Black pod, caused by Phytophthora spp. is one of the most important diseases of cacao occurring worldwide. Losses due to black pod caused by P. palmivora are still moderate in Côte d'Ivoire but P. megakarya causes high losses in Ghana and other Central African countries. Variation in field attack has been observed between cacao genotypes, but evaluation of pod losses is unsuitable for obtaining rapid progress in breeding. Results of inoculation tests using young detached leaves, twigs and roots, obtained from field and nursery plants, are presented here and compared to field resistance of similar genotypes observed over a 10-year period. Nine different Upper Amazon Forastero genotypes were tested together with progenies obtained by crossing these with the susceptible check IFC5 (Amelonado genotype). Rank correlations between the early screening tests and the level of field attack were positive and mostly significant (r=0.58–0.95). The coefficient of correlation was slightly higher for leaves (r=0.88) and roots (r=0.89) than for twigs (r=0.76). Also, resistance of the different plant organs was correlated (r=0.6–0.9). Resistance of the Upper Amazon parents was well correlated with the resistance of their cross progenies (r=0.7–0.9), suggesting that resistance is highly heritable. Resistance of leaves and twigs from the nursery was better correlated with field resistance than resistance of leaves and twigs from the field, which might result from more uniform growing conditions in the nursery. Inoculation of leaves appears the most suitable early screening method for black pod resistance. Application of this test in breeding more resistant cacao cultivars is discussed.  相似文献

Thirty-nine genotypes ofCucumis melo (plant introduction entries, open-pollinated cultivars and F1 hybrids) were evaluated for resistance to powdery mildew under either natural field conditions or artificial inoculation in growth chambers at the cotyledonary stage and the 2-true-leaf stage. Results confirmed that susceptibility in cotyledons was not necessarily associated with susceptibility in either true leaves in growth chambers or adult plants in the field. However, resistance at the 2-true-leaf stage in growth chambers was highly correlated with resistance of field-grown plants. Results also showed that 20 muskmelon genotypes resistant to race 1 at the cotyledonary stage were also resistant at the 2-leaf-stage and as adult plants in the field. The same was true for ten genotypes with race 2 inoculations. Because muskmelon genotypes expressing resistance in cotyledons were also resistant in true leaves in growth chambers or the field, the use of plants at the cotyledonary stage is recommended for screening for powdery mildew resistance caused by race 1 or race 2 ofS. fuliginea. When cotyledons are susceptible, screening should be done at the 2-true-leaf stage.  相似文献

It has been suggested that if plants in the field are already induced, their ability to further enhance induced resistance might be compromised. This was examined in barley by inoculating the first two leaves with Rhynchosporium secalis prior to treatment of leaves three and four with an elicitor combination, followed by inoculation with R. secalis. The elicitor combination used consisted of acibenzolar-S-methyl, β-aminobutyric acid, and cis-jasmone, which was shown previously to provide higher levels of disease control in barley than any of the components used individually. The elicitor combination reduced infection by R. secalis, and led to an up-regulation of PR1-b, a marker gene for systemic acquired resistance, and increased activities of the defence-related enzymes cinnamyl alcohol dehydrogenase (CAD), peroxidase (POX), and β-1,3-glucanase. It also led to down-regulation of LOX2, a gene involved in biosynthesis of jasmonic acid. In plants where the first two leaves were inoculated with R. secalis prior to treatment of leaves three and four with elicitor, these increased defence responses did not occur, and control of R. secalis infection on leaves three and four was also reduced. These results suggest that, at least in young barley plants, prior infection with R. secalis compromises their ability to respond effectively to elicitors. The results might help to explain the relatively poor performance of induced resistance in the field, particularly in cereals, compared to plants grown under controlled conditions.  相似文献

A major screening programme was conducted to determine which Lycopersicon spp. (if any) have practical levels of resistance to glyphosate; the same lines were also tested for resistance to Orobanche aegyptiaca. A majority of the lines investigated showed high susceptibility to glyphosate applied at 37?5 g a.i. ha?1 in a spray volume of 250 1 ha?1. Only 41 of 1457 lines screened showed fresh weights of treated plants not significantly different from those of untreated plants at a probability level of 80%. Repeated screening of some selected Lycopersicon lines in the greenhouse and the field, however, indicated that only a few had inherent partial resistance to glyphosate. Giyphosate was less toxic to the test plants with increases in spray volume. Among 1361 Lycopersicon lines tested for resistance to O. aegyptiaca, only minor differences in susceptibility were observed. All lines were susceptible to varying degrees. The chances, therefore, of finding complete resistance in tomato to either glyphosate or Orobanche spp. alone are small. However, combining partial resistance to glyphosate and O. aegyptiaca in a breeding programme to produce new varieties might still prove to be economically useful even in fields heavily infested with the parasite.  相似文献

CAP-34, a protein from Clerodendrum aculeatum inducing systemic antiviral resistance was evaluated for control of Papaya ringspot virus (PRSV) infection in Carica papaya. In control plants (treated with CAP-34 extraction buffer) systemic mosaic became visible around 20 days that intensified up to 30 days in 56% plants. During this period, CAP-34-treated papaya did not show any symptoms. Between 30 and 60 days, 95% control plants exhibited symptoms ranging from mosaic to filiformy. In the treated set during the same period, symptoms appeared in only 10% plants, but were restricted to mild mosaic. Presence of PRSV was determined in induced-resistant papaya at the respective observation times by bioassay, plate ELISA, immunoblot and RT-PCR. Back-inoculation with sap from inoculated resistant plants onto Chenopodium quinoa did not show presence of virus. The difference between control and treated sets was also evident in plate-ELISA and immunoblot using antiserum raised against PRSV. PRSV RNA was not detectable in treated plants that did not show symptoms by RT-PCR. Control plants at the same time showed a high intensity band similar to the positive control. We therefore suggest that the absence/delayed appearance of symptoms in treated plants could be due to suppressed virus replication.  相似文献

The coat protein (CP) gene mediated transgenic resistance is found to be the best approach for protecting papaya plants against the destructive disease caused by Papaya ringspot viruses(PRSV). In order to study the variability of PRSV and the potential threat to the CP-transgenic resistance, five virus isolates were collected from transgenic plants of papaya line 16-0-1, which carry the CP gene of the typical mosaic strain of Taiwan PRSV YK, in an approved test field and fourteen from untransformed papaya plants in different areas of Taiwan. The results of biological, serological, and molecular characterization indicated that all isolates are related to PRSV YK. Among them, the isolate 5--19 from the transgenic line and the isolates CS and TD2 from untransformed papaya were able to overcome the YK CP gene-mediated resistance of papaya lines 18--2--4, 17-0-5, and 16-0-1, which provide high degrees of resistance to different geographic PRSV strains of Hawaii (HA), Mexico (MX), and Thailand (TH). These three isolates were also able to cause symptoms on untransformed papaya plants more severe than those induced by YK. In addition to the host reactions, the variability of the collected 19 isolates was also analyzed and compared with YK and other geographic strains by heteroduplex mobility assay (HMA) and sequence analyses. The results of HMA indicated that the CP genes of isolates 5--19 and TD2 are more divergent than those of other isolates when compared with YK. However, sequence analyses of the transgenic-resistance overcoming isolates 5-19, CS, and TD2 revealed that their CP coding regions and the 3 untranslated regions (UTRs) share nucleotide identities of 93.9–96.6% and 94.2–97.9% with those of YK, respectively; whereas the other geographic strains of HA, MX, and TH that could not overcome the transgenic resistance share lower nucleotide identities of 89.8–92.6% and 92.3–95.3% with those of YK, respectively. Our results indicate that the ability for overcoming the transgenic resistance is not solely correlated with higher degrees of sequence divergence from the transgene. The possible mechanism for overcoming the transgenic resistance and the potential threat of these PRSV strains to the application of the transgenic papaya lines carrying PRSV YK CP gene are discussed.  相似文献

In 2008 and 2009 seasons, a sudden increase in Potato virus Y (PVY) incidence was recorded in foundation seed potatoes in Hokkaido, northern Japan. This increase was obvious during the field inspection and the postharvest indexing. Molecular typing revealed that besides the previously reported strains of PVYO and PVYNA‐N, the most common strain identified was the recombinant PVYNTN, with three characteristic recombinant junctions at the HC‐Pro, VPg and CP regions. No potato tuber necrotic ringspot disease (PTNRD) was observed in foundation seed potatoes in correlation with the presence of PVYNTN. Moreover, an isolate with a typical PVYNTN recombinant genome, namely Eu‐12Jp, did not induce PTNRD in 62 Japanese potato cultivars tested in both primarily and secondarily infected plants. Two cultivars carrying the extreme resistance gene Rychc were resistant to the infection with Eu‐12Jp, which presents potential sources of resistance to PVYNTN. Eu‐12Jp induced systemic mottle in potato cultivars Desiree and King Edward carrying resistance genes Ny and Nc, respectively, but induced a hypersensitive reaction in potato cultivar Maris Bard, with the Nz hypothetical resistance gene typical of the PVYZ strain group. Therefore, based on the genome structure and the reaction of the potato N resistance genes, Eu‐12Jp should be classified as PVYZ‐NTN, as described for isolates from Idaho, USA recently. This is the first report of PVYZ‐NTN in Japan and the sudden and increased occurrence of PVYNTN/PVYZ‐NTN represents a potential risk of PTNRD developing and increases the significance of PVY in Japan.  相似文献

Abstract

A zebra disease resistance factor has been incorporated into the high-yielding Agave hybrid no. 11648 by crossing this hybrid with A. lespinassei. The progeny of three crosses have been screened for resistance to Phytophthora nicotianae, the chief pathogen of zebra disease and results show that 56.7% of the plants have sufficient resistance to be of economic value. The two screening techniques used are described, and the nature of the resistance factor discussed.  相似文献

Physiological and molecular research on resistance responses of Solanum tuberosum cultivars and partially resistant Solanum species to Phytophthora infestans requires a reliable resistance test that can be used in the laboratory. Laboratory tests performed on detached leaves and intact plants were compared with field tests for similarity of late blight reactions. Detached leaves from field-grown plants were as resistant as detached leaves from climate chamber-grown plants when challenged with P. infestans. However, detached leaves incubated in covered trays at high relative humidity were more susceptible than detached leaves kept in open trays or leaves on intact plants. The incubation conditions of detached leaves in covered trays rather than detachment itself appeared to affect the resistance expression. Detached leaves of some wild Solanum genotypes became partially infected, whereas intact plants were completely resistant when inoculated. Inoculation of leaves on intact plants, however, resulted in lower infection efficiencies. These limitations should be taken into account when choosing the appropriate inoculation method for specific purposes. For resistance screening, laboratory tests proved to be a good alternative for field tests. The ranking of resistance levels for twenty plant genotypes was similar under laboratory and field conditions.  相似文献

Evolution of resistance to herbicides in weeds is becoming an increasing problem worldwide. To develop effective strategies for weed control, a thorough knowledge of the basis of resistance is required. Although non‐target‐site‐based resistance is widespread, target site resistance, often caused by a single nucleotide change in the gene encoding the target enzyme, is also a common factor affecting the efficacies of key herbicides. Therefore, fast and relatively simple high‐throughput screening methods to detect target site resistance mutations will represent important tools for monitoring the distribution and evolution of resistant alleles within weed populations. Here, we present a simple and quick method that can be used to simultaneously screen for up to 10 mutations from several target site resistance‐associated codons in a single reaction. As a proof of concept, this SNaPshot multiplex method was successfully applied to the genotyping of nine variable nucleotide positions in the CT domain of the chloroplastic ACCase gene from Lolium multiflorum plants from 54 populations. A total of 10 nucleotide substitutions at seven of these nine positions (namely codons 1781, 1999, 2027, 2041 2078, 2088 and 2096) are known to confer resistance to ACCase‐inhibiting herbicides. This assay has several advantages when compared with other methods currently in use in weed science. It can discriminate between different nucleotide changes at a single locus, as well as screening for SNPs from different target sites by pooling multiple PCR products within a single reaction. The method is scalable, allowing reactions to be carried out in either 96‐ or 384‐well plate formats, thus reducing work time and cost.  相似文献

Three cultivars of white cabbage with different levels of resistance toMycosphaerella brassicicola were tested for seedbed infection. Seedlings grown in seedbeds, to which infected plant debris was added as an inoculum, showed typical ring spot lesions on the cotyledons and first two leaves before seedlings reached the transplanting stage, whereas non-inoculated controls had few lesions only. Differences in levels of resistance between cultivars were present in seedlings grown under field conditions. Disease severity of transplants at the end of the season reflected disease severity of seedlings before transplanting in each cultivar.To lower the risk of a severe epidemic of ringspot at the end of the growing season, the seedbed should be protected from infection byM. brassicicola.  相似文献

Resistance to pea bacterial blight (Pseudomonas syringae pv. pisi) in different plant parts was assessed in 19 Pisum sativum cultivars and landraces, carrying race-specific resistance genes (R-genes) and two Pisum abyssinicum accessions carrying race-nonspecific resistance. Stems, leaves and pods were inoculated with seven races of P. s. pv. pisi under glasshouse conditions. For both race-specific and nonspecific resistance, a resistant response in the stem was not always associated with resistance in leaf and pod. Race-specific genes conferred stem resistance consistently, however, there was variability in the responses of leaves and pods which depended on the matching R-gene and A-gene (avirulence gene in the pathogen) combination. R2 generally conferred resistance in all plant parts. R3 or R4 singly did not confer complete resistance in leaf and pod, however, R3 in combination with R2 or R4 enhanced leaf and pod resistance. Race-nonspecific resistance conferred stem resistance to all races, leaf and pod resistance to races 2, 5 and 7 and variable reactions in leaves and pods to races 1, 3, 4 and 6.Disease expression was also studied in the field under autumn/winter conditions. P. sativum cultivar, Kelvedon Wonder (with no R genes), and two P. abyssinicum accessions, were inoculated with the most frequent races in Europe under field conditions (2, 4 and 6). Kelvedon Wonder was very susceptible to all three races, whereas P. abyssinicum was much less affected. The combination of disease resistance with frost tolerance in P. abyssinicum enabled plants to survive through the winter. A breeding strategy combining race-nonspecific resistance derived from P. abyssinicum with race-specific R-genes should provide durable resistance under severe disease pressure.  相似文献

Xanthomonas fragariae spreads in symptomlessly infected strawberry plantlets and a method for detection of latent infections is necessary. It is a very slow‐growing bacterium in culture and is easily overgrown by saprophytic bacteria. Therefore, plating is not a suitable method for detecting low numbers of bacteria in symptomless plants. In addition, selective media are not available. Serological assays like immunofluorescence are useful for testing in‐vitro plants, but they are not suitable for field‐grown plants, as cross reactions are common with the available antisera. For these plants, nested PCR with primers from Pooler ( Pooler et al., 1996 ) and Zimmermann ( Zimmermann et al., 2004 ) has proved to be a valuable method. The method was successfully applied for a survey of strawberry plants from fields in Germany and for testing imported plants (frigo and green plants).  相似文献

Chocolate spot is an important disease of faba bean (Vicia faba) caused by the necrotrophic fungus Botrytis fabae. The aims of this work were: i) to compare different methods of screening for resistance; ii) to assess the influence of the age of host tissue and temperature on this pathosystem. To this effect, a collection of 42 faba bean accessions was evaluated in mature plant stage in the field in Cordoba (Southern Spain) and in detached leaflet and whole plant tests under controlled conditions. Field results correlated better with those of the whole plant test than with those of the detached leaflet assay. Integration of results from the field and whole plant experiments resulted in the selection of six accessions of interest as sources of resistance. Influence of leaf age on disease development was found to be genotype dependent. Older leaves were more susceptible than younger ones in 23 accessions, while no difference between leaf ages was detected in the remaining accessions. The effects of plant age and temperature were assessed by a whole plant test on seven accessions at two plant ages (4 and 7 weeks) and three temperatures (13, 20, and 25°C). Results showed that the differential genotypic responses to B. fabae were not significantly influenced by either plant age or temperature, although there was a tendency towards lower susceptibility to chocolate spot in faba bean plants as they become older. Further, a partial high-temperature, young-plant resistance was detected.  相似文献

The inheritance of sulfonylurea (SU) resistance in Monochoria vaginalis was investigated based on the bensulfuron‐methyl response phenotypes of F1 plants between SU‐resistant (R) and ‐susceptible (S) and segregation analysis in F2 progenies. Differences of SU resistance between SU‐R biotypes and F1 plants at the recommended field dose were also investigated by comparing shoot dry weight. All F1 plants survived the treatment with 25 g a.i. ha?1 bensulfuron‐methyl, one‐third of the recommended field dose, and showed similar responses to SU‐R plants. Conversely, all F1 plants died or showed extreme necrosis at 225 g a.i. ha?1, three times the recommended field dose, as SU‐S plants. F2 plants were classified as either R or S phenotype. Segregation for resistance to bensulfuron‐methyl in F2 families did not differ from the expected 3:1 (R:S) ratio at 25 g a.i. ha?1. At 225 g a.i. ha?1, the F2 families segregated in a 1:3 (R:S) ratio. These results suggest that SU resistance in M. vaginalis is controlled by a single nuclear allele with resistance being dominant at low dose and susceptibility dominant at high dose. Moreover, F1 plants died or were extremely injured after application of bensulfuron‐methyl at the recommended field dose, although SU‐R biotypes grew normally.  相似文献

We analysed the kinetics of virus distribution in Tobacco ringspot virus (TRSV)-infected tobacco. Infected plants developed asymptomatic leaves above the symptomatic leaves that exhibited ringspots. We detected virus in the shoot apical meristem (SAM). Time-course immunohistochemical microscopy showed that TRSV continuously distributed in the centres of the SAM and in leaf primordia. In contrast, TRSV infection was transient in the root meristem. In western blot and RT-PCR analyses, TRSV infection occurred only within and around the ringspot tissues of the symptomatic leaf; however, TRSV was not or hardly detected in the asymptomatic leaves of inoculated tobacco plants. These results indicate that TRSV persists in the SAM, but that TRSV infection is transient in the root meristem and asymptomatic leaves.  相似文献

The residual toxicity of two selective insecticides, teflubenzuron (acylurea) and Bacillus thuringiensis Berliner ssp. aizawai (microbial), to laboratory and field strains of Plutella xylostella L. was shown in the laboratory to be significantly affected by leaf nutritional status, other host-plant resistance factors, cultivation method and plant age. With plants offering some degree of host-plant resistance, the toxicity of the insecticides was either increased or decreased compared with highly susceptible plants, depending on the specific nature of the plant-herbivore interaction. Differences in residual toxicity of the insecticides varied up to nine-fold on different host plants (=host-plant- + insecticide-induced mortality) despite less than four-fold differences in P. xylostella mortality in controls (=host-plant-induced mortality alone). Host-plant nutritional status also had a substantial effect on the damage potential of larvae. Different response times by P. xylostella to the two insecticides tested on host plants of varying nutritional status were related to the contrasting modes of action of the respective active ingredients. The present studies suggest that insecticides applied to Brassica oleracea L. var. capitata with partial plant resistance may contribute to improved control of P. xylostella. A conceptual model is used to describe likely mortality responses by macrophagous larvae to insecticides applied to plants of varying resistance status. The implications of the findings in relation to the integrated management of P. xylostella are considered.  相似文献

Beet cyst nematode-resistant cultivars, which were introduced recently, originated from the homozygous inbred line B883. This translocation product was unstable and the transmission of resistance when crossed with a susceptible cultivar did not exceed 94%. Tests with the resistant cultivars in climate cabinets showed a wide variety of resistance against Heterodera schachtii and beet necrotic yellow vein virus (BNYVV), expressed as average numbers of infective units per plant and percentages of resistant plants. In a series of field trials at different levels of infection of H. schachtii, their multiplication rates on all resistant cultivars depended on the initial density, which was caused by the presence of small numbers of susceptible plants. Since tolerance to wilting was also incorporated in B883, reasonable yields were obtained in the presence of H. schachtii. However, at increasing initial densities of H. schachtii, yields decreased considerably, since penetrating juveniles cause a hypersensitivity reaction in resistant plants. Based upon the results of three series of field trials, it was concluded that resistant cultivars should preferably be applied at population densities between 500 and 2000 eggs and juveniles of H. schachtii per 100ml of soil. Cultivars with double resistance against H. schachtii and BNYVV behaved like those with H. schachtii resistance in soils infected with beet cyst nematodes, but not with BNYVV. In soils with a combined infection of H. schachtii and BNYVV double resistant cultivars were far superior to single resistant ones, since damage caused by BNYVV was far more serious than damage caused by H. schachtii. No substantial interaction between soil pathogens nor types of resistance could be detected.  相似文献

 西瓜花叶病毒(watermelon mosaic virus,WMV)是危害我国葫芦科作物生产的重要病原。培育和种植抗病品种是防治病毒病最经济有效的措施。本研究利用RT-PCR和血清学方法在表现环斑症状的西葫芦果实上检测到WMV,未检测到番木瓜环斑病毒(papaya ringspot virus,PRSV)。接种WMV侵染性克隆到西葫芦品种绿源冬宝,果实表面产生环斑症状,表明WMV是引起西葫芦果实表面环斑症状的重要病原。利用间接ELISA方法对山东泰安的西瓜、黄瓜、甜瓜、西葫芦和南瓜等46个样品检测发现33个样品表现WMV血清学阳性(检出率71.74%),薛庄采集的带有环斑症状的西葫芦均检测到WMV。对山东省81份葫芦科作物种质资源抗WMV鉴定发现西葫芦品种万盛丰宝、盛丰金珠表现中抗(MR);西瓜品种绿宝新秀和浪潮一号表现中抗(MR);甜瓜品种黄皮面瓜表现抗病(R);黄瓜品种星君贝贝表现中抗(MR);南瓜品种爱维80南瓜表现高抗(HR),蜜本南瓜、传统蜜本南瓜、七叶早南瓜、玲珑二号和绿贝贝迷你南瓜等5个品种表现抗病(R);瓠瓜品种均表现为感病(S)。研究结果对合理布局葫芦科作物品种防控WMV具有重要意义。  相似文献
