为明确PG基因在菠萝蜜果实后熟软化过程中的作用,本研究以‘海大2号’菠萝蜜果实为材料,采用0.5 mg/L 1-MCP和1000 mg/L ETH处理,研究了室温(20℃)条件下果实成熟过程中硬度和果胶的动态变化,克隆获得4个PG基因,并对其进行了生物信息学和表达分析。结果表明:随着菠萝蜜果实的成熟,果肉硬度快速下降,可溶性果胶和离子型果胶不断增加,共价态果胶有所下降;ETH处理促进了果实WSP和ISP含量的上升,加速了软化进程,1-MCP处理抑制了贮藏前期果肉硬度的下降,推迟了软化进程,但明显提高了3种果胶的含量。AhePG1~AhePG4基因的开放阅读框(ORF)长度1221~1434 bp,编码406~477个氨基酸。AhePG1蛋白含有4个保守结构域(Ⅰ~Ⅳ),AhePG2和AhePG3只含结构域Ⅰ和Ⅱ,AhePG4缺失结构域Ⅲ;AhePG基因分别与桃(AF095577.1)、菜豆(XM_007162208.1)、葎草(MN971583.1)、菜豆(XM_007151391.1)PG基因编码的氨基酸序列的亲缘关系较近,相似度分别达75.63%、73.11%、79.71%、70.75%。qRT-PCR分析结果显示:AhePG1基因在果实成熟前期表达量低,在后期高表达,而AhePG2/3/4基因的表达量在果实成熟过程中总体较低。ETH处理抑制了AhePG1基因的表达量,而1-MCP处理延缓了4个AhePG基因表达量的增加,但增加了成熟后期AhePG2、AhePG3、AhePG4基因的表达。相关性分析发现,果肉硬度与水溶性果胶含量和AhePG1基因表达呈显著和极显著负相关,而水溶性果胶含量又与AhePG1基因表达呈显著正相关。本研究说明,菠萝蜜果实的软化与果胶降解有关,AhePG1可能是菠萝蜜果实果胶降解和果实软化的关键PG基因之一,控制着果实成熟后期的软化;1-MCP处理能延缓菠萝蜜果实的成熟,但并不影响果实后期成熟时的软化,AhePG1、AhePG2、AhePG3、AhePG4基因可能对其软化均有作用。  相似文献

Mature cactus pears from Opuntia stricta have a dark purple color due to high betacyanin concentration, whose biosynthesis is initiated with the amino acid L-tyrosine as a primary precursor. This study followed the maturation and ripening processes of Opuntia stricta fruits to harvest them at high betacyanin and other antioxidant concentrations. Fruits lasted 9 months for final ripening. Physical and compositional changes at different maturation and ripening stages have been determined. Thus, ripe fruits were around 4.72?±?0.10 cm length, 2.94?±?0.05 cm diameter and 22.71?±?0.20 g weight; moisture and pH were maintained at 87.05?±?0.19 % and 3.37?±?0.12, respectively. Purple pigment production started in the ovary of immature fruits four months after anthesis (MAA). Concentration of all analyzed metabolites increased from immature (4 MAA) until ripe (9 MAA) stage. In ripe fruits, reducing sugars were 4.72?±?0.54 g/100 g ff and total phenols 135.17?±?0.68 mg gallic acid/100 g ff. Metabolites identified by HPLC were the betacyanins: betanin (60.17?±?1.08 mg/100 g ff), isobetanin (7.58?±?0.94 mg/100 g ff) and betanidin (13.48?±?0.87 mg/100 g ff). Also, L-ascorbic acid (35.03?±?1.06 mg/100 g ff) and L-tyrosine (4.43?±?0.73 mg/100 g ff) were determined. Furthermore, the addition of L-tyrosine or L-dopa to fruit pulp of moderately ripe fruits, increased betacyanin concentrations 17 (103.3?±?3.8 mg/100 g) and 32 % (114.3?±?4.1 mg/100 g), respectively.  相似文献

The composition of sunflower (Helianthus annuus) seed heads was investigated. High amounts of lignin (12%) were found while glycosides comprised a minor component of the plant material. Lipids were isolated and a total sterol content of 1.05% was found. A protein content of 12.5% was also found. Acid hydrolysis of the plant material showed the presence of combined galacturonic acid (8%), galactose (3.4%), glucose (1.7%), arabinose (0.63%), xylose (0.7%) and rhamnose (21%). The factors influencing the extraction of pectin (11.85%) were studied.  相似文献

Changes in fatty acids were studied during maturation of coriander (Coriandrum sativum L.) fruits cultivated in the North-East of Tunisia (Charfine). The fruits matured in 55 days after flowering (DAF). Oil and petroselinic acid synthesis proceeded at a steady rate up to 32 DAF. The first results showed a rapid oil accumulation started at newly formed fruits (9.6 ± 0.2%) and continued until their full maturity (26.4 ± 0.5%). During fruit maturation, fatty acid profiles varied significantly among the nine stages of maturity. At the 32th DAF, palmitoleic, gadoleic, erucic and docosahexenoic acids were not detected and petroselinic acid had a highest amount (84.8 ± 4.5%). Fruits development resulted mainly in an increase of petroselinic acid and a decrease of palmitic acid (C16:0). At full maturity, the main fatty acids were petroselinic acid (80.9 ± 5.7%), followed by linoleic (13.6 ± 2.9%), palmitic (3.6 ± 0.1%) and stearic (0.7 ± 0.1%) acids. Saturated and polyunsaturated fatty acids decreased significantly and monounsaturated fatty acids increased during maturation of coriander fruit. Coriander fruits at the first four stages of maturity have a healthy nutritional value and the last five stages were with important economic and industrial applications. Results of this study indicate that the variation in the fatty acid composition of coriander fruit during maturation may be useful in understanding the source of nutritionally and industrially important fatty acids in this fruit. Coriander fruit is potentially an important source of petroselinic acid which has numerous industrial applications.  相似文献

The polysaccharide composition of cell wall of sour cherry (Prunus cerasus var. Schattenmorelle) fruit and pomace was investigated. Furthermore, the alcohol insoluble solids composition of ‘Kelleriis’ and ‘Dobreczyn Botermo’ varieties were studied too. Yield of alcohol insoluble solids for fruits was lower than 10%, and for pomaces circa 50%. Uronic acid was the main pectin component of alcohol insoluble solids. Enzymes used as juice processing aids decreased the content of uronic acid. Araban and galactan side chains bonded tightly to cellulose presence was suggested by high content of arabinose and galactose in hemicellulose fraction. The process of drying at below 70 °C did not influence polysaccharide composition of sour cherry pomaces. Alcohol insoluble solids of fruits expressed higher hydration properties than of pomaces.  相似文献

In the present study, acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and tyrosinase inhibitory and antioxidant activities of the fruit and leaf extracts of Melia azedarach L. (Meliaceae) of Turkish origin were evaluated. Enzyme inhibitory activity of the extracts was tested in vitro using ELISA microplate reader. Antioxidant activity of the extracts was tested using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, ferrous ion-chelation, and ferric-reducing antioxidant power (FRAP) assays. Phenolic composition of the extracts was elucidated by high performance liquid chromatography (HPLC) and fatty acid compositions of the fatty oils of the fruits and leaves were elucidated by gas chromatography-mass spectrometry (GC-MS). The ethyl acetate extract from the leaves showed the highest inhibition against AChE (33.63 ± 1.40%) and BChE (92.89 ± 3.05%). The methanol extract from the leaves exerted the best antioxidant activity in DPPH radical scavenging and FRAP assays, while the ethyl acetate extracts of the fruits and leaves had the most notable effect in metal-chelation assay.  相似文献

Influence of amino acid compositions and peptide profiles on antioxidant capacities of two protein hydrolysates from skipjack tuna (Katsuwonus pelamis) dark muscle was investigated. Dark muscles from skipjack tuna were hydrolyzed using five separate proteases, including pepsin, trypsin, Neutrase, papain and Alcalase. Two hydrolysates, ATH and NTH, prepared using Alcalase and Neutrase, respectively, showed the strongest antioxidant capacities and were further fractionated using ultrafiltration and gel filtration chromatography. Two fractions, Fr.A3 and Fr.B2, isolated from ATH and NTH, respectively, showed strong radical scavenging activities toward 2,2-diphenyl-1-picrylhydrazyl radicals (EC50 1.08% ± 0.08% and 0.98% ± 0.07%), hydroxyl radicals (EC50 0.22% ± 0.03% and 0.48% ± 0.05%), and superoxide anion radicals (EC50 1.31% ± 0.11% and 1.56% ± 1.03%) and effectively inhibited lipid peroxidation. Eighteen peptides from Fr.A3 and 13 peptides from Fr.B2 were isolated by reversed-phase high performance liquid chromatography, and their amino acid sequences were determined. The elevated antioxidant activity of Fr.A3 might be due to its high content of hydrophobic and aromatic amino acid residues (181.1 and 469.9 residues/1000 residues, respectively), small molecular sizes (3–6 peptides), low molecular weights (524.78 kDa), and amino acid sequences (antioxidant score 6.11). This study confirmed that a smaller molecular size, the presence of hydrophobic and aromatic amino acid residues, and the amino acid sequences were the key factors that determined the antioxidant activities of the proteins, hydrolysates and peptides. The results also demonstrated that the derived hydrolysates and fractions from skipjack tuna (K. pelamis) dark muscles could prevent oxidative reactions and might be useful for food preservation and medicinal purposes.  相似文献

The essential oil and fatty acid composition of two provenances of Ruta chalepensis from four organs (leaves, flowers, stems and fruits) was determined. The effect of the plant part on total fatty acid contents, essential oil yields, fatty acid and volatile constituents was significant.Fatty acid profiles varied significantly among the studied provenances and organs. Linolenic acid had the highest amount in leaves of the two provenances. From R. chalepensis, in all organs, the main fatty acids were palmitic (13.10-25.31%), followed by palmitoleic (0-15.72%), stearic (1.03-6.85%), oleic (1.90-24.04%), arachidic (0.11-4.03%), eicosatetraenoic (0.10-5.60%) and behenic (0.47-6.09%) acids. Saturated fatty acids had the highest amounts in growing wild R. chalepensis flowers, and cultivated R. chalepensis stems were characterized by the predominance of polyunsaturated fatty acids. Oil composition of all studied organs has a healthy and nutritionally value. Essential oil yields varied from 0.39% to 2.46% and showed a remarkable variation with plant organs. Thirty-six volatile compounds were identified in different analyzed essential oils; 2-undecanone, 2-nonanol and 2-dodecanone had the highest percentages.  相似文献

Changes in chemical composition and the activities of hydrolytic enzymes during four different stages of maturity, viz. mature green (MG), color turning (CT), ripe (R), and overripe (OR), have been studied in guava fruits cv. Banarsi Surkha. Chlorophyll content decreased while carotenoid content increased during ripening. Starch content decreased with concomitant increase in alcohol-soluble sugars. Cellulose, hemicellulose, and lignin also decreased up to ripe stage, while pectin continued to decrease up to OR stage. PG (polygalacturonase) and cellulase exhibited progressive increase in activity throughout ripening, whereas pectin methyl esterase (PME) activity increased up to CT stage and decreased at R stage. The activities of alpha-amylase and beta-amylase decreased significantly with ripening. The most notable metabolic changes occurred between MG and CT stage, implying that for improved postharvest handling, guava fruits may be harvested at CT stage.  相似文献

Antifungal potential of Streptomyces hygroscopicus metabolites was tested against the isolates of Colletotrichum spp., obtained from rotten apple fruits. Experimental factorial design and response surface methodology were employed to optimize medium composition and enhance antifungal components productivity by S. hygroscopicus. Results of optimization suggest that the optimal medium composition is (g/L): glycerol (13.92), yeast extract (5.00), phosphates (0.63), CaCO3, (3.0), NaCl, (3.0), MgSO4, (0.5). Further research was performed to validate the obtained results and confirm their applicability in planta on artificially inoculated apple fruits. The obtained efficacy after 14 days of incubation was 99.3% against Colletotrichum acutatum and 99.8% against Colletotrichum gloeosporioides. Application of supernatant of S. hygroscopicus cultivation medium, produced under defined conditions, in biological control of apple rot could offer safe and healthy apple fruits on the market, while simultaneously helping to solve the problem of waste glycerol disposal in the biodiesel industry.  相似文献

Ascorbic acid content in green and ripe fruits and zinc, copper, manganese, iron, sodium, potassium, calcium, magnesium and phosphorus content in dry fruits were found to vary significantly in different varieties of chilli (Capsicum annuum L.). Ascorbic acid content in ripe fruits of all of the eleven chilli varieties was higher than in the corresponding green fruits. Path coefficient analysis showed that number of days to fruit ripening, fruit surface area, fruit length, dry and fresh fruit yield, ascorbic acid content in green fruits and iron, calcium, magnesium and copper content in dry fruits influenced ascorbic acid content in ripe fruits of chilli.  相似文献

The biochemical composition of three novel selected microalgae strains (Chlorophyta) was evaluated to confirm their potential possibilities as new sustainably produced biomass with nutritional, functional, and/or biomedical properties. Extracts from cultured Pseudopediastrum boryanum, Chloromonas cf. reticulata, and Chloroidium saccharophilum exhibited higher radical scavenging activity of DPPH (1,1-diphenyl-2-picrylhydrazyl) when compared to butylated hydroxytoluene (BHT), but lower than butylated hydroxyanisole (BHA). Total phenolic compounds and amino acids were determined by newly developed RP-HPLC methods. Total phenolic contents, as µg g−1 of dry biomass, reached 27.1 for C. cf. reticulata, 26.4 for P. boryanum, and 55.8 for C. saccharophilum. Percentages of total analysed amino acids were 24.3, 32.1, and 18.5% of dry biomass, respectively, presenting high values for essential amino acids reaching 54.1, 72.6, and 61.2%, respectively. Glutamic acid was the most abundant free amino acid in all microalgae samples, followed by proline and lysine in C. saccharophilum and P. boryanum, and methionine and lysine in C. reticulata. Soluble carbohydrates in aqueous extracts ranged from 39.6 for C. saccharophilum to 49.3% for C. reticulata, increasing values to 45.1 for C. saccharophilum and 52.7% for P. boryanum in acid hydrolysates of dried biomass. Results confirmed the potential possibilities of these microalgae strains.  相似文献

Acid soluble collagen (ASC) from scales of croceine croaker (ASC-C) was successfully isolated with the yield of 0.37% ± 0.08% (dry weight basis), and characterized as type I collagen on the basis of amino acid analysis and electrophoretic pattern. The antioxidant hydrolysate of ASC-C (ACH) was prepared through a two-stage in vitro digestion (4-h trypsin followed by 4-h pepsin), and three antioxidant peptides (ACH-P1, ACH-P2, and ACH-P3) were further isolated from ACH using ultrafiltration, gel chromatography, and RP-HPLC, and their amino acid sequences were identified as GFRGTIGLVG (ACH-P1), GPAGPAG (ACH-P2), and GFPSG (ACH-P3). ACH-P1, ACH-P2, and ACH-P3 showed good scavenging activities on hydroxyl radical (IC50 0.293, 0.240, and 0.107 mg/mL, respectively), DPPH radical (IC50 1.271, 0.675, and 0.283 mg/mL, respectively), superoxide radical (IC50 0.463, 0.099, and 0.151 mg/mL, respectively), and ABTS radical (IC50 0.421, 0.309, and 0.210 mg/mL, respectively). ACH-P3 was also effectively against lipid peroxidation in the model system. The antioxidant activities of three collagen peptides were due to the presence of hydrophobic amino acid residues within the peptide sequences. The collagen peptides might be used as antioxidant for the therapy of diseases associated with oxidative stress, or reducing oxidative changes during storage.  相似文献

A new in vitro anti-tumor polypeptide, coded as J2-C3, was isolated from Arca inflata Reeve and purified by diethyl-aminoethanol (DEAE)-sepharose Fast Flow anion exchange and phenyl sepharose CL-4B hydrophobic chromatography. J2-C3 was identified to be a homogeneous compound by native polyacrylamide gel electrophoresis (Native-PAGE). The purity of J2-C3 was over 99% in reversed phase-high performance liquid chromatography (RP-HPLC). The molecular weight was determined as 20,538.0 Da by electrospray-ionization mass spectrometry (ESI-MS/MS). J2-C3 was rich in Glx (Gln + Glu), Lys, and Asx (Asp + Asn) according to amino acid analysis. Four partial amino acid sequences of this peptide were determined as L/ISMEDVEESR, KNGMHSI/LDVNHDGR, AMKI/LI/LNPKKGI/LVPR and AMGAHKPPKGNEL/IGHR via MALDI-TOF/TOF-MS and de novo sequencing. Secondary structural analysis by CD spectroscopy revealed that J2-C3 had the α-helix (45.2%), β-sheet (2.9%), β-turn (26.0%) and random coil (25.9%). The anti-tumor effect of J2-C3 against human tumor cells was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, and the IC50 values of J2-C3 were 65.57, 93.33 and 122.95 µg/mL against A549, HT-29 and HepG2 cell lines, respectively. Therefore, J2-C3 might be developed as a potential anti-tumor agent.  相似文献

Both the pericarp and the unripe seeds of the unripe fruits ofGlossonema edule were separately analysed. The crude protein content of the unripe seeds is double that of the pericarp. Investigation of the amino acids of both parts revealed slight quantitative differences with chemical scores of 40, 40 for each of methionine and tryptophan and 65 for lysine. The ratio of essential total: amino acid averages 34.5, compared to a corresponding value of 47.4 for the reference egg proteins. The analysis of the fatty acids showed that the ratio of saturated to non-saturated fatty acids averaged 73:27; palmitic acid makes up 60% of the total fatty acids. The soluble carbohydrate content of the pericarp is slightly higher than the corresponding value in the unripe seeds. Paper chromatography of the free sugars of both parts revealed qualitative differences. The oligosaccharide reffinose was detected in both parts. The ash was analysed for its content in four trace minerals: iron, zinc, manganese and copper.  相似文献

Juçara (Euterpe edulis M.) fruits are an interesting source of phenolic compounds, mainly anthocyanins, making them valuable to the food and pharmaceutical industries. Juçara fruits were harvested along the on-tree ripening process between March and June as practiced in Paraná state, Brazil and examined for their total anthocyanin content (TAC), total phenolic content (TPC), total phenolic acid (TPA) and total antioxidant capacity (TAA). Overall, TAC increased (91.52–236.19 mg cyanidin-3-glucoside equivalent/100 g dm) whereas TPC (81.69–49.09 mg GAE/g dm) and TPA (44.27–30.95 mg/100 g dm) decreased during ripening of juçara fruits. Use of tandem mass spectrometry allowed the identification of cyanidin-3,5-diglucoside, peonidin-3-glucoside and peonidin-3-rutinoside for the first time in juçara fruits. The analysis of the phenolic acids by HPLC-MS/MS indicated the presence of gallic, protocatechuic, p-hydroxybenzoic, vanillic, chlorogenic, caffeic, syringic, p-coumaric, sinapinic and ferulic acids. The high antioxidant capacity using DPPH radical scavenging capacity (655.89–745.32 μmol TE/g dm) and ORAC assays (1088.10–2071.55 μmol TE/g dm) showed that juçara fruits have potential as a source of novel natural antioxidants for disease prevention and health promotion, and also as natural food additives for developing new functional food products.  相似文献

Results are reported on the nutritional quality of prickly pear seeds,Opuntia ficus-indica. The seeds contained 16.6% protein, 17.2% fat, 49.6% fiber and 3.0% ash. The meal showed a high amount of iron (9.45 mg %). The contents of Mg, P, K, Zn and Cu were nutritionally significant contributing approximately 10–20% of the Recommended Dietary Allowances (RDA) of these elements per 100 g of dry weight. The amount of Ca represented less than 10% of the RDA for that element. Aspartic acid, glutamic acid, arginine and glycine were the most abundant amino acids making nearly half of the total amino acids content. The seeds were rich in sulfur amino acids (methionine + cystine). Lysine was the first limiting amino acid resulting in a chemical score of 62 for the protein. The in-vitro protein digestibility and the calculated protein efficiency ratio were 77% and 1.82 compared to 90% and 2.50, respectively, for ANRC casein.  相似文献

Fruits with red and orange flesh of the columnar cactus pitaya (Stenocereus pruinosus) were studied to evaluate physical characteristics, total soluble solids, betalains and soluble phenols content, and antioxidant activity. Fruits had, in average, weight of 179.0 g, 9.8 °Brix, 9.4 % carbohydrates, 1.25 % protein, 0.11 % ethereal extract, 0.60 % crude fiber, and 0.62 % ash. Also, fruits resulted rich in Fe (22.8–27.8 mg/kg). Hue angle and contents of betacyanins, betaxanthins (μg/g dry sample), and total soluble phenols (mg GAE/g fresh sample) were 19.8°, 2860.0, 3210.0, and 1.6 in the red material, and 28.9°, 470.0, 2670.0, and 1.2, respectively, in the orange fruit. The antioxidant capacity was higher in the red material, since the ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) technique reported 1090.6 and 735.4 μmol of Trolox equivalents/g fresh flesh in red and orange fruits, respectively, while the oxygen radical absorbance capacity (ORAC) assay reported 7.84 and 5.16 μmol of Trolox equivalents/g fresh flesh, respectively. The chromatographic profile showed five betalains in red fruits, but only four of them were observed in those orange fleshed.  相似文献

An investigation of the composition of two varieties ofLupinus termis seeds showed the presence of proteins, free amino acids, lipids, lignin, polysaccharides and mannitol. The polysaccharide materials (27.47 – 34.33%) consisted of starch, combined galacturonic acid, galactose, arabinose, xylose and rhamnose. Combined galactose and rhamnose comprised the major components of polysaccharide materials. Protein's constituted 46.11 – 50.57% ofL. termis seeds. The amino acid composition of the seeds as well as those of the proteins isolated with different agents were determined. The amino acid patterns of free and combined amino acids varied according to the extracting agent.  相似文献

Cryptostegia grandiflora, a member of Asclepiadaceae, was evaluated as a potential multi-use crop. The plant contained 14.0% protein, 6.5% oil, 6.9% polyphenol, and 2.13% hydrocarbon. The gross heat value of the species was 3878.0 cal/g, while the oil fraction was 7350.1 cal/g, and the hydrocarbon fraction was 9300.0 cal/g. The NMR spectra of the hydrocarbon fraction reveals the presence of cis-polyisoprene (natural rubber). The oil fraction contains both saturated and unsaturated fatty acids including: lauric acid (trace), myristic acid (15.24%), palmitic acid (25.90%), stearic acid (3.8%), oleic acid (8.0%), linoleic acid (24.76%), and arachidic acid (22.28%). The high proportion of saturated fatty acids and the high oil content (>5.0%) make C. grandiflora a potential source for industrial raw material and alternative for conventional oil.  相似文献
