This investigation examined the intestinal microbial flora among healthy and intestinal‐diseased seahorses Hippocampus trimaculatus, Hippocampus erectus, and Hippocampus spinosissimus by utilizing polymerase chain reaction‐denaturing gradient gel electrophoresis (DGGE) and densitometric analysis. Results demonstrated that 16 disparate DGGE bands belong to six major bacterial groups, which were Vibrionace, Enterobacteriacea, Rhodobacteraceae, Sphingomonadaceae, Flavobacteriaceae, and Alcaligenaceae. It was found that Vibrionaceae was the dominant population among the healthy and intestinal‐diseased seahorses. Vibrio ponticus strain XJ3 and Vibrio neptunius strain WT82, especially V. ponticus strain XJ3 of high abundance, were identified for the first time in seahorses and concluded to be intestinal disease pathogens because of their co‐existence in three intestinal‐diseased seahorse species and other reported fish or oyster. In comparison, uncultured bacterium clone Alcaligenaceae, Vibrio sp., uncultured bacterium clone Rhodobacteraceae, Serratia nematodiphila strain, and Serratia marcescens strain comprised the basic intestinal bacterial flora of all healthy seahorses. This study is the first to report the presence of S. nematodiphila in seahorses.  相似文献

Using polymerase chain reaction amplification of 16S rDNA coupled to denaturing gradient gel electrophoresis (DGGE) and sequencing of isolated amplicons, we investigated the microbiota of the intestinal digesta and mucosal surface in mangrove red snapper cultured in a cage aquaculture area in Daya Bay. A total of 14 sequences were characterized by phylogenetic analysis. Among the bacterial species determinated from sequences, the γ‐Proteobacteria group (64.25%, nine species) dominated absolutely in fish intestines. Others belonged to Spirochaetes (14.3%, two species), Cyanobacteria (14.3%, two species) and Firmicutes (7.15%, one species). However, the bacteria were identified as uncultured accounting for 28.6% (four species). The apparent bacterial richness (calculated as the numbers of DGGE bands) was significantly higher in digesta than that in mucosal tissue samples (P<0.05). There existed five dominant individual populations including one unknown species of Firmicutes, Arthrospira sp., Vibrio metschnikovii, Vibrio harveyi and Vibrio sp. in intestinal digesta, and in contrast, only three dominant individual populations including Vibrio natriegens, V. harveyi and Vibrio sp. in intestinal mucosal surface. The results indicated that the microbiota in intestinal digesta was significantly different from that in mucosal surface.  相似文献

The microenvironment of bacteria associated with the gastrointestinal tract of an animal influences the host in many ways, including the metabolism of several nutrients. Isolation, molecular characterization and enzymatic activities of culturable bacteria associated with the gastrointestinal tract of hatchery‐reared oceanic yellow seahorse, Hippocampus kuda, were investigated in the present study. 16S rRNA sequencing of bacterial isolates yielded 10 different bacterial genera belonging to two major groups: proteobacteria (62.9%) and firmicutes (37.03%). The predominant microbial flora belonged to the genera Vibrio spp., Enterovibrio sp. and Bacillus spp. Burkholderia cenocepacia, hitherto unreported from the gastrointestinal tract of marine fish, was retrieved from the seahorse gut, albeit in small numbers. Enzymatic studies indicated that the bacterial isolates possess the ability to degrade lipids, cellulose, xylan, starch and proteins. Lipase activity was exhibited by a majority of the isolates (70.4%), indicating that the bacterial flora associated with the intestinal tract of seahorses is capable of digesting foods rich in lipids. The possible implications for the role of these bacteria in the digestion and physiological processes are discussed. To our knowledge, this is the first time that the characterization and enzymatic activities of bacteria associated with the intestinal tract of farmed seahorses have been reported.  相似文献

在育肥饲料中添加虾青素会影响中华绒螫蟹肠道及鳃内的可培养优势菌群以及机体的免疫力,本实验旨在探讨饲料中虾青素含量及养殖水体对其肠道和鳃部菌群的影响。采用不同虾青素含量的饲料(0.00、26.60、41.62、81.37和75.35 mg/kg,分别对应饲料1#~5#)对雄性中华绒螫蟹进行育肥70 d后,利用基础培养基和选择性培养基对其肠道、鳃及养殖水体中的细菌进行传统分离和纯化,所得菌株进行16S rRNA测序,再进行同源性分析后做系统进化树。结果发现,在分离获得的106株细菌(登录号:KU570293~KU570368,KU570370,KU570372~KU570383,KU601302~KU6013 16,KU720553)中,92株为优势菌株,其中,肠道、鳃部和水体各29、32和31株。肠道中的可培养优势细菌属于柠檬酸杆菌属、假单胞菌属和气单胞菌属;而鳃中可培养优势细菌属于柠檬酸杆菌属、芽孢杆菌属、假单胞菌属和气单胞菌属;养殖水体中可培养优势细菌属于芽孢杆菌属、气单胞菌属、柠檬酸杆菌属、假单胞菌属和黄杆菌属。饲料2#组肠道中可培养细菌总数最高(1.06×10~8cfu/g);饲料5#肠道中的潜在致病菌数量最低,但其可培养细菌总数显著高于其他饲料组;饲料3#组肠道和鳃中潜在致病菌数量相对较高。研究表明,饲料中添加不同含量的虾青素能够显著影响雄性中华绒螫蟹肠道和鳃中的菌群构成,但对水体中可培养细菌数量无显著影响。本研究首次对投喂不同含量虾青素饲料的育肥期雄性中华绒螫蟹肠道、鳃及养殖水体可培养的优势细菌数量和组成进行分析,探究了饲料中虾青素含量以及养殖水体与蟹肠道内可培养细菌之间的联系,为中华绒螫蟹营养免疫和菌群调控提供了理论基础。  相似文献

The trade in seahorses for aquarium fishes is contributing to the depletion of many wild populations of these animals. Many seahorses are sold to replace those that have died in captivity as a result of husbandry problems. It can be particularly difficult to rear the young seahorses, because of their need for varied live food and their vulnerability to disease. We here report a pilot study on rearing broods from males of three species (H. fuscus, H. barbouri, and H. kuda) that had mated in the wild and gave birth in captivity. The new-born seahorses were fed an initial diet of enriched Artemia until 7 days, after which copepods were added to the diet. From 5 weeks, frozen mysids were gradually phased in to replace both other food items. Scrupulous hygiene was maintained. We achieved 100% survival of the partial broods we reared for all three species and achieved life cycle closure in two of these during the experimental period. Of the three species, H. kuda grew to be largest and longest, and H. barbouri grew least. However, H. kuda were the slowest to mature and reproduce while H. fuscus (intermediate in growth) were the fastest. Techniques used in this work should be more generally applicable, both for aquarium husbandry and for small-scale aquaculture to help provide alternative incomes for small-scale fishers who are otherwise dependent on catching wild seahorses.  相似文献

为研究尼罗罗非鱼肠道和池塘养殖环境中菌群结构变化与链球菌病暴发的相关性,实验采用16S r DNA高通量测序方法对比分析发病和健康池塘水体、底泥和尼罗罗非鱼肠道菌群的结构特征。结果显示,底泥中微生物多样性最高,水体和肠道中微生物多样性次之;肠道菌群与水体中菌群的相似性较高,而与底泥中菌群相似性较低;发病组的尼罗罗非鱼肠道和池塘底泥中微生物的多样性均低于健康组,而发病组池塘水体中微生物多样性高于健康组。OTU聚类分析发现,发病与健康尼罗罗非鱼肠道微生物差异极显著,大部分健康尼罗罗非鱼肠道微生物样品单独聚为一支。菌群结构组成分析结果表明,虽然发病组和健康组水体或底泥中优势菌群结构组成的差异较小,但在非优势菌群中存在一定差异,发病组水体中变形菌门和梭杆菌门的比例显著高于健康组水体,疣微菌门和浮霉菌门的丰度显著低于健康组水体;发病组池塘底泥中厌氧粘细菌属、甲烷丝菌属和枝芽孢菌属等具有降解有机质和生态修复功能,菌群的丰度显著低于健康组底泥,而具有致病能力的曼氏杆菌属丰度却显著高于健康组底泥。发病组尼罗罗非鱼肠道菌群中链球菌属、分枝杆菌属和曼氏杆菌属等致病菌群的丰度显著高于健康组尼罗罗非鱼肠道,而乳球菌属、鲸杆菌属和红球菌属等益生菌的丰度显著低于健康尼罗罗非鱼肠道。无乳链球菌普遍存在于发病和健康养殖环境,以及尼罗罗非鱼肠道中,无乳链球菌的丰度在发病和健康养殖环境之间无显著差异,但其在发病尼罗罗非鱼肠道中的丰度要显著高于健康组。研究表明,尼罗罗非鱼链球菌病发病池塘水体和底泥中有益菌丰度降低和致病菌丰度的升高反映其养殖环境出现恶化,尼罗罗非鱼链球菌病的暴发与肠道微生态平衡破坏后无乳链球菌丰度增加密切相关,但肠道中无乳链球菌丰度的剧增与养殖水体和底泥中该病原菌的丰度水平之间并无直接关联。  相似文献

To assess the effect of addition of probiotics and carbon source on the microbial community structure within the shrimp culture, an enclosure system simulating a high density, intensive aquaculture of Litopenaeus vannamei was set up in Dianbai, Maoming, Guangdong province, China. The phylogenetic profiles of microbial communities in the enclosure system after addition of Bacillus and molasses were studied in the middle and final culture season (on the 45th, 75th and 95th day) using polymerase chain reaction‐denaturing gradient gel electrophoresis. A total of four groups, each containing a triplicate were set up: Group A (control), Group B (64% of daily feed was provided as molasses with Bacillus sp.), Group C (50% of daily feed amount provided as molasses with Bacillus sp.) and Group D (64% of daily feed amount as molasses alone). The results showed that α‐Proteobacteria, δ‐Proteobacteria, Bacteroidetes and Actinobacteria were the dominant bacteria. Moreover, the relative proportion of α‐Proteobacteria increased and that of Bacteroidetes decreased gradually with time. Pathogenic Flexibacter was the dominant genus in Group A on the 45th day, while the functional strains (Sphingomonas sp., Erythrobacter sp., Roseobacter sp., and Rhodobacter sp.) were detected multiple times in Groups B and C. Bacillus sp. was the cultivable, dominant bacterium in Groups B and C on the 95th day. Therefore, the combined use of Bacillus and molasses helped increase diversity of the microbial community, effectively inhibit pathogens, and promote the formation and development of a beneficial microbial community structure in the high density, intensive aquaculture of L. vannamei.  相似文献

To characterize the composition of the intestinal bacterial flora in cage‐cultured adult small abalone, Haliotis diversicolor, we used both culture‐dependent and culture‐independent approaches. In addition, in order to investigate the presence of probable pathogens, the intestinal bacterial community structures of healthy and diseased adult abalones were compared using the denaturing gradient gel electrophoresis (DGGE) fingerprinting technique. Bacteria isolated from the healthy abalone intestine belonged to the genera Vibrio, Flammeovirga, Shewanella, Persicobacter and Paraferrimonas. From the 16S rRNA gene clone library analysis, the intestinal bacterial community comprised two groups: the Proteobacteria and Tenericutes. Betaproteobacteria (18.48%), Deltaproteobacteria (30.43%) and Mollicutes (35.87%) were the most dominant components in the clones and the Mollicutes were mainly Mycoplasma‐affiliated clones. The DGGE band pattern demonstrated that more bands appeared in the healthy abalones (H group) than in the diseased abalones (D group). Although few potential pathogens were detected from the D group, a clustering dendrogram indicated that individuals of the H and D groups (except for D6) formed two separate clusters. An unidentified Deltaproteobacteria species and Mycoplasma sp. were the dominant components in the intestine of both the H and the D group.  相似文献

The lined seahorse, Hippocampus erectus, is an Atlantic species, which mainly inhabits shallow sea beds or coral reefs. Seahorse has become very popular in China due to its wide use in traditional Chinese medicine (TCM). However, the aquaculture of the lined seahorse has been threatened by a variety of diseases, especially bacterial infections. Skin ulcer syndrome becomes more and more common in the culture of seahorses, leading to skin ulcer, liver erosion and minority tail rotting. These diseased seahorses die in only 72 hr, causing huge economic losses in aquaculture. Pathogen HMT‐1 was isolated from the lined seahorses with skin ulcer syndrome, which was identified as Vibrio harveyi. The median lethal dose (LD50) of HMT‐1 was 2.88 × 108 cfu/ml. The pathogenic mechanism of HMT‐1 was preliminarily studied, and the pathological changes of the diseased lined seahorses were also investigated. Moreover, the activities of ACP, complement 3, SOD and LZM at different time post injection were measured by commercial kits. Our findings can provide reliable reference for diagnosis and treatment in the future.  相似文献

The effect of stocking density and photoperiod on Hippocampus abdominalis was examined in this study. Stocking densities of 45, 30, 15 and 5 seahorses 3 L?1 were tested on newborns. Growth and survival were independent of stocking density. A second stocking‐density experiment aimed to remove the effect of an early mortality experienced in experiment 1 using older juveniles at 25, 15 and 5 seahorses 3 L?1. There were no differences in the parameters measured. Also, the effect of photoperiod was investigated on seahorses cultured under 24:00, 16:08 and 08:16 (L:D) photoperiods. A single Artemia meal was delivered at 10:00 hours. Survival and body growth in 16:08 and 08:16 hours were higher than in 24:00 hours. In a second experiment, seahorses were cultured in the photoperiods and conditions described for experiment 1, except they were fed twice the amount in two meals delivered at 10:00 and 16:30 hours. The seahorses in 16:08 hours showed better growth than the other treatments, but there were no differences in survival. These results suggest that early juvenile H. abdominalis can be cultured at higher stocking densities than previously reported, without compromising growth and survival, and when feeding was not limiting, grew better in an extended photoperiod (16:08) but not in 24:00 hours.  相似文献

The seahorse has been used for thousands of years as an important traditional Chinese medicine (TCM) in China. Many species of wild seahorse have become endangered, but demand for them continues to grow rapidly. Therefore, it is urgent to cultivate seahorses to relieve the strain on natural populations and meet the market demand for this TCM. In this study, medicinal components of wild and cultured yellow seahorses (Hippocampus kuda Bleeker) were analysed and compared. Different parts of the body (cortex and bone), male and female specimens, and seahorses of different ages were analysed. Crude protein, crude fat, trace elements, calcium, amino acid, fatty acid, steroid and cholesterol content were measured. The results showed that moisture content accounted for 70% of wet weight and ash content increased with growth. Crude protein and fat contents were higher in cultured seahorses than in wild specimens, and the content of Zn in cultured seahorses (93.26 ± 10.44%) was significantly higher than that of same‐aged wild seahorses (74.20 ± 3.83%). Amino acid and fatty acid contents of cultured seahorses also were higher than those of wild seahorses. These findings show that cultured seahorses can also be used in TCMs and that 1‐year‐old cultured seahorses have the best properties.  相似文献

为研究大豆皂甙对大菱鲆幼鱼生长性能、消化酶活性、肠道组织结构完整性和肠道菌群结构的影响,在以鱼粉为蛋白源的基础饲料中分别添加0%和0.3%的大豆皂甙,配制成鱼粉组和大豆皂甙组2种等氮等脂的实验饲料来投喂体质量为(4.63±0.01) g的大菱鲆进行12周的摄食生长实验。结果显示,饲料中添加0.3%的大豆皂甙对大菱鲆的生长性能没有产生显著影响,但显著降低了胃蛋白酶及肠淀粉酶活性;2组实验中大菱鲆肠道组织形态无明显差异,但大豆皂甙组肠道紧密连接蛋白的基因表达量显著降低。肠道菌群分析结果显示,大菱鲆肠道中相对丰度最高的门和属分别为变形菌门和盐单胞菌属。LEfSe和MetaStat分析显示,饲料中添加大豆皂甙后显著提高了大菱鲆肠道内优势菌(变形菌门及希瓦氏菌属),皂甙水解相关的肠道微生物(鞘脂单胞菌属、普氏菌属、栖瘤胃普雷沃菌以及普通拟杆菌)及潜在致病菌(莫氏杆菌属和发光杆菌属)的相对丰度,同时显著降低了CaenimonasNiastella和条件致病菌罗尔斯通菌属的相对丰度。研究表明,0.3%大豆皂甙抑制了大菱鲆消化酶活性及肠道紧密连接蛋白的基因表达,且引起了大菱鲆肠道菌群结构的显著改变。因此,大豆皂甙对鱼类肠道健康尤其是肠道菌群的影响不容忽视,值得进一步研究。  相似文献

  1. Rafting has been proposed as a dispersive mechanism for some species of seahorses. Juvenile Hippocampus patagonicus rafting on the sea surface have been reported. This life stage has high mortality in nature due to ecological and environmental factors usually affecting juvenile planktonic teleosts (e.g. predation).
  2. In this study, 50 juvenile seahorses were captured while rafting at the beginning of the reproductive season (December). They were grown out under a standard protocol in a recirculating aquaculture system for 120 days until they reached maturity.
  3. During this period, only three seahorses died (6%). Mature seahorses were 65 ± 7 mm average height and showed variable coloration and pigmentation pattern (n = 47; 94%).
  4. The availability of juvenile seahorses during the reproductive season and their high survival performance ex situ highlight the feasibility of developing protocols in which juvenile seahorses serve as a novel source of broodstock.

There are few reports in the literature about the isolation of bacteria from whale intestine. In this report, we counted colony-forming units in the feces obtained from three female common minke whales (Balaenoptera acutorostrata). The number of colony-forming units ranged from (2.2 ± 0.4) × 105 to (8.9 ± 2.0) × 108 per gram (wet weight) of excrement. 16S rRNA gene sequences of 141 isolates were determined. These strains were identified as Enterococcus faecalis, Enterococcus sp., Enterobacter cloacae, Enterobacter sp., Escherichia coli, Edwardsiella ictaluri or Clostridium sp. The data suggested that the facultative anaerobic population of the intestinal bacterial flora of the minke whale was similar to that of ground mammals.  相似文献

This study aims at evaluating the Patagonian seahorse, Hippocampus patagonicus, feeding behavior in captivity with regard to type and size of prey selectivity and time‐investment‐related activities for searching and capture. Experiments in aquaria were carried out under two different treatments, with and without refuge for prey. Different diets composed of amphipods (Melita palmata and Ampithoe valida), juvenile decapods (Neohelice granulata) from natural environments, and artificially cultured Artemia salina. The eaten prey’s size was analyzed and experiments with size‐controlled prey were carried out. There were neither significant differences in the prey quantity ingested between males and females nor between treatments (with or without refuge). However, significant differences were found between the quantities of each prey consumed. Amphipods and A. salina were consumed in greater quantity (P < 0.05) when compared with juvenile decapods. Experiments with A. salina of different sizes did not show significant differences on seahorses’ preference. N. granulata was highly avoided and seahorses had lower capture efficiency over this prey. Behavioral observations showed that more than 50% of the times seahorses were resting and ambushing their prey. In captivity, with optimal conditions for predation, the Patagonian seahorse modifies its opportunistic behavior, feeding selectively, and bases its food preferences on prey size and shape .  相似文献

In Tasmania, commercial seahorse culture takes place in tank systems in which approximately 75% of the water is exchanged daily from the Tamar River estuary. The aim of this study was to examine the effect on survival of early juvenile pot‐bellied seahorses, Hippocampus abdominalis, directly transferred and cultured for 9 d in salinities of 5, 10, 15, 20, 25, and 32 g/L. Direct transfer of seahorses to 5 g/L salinity resulted in 100% mortality within 7 d. After 9 d, an improvement in survival was recorded in seahorses cultured in 10 and 15 g/L compared to those cultured in 25 and 32 g/L salinities. This study is the first to report on the tolerance of H. abdominalis in captivity at salinities as low as 10 g/L in early life stages without compromising survival.  相似文献

为探究江鳕(Lota lota)肠道菌群结构特征,采用MiSeq 16S rRNA高通量测序技术对3龄、4龄、5龄野生江鳕的雌、雄样本进行了微生物测序和分析。结果显示,3个年龄组江鳕的肠道菌群丰度及多样性差异不显著(P>0.05),同一年龄雌、雄江鳕间的肠道菌群丰度及多样性差异不显著(P>0.05)。江鳕肠道内容物的优势菌门为变形菌门和厚壁菌门,与鱼类肠道中最普遍和共存的细菌类群相同。Mycoplasma(支原体属)存在于所有样品中,且为优势菌群,其功能有待进一步研究。  相似文献

为获得鱼源益生乳酸菌,本研究从健康鲫肠道内分离鉴定得到38株乳酸菌,并选择其中8株乳酸菌进行体外益生特性分析。结果显示,8株乳酸菌均能在p H=4.5和10%鲫胆汁环境中存活,对嗜水气单胞菌、豚鼠气单胞菌、无乳链球菌和副溶血弧菌均有较强的抑菌能力,但菌体表面疏水性和自凝集能力具有菌株特异性。选择5株疏水性较高(63%~89%)、自凝集能力强(37%~45%)的乳酸乳球菌进行体外黏附能力测定,结果显示,黏附能力在菌株间具有差异性(4.5%~7.9%),但均能显著降低嗜水气单胞菌NJ-35的体外黏附率,黏附抑制率达30%~35%;最后对筛选出的3株高黏附力的乳酸乳球菌进行安全性评价,发现3株菌对鲫均无致病力。鱼源乳酸菌的筛选,为鲫养殖生产中益生菌的实际应用提供了理论依据。  相似文献

为了发展渔业生产,人们将大量自然湖泊通过围网的方式投放鱼苗进行养殖。目前,这种养殖模式对鱼类群落的影响没有得到有效的评估。本研究于2016年在华阳河湖围网养殖区和非养殖区分别设置采样点对鱼类进行季度调查,探讨围网养殖对鱼类群落结构的影响。结果显示,在围网养殖区共采集鱼类6目12科35属46种,非养殖区采集鱼类6目11科40属57种,其中以鲤形目种类最多,分别占养殖区和非养殖区鱼类总数的65.22%和63.16%。非养殖区的优势种为?和短颌鲚,而鲢、鳙是围网养殖区的优势种。与非养殖区相比,围网养殖区山溪河流性鱼类百分比从21.05%降低至15.22%。通过双因素方差分析解析养殖与季节对鱼类群落结构的影响,发现围网养殖区的鱼类密度、物种数、优势度指数都显著低于非养殖区,而均匀度指数显著高于非养殖区。通过SIMPER分析得出,造成养殖区和非养殖群落结构差异的重要贡献物种为短颌鲚、?、鲤、太湖短吻银鱼、鲫、鱵、鲢和鳙。  相似文献
