Pedigree information and 179 mtDNA sequences from two endangered Spanish horse breeds, the Asturcón pony (143) and the Mallorquí horse (36), were analysed to asses: (i) the pedigree and molecular maternal genetic diversity of the two breeds; (ii) the concordance between the dam lines recorded in the corresponding studbooks and the mtDNA haplotypes identified; and (iii) to assess the losses of maternal genetic variability occurred from the foundation of the studbooks to present. Up to 50 Asturcón and 18 Mallorquí founder dam lines were identified in the studbooks analysed. Up to 315 Asturcón mares and 51 Mallorquí mares that foaled in the last 5 years of recording formed a reference population. Only 35 Asturcón and 13 Mallorquí founder dam lines were represented in their reference populations. Sequences from a total of 38 Asturcón and 12 Mallorquí dam lines could be obtained. The 179 sequences obtained gave 15 different haplotypes, 11 and 9 of them being identified, respectively, in the Asturcón pony and in the Mallorquí horse. Five different haplotypes (roughly two-thirds of the sequences) were shared by the two horse breeds. Most dam lines analysed had a single mtDNA haplotype. However, more than one haplotype was detected within eight of the dam lines in Asturcón pony. The found inconsistencies are likely to result from deficiencies in genebank management. The maternal N(e) (mN(e)) computed using the dam line information was higher in the Asturcón pony (20.5) than in the Mallorquí horse (15.9), while these figures were on the opposite direction for the haplotypic line information (6.4 and 9.4, respectively). The ratio of the computed mN(e) values to the actual number of founder dam lines were always higher in the Mallorquí horse probably due to a more balanced distribution of individuals kept for reproduction among studs. Consequences for the conservation programmes of the analysed breeds are discussed.  相似文献

Abstract

Young horse performance test data from two warmblood riding horse populations, Norwegian warmblood (NWB) and Swedish warmblood (SWB), were analysed to examine whether including information from a related studbook would increase the accuracy of the genetic evaluations within a population. Ten conformation and performance traits from 31,588 horses, 774 NWB and 30,814 SWB were analysed separately and jointly using single trait animal models. Heritabilities were moderate to high, and varied from 0.15 (conformation, joint data) to 0.74 (jumping technique, NWB data). The genetic similarity (GS) between populations was 31%, with the SWB, as expected given the size of the populations, contributing most to the GS (98%). Genetic correlations between the same traits in the two populations were 0.43–0.90 but with large standard errors (0.2–0.3). Including information from the other population increased the average accuracy of estimated breeding values for common stallions, on average 4% for SWB and 110% for NWB.  相似文献

A data set constituting a total of 310,109 Hanoverian warmblood horses was analyzed to ascertain the genetic variability, coefficients of inbreeding, and gene contributions of foreign populations. The reference population contained all Hanoverian horses born from 1980 to 2000. In addition, Hanoverian stallions born from 1980 to 1995 and Hanoverian breeding mares from the birth years 1980 to 1995 with registered foals were analyzed for the same genetic parameters. The average complete generation equivalent was approximately 8.43 for the reference population. The mean coefficient of inbreeding was 1.33, 1.19, and 1.29% for the reference population, stallions, and breeding mares, respectively. The effective number of founders was largest in stallions (364.3) and smallest in the reference population (244.9). The ratio between the effective number of founders and the effective number of ancestors was 3.15 for the reference population, 3.25 for the stallions, and 3.06 for the breeding mares. The effective population size in the Hanoverian warmblood reference population was 372.34. English Thoroughbreds contributed nearly 35% of the genes to the Hanoverian reference population and even slightly greater contributions (39%) to the stallions. Trakehner and Arab horses contributed approximately 8 and 2.7%, respectively, to the Hanoverian gene pool. The most important male ancestors were Aldermann I from the A/E line, Fling from the F/W line, and Absatz from the Trakehner line, whereas the breeding mare Costane had the greatest contribution to the reference population, stallions, and breeding mares. From 1996 onward, the stallions Weltmeyer and Donnerhall had the largest genetic impact on the Hanoverian horse population.  相似文献

It is well known that horse breeding in Bulgaria is a cultural heritage in Bulgaria, dating from prehistoric and historic times. Until now, molecular data on Bulgarian horses from the plain regions of the country were not available. Therefore, for the first time, we have collected genetic information about some modern horse breeds from the plain regions in Bulgaria. A total of 50 horses originating from different families from two different breeds were investigated: the first one was the Pleven horse (n = 11, breeding in the Danubian Plain), and the second one was the East Bulgarian horse breed (n = 39, Shumen district). These breeds were genotyped according to the mitochondrial D-loop region. The results showed that the Pleven horse particularly carries the European haplogroup L (45.45%), followed by the Middle East haplogroup C (27.27%). In contrast to the Pleven horse, the East Bulgarian horse breed revealed almost equal frequencies of the European specific haplogroup L (33.33%) and the Central Asiatic haplogroup Q (35.90%). Analyses of these two horse breeds revealed a specific genetic profile, but it is obvious that the East Bulgarian horse showed an unusual, mixed profile—a massive admixture with the Asiatic-type haplogroup Q and a high haplogroup diversity. In conclusion, the differences in genetic structure of the two plain horse breeds may be explained with the various horse breeds involved in their formation.  相似文献

Summary We analysed a 610-bp mitochondrial (mt)DNA D-loop fragment in a sample of German draught horse breeds and compared the polymorphic sites with sequences from Arabian, Hanoverian, Exmoor, Icelandic, Sorraia and Przewalski's Horses as well as with Suffolk, Shire and Belgian horses. In a total of 65 horses, 70 polymorphic sites representing 47 haplotypes were observed. The average percentage of polymorphic sites was 11.5% for the mtDNA fragment analysed. In the nine different draught horse breeds including South German, Mecklenburg, Saxon Thuringa coldblood, Rhenisch German, Schleswig Draught Horse, Black Forest Horse, Shire, Suffolk and Belgian, 61 polymorphic sites and 24 haplotypes were found. The phylogenetic analysis failed to show monophyletic groups for the draught horses. The analysis indicated that the draught horse populations investigated consist of diverse genetic groups with respect to their maternal lineage.  相似文献

试验旨在以线粒体DNA(mitochondrial DNA,mtDNA)为切入点,研究建昌马的母系遗传多样性与系统进化。从建昌马(n=39)血液中提取基因组DNA,用PCR方法扩增mtDNA D-loop区并直接测序,分析其高变区247 bp序列信息,统计mtDNA D-loop区的单倍型及变异位点,计算单倍型多样性(haplotype diversity,Hd)、核苷酸多样性(nucleotide diversity,Pi)和平均核苷酸变异数(average number of nucleotide differences,K)。构建包括建昌马在内的19个品种马的NJ系统进化树,计算各品种间的遗传距离。结果显示,试验获得了清晰的PCR扩增产物,并通过直接测序方法获得了约1200 bp的序列。39匹建昌马mtDNA D-loop区247 bp序列(其中1个样品缺失1 bp)的AT碱基含量为61.45%,属AT碱基对富集区,检测到33个多态性位点,共显示26种单倍型,其中4种为共享单倍型,且Hap7和Hap1为优势单倍型,单倍型多样性为0.947,核苷酸多样性为0.02399,平均核苷酸变异数为5.901,显示丰富的母系遗传多样性;NJ系统进化树显示,建昌马分布在A、C、D、E、F、G共6个支系中,约50%的样品分布在A支系,显示出复杂的母系起源;建昌马与关中马的遗传距离最小(0.021),其次是三河马、文山马、韩国车巨马(0.024),与韩国济州岛马遗传距离最大(0.032)。本研究结果表明,建昌马的mtDNA D-loop高变区遗传多样性丰富,具有多个母系起源,且A支系占有明显优势,与关中马、文山马可能有共同的母系起源。  相似文献

BACKGROUND: Cervical vertebral compressive myelopathy (CVCM) is a common cause of myelopathy in horses aged 6 months to 4 years. Little information is available regarding the types of lesions, treatment, and outcomes in horses with CVCM that are > or =4 years old. ANIMALS: Twenty-two affected horses (10 with a confirmed diagnosis of CVCM and 12 presumptive cases) and 210 contemporaneous control horses. METHODS: Horses > or =4 years old that were diagnosed with CVCM between January 1992 and January 2004 were identified from medical records at Texas A&M University and the University of Florida. Data analyzed included history, signalment, neurologic examination findings, lesion location, treatment, and outcome. Signalment was also recorded in a population of contemporaneous controls. RESULTS: Horses identified had a median age of 8.4 years, and there was a greater percentage of male horses among the cases than among the controls. The most common breeds represented were warmblood (n=6) and quarter horse (n=5) types; warmbloods were significantly (P < .05) overrepresented relative to control horses. The caudal cervical vertebral column was the most common site of CVCM lesions, and the C5-C6 (4/9) and C6-C7 (3/9) articulations were most often identified as abnormal via myelography. The most common lesions seen with radiography and myelography were articular process osteophytes. Of the 22 affected horses, 8 were euthanized and a diagnosis of CVCM was confirmed by necropsy for all; 5 of 8 of these horses had spinal cord compression caused, entirely or in part, by articular process osteophytes. Medical management was the therapy chosen in all horses, and administration of corticosteroids and nonsteroidal anti-inflammatory drugs resulted in improvement in the greatest number of horses. CONCLUSIONS AND CLINICAL IMPORTANCE: CVCM should be a differential diagnosis in older horses with cervical myelopathy. Articular process osteophytes are the most frequently identified cause of spinal cord compression in this group. Male horses and horses of warmblood or Tennessee Walking Horse breeds may be predisposed to this condition.  相似文献

Criollo horse populations descend from horses brought from the Iberian Peninsula over the period of colonization (15th to 17th century). They are spread throughout the Americas and have potentially undergone genetic hybridization with other breeds in the recent past. In this study, 25 autosomal microsatellites were genotyped in 50 horse breeds representing Criollo populations from 12 American countries (27 breeds), breeds from the Iberian Peninsula (19), one breed each from France and Morocco and two cosmopolitan horse breeds (Thoroughbred and Arabian). The genetic relationships among breeds identified five clusters: Celtic; Iberian; North American with Thoroughbred influence; most Colombian breeds; and nearly all other Criollo breeds. The group of “all other Criollo breeds” had the closest genetic relationship with breeds originating from the Iberian Peninsula, specifically with the Celtic group. For the whole set of Criollo breeds analysed, the estimated genetic contribution from other breeds was approximately 50%, 30% and 20% for the Celtic, Iberian and Arab-Thoroughbred groups, respectively. The spatial distribution of genetic diversity indicates that hotspots of genetic diversity are observed in populations from Colombia, Ecuador, Brazil, Paraguay and western United States, possibly indicating points of arrival and dispersion of Criollo horses in the American continent. These results indicate that Criollo breeds share a common ancestry, but that each breed has its own identity.  相似文献

Navicular disease or podotrochlosis has long been known to cause forelimb lameness in horses. It had been proposed that the development of podotrochlosis has similarities to the human osteoarthritis (OA) complex. Alterations of the navicular bone can be made visible early in life only on the basis of radiographs. Reports on the prevalences of navicular disease indicate that radiological alterations in the navicular bone are present in different warmblood populations at frequencies of between 14.9% and 87.6%. Genetic factors play an important role in the development of the radiological signs. Estimates of heritability using animal threshold models range from h2 = 0.09 to h2 = 0.40. Estimated additive genetic correlations between radiological changes in the navicular bone and other orthopaedic health traits indicated that they mostly develop genetically independently of each other. There was a negative genetic correlation between radiological changes in the navicular bone and the number of tournament entries and placings. It has also been shown that reduction of radiological changes of navicular bones and improvement of breeding values for performance of riding horses can be achieved if selection is based on breeding values for these traits simultaneously. An optimised markerset was developed to detect quantitative trait loci (QTL) for pathologic changes in the navicular bone of Hanoverian warmblood horses. The horse genome was scanned using 214 highly polymorphic microsatellites chromosome-wide significant QTL were located on equine chromosomes (ECA) 2, 3, 4, 10, and 26. Genome-wide significant QTL were on ECA2 and on ECA10. Unravelling QTL associated with navicular disease will enhance selection progress for a healthy limb constitution in horses.  相似文献

Between 1994 and 2001, guttural pouch tympany was diagnosed in 51 foals; there were approximately three times as many fillies as colts, of Arabian, different German warmblood breeds and Western horse breeds. There were significantly more Arabian and paint horse foals than expected in comparison with the breed distribution of the foals hospitalised at the Clinic for Horses. The foals' breed and sex did not influence the age of onset, the type and severity of the clinical signs or the recurrence rate. A surgical laser technique was used on 50 of the foals; in 35 cases only one surgical treatment was necessary, in seven cases a second operation was required during the foal's initial period of hospitalisation, and in eight cases a second operation was performed during a second period of hospitalisation. Long-term follow-up information was obtained for 44 of the 50 treated horses; 24 of them were under two years of age and 20 were over two years of age. In six horses, no follow-up information was available. Four horses were euthanased for reasons unrelated to the condition or its treatment. The horses over two years of age were in training or were being used for competitions in dressage or jumping or for breeding purposes, and in only one of them was an adventitious respiratory noise reported. All the horses up to two years of age were reported to be healthy.  相似文献

Genetic variation at 7 blood-group and 10 biochemical genetic loci was examined in 30 horses from a feral herd from the Namib Desert of Namibia, Africa. The observed genetic variability was extremely low compared with that found in domestic horse breeds. The low variation was most probably a result of recent small population size and a small founding population size. Genetic comparison of the Namib horses, which were of unknown origins, to domestic horse breeds, showed that the Namib horses had the highest genetic similarity to Arabian type horses, although they did not closely resemble this type of horse in conformation.  相似文献

The liberalization of European animal breeding legislation and an increasing diversity of equestrian sports have led to a constant rise in the number of horse breeds and breed registries. In addition to the trend towards more and smaller breed registries, there is another trend towards an international expansion of the bigger established sport horse breeds. Regional breeds, at least in smaller countries, may no longer be able to run an independent breeding programme. The typical horse breeder, in the future, will be a female and qualified in equestrian sports. Artificial insemination (AI) mainly with fresh or cooled-transported semen has become a major breeding tool, allowing breeders all over Europe to benefit from the best stallions of most breeds. New AI techniques such as low-dose insemination may remain restricted to individual stallions and also the interest of breeding programmes in sex determination of foals via semen sorting is limited. Embryo transfer and associated techniques, although allowed by most breeds, have not contributed significantly to genetic progress in European sport horses so far. A potential use of cloning may be to produce gonad-intact copies from geldings that have performed to a superior level. With a more open and international structure of horse breeding and increased use of AI, equine reproduction and biotechnology should be emphasized by veterinary curricula and continuing professional education programmes.  相似文献

Researchers and practitioners of assisted reproduction in horses have borrowed heavily from similar bovine practices over the years. However, most procedures required species-specific experiments for optimization. Furthermore, the flow of information occurs in both directions; some procedures, for example, vitrification of embryos and intracytoplasmic sperm injection, were made efficacious in horses before cattle. The great advantage of research in cattle is the availability of large numbers of animals and embryos for testing hypotheses economically. Objectives of cattle breeding generally are more scientific than those concerning horses, and it is likely that much can be learned in the future from genetic tools being developed for cattle breeding. A genetic advantage of cattle breeding over horse breeding is the extremely large genetic and phenotypic databases of some breeds, which can be matched to genomic information.  相似文献

The genetic variability within the Uruguayan Creole horse and its relationship to a group of geographically or historically related breeds (Spanish Pure-bred, Barb, Quarter horse, Paso Fino, Peruvian Paso, Arabian and Thoroughbred horse), was evaluated using 25 loci (seven of blood groups, nine of protein polymorphisms and nine microsatellites) analyzed on a total of 145 Uruguayan Creole horses. In this study, blood group and protein polymorphism variants that are considered to be breed markers of Spanish Pure-bred and Barb horses were detected in the Creole breed. Conversely, some microsatellites and protein polymorphisms alleles were found uniquely in the Creole horse. American horse breeds together with Barb and Arabian horses clearly formed a separate cluster from the Spanish pure-bred and Thoroughbred breeds, as shown by an UPGMA dendrogram based on Nei's standard genetic distance. Data in this study provided evidence for considerable genetic variation within Uruguayan Creole horses and of a distinctive breed profile. Both traits were most likely inherited from the XVIth century Spanish horses, more closely related to Barb than to Spanish Pure-bred.  相似文献

Sport performance in dressage and show jumping are two important traits in the breeding goals of many studbooks. To determine the optimum selection scheme for jumping and dressage, knowledge is needed on the genetic correlation between both disciplines and between traits measured early in life and performance in competition in each discipline. This study aimed to estimate genetic parameters to support decision‐making on specialization of breeding horses for dressage and show jumping in Dutch warmblood horses. Genetic correlations between performance of horses in dressage and show jumping were estimated as well as the genetic correlation between traits recorded during studbook‐entry inspections and performance in dressage and show jumping competitions. The information on competition comprised the performance of 82 694 horses in dressage and 62 072 horses in show jumping, recorded in the period 1993–2012. For 26 056 horses, information was available for both disciplines. The information on traits recorded at studbook‐entry inspections comprised 62 628 horses, recorded in the period 1992–2013. Genetic parameters were estimated from the whole dataset and from a subset without horses recorded in both disciplines. Additionally, the genetic parameters were estimated in three different time periods defined by horses' birth year. The genetic correlation between dressage and show jumping in the whole dataset was ?0.23, and it was ?0.03 when it was estimated from horses recorded in only one discipline. The genetic correlation between dressage and show jumping was more negative in the most recent time period in all the cases. The more negative correlation between disciplines in more recent time periods was not reflected in changes in the correlations between competitions traits and the traits recorded in the studbook‐first inspection. These results suggest that a breeding programme under specialization might be most effective defining two separate aggregate breeding goals for each of the disciplines.  相似文献

Genetic correlations for body measurements and subjectively scored traits between foals and studbook horses were estimated using bivariate linear mixed models. Observations for nine foal and eleven studbook traits in Finnhorses on 6529 foals and 6596 studbook horses and in Standardbred trotters on 3069 foals and 2112 studbook horses were available from the Finnish horse breeding shows. The number of sires with progeny in both foal and studbook data was 203 in Finnhorse and 145 in Standardbred trotters. Estimates of heritability for body measurements in foals and studbook horses using univariate models were high in both breeds (0.41-0.84). Heritability estimates for subjectively scored traits using univariate models were generally higher for foals (0.08-0.46) than for studbook horses (0.06-0.21) in both breeds. Genetic correlations between foals and studbook horses for body measurements were highly positive ranging from 0.74 to 0.96 in Finnhorses and from 0.79 to 0.99 in Standardbred trotters. Low to highly positive genetic correlations between foals and studbook horses for subjectively scored traits were obtained in Finnhorse trotters, whereas in Standardbred trotters genetic correlations for subjectively scored traits varied from moderately negative to highly positive. Higher estimates of heritability for foal traits and generally high genetic correlations between the foal and studbook traits indicate that an early selection for conformation traits would be efficient in the breeding programmes.  相似文献

The major histocompatibility complex (MHC) genes play well‐defined roles in eliciting immune responses and combating infectious diseases. This genetic system is among the most polymorphic. The extent of genetic variation within a population has been directly correlated with fitness for many traits. The MHC class II locus DRA polymorphism was analysed in the endangered Sorraia horse, two other Portuguese and four New World horse breeds considered to be historically close to the Sorraia. Comparison of the Sorraia with other breeds demonstrated less MHC variation among Sorraia horses. If DRA polymorphism provides greater disease resistance, selective breeding to increase MHC polymorphism may increase fitness of this population.  相似文献

旨在揭示康西草原马匹的品种来源以及群体遗传结构。本研究采集了当地60匹健康状况良好、体重适中、3~6岁之间不区分公母马的血液样品,试验分为6个种群并以纯血马等4个国外马品种、1个我国地方马品种作为对照, 通过设计引物并提取基因组DNA结合微卫星扩增法对染色体上12个微卫星位点的等位基因进行检测,并对各种群遗传多样性参数、群体分化指数、瓶颈效应以及群体结构进行分析。试验按照群体个数为依据分为6组,每组1个重复,每个重复以该组样本量为标准。分析6个种群马的遗传参数结果表明,其中共检测到221个等位基因,平均Shannon指数为1.691;有效等位基因数(Ne)为3.649~5.397;期望杂合度(He)为0.681~0.798;观测杂合度(Ho)为0.632~0.780;平均多态信息含量(PIC)为0.643~0.772。这些结果表明,6个群体都具有很高的遗传多样性。通过微卫星DNA位点的连锁不平衡及哈迪-温伯格平衡分析发现, 大多数位点的P值都大于校正值(P=0.006 5),表示较多位点都处于哈迪-温伯格及连锁平衡状态。各群体间群体分化系数(Fst)表明,群体间分化程度较低(Fst<0.15)。基于Nei’s遗传距离与地理距离之间Mantel相关性检验显示二者之间呈现正相关性(R=0.502),但未达到显著水平(P=0.310);各马匹种群瓶颈效应分析表明,P值极显著(P(IAM)<0.01)说明该地区的马群体受到过瓶颈效应的影响且该马群体历史上数量有大规模的减少。通过 FCA因子分析、基于Nei’s标准遗传距离UPGMA树状图以及Structure贝叶斯聚类分析发现,该地区马种群与纯血马和温血马聚为一类,说明该种群最有可能主要来源于温血马和纯血马。这些结果揭示,康西草原地区马匹具有较高的遗传多样性,该地马群体的血统与纯血马和温血马相近,这为当地马遗传资源评估以及开发利用打下了坚实的基础。  相似文献

In this study, genetic analyses of diversity and differentiation were performed on five horse breeds raised in Algeria (Barb, Arab‐Barb, Arabian, Thoroughbred and French Trotter). All microsatellite markers were highly polymorphic in all the breeds. A total of 123 alleles from 14 microsatellite loci were detected in 201 horses. The average number of alleles per locus was the highest in the Arab‐Barb horses (7.86) and lowest in the thoroughbred breed (5.71), whereas the observed and expected heterozygosities per breed ranged from 0.71 (Thoroughbred) to 0.752 (Barb) and 0.71 (Thoroughbred) to 0.77 (Arab‐Barb), respectively. The genetic differentiation between the breeds was significant (p < 0.01) based on the infinitesimal model (FST). Three different approaches for evaluating the genetic relationships were applied. Genetic distances, the factorial correspondence analysis and structure analysis showed that a significant amount of genetic variation is maintained in the native horse populations and the other breeds. The Barb and Arab‐Barb breeds seem to be the most genetically related and support the decision to consider the breeds as same population.  相似文献
