During storage of potato tubers (Solanum tuberosum L., var. Bintje), important changes appear which affect respiratory control, ADPO, intensity of O2 consumption in the presence of different substrates, and NAD+ dependence, In mitochondria extracted under strictly similar conditions, from patato tubers stored at 4°C, the respiratory control (RC) maintains a value near 4 for 4 to 5 months. It then declines progressively to low values. At 20°C, a stable RC of 4 can be observed for several months, which then decreases at the end of dormancy. Then, the RC increases sharply; at this stage, ADPO are abnormally low, and, some time later, NAD+ dependence disappears. Mitochondria treated with 250 μM chlorpropham show a 50% inhibition of the electron transfer with exogenous NADH as substrate. After tuber treatment with 1% chlorpropham, sprouting is inhibited for several months. The activities of mitochondria extracted from such tubers remain unaffected by the treatment. The use of this phenylcarbamate for potato tuber treatment permits obtaining functional mitochondria from tubers after a slightly longer period of storage.  相似文献

The parasitoids of Saperda populnea (L.) (Col.: Cerambycidae) were studied in Bulgaria during the period 1997–2001. Galls with pest larvae were collected from young aspen trees at eight locations (Sofia, Kokaliane, Plana, Churek, Gorni Lom, Gintzi, Dolno Kamartzi and Klisura) and examined under laboratory conditions. Four species were reared from 1118 galls containing overwintering pest larvae: Iphiaulax impostor (Scop.) (Hym.: Braconidae), Dolichomitus populneus (Ratz.), Schreineria populnea (Gir.) (Hym.: Ichneumonidae) and Billaea irrorata (Meig.) (Dipt.: Tachinidae). All of the parasitoids were solitary. Schreineria populnea was found as a new parasitoid of the host in Bulgaria. B. irrorata emerged mainly from late-stage S. populnea larvae; the remaining species from both early- and mid-stage larvae. In the parasitoid complex of S. populnea, the most numerous were B. irrorata (59.8%) and D. populneus (29.4%). The percentage of host larval parasitism varied from 2.4 to 33.3, with an average of 9.7%. B. irrorata was the most important in reducing the pest number. The average mortality of S. populnea caused by the tachinid was 5.8%, and the highest mortality observed in individual study was 20.2%.  相似文献

(Lep., Plutellidae) and its predators, e.g. Amblyseius longispinosus (Evans) (Acari, Phytoseiidae), Erigonidium graminicola (Sundvall) (Araneae, Linyphiidae), Orius similis Zheng (Het., Anthocoridae) and Coccinella septempunctata L.(Col., Coccinellidae), in comparison to the commercial insecticides, e.g. Abamectin, Tebufenozide, Dichlorvos, Cartap and Lambda-cyhalothrin.The results showed that GCSC-BtA was highly toxic to the 3rd instar of P. xylostella with 91.18% mortality, followed by Cartap with 84.38%, Abamectin with 78.00%, Tebufenozide with 75.57%, Lambda-cyhalothrin with 63.75% and Dichlorvos with 50.86% mortality. On the other hand, GCSC-BtA was found to be comparatively less toxic to the predators, causing 31.11%, 13.33%, 11.54% and 6.00% mortalities in A. longispinosus, E. graminicola, O. similis and C. septempunctata, respectively. For comparison, the mortalities recorded for Abamectin, Tebufenozide, Dichlorvos, Cartap, Lambda-cyhalothrin were 72.94%, 55.55%, 70.00%, 53.26% and 98.85% in A. longispinosus, 46.51%, 55.10%, 60.00%, 46.00% and 73.68% in E. graminicola, 22.00%, 16.00%, 35.71%, 26.78% and 81.03% in O. similis, 15.55%, 19.64%, 28.00%, 16.66% and 41.79% in C. septempunctata, respectively.  相似文献

The 1st group consisted of GCSC-BtA characterized with low toxicity to all the predators tested with 15.49% mortality on average (highest 31.11% and lowest 6.00%). The 2nd group consisted of Abamectin, Tebufenozide, Dichlorvos and Cartap with moderate toxicity to the predators with 39.96% mortality on average. The 3rd group included Lambda-cyhalothrin with high toxicity to the predators with 73.83% mortality on average (highest 98.85% and lowest 41.70%). The susceptibility of the pest and its predators to GCSC-BtA and the insecticides is discussed. GCSC-BtA as a biological control agent is recommended for use in the integrated pests control programs in the vegetable fields.  相似文献

The relationship between Xenorhabdus nematophilus and Photorhabdus luminescens, the bacterial symbionts of the entomopathogenic nematodes Steinernema carpocapsae and Heterorhabditis bacteriophora, against the diamondback moth, P. xylostella (L.), pupae showed the pathogenic capability of P. luminescens to be over that of X. nematophilus. They gave 60 and 40% mortality with LC50 values of 5×104 and 5.5×105 cells/ml, respectively.The number of bacterial cells influences the rate of killing the pupae of P. xylostella and a direct correlation between mortality of the pupae, percentage of deformed adults and the spraying dose with significant differences were observed. An inverse correlation found between the no. of eggs laid/female, percentage of hatching, adult survival and the spraying dose. These bacterial effects have been attributed to different toxic bacterial enzymes which damage the haemocytes and inhibit activation of the humoral immune system of the insect.  相似文献

sec-Butylamine at 5 mM inhibited the oxidation of pyruvate by mitochondria isolated from hyphae of Penicillium digitalum, but had little effect on the oxidation of citrate, isocitrate, succinate, malate, acetyl-coenzyme A, or reduced nicotinamide adenine dinucleotide. sec-Butylamine did not interfere with oxidative phosphorylation, as evidenced by similar PO ratios in treated and control mitochondria. The pyruvate dehydrogenase complex (EC 1.2.4.1) isolated from young hyphae of P. digitatum was inhibited strongly by 20 mM sec-butylamine, whereas other tricarboxylic acid cycle enzymes were only slightly affected at most. Inhibition of the pyruvate dehydrogenase complex by sec-butylamine was competitive with respect to pyruvate. The Ki for sec-butylamine in the reaction was 1.38 × 10?2M, and the Km for pyruvate was 2.28 × 10?4M. These observations and other evidence derived from studies with intact hyphae support the hypothesis that the pyruvate dehydrogenase complex is the primary site of the fungistatic action of sec-butylamine.  相似文献

This paper deals with the influence of the mixed biocide GCSC-BtA on the pupal and adult stages of Apanteles plutellae Kurd. (Hym., Braconidae) and its host, Plutella xylostella (L.) (Lep., Plutellidae). The results show that mortalities of the pupae of P. xylostella in the direct-dip bioassay were 84.67%, that of the adults in the residue bioassay at 1.2500mg/ml concentration of GCSC-BtA were 78.00% which were significantly higher than the mortality values for the pupae with 54.62% and adults with 48.13% of A. plutellae. In contrast, cypermethrin showed extremely high toxicity to the pupae with 94.58% and adults with 86.00% mortality values of A. plutellae as compared to the low mortality values of 42.14% for the pupae and 32.11% for the adults of P. xylostella, with the same concentrations and bioassay methods. The LC50 values of GCSC-BtA were 0.3402, 0.5516 and 1.2405, 1.9480mg/ml for the pupae and adults of P. xylostella and A. plutellae, respectively, while the LC50 values for cypermethrin were 1.5652, 2.3471 and 0.1096, 0.1152mg/ml, respectively. GCSC-BtA was found more toxic to the pupae and adults of P. xylostella and safer to the pupae and adults of A. plutellae than cypermethrin. The possibilty of using GCSC-BtA against P. xylostella under partial control by A. plutellae in vegetable fields is discussed.  相似文献

The effects of ratio between sex pheromone components, pheromone dose in the dispenser, aging of dispenser in the field, and trap type on trapping efficiency of males of the peach twig borer,Anarsia lineatella Zeller (Lepidoptera: Gelechiidae), were investigated. To attract males, the optimal ratio between pheromonal components in a binary blend containing (E)-5-decenyl acetate (E5-10:Ac): (E)-5-decenol (E5-10:OH) was 72:28 or 83:17. Dosages of 7 or 0.7 mg of the binary blend containing E5-10:Ac: E5-10:OH (72:28) were equally effective in attracting males. The effect of aging of dispenser (Israeli dispensers, loaded with 7 mg pheromone) in the field on trapping efficiency was moderate. Captures in traps baited with 4-week-old septa did not differ from those in traps baited with 1-week-old septa. The fairly slow loss of attractancy exhibited by rubber septa indicates that septa may be used for at least 4–5 weeks. At high population levels, the nonsticky IPS trap was significantly more effective in capturing males than the sticky Pherocon 1C trap; at low populations, however, the Pherocon 1C trap was better.  相似文献

The parasitoids of Gelechia senticetella (Stgr.) (Lepid.: Gelechiidae), a pest of Juniperus excelsa M.B., were studied in the Tissata reserve in Bulgaria. They were reared in laboratory conditions from pest larvae and pupae collected in 1991, 1992 and 2000. As a result of this study, 16 species of parasitoids of G. senticetella were recorded: Habrobracon stabilis Wesm., Hormius moniliatus (Nees), Phanerotoma tritoma Marsh. (Hym.: Braconidae), Diadegma consumptor (Grav.), Liotryphon cydiae (Perk.), Scambus pomorum (Ratz.), Scambus foliae (Cush.), Pimpla turionellae (L.), Eudelus simillimus (Tasch.), Mesochorus sp. (Hym.: Ichneumonidae), Sympiesis acalle Walk., Aprostocetus sp. (Hym.: Eulophidae), Eupelmus urozonus Dalm. (Hym.: Eupelmidae), Copidosoma thompsoni Merc. (Hym.: Encyrtidae), Calliprymna bisetosa Grah., and Mesopolobus sp. (Hym.: Pteromalidae). H. stabilis and P. tritoma were discovered as new species for the fauna of Bulgaria. Three species (E. simillimus, Mesochorus sp. and Aprostocetus sp.) are hyperparasitoids, and the remaining species are primary parasitoids. Most of the parasitoid species attack the larvae of G. senticetella and kill the hosts before pupation. In the studied parasitoid complex, D. consumptor and H. moniliatus were the dominant species. In 1991, the level of total parasitism of the host in the Tissata reserve was 24.3%, in 1992 – 57.9% and in 2000 – 12.7%. D. consumptor was the most important parasitoid destroying 26.6% of the pest larvae in 1992.  相似文献

Rhizoctonia solani AG-1 IA causes leaf blight on soybean and rice. Despite the fact that R. solani AG-1 IA is a major pathogen affecting soybean and rice in Brazil and elsewhere in the world, little information is available on its genetic diversity and evolution. This study was an attempt to reveal the origin, and the patterns of movement and amplification of epidemiologically significant genotypes of R. solani AG-1 IA from soybean and rice in Brazil. For inferring intraspecific evolution of R. solani AG-1 IA sampled from soybean and rice, networks of ITS-5.8S rDNA sequencing haplotypes were built using the statistical parsimony algorithm from Clement et al. (2000) Molecular Ecology 9: 1657–1660. Higher haplotype diversity (Nei M 1987, Molecular Evolutionary Genetics Columbia University Press, New york: 512p.) was observed for the Brazilian soybean sample of R. solani AG-1 IA (0.827) in comparison with the rest of the world sample (0.431). Within the south-central American clade (3-2), four haplotypes of R. solani AG-1 IA from Mato Grosso, one from Tocantins, one from Maranhão, and one from Cuba occupied the tips of the network, indicating recent origin. The putative ancestral haplotypes had probably originated either from Mato Grosso or Maranhão States. While 16 distinct haplotypes were found in a sample of 32 soybean isolates of the pathogen, the entire rice sample (n=20) was represented by a single haplotype (haplotype 5), with a worldwide distribution. The results from nested-cladistic analysis indicated restricted gene flow with isolation by distance (or restricted dispersal by distance in nonsexual species) for the south-central American clade (3-2), mainly composed by soybean haplotypes.  相似文献

The aim of this study was to assess the influence of transgenic soybean [Glycine max L. (Fabaceae)] cultivars on the biological cycle of Tetranychus ludeni Zacher (Acari: Tetranychidae) in laboratory conditions. We used three cultivars: a conventional soybean (CO), a glyphosate-resistant with insertion of genes of Agrobacterium sp., Roundup Ready (RR), and a glyphosate-resistant with insertion of genes of Bacillus thuringiensis var. israelensis (BT). Duration of developmental stage, mite viability, number of laid eggs, and female longevity of T. ludeni were evaluated. The duration of the deutonymph phase was shorter in RR (2.1?±?0.12 days) than in CO (2.6?±?0.12) and BT (2.5?±?0.08). The duration of egg, larval, and protonymph phases were similar for T. ludeni on the three cultivars. The mean duration of each generation (T), net reproduction rate (Ro), intrinsic rate of natural increase (rm), and doubling time (DT) were similar on the three cultivars. Results showed that population growth are not influenced by transgenesis at the laboratory level. Females died without a post-oviposition period on all soybean cultivars. The intrinsic rate of natural increase of T. ludeni on soybean cultivars was low (between 0.15 and 0.17).  相似文献

An international test performance study (TPS) was organised to generate validation data for three molecular Synchytrium endobioticum tests: van den Boogert et al. (European Journal of Plant Pathology 113, 47–57, 2005), and van Gent-Pelzer et al. (European Journal of Plant Pathology, 126, 129-133, 2010) for the detection of S. endobioticum, and the pathotype 1(D1) identification test described by Bonants et al. (European Journal of Plant Pathology, 143, 495-506, 2015). Two TPS rounds were organised focussing on different test matrices, i.e. round 1: warted potato tissue, and round 2: resting spore suspensions. When using the tests for detection and identification of S. endobioticum in warted potato tissue, no significant differences were observed for diagnostic sensitivity, diagnostic specificity, overall accuracy, analytical sensitivity and robustness. When using the tests for detection and identification of S. endobioticum in resting spore suspensions, the van den Boogert and van Gent-Pelzer tests significantly outperform the Bonants test for diagnostic sensitivity and diagnostic specificity. For overall accuracy and analytical sensitivity, the van Gent-Pelzer significantly outperforms the van den Boogert and Bonants tests and is regarded as the test of choice when identifying S. endobioticum from resting spores. Tests regarded fit for purpose for routine testing of wart material and resting spore suspensions are proposed for the update of EPPO standard PM7/28(1) Synchytrium endobioticum.  相似文献
