The present report describes an intestinal disease which causes important losses in farmed turbot. Mortality rates were higher in summer and reached 100% in all tanks where the disease was confirmed. Affected fish showed external signs consisting of anorexia, sunken eyes and a typical prominent bony ridge on the skull. These signs can be considered the pathognomonic signs of the disease, together with the gut lesions seen in the histological study. Pallor of the internal organs, intestinal haemorrhages and the presence of liquid in the intestine were also observed, with ascites in heavily infected fish. Histopathological damage was evident in the gut, with severe enteritis, detachment of epithelium, haemorrhages and inflammation of the subepithelial connective tissue. The myxosporean aetiology was demonstrated in all the fish showing the characteristic signs of disease. Myxosporean stages, including scarce spores, were found in the affected epithelium or free in the intestinal lumen together with epithelial debris. The present study demonstrates the importance of this myxosporean disease which represents a serious threat for turbot culture. This is also the first record of a member of the Myxosporea in turbot.  相似文献

Macroscopic and histopathological changes in cultured turbot, Scophthalmus maximus (L.), in Spain caused by infection with Edwardsiella tarda are described. Eye tumefaction, inflammation, haemorrhages, ascites and the presence of a purulent fluid were the main macroscopic lesions observed. Histopathological lesions were found in the kidney, spleen and liver. In the kidney and spleen these were characterized by a severe apostematous inflammatory reaction, with a large number of abscesses. The liver was affected to a lesser degree and only some phagocytes loaded with bacteria were observed. Ultrastructural observations indicated that macrophages were the main cell type implicated in the inflammatory response. Most of the bacteria observed within the phagocyte cytoplasm showed no degenerative changes and some were dividing. Degenerative changes observed in macrophages indicate their failure in preventing the infection.  相似文献

Turbot was shown to be sensitive to injection challenges by Aeromonas salmonicida subsp. achromogenes (Asa). A systemic disease was induced and the bacterium was isolated from various internal organs. Histopathological changes involved haemorrhages, necrosis and degeneration in skin and muscle, haemorrhages and necrosis in kidney, degeneration in the heart muscle, and fusion of the secondary gill lamellae. A polyvalent commercial salmon vaccine, containing A. salmonicida subsp. salmonicida as one of five antigens, did not confer protection in turbot against an experimental Asa infection 13 weeks post-vaccination. Vaccination induced a significant antibody response against Asa cells but not against extracellular products of the bacterium. The results of the study indicate that Asa may be a potential threat to turbot farming and that the development of new turbot vaccines is needed.  相似文献

Several strains of Garm-positive short rod (coccibacilli)-shaped bacteria were isolated from diseased cultured turbot, Scophthalmus maximus (L.), in the North of Spain with lesions and signs of Spain with lesions and signs of streptocecosis. The α-haemolytic streptoceoccus-like organisms from diseased turbot were identified by physiological, biochemical and 16S rRNA gene sequence analysis as Streptococcus parauberis. This is the first report of S. parauberis associated with fish disease.  相似文献

Haemorrhagic areas in the mouth of farmed turbot, Scophthalmus maximus (L.)   总被引:4,自引:0,他引:4  
Abstract. An epizootic in juvenile turbol reared on a farm located in The Ria de Vigo, Galicia, Northwest Spain, is described. The diseased turbot did not display unusual swimming behavior; the external signs of the disease were abdominal distension and haemorrbagic areas in the mouth. Internal examination of the fist showed an accumulation of reddish fluid in the peritoneal cavity. Microbiological analysis of the diseased fish revealed the presence, in pure culture in all the organs and lesions examined, of a bacterium which was characterized biochemically as Vibrio splendidus biotype I. The virulence tests showed that the V. splendidus biotype I isolate was pathogenic for rainbow trout (LD50: 2.2 × 104) and also for turbot (LD50: 1.2 × 104). The treatment of the fish using flumequine incorporated into the feed was effective in offsetting the mortality rate.  相似文献

The susceptibility of turbot, Psetta maxima, to infection with two strains of viral haemorrhagic septicaemia virus (VHSV) obtained from wild Greenland halibut, Reinhardtius hippoglossoides, and from farmed turbot was examined. A marine VHSV strain known to be highly pathogenic for turbot was also utilized for comparative purposes. Fish were infected by intra-peritoneal (i.p.), immersion or cohabitation, and maintained at two different temperatures (8 and 15 degrees C). Infection trials showed that the three VHSV isolates were pathogenic for turbot fingerlings by i.p. injection at both temperatures, with high levels of mortality. Virus was recovered from most pools of dead fish i.p. challenged, but not from surviving fish. Although clinical signs were not induced following waterborne exposure, viral growth was obtained from some pools of surviving fish challenged by immersion with strain GH40 from Greenland halibut, which indicates that the virus can survive in sea water and infect other fish via horizontal transmission. Furthermore, although low, the clinical signs and mortality observed in fish cohabitating with turbot challenged with strain GH40 confirms horizontal transmission and indicates that the passage through fish increases the virulence of this strain for turbot. These findings indicate that Greenland halibut, as other wild fish, may play an important role in the epizootiology of VHSV and suggest a potential risk for the turbot farming industry.  相似文献

Abstract. Turbot, Scophthalmus maximus (L.), and sea bass, Dicentrarchus labrax (L.), are susceptible to vibriosis, a septicaemia caused in France by Vibrio anguillarum 408. A bacterin produced with this strain by Rhône–Merieux has been tested orally in both species and, as a comparison, by intraperitoneal injection. During a challenge carried out 4 weeks later by intraperitoneal inoculation, the protection observed in orally vaccinated fishes was significant (72 % in sea bass and 70 % in turbot). Eleven weeks after vaccination, this protection is still significant. The mean titre of serum agglutinins was weakly increased in orally vaccinated fishes. Bacteriostatic antibodies have also been searched for in serum, but compared to agglutination, the applied technique leads to lower titres and it has not been possible to prove the presence of such antibodies after oral vaccination. Passive immunizations using serum from orally vaccinated fishes (2 months after vaccination) confers some protection against a challenge carried out by inoculation of virulent vibrio. Therefore, the production of serum factors following oral vaccination of sea bass and turbot may be involved in generalized protection of fish.  相似文献

采用基于Illumina测序平台的高通量测序技术,对大菱鲆(Scophthalmus maximus)幼鱼肠道及其养殖水体、生物饵料中细菌种类及丰度进行研究。测序结果显示,养殖水体、生物饵料和大菱鲆幼鱼肠道等19个样品共获得有效序列547621条,可聚类于3771个可分类操作单元(OTUs),归属于养殖水体、生物饵料、健康幼鱼和发病幼鱼的操作分类单元(OTU)个数分别为3038、1090、87和777,其中,健康幼鱼与生物饵料、健康幼鱼与养殖水体特有的OTU个数分别为57和0,发病幼鱼与生物饵料、发病幼鱼与养殖水体特有的OTU个数分别为481和31。表明幼鱼肠道微生物多样性与生物饵料密切相关。根据细菌注释结果,拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)和变形菌门(Proteobacteria)在大菱鲆幼鱼肠道中占优势地位,其中,健康幼鱼肠道微生物共聚类为8个门,发病幼鱼的肠道微生物可聚类为19个门。与健康幼鱼相比,发病幼鱼肠道门水平上的3种主要优势菌群落结构出现失衡。此外,对各样品中丰度最高的100位OTU分析显示,幼鱼肠道优势菌种类与生物饵料中的优势菌种类密切相关,而每个发病幼鱼肠道优势菌种类具有一定的独立性。本研究旨在为大菱鲆健康养殖和微生态调控提供实验依据。  相似文献

Whole-genome next-generation sequencing was used to investigate the local evolution of viral haemorrhagic septicaemia virus, a serious pathogen affecting economically important fish such as rainbow trout and turbot in Europe and olive flounder in Asia. Sequence analysis showed that all isolates were genotype IVa, but could be classified further into four subgroups (K1–K4). In addition, genomic regions encompassing the nucleoprotein, phosphoprotein, matrix protein and non-virion protein genes, as well as the seven non-coding regions, were relatively conserved, whereas glycoprotein and RNA-dependent RNA polymerase genes were variable in the coding region. Taken together, the data demonstrate that whole-genome next-generation sequencing may be useful for future surveillance, prevention and control strategies against viral haemorrhagic septicaemia.  相似文献

The susceptibility of Atlantic halibut, Hippoglossus hippoglossus (L.), to viral haemorrhagic septicaemia virus (VHSV) was tested. Juvenile halibut of approximately 5 g weight were subjected to challenge by intraperitoneal injection, cohabitation and immersion to a VHSV isolate from an outbreak of the disease in turbot, Scophthalmus maximus (L.). The intraperitoneal injection gave the highest mortality rate of 28% after 50 days. The cohabitee group suffered 19% mortality rate and the immersion group only 2%. Control groups included turbot exposed either by intraperitoneal injection or immersion which suffered mortality rates of 93 and 50%, respectively. The results suggest that halibut are markedly less susceptible to VHSV than turbot.  相似文献

Abstract. Since 1992, frequent erythrocytic viral infections have occurred in cultured turbot, Scophthalmus maximus (L.), on several fish farms in Galicia, Spain, causing important mortalities. Affected fish showed haemorrhages in the head, mouth and fins, exophthalmia and abdominal distension. The disease was also characterized by the presence of pale blue inclusions in affected erythrocytes. The examination of these erythrocytes by electron microscopy showed virus particles of 50–160 nm in size in the cytoplasm, budding av the cell membrane and extracellularly. The histopathological study revealed occasional necrosis of kidney haematopoietic tissue, an increase in the size and number of melanomacrophage centres of the spleen and kidney, and oedematous areas with hepatocytes containing eosinophilc bodies in the liver. Degeneration of muscle fibres was accompanied by an intense inflammatory oedema. Dermatitis as well as uveitis and corneal oedema were consistent lesions.  相似文献

Sixteen specimens of female crucian carp, Carassius carassius (L.), during the breeding season, were investigated for post‐mortem and full diagnostic examination during a mortality outbreak in a tributary stream of the Arno River in Tuscany in 2011. Necropsy highlighted the presence of a swollen anus and widespread haemorrhages in the body, fins, gills and eyes. Haemorrhages in internal organs and spleen granulomas were also observed. Bacteria isolated from the brain, kidney and spleen of affected fish were identified as A. sobria. Microscopic lesions observed in gills were characterized by necrosis of the secondary lamellae, congestion and multifocal lamellar fusion. The kidney showed necrosis, oedema, fibrin exudation and areas of haemorrhages, while in the spleen the main lesions were by multifocal necrosis of the lymphoid tissue. In the gills, transmission electron microscopy revealed herpesvirus‐like particles, subsequently identified as Cyprinid herpesvirus‐2 (CyHV‐2) with a nested PCR protocol. Although it was not possible to attribute a pathogenic role to CyHV‐2 in this mortality event, the identification of this herpesvirus in crucian carp increases the concern about its potential role in this species.  相似文献

Streptococcosis, lactococcosis and enterococcosis are among the most important bacterial diseases affecting tilapia farms in Kafr Elsheikh governorate, Egypt. A number of clinically diseased fish were collected and submitted to our laboratory during disease outbreak in 2018. They were characterized by nervous swimming behaviour, skin darkness, exophthalmia, ocular opacity and haemorrhages. Necropsy findings were splenomegaly, congestive hepatomegaly, liquefied brain and enteritis. The phenotypic and molecular characterizations of the bacterial strains isolated from naturally infected fish identified three genera of Gram‐positive cocci: Streptococcus agalactiae, Enterococcus faecalis and Lactococcus garvieae. Infectivity trials were conducted in four groups of Nile tilapia inoculated with S. agalactiae Egy‐1, E. faecalis Egy‐1 and L. garvieae Egy‐1 strains and saline. Mortalities, clinical signs and pathological findings were recorded daily 14 days post infection. Experimentally infected tilapia showed similar clinical signs, postmortem lesions, but varied in the severity and experienced high mortalities up to 70% in case of S. agalactiae and L. garvieae infections and 30% in case of E. faecalis infection. Pathological examination of infected tissue sections stained with modified Brown–Brenn and immunohistochemistry revealed an important direct correlation between the distribution of each bacterial isolate and the lesions developed in different organs. Furthermore, the isolates were subjected to profiling against 11 antibiotics, and they showed resistance against several types of antibiotics, which implicate potential risk to human health and emphasize the urgent need for alternate bio‐control strategies to prevent the diseases and the problem of multidrug resistance in aquatic environment.  相似文献

The chemokines are a superfamily of chemotactic cytokines playing an important role in leukocyte chemotaxis. Here, a turbot head kidney cDNA library was constructed in which KC70 was identified as a CC chemokine. Unknown 5′ and 3′ parts of the cDNA were amplified by 5′ and 3′ rapid amplification of cDNA ends (RACE). The complete cDNA of KC70 contains a 59-bp 5′ UTR, a 336-bp ORF, and a 152-bp 3′ UTR. Four exons and three introns were identified in KC70. Phylogenetic analysis showed that KC70 was similar to CCL19. In normal turbot KC70 was expressed in all tissues except brain and skin. Infection of turbot with pathogenic bacteria significantly increased expression of KC70 in the liver. Expression of KC70 in head kidney first increased and then decreased after bacterial challenge. No significant change was observed in the spleen after bacterial challenge. During embryonic development, KC70 was highly expressed after the gastrula stage. These results indicated KC70 plays important and multiple roles in turbot immune response.  相似文献

Abstract. Fingerling channel catfish, Ictalurus punctatus (Rafinesque), were exposed experimentally to Edwardsiella ictaluri by immersion for 1 h in water containing 5 × 108 colony-forming units (CFU) of the bacterium per ml. Ninety per cent of the fish developed lesions typical of enteric septicaemia of catfish (ESC), 93% of affected fish developed the acute form of ESC and 7% developed chronic ESC. Acute disease was characterized grossly by cutaneous haemorrhage and ulceration, and microscopically by enteritis, olfactory sacculitis, hepatitis and dermatitis. The earliest lesions of acute ESC, i.e. enteritis and olfactory sacculitis, were observed microscopically at 2 days post-exposure (PE); gross lesions, primarily mild subcutaneous haemorrhage at the base of fins, were first apparent at 4 days PE. Chronic ESC, seen most commonly 3–4 weeks PE, was characterized by dorsocranial swelling and ulceration, granulomatous olfactory neuritis/perineuritis, and meningoencephalitis involving the olfactory bulbs, olfactory tracts and olfactory lobes of the brain. Gross and microscopic lesions in the acute and chronic forms of experimental ESC were similar to the lesions reported in naturally occurring ESC. Definitive pathogenesis of acute and chronic ESC remains to be determined.  相似文献

Abstract. Viral haemorrhagic septicaemia (VHS) is reported for the first time in sea-water cultured rainbow trout. Heavy mortalities with typical signs and lesions A VHS virus (serotype 1) was isolated from the diseased fish. The mortalities were caused only by the VHS virus and 80 days post transfer of trout to sea-water the mortalities reached 85%, of the initial population.
The disease was experimentally transmitted to rainbow trout, both in sea-water 3·104 pfu/ml of virus or by intramuscular injection of various doses of VHS1 (7·101 7·104 or 7·104 pfu per fish). Death occurred in all infected groups and started earlier in sea-water. Typical signs of VHS were observed in moribund fish. Viral multiplication was demonstrated to have occurred in fish organs.  相似文献

2014年11月,大连地区某大菱鲆养殖场发生病害并伴有死亡,主要症状为吻部下颌明显出血,腹腔积水,肝脏充血,肠道出血有白便。从患病大菱鲆肠道中分离出1株可在硫代硫酸盐—柠檬酸盐—胆盐—蔗糖琼脂培养基上生长的菌株HZ-C1,人工回接感染试验证明其对健康大菱鲆具有较强致病性。通过形态学观察、16SrDNA序列同源性分析及生理生化试验确定菌株HZ-C1为大菱鲆弧菌。药敏检测结果显示,大菱鲆弧菌HZ-C1对氯霉素、丁胺卡那及头孢曲松等11种抗生素敏感,但对青霉素、四环素及诺氟沙星等15种抗生素耐药。本研究为大连地区养殖大菱鲆细菌性病害防治提供了一定病原学依据。  相似文献

Abstract. Cultured turbot Scophthalmus maximus (L.) from three farms at Hunterston, Scotland were found to be infected with Haemogregarina sachai . Up to 6% of some populations were affected with gross tumorous lesions of the musculature and viscera. The lesions consisted of necrotic tissue with a caseous centre. Histologically, there was an accumulation of parasitized reticulo-endothelial cells, cell debris and pyknotic nuclei. Parasitaemias of up to 36% of all blood cell types were observed. The majority of infected cells were neutrophils and monocytes. Infected leucocytes were hypertrophied. There was an erythrocytopenia of infected fish with increases in erythroblasts, monocytes and neutrophils but a decrease in thrombocytes and lymphocytes. Transmission experiments between fish were unsuccessful.  相似文献

Viral haemorrhagic septicaemia (VHS) is a well-characterized disease of rainbow trout, Oncorhynchus mykiss, which has also caused economic losses in marine turbot farms in the British Isles. We have previously demonstrated that turbot, Scophthalmus maximus, are susceptible to isolates of viral haemorrhagic septicaemia virus (VHSV) that are endemic in the marine environment, highlighting a potential risk to marine aquaculture. Given the increasing interest in the intensive rearing of additional aquaculture species such as Atlantic cod, Gadus morhua, and Atlantic halibut, Hippoglossus hippoglossus, this study aimed at investigating the susceptibility of these species to VHSV. Both species were found to be largely resistant to VHS following immersion challenge with a selection of 18 isolates, representing the known marine VHSV genotypes. Only one and two VHSV-associated mortalities occurred out of a total of 1710 and 1254 halibut and cod, respectively. These findings suggest that there is a low direct risk to the development of commercial cod and halibut aquaculture from the existing endemic reservoir of VHSV. This study, coupled to field observations has, however, highlighted the fact that both species can become infected with VHSV. The known adaptability of RNA viruses, together with the selection pressures associated with intensive aquaculture would thus advocate a cautious approach to VHSV surveillance within these emerging industries.  相似文献
