Samples of blood and milk were taken from clinically ketotic cows at 12-hour intervals before and after treatment The concentration of beta-hydroxybutyrate was determined in plasma and milk, and the concentration of acetone was determined in milk. Measurement of milk acetone had the optimal combination of sensitivity, specificity and positive predictive value to identify ketotic cows and was subsequently used in a comparison of the fertility of ketotic and normal cows. Two samples of milk were taken approximately two weeks apart, between 12 and 60 days after calving, from 410 cows on three farms and the concentration of acetone in the milk was measured. Thirty cows with milk acetone concentrations of at least 0.4 mmol/litre were compared with paired control cows with milk acetone concentrations less than 0.3 mmol/litre. The ketotic cows had a significantly longer calving-to-conception interval and a significantly higher culling rate due to a failure to conceive (P<0.05).  相似文献

The whole blood concentrations of acetoacetate (AA concn), β-hydroxybutyrate (BHB concn) and glucose (gluc concn) of 662 Ayrshire and Friesian dairy cows were measured and their milk yield during the indoor-housing period was recorded. Simple correlations among these parameters were evaluated. The correlation between the AA concn and the BHB concn (r = 0.869) was statistically highly significant (P < 0.001), as were the correlations of the logarithmic value of the AA concn with the gluc concn (r = —0.471) and with the milk yield (r = 0.259), and the correlation between the BHB concn and the glue concn (r = —0.288). The milk yield was found also to be associated with the BHB concn and the glue concn (P< 0.001). The associations between each pair of blood parameters were highly significant, too (P < 0.001). The AA concn was taken to be at least as good an indicator of the energy status of dairy cows as the BHB concn and the gluc concn. The AA concn is not diet-derived like the BHB concn and not as stress-sensitive as the blood concentration of free fatty acids and its diurnal variation is not as wide as the BHB concn and the gluc concn in subclinically ketotic cows. Propionate utilisation by the liver in spontaneously ketotic dairy cows was investigated by determining blood glucose levels after an intravenous sodium propionate load (2.5 mmol kg-1). In addition, blood ketone body concentrations were measured after propionate loading. Cows were divided into three groups (control, mildly ketotic and severely ketotic) by their blood acetoacetate concentrations. Plasma glucose concentrations increased significantly after sodium propionate injection in all three groups (P less than 0.05). The maximum glucose concentration occurred earlier in the control group than in the ketotic groups. Changes in glucose concentrations following propionate loading of control and ketotic cows differed significantly at 20 minutes and beyond. Differences in the change in glucose concentration between mildly ketotic and severely ketotic cows were not significant. Acetoacetate concentration was significantly decreased at five minutes and beyond after the injection in ketotic cows, whereas beta-hydroxybutyrate concentration decreased more slowly. A decrease in beta-hydroxybutyrate concentration was significant at 40 minutes and beyond in the severely ketotic group and at 10 minutes and beyond in the mildly ketotic group after loading.  相似文献

The role of ketone bodies on chemotactic capacities of leukocytes was characterized in two experiments. Experiment I was performed to investigate the association between serum beta-hydroxybutyrate concentrations (BHB) and in vitro chemotaxis of leukocytes. Cows were divided into low-BHB, medium-BHB, and high-BHB ones and classified according to their BHB. Leukocytes from high-BHB cows had a significantly lower chemotactic differential than leukocytes from low-BHB cows (p < 0.01). The effect of adding ketone bodies into in vitro chemotaxis cultures on leukocytes chemotaxis was studied in Experiment II. Either individual or a combination of commercial ketone bodies - sodium salts of BHB (BHBA), lithium salt of acetoacetate (ACAC), and acetone (Acetone) - were diluted in culture media and divided into eight concentrations corresponding to concentrations of bovine subclinical and clinical ketosis. For leukocytes from medium- and high-BHB cow, the chemotactic indexes of leukocytes were reduced by ACAC and Acetone. Chemotactic differentials of cultures with ACAC and acetone supplementation from both sources of leukocytes were significantly lower than that of the control culture (p < 0.05). For leukocytes from high-BHB cows, chemotactic indexes were suppressed in a ketone-body environment. In conclusion, leukocytes from naturally-occurring ketotic cows have lower chemotactic differentials than those from non-ketotic cows, and a chemotactic capacity indicated by a chemotactic differential is impaired when leukocytes migrate in an environment with ketone bodies in vitro.  相似文献

Bovine Subclinical Ketosis in Dairy Herds in Iran   总被引:3,自引:0,他引:3  
Subclinical ketosis (SCK) is defined as elevated concentrations of ketone bodies in the absence of clinical signs of ketosis. It is an important metabolic disease in dairy cattle during early lactation and is associated with losses in milk production and several other periparturient diseases. Limited information is available regarding the prevalence of SCK in dairy herds in Iran. The objectives of this study were (i) to determine the incidence of SCK in the dairy herds in Kerman province of Iran using serum beta-hydroxybutyrate (BHB) concentrations, and (ii) to investigate the relationship between serum concentrations of BHB and glucose of cows with SCK. In the present study, 90 multiparous Holstein cows (4-6 years old) from 11 commercial dairy herds were evaluated 3-4 weeks after calving. The distribution of blood BHB concentrations seemed to suggest a cut-off point of 1200 micromol/L between cows with and without SCK. At this cut-off point, 14.4% of tested cows (13/90) were classified as subclinically ketotic, with the prevalence rate within herd ranging from 10% to 20%. Cows with SCK were detected in all the investigated dairies except one. Blood glucose concentrations in cows with SCK were significantly lower (p < or = 0.05) than in cows without SCK, and serum BHB and glucose concentration were inversely correlated (r = -0.43, p < or = 0.05). The results suggest that, using a cut-off of 1200 micromol/L, BHB concentrations can be used during early lactation for diagnosis and to make management decisions for prevention and treatment.  相似文献

The aim of this study was to evaluate the oxidative status in ketotic cows. We observed changes in the oxidative status and correlations between the oxidative and metabolic status in non‐ketotic (n = 10), subclinical ketotic (n = 10) and ketotic cows (n = 10). Plasma samples were analysed by standard biochemical techniques and ELISA to determine traditional metabolic parameters: triglyceride (TG), phosphonium (P), calcium (Ca), aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), total protein (TP), albumin (ALB), immune globulin (Ig), total cholesterol (TC), high‐density lipoprotein (HDL), very low‐density lipoprotein (VLDL) and lactate dehydrogenase (LDH); energy metabolism indices: glucose, β‐hydroxybutyrate (BHBA) and non‐esterified fatty acids (NEFA); and indices of oxidative status: malondialdehyde (MDA), hydrogen peroxide (H2O2), vitamin C, vitamin E, superoxide dismutase (SOD), glutathione peroxidase (GSH‐Px), catalase (CAT), xanthine oxidase (XOD) and total antioxidant capacity (TAOC). The results of this study showed that plasma glucose levels were lower in ketotic and subclinical ketotic cows than in non‐ketotic cows; however, the plasma NEFA and BHBA concentrations were higher. In addition, significant decreases in TC, HDL and VLDL and significant increases in AST, ALT and LDH were observed in the plasma of the ketotic cows. The ketotic cows showed decreased plasma SOD, CAT, vitamin C and vitamin E, inhibited hydroxyl radical capacity and increased plasma H2O2 and MDA. There were positive correlations between the plasma NEFA and ALT, AST, LDH and MDA and negative correlations between the plasma NEFA and TC, HDL, VLDL, SOD, vitamin C, vitamin E, 1542280 uric acid and inhibited hydroxyl radical capacity. In addition, there were positive correlations between BHBA concentrations and ALT, AST and LDH and negative correlations between plasma BHBA concentrations and TC, HDL, VLDL, vitamin E and inhibited hydroxyl radical capacity. Overall, ketotic dairy cows experience oxidative stress, which is presumably associated with hyperketonemia and higher NEFA.  相似文献

Multiple logistic regression was applied to milk yield and composition data for 632 records of healthy cows and 61 records of ketotic cows in Hokkaido, Japan. The purpose was to diagnose ketosis based on milk yield and composition, simultaneously. The cows were divided into two groups: (1) multiparous, including 314 healthy cows and 45 ketotic cows and (2) primiparous, including 318 healthy cows and 16 ketotic cows, since nutritional status, milk yield and composition are affected by parity. Multiple logistic regression was applied to these groups separately. For multiparous cows, milk yield (kg/day/cow) and protein-to-fat (P/F) ratio in milk were significant factors (P<0.05) for the diagnosis of ketosis. For primiparous cows, lactose content (%), solid not fat (SNF) content (%) and milk urea nitrogen (MUN) content (mg/dl) were significantly associated with ketosis (P<0.01). A diagnostic rule was constructed for each group of cows: (1) 9.978 × P/F ratio + 0.085 × milk yield <10 and (2) 2.327 × SNF − 2.703 × lactose + 0.225 × MUN <10. The sensitivity, specificity and the area under the curve (AUC) of the diagnostic rules were (1) 0.800, 0.729 and 0.811; (2) 0.813, 0.730 and 0.787, respectively. The P/F ratio, which is a widely used measure of ketosis, provided the sensitivity, specificity and AUC values of (1) 0.711, 0.726 and 0.781; and (2) 0.678, 0.767 and 0.738, respectively.  相似文献

The objective of this study was to determine the effect of evaporative cooling and dietary supplemental Zn source on blood metabolites, insulin and mineral concentrations, and milk mineral concentrations following intramammary lipopolysaccharide (LPS) infusion. Seventy-two multiparous Holstein cows were assigned to one of four treatments with a 2 × 2 factorial arrangement. Treatments included two environments: with or without evaporative cooling using fans and misters over the freestall and feedbunk, and two dietary sources of supplemental Zn: 75 mg/kg of dry matter (DM) supplied by Zn hydroxychloride (inorganic Zn; IOZ) or Zn hydroxychloride (35 mg of Zn/kg of DM) + Zn–Met complex (ZMC; 40 mg of Zn/kg of DM). A subset of cows (n = 16; 263 ± 63 d in milk) was infused with 10 μg of LPS or a saline control in the left or right rear quarters on day 34 of the environmental treatment. Individual milk samples collected from LPS-infused quarters at −4, 0, 6, 12, 24, 48, 72, 96, and 144 h relative to infusion were analyzed for minerals. Blood samples were collected at the same time with an additional sample collected at 3 h post-infusion to analyze glucose, nonesterified fatty acids (NEFA), insulin, and minerals. Cooling by time interactions (P ≤ 0.07) were observed for plasma glucose, NEFA, and serum insulin. Compared with cooled cows, non-cooled cows had lower concentrations of plasma glucose except at 3 h following intramammary LPS infusion, greater serum insulin at 3 and 12 h, and lower plasma NEFA at 24 and 48 h after infusion. Relative to cooled cows, non-cooled cows tended (P = 0.07) to have lower serum K concentration and had lower (P < 0.01) serum Zn 6 h following infusion (cooling by time interaction: P < 0.01). Relative to ZMC cows, IOZ cows had greater (P ≤ 0.09) concentrations of plasma Se, skim milk Na and Se, and skim milk Na to K ratio. Regardless of treatment, intramammary LPS infusion reduced (P < 0.01) serum or plasma concentrations of Ca, Mg, Zn, Fe, and Se, but increased (P < 0.01) their concentration in skim milk. In conclusion, deprivation of cooling resulted in more rapid and prolonged insulin release and influenced the systemic and mammary mineral metabolism during mammary inflammation induced by LPS of lactating dairy cows. Dietary supplementation of Zn–Met complex reduced blood and milk Se concentrations compared with cows fed Zn from an inorganic source.  相似文献

The authors' hypothesis is that the members of the tricarboxylic acid cycle (TCA cycle) such as citrate decrease in association with increased ketone body formation. To prove this hypothesis the connection between ketone bodies and citrate formation of milk was studied. A fluorimetric method was used to determine citrate and a headspace sampling gas chromatographic (GC) method was developed for determination of ketone bodies. Under real conditions of milk sampling, transport and storage, preserved milk samples of 119 clinically healthy dairy cows obtained in the 48 hours after milking were investigated. A low level of acetoacetate (ACAC) was found in all samples. This fact can be explained by the spontaneous decarboxylation of acetoacetate during sample storage (previously decarboxylised acetoacetate = pdACAC) and, consequently, the majority of the amount of acetoacetate in the samples (AC + pdACAC) appeared in the measured acetone concentrations. Based on the measured acetone concentration of milk samples two groups were formed retrospectively: HA (high-acetone) group (n = 41) with an AC + pdACAC concentration of > 0.4 mmol/l and a LA (low-acetone) group (n = 78) with an AC + pdACAC level of < or = 0.4 mmol/l. In the milk of cows of Group HA a positive correlation (r = +0.623) and linear connection between acetone (AC + pdACAC) and beta-hydroxybutyrate (BOHB) levels was found [BOHB = 2.491 + 0.586 x (pdAC + ACAC)]. Furthermore, in this group a negative correlation between citrate and BOHB and AC + pdACAC was also established (r = -0.579). Focusing on the results of this group the authors found a significant drop of AC + pdACAC and citrate during the metabolically critical first 1-4 weeks of lactation. For this reason they suggest that simple, easy, automated methods (i.e. flow injection analysis, Fourier transformation infrared analysis) should be introduced for the simultaneous determination of acetone and citrate concentration in milk to make the evaluation of the energy status of high-producing dairy cows easier and more certain.  相似文献

This study aimed to assess the effect of ketone body status early postpartum on time empty in 74 multiparous dairy cows under field conditions. Animals were equally distributed across eight farms and were controlled by the same herd‐fertility‐monitoring programme. Cows were visited twice antepartum and six times postpartum at weekly intervals between 0530 and 0830 a.m. On these occasions, body condition scores and milk yields were measured, blood and milk samples were taken, cows were gynaecologically examined, and parameters of reproduction were recorded. Cows with a time empty of less or more than 80 days were classified as early and late conceiving cows (EC and LC, respectively). A time empty of 80 days results in calving‐to‐calving intervals of 1 year and classification based on this threshold value resulted in groups of equal size and equal distribution of EC and LC within farms. Ketone bodies measured were β‐hydroxybutyrate in blood and acetoacetate and acetone in blood and milk. Blood and milk ketone body concentrations, as well as the ratios of acetoacetate and acetone to β‐hydroxybutyrate, over the first 6 weeks postpartum were higher in LC than in EC, whereas plasma glucose and non‐esterified fatty acids and milk fat, protein and urea concentrations did not exhibit clear differences between groups. Ketone body concentrations were as good predictors of time empty as ketone body ratios and might have practical impact in herd‐fertility‐monitoring programmes.  相似文献

The present study examined the Serum 3-methylhistidine concentrations and energy-associated variables of 5 healthy Holstein cows and 5 Holstein cows with ketosis. The serum total cholesterol and apolipoprotein B-100 concentrations and lecithin-cholesterol acyltransferase (LCAT) activity of the ketotic cows were lower than those of the healthy cows 14 days before parturition. The serum non-esterified fatty acid (NEFA) concentration on the day of parturition and 3-methylhistidine concentration 14 days after parturition were higher in the ketotic cows. The serum 3-methylhistidine concentration 14 days after parturition was negatively correlated with the serum LCAT activity 14 days before parturition and was positively correlated with the serum NEFA concentration on the day of parturition. Insufficiency of cholesterol metabolism and acceleration of body fat degradation occur before parturition in cows with ketosis, and these characteristics are correlated with acceleration of protein degradation after parturition.  相似文献

AIM: To determine the concentration of the anti-theilerial drug buparvaquone in the milk and tissue of dairy cattle following treatment with two different formulations, and to assess the effect of clinical theileriosis on the concentration of buparvaquone in milk.

METHODS: Healthy lactating dairy cows (n=25) were injected once (Day 0) I/M with 2.5?mg/kg of one of two formulations of buparvaquone (Butalex; n=12 or Bupaject; n=13). Milk samples were collected from all cows daily until Day 35. Five cows were slaughtered on each of Days 56, 119, 147, 203 and 328, and samples of liver, muscle and injection site tissue collected. Milk samples were also collected from cows (n=14) clinically affected with theileriosis for up to 21 days after treatment with buparvaquone. Milk and tissue samples were analysed by liquid chromatography-mass spectrometry; limits of detection (LOD) were 0.00018?mg/kg for muscle and 0.00023?mg/L for milk. Concentrations of buparvaquone in milk and tissues were log10-transformed for analysis using multivariate models.

RESULTS: In healthy cows, concentrations of buparvaquone in milk declined with time post-treatment (p<0.001), but were above the LOD in 11 of 25 cows at Day 35. Concentration in milk was higher one day after treatment in cows treated with Butalex than in cows treated with Bupaject, but not different thereafter (p=0.007). Concentrations of buparvaquone in muscle were below the LOD for four of five animals at Day 119 and for all animals by Day 147, but were above the LOD at the injection site of one cow, and in the liver of three cows at Day 328. Tissue concentrations did not differ with formulation nor was there a formulation by time interaction (p>0.3).

Concentrations of buparvaquone in the milk of clinically affected animals were not different from those of healthy animals at 1 and 21 days post-treatment (p=0.72). Between 21 and 25 days post-treatment concentrations were below the LOD in 9/14 milk samples from clinically affected cows.

CONCLUSIONS: Detectable concentrations of buparvaquone were found in the milk of some cows for at least 35 days and in the liver and injection site of some cows until at least 328 days after injection. There were no biologically meaningful differences in milk or tissue concentrations between the formulations, or in the milk concentrations for cows that were clinically affected compared with those that were healthy at the time of treatment.  相似文献

Abstract

AIM: To evaluate the macromineral status of field cases of dairy cows surgically treated for left abomasal displacement (LDA), with concurrent fatty liver of different severity, and compare this for animals that died or recovered.

METHODS: Sixty-eight Holstein dairy cows with LDA and 110 control cows, from 28 farms, were used in the study. Blood samples and liver biopsies were obtained during standing surgery for correction of LDA, and from control cows. The concentration of macrominerals in serum, and of total lipids (tLPD) and triglycerides (TG) in liver were determined. Liver was examined histologically, and classified for its severity of fatty liver. Cows with LDA were grouped according to severity of fatty liver. Cows in Groups 1 to 3 recovered, whereas those in Group 4 died within 4 weeks of surgery. Group 1=mild (n=4) or moderate (n=6 cows, n=4 heifers) fatty liver, Group 2 = moderate to severe fatty liver (n=13), Group 3=severe fatty liver (n=15 cows, n=5 heifers), Group 4 = severe fatty liver (n=17 cows, n=4 heifers).

RESULTS: The concentration of macrominerals in serum was affected by the concurrence of fatty liver and LDA; Ca, K and Mg were significantly (p<0.05) lower in animals that died than those that survived. For cows with severe fatty liver, concentrations of tLD and TG were higher in the animals that died compared with those that recovered (p<0.01). Cows with LDA and severe fatty liver that died were earlier in lactation (median days in milk (DIM) 13 days) compared with the other cows with LDA (median DIM 21–26 days) (p<0.05); they were also significantly older (median 6 years old) than cows in the other groups (median 4 or 5 years old) (p–0.05).

CONCLUSIONS AND CLINICAL RELEVANCE: Concentrations of macrominerals in serum were influenced by the concurrence of LDA and fatty liver. Animals with low concentrations of Ca, K and Mg had a guarded prognosis. The concentration of K should always be evaluated in cows with LDA and concurrent fatty liver when providing a prognosis. Most cows with severe fatty liver were detected in the first 4 weeks of lactation, but older animals and those that had more recently calved had a worse prognosis.  相似文献

Bovine aorta endothelial cells (BAECs) were used to determine the effect of ketone bodies and glucose on in vitro interferon (IFN), tumor necrosis factor (TNF) and nitric oxide (NO) production. BAECs were incubated for 4 and 24h with the ketone bodies: 3.8mmol/l beta-hydroxybutyrate (BHB), 1mmol/l acetoacetate (AcAc) and 5. 2mmol/l acetone (Ac), used separately or in a mixture together with cytokine inducers: Newcastle disease virus (NDV) and lipopolysaccharide (LPS). BHB alone (but not AcAc or Ac) and a mixture of ketone bodies caused a significant decrease in IFN titers induced by NDV and LPS and in TNF titers induced by LPS. Glucose used at concentrations of 5.55, 3.33 and 1.66mmol/l did not influence cytokine production.NO measured by the nitrite content in culture medium was released spontaneously from BAECs. A slight enhancement of NO release was observed after infection of BAECs with NDV; however, treatment with LPS caused inhibition of the release. The mixture of ketone bodies used with NDV or LPS enhanced NO release. However, when cells were incubated in the medium with 1. 66mmol/l glucose (mimicking low plasma glucose level in ketotic cows) a significant decrease in NO release was observed. This enhancing effect of ketone bodies and inhibition by low glucose in the final effect balanced each other, and the amounts of NO released in the medium with 1.66mmol/l glucose and with the mixture of ketone bodies resembled those produced at 3.33mmol/l glucose without ketone bodies. The significance of these effects of ketone bodies and glucose concentrations on cytokine and NO production in the immunity of ketotic cows has been discussed.  相似文献

Thirty-six primiparous heifers were used to determine the effect of dietary energy on postpartum interval, milk production, serum concentrations of insulin, insulin-like growth factor I (IGF-I), growth hormone, and cerebrospinal fluid (CSF) concentrations of neuropeptide-Y (NPY). Low-quality hay was fed during the last trimester of pregnancy to achieve suboptimal calving weight (370 +/- 5 kg) and condition score (4.0 +/- .1). After calving, cows were allotted to one of four dietary treatments that differed in metabolizable energy. Experimental diets were fed at 2.5% of shrunk body weight and formulated to provide 1.8 (low), 2.1 (maintenance), 2.4 (maintenance high), or 2.7 (high) Mcal of ME/kg DM. Daily milk production was estimated at approximately 30, 60, and 90 d postpartum. Condition score change and weight change were defined as change from calving to d 90 postpartum. As energy intake increased, condition score change (P < .001), IGF-I (P < .001) and insulin (P < .01) increased and postpartum interval decreased (P = .04). No relationship existed between postpartum interval and CSF concentration of NPY (P > .1). Condition score change was positively associated with IGF-I and insulin (r = .71, P < .001; r = .38, P = .02, respectively) and negatively associated with GH (r = -.67, P < .001). Weight change and serum concentrations of GH did not differ (P > .10) among treatments. Increasing dietary energy intake was associated with a curvilinear increase in milk yield (P = .04) and percentage milk fat (P = .03) and a linear increase (P = .04) in energy available for milk production. Greater milk yield at d 30 was associated with a longer postpartum interval (r = .34, P < .05). In conclusion, a greater proportion of net energy is partitioned to maternal tissue realimentation when cows receive high compared to low-, maintenance-, or moderate-energy diets.  相似文献

AIMS: To determine the effects of oral Ca bolus administration in the early postpartum period of cows on milk yield and composition, blood metabolites, early-lactation health status, and reproductive performance.

METHODS: Multiparous Holstein dry cows (n=66) with a mean parity of 3.1 (SD 0.35) were fed a diet with a positive dietary cation–anion difference (DCAD) prior to calving. They were randomly assigned to receive no treatment (Control; n=33) or two oral Ca boluses (n=33, 45?g of Ca per bolus); one was administered immediately after calving (Day 0) and the second 24 hours (±30 minutes) later. Blood samples were collected at calving, and on Days 2 and 7 to determine concentrations in serum of Ca, P, Mg, glucose, non-esterified fatty acids (NEFA), and β-hydroxybutyric acid (BHBA). Milk yield was recorded daily and milk composition was determined weekly from calving until 28 day postpartum. Health and outcomes were determined during the first 30 days postpartum and reproductive outcomes to 180 days postpartum.

RESULTS: Mean milk yields and composition over the first month of lactation were similar between cows in the two treatment groups (p>0.1). Mean concentrations of Ca in serum were not different between treatment groups on Day 0, but were higher on Day 2 for cows that received oral Ca boluses (1.77 (SE 0.07)) compared with Control cows (1.54 (SE 0.08)) (p=0.04). Concentrations in serum of P, Mg, glucose, NEFA and ΒHBA did not differ between treatment groups on any day of measurement. Fewer cows that received oral Ca were diagnosed with hypocalcaemia (total concentrations of Ca in serum <1.5?mmol/L) by Day 2 (2/33; 6%) compared with Control cows (12/33; 36%) (p=0.01). There was no difference in the prevalence of other health outcomes between treatment groups. The proportion of cows conceiving to first insemination was greater in cows that received an oral Ca bolus (19/29; 65%) than Control cows (12/29; 41%) (p=0.01).

CONCLUSIONS: Oral Ca bolus administration increased concentrations of Ca in serum on Day 2 postpartum, and increased first service conception rates, in cows fed a diet with a positive DCAD prior to calving compared to cows that received no oral Ca bolus supplementation. Because of the small number of cows used in this study, further studies in large-scale dairy farms should be carried out to confirm these findings.  相似文献

The aim of the study was to evaluate the concentrations of amyloid A in serum (SAA) and in milk (MAA) of cows with mastitis caused by Streptococcus agalactiae, Streptococcus dysgalactiae and Streptococcus uberis and healthy cows. The blood and milk samples were obtained from Holstein-Friesian cows with clinical signs of mastitis from two tie-stall housing systems herds in the Lublin region in Poland. A total of 80 milk and serum samples from 30 cows with mastitis and 10 healthy cows were selected for study. In the quarter milk samples from cows with mastitis Streptococcus strains were isolated: Strep. agalactiae (7 cows), Strep. dysgalactiae (9 cows) and Strep. uberis (14 cows). The present study indicates that amyloid A concentration was significantly higher in milk of cows with mastitis compared to control cows (1134.25 ng/mL and 324.50 ng/mL, P < 0.001). The highest concentration of amyloid A was found in milk of cows with mastitis caused by Strep. agalactiae and Strep. uberis whereas lowest in the milk of cows with mastitis caused by Strep. dysgalactiae (3882.50 ng/mL, 2587.75 ng/mL and 812.00 ng/mL, respectively). No statistically significant difference in amyloid A concentration in serum was revealed between all unhealthy cows and control group (2140.00 ng/mL and 2510.00 ng/mL, P > 0.05). There was also no statistically significant difference between the level of amyloid A in serum and in milk of cows with mastitis caused by Strep. agalactiae and Strep. uberis. Whereas, in the case of Strep. dysgalactiae, like in the group of healthy cows, the level of amyloid A was significantly higher in serum compared to this in milk (2100 ng/mL and 812.00 ng/mL, P < 0.001; 2510.00 ng/mL and 324.50 ng/mL, P < 0.001; respectively).  相似文献

Subclinical ketosis (SCK) is simply a condition marked by increased levels of circulating ketone bodies without the presence of the clinical signs of ketosis. Subclinical ketosis can cause economic losses through decreased milk production and association with preparturient diseases. Limited information is available regarding the prevalence of SCK in dairy herds in Southwestern Iran. The objectives of this study were (i) determination of the cutoff point of nonesterified fatty acids (NEFAs) and glucose concentrations for diagnosis of SCK using receiver operating characteristic (ROC) analysis, and (ii) determination of prevalence of subclinical ketosis in apparently healthy dairy cattle in Southwestern Iran. From October to December 2009, a total of 100 clinically healthy multiparous Holstein cows (3-8 years old) were randomly selected from 16 dairy herds around Kazerun, Fars Province, Iran. The cows had two-six lactations, with body weight ranging from 500 to 650 kg. Blood samples for each cow were taken at 2, 4 and 6 weeks post parturition and 3-4h after the morning feeding. The optimal cutoff point was set, by the ROC method, to >0.26 mmol/L for NEFA, and < 2.26 mmol/L for glucose with corresponding 82.54% sensitivity and 91.89% specificity for NEFA and 44.44% sensitivity and 78.38% specificity for glucose. Cows with BHB concentrations higher than 1200 μmol/L were classified as having SCK. In 2, 4 and 6 weeks post parturition 63%, 68% and 59% of the tested cows were subclinically ketotic. Overall, 97% of tested cows (97/100) were considered subclinically ketotic in at least one sample period. Thirty percent of tested cows (30/100) suffered from subclinical ketosis in all of the 2, 4 and 6 weeks postpartum. The results suggest that, a cut-off point of 0.26 mmol/L for NEFA concentrations can be used during early lactation for diagnosis of subclinical ketosis and making management decisions for prevention and treatment. Glucose cannot be a good criterion for diagnosis of SCK and it does not appear to be useful for monitoring subclinical ketosis.  相似文献

The objective of this study was to determine the effect of a monensin controlled-release capsule on the proportion of cows with subclinical ketosis (SK). During July to August 2001, 300 cows dried-off 50-70 days before expected parturition were randomly assigned to either a treatment (n = 150, oral capsule, 335 mg/d of monesin for 95 d) or control group (no capsule, n = 150). At 14 days postpartum, a milk sample was obtained and evaluated for beta-hydroxybutyrate (BHBA) using a semi-quantitative ketone test strip. In a sub-sample of 50 cows per group a blood sample was taken and analyzed for BHBA using an ELISA kit. Milk BHBA > or = 200 micromol/L was used as the cut-off value for diagnosis of SK. The incidence of SK based on the milk test was statistically different between groups (P < or = 0.05) with a value of 26.6% for control and 14.5% for cows treated with monensin, respectively. Cows treated with monensin were 0.68 times less likely to give a positive result for milk BHBA than non-treated cows (0.53-0.80; 95% CI). Serum BHBA concentrations did not differ between groups (0.81 +/- 0.09 mmol/L versus 0.70 +/- 0.07 mmol/L for controls and treated, respectively; P > 0.05). However, for each incremental increase in serum BHBA of 0.1 mmol/L occurrence of SK increased 52% (OR = 1.52; 1.21-1.91; 95% CI).  相似文献

ABSTRACT

Aims: To assess the change in body condition score (BCS) during the early and late dry periods and its association with postpartum diseases and milk yield in grazing dairy cows from central Argentina.

Methods: BCS assessments during the dry period, and cow health and milk production records up to 90 days in milk (DIM), were collated for cows from 28 farms at monthly visits between 2007 and 2008. Cows were categorised into four groups; those in Group 1 (n=7,067) maintained or gained BCS during the early and late dry periods; Group 2 (n=2,615) maintained or gained BCS during the early dry period and lost BCS during the late dry period; Group 3 (n=1,989) lost BCS during the early dry period and maintained or gained BCS during the late dry period; and Group 4 (n=5,144) lost BCS during the early and late dry periods.

Results: Cows in Group 1 had reduced odds of having retained fetal membranes (RFM), metritis, and clinical mastitis up to 90 DIM than cows in Group 2 (p<0.001), but the odds of disease were similar to cows in Group 3. The odds of having RFM or clinical mastitis tended to be lower in cows in Group 1 than cows in Group 4 (p=0.08). The odds of cows being culled or dying during the first 90 DIM were lower for cows in Group 1 than for those in Groups 2, 3, and 4 (p≤0.05). Mean accumulated milk yield up to 90 DIM was higher in cows in Group 1 than Group 2 and Group 4 (p<0.001), but was similar to that of cows in Group 3 (p=0.28).

Conclusions and clinical relevance: Cows that lost BCS during the late dry period had increased odds of being diagnosed with several postpartum diseases and had decreased milk yield compared to cows that maintained or gained BCS during the entire dry period. Loss of BCS during any stage of the dry period was also associated with increased incidence of culling or death during the first 90 DIM. These results should raise awareness among dairy cattle producers of the importance of properly managing cow body condition during the dry period, especially during the late dry period.  相似文献
