Fetal intrauterine position relative to the sex of adjacent fetuses has an effect on reproductive performance in rodents. An experiment was conducted to determine whether sex of adjacent fetuses in utero has an influence on fetal and placental weights and whether the hormonal mechanisms documented in rodents are similar in fetal pigs. Sows were slaughtered at 70.1 +/- 1.7 d (n = 123) and 104.5 +/- .05 d (n = 135) of gestation. The fetuses and placentas were removed from the uterus and the position and sex of each fetus was recorded to indicate whether the fetus was between two males, two females, or a male and a female. Fetal blood was sampled for later hormonal analysis. At 70 d of gestation, male fetal and placental weights were heavier than those of females (P less than .05), but no differences were detected relative to the sex of adjacent fetuses. At 104 d of gestation, a fetus surrounded on each side in utero by fetuses of the opposite sex (two males or two females) was lighter in weight than a fetus surrounded by fetuses of the same sex (P less than .01). Differences in fetal weight due to the sex of adjacent fetuses were not related to placental function because placental weights were generally not different at 104 d of gestation. By 104 d of gestation, most placentas were not separated by necrotic regions and were in close apposition with surrounding placentas. Gonadal steroid hormones influence an animal's sexual behavior through two primary means. During development they affect differentiation of the brain (primarily in males), and after puberty, circulating concentrations of steroids influence expression of sexual behaviors. In mammals, sexual behaviors of females are regarded as inherent (independent of steroids secreted by the developing ovary). Males, on the other hand, must undergo active differentiation that is brought about by actions of testicular steroids on the brain during discrete sensitive periods of early development. Sexual differentiation in mammals is referred to as processes of defeminization and masculinization. Defeminization is loss of behavioral traits inherent to females and occurs prenatally in sheep but postnatally in swine. Comparative data are lacking for cattle, but preliminary evidence indicates prenatal defeminization of sexual behaviors. Masculinization is acquisition of behavioral traits characteristic of males. Adult females of all three species show mounting behavior after prolonged treatment with testosterone, and in cattle and swine, there is, as yet, no strong evidence that males undergo masculinization of sexual behaviors. In sheep, limited observations are suggestive of two sensitive periods for masculinization; one prenatally and one postnatally. The earlier concept that differentiation of sexual behaviors in mammals occurs prenatally in species with a long gestation and postnatally in species with a short gestation must now be modified in view of recent findings in swine, in which defeminization occurs postnatally. Further studies will likely identify additional species differences with respect to differentiation of sexual behaviors.  相似文献

Pigs of known intrauterine position were obtained from 31 litters by a procedure in which donor sows were slaughtered at d 112 of gestation, their uteri removed and piglets delivered manually. Uterine position was recorded for each piglet as being positioned between two female fetuses (OM), between a male and female fetus (1M), between two male fetuses (2M), between a female fetus and the tip of the uterine horn (OE) or between a male fetus and the tip of the horn (1E). Piglets were fostered as litter groups to recipient sows and reared in these groups until 120 d of age. They then were regrouped and housed as groups of three and six for males and females, respectively. Intrauterine position had no effect on birth weight or survivability of pigs of either sex, although pigs positioned in utero nearest the ovaries (OE and 1E) tended to be heavier at birth. Body weights were similar among groups in each sex at 120 and 175 d of age when given ad libitum access to feed; however, 2M males gained more weight from d 175 to 270 under restricted feeding conditions (P less than .05). Intrauterine position had no effect on anogenital distances either at birth or 120 d of age, and predicted testes weights were similar among males from different positions. Semen characteristics at 220 d of age did not appear to vary due to prenatal environment. It is well established that sexual dimorphism exits within the immune system. Females have higher levels of immunoglobulins, greater antibody response to antigens, and higher incidence of autoimmune diseases, such as systemic lupus erythematosus, Grave’s disease, and Hashimoto thyroiditis than males. Spontaneous autoimmune syndromes in mice are more prevalent and of greater severity in females compared with males, and the course of the disease can be modulated by changes in levels of gonadal steroids. A sexual dimorphism is also present in the pituitary-adrenal function: females have higher corticosterone levels and higher corticosteroidogenesis.In the context of the immune-neuroendocrine interactions, we investigated the effects of gonadectomy and sex hormone therapy on endotoxin-stimulated hypothalamo-pituitary-adrenal axis. Whereas endotoxin-induced corticosterone release is invariable throughout the different stages of the oestrus cycle, gonadectomy in both male and female mice leads to enhanced adrenal and immune responses to endotoxin. Interestingly, these enhanced adrenal and immune responses can be completely reversed by testosterone treatment regardless of the sex of the mice. Studies performed over development confirm the role of endogenous testosterone in modulating the endotoxin-induced corticosterone secretion. Indeed, corticosterone response to endotoxin is maximal before puberty when endogenous testosterone levels are low and declines in postpubertal and adult mice.In conclusion, all these data support a sex steroid hormone basis for a neuroendocrine-immunologic sexual dimorphism.  相似文献

The purpose of this study was to determine steroid hormone concentration profiles in healthy intact and neutered male and female dogs. Seventeen intact female dogs, 20 intact male dogs, 30 spayed female dogs, and 30 castrated male dogs were used in this study. Serum samples were collected before and 1h after cosyntropin administration, and serum concentrations were determined for cortisol, progesterone, 17-OH progesterone (17-OHP), dehydroepiandrosterone sulfate (DHEAS), androstenedione, testosterone, and estradiol. Intact male dogs had greater concentrations of DHEAS, androstenedione, and testosterone. Intact female dogs had greater concentrations of progesterone. There was no significant difference in estradiol concentration among the four groups. Intact male dogs had lower concentrations of cortisol post-stimulation. DHEAS and testosterone did not increase in response to ACTH in intact males, and estradiol concentrations did not increase in response to ACTH in any group. Results from this study will enhance interpretation of suspected adrenal and/or gonadal disorders of dogs. Because estradiol concentrations were similar in all groups of dogs, measuring estradiol may not be a useful diagnostic test. Cortisol concentrations for intact male dogs with hyperadrenocorticism may be lower than those of female or neutered dogs.  相似文献

The longitudinal changes in fecal steroid hormone concentrations and sexual behavior in 2 mated/pregnant and 3 non-mated female Hokkaido brown bears were investigated during the breeding season. Behavioral estrus (standing) lasted for 14 and 32 days in the mated females and for 25 to 36 days in the non-mated females. In non-mated females, sexual behavior, such as female-female mounting and masturbation, was observed for several days before and after the estrous period. In mated females, mean fecal estradiol-17beta concentrations were higher in the estrous period than in the post-estrous period, while fecal progesterone concentrations were higher in the post-estrous period than in the estrous period. The similar trends of steroid hormone changes were observed in the non-mated females.  相似文献

Two experiments were conducted to determine the response of Creole male goats treated with long days and melatonin implants, and the response of the anovulatory does to male effect using males treated only with artificially long days. All animals were allocated to open sheds. In Exp. 1, one group of males was under natural photoperiod (CG; n = 7); the second group was submitted to 2.5 mo of long days followed by the insertion of two s.c. melatonin implants (LD+MEL; n = 7); the third group was subjected only to 2.5 mo of long days (LD; n = 7). Testicular weight was measured every 2 wk. Plasma testosterone concentrations were determined weekly. A treatment x time interaction was detected (P < 0.001) for testicular weight and plasma testosterone concentration. In the LD+MEL and LD groups, testicular size and plasma testosterone levels varied in a similar way, but differed from those observed in CG (P < 0.001). In this latter group, testicular weight displayed seasonal variations and peaked in June, whereas in treated groups this peak occurred in March. In CG, testosterone varied in a seasonal manner and plasma concentrations increased in June and remained elevated throughout the study. In experimental groups, testosterone increased in February and peaked in March. In Exp. 2, one group of males was left under natural photoperiod (CG, n = 5) and the other one was submitted to 2.5 mo of artificially long days (LD, n = 4). On March 16, two control and two treated males were put in contact with 20 and 19 females, respectively. Sexual behavior of the bucks was observed during the 5 d following male introduction. Progesterone assays and estrous behavior were used to determine ovarian and behavioral responses of the females to teasing. The anogenital sniffing, nudging, and mount instances registered in LD-treated males were greater than those observed in CG (P < 0.05). Of the does exposed to CG, none ovulated and only two of 20 females displayed estrous behavior. All does in contact with LD-treated males ovulated and showed at least one estrous behavior during the 15 d following joining (P < 0.001). These results indicate that the sexual activity of male goats from subtropical latitudes can be induced using only artificially long days. In addition, males treated in this way are capable of stimulating sexual activity in anovulatory females by the male effect.  相似文献

During the breeding season three experiments were conducted to evaluate the LH and testosterone (T) response of rams with male sexual orientation (e.g., male-oriented homosexual rams) to female sheep, to male sheep, and to treatment with LHRH. Male-oriented rams were identified through a series of sexual performance and sexual preference tests. Treatments included exposure to estrous females and to males for 15 min (Exp. 1) and exposure to estrous females and to males for 8 h (Exp. 2). Behavioral responses to stimulus animals were recorded. In Exp. 2 homosexual rams mounted males more than females (P less than .02) and exhibited more flehmen (P less than .002) and investigatory sniffs (P less than .01) when exposed to males vs females. Acts of aggression (butting the stimulus animals) did not differ by gender (P greater than .1). Flehmen and butting were positively correlated to LH secretion (P less than .02) of rams exposed to females but not to males. In Exp. 1, LH concentration determined every 15 min for 6 h was not affected (P greater than .05) by the gender of the stimulus animal. In Exp. 2, LH pulse frequency and concentration were similar (P greater than .05) by treatment. In the present study, we investigated the sexual dimorphisms, effects of castration and sex hormone treatment, and strain differences in the morphology of mouse kidney by measuring the percentage of renal corpuscles with cuboidal cells in the parietal layer of the glomerular capsule, the diameter of renal corpuscles and the number of nuclei of epithelial cells in the proximal convoluted tubule. In ICR mice, the percentage of renal corpuscles with cuboidal cells and the diameter of renal corpuscles were larger in males than in females, and the numbers of nuclei were lower in males than in females. All of these parameters became similar to those of intact females by orchiectomy, and restored to levels similar to those in intact males after testosterone treatment to orchiectomized males. On the other hand, ovariectomy and estradiol treatment gave no effects to any of these parameters. Similar sexual dimorphisms were observed in BALB/c, C57BL/6, C3H/He and DBA/2 mice as in ICR mice, although the diameter of renal corpuscles was not significantly different between males and females in BALB/c mice. Strain differences were observed in each of these three parameters among the 5 strains. In conclusion, the present study demonstrated sexual dimorphisms, effects of castration and sex hormone treatment, and strain differences in the morphology of mouse kidney by morphometrical analysis.  相似文献

Gender identification of great sturgeon during different life stages was conducted using morphology, sex steroid concentrations, histological and endoscopic techniques. One hundred and forty great sturgeons from two sturgeon farms at ages between 3‐16 years were selected for examination. Most of the morphological indices showed no significant sex‐related differences except for the ratio of fork length to distance of snout to anterior of dorsal fin (PD). The urogenital opening had no difference in shapes for both sexes. Mean sex steroid concentrations showed no significant difference in levels of testosterone, estradiol and progesterone between males and females in the stage II of maturity. 95.8% of fish were correctly sexed using endoscopy at this stage. Histological investigation indicated the ovaries of 3‐ to 9‐year‐old females were at the different stages of pre‐vitellogenic, and most males were at the early spermatogenesis for age 3. The result of this study suggests that endoscopy is a reliable tool for sex and stage determination after the age of 3 years. This method is useful in providing a quick discernment of sexes and stages compared with utilizing morphometric and blood indices. Direct observation of gonads can give us important information about the gonadal and maturity stages in sturgeon aquaculture and wild population management, with minimal damage or stress to this endangered species.  相似文献

Two experiments were conducted to evaluate the effect of prenatal testosterone treatment in combination with postnatal steroid implantation (Exp. 1) and to assess the effect of time of prenatal testosterone treatment in conjunction with postnatal steroid implantation (Exp. 2) on animal performance and carcass characteristics. In Exp. 1, seventy-six pregnant cows were assigned randomly to a control group or implanted with testosterone propionate (TP) silastic implants between d 40 and 80 of gestation. Half the heifer calves were selected randomly to be implanted with 200 mg TP plus 20 mg estradiol benzoate (EB); the other half of the steer calves were implanted with 200 mg progesterone plus 20 mg EB on d 1 and 85 of the feedlot trial. Daily gain of heifers was increased 10.4% (P less than .08) due to prenatal testosterone treatment (P) and 16.4% (P less than .05) by postnatal steroid implantation (I). Feed efficiency was 12.9% greater (P less than .05) due to P and 9.5% greater (P less than .05) due to I. Prenatal testosterone treatment decreased (P less than .05) kidney, pelvic and heart fat and final yield grade but increased (P less than .05) ribeye area of heifers. Heifers had greater (P less than .07) liver weights per unit of carcass weight due to P. In Exp. 2, one hundred seventy-four pregnant cows were assigned randomly to a control group or implanted with TP silastic implants on d 42, 63, 84 or 105 of gestation. Half the heifer and steer calves were selected randomly to be implanted on d 1 and 112 of the feedlot trial. Time of P caused a quadratic effect (P less than .08) on birth weight of heifers. There was a quadratic effect (P less than .05) of time of P on daily gain and final weight per day of age of heifers. Feed efficiency of heifers was improved (P less than .05) due to P. Postnatal steroid implantation increased (P less than .05) daily gain and feed efficiency of heifers by 9.6% and 8.6%, respectively. No changes were observed in growth performance of steers due to P. Results from these two trials suggest that the combination of prenatal testosterone treatment and postnatal testosterone and estradiol implantation produced an additive improvement of daily gain, feed efficiency and carcass merit of heifers.  相似文献

Serum testosterone and estradiol 17-beta concentrations, and serum testosterone-to-estradiol ratio were evaluated in 15 dogs (greater than or equal to 5 years old) with perineal hernia (9 sexually intact males and 6 castrated males) and in 9 clinically normal sexually intact male dogs greater than or equal to 5 years old. There was no significant difference in serum testosterone-to-estradiol ratio between sexually intact male dogs with perineal hernia and clinically normal sexually intact male dogs. In castrated dogs with perineal hernia, serum testosterone concentration and testosterone-to-estradiol ratio were significantly (P less than 0.05) lower, compared with those values in sexually intact dogs with perineal hernia and in clinically normal sexually intact male dogs. There was no significant difference in serum estradiol 17-beta concentration among sexually intact male dogs with perineal hernia, castrated dogs with perineal hernia, and clinically normal sexually intact male dogs. Serum testosterone and estradiol 17-beta concentrations in dogs with perineal hernia did not differ from those values in clinically normal male dogs of the same age. Castration cannot be recommended for the treatment of perineal hernia unless a castration-responsive contributing factor such as prostatomegaly is identified, unless the pelvic diaphragm of dogs with perineal hernia has high sensitivity to normal or low serum testosterone and estradiol 17-beta concentrations, or unless there is documentation that other androgens and/or estrogens are involved.  相似文献

The enzyme 20alpha-hydroxysteroid dehydrogenase (20alpha-HSD) catabolizes progesterone into a biologically inactive steroid, 20alpha-dihydroprogesterone (20alpha-OHP). In the corpora lutea of rats and mice, 20alpha-HSD is considered to be involved in functional luteolysis. It is also distributed in other tissues including the placenta, endometrial epithelia and fetal skin, although the roles it plays in these tissues remain to be elucidated. In the present study, we investigated the role of 20alpha-HSD in the maintenance of pregnancy using mice with targeted disruption of the 20alpha-HSD gene. We first confirmed that the number of pups was significantly smaller in 20alpha-HSD-/- pairs than in 20alpha-HSD+/+ pairs. We then mated 20alpha-HSD+/- males and females so that each pregnant female produced 20alpha-HSD+/+, 20alpha-HSD+/- and 20alpha-HSD-/- offspring. The genotype ratio of the offspring did not match the Mendel's law of inheritance, and the numbers of 20alpha-HSD+/- and 20alpha-HSD-/- offspring were smaller than expected values. Although the genotype ratio of fetuses on days 13, 15 and 18 of pregnancy matched the Mendel's law, the total number of fetuses on day 18 was significantly smaller than that on day 13, suggesting that fetal loss occurred during late pregnancy. Next, we transferred 20alpha-HSD+/+ embryos to 20alpha-HSD+/+ or 20alpha-HSD-/- females and found that the number of offspring was significantly smaller in 20alpha-HSD-/- dams than in 20alpha-HSD+/+ dams. Expression of 20alpha-HSD mRNA in the fetus, placenta and uterus progressively increased from day 11 to 18 of pregnancy. In addition, concentrations of progesterone were significantly higher in the 20alpha-HSD-/- fetuses than in the 20alpha-HSD+/+ fetuses, while those of 20alpha-OHP were lower in the 20alpha-HSD-/- fetuses than in the 20alpha-HSD+/+ fetuses. These results suggest that both maternal and fetal 20alpha-HSD play a role in maintaining normal pregnancy at least partially by reducing progesterone concentrations in fetuses.  相似文献

The fossa is an endangered, mongoose-like carnivore species endemic to Madagascar with a breeding season (BS) in the southern hemisphere spring. For the present study, faecal samples of captive fossas were collected for over 1 year for five males and four non-pregnant females, and for two pregnant animals during the reproductive period. The goal was to assess gonadal activity using non-invasive hormone measurements of faecal testosterone (T) and gestagen metabolites using assays previously established in our laboratory and further validated in this study. All study animals were housed in northern hemisphere zoos. In males, the seasonal T metabolite profile revealed a peak in spring (March). High performance liquid chromatography (HPLC) analysis was used to characterize the faecal hormone metabolite composition. The highest immunoreactivity was detected in the position of dihydrotestosterone, whereas native T was not detected. In the two pregnant females, gestagen metabolite concentrations increased 4–9 days after the initial matings and remained elevated throughout gestation with concentrations dramatically higher than those of the non-pregnant females during the BS. In these females, gestagen metabolite analyses did not reveal a seasonal pattern similar to the males. The HPLC-analysis revealed that the major proportion of immunoreactivity was associated with an unknown metabolite, whereas native progesterone was undetectable. The seasonal hormone pattern of male fossas gives proof of the reproductive seasonality of this species.
The elevated 5α-pregnan-3β-ol-20-one levels in pregnant animals allows for the characterization of pregnancy in the fossa based on analysis of faecal steroid metabolite concentrations.  相似文献

Northern fur seals (Callorhinus ursinus) have a distinct life history pattern comprising annual terrestrial breeding and oceanic migration, and the physiological changes associated with these patterns are of particular interest for understanding their environmental adaptations. However, owing to their oceanic distribution, limited information is available on the reproductive physiology of wild individuals during the immature stage and the winter migration period. This study aimed to determine the relationships among the seasonal hormone profiles, body growth, age, and pregnancy using monthly serum samples collected over 3–5 years from two male and two female captive individuals during pubescence and sexual maturation. Small increases in the serum testosterone signaled puberty in males aged 3 and 4 years. Thereafter, males showed considerable increases in testosterone during breeding seasons, indicating sexual maturity. Immature female serum progesterone was maintained at low levels, but after pubescence, females showed an increase in serum progesterone in August, the month next to the peak of delivery, followed by a decrease. In non-pregnant females, progesterone did not increase significantly until the next breeding season, but in pregnant females, they increased again from February to March and then gradually decreased. Immature males increased body mass constantly and reached puberty when their body mass exceeded 20 kg, and they showed seasonal weight fluctuations after puberty. These results provide fundamental information for determining sexual maturity and pregnancy in this species based on sex steroid hormones and body mass measurements.  相似文献

Data were collected from intact males, castrated males and ewe lambs to investigate the effect of presence or absence of testosterone prenatally and during the postweaning period on postweaning growth, feed intake and carcass chemical composition. Half the lambs from each sex were the progeny of dams that had received five injections of testosterone cyprionate from d 32 through d 87 of gestation. Linear contrasts were used to detect differences. Postweaning daily gain of intact males was greater (P less than .01) than that of male castrates. Ewe lambs from treated dams had approximately 12% greater rate of growth (P less than .04) than ewe lambs from control dams. Ewe lambs from dams that had been treated were 28% more efficient (P less than .01) in the conversion of food to weight than those from untreated dams. Ewe lambs from treated dams had heavier livers (P less than .07). Carcass protein for intact males was greater (P less than .11) than for castrates, and extractable fat was less (P less than .05). Masculinization of growth characteristics of ewe lambs affected the quantity of carcass fat relative to control ewes (7.59 vs 8.92 kg). These ewe lambs also had more water in the carcass than did the control ewes (13.93 vs 12.29 kg). Administration of exogenous testosterone to pregnant ewes over an interval of time approximating time of sexual differentiation in the fetus enhances postweaning growth rate, feed conversion efficiency and chemical composition of genetic females.  相似文献

In this study, the annual cycle of the gonadal steroids testosterone (T), 11‐ketotestosterone (11‐KT), 17β‐estradiol (E2) and 17α, 20β‐dihydroxy‐4‐pregnen‐3‐one (DHP) was determined using radioimmunoassay and then compared for two populations of rainbow trout, XX diploid females (n = 40) and XXX triploid females (n = 15). In females, E2 and DHP levels were found to be significantly related to body weight (r = 0.22513; p < 0.0001 and r = 0.15831; p > 0.001, respectively). In this group, E2 concentrations peaked in November (25.05 ng /ml), while maximum DHP levels, only measurable from October to April, were attained in February (64.14 ng /ml). No significant differences in hormone ranges related to egg output ability were observed. Finally, sex steroid concentrations were low in the triploid female XXX fish compared to the female XX population. Nevertheless, maximum T (33.85 ng /ml) and 11‐KT (32.35 ng /ml) levels were recorded in January, for XXX. The levels for these two hormones are relatively high and are also significantly associated (r = 0.8430; p < 0.0001). Diploid females showed significantly higher levels of E2 than triploids over the 12‐month study period. The female triploid fish produced the lowest steroid hormone levels, such that these would be the most suitable for human consumption.  相似文献

Postnatal growth, steroid hormone profiles and response to steroid hormone treatment were studied in 18 freemartins and one normal female born co-twin with a bull. They were either treated postnatally with testosterone or estrone at one week of age, or left untreated until 50 weeks of age when they received silastic implants calculated to release either 12.9 microgram of estrone or 2.6 microgram of estradiol per day per kg of body weight. Later a dihydrotestosterone-treated group was added. Reproductive development was studied by palpation per rectum and by examination when animals were slaughtered at 79 weeks of age. Treated animals grew slightly faster than untreated animals. Testosterone in untreated freemartins averaged 76 and 87 pg/ml of blood plasma during weeks 1 to 48 and 52 to 56. Corresponding values for those animals with small testosterone implants (weeks 1 to 48) and with larger implants (weeks 52 to 56) were 130 and 272 pg/ml. Estrone and estradiol values appeared to fluctuate between 10 and 50 pg/ml but values are uncertain because they were below the sensitivity of the assay then available. Thus, circulating steroid hormone concentrations were similar to those reported for castrates. Testosterone stimulated clitoral development prenatally and postnatally. None of the treatments influenced vaginal depth, which averaged 4.0, 9.0 and 10.9 cm at 1, 24 and 52 weeks of age. Vaginal depth at birth was not a reliable indicator of freemartinism. Androgen may have inhibited udder development, whereas estrogen appeared to be stimulatory. The reproductive organs of the freemartin were characterized by differences in underdevelopment and the general presence of seminal vesicles. The latter structures, plus clitoral development at birth in 3 animals and postnatal response of the clitoris to testosterone is interpreted to indicate that the presence of androgens is one factor in abnormal development. 1. Radioimmunoassays were validated for oestradiol and testosterone in faecal and plasma samples from domestic fowls. Faecal samples were prepared in phosphate buffer. A delay of up to 24 h between defaecation and the freezing of a dropping did not affect faecal steroid concentrations.

2. Differences in plasma concentrations of testosterone and oestradiol between cockerels and hens were clearly reflected in faecal hormone concentrations. Faecal concentrations of oestradiol were low in males and did not overlap with much higher concentrations in females, whereas there was some overlap between the sexes in faecal concentrations of testosterone.

3. The correlation coefficients (r2) between plasma and faecal steroid concentrations were 0.464 for testosterone and 0.852 for oestradiol (log‐transformed data). The coefficients did not increase when the mean hormone concentrations for several droppings produced by each bird during a collection period were used.

4. Faecal steroid concentrations can be used as a measure of plasma steroid concentrations and hence of gonadal activity in chickens. The resolution of the faecal steroid method is less than if plasma measurements were used, but it offers a practical alternative to blood sampling that is non‐invasive and does not require birds to be handled.  相似文献
