Molecular characterization of germplasm is important for sustainable exploitation of crops. DNA diversity was measured using inter-retrotransposon-amplified-polymorphism and inter-simple-sequence-repeat markers in 203 Ethiopian landraces and reference varieties of linseed (flax, Linum usitatissimum) and wild Linum species. Molecular diversity was high (PIC, 0.16; GD, 0.19) compared to other reports from the species. Genotyping separated reference from landrace accessions, and clustered landrace accessions from different altitudes and geographical regions. Collections showed evidence for recent introduction of varieties in some regions. The phylogeny supported L. bienne Mill. as the progenitor of domesticated L. usitatissimum. Markers developed here will be useful for genetic mapping and selection of breeding lines. The results show the range of characters that can be exploited in breeding lines appropriate for smallholder and commercial farmers in Ethiopia, producing a sustainable, secure, high-value crop meeting agricultural, economic and cultural needs.  相似文献

Abstract

Barley (Hordeum vulgare L.) landraces display a high degree of variability in morphological and developmental traits, in disease resistance, and in protein content. Representatives of 29 barley landraces from southeast Turkey were collected from farmers’ fields, for a total of 800 accessions. The objectives of this study were to characterize these accessions over four years for morphological and agronomical traits to be used for future selection and breeding program. The observed variation between landraces was very large for all traits. In the first year of testing the accessions showed average grain yields ranging from 197–2225 kg ha?1. After three years of selection, promising accessions were tested at two different geographical regions and using two different irrigation methods. One line was identified which significantly out-yielded the local landrace in all of the testing years and had a higher average yield than the check genotypes.  相似文献

Landraces are thought to be well adapted to the environmental conditions of the region where they have evolved and to be highly tolerant to biotic and abiotic stress. There is indeed experimental evidence of that for several cereal species, but to our knowledge only scarce data is available on rye and none on rye landraces from South Tyrol in particular. For this region, assumptions rely mainly on historic documents or reports from the owners of the landraces. In a 3-year field experiment we described 13 landraces from Val Venosta/Vinschgau (South Tyrol, Italy), along with six modern population cultivars (CUL), at a mountain location. Depending on the available passport data, the landraces were assigned to two variety types: well documented landrace status (DLS) and uncertain landrace status (ULS). 14 traits concerning phenology, agronomy, indirect milling and baking quality as well as seed colour were investigated. Most of the traits representing the usual target of modern breeding were found to be dependent on the variety type. The results reflect well the expected differences between CUL and DLS, while ULS had an intermediate character. Winter hardiness and ripening time of both DLS and ULS, however, were found to be affected by the altitude of the site at which the landraces were collected, suggesting a certain adaptation of the landraces to the climatic conditions at the sites of origin.  相似文献

In Ethiopia, barley is generally grown in two different planting seasons per year: during the long rainy season (Meher) and the short rainy season (Belg). The aim of the present study was to assess for the first time the role of this ‘two-season system’ on the structure of the genetic diversity of the Ethiopian barley landraces. We characterised 3,170 individual genotypes from 106 landrace populations using eight morphological spike traits. The diversity within population was higher in the season where barley is more important (Belg), and in general, where its cultivation is in larger plots because of weaker ‘competition’ with others crops. This indicates that barley diversity has a complex relationship with variations in the surrounding agro-ecosystem. Overall, the divergence between the two seasons was quite small (3.4%), suggesting that seed flow does not occur independently across the years within the two seasons. This would affect the amount of mutations and historic recombination that have accumulated within these populations.  相似文献

An aged seed sample of a single oat landrace resulted in four plants when germinated. The plants differed from each other morphologically and genetically. They represented four grain types, three lemma colours, three avenin and three residual grain protein patterns and two molar ratios of C18:C16 fatty acids. These old oats grew high under current conditions and some also had yielding potential. Landraces with wide residual to avenin protein ratios would have been good sources in breeding food cereals for coeliac patients. Literature data since the 1700s indicate that Finnish oat landraces included morphological mixtures. Some were intentionally mixed. As also noted for Finnish barley landraces, oat landraces appear to have approached mixtures of genetically unique plants. Several reasons for the variation are presented. Active replacement of landraces (genetic polymorphism) with only a few pure-line cultivars was encouraged by authorities during the Mendelian period. Genetic variation was lost earliest in oats among Finnish cereal landraces. The proportion of oat landraces declined from 100% in 1902 to 34.4% in 1922 and to 0.2% in 1955. Genetic conservation did not occur in time, resulting from lack of foresight by authorities.  相似文献

Genetic relationships, agronomic, nutritional and technological traits of ten Italian landraces, two improved lines and two cultivars of lentil (Lens culinaris Medik.) were investigated using a multi-disciplinary approach. Seed storage proteins, used as biochemical markers, were able to detect polymorphisms with variability mainly related to the polypeptide abundance. Microsatellite (SSR) molecular markers provided very useful information on genetic variation and relationships among landraces, with polymorphic fragments able to discriminate all the accessions. Lentil landraces were grouped in different clusters and sub-clusters principally on the basis of their geographical origin. The highest levels of genetic diversity were observed for lentils from ‘Castelluccio di Norcia’, ‘Colliano’ and ‘Villalba’. Field trials, performed in two locations of Southern Italy, revealed a high influence of location on yield. Comparing performances at both tested locations, the best landraces were ‘Linosa’ and ‘Valle di Nevola’ suggesting that these have the highest adaptability. Technological and nutritional data together with the agronomic ones evidenced that ‘Linosa’ lentil is the best landrace, however also ‘San Gerardo’ deserves some attention.  相似文献

The introduction of novel quality characteristics from wheat (Triticum aestivum L.) landraces can enhance the genetic diversity of current wheat breeding programs. The composition of starch and protein in wheat is important when determining the end‐product quality, particularly for white salted noodles (WSN). Quality characteristics that contribute to the production of improved WSN include high starch pasting peak viscosity, low amylose content, high proportion of A‐type granules, low protein content, soft grain texture, and high protein quality as measured by SDS sedimentation volume. A survey of 133 wheat landraces from Afghanistan, China, Egypt, Ethiopia, India, Iran, Syria, and Turkey was conducted to examine the genetic variability of starch and protein quality characteristics. Two wheat cultivars, Rosella and Meering, were used as the quality controls. The variation in starch pasting peak viscosities observed among the wheat landraces had a range of 175–295 Rapid Visco Analyser units (RVU), where 52 of the landraces were not significantly different from Rosella, a commercial soft grain wheat with high pasting properties. The amylose content of the landrace population was 23.4–30.2%, where 17 landraces had significantly lower values than Rosella. The proportion of A‐type granules was 60.5–73.9%, where 112 landraces had significantly higher values than Rosella. The grain texture hardness score was 28.0–99.3, the total protein content was 8.0–15.1%, and the adjusted SDS sedimentation volume (SDS/protein) was 1.6–7.0 mL/%P. The landrace AUS4635 had high starch pasting peak viscosity, high breakdown, low amylose content, low protein content, soft grain texture, and high protein quality flour. This wheat is an ideal parent to use in a breeding program that increases the genetic variation available to develop cultivars with high‐quality WSN characteristics.  相似文献

Apart from their significance in genetic conservation, barley landraces are still favoured by local farmers in low-input farming systems. They often perform more predictably under adverse conditions than modern cultivars due to local adaptation. Increased seed exchange between farmers may put adaptation of local populations of barley landraces at risk. Isozyme markers were used to investigate differentiation and infer gene flow between local populations of barley landrace Arabi Aswad in Northern Syria. Inferred gene flow decreased exponentially with geographic distance and may imply that seed exchange between farmers is limited to a regional scale and diminishes over longer distance. Gene flow seems to be a suitable index for estimating seed exchange in highly inbreeding crop species such as barley. In the future, improved mobility of farmers and extension work may facilitate seed exchange over longer distances, and consequently jeopardise preservation of locally adapted landraces in barley.  相似文献

Field survey data of sorghum landrace populations grown by traditional farmers in four adjacent communities in the Ethiopian Highlands are used to analyze and project the local risk of loss of individual landraces under three scenarios: wild population, traditional farming and agricultural modernization. The risk of loss in the wild population scenario is based on landscape ecology theory which evaluates total population size, spatial distribution and patch occupancy, with high values for each factor and the sum of the factors decreasing the risk. This wild population analysis forms the basis for comparison with the other scenarios. In the traditional farming scenario, the deliberate actions of farmers must be taken into account through the recognition that the farmers favour various traits of the sorghum for specific reasons – yield, taste, storability, etc. – and will balance plant numbers according to quantity and quality requirements. In the agricultural modernization scenario, which includes the introduction of high-yield varieties and the tendency toward monocultures and agricultural chemical use, the traits of the landraces that are most likely to be displaced are hypothesized, as are those of the landraces that are likely to be retained on the basis of cultural or other factors. It is planned to follow up this baseline study with a long-term time-series study of the same communities to assess the validity of our projections.  相似文献

Thirty-two unimproved landraces of barley ( Hordeum vulgare L.) based on single plant selections in SE Finland were studied. Some of the lines still had ancient features, e.g., rachis brittleness in the tip of the spike. The lines showed 17 different and 16 unique hordein patterns and three lines showed more than one pattern, suggesting heterozygosity, and six different residual grain protein patterns. The three enzyme activities (-amylase, -amylase and -glucanase) of grains germinated aseptically for 120 h were determined. The average activity levels were high compared with a standard of five global barleys and with those determined previously in wild barley (H. vulgare ssp. spontaneum (Koch) Archers et Graebn.) grown in Finland. The ssp. spontaneum sample of 257 accessions showed significantly (P<0.001) less variation in -amylase and significantly (P<0.001) lower mean activity of all three enzymes. The high variation of these chemotypes indicates great potential variation of possible use by breeders has been lost by the disappearance and displacement of local barleys with commercial cultivars since 1950 in Finland before which barley cultivation and adaptation to the local environment had occurred over more than 3000 years. Selection for currently preferable plant characteristics in the descendants of the cross of HA52 (a landrace selection) × Adorra discriminated the hordein pattern of HA52 not being directly selected. The best landraces outyield the standard cultivars especially when there was no lodging. Top yield and small grains appeared to be associated characteristics under the environmental selection pressure, conflicting with the man-made regulations of the EU.  相似文献

There is considerable wealth of genetical and morphological variation in tetraploid wheat collected from Ethiopia by past expeditions. Several authors have speculated on the reasons for this concentration of diversity so far removed from the centre of origin of wheats in West Asia. The present study reports results of evaluation carried out on material collected in the early 1970's from Ethiopia. The landraces from the two provinces Shewa and Tigray were found to be distinctly different. This divergence was attributed to the differences in environmental conditions between them. Wide differentiation among landraces within each province was also present. The Ethiopian farmer's practice of growing composite cultivars in the field has resulted in bulk samples of Ethiopian landraces containing several agrotypes. The proportion of total variance due to differences among agrotypes within landraces was by far the greatest found in this study, followed in most cases by the variation among landrace populations within provinces which exceeded, in turn, the variation between province gene pools. The optimal strategy for further germplasm collections and implications for improvement and conservation in locally-based community gene banks are discussed.  相似文献

On-farm varietal diversity is now highest in marginal agricultural environments because, unlike the case in more favourable areas, modern varieties have not been a sufficiently attractive option for farmers to replace their landraces. However, the continued survival of landraces on farm is dependent on the continuing failure of plant breeding to provide better alternatives. Highly client-oriented breeding can produce suitable modern varieties for areas that were dominated by landraces. We examine here the case of the adoption of two upland rice varieties in three states in eastern India, Jharkhand, West Bengal and Orissa, where our surveys had confirmed low adoption of modern varieties in the upland ecosystem. The two new upland varieties were readily accepted by farmers who rapidly adopted them on large proportions of their land. On-farm diversity is maintained by the need to trade-off among varieties but once a variety with overall superiority was found this incentive was removed. The new varieties from the client-oriented breeding did not have weaknesses that farmers had to trade-off against their landraces so they were more likely to replace them. They also replaced older modern varieties. Farmers had previously maintained them along with landraces because they had to trade-off the higher yield of Kalinga III against the more stable yield of landraces or the higher yield of Vandana against the higher gain quality of the landraces. Only mean count per household could be used to test the significance of the differences between individual years. In all states, this revealed a significant reduction in landrace diversity with the adopting farmers when the addition of the two new varieties was not considered. Trends across years for mean varietal count, total count and Shannon–Wiener index showed a significant decline in Orissa, irrespective of whether the two new varieties were included in the analysis or not. In Jharkhand there was a significant decline only for mean count when the new varieties were not included. In Orissa, it was possible to test if rare landraces were as readily replaced as more common ones. Landrace replacement was unmitigated by increasing rarity yet the most rare landraces have the highest priority for genetic conservation. Some landraces were completely replaced by the farmers in the sample despite a diverse variety portfolio being a risk-reducing strategy in a region where there is a high risk of crop failure. There was little environmental heterogeneity in the marginal, drought-prone areas to slow this decline. The new varieties had an impact on the farming system because sometimes farmers brought additional upland into cultivation. They also introduced these varieties into more favourable, medium land. The strategy of releasing two new upland varieties met with some success in maintaining diversity.  相似文献

Ninety-five common bean (Phaseolus vulgaris L.) landraces from Spain were evaluated in three different environments in northern Spain for their agronomic performance and seed quality. Significant differences among landraces were found for 14 quantitative traits related to phenology, yield and its components, and seed quality traits. Environmental effects were significant for all traits evaluated except for seeds pod?1, seed width/thickness, seed weight, and seed water absorption. Landrace by environment interactions were significant for all traits except for seeds pod?1 and seed water absorption. Selection of new breeding lines for agronomic performance and seed quality within landraces should be reliable because many of them are mixture of lines. Some heirloom varieties belonging to the types faba, caparrón,riñón,ganxet and tolosana had the best performances regarding to seed quality and yield. Principal component analysis revealed differences among environments affecting the performance of the bean landraces. Variation in the landraces seems to be organized in a different way in each one of the test environments, therefore, low plasticity and specific adaptation of Spanish bean landraces to different environments is derived from this study. Some landraces, especially those large and white seeded should be an useful resource for sustainable farming systems in different biogeographical areas and a worthy germplasm for the genetic improvement of agronomic value and seed quality.  相似文献

Cowpea landraces belonging to Vigna unguiculata (L.) Walp. subsp. unguiculata cv.-gr. unguiculata and cv.-gr. sesquipedalis collected from part of the Deccan Plateau and West Coast of India were evaluated to (i) identify the diverse source(s) of variation for improved characters like pods/peduncle and seed index (ii) study the response of landraces for adaptation to drought and heat stress and (iii) understand the breeding value of a landrace in the genetic improvement of a popular cowpea cultivar. Landraces were evaluated for various morphological characters, pods/peduncle, seed index and other economically important agronomic traits, rust resistance and drought and heat tolerance in different years and environments. Landraces were found as an important source of genetic variability for pods/plant, pods/peduncle, better pod filling ability (seed index), grain yield/plant as well as drought and heat tolerance and rust resistance. Hybridization between C 152 (cv.) and DWDCC 016 (landrace) resulted in release of new variation not present in the two parents. Thus the landrace, DWDCC 016, can be utilized to improve cultivated varieties by transferring to them the economically valuable traits like pods/peduncle and seed index thereby enhancing realisation of sink potential and ultimately grain yield in a sustainable way.  相似文献

Forty-eight populations of barley landraces collected from Morocco were screened for resistance to powdery mildew and a number of different resistance genes were detected. Landraces originated from the collection of the International Center for Agricultural Research in the Dry Areas – ICARDA, Aleppo, Syria. Twenty populations of tested landraces (about 42%) showed resistance reactions and 46 single plant lines were selected. Fourteen of these lines were tested in the seedling stage with 17 and another 32 lines with 23 differential isolates of powdery mildew, respectively. The isolates were chosen according to their virulence spectra observed on the Pallas isolines differential set. Five lines originating from five populations of landraces showed resistance to all prevalent in Europe powdery mildew virulence genes. Thirty-five lines (76%) showed resistance reaction type 2. The distribution of reaction type scores indicated that about 81% of all reaction types observed were classified as powdery mildew resistance (scores 0, 1 and 2). In forty-one lines (89%) the presence of unknown genes alone or in combination with a specific one was detected. Four different resistance alleles (Mlat, Mla6, Mla14 and Mla22) were postulated to be present in the tested lines alone or in combination. Among specific resistance alleles the most common was allele Mlat (resistance Atlas). This allele was postulated to be present in twenty-three (50%) tested lines. The use of new identified sources of resistance to powdery mildew in barley breeding is discussed.  相似文献

Chawali and Lekali are two common farmer’s barley varieties or landraces in Jumla, Nepal (2,240–3,000 m) with small to bold grains and wide adaptation from irrigated low lands to high hills. This study was undertaken to test whether features of the traditional seed system can significantly influence the diversity of a crop and its conservation on-farm. In Jumla (high-hill), the barley seed system is completely informal and is mainly from farmer to farmer. In the present study, the seed flows and the pattern of genetic diversity in barley were investigated to detect differences between the two varieties and test the divergence among populations of each variety These data suggested that Chawali, the more common variety, was less subject to homogenising gene flow between farms than was Lekali. A total of 128 farming households were surveyed for seed supply information and 128 populations for each landrace from two villages: Kartikswami and Talium were collected for SSR diversity analysis. Some 92 SSRs were screened in an initial sample of 20 barley populations of both landraces and 2 improved varieties (LG-51 and Soluwa). Of the 81 SSRs that consistently amplified, only 15 SSRs (19%) were polymorphic with gene diversity values ranging from 0.09 to 0.71. A medium to low diversity was detected among the landrace populations of barley varieties. Chawali populations were less polymorphic within ecological groups, and more divergent between than were Lekail populations. This result accords with Chawali having a more conservative local seed system.  相似文献

Phaseolus vulgaris L. is an important species that originated in Mesoamerica. A Mesoamerican and an Andean gene pool are usually distinguished in the domesticated forms. Many bean landraces are still cultivated in Italy and the Department of Applied Biology maintains an ex situ collection of 146 landraces. Although protection schemes are being developed in Italy, most landraces are extinct or at risk of extinction. To facilitate their conservation and use, geographical, morphological, biochemical and molecular (SSR) data were collected and analysed to estimate the diversity and the genetic structure of the collection. Data confirmed that both the Mesoamerican and the Andean gene pools were introduced in Italy and, although a distinction between the two gene pools exists, the Italian landrace diversity is clearly structured in three clusters that are not simply ascribable to the original gene pools. The observed structure appears also to be due to adaptation to the different environmental conditions determined by altitude. This was confirmed by assessing the presence of selective effects for some of the SSR used in this study. Finally, a certain extent of admixture in Italian landrace diversity suggests past (or recurring) hybridisation events among gene pools. The combined use of morphological, biochemical and molecular data clearly distinguished almost all the landraces. The data gathered here can assist landrace in situ protection schemes that are being developed in Italy, be used to register landraces in the European common catalogue of ‘conservation varieties’ for seed commercialisation and contribute to a better use of Italian common bean diversity in breeding for organic and conventional production systems.  相似文献

Temporal variation of diversity in Italian durum wheat germplasm   总被引:1,自引:0,他引:1  
The aim of this work is to analyse the temporal change of genetic diversity in Italian durum wheat germplasm. The germplasm deployed in this study (158 accessions), belonging to 5 different historical classes, was characterised for its microsatellite and gliadin markers. The level of genetic diversity (He), based on gliadin and SSR markers results – on average – greater in indigenous landraces present in Italy before 1915, with the exception of pure line material which had been selected from landraces (showing highest level of heterozigosity for gliadin markers). Genotypes obtained from crosses or mutagenesis (referring to the 1950–1960 period) along with those resulting from crosses between CIMMYT lines and old materials (1970s and beyond) were also genetically more diverse. Forty-nine percent of indigenous landraces were genetically heterogeneous. Nine out of 53 landrace accessions were able to capture 4 different SSR private alleles. It is speculated that the reduction of allele richness is an indicator of the genetic erosion of the pre-breeding germplasm and it is pointed out that the implementation of appropriate methods of genetic conservation of this germplasm is a priority for breeding and food safety.  相似文献

An European maize (Zea mays L.) landrace core collection (EMLCC) was formed with samples from several countries. Evaluation of the EMLCC may contribute to broad the genetic base of maize breeding programs. The objective of this study was to assess the variability of EMLCC under low nitrogen (N) in relation to high N input. Eighty-five landraces of the EMLCC, grouped in four maturity groups, and three check hybrids were evaluated for response to low (0 kg ha−1) and high (150 kg ha−1) N in Spain and Greece. Five plant size traits (plant height, ear height, leaf length, leaf width and leaf area index), two grain traits (1000-kernel weight and grain yield), and two agronomic traits [growing degree units (GDU) and lodging] were studied. Overall means of plant size and grain traits increased when genotypes were grown at 150-N relative to 0-N input. The relative increase for grain traits was smaller in landraces than in hybrids. This suggests that landraces had lower grain yield response to N supply compared to hybrids. Linear regressions of plant size traits on GDU indicated that vegetative development was primarily associated with flowering lateness. The maturity group was the main source of variation for all traits. Landrace variability within maturity groups was significant for all traits across environments, despite significant landrace × environment interactions. Estimates of genetic and genotype × environment variances, and heritabilities at both high and low N inputs were not significantly different from each other. However estimates were generally larger at high N. Genetic and phenotypic correlation coefficients between the two N levels were very high for all traits.  相似文献

Many barley landraces are purple-or black-seeded, but the chemical composition of these purple-and black- seeded barley is rarely examined. Therefore, studies were conducted to determine if the chemical composition of purple and black barleys differs from that of yellow barleys. Four sets of genetic materials were used for these studies: 96 doubled-haploid (DH) lines, 10 near-isogenic lines, 40 landraces, and four F3 bulks. The results showed that purple DH lines contained a similar amount of lignin as yellow DH lines and that anthocyanin-containing isogenic lines did not differ from anthocyanin-free lines in lignin content. Therefore, these results suggest that anthocyanin biosynthesis is independent of lignin biosynthesis in barley. The results also showed that black barley contained more protein and more lignin than yellow barley in two crosses and that among the 40 landraces studied, the three with the highest concentration of lignin happened to be black seeded. Lignin content was not associated with seed weight except in defective endosperm lines. The presence of pigments and a relatively high concentration of lignin might help reduce the level of kernel blight severity in black barley. More studies are needed to determine the chemical composition and pest resistance associated with purple and black barleys.  相似文献
