol and water) indicated ethanol in aqueous solution as the best solvent for antioxidants recovery. Then, a Box-Behnken design was applied considering as independent variables the solvent composition (30–70% ethanol in water), temperature (30–70 °C) and time (30–60 min), in order to evaluate the effects of these factors on antioxidant activity in juçara extract. Results showed that the extracts with higher antioxidant activity were obtained using 30% ethanol at 70 °C for 60 min; measurements included ABTS and DPPH assays, determination of total phenolic content and total monomeric anthocyanins. Furthermore, the effect of pH in antioxidants recovery was evaluated. For this purpose, the 30% ethanol solution was acidified to pH 1 and 2 with HCl. Principal component analysis showed the formation of three distinct groups: one characterized by high bioactive compounds content (pH 1.0), another with superior antioxidant activity (pH 5.75, non-acidified), and finally the group at pH 2 presenting the worst concentrations in the evaluated responses. HPLC analysis showed the presence of cyanidin-3-O-rutinoside and cyanidin-3-O-glucoside in the extracts. Therefore, the conventional solid-liquid extraction using renewable solvent can be successfully applied to recover bioactive compounds from juçara residue, which can be used by different food industries.  相似文献
