Fifteen isolates of rust fungi were collected in the United Kingdom and in the Netherlands from severalAllium species. The samples represented three putative rust species. The morphology of the telia, teliospores and urediniospores was investigated. From urediniospores infection structures were induced on leek, and their morphology was described. Telia and teliospores were not available on every sample. Morphology of the infection structures clearly differentiated between ‘leek’ type and ‘chive’ type isolates. The morphology of infection structures ofPuccinia mixta is very similar to that ofUromyces ambiguus, but clearly distinct fromPuccinia allii sensu Jennings from leek. Quantitative differences in urediniospore characters differentiated between the putative species, but there was overlap between the taxa. We conclude that morphology of infection structures of urediniosporelings is a useful trait for identification of rust isolates fromAllium. This is especially true where more than one rust species may occur on the same host species, as withAllium fistulosum.  相似文献

The legume species Medicago truncatula is gaining interests as a plant for structural and functional genomics that can be used to identify agronomically important genes in crop legumes. Resistance to the alfalfa rust (Uromyces striatus) was studied in a germplasm collection of M. truncatula. Accessions varied in resistance, as expressed by disease severity, but none showed macroscopically visible necrosis. Histological investigations, in selected lines covering the whole range of resistance reactions, revealed little difference in spore germination and none in orientation of germtubes on the leaf surface. However, appressorium formation on the stoma was significantly reduced in some accessions. Differences in resistance among accessions were more evident once the stoma were penetrated by the infection structures. Resistance was mainly due to a restriction of haustorium formation with varying levels of early abortion of the colonies, a reduction in the number of haustoria per colony, and hampered colony growth. In addition, necrosis of the host cells associated with infection hyphae was detectable in some accessions from the beginning of colony development. This information will be useful for eventual mapping and cloning analyses of resistance genes in M. truncatula that will in turn be useful for understanding other legume/rust interactions.  相似文献

Several new isolates ofBacillus thuringiensis Berliner have shown significantly higher levels of insecticidal activity than the previous best isolates (Bt24-entomocidus and IH-A-aizawai) against bothSpodoptera littoralis Boisd. andSpodoptera exempta Walker. The best ones, in the following descending order of potency, were: againstS. littoralis — K26–21, MF4B–2, MR1–37, K28–30, K26–8; againstS. exempta — Bt-3, K30–3, HD-133, MR1–37, Bt-17, K26–8, K26–21, MF4B-2, HD-593. Larvae ofS. exempta were considerably more susceptible (4.12–1.25 fold) to some of the isolates (Bt-3, K30–3, HD-133, MR1–37, Bt-17, K26–8, HD-593) thanS. littoralis.  相似文献

Severe blight of stems, leaves and pods caused by Botrytis cinerea was found on pearl lupine (Lupinus mutabilis), a legume crop, grown in Kagawa Prefecture, Japan, in March–June 1996–2002. This disease was named “gray mold of pearl lupine” as a new disease. One of the fungal isolates obtained in this study was deposited in Genebank, National Institute of Agrobiological Sciences as accession MAFF238557.  相似文献

A gene that controls resistance to chickpea rust (Uromyces ciceris-arietini) has been identified in a recombinant inbred line (RIL) population derived from an interspecific cross between Cicer arietinum (ILC72) × Cicer reticulatum (Cr5-10), susceptible and resistant to rust, respectively. Both parental lines and all RILs displayed a compatible interaction but differed in the level of infection measured as Disease Severity (DS) and Area Under the Disease Progress Curve (AUDPC). Histological studies of the seedlings of resistant parental Cr5-10 line revealed a reduction in spore germination, appressorium formation, number of haustoria per colony and colony size, with little host cell necrosis, fitting the definition of partial resistance. A Quantitative Trait Locus (QTL) explaining 31% of the total phenotypic variation for DS in seedlings and 81% of the AUDPC in adult plants in the field was located on linkage group 7 of the chickpea genetic map. The AUDPC displayed a bimodal distribution with high frequency of susceptible lines and both the AUDPC and markers showed the same distorted segregation. Consequently, it was hypothesised that a single dominant gene (proposed as Uca1/uca1) controlled resistance to rust in adult plants. This allowed us to locate the gene on the genetic linkage map. Two Sequence Tagged Microsatellite Sites (STMS) markers, TA18 and TA180 (3.9 cM apart) were identified that flank the resistance gene. These findings could be the starting point for a Marker-Assisted Selection (MAS) programme for rust resistance in chickpea.  相似文献

Leaves of powdery mildew-susceptible barley (Hordeum vulgare cv. Ingrid) and related near-isogenic lines bearing various resistance genes (Mla12, Mlg or mlo5) were inoculated with Blumeria graminis f. sp. hordei race A6. Fungal attack induced several-fold increases in ethylene emission and electrolyte leakage in leaves of susceptible Ingrid beginning 3 days after inoculation. Activities of peroxidase, superoxide dismutase, glutathione S-transferase, ascorbate peroxidase and glutathione reductase enzymes were induced markedly in susceptible leaves 5–7 days after inoculation. Similar, but less pronounced pathogen-induced changes were detected in inoculated leaves of Mla-type resistant plants that show hypersensitive cell death upon inoculation, and, to an even lesser extent, in the Mlg and mlo lines, where no visible symptoms accompanied the incompatible interaction. Glutathione content increased only in susceptible barley 7 days after inoculation. Catalase activity, total ascorbate content and redox state were not influenced by inoculation in any of the genotypes. The activity of dehydroascorbate reductase was significantly reduced 3–5 days after inoculation in the susceptible parental plants and after 5 days in Mla and Mlg lines, while it was stable in the mlo barley. Slightly elevated levels of H2O2 were observed in the inoculated resistant plants. In contrast, H2O2 content decreased in the susceptible line 7 days after pathogen attack. These data indicate that high levels of antioxidants are involved in the compatible interaction of susceptible barley and powdery mildew by protecting the pathogen from oxidative damage.  相似文献

The effect of microclimate variables on development ofClonostachys rosea and biocontrol ofBotrytis cinerea was investigated on rose leaves and crop residues. C.rosea established and sporulated abundantly on inoculated leaflets incubated for 7–35 days at 10°, 20° and 30°C and then placed on paraquat—chloramphenical agar (PCA) for 15 days at 20°C. On leaflets kept at 10°C, the sporulation after incubation on PCA increased from 60% to 93% on samples taken 7 to 21 days after inoculation, but decreased to 45% on material sampled after 35 days. A similar pattern was observed on leaves incubated at either 20° or 30°C. The sporulation ofC. rosea on leaf disks on PCA was not affected when the onset of high humidity occurred 0, 4, 8, 12 or 16 h after inoculation. However, sporulation was reduced to 54–58% on leaflets kept for 20–24 h under dry conditions after inoculation and before being placed on PCA. The fungus sporulated on 68–74% of the surface of leaf disks kept for up to 24 h at high humidity after inoculation, but decreased to 40–51% if the high humidity period before transferral to PCA was prolonged to 36–48 h. The growth ofC. rosea on leaflets was reduced at low inoculum concentrations (103 and 104 conidia/ml) because of competition with indigenous microorganisms, but at higher concentrations (105 and 106 conidia/ml) the indigenous fungi were inhibited. Regardless of the time of application ofC. rosea in relation toB. cinerea, the pathogen’s sporulation was reduced by more than 99%. The antagonist was able to parasitize hyphae and conidiophores ofB. cinerea in the leaf residues. AsC. rosea exhibited flexibility in association with rose leaves under a wide range of microclimatic conditions, and in reducingB. cinerea sporulation on rose leaves and residues, it can be expected to suppress the pathogen effectively in rose production systems.  相似文献

The luteovirus Barley yellow dwarf virus–PAV (BYDV–PAV) and its vector, the aphid Sitobion avenae are two major sources of yield losses in cereal crops. We report in this paper the effects of a Triticum monococcum line (TM44), resistant by antibiosis to S. avenae, on the different steps of transmission of one BYDV–PAV isolate by the aphids. First, it was shown that TM44 is strongly resistant to BYDV–PAV transmission, but exclusively when S. avenae is the vector. Second, that TM44 is resistant (1) to BYDV–PAV acquisition by S. avenae and (2) to its inoculation, whatever the respective duration of these two periods. Third, that both resistances have partially additive effects. In the discussion, several lines of evidence are given to support the hypothesis that resistance of TM44 to PAV transmission is due to the same disturbances to S. avenae feeding behaviour that are involved in its antibiosis against this aphid species. Reasons for caution in releasing resistant material in the field are presented.  相似文献

Dry mycelium (DM) of killedPenicillium chrysogenum and its water extract (DME) were used to induce resistance in cotton plants againstFusarium oxysporum f.sp.vasinfectum (Fov). Results showed that the efficacy of either DM or DME in controlling the disease depends on both the concentration and the mode of application. DM amended to the soil at 0.25–2% (w/w) provided 32–75% protection againstFov. Soil drench with 2–5% DME (w/v) and pre-sowing seed soakage with 5–10% DME provided 51–77% and 28–35% protection against the wilt disease, respectively, whereas no protection was obtained with foliar sprays of 1–10% DME. DM and its water extract had no direct antifungal activity on growth ofFov in vitro, suggesting that disease control with DM or DME resulted from the induction of natural defense mechanisms in the cotton plants. Soil drench with 5% DME was as effective as 2% DM powder in inducing resistance againstFov, implying that the resistance-inducing substances were mostly water-soluble. Four cotton cultivars with various genetic resistance levels againstFov were tested at the seedling stage: two resistant ‘Pima’ cultivars and two susceptible ‘Acala’ cultivars. The level of protection achieved in the two susceptible cultivars with DME was equal to, or higher than, that of the two resistant cultivars treated with water. Innate and induced peroxidase activity in cotyledons or hypocotyls and roots coincided with the level of genetic resistance and DME-induced resistance, respectively. Based on our results, an integrated control strategy ofFov with both genetic resistance and induced resistance is suggested.  相似文献

Identifying the Fusarium species cause Fusarium head blight (FHB) and produces mycotoxins in wheat and other cereal is difficult and time consuming because of confusing phenotypic classification systems. In Japan, the F. graminearum complex, F. culmorum, F. avenaceum, and Microdochium nivale predominantly cause FHB. The internal transcribed spacer (ITS) and 5.8S of rDNA, a partial sequence of β-tubulin and mitochondrial cytochrome b (cytb) genes of the four species were PCR-amplified and analyzed. On the basis of the ITS, β-tubulin and cytb sequences, F. avenaceum and M. nivale are distinct from the F. graminearum complex and F. culmorum, whereas the F. graminearum complex is closely related to F. culmorum. Moreover, thiophanate–methyl-resistant isolates of the F. graminearum complex and F. culmorum did not have an amino acid substitution at amino acid codon 198 or 200 of β-tubulin. In contrast, very highly or highly thiophanate–methyl-resistant isolates of M. nivale had Glu (GAG) substituted with Ala (GCG) or Lys (AAG) at codon 198, respectively. The allele-specific PCR assay was used to identify the F. graminearum complex and F. culmorum, and these Fusarium species could be distinguished rapidly.  相似文献

Forty-five onion (Allium cepa) cultivars were evaluated for disease resistance to the fungal pathogen Pyrenochaeta terrestris (pink root disease). Forty-three Iranian cultivars and two commercially exotic resistant cultivars (Texas Early Grano and Yellow Sweet Spanish) were grown in a glasshouse for two successive years (2003 and 2004) in Isfahan, Iran. Susceptibility of the cultivars was determined using a scale of 0–3. Six cultivars had significantly lower susceptibility of pink root compared to the exotic ones. High susceptibility tended to be associated with high mean scores (2–3), and the highly resistant cultivars had the lowest scores (0–1). In addition, the 45 examined cultivars were ranked from 1 to 45 according to their markedly differing reactions to P. terrestris, which differed markedly. Of 32 cultivars that were less resistant, 30 with an intermediate status or tolerance (1–2) differed considerably in their rank order. No pink root symptoms were seen in the early stages, but on blotter paper or wheat-straw agar some roots turned pink with no pycnidia formation, whereas on potato dextrose agar medium (PDA) with a few fresh healthy and sterile onion roots on the surface, mycelia and pycnidia were recovered. Variance and cluster analysis showed similar results among the cultivars with various levels of resistance, tolerance and susceptibility as determined by scoring scales.  相似文献

Yeast-like fungi were isolated from lesions on azuki bean (cv. Shin-Kyotodainagon) seeds that had been sucked by bean bugs in Kyoto Prefecture, Japan. On the basis of morphological and physiological characteristics and sequence data of the internal transcribed spacer (ITS) regions including the 5.8S rDNA, these yeasts were identified as Eremothecium coryli and E. ashbyi. Pathogenicity of those yeasts was confirmed by a reinoculation test. To our knowledge, this is the first report of the occurrence of yeast spot in azuki bean in Japan. The nucleotide sequence data reported are available in the GeneBank/EMBL/DDBJ database as accessions AB478291–AB478309 for E. coryli AZC1–19 and AB478310–AB478317 for E. ashbyi AZA1–8.  相似文献

A survey of common and uncommon weed species usually showing Verticillium wilt symptoms was carried out during 1992–2000 in Crete, Greece.Verticillium dahliae was isolated in 48 out of 182 sampled fields, in which several weed species were grown, from several locations in Oropedio, Lasithi. Altogether, 124 isolates ofV. dahliae were recovered from the vascular stem-tissue of 19 weed species, belonging to ten botanical families. Pathogenicity trials with 13 out of 19 weed species that have never been reported as hosts of the fungus, using for inoculation isolates which originated from the same weed species, resulted in infection of all of them, showing various disease symptoms. Seven weed species (Anthemis melanolepis, Cardaria draba, Convolvulus arvensis, Erodium sp.,Euphorbia helioscopia, Helminthotheca echioides andSinapis alba) are new hosts worldwide, and six additional species (Euphorbia sp.,Lactuca serriola, Raphanus raphanistrum, Sinapis arvensis, Sonchus oleraceus andTrifolium sp.) are new hosts for Greece. The most susceptible (isolation frequency: 27.9–52.8%, moderate disease severity) species were:Capsella bursa-pastoris, C. draba, Chenopodium album, Senecio vulgaris andSolanum nigrum. Less susceptible (isolation frequency: 4.8–17.8%, slight disease severity) were:Amaranthus sp.,A. melanolepis, C. arvensis, Erodium sp.,Euphorbia sp.,E. helioscopia, H. echioides, L. serriola, Malva sylvestris, R. raphanistrum, S. alba, S. arvensis, S. oleraceus andTrifolium sp. Some species —C. draba, C. album, L. serriola andS. nigrum L. — that usually showed external and vascular wilt symptoms, occasionally exhibited only reduced growth. Visible symptoms under natural field conditions in all 13 weed species that had never been reported as hosts ofV. dahliae were similar to those observed after their artificial inoculation. Dry fungal biomass ofPenicillium chrysogenum (dry mycelium), a waste product of the pharmaceutical industry, was extracted with water and applied to the roots of melon plants before or after inoculation withFusarium oxysporum f.sp.melonis (Font). Seedlings (4–6 days after emergence) treated with either acidic dry mycelium extract (DME) or neutralized dry mycelium extract (NDME) were protected against challenge infection withFom. A single drench with 2–5% DME applied 12–72 h before inoculation provided significant control of the disease compared with water-drenched, challenged seedlings. No protection was seen in plants treated 0–6 h before inoculation or 0–48 h after inoculation. Neither DME nor NDME (0.5–5%) had any effect on fungal growthin vitro, which implied that disease controlin vivo was mediated by induced resistance. The resistance induced by DME protected melon plants not only against race 1,2, but also against the three other races of the pathogen, indicating a race-non-specific resistance againstFom. Both DME and NDME significantly increased peroxidase activity and free L-proline content in seedlings 12 h and 48 h after soil drench, respectively. Resistance to Fusarium wilt was significantly associated with elevated levels of peroxidase activity but not with free L-proline content. Anthracnose, caused by Colletotrichum spp., is a major disease of cultivated strawberry, Fragaria × ananassa. This study identifies the Colletotrichum spp. which causes strawberry anthracnose in the southwest of Spain. A survey of the region was carried out, and the strains isolated were identified as C. acutatum by using the polymerase chain reaction (PCR) with genus and species-specific primers, demonstrating that this species is currently the causal agent of strawberry anthracnose in the studied region. The pathogenicity of C. acutatum and C. gloeosporioides strains was evaluated on two principal strawberry cultivars (cvs Camarosa and Ventana) under field conditions, the latter being more pathogenic than the former.  相似文献

Aeciospores in Uromyces fabae were found to be repeating spores and play an important role in pea rust outbreaks in the North Eastern Plain Zone (NEPZ) of India. Experiments conducted on pea rust from 2001 to 2004 revealed the dominance of aeciospores at all growth stages of pea in this region. Urediospore production was erratic and was only observed in a few samples of stems and tendrils (5–10%). Inoculation of pea plants either by aeciospores or urediospores resulted in the production of aeciospores. Production of aeciospores was observed at a temperature range of 10–25 °C, with a maximum at 25 ± 2 °C. Among the different growth stages of pea, the pod formation stage was highly susceptible and produced the maximum number (744) of aecidia/leaf at 20–25 °C. Significant effects of growth stages and temperature were also noticed for pustule number. Urediospore production mainly coincided with the senescence of the pea plants. Maximum germination (2%) of aeciospores was observed at 25 °C, whereas maximum urediospore germination (3.5%) was at 15 °C. Temperatures > 15 °C decreased urediospore germination. A relative humidity (RH) of 100% was favourable for aeciospore germination while 98% RH favoured urediospore germination. The toxicity of allyl alcohol as a fumigant was assessed for adults ofTribolium castaneum (Herbst.),Rhyzopertha dominica (F.) andSitophilus oryzae (L.). Fumigations were carried out in 3.4–1 glass containers at concentrations of 1–15 mg/l for 3 h. The most sensitive species wasS. oryzae and the most tolerant wasT. castaneum. A dose of 15 mg/l for 3 h was lethal to all three species tested. The feasibility of proposing allyl alcohol as a candidate fumigant is discussed. The rDNA-ITS sequences of ten single-sporangium isolates of Olpidium virulentus (a noncrucifer strain of Olpidium brassicae), which transmits Mirafiori lettuce big-vein virus (MLBVV) and tobacco stunt virus (TStV), were compared with those of six single-sporangium isolates of O. brassicae. The sequence similarity within isolates of O. virulentus or O. brassicae was almost identical (98.5%–100.0%), but was low between the two species (79.7%–81.8%). In a phylogenetic analysis of the rDNA-ITS region, O. virulentus and O. brassicae fell into two distinct clusters, indicating that O. virulentus, a vector of MLBVV and TStV, is a distinct species rather than a strain of O. brassicae.  相似文献

The red palm weevil (RPW,Rhynchophorus ferrugineus (Olivier) (Coleoptera: Curculionidae) is one of the most severe pests of various palm species, including date palms. While examining the susceptibility of RPW to two entomopathogenic fungi,Metarhizium anisopliae andBeauveria bassiana, strains of the former were found to be more virulent than those of the latter, achieving 100% larval mortality within 6–7 days. The most virulent strains ofM. anisopliae were then tested on RPW eggs and adults. Incubation in a substrate treated withM. anisopliae spores increased egg mortality and reduced their hatchability. The total percentage mortality of eggs and hatched larvae was 80–82%, compared with 34% in the controls. RPW adults were challenged with two types of fungal formulation: dry powder and aqueous suspension. Cumulative adult mortality of 100% was achieved in 2–3 weeks for the dry rice-based formulation and in 4–5 weeks for the spore suspension. As a result of decreased longevity, treated females had a shorter oviposition period and three times lower fertility than the controls.
