An observational study was conducted in 63 Canadian dairy farms to evaluate the association of bulk tank milk selenium (Se) concentration (BTSe) with average daily milk yield, milk fat and milk protein, bulk tank somatic cell count (BTSCC), and the probability of being a Staphylococcus aureus-positive herd. Bulk tank milk samples collected between March 2007 and February 2008 were evaluated for BTSCC, S. aureus culture status, and BTSe. Mean BTSe was 0.51 ± 0.15 μmol/L; no herds were classified as deficient or marginal based on BTSe. Bulk tank milk Se was unconditionally associated with milk production; however, adjusting by region indicated a confounding effect of this variable. There was no relationship between BTSe and BTSCC. Higher values of BTSe were associated with lower risk of being a S. aureus-positive herd, possibly as a result of a more robust udder immune response, or as a result of providing Se at a higher rate as one component of an udder health program that reduces risk of being S. aureus positive.  相似文献

In cows from 15 dairy herds (n = 210), serum selenium (Se) concentrations ranged from 0.021 to 0.789 microgram/ml, whereas 0.05 to 0.40 microgram/ml is the reported range for adequate serum Se concentrations in cattle. Serum Se concentrations of dairy cattle appeared to follow a geographic distribution pattern. On the basis of herd mean serum Se concentrations, adequate serum Se concentrations were found in cattle from only 1 of 5 herds grazing forage in the geographic area classified as Se deficient for cattle. Adequate mean serum Se concentrations were found in cattle from 4 of 5 herds located in geographic areas described as having variable forage Se concentrations (Se-marginal areas). Of the 10 herds from these 2 areas, there were only 2 herds in which 95% of the cattle had serum Se concentrations in the Se-adequate range (0.05 to 0.40 microgram/ml). In 2 selected neighboring farms in the Se-deficient area, cattle in 1 herd had adequate serum Se concentrations and cattle in the other herd had less than adequate serum Se concentrations (less than 0.05 microgram/ml). Therefore, more cattle are at risk of developing Se-deficiency disease than is commonly believed and forage of neighboring farms may have different Se concentrations. Serum Se concentrations (up to 0.789 microgram/ml) correlated with glutathione peroxidase enzyme activity; this serum Se concentration (0.789 microgram/ml) is approximately 6.2 times higher than previously reported in dairy cattle. Therefore, RBC glutathione peroxidase activity may be useful in determining the diagnosis of chronic Se toxicosis.  相似文献

We evaluated selenium determination of bulk milk tank samples as an alternative to testing blood selenium for evaluating herd selenium status in DHIA dairy herds in the San Joaquin Valley of California. A method of determining milk selenium levels using inductively coupled plasma spectrometry is described. Mean bulk tank milk selenium levels were 0.0224 mg/L (Range 0.0126-0.0418 mg/L). No statistically significant relationships were found between bulk tank milk selenium levels of a herd and calving interval, days open or log somatic cell counts. Mean herd blood and milk levels were directly proportional to bulk tank milk selenium levels. Within a herd milk selenium levels of a cow were directly proportional to the cow's blood selenium level. Herd selenium levels were not significantly related to soil selenium levels. Determination of bulk tank milk selenium levels has the potential to be a low cost, non-invasive means of evaluating herd selenium levels in order to determine selenium deficiency. Further studies with this technique in areas which are deficient in selenium may provide estimates of the sensitivity, specificity and predictive value of bulk milk tank selenium for determining selenium deficiency in dairy herds.  相似文献

The objectives of this study were: 1) to determine if bulk tank milk urea nitrogen (BTMUN) and whole herd weighted average of the individual cow MUN levels (WHMUN) were equivalent measurements of herd MUN status; and 2) to determine the seasonal variation in BTMUN concentrations in Prince Edward Island (PEI) dairy herds. For BTMUN-WHMUN correlation testing, bulk tank milk samples from 176 herds were tested for MUN once every 1 to 2 wk between September 1999 and August 2002, as part of routine BTM testing for milk components. During this 3-year period, all herds had all milking cows tested for MUN once a month at the same lab. The WHMUN levels (weighted for milk production) were calculated for each month, and were compared to BTMUN levels using a concordance correlation coefficient (CCC) and a graphic procedure. Tests were only compared if they occurred on the same date, producing a final dataset of 669 comparisons. The BTMUN had good (but not perfect) correlation with WHMUN (CCC = 0.91). This high reliability extended to both the pasture and non-pasture seasons, various milk sampling protocols, and all herd sizes seen in PEI. For evaluating the seasonal variation of BTMUN, the 3 y worth of data (24 803 observations) were divided into 15 seasonal categories, 5 seasons per year (early, mid, and late pasture, and early and late stable). Using linear mixed modelling, significantly (P < 0.05) higher BTMUN values were found during the mid and late pasture seasons of 2000, likely because the precipitation was unusually high during this period, enhancing pasture growth.  相似文献

Bulk tank milk from 1,429 herds were collected in 3 rounds from 19 different geographic areas. The milk samples were tested by use of indirect LPS-ELISA procedure to detect Salmonella dublin antibodies. From the obtained OD-values herd seroprevalence in the given area was determined and GR-scores calculated for each herd by addition of the number of positive sampling rounds by the 5 geographically closest neighbour herds. In the 19 different areas the calculated prevalence ranged from 0.01 to 0.41. Totally 3,697 GR-scores were given. The mean GR-scores in the areas ranged from 0.0 to 6.5. Higher GR-scores were found in herds changing to seropositive status compared with herds seronegative throughout the study period. The results indicate that the risk for a dairy herd to receive S. dublin infection increases with the disease status among the nearest neighbours and with the prevalence of seropositive herds in the geographic area.  相似文献

A retrospective study using analysis of plasma copper and zinc, and erythrocyte glutathione peroxidase from 2 080 dairy and beef cow herds was conducted to evaluate the relationship between trace-element status and production, reproduction and health in cows and their calves. Classification of the herd status as deficient, marginal, low-adequate or high-adequate was based on the lower tercile of individual values. Odds ratios for each disorder in herds were calculated by multivariable stepwise logistic regression. Inadequate copper status was not associated with adult disorders, but was an important risk factor for poor calf performance or health. Selenium deficient status was associated with most studied disorders in cows, and both deficient and marginal herd status were strongly associated with poor health of calves, particularly with increased risks of myopathy and infectious diseases. Zinc insufficiency was strongly associated with low milk production and impaired locomotion in dairy herds, and was also associated with diarrhoea and poor growth in calves. Because a low-adequate status increased the risk of many disorders in adults and calves, we propose to classify herds as deficient and marginal when the lower terciles of plasma zinc concentration are below 12 and between 12 and 14 mumol/l respectively.  相似文献

Mycoplasma bovis (M. bovis) is a highly infectious pathogen of cattle causing pneumonia, polyarthritis, otitis, and less frequently, subcutaneous abscesses, abortions and meningitis. Ineffective drugs treatments, culling of infected cows and loss of milk production can lead to significant economic loss on dairy farms. The early detection of cows excreting M. bovis bacteria to prevent mastitis outbreaks is warranted. Reports suggest that the risk of M. bovis mastitis is higher in larger dairy herds. The objective of this study is to estimate the herd-level prevalence of M. bovis in Flanders, Belgium by culturing bulk tank milk samples taken from dairy farms. Three bulk tank milk samples per dairy herd were taken over four weeks, with collection intervals of two weeks. Culturing was done after pre-incubation using modified Hayflicks media to increase the chances of recovery of bacteria. For the identification of M. bovis, tDNA intergenic spacer PCR was used. In three herds (1.5%) of the 200 herds sampled, M. bovis was isolated from one of the three consecutive bulk tank milk samples. We conclude that in Flanders in 2009 at least 1.5% of the dairy herds had one or more cows excreting M. bovis in the milk. The frequent monitoring of bulk tank milk to detect the presence of M. bovis, especially in expanding herds on farms that often purchase replacement animals, should be encouraged in order to detect the presence of M. bovis and to monitor the success of control procedures following an outbreak of mycoplasmal mastitis in the herd.  相似文献

The purpose of this study was to 1) estimate the herd prevalence of contagious mastitis pathogens in bulk milk from Prince Edward Island (PEI) dairy farms, 2) determine the association between bulk milk culture results and mean bulk milk somatic cell count (BMSCC), and 3) investigate the agreement of repeated bulk milk cultures. Three consecutive bulk milk samples were obtained at weekly intervals from all 258 PEI dairy herds and were cultured using routine laboratory methods. Cumulative prevalence of Staphylococcus aureus, Streptococcus agalactiae, and Mycoplasma spp. (M. bovis and M. alkalescens) was 74%, 1.6%, and 1.9%, respectively. Bulk milk somatic cell count of Staph. aureus-positive herds was higher than that of negative herds. Agreement for Staph. aureus isolation between 3 consecutive tests was moderate (kappa = 0.46). Mycoplasma bovis and M. alkalescens in bulk milk are being reported for the 1st time in PEI ever and in Canada since 1972.  相似文献

The primary objective of this study was to determine the contribution of several management variables to average production per day of life (PPDL). A secondary objective was to compare PPDL between two geographic regions in North America and between dairy herds within the two regions. PPDL was determined for animals culled from four ‘large’ dairy farms in Florida, USA (FL), and eight ‘small’ dairy farms in Prince Edward Island, Canada (PEI). Average milk PPDL was calculated by summing the kilograms of milk produced per cow and dividing by the total number of days from birth until culling. PPDL for the four FL herds (9.50 kg day−1) was significantly higher than PPDL for the eight PEI herds (8.64 kg day−1). PPDL increased in a quadratic fashion with number of lactations. PPDL did not differ between the eight PEI herds. Significant differences in PPDL were identified between the four FL dairy farms.

The regression of PPDL on average lactational milk yield (AVGFCM), number of lactations (LACT), age at first calving and average days to conception explained 94.5% and 92.4% of the variation of PPDL for multiparous cows in the PEI and FL herds, respectively. One variable (AVGFCM) accounted for 68% and 55% of the variance in PPDL for PEI and FL, respectively. The addition of LACT to the model explained 87% and 81% of the variance in PPDL for PEI and FL, respectively.  相似文献

A case-control study was undertaken during the summer of 1989 in central Alberta dairy herds to identify independent predictors of nocardial mastitis. Thirty-seven herds with nocardial mastitis were matched with control herds based on herd size, milk production, and enrolment in Alberta Dairy Herd Improvement Services. Control herds were considered free of nocardial mastitis based on negative cultures of four weekly bulk tank milk samples and one composite milk sample collected during the same period from each lactating cow in the herd. A detailed questionnaire on herd management was completed during farm visits. The use of blanket dry cow therapy was not found to be a risk factor for nocardial mastitis. Dry cow therapy with intramammary products containing neomycin and the use of multidose vials of dry cow medications were the only predisposing factors identified as being significantly associated with nocardial mastitis in central Alberta dairy herds. Use of neomycin as a dry cow therapy increased the odds of nocardial mastitis occurring in these dairy herds by 169 times.  相似文献

Milk urea determination is being used as a broad indicator of protein/energy imbalance in dairy herds. The main purpose of this study was to compare blood and bulk milk urea values in grazing herds, to evaluate their seasonal variation under South Chilean conditions, and to examine their potential relationships with herd fertility. The association between herd blood urea concentration (mean of seven lactating cows) and bulk milk urea concentration (tank containing milk from the previous 24 h) was determined in 21 diary herds. Reference values, seasonal and herd variance, and the frequency of herds with values outside a range of 2.5 to 7.3 mmol/l were determined in bulk milk samples obtained monthly for a period of one year from 82 suppliers at two creameries located in southern Chile. Finally, bulk milk urea was measured every two weeks in samples from 24 herds, and the first service conception rate (FSCR) from 2153 dairy cows was determined. Mean bulk urea concentration was highly correlated with mean herd blood urea concentration (r = 0.95; p < 0.01). Mean urea concentration in the bulk milk samples obtained during one year from 82 herds was 4.9 +/- 1.2 mmol/l, with a range of 1.5 to 11.6 mmol/l. The highest values were found during spring and the lowest values during the summer. There was a high seasonal variation (CV = 13-47%) and between-herd variation (CV = 20-31%). Out of a total of 984 samples, 5.4% had urea values > 7.3 mmol/l and 3.8% had values < 2.5 mmol/l. Of the 82 herds, 27% had values outside the reference interval (2.5-7.3 mmol/l) on two or more occasions. FSCR was lower in herds when the bulk milk urea was > 7.3 mmol/l (50.7%) than in cows, where the urea concentration was < 5.0 mmol/l (73.8%) at the time of insemination. The study concluded that bulk milk urea concentrations provided information similar to herd blood urea concentrations in local grazing dairy herds. There was a high frequency of herds with abnormal values, with large variations between herds and between seasons. Increased milk urea concentrations during spring were associated with lower conception rates.  相似文献

AIM: To determine the relationships between blood selenium (Se) concentrations or glutathione peroxidase activity (GSH-Px), and milk Se concentrations in dairy cows. METHODS: Seventy-two Friesian dairy cows were either untreated or injected with 0.5, 1.0 or 2.0 mg Se/kg liveweight as barium selenate (BaSeO4) formulations, resulting in 6 groups of animals with mean blood Se concentrations that varied from 212 to 2272 nmol/l. Milk samples were collected on Days 104 and 188, and blood samples were collected prior to treatment and on Days 41, 76, 104, 188, 244, and 292 after Se injection. RESULTS: Significant quadratic relationships between blood Se and milk Se concentrations, as well as blood GSH-Px activity and milk Se concentrations, were evident at Days 104 and 188. Using combined data, these were represented by the equations: milk Se = 27.3 + 0.073 blood Se -0.00001 (blood Se)2; R2=0.79, p<0.005, and; milk Se = 34.8 + 4.99 GSH-Px -0.068 (GSHPx)2; R2=0.79, p<0.005. CONCLUSIONS: The Se status of dairy cows can be assessed from milk Se concentrations. CLINICAL SIGNIFICANCE: Bulk-tank milk Se concentrations could be evaluated as a method to assess the Se status of dairy herds.  相似文献

Environmental factors that enhance either the survivability or dispersion of Salmonella enterica serovar Typhimurium (S. Typhimurium) could result in a spatial pattern of disease risk. The objectives of this study were to: (1) describe herd status based on antibody response to Salmonella Typhimurium as estimated from bulk tank milk samples and (2) to describe the resulting geographical patterns found among Texas dairy herds. Eight hundred and fifty-two bulk milk samples were collected from georeferenced dairy farms and assayed by an indirect enzyme-linked immunosorbent assay (ELISA) using S. Typhimurium lipopolysaccharide (LPS). ELISA signal-to-noise ratios for each bulk tank milk sample were calculated and used for geostatistical analyses. Best-fit parameters for the exponential theoretical variogram included a range of 438.8 km, partial sill of 0.060 and nugget of 0.106. The partial sill is the classical geostatistical term for the variance that can be explained by the herd's location and the nugget is the spatially random component of the variance. We have identified a spatial process in bulk milk tank titers for S. Typhimurium in Texas dairy herds and present a map of the expected smoothed surface. Areas with higher expected titers should be targeted in further studies on controlling Salmonella infection with environmental modifications.  相似文献

The Danish government and cattle industry instituted a Salmonella surveillance program in October 2002 to help reduce Salmonella enterica subsp. enterica serotype Dublin (S. Dublin) infections. All dairy herds are tested by measuring antibodies in bulk tank milk at 3-month intervals. The program is based on a well-established ELISA, but the overall test program accuracy and misclassification was not previously investigated. We developed a model to simulate repeated bulk tank milk antibody measurements for dairy herds conditional on true infection status. The distributions of bulk tank milk antibody measurements for infected and noninfected herds were determined from field study data. Herd infection was defined as having either >or=1 Salmonella culture-positive fecal sample or >or=5% within-herd prevalence based on antibody measurements in serum or milk from individual animals. No distinction was made between Dublin and other Salmonella serotypes which cross-react in the ELISA. The simulation model was used to estimate the accuracy of herd classification for true herd-level prevalence values ranging from 0.02 to 0.5. Test program sensitivity was 0.95 across the range of prevalence values evaluated. Specificity was inversely related to prevalence and ranged from 0.83 to 0.98. For a true herd-level infection prevalence of 15%, the estimate for specificity (Sp) was 0.96. Also at the 15% herd-level prevalence, approximately 99% of herds classified as negative in the program would be truly noninfected and 80% of herds classified as positive would be infected. The predictive values were consistent with the primary goal of the surveillance program which was to have confidence that herds classified negative would be free of Salmonella infection.  相似文献

An enzyme-linked immunosorbent assay (ELISA) using bulk tank milk samples was evaluated as a screening test for bovine tuberculosis (TB), a contagious chronic disease of cattle. An ELISA with MPB70, a major antigen of Mycobacterium bovis was performed using paired sets of milk and sera samples from 33 tuberculin-positive and 43 tuberculin-negative cattle. Anti-MPB70 antibodies were detected in milk samples and there was a significant correlation between seroreactivities of milk and sera samples (R2 = 0.83). Using the tuberculin skin test as the reference test, the sensitivities of ELISA using milk and sera samples were 87.8% and 81.8%, respectively, and the specificities were 97.7% and 100%, respectively.In the screening test using bulk tank milk samples from 931 dairy herds in Whasung, Gyeonggi-do, Korea, the positive rate for anti-MPB70 antibody was 4.5% (42/931) and the tuberculin-positive rate was 2.8% (26/931). Individual milk samples (n = 253) were collected from randomly selected 8 problematic and 3 negative herds (positive and negative in the screening test by MPB70 ELISA using bulk tank milk samples, respectively) and tested by MPB70 milk ELISA. In the problematic herds, positive rates were 10.5% (20/190) for anti-MPB70 antibodies in milk ELISA and 2.1% (4/190) in the tuberculin skin test. More than one dairy cows were positive by milk ELISA among the problematic herds, and all tuberculin-positive dairy cows were positive in the milk ELISA. Further, no positive cows were detected in negative herds both by milk ELISA and tuberculin skin test. These results suggest that an ELISA, using bulk tank milk samples, might be a potential efficient screening test for bovine TB of dairy cows.  相似文献

Streptococcus agalactiae mastitis persists as a significant economic problem for the dairy industry in many countries. In Denmark, the annual surveillance programme for this mastitis pathogen initially based only on bacteriological culture of bulk tank milk (BTM) samples, has recently incorporated the use of the real-time PathoProof Mastitis PCR assay with the goal of improving detection of infected herds. The objective of our study was to estimate the herd sensitivity (Se) and specificity (Sp) of both tests of BTM samples using latent class models in a Bayesian analysis while evaluating the effect of herd-level covariates on the Se and Sp of the tests. BTM samples were collected from all 4258 Danish dairy herds in 2009 and screened for the presence of S. agalactiae using both tests. The highest Se of PCR was realized at a cycle threshold (Ct) cut-off value of 40. At this cut-off, the Se of the PCR was significantly higher (95.2; 95% posterior credibility interval [PCI] [88.2; 99.8]) than that of bacteriological culture (68.0; 95% PCI [55.1; 90.0]). However, culture had higher Sp (99.7; 95% PCI [99.3; 100.0]) compared to PCR (98.8; 95% PCI [97.2; 99.9]). The accuracy of the tests was unaffected by the herd-level covariates. We propose that screenings of BTM samples for S. agalactiae be based on the PCR assay with Ct readings of <40 considered as positive. However, for higher Ct values, confirmation of PCR test positive herds by bacteriological culture is advisable especially when the between-herd prevalence of S. agalactiae is low.  相似文献

Between January 1972 and December 1990, bulk-tank (n = 721) and cow (n = 9,163) milk samples from dairy herds in New York State were examined by bacteriologic procedures for Mycoplasma. The organism was found in 165 herds in 42 counties, and in 2.3 and 11.7% of the tank and cow samples, respectively. Mycoplasma bovis was isolated in 164 herds, M. californicum was isolated in 1. Highest incidence of mycoplasmal clinical mastitis occurred during the winter. The disease resulted in culling of 30-70% of the cows in several herds. Eighty-six of the positive herds were located in the western part of the state. This area had more large herds (greater than 200 cows) compared to the rest of the state; however, herd size was not a risk factor. Purchased animals added to herds without quarantine, poor hygiene during mastitis treatment, and personnel in contact with mastitic cows or infected milk were involved in outbreaks and disease transmission.  相似文献

Caused by imported beef cattle new outbreaks of Brucellosis (subtype III) were observed in the government district of Hannover, which was "Brucellosis free" over a long period. With the aim to interrupt the series of infections from herd to herd it seemed to be necessary to introduce a herd screening system including frequent tests. It was not possible to screen the herds by the usual way of blood serum testing because of reasons of practicability and economy. A practicable alternative was the ELISA supported tank investigation. Four of the last six outbreaks were detected by this milk sampling. The other two infected herds were detected by only clinical symptoms because the lactating cows were not infected and had no contacts to the infected separately held heifers. The required test frequency in the dairy cattle herds could only be realized applying an ELISA supported highly sensitive tank milk test method. This method offered the chance of discovering infected herds as soon as possible and prevented greater economical losses and animal health risks. The tank milk screening of the dairy cattle herds to detect antibodies against Brucellosis (and additional EBL and IBR) has now become a standard method to continue the official status "free from..." This is a safer and a more economic alternative to the previous blood sampling of cattle older than two years with a three years interval because this method guarantees a safe information about the serological status of each cow which is now tested at least once a year, depending on the time of dry standing status.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

OBJECTIVE: To monitor patterns of Salmonella fecal shedding in naturally infected dairy herds, determine the association between fecal shedding and individual animal production measures, and evaluate potential risk factors for shedding of Salmonella organisms among cattle in dairy herds. DESIGN: Longitudinal study. SAMPLE POPULATION: 5 Ohio dairy herds. PROCEDURE: For 3 herds, fecal samples were collected from all mature cows and unweaned calves 7 times during an 18-month period. For the remaining 2 herds, fecal samples were collected from 50 lactating cows 6 times during a 12-month period. Individual animal production records for 3 herds were used to examine associations between individual fecal Salmonella shedding status and 305-day mature-equivalent milk production, somatic cell count, milk fat content, and milk protein content. Multivariable logistic regression was used to test for associations between fecal shedding status and breed, lactation status, lactation number, and duration of lactation. RESULTS: None of the adult animals had clinical signs of salmonellosis, but prevalence of fecal Salmonella shedding at individual collection times ranged from 0 to 99% for cows and from 0 to 67% for unweaned calves. Mature cows were more likely to be shedding Salmonella organisms than were unweaned calves. Within herds, lactation status and duration of lactation for individual animals were associated with Salmonella shedding status. Salmonella fecal shedding status was not associated with individual cow production measures. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that subclinical fecal Salmonella shedding can persist in dairy herds for up to 18 months with no measurable effects on health or production of individual cows.  相似文献

The geographical and temporal dynamics of the outbreak of off-flavours in bulk-tank milk that occurred between September 2000 and June 2002 in Prince Edward Island (PEI) dairy herds were described using spatial, temporal and space-time scan statistics. Results suggested clustering both in time and space. We found one primary cluster (six case herds, feed off-flavour, October 2000 to January 2001) and two secondary clusters formed of feed (5 case herds, October 2000 to January 2001)) and rancid (five case herds, December 2000 to March 2001) off-flavours, respectively. The relative risk (RR) for producing off-flavoured milk was similar for these three clusters (RR=6.4).  相似文献
