基于前表面荧光光谱鉴别新鲜与冻融大黄鱼   总被引:3,自引:3,他引:0  
为研究反复冻融对水产品品质的影响,通过理化方法检测了不同冻融次数处理对大黄鱼解冻损失、pH 值、色泽、硫代巴比妥酸值、羰基含量等指标的影响,并采用前表面荧光光谱结合主成分分析(principal component analysis,PCA)和 Fisher 线性判别分析法(Fisher linear discriminant analysis,FLDA)对不同冻融次数的大黄鱼进行区分。结果显示随着冻融次数增加,大黄鱼的解冻损失显著增加(P<0.05);pH 值呈先上升后下降的趋势;L*(亮度)值、b*(黄度)值均有不同程度的增加(P<0.05),a*(红度)值下降(P<0.05);羰基含量和硫代巴比妥酸反应物值(thiobarbituric acid reactive substances,TBARS)增加(P<0.05),反复冻融导致大黄鱼的品质下降。色氨酸和烟酰胺腺嘌呤二核苷酸磷酸(nicotinamide adenine dinucleotide,NADH)的荧光光谱分别结合 PCA 和 FLDA 对不同冻融处理组进行分析,结果表明 FLDA 识别效果优于 PCA。通过 FLDA 建立了新鲜大黄鱼与冻融大黄鱼荧光光谱判别模型,发现色氨酸原始判别的准确率和交叉验证的准确率分别为68.3%和66.7%,NADH 原始判别的准确率和交叉验证的准确率均达到100%。由此可见,利用 NADH荧光光谱结合化学计量分析可以鉴别不同冻融处理的大黄鱼。研究结果为水产品新鲜度的快速评价提供参考。  相似文献

蛋壳品质对蛋品孵化、贮存和运输均有重要影响。为了探索近红外光谱技术快速检测蛋壳品质的方法,该文在鸡蛋蛋壳品质指标相关性分析的基础上进行了蛋壳品质的近红外光谱检测分析,研究比较了不同建模方法、不同光谱预处理方法和不同波段范围对预测结果的影响。结果表明:在5段特征波长范围内建立的经过多元散射校正的偏最小二乘回归(PLSR,partial least squares regression)模型对蛋壳强度的预测结果最好,相关系数r为0.86,校正、预测均方根误差分别为4.42、7.53 N;同时蛋壳百分比(蛋壳质量/蛋质量)的PLSR模型的相关系数r为0.92,校正、预测误差分别为0.313%、0.529%;蛋壳厚度的PLSR模型的相关系数r为0.81,校正、预测误差分别为0.0176、0.0234 mm。研究结果表明应用近红外光谱技术预测蛋壳品质是可行的,为蛋壳品质的快速无损检测提供了一种新的方法。  相似文献

高温老化下甜玉米种子活力近红外光谱检测技术研究   总被引:3,自引:0,他引:3  
为利用近红外光谱(NIRS)技术建立一种用于快速无损检测甜玉米种子发芽率、发芽指数、活力指数等活力指标的新方法,选取8个代表性甜玉米品种种子为试材,分别将每品种分成5份(共40份样品);将所有样品置于50℃干燥箱内,分别处理1、2、3、4、5 d后取出,进行生理检测和光谱采集;利用偏最小二乘法回归(PLSR)建立甜玉米种子活力相关指标的定量模型。结果表明,发芽率定量分析模型的性能参数校正集相关系数(Rc)、预测集相关系数(Rp)、交叉验证相关系数(Rcv)、校正集均方根误差(RMSEC)、验证集均方根误差(RMSEP)、交叉验证均方根误差(RMSECV)分别为0.8952、0.8605、0.8503、6.6490、6.6270、7.9091。发芽指数的PLSR模型参数Rc、Rp、Rcv、RMSEC、RMSEP、RMSECV分别为0.9710、0.9366、0.9427、7.4373、24.7921、10.5003。活力指数PLSR模型参数分别为0.9109、0.8962、0.8711、0.8674、0.9489、1.0393。综上,NIRS技术简便、快速、准确可靠,适用于甜玉米发芽率、发芽指数、活力指数等多参数的快速评价。本研究结果为高活力玉米种子筛选提供了一种新的快速分析技术。  相似文献

为建立三疣梭子蟹鲜度的快速评价方法,采用电子鼻获取不同贮藏时间三疣梭子蟹肌肉的气味信息,通过主成分分析(PCA)、线性判别分析(LDA),并结合p H值、TVB-N含量和菌落总数分析三疣梭子蟹蟹肉贮藏不同时间的鲜度变化情况。结果表明,随着贮藏时间的延长,电子鼻传感器的响应强度逐渐增大,PCA和LDA均能有效区分不同贮藏时间的蟹肉,且LDA在以均值体现分类信息方面区分效果更好。基于电子鼻传感器响应值与p H、TVB-N含量和细菌菌落总数的偏最小二乘回归模型(PLSR)的相对分析误差(RPD)分别为2.73、2.50、3.06,说明PLSR分析方法适于对p H值、TVB-N含量和细菌菌落总数进行定量预测;该方法的校正决定系数、预测决定系数均大于0.85,表明电子鼻传感器响应值与p H值、TVB-N含量、细菌菌落总数的相关性较好。本研究为利用电子鼻评价水产品鲜度及保鲜技术效果提供了技术手段及科学依据。  相似文献

含水率是影响板栗贮藏、加工的关键指标之一,该文应用近红外光谱技术对板栗含水率进行快速无损检测。试验对240个板栗样本的带壳光谱和栗仁板栗光谱采用SPXY算法进行样本集划分,利用偏最小二乘法建立含水率定量检测模型,并对微分、多元散射校正、变量标准化等多种预处理方法对建模结果的影响进行比较。结果表明:栗仁和带壳板栗的光谱经一阶微分预处理后所建模型性能最佳,其中栗仁的水分检测模型校正集和验证集的相关系数分别为0.9359和0.8473,校正均方根误差为1.44%,验证均方根误差为1.83%;带壳板栗光谱所建模型校正集和验证集的相关系数分别为0.8270和0.7655,校正均方根误差为2.27%,验证均方根误差为2.35%。受栗壳的影响,带壳板栗光谱模型对含水率的预测精度低于栗仁光谱模型的预测精度。研究表明,近红外光谱分析技术可用于板栗含水率的快速无损检测。  相似文献

为实现婴幼儿配方乳粉中低聚半乳糖(GOS)和低聚果糖(FOS)的快速检测,本研究分别采用标准正态变换(SNV)、多元散射校正(MSC)、归一化(Nor)和Savitzky-Golay平滑(SG)4种方法对获取的乳粉原始光谱进行预处理,再使用变量空间迭代收缩算法(VISSA)和竞争自适应重加权采样算法(CARS)提取具有代表性的特征波长,并建立线性偏最小二乘回归(PLSR)模型和非线性支持向量回归(SVR)模型对婴幼儿配方乳粉中的低聚半乳糖(GOS)和低聚果糖(FOS)含量进行预测。结果表明,乳粉的原始光谱经SNV预处理后,再使用VISSA算法能够有效提取GOS和FOS特征波长,建立的VISSA-SVR非线性模型能够得到较优的GOS和FOS预测结果,GOS的VISSA-SVR模型校正集相关系数为0.998 1,均方根误差为0.050 5,预测集相关系数为0.985 0,均方根误差为0.219 3;FOS的VISSA-SVR模型校正集相关系数为0.994 3,均方根误差为0.053 3,预测集相关系数为0.948 7,均方根误差为0.135 7。本研究可为实现婴幼儿配方乳粉生产过程营养成分...  相似文献

对灰霉病胁迫下茄子叶片过氧化氢酶(CAT)活性的高光谱图像特征进行了研究。采用380~1030nm范围的高光谱图像摄像仪获取健康、轻度、中度、严重染病茄子叶片的高光谱图像信息,基于ENVI软件处理平台提取高光谱图像中对象的漫反射光谱响应特性,并采用平滑、中值滤波、归一化法等预处理方法提高光谱的信噪比。然后采用偏最小二乘回归(PLSR)、最小二乘支持向量机(LS-SVM)和BP神经网络算法来建立叶片高光谱响应特征与CAT活性之间的关系模型。在PLSR模型中,前2个隐含变量能够实现健康、轻度、中度、严重染病茄子叶片的直观定性区分,而基于PLSR模型推荐的9个隐含变量建立的BP神经网络模型的预测集决定系数R2为0.8930,均方根误差为2.17×103。表明基于高光谱图像特性可以实现灰霉病胁迫下茄子病害程度的有效区分,同时证明基于高光谱图像特性的茄子叶片CAT活性的定量检测是可行的。  相似文献

为快速、无损和准确地诊断水稻营养状况,开展了基于高光谱成像技术的寒地水稻叶片氮素含量预测研究。以不同施氮水平下的水稻叶片为研究对象,利用高光谱成像技术,分析拔节期水稻叶片光谱,采用全波段高光谱数据、连续投影算法及分段主成分分析(segmented principal components analysis,SPCA)与相关分析(correlation analysis,CA)相结合的方法建立多种回归分析模型,并对模型进行检验和筛选。结果表明:随着施氮水平提高,水稻叶片反射率在可见光区域降低,在近红外区域升高。在校正集决定系数上,基于多元逐步回归分析的全波段模型较好,校正集决定系数为0.821,校正集均方根误差RMSEC=0.079;在预测集决定系数上,基于SPCA-CA结合多元回归分析的多变量单波段指数、差值指数、双差值指数模型较好,预测集决定系数为0.869,预测集均方根误差RMSEP=0.085。该研究结果为快速检测水稻叶片氮素含量及水稻生长期间精确施肥管理提供了参考。  相似文献

为研究高光谱技术检测霉变玉米中黄曲霉毒素B_1含量的可行性,选择5种不同霉变程度的玉米为试验材料,利用高光谱图像采集系统获得了250个霉变玉米样本的高光谱数据,并进行多元散射校正(MSC)预处理;运用偏最小二乘回归(PLSR)系数来选择特征波长,筛选出7个特征波长,然后利用Fisher判别分析(FDA)分别对全波长和特征波长下霉变玉米进行鉴别分析。结果表明,5组样本在全光谱波段下的FDA鉴别正确率在85%~88%之间,而在特征光谱下的FDA鉴别正确率均在98%以上,说明特征波长能较好地表征不同霉变等级的玉米。神经网络模型优于PLSR模型,其预测集相关系数和均方根误差分别为0.999 9、0.180 9。因此,可认为利用高光谱技术来检测不同霉变程度玉米中的黄曲霉毒素B_1含量是可行的。本研究结果为高光谱鉴别其他农产品提供了重要参考。  相似文献

利用鲜肉的近红外光谱中少量特征波长对其pH值进行预测,可以大幅度降低模型复杂性和计算量,对开发无损检测装置, 实施肉品生产加工过程中pH值监测有重要意义。该文通过连续投影算法(SPA)选择特征波长建立简单多元线性回归模型(SPA-MLR),并对比了SPA-MLR模型与全波段(5 000~10 440 cm-1)偏最小二乘回归模型(PLSR)及逐步线性回归(SMLR)、遗传算法(GA)选择特征波长所建模型的性能。结果表明经连续投影算法提取37个特征波长建立的模型,所用变量数仅占全波段的2.6%,校正集相关系数0.870,校正集均方根误差为0.094,验证集相关系数0.892,验证集均方根误差为0.085;性能与经多元散射校正预处理的PLSR模型接近,但采用变量数明显减少,优于逐步线性回归和遗传算法选择特征波长建立的模型,表明该方法可较好的选择特征波长,建立简单的预测模型。  相似文献

The potential of intrinsic fluorescence spectroscopy was investigated for differentiating between processed grains (flours, pasta, and semolinas) of different wheat cereal products. A total of 59 samples (e.g., 20 complete Kamut, semicomplete Kamut, and soft wheat flours, 28 pasta, and 11 semolinas manufactured from complete Kamut, semicomplete Kamut, and hard wheat flours) were analyzed by front-face fluorescence spectroscopy. Tryptophan fluorescence spectra were scanned between 305 and 400 nm on samples following excitation at 290 nm. The principal component analysis (PCA) performed on flour spectra clearly differentiated complete Kamut and semicomplete Kamut samples from those produced from complete and semicomplete soft wheat flours. The PCA performed on pasta spectra discriminated samples manufactured from complete Kamut and complete hard wheat flours from those made with semicomplete Kamut and semicomplete hard wheat flours. The best discrimination was obtained from tryptophan spectra recorded on semolinas since the four groups were well discriminated. Correct classification amounting to 61.9% was obtained for pasta spectra. A better classification was obtained for flour and semolina spectra since correct classification amounted to 86.7% and 87.9%, respectively. Front-face fluorescence spectroscopy has the potential to be a rapid, low-cost, and efficient method for the authentication of cereal products.  相似文献

Light-induced oxidation of semihard cheese has been evaluated by fluorescence spectroscopy. The cheese was packaged in two packaging materials and exposed to different storage conditions, which included light/dark storage, oxygen availability, and storage time (0, 4, 7, 14, 21, 42, 70, or 84 days). Fluorescence excitation-emission matrices (EEM) were analyzed by PARAFAC, which gave an estimation of the pure excitation and emission spectra of the fluorophores and the concentrations of these. This analysis showed the presence of components such as tryptophan, tyrosine, vitamin A, fluorescent oxidation products, and riboflavin. Effects of packaging material, light or dark storage, and storage time were seen. However, there was no effect of the oxygen availability on the fluorescence measurements. The score values obtained by the PARAFAC models and chemical and physical measurements were analyzed together by principal component analysis (PCA). The loadings showed a separation of the variables into three groups; the first group was related to oxidation, the second group was related to the degradation of both riboflavin and vitamin A, and the third group was linked to the protein structure.  相似文献

The potential of front-face fluorescence spectroscopy for the authentication of unifloral and polyfloral honey types (n = 57 samples) previously classified using traditional methods such as chemical, pollen, and sensory analysis was evaluated. Emission spectra were recorded between 280 and 480 nm (excit: 250 nm), 305 and 500 nm (excit: 290 nm), and 380 and 600 nm (excit: 373 nm) directly on honey samples. In addition, excitation spectra (290-440 nm) were recorded with the emission measured at 450 nm. A total of four different spectral data sets were considered for data analysis. After normalization of the spectra, chemometric evaluation of the spectral data was carried out using principal component analysis (PCA) and linear discriminant analysis (LDA). The rate of correct classification ranged from 36% to 100% by using single spectral data sets (250, 290, 373, 450 nm) and from 73% to 100% by combining these four data sets. For alpine polyfloral honey and the unifloral varieties investigated (acacia, alpine rose, honeydew, chestnut, and rape), correct classification ranged from 96% to 100%. This preliminary study indicates that front-face fluorescence spectroscopy is a promising technique for the authentication of the botanical origin of honey. It is nondestructive, rapid, easy to use, and inexpensive. The use of additional excitation wavelengths between 320 and 440 nm could increase the correct classification of the less characteristic fluorescent varieties.  相似文献

为了探究表面增强拉曼光谱(SERS)法在大肠杆菌(ATCC 25922)、肠炎沙门氏菌(CICC 21540)和单核增生李斯特菌(CICC 10982)快速检测中的可行性,本研究利用两步生长法制备银包金纳米颗粒(Au@Ag NPs)作为增强基底,研究Au@Ag NPs分别与3种致病菌偶联后的拉曼增强效果和稳定性,并对3种致病菌的特征拉曼光谱结合主成分分析(PCA)和层次聚类分析(HCA)进行对比分析。结果表明,合成的Au@Ag NPs颗粒大小均匀,拉曼增强效果明显,可与3种致病菌有效结合,且均有稳定的拉曼响应度;基于大肠杆菌、肠炎沙门氏菌和单核增生李斯特菌的特征拉曼光谱的PCA和HCA分析得出,3种致病菌的拉曼特征光谱之间差异明显,能有效区分。本研究结果为利用Au@Ag NPs作为增强基底的SERS方法快速检测大肠杆菌、肠炎沙门氏菌和单核增生李斯特菌提供了参考基础。  相似文献

Dynamic headspace/gas chromatography-mass spectrometry (GC-MS), front-face fluorescence spectroscopy, and a gas-sensor array technique (electronic nose) have previously detected lipid oxidation in pork back fat or mechanically recovered poultry meat earlier than or at the same time as a sensory panel. The present study was focused on measurement of early lipid oxidation in a more complicated product (freeze-stored, smoked sausages with spices). During the storage time, formation of components contributing to rancid odor and flavor (e.g., hexanal and 1-penten-3-ol) could be monitored with dynamic headspace/GC-MS. The GC-MS data also showed a decrease in 2-furancarboxaldehyde, which could indicate loss of Maillard type components often associated with acidic or meat odor and flavor. The fluorescence spectra were difficult to interpret, probably due to the simultaneous influence from increasing levels of lipid oxidation products and loss of fluorescent Maillard or spice components. The gas-sensor array responses were dominated by signals from, e.g., spice and smoke compounds.  相似文献

Emission and excitation spectra of intrinsic fluorophores present in milk were used to evaluate changes in milk following thermal treatments in the 57-72 degrees C temperature range from 0.5 min up to 30 min. Alternatively, the concentrations of native alkaline phosphatase, lactoferrin, immunoglobulin G, bovine serum albumin, beta-lactoglobulin, and alpha-lactalbumin were determined in the same samples by enzymatic and immunochemical techniques. As principal component analysis applied to the normalized fluorescence spectra successfully discriminated different milk samples according to the temperature and time of thermal treatment, principal component regression was applied to predict the amounts of the native proteins investigated using fluorescence data. The results showed strong correlations between measured and predicted data for alkaline phosphatase and beta-lactoglobulin. This study has demonstrated that front-face fluorescence spectroscopy has a promising potential to become a rapid and nondestructive analytical technique for the evaluation of physicochemical changes in milk induced by low thermal treatment.  相似文献

为了制备风味良好的鲣鱼调味基料,本试验以鲣鱼蒸煮液(STCL)为原料,在酶解和脱腥基础上,采用感官评价、电子鼻技术结合主成分分析(PCA)及线性判别分析(LDA),通过正交试验优化美拉德工艺,并对鲣鱼蒸煮液酶解液美拉德产物进行氨基酸组成成分分析和必需氨基酸营养评价。结果表明,电子鼻技术结合LDA优于PCA,更能客观地区分不同条件下鲣鱼蒸煮液酶解液美拉德反应产物的风味差异;鲣鱼蒸煮液酶解液美拉德反应最优工艺条件为:添加木糖-葡萄糖1:2,在110℃下加热45 min,在此条件下,美拉德产物鱼香味浓郁,色泽呈黄棕色,氨基酸总量为7.036 g·100g-1,必需氨基酸含量为2.888 g·100g-1,必需氨基酸指数(EAAI)为64.034,说明鲣鱼蒸者液美拉德反应产物营养价值较高,是研发海鲜调味料的理想基料。本研究结果为鲣鱼调味基料的开发提供了一定的理论依据。  相似文献

为探讨草麻黄对盐胁迫的适应性,以2年生草麻黄苗为试验材料,采用盆栽法测定在不同NaCl浓度、不同胁迫时间下草麻黄苗各项电阻抗图谱参数(EIS)变化及5种矿质离子含量,并分析各电阻抗参数、相对电导率及矿质离子含量之间的相关性。结果表明,随着NaCl浓度的增加和胁迫时间的延长,草麻黄地上部相对电导率逐渐升高,其中200 mmol·L-1NaCl胁迫28 d以上时的相对电导率显著高于对照,200、300、400 mmol·L-1盐胁迫下的相对电导率分别为对照的1.43、1.52、1.65倍;电阻抗图谱参数高频电阻率(r)、低频电阻率(r1)、胞内电阻率(ri)、胞外电阻率(re)、弛豫时间(τ)及弛豫时间分布系数(Ψ)均呈先下降后上升再下降或先下降后上升的变化趋势,其中r、r1、re、ri、Ψ均在NaCl浓度超过200 mmol·L-1时出现再次下降,且呈不可逆的下降趋势。随着胁迫天数的增加,草麻黄地上部和根中Na+含量逐渐增加,K+含量逐渐下降,K+/Na+显著降低。盐胁迫下,草麻黄地上部Zn、Fe、Mg含量均有不同程度的降低。相关性分析结果表明,K+/Na+与re、r、r1均呈极显著正相关,与ri呈显著正相关,其中 K+/Na+ 与re的相关性最高,相关系数为0.985;5种阳离子总量与re、r、r1均呈显著负相关。盐胁迫影响了草麻黄体内离子的运输和平衡,细胞内外离子浓度的变化引发电阻抗参数的变化,分析相对电导率电阻抗图谱参数的变化发现NaCl胁迫下,草麻黄的忍耐极限浓度为200 mmol·L-1,表明草麻黄具有较高的耐盐能力。本研究结果为盐碱地区草麻黄的推广栽培提供了理论依据。  相似文献

为了控制采后香菇在贮藏过程中的褐变并提高其贮藏品质,本试验采用不同剂量(2、4、6和8 kJ·m-2) 短波紫外线(UV-C)对新鲜香菇进行预处理,研究不同剂量UV-C处理对香菇色泽、总酚含量、多酚氧化酶(PPO)活性、过氧化物酶(POD)活性、超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性、苯丙氨酸解氨酶(PAL)活性、可溶性蛋白含量、还原糖含量、游离氨基酸含量以及香菇表面微生物的影响。结果表明,UV-C处理延缓了香菇色泽的下降,保持了较高的总酚含量,并且有利于POD、SOD、CAT和PAL活性维持在较高水平,抑制了PPO活性,同时可以保持较高的还原糖和可溶性蛋白含量,在一定程度上延缓了蛋白质的降解,而且可以有效抑制微生物的生长,其中4 kJ·m-2 UV-C处理效果最佳。综上所述,UV-C处理可以有效地抑制香菇褐变,延长其贮藏期。本研究结果为香菇的贮藏保鲜提供了理论依据。  相似文献
