Results are presented of a zero‐discharge marine recirculating system used for the culture of gilthead seabream Sparus aurata. Operation of the system without any discharge of water and sludge was enabled by recirculation of effluent water through two separate treatment loops, an aerobic trickling filter and a predominantly anoxic sedimentation basin, followed by a fluidized bed reactor. The fish basin was stocked for the first 6 mo with red tilapia Oreochromis niloticus × O. aureus at an initial density of 16 kg/m3. During this period salinity was raised from 0 to 20 parts per thousand. Then, gilthead seabream, stocked at an initial density of 21 kg/m3, replaced tilapia at day 167 and were cultured for an additional 225 d. Non steady‐state inorganic nitrogen transformations occurred as a result of these salinity changes. After day 210, the system operated at all times with those water quality parameters considered critical for successful operation of mariculture systems, within acceptable limits. Thus ammonia, nitrite, and nitrate concentrations did not exceed 1.0‐mg total ammonia‐N/ L, 0.5‐mg NO2:‐N/L and 50‐mg NO3‐N/L, respectively. Sulfide levels in the fish basin were below detection limits and oxygen > 6 mg/L after the oxygen generator was added at day 315. Ammonia, produced in the fish basin and to a lesser extent in the sedimentation basin, was converted to nitrate in the aerobic trickling filter. Nitrate removal took place in the sedimentation basin and to a lesser extent in the fluidized bed reactor. Sludge, remaining in the sedimentation basin at the end of the experimental period, accounted for 9.2% of the total feed dry matter addition to the system. The system was disease‐free for the entire year and fish at harvest were of good quality. Water consumption for production of 1 kg of tilapia was 93 L and 214 L for production of 1 kg of gilthead seabream. Additional growth performance data of gilthead seabream cultured in a similar but larger system are presented. During 164 d of operation of the latter system, maximum stocking densities reached 50 kgl M3 and fish biomass production was 27.7 kg/m3. Relatively poor fish survival and growth resulted from occasional technical failures of this pilot system.  相似文献

A 70 days feeding trial was carried out on juvenile gilthead seabream (Sparus aurata) (mean initial body weight: 362 ± 59 g) to evaluate whether the simultaneous replacement of fishmeal and fish oil by vegetable ingredients affected the muscle's proteolytic potential. Fish were fed either a fishmeal/fish oil diet, a fishmeal/vegetable oil diet, a plant protein/fish oil or a plant protein/vegetable oil diet. The use of dietary plant proteins resulted in lower sulphated glycosaminoglycans' content and lower glycogen phosphorylase activity in the muscle, while both plant proteins and vegetable oil increased muscle pH and reduced cathepsin B activity. Our results indicate that high replacement of fish meal and/or fish oil with plant ingredients has a significant impact on early postmortem metabolic processes and proteolytic potential of seabream muscle, especially when replacing both fish meal and fish oil.  相似文献

This study evaluates the effects of the dietary administration of the live yeast Debaryomyces hansenii strain L2 on the immune responses of gilthead seabream for 4 weeks. Cellular immune parameters were measured from serum and head‐kidney leucocytes respectively. The expression levels of immune‐associated genes were quantified by real‐time polymerase chain reaction. In addition, the profile of intestinal microbiota was studied by denaturing gradient gel electrophoresis. The results showed that seabream fed a diet containing D. hansenii had significantly increased cellular immune parameters. The yeast‐supplemented diet up‐regulated the expression of most seabream genes at week 2 and down‐regulated all of them at week 4, except in the head‐kidney. Finally, a reduction in the diversity of the intestinal microbiota was detected in those specimens receiving the yeast‐supplemented diet. These results support the idea that the live yeast D. hansenii strain L2 stimulates the immune system of gilthead seabream.  相似文献

Four isolates obtained from gilthead seabream have been tested for their adhesion to the skin, gill and intestinal mucus of gilthead seabream, and for their ability to interfere with Listonella anguillarum, an important pathogen of farmed gilthead seabream. The ability to adhere to mucus was higher than 7% for all isolates. Three isolates showed an antagonistic effect against some of the pathogenic strains tested. They were assayed to interfere with the attachment of L. anguillarum to the mucus of gilthead seabream. Only two isolates significantly reduced the adhesion of L. anguillarum to all of the mucus assayed under exclusion, competition and displacement conditions. According to the criteria applied, the isolate Pdp11 was selected and its in vivo probiotic potential was assessed by oral administration followed by challenge with the pathogen L. anguillarum. For the feeding trial, a group of 50 gilthead seabreams received a commercial diet supplemented with lyophilized 108 CFU g?1 of this isolate for 15 days. An other group of similar characteristics received the non‐supplemented commercial diet. After the challenge, the mortality of the fish receiving the diet supplemented with the potential probiotic was significantly lower than that observed in the groups of fish receiving the non‐supplemented commercial diet.  相似文献

To date, the mechanisms of inflammation have been poorly studied in fish of commercial interest, due to the lack of development of appropriate experimental models. The current study evaluated a local inflammation triggered by a polymeric carrageenin mixture (a mucopolysaccharide derived from the red seaweed Chondrus crispus) in the skin of gilthead seabream (Sparus aurata). Fish were injected subcutaneously with phosphate-buffered saline (as control) or λ/κ-carrageenin (1%), and skin samples from the injection sites were collected 1.5, 3 and 6 hr post-injection, processed for inclusion in paraplast and stained with haematoxylin–eosin, Alcian blue or periodic acid–Schiff. Furthermore, immunohistochemistry and expression analyses of several cells’ markers and proinflammatory genes were also analysed in samples of the injected sites. Microscopic results indicated an increased number of skin mucus-secreting cells and acidophilic granulocytes in the skin of fish studied at 1.5 hr and 3 hr post-injection with carrageenin, respectively, with respect to the data obtained in control fish. Otherwise, both the gene expression of the non-specific cytotoxic cell marker (granzyme B, grb) and the proinflammatory cytokine (interleukin-1β, il-1β) were up-regulated at 1.5 hr in the skin of fish injected with carrageenin compared with the control fish, whilst the gene expression of acidophilic granulocyte markers (NADPH oxidase subunit Phox22 and Phox40, phox22 and phox40) was up-regulated at 3 and 6 hr in the carrageenin group, compared with the control group. In addition, the gene expression of myeloperoxidase (mpo) was also up-regulated at 6 hr in the skin of fish injected with carrageenin in comparison with control samples. The present results indicate the chronological participation of two important immune cells involved in the resolution of the inflammation in the skin of gilthead seabream.  相似文献

To improve the unnatural fade-pigmented skin of cultivated gilthead seabream, Sparus aurata, (if shown) the present study was initiated. The effects of either red bell-pepper (Capsicum annum) meal or carrot (Daucus carota) meal as a natural dietary carotenoid source, on growth and skin coloration of gilthead seabream growers were investigated. A basal/control diet (D1/CTR) was firstly formulated to contain 48% crude protein and 14% lipids, with no added-carotenoids. With this basal diet, two other test diets were similarly prepared and supplemented each with about 40mg/Kg total carotenoids from either red-pepper meal (D2) or carrot meal (D3). In a feeding trial, fish (mean IW, 94.86±0.3g) were fed one of the three diets (D1, D2, D3), in triplicates for each treatment, for 6 weeks in light-blue background PVC tanks supplied with natural seawater flow. Total carotenoids content of skin was determined spectrophotometerically at initiation and end of the experiment. Neither growth nor feed utilization were significantly (P<0.05) affected by the red pepper-added diet (D2) as compared to CTR diet. However, the carrot fed fish recorded the lowest and significant (P<0.05) weight gain (g/fish) and specific growth rate (SGR, %/d) among dietary treatments. There were no considerable (P>0.05) differences in major nutrients composition between fish fed the experimental diets. Total carotenoids content was significantly (P<0.05) increased, in the skin-opercle area, of fish fed the red pepper diet (D2) as compared to initial fish and to either carrot fed fish or CTR fish. Results have suggested that gilthead seabream can effectively bio-absorb natural carotenoid pigments (mainly capsansin and capsorbin) in red-pepper but not in carrot (mainly β-and α-carotene).  相似文献

A feeding experiment was conducted over 9 weeks with seven groups of 30 (fish per group) unpigmented gilthead seabream, Sparus aurata (L. 1875) (initial mean weight = 145.2 ± 12.3 g). Three experimental diets were prepared by adding to a basal diet free of carotenoid (final pigment content of around 40 mg per kg feed): (i) a biomass of the carotenogenic Chlorella vulgaris (Chlorophyta, Volvocales); (ii) a synthetic astaxanthin; and (iii) a mixture (1:1) of microalgal biomass and synthetic astaxanthin. At 3‐week intervals, five fish were sampled from each tank for total carotenoids analysis in skin and muscle. The carotenoid pigments (total amount = 0.4%) identified in the carotenogenic alga were lutein (0.3%), β‐carotene (1.2%), canthaxanthin (36.2%), astaxanthin, free and esterified forms (55.0%), and other pigments (7.3%). Carotenoid pigments were significantly deposited in the four skin zones studied during the feeding trial: the forefront between the eyes, the opercule, along the dorsal fin and in the abdominal area. In the muscle, regardless of the astaxanthin source, the amount of carotenoids measured was very low (less than 1 mg kg?1) and differences not significant. Moreover, no muscle pigmentation was evident, and there was no variation in the amount of carotenoid analysed in skin tissue, through the trial, for each treatment. It was concluded that supplementing the feed with C. vulgaris would be an acceptable practice in aquaculture to improve the market appeal of the gilthead seabream.  相似文献

Two rearing trials were conducted to evaluate the effects of dietary lupin (LM) and rapeseed (RM) meals in gilthead seabream (Sparus aurata L.) and goldfish (Carassius auratus L.) juveniles. Each plant meal was incorporated at the rate of 200 g kg?1 in two distinct diets, which were compared with a fishmeal‐based diet as control. After 1 month, the two plant diets did not influence the whole body growth, but the digestive systems were affected. The splenosomatic index was reduced with two plant meals in goldfish and with RM in seabream. The hepatosomatic index was only reduced in LM‐fed seabream. Cellular characteristics were also affected. The largest liver cells were observed in RM‐fed goldfish suggesting changes in metabolic function. The LM and RM diets stimulated in seabream, especially the reaction in haematopoietic tissues with the proliferation of melano‐macrophages centres, and a tendency for elongated villus height in the anterior intestine thus that possibly compensated for a reduction in digestive function. Such adaptive structural modifications and the absence of degenerative signs allowed concluding that the integrity of the digestive system was maintained in fish fed plant meals.  相似文献

The concept of nutritional programming raises the interesting possibility of directing specific metabolic pathways or functions in juvenile fish, for example, to improve the use of substitutes to fishmeal and oil, and hence to promote sustainability in aquaculture. The aim of the study was to determine effects of early nutritional stimuli of gilthead seabream larvae and check if nutritional programming of gilthead sea bream is possible between 16 days post hatching (dph) and 26 dph. A trial was conducted to determine the effects of early nutritional stimuli of gilthead seabream larvae. Five experimental microdiets (pellet size <250 μm) were formulated containing five different proportions of a marine lipid source rich in long‐chain polyunsaturated fatty acids (LC‐PUFA) and two vegetable lipid sources rich in linolenic and linoleic acids. The results of this study demonstrate that dietary n‐3 LC‐PUFA levels increased larval growth and survival affecting Δ6 desaturase gen (fads2) expression and retinal neurons density. However, the high mortalities obtained along on‐growing in fish fed low n‐3 LC‐PUFA at 16 dph constrained the feasibility of nutritional programming of gilthead seabream during this late developmental window and needs to be further investigated.  相似文献

This study evaluated the ambient noise effect of onshore and offshore aquaculture systems on oxidative status and on some immune parameters in gilthead sea bream (Sparus aurata) juveniles. The experiment was performed for 40 days, projecting two different playlists of acoustic stimuli: offshore aquaculture noise condition that created the typical acoustic field in proximity of an offshore sea cage for fish farming, using a random sequence of quite sea background and boat noises; and onshore aquaculture noise condition that represented the acoustic field inside an open concrete tank for fish farming. A third group without any acoustic projection was used as a control. After 40 days from the beginning of acoustic noise, fish exposed to the offshore aquaculture condition showed significantly higher levels of total oxidant status, lysozyme activity, antiprotease activity and white blood cells together with a lower albumin/globulin ratio in comparison to fish from the control group; whereas fish exposed to the onshore condition showed analytical data placed between the other groups, though never significantly different. These results demonstrate that the offshore aquaculture noise, and in particular the sea soundscape, adversely influences the oxidative status and the immune function of gilthead sea bream determining a mild stress condition that could affect the sea bream welfare.  相似文献

The first attempts at applying aquacultural techniques to sharpsnoutseabream (Diplodus puntazzo) have proved very promising.One of the factors to keep in mind when analysing the viability of a speciesforindustrial farming is that of consumer acceptance of the product, a variablethat can be measured by sensory evaluation tests. To that end, 107 participantsfrom a number of work centres were assessed with hedonic sensory tests usingsharpsnout seabream samples. For reference purposes, the tasting also includedgilthead seabream samples. Participants were asked to fill out a questionnairethat included questions concerning their preferences for any one fish. Weanalysed the sensory attributes of both fish species using a hedonic scale, andalso their acceptability (disposition to purchase) to consumers. The resultsproved fairly encouraging with a view to sharpsnout seabream large-scaleproduction. All attributes tested in the sensory profile analysis werepositively rated. About 60% of the surveyed population would consider buyingsharpsnout seabream at a price similar to that of gilthead seabream (10),and 85% found it to be pleasing to the eye. Aside from flavour, its texture,juiciness and fat level are the most important appraisal attributes. Efforts toimprove the organoleptic characteristics of the species should focus mainly onthese attributes.  相似文献

The transmission of lymphocystis disease virus (LCDV) to gilthead seabream, Sparus aurata L., larvae was investigated using fertilized eggs from a farm with previous reports of lymphocystis disease. LCDV genome was detected by PCR‐hybridization in blood samples from 17.5% of the asymptomatic gilthead seabream broodstock analysed. Using the same methodology, eggs spawned from these animals were LCDV positive, as well as larvae hatched from them. The presence of infective viral particles was confirmed by cytopathic effects development on SAF‐1 cells. Whole‐mount in situ hybridization (ISH) and immunohistochemistry (IHC) showed the presence of LCDV in the epidermis of larvae hatched from LCDV‐positive eggs. When fertilized eggs were disinfected with iodine, no viral DNA was detected either in eggs (analysed by PCR‐hybridization) or in larvae (PCR‐hybridization and ISH). These results suggest the vertical transmission of LCDV, the virus being transmitted on the egg surface. Larvae hatched from disinfected eggs remain LCDV negative during the endotrophic phase, as showed by PCR‐hybridization, ISH and IHC. After feeding on LCDV‐positive rotifers, viral antigens were observed in the digestive tract, which suggests that viral entry could be achieved via the alimentary canal, and that rotifers can act as a vector in LCDV transmission to gilthead seabream larvae.  相似文献

Despite carotenoids and essential fatty acids seem to play important roles in fish reproduction, no studies have yet been conducted to determine the effect of dietary carotenoids on gilthead seabream broodstock performance and their relation as antioxidants with dietary n‐3 highly unsaturated fatty acid (HUFA) levels. In addition, the high cost of synthetic sources of carotenoids is leading to the search for new natural carotenoid sources such as paprika oleoresin. Four experimental diets containing two combined levels of carotenoids from paprika oleoresin (40 and 60 mg kg?1) and n‐3 HUFA (25 and 40 g kg?1) were respectively fed to triplicate groups of gilthead seabream (Sparus aurata) broodfish. Elevation of n‐3 HUFA dietary levels from 25 to 40 g kg?1 significantly improved gilthead seabream broodstock performance in terms of egg viability, hatching rates and fecundity. Besides, it markedly increased egg contents in HUFAs which play important energetic and structural roles and improve embryo development. Both arachidonic acid and eicosapentaenoic acid (20:5n‐3) egg contents were more readily affected by dietary n‐3 HUFA than docosahexaenoic acid. HUFA levels did not caused any negative effect suggesting an optimized content of antioxidants in broodstock diets. Increase in dietary carotenoids from 40 to 60 mg kg?1 increased carotenoid contents in eggs and significantly improved egg fertilization rates suggesting an important sperm cell’s protective role by reducing the risk of lipid peroxidation which is detrimental for sperm motility. The increased inclusion of dietary paprika oleoresin enhanced egg carotenoid deposition and improved fish reproductive performance, denoting the high nutritional value of this product as a source of carotenoids for broodstock of this species.  相似文献

The aim of this study was to determine if algal products rich in DHA or ARA are able to completely replace fish oil in microdiets for marine fish larvae, gilthead seabream and if extra supplementation with EPA may further enhance larval performance. For that purpose, 20 day‐old gilthead seabream larvae of 5.97 ± 0.4 mm mean total length and 0.12 ± 0.001 mg mean dry body weight were fed with five microdiets tested by triplicate: a control diet based on sardine oil; a diet containing AquaGrow® DHA (diet DHA) to completely substitute the sardine oil; a diet containing AquaGrow® ARA (diet ARA); a diet containing both products, AquaGrow® DHA and AquaGrow® ARA to completely substitute the fish oil; and, a diet containing both products, AquaGrow® DHA and AquaGrow® ARA, together with an EPA source. Temperature, air and salinity activity tests were also performed to detect larval resistance to stress. At the end of the experiment, final survivals did not differ among groups. The microorganism produced DHA was able to completely replace fish oil in weaning diets for gilthead seabream without affecting survival, growth or stress resistance, whereas the inclusion of microorganism produced ARA did not improve larval performance. Moreover, addition of EPA to diets with total replacement of fish oil by microorganism produced DHA and ARA, significantly improved growth in terms of body weight and total length. The results of this study denoted the good nutritional value of microorganisms produced DHA as a replacement of fish oil in weaning diets for gilthead seabream, without a complementary addition of ARA. However, dietary supplementation of EPA seems to be necessary to further promote larval performance.  相似文献

There is an increasing trend towards the replacement of fish oil (FO) by vegetable oils in feeds for farmed fish, particularly marine carnivores. Previous researches on partial or complete replacement of FO by alternate vegetable oils for young gilthead seabream (Sparus aurata) were successful. The aim of the present study was to investigate the effects of different dietary vegetable oils on seabream growers' health and liver structure. Two dietary vegetable oil blends (Mix 1 and Mix 2), namely sunflower oil, cottonseed oil and either linseed oil (LO) or soybean oil (BO), were tested as 60% FO substitutes versus the 100% FO control (CTR) diet. Three isonitrogenous (∼47% CP) and isolipidic (18.2% L) diets were fed, twice a day, 6 days a week to apparent satiation, to triplicate groups of seabream growers (mean IW, 130.85 g) in PVC tanks. The growth trial lasted for 20 weeks at an average ambient water temperature of 27.0±1.8°C. At the end of the experiment, fish attained market size (300–400 g) and their health status was evaluated by blood analyses, head kidney prints and liver histology. Haemoglobin and haematocrit contents, erythrocyte (RBC) and leucocyte (WBC) count remained significantly unaffected by any dietary treatments. Despite the apparent relatively higher thrombocyte counts for fish fed vegetable oil diets, especially Mix 2 as compared with those of CTR fish, variations were found to be insignificant (P>0.05). However, a slight deformation in erythrocyte shape was noticed in the blood films of fish fed the vegetable oil diets. Inclusion of either dietary vegetable oil blend had also elevated the number of both developing myelocytes and phagocytes in fish head kidney prints examined compared with those of CTR fish particularly for Mix 2 group. Liver from seabream fed vegetable oils showed slightly swelled hepatocytes due to accumulation of oil droplets in the cytoplasm. In the liver sections investigated ultra‐structurally, nuclei and cytoplasmic organelles were shifted towards the cell periphery. Hepatocytes of fish fed Mix 2 diet showed relative shrinkage and picnotic nuclei, compared with those of CTR fish, suggesting an effect on lipid transport and/or metabolism in fish. The comparative hepatic features between experimental fish groups were described. Accordingly, Mix 1 diet (with LO) was considered better than Mix 2 (with BO) in terms of influence on seabream growers' health status.  相似文献

A trial was conducted to determine the effect of ascorbyl‐2‐monophosphate Na/Ca (AMP‐Na/Ca) on blood chemistry and nonspecific immune response of red sea bream juveniles. Test diets with three levels of AsA (free, 107, and 325 mg/kg diet) were fed to juvenile red sea bream (36.0 ± 1.3 g) two times a day for 3 wk. There were no significant differences in hematocrit, glucose, and blood urea nitrogen. Total cholesterol and triglyceride in plasma of fish fed AsA‐free diet was significantly (P < 0.05) higher than that of fish fed two other diets. There were no significant differences in serum albumin, total bilirubin, and total serum protein. Glutamyl oxaloacetic transaminase in serum of fish fed diets containing 107 and 325 mg of AsA were significantly (P < 0.05) lower than that of fish fed AsA‐free diet. Serum lysozyme activity (LA) of fish fed diets containing 107 and 325 mg of AsA were significantly (P < 0.05) higher than that of fish fed AsA‐free diet. There was no significant difference in mucus LA. The results mentioned above demonstrated that AMP‐Na/Ca is a bioavailable AsA source for red sea bream juveniles. Supplement of more than 107 mg AsA/kg in diets improved blood chemistry and nonspecific immune function of red sea bream juveniles.  相似文献

ABSTRACT

Market integration occurs when prices among different locations or related goods follow similar patterns over time. Current knowledge on market integration between aquaculture and wild-caught fish is based on a small number of species and markets. Most studies show the existence of market integration between wild and farmed conspecifics, with the clear exception of European seabass and gilthead seabream in Southern European countries. In this study, we investigate whether this lack of market integration between wild and farmed conspecifics for European seabass and gilthead seabream in Southern European countries is specific for these species or is representative for the area. Therefore, we investigate the existence of market integration in Spain between wild and farmed conspecifics for a large variety of different species: turbot (Scophthalmus maximus), sole (Solea spp.), blackspot (red) seabream (Pagellus bogaraveo), Atlantic cod (Gadus morhua), and meagre (Argyrosomus regius).  相似文献

Dehydrated lemon peel (DLP) at two levels (1.5% and 3%) was included for 30 days in the diet of gilthead seabream (Sparus aurata L.) and after 15 and 30 days the serum and skin mucus of fish were analysed. In serum, both experimental diets led to an initial decrease in glucose and lactate after which the levels became comparable to the control. Dietary DLP modulated the activity of transaminases, alanine aminotransferase and aspartate aminotransferase. The total antioxidant status showed a progressive increase in relation to levels of inclusion of DLP and time of administration (p < .05). In skin mucus, some biomolecular markers related to general stress, oxidative stress and apoptosis exhibited variations that suggest an adaptive response of fish to the new metabolic situation, resulting from the experimental diet. The inclusion of DLP in the diet seems to positively modulate the central metabolism and welfare of gilthead seabream.  相似文献

Different antibiotic‐based drugs are being used for the treatment of Aeromonas hydrophila infection in rainbow trout, and several studies emphasize the use of medicinal plants as immunostimulants for prophylactic measure against Aeromoniasis disease. However, therapeutic effects of aqueous methanolic extracts of tetra (Cotinus coggygria) against A. hydrophila in rainbow trout were not investigated. Four different concentrations of tetra extract (0 [control], 4, 8 and 12 mg/100 µl) and also two different positive control groups (florfenicol and doxycycline antibiotics) were administered orally using feeding needles to individual rainbow trout, Oncorhynchus mykiss of all experimental groups twice a day after intramuscular inoculation of A. hydrophila. The study period was for 10 days. On 0th, 3rd, 7th and 10th day, blood and tissues were collected from the fish and changes in humoral immune responses, haematology and immune‐related gene expressions were determined. In the study, superoxide radical production was decreased generally in all experimental groups except in 12 mg tetra and florfenicol treatments compared to control (p < .05). Lysozyme activity was generally decreased (p < .05), or no differences were observed in all experimental groups compared to the control. Myeloperoxidase activity was significantly increased in florfenicol‐treated fish group on 7th day (p < .05). Generally, myeloperoxidase activity showed an increase in almost all tetra‐treated groups. Haematological parameters increased but were not significantly high enough in treatments. Almost all immune‐related gene expressions were significantly enhanced on 3rd and 10th day of the study. Survival rate of 53.33% was found in control group. There were no significant differences in survival between control and 4 mg tetra‐treated group (p > .05). All the other groups' survival rate was significantly increased compared to control. The highest survival rate was found in florfenicol group (80%). In 12 mg tetra‐, doxycycline‐ and 8 mg tetra‐treated groups, survival rate was recorded as 74.44%, 70% and 70%, respectively. Our results suggest that tetra methanolic extract is an effective therapeutic remedy against A. hydrophila infection in rainbow trout at the dose of 24 mg/32.34 g body weight/day.  相似文献
