Due to its high sorption affinity for organic compounds, biochar may interfere with extraction procedures involving such compounds used for microbially-related assays commonly applied to soils. Here we assessed the impact of two biochars (derived from pine bark and produced at 300 and 600 °C) at three concentrations (0, 12.5, and 50 g kg−1) in three distinct arable soils with contrasting textural classes (loamy sand, sandy loam, and clay) on the determination of soil microbial biomass C by fumigation–extraction, fungal biomass by ergosterol analysis, and microbial community structure as defined by phospholipid fatty acid (PLFA) profiling. Biochar did not affect the apparent concentration of soil microbial biomass C and had no significant impact on apparent PLFA profiles. By contrast, the apparent extraction efficiency of ergosterol was affected dependent on soil type, biochar production temperature, and biochar concentration. Nonetheless, ergosterol contents of biochar-amended soils can be accurately estimated by correcting for reduced recovery using an ergosterol spike.  相似文献

The dynamics of soil water-stable aggregation (WSA) following organic matter (OM) addition are controlled by microbial activity, which in turn is influenced by carbon substrate quality and mineral N availability. However, the role of microbial communities in determining WSA at different stages of OM decomposition remains largely unknown. This study aimed at evaluating the role of microbial communities in WSA during OM decomposition as affected by mineral N. In a 35-day incubation experiment, we studied the decomposition of two high-C/N crop residues (miscanthus, C/N = 311.3; and wheat, C/N = 125.6) applied at 4 g C kg−1 dry soil with or without mineral N addition (120 mg N kg−1 dry soil). Microbial characteristics were measured at day 0, 7, and 35 of the experiment, and related to previous results of WSA. Early increase in WSA (at 7 days) was related to an overall increase of the microbial biomass (MBC) with wheat residues showing higher values in MBC and WSA than miscanthus. In the intermediate stage of decomposition (from day 7 to 35), the dynamics of WSA were more associated with the dynamics of microbial polysaccharides and greatly influenced by mineral N addition. Mineral N addition resulted in a decrease or leveling off of WSA whereas it increased in its absence. We suggest that opportunistic bacterial populations stimulated by N addition may have consumed binding agents which decreased WSA or prevented its increase. To the contrary, microbial polysaccharide production was high when no mineral N was added which led to the higher WSA in the late stage of decomposition in this treatment. The late stage of decomposition was associated with a particular fungal community also influenced by the mineral N treatment. We suggest that WSA dynamics in the late stage of decomposition can be considered as a « narrow process³ where the structure of the microbial community plays a greater role than during the initial stages.  相似文献

To investigate the uptake by the microbial community of easily decomposable exogenous organic C and the proportion of this organic C remaining in soils under long-term fertilization schemes, 13C-glucose was supplied to arable soils (aquic inceptisol) following a 20-year (1989–2009) application of compost (CM) or inorganic NPK (NPK), along with a control (no fertilizer). Phospholipid fatty acids (PLFAs) were used as biomarkers for actinobacteria, bacteria and fungi. Gas chromatography–combustion–stable isotope ratio mass spectrometry (GC–C–IRMS) was used to determine the incorporation of 13C into individual PLFAs. The concentrations of soil microbial PLFAs significantly (P < 0.05) increased in all three soils after the addition of 13C-glucose. Over a 30-day incubation period, the highest PLFA concentrations were on day 7 (control) or day 15 (NPK and CM) for bacteria, and on day 30 for both fungi and actinobacteria. The added 13C-glucose was incorporated into bacterial PLFAs first, whilst an increase of 13C in fungal and actinobacterial PLFAs was measured on day 7 and 15, respectively. The mean amounts of 13C in bacterial, actinobacterial and fungal PLFAs in CM-treated soil during the 30-day incubation period were 0.589, 0.030 and 0.056 μg g−1 soil, respectively, which were significantly (P < 0.05) higher than levels measured in the NPK and control soils. Among the bacterial groups, the amount of 13C in Gram-positive (G+) bacteria over the entire incubation ranged from 0.326 to 0.440 μg g−1 soil in the CM scheme, which was significantly (P < 0.05) higher than levels detected in the NPK and control regimes. In contrast, 13C concentrations in monounsaturated PLFAs (aerobic microorganisms) in the CM-treated soil were 0.030–0.045 μg g−1 soil, which was significantly (P < 0.05) lower than in the NPK schemes. The proportion of glucose-derived 13C remaining in soils was ranked as follows: CM (53%) > NPK (41%) > control (28%) after 30 days of incubation. Easily decomposable exogenous organic C was thus more effectively maintained under the CM regime, primarily because, after 20 years, CM had altered the microbial community by reducing the ratio of aerobic to anaerobic microorganisms whilst increasing levels of G+ bacteria in soil compared to the control and NPK soils. This study aids our understanding of the transformation and maintenance of easily decomposable organic C in soil over long-term fertilization regimes.  相似文献

In Eastern Spain, almond trees have been cultivated in terraced orchards for centuries, forming an integral part of the Mediterranean forest scene. In the last decades, orchards have been abandoned due to changes in society. This study investigates effects of changes in land use from forest to agricultural land and the posterior land abandonment on soil microbial community, and the influence of soil physico-chemical properties on the microbial community composition (assessed as abundances of phospholipids fatty acids, PLFA). For this purpose, three land uses (forest, agricultural and abandoned agricultural) at four locations in SE Spain were selected. Multivariate analysis showed a substantial level of differentiation in microbial community structure according to land use. The microbial communities of forest soils were highly associated with soil organic matter content. However, we have not found any physical or chemical soil property capable of explaining the differences between agricultural and abandoned agricultural soils. Thus, it was suggested that the cessation of the perturbation caused by agriculture and shifts in vegetation may have led to changes in the microbial community structure. PLFAs indicative of fungi and ratio of fungal to bacterial PLFAs were higher in abandoned agricultural soils, whereas the relative abundance of bacteria was higher in agricultural soils. Actinomycetes were generally lower in abandoned agricultural soils, while the proportions of vesicular–arbuscular mycorrhyzal fungi were, as a general trend, higher in agricultural and abandoned agricultural soils than in forests. Total microbial biomass and richness increased as agricultural < abandoned agricultural < forest soils.  相似文献

Afforestation and deforestation are key land-use changes across the world, and are considered to be dominant factors controlling ecosystem functioning and biodiversity. However, the responses of soil microbial communities to these land-use changes are not well understood. Because changes in soil microbial abundance and community structure have consequences for nutrient cycling, C-sequestration and long-term sustainability, we investigated impacts of land-use change, age of stand and soil physico-chemical properties on fungal and bacterial communities and their metabolic activities. This study was carried out at four sites in two geographical locations that were afforested on long-established pastures with Pinus radiata D. Don (pine). Two of the sites were on volcanic soils and two on non-volcanic soils and stand age ranged from 5 to 20 y. Microbial communities were analysed by biochemical (phospho-lipid fatty acids; PLFA) and molecular (multiplex-terminal restriction fragment length polymorphism; M-TRFLP) approaches. Both site and stand age influenced microbial properties, with changes being least detectable in the 5-y-old stand. Land use was a key factor influencing soil metabolic activities as measured by physiological profiling using MicroResp. Pasture soils had higher microbial biomass (P < 0.001), and metabolic activities (P < 0.001), and basal respiration rates were up to 2.8-times higher than in the pine soils. Microbial abundance analysis by PLFA showed that the fungal to bacterial ratio was higher in the pine soils (P < 0.01). Community analysis suggested that soil bacterial communities were more responsive to site (principal component 1; P < 0.001) than to land use (principal component 5; P < 0.001). In contrast, the fungal community was more affected by land-use change (principal component 1; P < 0.001) than by site, although site still had some influence on fungal community structure (principal component 2; P < 0.001). Redundancy analysis also suggested that bacterial and fungal communities responded differently to various soil abiotic properties, land-use change and location of sites. Overall, our results indicate that the change in land use from pasture to P. radiata stands had a direct impact on soil fungal communities but an indirect effect, through its effects on soil abiotic properties, on bacterial communities. Most of the changes in bacterial communities could be explained by altered soil physico-chemical properties associated with afforestation of pastures.  相似文献

This study aimed to evaluate the interaction between filter cake (FC), and phosphorus fertilizers with differing solubility on the growth and P nutrition of sugarcane. Effects of soil amendment with FC on different soil P fractions and influence on microbial community structure in the rhizosphere were also assessed. Two glasshouse experiments were conducted with completely randomized block designs. The first experiment evaluated rates of FC using a factorial design (5 × 2): 0, 2.5, 5, 10 and 15 g FC kg?1 soil applied as either broadcast in bulk soil or in the planting furrow. The second experiment used a factorial design (2 × 3): without and with FC (5 g kg?1 soil, dry basis), both without P (NP) and with P supplied as either triple superphosphate (TSP) or as rock phosphate (RP), both at the rate of 78.4 mg kg?1 based on total P. Microbial community structure was determined using TRFLP and dynamics of soil P by Hedley fractionation. Filter cake applied at increasing rates in the absence of P was effective in increasing shoot growth and P uptake by plant, particularly when applied to bulk soil as compared to furrow application. Also, FC improved P uptake and increased the availability of labile inorganic P in the rhizosphere and modified the structure of fungal and bacterial communities, whereas only bacterial and archaea communities were influenced by P fertilizer use. Filter cake was more effective when combined with RP, with increased growth and P utilization and thus can be considered as a feasible and practical option for farmer use in substitution to TSP, a more expensive source.  相似文献

The need to identify microbial community parameters that predict microbial activity is becoming more urgent, due to the desire to manage microbial communities for ecosystem services as well as the desire to incorporate microbial community parameters within ecosystem models. In dryland agroecosystems, microbial biomass C (MBC) can be increased by adopting alternative management strategies that increase crop residue retention, nutrient reserves, improve soil structure and result in greater water retention. Changes in MBC could subsequently affect microbial activities related to decomposition, C stabilization and sequestration. We hypothesized that MBC and potential microbial activities that broadly relate to decomposition (basal and substrate-induced respiration, N mineralization, and β-glucosidase and arylsulfatase enzyme activities) would be similarly affected by no-till, dryland winter wheat rotations distributed along a potential evapotranspiration (PET) gradient in eastern Colorado. Microbial biomass was smaller in March 2004 than in November 2003 (417 vs. 231 μg g−1 soil), and consistently smaller in soils from the high PET soil (191 μg g−1) than in the medium and low PET soils (379 and 398 μg g−1, respectively). Among treatments, MBC was largest under perennial grass (398 μg g−1). Potential microbial activities did not consistently follow the same trends as MBC, and the only activities significantly correlated with MBC were β-glucosidase (r = 0.61) and substrate-induced respiration (r = 0.27). In contrast to MBC, specific microbial activities (expressed on a per MBC basis) were greatest in the high PET soils. Specific but not total activities were correlated with microbial community structure, which was determined in a previous study. High specific activity in low biomass, high PET soils may be due to higher microbial maintenance requirements, as well as to the unique microbial community structure (lower bacterial-to-fungal fatty acid ratio and lower 17:0 cy-to-16:1ω7c stress ratio) associated with these soils. In conclusion, microbial biomass should not be utilized as the sole predictor of microbial activity when comparing soils with different community structures and levels of physiological stress, due to the influence of these factors on specific activity.  相似文献

Characterizing functional and phylogenetic microbial community structure in soil is important for understanding the fate of microbially-derived compounds during the decomposition and turn-over of soil organic matter. This study was conducted to test whether amino sugars and muramic acid are suitable biomarkers to trace bacterial, fungal, and actinomycetal residues in soil. For this aim, we investigated the pattern, amounts, and dynamics of three amino sugars (glucosamine, mannosamine and galactosamine) and muramic acid in the total microbial biomass and selectively cultivated bacteria, fungi, and actinomycetes of five different soils amended with and without glucose. Our results revealed that total amino sugar and muramic acid concentrations in microbial biomass, extracted from soil after chloroform fumigation varied between 1 and 27 mg kg−1 soil. In all soils investigated, glucose addition resulted in a 50-360% increase of these values. In reference to soil microbial biomass-C, the total amino sugar- and muramic acid-C concentrations ranged from 1-71 g C kg−1 biomass-C. After an initial lag phase, the cultivated microbes revealed similar amino sugar concentrations of about 35, 27 and 17 g glucosamine-C kg−1 TOC in bacteria, fungi, and actinomycetes, respectively. Mannosamine and galactosamine concentrations were lower than those for glucosamine. Mannosamine was not found in actinomycete cultures. The highest muramic acid concentrations were found in bacteria, but small amounts were also found in actinomycete cultures. The concentrations of the three amino sugars studied and muramic acid differed significantly between bacteria and the other phylogenetic microbial groups under investigation (fungi and actinomycetes). Comparison between the amino sugar and muramic acid concentrations in soil microbial biomass, extracted after chloroform fumigation, and total concentrations in the soil showed that living microbial biomass contributed negligible amounts to total amino sugar contents in the soil, being at least two orders of magnitude greater in the soils than in the soil inherent microbial biomass. The main aim of this study is to analyze the influence of 4-nonylphenol (NP) on soil water retention and biological activity. Two doses of 4-nonylphenol (25 and 50 mg kg−1) were tested in a loam soil with and without peat amendment. In general, one week after the start of the experiment, the soil water content retained at −0.75 MPa of soil suction was 18% higher in the soil amended and its basal respiration (BR) was 15% higher than soil without peat. In contrast, the microbial activity indices (CM: coefficient of mineralization or BR:total organic carbon (TOC) ratio; Cmic:Corg: microbial biomass carbon (MBC):TOC ratio; qCO2: metabolic quotient or BR:MBC ratio) were higher in the soil without peat, compared to the soil amended with peat. On the other hand, the addition of NP to soil was able to modify soil biological but not physical (water retention, desorption) properties. When soil was amended with peat, MBC was reduced one week after applying NP. In contrast, no effects of NP on MBC were observed in the soil without peat. BR was reduced by 16% one week after applying 50 mg kg−1 of NP to soil with peat, and was increased by 46% one week after applying 25 mg kg−1 of NP to soil without peat. The effects of NP on MBC and BR could be associated more with the adsorption of NP by soil organic matter, while changes in CM or Cmic:Corg ratio were more closely related to changes in soil water retention. The potential toxic effects of NP (high qCO2 values) were only observed in the absence of peat amendments. Peat addition reduced NP toxic effects on microorganisms.  相似文献

Many studies have shown that changes in nitrogen (N) availability affect the diversity and composition of soil microbial community in a variety of terrestrial systems, but less is known about the responses of microbes specific to biological soil crusts (BSCs) to increasing N additions. After seven years of field experiment, the bacterial diversity in lichen-dominated crusts decreased linearly with increasing inorganic N additions (ambient N deposition; low N addition, 3.5 g N m−2 y−1; medium N addition, 7.0 g N m−2 y−1; high N addition, 14.0 g N m−2 y−1), whereas the fungal diversity exhibited a distinctive pattern, with the low N-added crust containing a higher diversity than the other crusts. Pyrosequencing data revealed that the bacterial community shifted to more Cyanobacteria with modest N additions (low N and medium N) and to more Actinobacteria and Proteobacteria and much less Cyanobacteria with excess N addition (high N). Our results suggest that soil pH, together with soil organic carbon (C), structures the bacterial communities with N additions. Among the fungal communities, the relative abundance of Ascomycota increased with modest N but decreased with excess N. However, increasing N additions favored Basidiomycota, which may be ascribed to increases in substrate availability with low lignin and high cellulose contents under elevated N conditions. Bacteria/fungi ratios were higher in the N-added samples than in the control, suggesting that the bacterial biomass tends to dominate over that of fungi in lichen-dominated crusts after N additions, which is especially evident in the excess N condition. Because bacteria and fungi are important components and important decomposers in BSCs, the alterations of the bacterial and fungal communities may have implications in the formation and persistence of BSCs and the cycling and storage of C in desert ecosystems.  相似文献

Biochar’s role on greenhouse gas emission and plant growth has been well addressed. However, there have been few studies on changes in soil microbial community and activities with biochar soil amendment (BSA) in croplands. In a field experiment, biochar was amended at rates of 0, 20 and 40 t ha−1 (C0, C1 and C2, respectively) in May 2010 before rice transplantation in a rice paddy from Sichuan, China. Topsoil (0–15 cm) was collected from the rice paddy while rice harvest in late October 2011. Soil physico-chemical properties and microbial biomass carbon (MBC) and nitrogen (MBN) as well as selected soil enzyme activities were determined. Based on 16S rRNA and 18S rRNA gene, bacterial and fungal community structure and abundance were characterized using terminal-restriction fragment length polymorphism (T-RFLP) combined with clone library analysis, denaturing gradient gel electrophoresis (DGGE) and quantitative real-time PCR assay (qPCR). Contents of SOC and total N and soil pH were increased but bulk density decreased significantly. While no changes in MBC and MBN, gene copy numbers of bacterial 16S rRNA was shown significantly increased by 28% and 64% and that of fungal 18S rRNA significantly decreased by 35% and 46% under BSA at 20 and 40 t ha−1 respectively over control. Moreover, there was a significant decrease by 70% in abundance of Methylophilaceae and of Hydrogenophilaceae with an increase by 45% in Anaerolineae abundance under BSA at 40 t ha−1 over control. Whereas, using sequencing DGGE bands of fungal 18S rRNA gene, some bands affiliated with Ascomycota and Glomeromycota were shown inhibited by BSA at rate of 40 t ha−1. Significant increases in activities of dehydrogenase, alkaline phosphatases while decreased β-glucosidase were also observed under BSA. The results here indicated a shift toward a bacterial dominated microbial community in the rice paddy with BSA.  相似文献

The activity of heterotrophic soil microorganisms is usually limited by the availability and quality of carbon (C). Adding organic substances will thus trigger a microbial response. We studied the response in bacterial growth and respiration after the addition of low amounts of glucose. First we determined if additions of glucose, at concentrations which did not result in an exponential increase in respiration after the lag phase, still stimulated bacterial growth. The second aim was to determine the threshold concentration of glucose needed to induce bacterial growth. Adding glucose-C at 1000 μg g−1 soil resulted in an increased respiration rate, which was stable during 12 h, and then decreased without showing any exponential increase in respiration. Bacterial growth, determined as leucine incorporation, did not change compared to an unamended control during the first 12 h, but then increased to levels 5 times higher than in the control. Thus, after the lag phase, a period with increasing bacterial growth, but at the same time decreasing respiration rates, was found. Similar results, but with a more modest increase in bacterial growth, were found using 500 μg glucose-C g−1 soil. Adding 50–700 μg glucose-C g−1 resulted in increased respiration during 24 h correlating with the addition rate. In contrast, bacterial growth after 24 h was only stimulated by glucose additions >200 μg C g−1 soil. Thus, there was a threshold concentration of added substrate for inducing bacterial growth. Below the threshold concentration growth and respiration appear to be uncoupled.  相似文献

This study assessed the effect of mycorrhizal colonization by Glomus intraradices (Gi) and G. versiforme (Gv) on the bacterial community composition in the rhizosphere of canola, clover and two tomato genotypes (wild type (76R) and its mutant with reduced mycorrhizal colonization (rmc)). Additionally, the effect of light intensity on the rhizosphere bacterial community composition of the tomato genotypes was studied. The bacterial community composition was assessed by denaturing gradient gel electrophoresis (DGGE). In canola, which is considered to be a non-mycorrhizal species, inoculation with Gi increased the shoot dw compared to Gv and the non-mycorrhizal control plants and also induced changes in the bacterial community composition in the rhizosphere. These fungal effects were observed although less than 8% of the root length of canola was colonized. On the other hand, about 50% of the root length of clover was colonized and inoculation with Gv resulted in a higher shoot dw compared to Gi or the control plants but the rhizosphere bacterial community composition was not affected by inoculation. Plant growth, mycorrhizal colonization and bacterial community composition of the two tomato genotypes were affected by a complex interaction between tomato genotype, AM fungal species and light intensity. Low light intensity (photosynthetic photon flux 200–250 μmol m−2 s−1) increased the shoot–root ratio in both genotypes and reduced colonization in the wild type. The differences in bacterial community composition between the two genotypes were more pronounced at low than at high light intensity (550–650 μmol m−2 s−1).  相似文献

Veterinary medicines enter agricultural soils by the use of animal excrements as fertilizers. To study their impact on soil bacterial communities, microcosms containing orthic luvisol soil were spiked with the antimicrobial agents sulfadiazine (SDZ) and chlorotetracycline (CTC) at three different concentrations (1, 10, 50 mg kg−1 soil) and incubated for 48 days at 20 °C. The impact on the microbial respiratory activity was measured continuously in a respirometer (Sapromat). Changes in bacterial community structure were visualized by means of PCR-denaturing gradient gel electrophoresis (DGGE) of 16S rDNA derived from soil samples after 1, 7, 11 and 48 days. Additionally, growth inhibitory effects of SDZ and CTC on bacteria previously isolated from the same soil were tested in agar diffusion tests. In microcosms with soil and antibiotics only, no effects could be observed, either on respiratory activity or on bacterial population structure. Therefore, further incubations were conducted in the presence of an additional assimilable carbon source (5 g glucose kg−1 soil). In the presence of glucose, SDZ affected soil respiration as well as the bacterial community structure: Additional bands appeared and some bands already visible at the beginning of incubations increased in intensity. A clear relationship between SDZ concentrations and changes in DGGE patterns became visible. During 48 days of incubation, changes in DGGE patterns were minimal in microcosms with 50 mg SDZ kg−1soil indicating an inhibition of strains, which were capable of growing on glucose in the presence of lower SDZ concentrations. Only a few soil bacterial isolates (5 out of 47 strains tested) were weakly inhibited by SDZ in agar diffusion disk tests. Contrastingly, CTC inhibited growth of 12 soil bacterial isolates significantly in disk tests, but no effects on soil respiration and bacterial community structure could be observed. In the presence of the soil matrix the growth inhibitory potential of CTC decreased due to adsorption or complexation. This was confirmed in growth inhibition experiments with soil suspensions and time-dependent sampling.  相似文献

Identifying amino sugar pools from different land-use systems may advance our knowledge of land-use effects on the fate of microbially-derived substances. Surface soils (0–10 cm) from (1) native pasture, (2) a >80-years-arable site, and (3) a >80-years-afforested site were fractionated into clay, silt, fine-, and coarse-sand fractions. Then, soil organic carbon, N, glucosamine, galactosamine, mannosamine, and muramic acid were analyzed.Afforestation did not influence the amino sugar content in bulk soil, whereas cultivation reduced the content by 54%. The concentrations of amino sugars in g kg−1 SOM declined after both long-term cropping and afforestation by 6% and 13%, respectively, relative to that in the grassland. The amino sugar depletion at the forest site occurred mainly from the silt fraction (by 25%), while that in the cultivated site was mainly due to preferential loss of amino sugars from clay (by 19% compared with the grassland). Both ratios of glucosamine to galactosamine and glucosamine to muramic acid increased when the prairie was converted to forest or cultivated land, suggesting that bacterial N especially is better preserved than fungal N under prairie conditions.  相似文献

Nitrogen (N) is an essential element associated with crop yield and its availability is largely controlled by microbially-mediated processes. The abundance of microbial functional genes (MFG) involved in N transformations can be influenced by agricultural practices and soil amendments. Biochar may alter microbial functional gene abundances through changing soil properties, thereby affecting N cycling and its availability to crops. The objective of this study was to assess the effects of wood biochar application on N retention and MFG under field settings. This was achieved by characterising soil labile N and their stable isotope compositions and by quantifying the gene abundance of nifH (nitrogen fixation), narG (nitrate reduction), nirS, nirK (nitrite reduction), nosZ (nitrous oxide reduction), and bacterial and archeal amoA (ammonia oxidation). A wood-based biochar was applied to a macadamia orchard soil at rates of 10 t ha−1 (B10) and 30 t ha−1 (B30). The soil was sampled after 6 and 12 months. The abundance of narG in both B10 and B30 was lower than that of control at both sampling months. Canonical Correspondence Analysis showed that soil variables (including dissolved organic C, NO3–N and NH4+–N) and sampling time influenced MFG, but biochar did not directly impact on MFG. Twelve months after biochar application, NH4+–N concentrations had significantly decreased in both B10 (4.74 μg g−1) and B30 (5.49 μg g−1) compared to C10 (13.9 μg g−1) and C30 (17.9 μg g−1), whereas NO3–N concentrations increased significantly in B30 (24.7 μg g−1) compared to B10 (12.7 μg g−1) and control plots (6.18 μg g−1 and 7.97 μg g−1 in C10 and C30 respectively). At month 12, significant δ15N of NO3–N depletion observed in B30 may have been caused by a marked increase in NO3–N availability and retention in those plots. Hence, it is probable that the N retention in high rate biochar plots was mediated primarily by abiotic factors.  相似文献

The forest–savanna transition zone is widely distributed on nutrient-poor oxisols in Central Africa. To reveal and compare the nutrient cycle in relation to soil microbes for forest and savanna vegetation in this area, we evaluated seasonal fluctuations in microbial biomass carbon (MBC), nitrogen (MBN), and phosphorus (MBP) for 13 months as well as soil moisture, temperature, soil pH levels, and nutrients for both vegetation types in eastern Cameroon. Soil pH was significantly lower in forest (4.3) than in savanna (5.6), and soil N availability was greater in forest (87.1 mg N kg−1 soil) than in savanna (32.9 mg N kg−1 soil). We found a significant positive correlation between soil moisture and MBP in forest, indicating the importance of organic P mineralization for MBP, whereas in savanna, we found a significant positive correlation between soil N availability and MBP, indicating N limitation for MBP. These results suggest that for soil microbes, forest is an N-saturated and P-limited ecosystem, whereas savanna is an N-limited ecosystem. Additionally, we observed a significantly lower MBN and larger MB C:N ratio in forest (50.7 mg N kg−1 soil and 8.6, respectively) than in savanna (60.0 mg N kg−1 soil and 6.5, respectively) during the experimental period, despite the rich soil N condition in forest. This may be due to the significantly lower soil pH in forest, which influences the different soil microbial communities (fungi-to-bacteria ratio) in forest versus savanna, and therefore, our results indicate that, in terms of microbial N dynamics, soil pH rather than soil substrate conditions controls the soil microbial communities in this area. Further studies should be focused on soil microbial community, such as PLFA, which was not evaluated in the present study.  相似文献

A high level of biological degradation is usually observed in soils under semiarid climate where the low inputs of vegetal debris constraint the development of microbiota. Among vegetal inputs, cellulose and lignin are dominant substrates but their assimilation by the microbial community of semiarid soils is yet not understood. In the present study, 13C-labeled cellulose and 13C-labeled lignin (75 μg 13C g−1 soil) were added to two semiarid soils with different properties and degradation level. Abanilla soil is a bare, highly degraded soil without plant cover growing on it and a total organic C content of 5.0 g kg−1; Santomera soil is covered by plants (20% coverage) based on xerophytic shrubs and has a total organic C content of 12.0 g kg−1. The fate of added carbon was evaluated by analysis of the carbon isotope signature of bulk soil-derived carbon and extractable carbon fractions (water and sodium-pyrophosphate extracts). At long-term (120 days), we observed that the stability of cellulose- and lignin-derived carbon was dependent on their chemical nature. The contribution of lignin-derived carbon to the pool of humic substances was higher than that of cellulose. However, at short-term (30 days), the mineralization of the added substrates was more related to the degradation level of soils (i.e. microbial biomass). Stable isotope probing (SIP) of phospholipid fatty acids (PLFA-SIP) analysis revealed that just a minor part of the microbial community assimilated the carbon derived from cellulose and lignin. Moreover, the relative contribution of each microbial group to the assimilation of lignin-derived carbon was different in each soil.  相似文献

This study investigated the possible effects of tree species diversity and identity on the soil microbial community in a species-rich temperate broad-leaved forest. For the first time, we separated the effects of tree identity and tree species diversity on the link between above and belowground communities in a near-natural forest. We established 100 tree clusters consisting of each three tree individuals represented by beech (Fagus sylvatica L.), ash (Fraxinus excelsior L.), hornbeam (Carpinus betulus L.), maple (Acer pseudoplatanus L.), or lime (Tilia spec.) at two different sites in the Hainich National Park (Thuringia, Germany). The tree clusters included one, two or three species forming a diversity gradient. We investigated the microbial community structure, using phospholipid fatty acid (PLFA) profiles, in mineral soil samples (0–10 cm) collected in the centre of each cluster.The lowest total PLFA amounts were found in the pure beech clusters (79.0 ± 23.5 nmol g−1 soil dw), the highest PLFA amounts existed in the pure ash clusters (287.3 ± 211.3 nmol g−1 soil dw). Using principle components analyses (PCA) and redundancy analyses (RDA), we found only for the variables ‘relative proportion of beech trees’ and ‘living lime fine root tips associated with ectomycorrhiza’ a significant effect on the PLFA composition. The microbial community structure was mainly determined by abiotic environmental parameters such as soil pH or clay content. The different species richness levels in the clusters did not significantly differ in their total PLFA amounts and their PLFA composition. We observed a tendency that the PLFA profiles of the microbial communities in more tree species-rich clusters were less influenced by individual PLFAs (more homogenous) than those from species-poor clusters.We concluded that tree species identity and site conditions were more important factors determining the soil microbial community structure than tree species diversity per se.  相似文献

2,4-Dichlorophenoxyacetic acid butyl ester (2,4-D butyl ester) is extensively applied for weed control in cultivation fields in China, but its effect on soil microbial community remains obscure. This study investigated the microbial response to 2,4-D butyl ester application at different concentrations (CK, 10, 100 and 1000 μg g?1) in the soils with two fertility levels, using soil dilution plate method and phospholipid fatty acid (PLFA) analysis. Culturable microorganisms were affected by the herbicide in both soils, particularly at the higher concentration. After treating soil with 100 μg g?1 herbicide, culturable bacteria and actinomycetes were significantly higher, compared to other treatments. Treatment of soil with 1000 μg g?1 2,4-D butyl ester caused a decline in culturable microbial counts, with the exception of fungal numbers, which increased over the incubation time. PLFA profiles showed that fatty acids for Gram-negative (GN) bacteria, Gram-positive (GP) bacteria, total bacteria and total fungi, as well as total PLFAs, varied with herbicide concentration for both soil samples. As herbicide concentration increased, the GN/GP ratio decreased dramatically in the two soils. The higher stress level was in the treatments with high concentrations of herbicide (1000 μg g?1) for both soils. Principal component analysis of PLFAs showed that the addition of 2,4-D butyl ester significantly shifted the microbial community structure in the two soils. These results showed that the herbicide 2,4-D butyl ester might have substantial effects on microbial population and microbial community structure in agricultural soils. In particular, the effects of 2,4-D butyl ester were greater in soil with low organic matter and fertility level than in soil with high organic matter and fertility level.  相似文献
