The aim of this study was to evaluate the effects of essential oil extracted from aerial parts of Artemisia sieberi in normal and alloxan induced diabetic rats. Fifty rats were divided into five groups of 10 each. Group I normal rats received 1 mL day(-1) of dimethyl sulfoxide (control); group II normal rats received a single dose (80 mg kg(-1) b.wt.) of essential oil extract of Artemisia sieberi; group III diabetic rats received 1 mL day-of dimethyl sulfoxide; group IV diabetic rats received the oil extract (80 mg kg(-1) b.wt.); group V diabetic rats received metformin (14.2 mg kg(-1) b.wt.). All treatments were orally administered once a day for six weeks. Changes in blood glucose concentration, body weight and food and water intake were measured and the data obtained were compared with that of metformin. The essential oil extract significantly (p < 0.05) lowered blood glucose level as well as food and water intake in diabetic rats accompanied by an increase in body weight gain with no apparent side effect when compared with untreated diabetic rats. These effects were found to be closely similar to that of metformin, a common antidiabetic drug. On other hand, no apparent improvement on body weight gain in diabetic rats treated with metformin. In addition, for all parameters measured, the oil extract showed no effect in normal rats. In conclusion, the essential oil of Artemisia sieberi exhibited antidiabetic activity in alloxan-induced diabetic rats. Present findings support the possible use of the essential oil of Artemisia sieberi as a remedy for diabetes mellitus in humans.  相似文献

To investigate hypoglycemic substances from pumpkin, protein-bound polysaccharide was isolated by activity-guided isolation from water soluble substances of the fruits. The protein-bound polysaccharide from pumpkin fruits (PBPP) was identified to consist mainly of polysaccharide (approximately 41.21%) and protein (approximately 10.13%) by anthrone test and Lowry-Folin test. Different doses of PBPP were evaluated for hypoglycemic activity and the effect on serum insulin levels in alloxan diabetic rats. The results indicated that PBPP can obviously increase the levels of serum insulin, reduce the blood glucose levels and improve tolerance of glucose. The hypoglycemic effect of big dose PBPP group (1000 mg/kg body weight) excelled that of small dose PBPP group (500 mg/kg body weight) and antidiabetic agent group. The results suggest that the hypoglycemic effect of PBPP depends on the dose and PBPP possesses the possibility of being developed from a new antidiabetic agent.  相似文献

Urtica dioica L. (Stinging nettle) has already been known for a long time as a medicinal plant in the world. This histopathological and morphometrical study was conducted to determine the effects of the hydroalcoholic extract of Urtica dioica leaves on testis of streptozotocin-induced diabetic rats. Eighteen male Wistar rats were allocated to equally normal, diabetic and treatment groups. Hyperglycemia was induced by Streptozotocin (80 mg kg(-1)) in animals of diabetic and treatment groups. One week after STZ injection (80 mg kg(-1)), the rats of treatment group received the extract of U. dioica (100 mg/kg/day) IP for 28 days. After 5 weeks of study, all the rats were sacrificed and testes were removed and fixed in bouin and after tissue processing stained with H and E technique. Tubular cell disintegration, sertoli and spermatogonia cell vacuolization and decrease in sperm concentration in seminiferous tubules were seen in diabetic and treatment groups group in comparison with control. External Seminiferous Tubular Diameter (STD) and Seminiferous Epithelial Height (SEH) significantly reduced (p < 0.05) in the diabetic rats compared with controls and these parameters in the treatment group were similar to diabetics animals. This study showed that hydroalcoholic extract of Urtica dioica leaves, after induction of diabetes; has no treatment effect on seminiferous tubules alterations in streptozotocin-induced diabetic rats.  相似文献

This study was designed to evaluate the effect of an anthocyanin-rich extract from black rice on hyperlipidemia and insulin resistance in fructose-fed rats. Rats fed fructose diet for 4 weeks exhibited significantly higher plasma insulin levels and lower insulin sensitivity than the control rats fed AIN-93G diet. Dietary supplementation with the anthocyanin-rich extract (5 g/kg of high-fructose diet) prevented the development of fructose-induced insulin resistance. After fructose-induced insulin resistance had been established, 4-week treatment with the anthocyanin-rich extract (5 g/kg of high-fructose diet) or pioglitazone (270 mg/kg of high-fructose diet) ameliorated the glucose intolerance and hyperlipidemia, but the extract failed to reverse the fructose-induced hyperinsulinemia as pioglitazone did. In addition, rats supplemented by the extract exhibited lower oxidative stress than the fructose-fed controls, as indicated by the lower concentrations of plasma thiobarbituric acid reactive substances and blood oxidized glutathione. Overall, these results suggest that the anthocyanin-rich extract from black rice improves certain metabolic abnormalities associated with diets high in fructose.  相似文献

The aim of the present investigation was to examine the effect of Flax Seeds (FS) and Date Palm Leaves (DPL) extracts on the concentration of serum glucose and lipids in alloxan diabetic rats. Rats were divided into six groups, normal control rats (without treatment with either FS or DPL), normal control rats treated with either FS or DPL extract for four weeks, diabetic control rats and diabetic rats treated with either FS or DPL extract for four weeks. The concentration of glucose in diabetic rats treated with FS (D+FS) for four weeks was significantly decreased from 17.20 +/- 2.33 to 8.14 +/- 0.54 mmol L(-1) (p < 0.001). Serum total cholesterol concentration (TC) of diabetic rats treated with either FS or DPL extract showed significant decrease by 40 and 31% respectively in the fourth week. Also, serum low density lipoprotein cholesterol (LDL-C) concentration was significantly decreased after two weeks of treatment (p < 0.001 and p < 0.01, respectively). However, no significant changes were obtained in serum high density lipoprotein cholesterol (HDL-C) concentration. Results of the present study showed that FS extract has a hypoglycaemic effect against alloxan diabetic rats. Present findings also showed that treatment of alloxan diabetic rats with either FS or DPL extract significantly decreased serum concentration of TC and LDL-C. Present findings suggest that both of FS and DPL extracts could have a protective effect against diabetes complications as well as against hyperlipidemia through improvement of lipid profile.  相似文献

BACKGROUND: Teucrium polium is an analgesic, antidiabetic and antilipeidemic herbal medicament. The aim of this survey was to evaluate the effect of aqueous extract T. polium on liver enzymes linked to liver dysfunction, serum lipids and glucose, in diabetic male rats. METHODS: A total of 20 Sprague-Dawly male rats became diabetic by intraperitoneal injection of streptozotocin (60 mg/kg). the animals were divided randomly into two groups. Experimental group was fed Teucrium polium (50 mg/kg) for a month but control group was received the same volume of distilled water. Liver enzymes, biochemical parameters (cholesterol, triglyceride, low density lipoprotein, alanine transaminase, aspartae transaminase) and glucose were measured by kinetic (Enzymatic) and colorimetric methods. Data obtained were analyzed and mean values were compared by paired student's t-test. The results were expressed as mean +/- SD. Significant differences were set at P < 0.05. RESULTS: Our results showed that in test group, serum glucose values decreased significantly (P < 0.05), but cholesterol, triglyceride, low density lipoprotein, alanine transaminase and aspartae transaminase increased significantly after use of T. polium (P < 0.05). This parameters value did not show any changes in control group. CONCLUSION: Although the aqueous extract of Teucrium polium has strong hypoglycemic properties in experimental animals, but because of some hepatotoxic effects, it is not suitable to use it in human as an antidiabetic agent.  相似文献

The aim of this study was to investigate the effect of N. sativa oil on impaired glucose tolerance and insulin insensitivity induced by high-fat diet and trauma. Three dietary groups were used in this study; Rat-Chow (RC), N. sativa oil diet (Combination 4% N. sativa oil and 16% butter oil) (NSOD) and 20% Butter Oil Diet (BOD). Each group was subdivided in two groups; control and trauma. Diets were supplemented for five consecutive weeks body weight increase per week was calculated. At end of the dietary treatments, single dose (2 mL kg(-1) body weight) of turpentine was injected in the dorso-lumber region. Intravenous glucose tolerance test (i.v. GTT) was performed, insulinogenic index and insulin sensitivity was measured. The results showed butter oil diet significantly increased the body weights and visceral fats compared other two groups, respectively. Fasting glucose levels did not change in trauma induced rats while insulin levels increased significantly and it found highest in butter oil diet fed animals. Impaired glucose tolerance was found sever in BOD fed traumatized rats. N. sativa oil diet protected impaired glucose tolerance and insulin insensitivity induced either via saturated fatty acids or injury. In conclusion, N. sativa oil may be used in post surgery diabetic patients to prevent the long going adverse effects from surgical trauma.  相似文献

Parsley is one of the medicinal herbs used by diabetics in Turkey. It has been reported to reduce blood glucose levels. In this study the effects of feeding parsley on diabetes-induced free radical mediated injury in rat aorta and heart tissues were investigated. Swiss albino rats were divided into six groups: Control, diabetic, parsley, diabetic + parsley, glibornurid, and diabetic + glibornurid. Rats were subjected to i.p. streptozotocin (STZ, 65 mg/kg) to induce diabetes. On the fourteenth day of the study, either parsley (2 g/kg) or glibornurid (5 mg/kg) were given for 28 days to the diabetic rats. Aorta and heart tissue lipid peroxidation and glutathione levels as well as blood glucose levels were determined. The results of the present study indicate that lipid peroxidation was increased and glutathione levels were decreased in both aorta and heart tissue of the diabetic rats. However, treatment of the diabetic rats with either parsley or glibornurid reversed the effects of diabetes on blood glucose, and tissue lipid peroxidation and glutathione levels.  相似文献

Background: Data shows vanadium protects pancreatic beta cells (BC) from diabetic animals. Whether this effect is direct or through the relief of glucose toxicity is not clear. This study evaluated the potential effect of oral vanadyl sulfate (vanadium) on glycemic status and pancreatic BC of normal and diabetic rats. Methods: Rats were divided into five groups of normal and diabetic. Diabetes was induced with streptozocin (40 mg/kg, i.v.). Normal rats used water (CN) or vanadium (1 mg/ml VOSO4, VTN). Diabetic rats used water (CD), water plus daily neutral protamine Hagedorn insulin injection (80 U/kg, ITD) or vanadium (VTD). Blood samples were taken for blood glucose (BG, mg/dL) and insulin (ng/dL) measurements. After two months, the pancreata of sacrificed rats were prepared for islet staining. Results: Pre-treated normal BG was 88 ± 2, and diabetic BG was 395 ± 9. The final BG in CD, VTD, and ITD was 509 ± 22, 138 ± 14, and 141 ± 14, respectively. Insulin in VTN (0.75 ± 0.01) and VTD (0.78 ± 0.01) was similar, higher than CD (0.51 ± 0.07) but lower than CN (2.51 ± 0.02). VTN islets compared to CN had larger size and denser central core insulin immunoreactivity with plentiful BC. CD and ITD islets were atrophied and had scattered insulin immunoreactivity spots and low BC mass. VTD islets were almost similar to CN. Conclusion: Besides insulin-like activity, vanadium protected pancreatic islet BC, and the relief of glucose toxicity happening with vanadium had a little role in this action. We sought to determine the ultrastructure of pancreatic islet beta cells of streptozotocin-induced diabetic rats treated with oral vanadyl sulphate. Diabetes was induced in male Wistar rats by intravenous injection of 40 mg kg(-1) streptozotocin. The same volume of normal saline was injected in sham animals. Animals were divided into treated and control groups. Vanadyl sulphate was added to the drinking water of the treated animals at a concentration of 1 mg mL(-1) up to three months. Two months after vanadyl sulphate withdrawal animals were killed. Ultrastructure of islet beta cells were studied by transmission electron microscope. In diabetic treated rats plasma glucose and fluid intake returned to normal levels within three months while control animals remained diabetic. Well granulated cytoplasm, well developed endoplasmic reticulum, increase in the number of immature granules in the cytoplasm with no clear signs of cell injury were found in the islet beta cells of diabetic treated rats. Lymphocyte filteration, nuclear picnosis, cytoplasmic vacuolization were found frequently in the islet beta cells of untreated diabetic rats. In conclusion as was evident in thin sections of panceatic islet beta cells of treated diabetic rats in this study, vanadyl sulphate through preserving islet beta cells structure and ultrastructure contributes in reversing diabetic signs and symptoms in streptozotocin induced diabetic rats.  相似文献

The root of Platycodon grandiflorum (Jacq.) A. DC has been reported to have a wide range of health benefits in oriental food. This study examined the hypoglycemic effects of Platycodon grandiflorum (Jacq.) A. DC aqueous-ethanol extract (PGE) in streptozotocin (STZ) -induced diabetic ICR mice (STZ diabetic mice) for the first time. The effects of PGE on blood glucose, plasma insulin levels and body weight were investigated. A significant decrease in blood glucose levels was observed after single administration of PGE. Furthermore, Glibenclamide and PGE significantly suppressed the rise in blood glucose after 30 min in the acute glucose tolerance test. Treatment with glibenclamide and PGE resulted in a reduction in blood glucose levels from the 2nd week, and this reduction was maintained until the 4th week of treatment. The body weight changed slightly in glibenclamide and PGE treated mice in comparison with the STZ control group. Plasma insulin levels were increased with glibenclamide treatment in STZ diabetic mice, whereas such effect was not observed with PGE. These results indicated that PGE could induce hypoglycemic effects without stimulating insulin secretion.  相似文献

The hypoglycemic effects of hexane, chloroform and methanol extracts from fruits and seeds of Byrsonima crassifolia were evaluated by oral administration to normoglycemic and streptozotocin-induced severe diabetic rats (SD). The anti-diabetic effect was examined by blood glucose, triglycerides, lipid peroxidation, total cholesterol levels in the serum, glycogen content of liver and skeletal muscles, superoxide dismutase (SOD), catalase (CAT), glutathione (GSH) and oxidized glutathione (GSSG) levels. The most active extracts were obtained with hexane. Hexane and chloroform extracts from fruits and seeds of Byrsonima crassifolia increased the levels of SOD, GSH, GSSG and CAT, hepatic glycogen content, glucose-6-phosphatase (G6Pase) and the plasma insulin levels. They also decreased glucokinase (GK) and TBAR (thiobarbituric acid assay). In conclusion, Byrsonima crassifolia possesses significant antihyperglycemic properties after 4 h of a single oral dose. It can also improve hyperlipidemia and hyperinsulinemia in streptozotocin-induced diabetic rats. Both extracts exhibited significant inhibitory activity against AGEs (advanced glycation end products) formation with IC50 values ranging from 94.3 to 138.7 μg/ml. Therefore, B. crassifolia can be considered as a potential safe anti-diabetic agent.  相似文献

The objective of this study was to evaluate the effect of reduced-calorie avocado paste on lipid serum profile, insulin sensitivity, and hepatic steatosis in rats fed a hypercholesterolemic-high fructose diet. Thirty five male Wistar rats were randomly separated in five groups: Control group (ground commercial diet); hypercholesterolemic diet plus 60 % fructose solution (HHF group); hypercholesterolemic diet plus 60 % fructose solution supplemented with avocado pulp (HHF+A group); hypercholesterolemic diet plus 60 % fructose solution supplemented with reduced-calorie avocado paste (HHF+P group); and hypercholesterolemic diet plus 60 % fructose solution supplemented with a reduced-calorie avocado paste plus fiber (HHF+FP group). The A, P, and FP were supplemented at 2 g/kg/d. The study was carried out for seven weeks. Rats belonging to the HHF group exhibited significantly (P?≤?0.05) higher total cholesterol, triglycerides, and insulin levels in serum as well as lower insulin sensitivity than the control group. Supplementation with reduced-calorie avocado paste showed a significant (P?≤?0.05) decrease in total cholesterol (43.1 %), low-density lipoprotein (45.4 %), and triglycerides (32.8 %) in plasma as well as elevated insulin sensitivity compared to the HHF group. Additionally, the liver enzymes alanine aminotransferase and aspartate aminotransferase decreased significantly in the HHF-P group (39.8 and 35.1 %, respectively). These results are likely due to biocompounds present in the reduced-calorie avocado paste, such as polyphenols, carotenoids, chlorophylls, and dietary fibre, which are capable of reducing oxidative stress. Therefore, reduced-calorie avocado paste attenuates the effects of a hypercholesterolemic-high fructose diet in rats.  相似文献

Background: The leaves of Eucalyptus globulus (eucalyptus) are used for treatment of diabetes mellitus in traditional medicine. The aim of this study was to evaluate the effects of eucalyptus in treatment of established systemic infection with Candida albicans in normal and streptozotocin-induced diabetic rats. Methods: Sixty normoglycemic male Wistar rats, weighing 200-250 g, were selected and randomly divided into six groups (n= 10): normal control, control + C. albicans, control + eucalyptus + C. albicans, diabetic control, diabetic + C. albicans, diabetic + eucalyptus + C. albicans. Diabetes was induced after a single intraperitoneal injection of streptozotocin (60 mg/kg body weight) and eucalyptus was added to the diet (62.5 g/kg) and drinking water (2.5 g/L) of treated animals for 4 weeks. The concerned groups were inoculated with C. albicans 15 days after diabetes induction. At the end of one month experiment, fasted rats were killed by cervical decapitation. Blood was collected from neck vein for estimation of glucose. C. albicans concentrations were estimated in liver and kidneys using serial dilution culture of tissue homogenates. Results: Eucalyptus administration significantly improved the hyperglycemia, polydipsia, polyphagia, and it also compensated weight loss of diabetic rats (P<0.05). Moreover, eucalyptus caused a significant reduction in C. albicans concentration in liver and kidney homogenates (P<0.01). Conclusion: The results revealed that eucalyptus improves Candidia infection in normal and diabetic rats that in some ways validates the traditional use of this plant in treatment of diabetic patients. Various clinical observations and experimental data from in vitro studies suggest that insulin and sex hormones interact. The main purpose of the present study was to examine the effects of testosterone and estradiol on serum insulin in rats. Seven week old male and female albino (Wistar) rats were used in our study. Testosterone enanthate (50 mg kg(-1) day(-1)) or estradiol valerate (200 microg kg(-1) day(-l)) were injected intraperiotenally or subcutaneously in orchidectomised or ovariectomised rats, respectively. In orchidectomised rats, serum insulin was decreased compared with control animals (p < 0.01), on the other hand, decreasing of serum insulin was prevented by testosterone replacement (p < 0.001). In ovariectomised rats, serum insulin was also decreased compared with control group (p < 0.01) and decreasing of serum insulin was prevented by estradiol replacement (p < 0.05). Conclusively, present findings indicated that testosterone or estradiol were serum insulin enhancer hormones in male or female rats, respectively.  相似文献

The aim of to study the effect of Ballota undulata (70% EtOH) extract on lipid profile on Rabbits. The plant extract was orally administered to the atherogenic rabbits (atherogenic diet + cholesterol powder supplement 400 mg/kg/body weight/day dissolved in 5 mL coconut oil) at dose of 1.2 g kg(-1) body weight/day. During the overall period of the experiment blood was collected and serum was analyzed for lipid profile. Animals were sacrificed; the heart and the liver were collected and kept at -20 degrees C until assayed. Biochemical analysis of blood serum and tissue (liver and heart muscle) level were made for cholesterol, phospholipids and triglycerides. In addition blood serum was analyzed further for HDL-Cholesterol. All the results were statistically analyzed using students t-test. Hypolipidaemic nature of Ballota undulata (70% EtOH) extract was studied in hyperlipidaemic Rabbits. The increased cholesterol levels were brought to normal by administration of Ballota undulata. Serum cholesterol levels dropped from 940.7 to 230.41 (75.55%) and further to 119.2 (87.32%) by the end of the experiment. Similarly, phospholipids and triglycerides levels were observed to be also reduced. The tissues lipids profiles of liver and heart muscle showed similar changes in those noticed in serum lipids. Ballota undulata possesses active hypolipidaemic constituents.  相似文献

Background: Although effects of trace elements on secretion of sex steroids and insulin have been studied, the effects of these hormones on serum level of trace elements have been rarely investigated. The aim of the present study was to evaluate the effect of testosterone and finasteride administration and castration on serum levels of testosterone, insulin, zinc and chromium. Methods: Male adult rats (n = 32) were divided into 4 groups (n = 8). Group 1, control; Group 2, castration, castration was done at the first day of the study; Group 3, finasteride (20 mg/kg/day, dissolved in drinking water) and Group 4, testosterone (5 mg/kg/day, i.p.). At the end of the period of the study (35 days), serum testosterone, insulin, zinc and chromium levels were determined in the blood samples collected directly from the right atrium of the heart of the animals. Results: The data indicated that the serum levels of testosterone, insulin and zinc were significantly increased (P<0.01) in testosterone-administrated and finasteride groups, but the level of chromium was decreased in both groups (P<0.01). Castrated group had the lowest serum levels of testosterone, insulin and zinc (P<0.05). Also, the levels of serum chromium in this group were increased. Conclusion: The study demonstrates that testosterone and finasteride increases insulin and zinc levels and decreases chromium levels in the serum of male adult rats. According to these data, it seems that testosterone may affect glucose cycle through effect on serum insulin levels and trace elements such as zinc and chromium. The present study was aimed to investigate the combined effects of green tea and vitamin E on heart weight, body weight, serum marker enzymes, lipid peroxidation, endogenous antioxidants and membrane bound ATPases in isoproterenol (ISO)-induced myocardial infarction in rats. Adult male albino rats, treated with ISO (200 mg/kg, s.c.) for 2 days at an interval of 24 h caused a significant (P < 0.05) elevation of heart weight, serum marker enzymes, lipid peroxidation and Ca+2 ATPase level whereas there was a significant (P < 0.05) decrease in body weight, endogenous antioxidants, Na+/ K+ ATPase and Mg+2 ATPase levels. Administration of green tea (100 mg/kg/day, p.o.) and vitamin E (100 mg/kg/day, p.o.) together for 30 consecutive days and challenged with ISO on the day 29th and 30th, showed a significant (P < 0.05) decrease in heart weight, serum marker enzymes, lipid peroxidation, Ca+2 ATPase and a significant increase in the body weight, endogenous antioxidants, Na+/K+ ATPase and Mg+2 ATPase when compared with ISO treated group and green tea or vitamin E alone treated groups. These findings indicate the synergistic protective effect of green tea and vitamin E during ISO induced myocardial infarction in rats.  相似文献

Background:

Oral vanadyl sulfate (vanadium) induces normoglycemia, proliferates beta cells and prevents pancreatic islet atrophy in streptozotocin-induced diabetic rats. Soteriological method is used to quantitate the proliferative effects of vanadium on beta-cell numbers and islet volumes of normal and diabetic rats.

Methods:

Adult male Sprague-Dawley rats were made diabetic with intravenous streptozotocin injection (40 mg/kg). Normal and diabetic rats were divided into four groups. While control normal and diabetic (CD) groups used water, vanadium-treated normal (VTN) and diabetic (VTD) groups used solutions containing vanadyl sulfate (0.5-1 mg/mL, VOSO4+5H2O). Tail blood samples were used to measure blood glucose (BG) and plasma insulin. Two months after treatment, rats were sacrificed, pancreata prepared, and stereology method was used to quantitatively evaluate total beta cell numbers (TBCN) and total islet volumes (TISVOL).

Results:

Normoglycemia persisted in VTN with significantly decreased plasma insulin (0.190.08 vs. 0.970.27 ng/dL, P<0.002). The respective high BG (53249 vs. 14446 mg/dL, P<0.0001) and reduced plasma insulin (0.260.15 vs. 0.540.19 ng/dL, P<0.002) seen in CD were reversed in VTD during vanadium treatment or withdrawal. While the induction of diabetes, compared to their control, significantly decreased TISVOL (1.90.2 vs. 3.030.6 mm3, P<0.003) and TBCN (0.990.1 vs. 3.20.2 x 106, P<0.003), vanadium treatment significantly increased TISVOL (2.90.8 and 4.071.0 mm3, P<0.003) and TBCN (1.50.3 and 3.80.6 x 106, P<0.03).

Conclusion:

Two-month oral vanadium therapy in STZ-diabetic rats ameliorated hyperglycemia by partially restoring plasma insulin. This action was through proliferative actions of vanadium in preventing islet atrophy by increasing beta-cell numbers.Key Words: Vanadium, Pancreas, Islet volumes, Rats  相似文献
