The ruminal degradability, intake, and metabolism of diets differing in their relative rate of energy and nitrogen release in the rumen were characterized prior to their use in a study of the effects of high peripheral levels of ammonia on reproductive function in cattle. In a 2 x 2 factorial experiment, replicated four times, 16 heifers were offered isocaloric and isonitrogenous diets containing two sources of fermentable carbohydrate, fiber (slow energy release, SE) or starch (fast energy release, FE), and two rates of nitrogen release, which were either synchronous (S) or asynchronous (A) to that of energy release. Throughout the experiment, the amount of feed offered was held constant, at a level equivalent to 1.5 x maintenance. Four ruminally fistulated sheep were used to determine the in situ degradability of these diets. The 16 heifers were bled before feeding at 0800 and at 0900, 1000, 1100, 1200, 1400, and 1600 on d 0 (introduction to dietary treatments) and on d 4, 7, 11, 14, 21, and 28. Diet refusals were recorded at hourly intervals after feeding. The rapidly degradable nitrogen fraction of the SE:A and FE:A diets was greater than that of the SE:S and FE:S diets. Postprandial jugular plasma ammonia levels rose to a peak of around 300 micromol/L in heifers offered the SE:A and FE:A diets but did not rise in heifers offered the SE:S and FE:S diets. All feed offered was consumed within 1 h on diets SE:S and FE:S throughout the experiment. The proportion of feed consumed within 1 h of feeding declined from 100% on d 0 to around 70 and 56% by d 21 for heifers given the SE:A and FE:A diets, respectively. Peak postprandial plasma ammonia levels were accordingly lower, at around 160 micromol/L. Plasma urea levels averaged 7 mmol/L and were unaffected by dietary treatment. High plasma ammonia levels were associated with a suppression in the normal postprandial rise in insulin. There was no significant metabolic adaptation to high-ammonia-generating diets, and heifers given these diets modified their pattern of intake in an apparent attempt to avoid excessively high levels of plasma ammonia.  相似文献

To determine whether long-term administration of growth hormone (GH)-releasing factor (GRF) and(or) thyrotropin-releasing hormone (TRH) alters ovarian follicular fluid (FFL) concentrations of insulin-like growth factor-I (IGF-I), progesterone, and estradiol (E2), and follicular growth, Friesian x Hereford heifers (n = 47; 346 +/- 3 kg) were divided into the following four groups: control (vehicle; n = 11); 1 micrograms GRF (human [Des NH2 Tyr1, D-Ala2, Ala15] GRF [1-29]-NH2).kg-1 BW.d-1 (n = 12); 1 microgram TRH.kg-1 BW.d-1 (n = 12); or GRF + TRH (n = 12). Daily injections (s.c.) continued for 86 d. On d 89, heifers that had been synchronized were slaughtered and ovaries were removed. Follicles were grouped by magnitude of diameter into the three following sizes: 1 to 3.9 mm (small, n = 55), 4.0 to 7.9 mm (medium, n = 63), and greater than or equal to 8 mm (large, n = 71). Growth hormone-releasing factor and(or) TRH did not affect (P greater than .10) IGF-I concentrations in FFL of any follicle size group. Growth hormone-releasing factor increased (P less than .06) size (means +/- pooled SE) of large follicles (14.7 vs 13.0 +/- .6 mm). Growth hormone-releasing factor also increased (P less than .05) progesterone concentrations 4.4-fold above controls in FFL of medium-sized follicles but had no effect on progesterone in FFL of the small or large follicles. Thyrotropin-releasing hormone did not alter FFL progesterone or E2 concentrations in any follicle size group. We conclude that the GRF and(or) TRH treatments we employed did not affect intra-ovarian IGF-I concentrations, but GRF may alter steroidogenesis of medium-sized follicles and growth of large follicles.  相似文献

Postpubertal beef heifers (n = 55) were used to examine the effects of high-fat diets, independently of energy intake, on nonesterified fatty acid and lipoprotein metabolic patterns, ovarian follicular dynamics, and embryo recovery/viability after FSH superstimulation. High-lipid (HL) diets (5.4% added fat) increased (P < .01) serum concentrations of cholesterol, but not of nonesterified fatty acids, during the 35-d period before FSH treatment. Development of medium-sized (5 to 9.9 mm) follicles was enhanced (P < .05) during this period in heifers fed the HL diet. The HL diet increased total cholesterol (P < .05) and progesterone (P = .14) concentrations in follicular fluid obtained at ovariectomy (n = 10) 60 h after the onset of FSH treatment, but neither estradiol-17 beta nor androstenedione was affected. Granulosa cells recovered from FSH-induced, estrogen-active follicles in heifers fed the HL diet produced greater quantities of progesterone (P = .06) and less estradiol-17 beta (P < .05) in vitro than did granulosa cells from heifers fed the normal lipid diet. Dietary treatment did not influence FSH-stimulated recruitment of medium and large follicles, number of ovulations, embryo recovery, or embryo viability. Data suggest that increments in dietary fat intake can alter specific aspects of ovarian steroidogenic potential and can increase the population of medium-sized follicles theoretically available for maturation and harvest during the estrous cycle. However, conditions that limited the latter process in the current experiment are not understood and require further investigation.  相似文献

Peripheral serum progesterone concentrations were evaluated in beef heifers following breeding collected on d 6 +/- 1, 9 +/- 1 collected on d 6 +/- 1, 9 +/- 1 and 12 +/- 1 (estrus = d 0) after the puberal estrus of all heifers and after the third estrus of E3 heifers. Progesterone concentrations were higher (P less than .05) for heifers in E1 compared with heifers in E3 on d 6, 9 and 12 after breeding to a fertile bull. Progesterone concentrations on d 6, 9 and 12 did not differ (P greater than .10) between pregnant heifers in E1 and E3; however, non-pregnant heifers in E1 had higher (P less than .05) concentrations of progesterone compared with non-pregnant heifers in E3 on each day. Concentrations of progesterone did not differ (P greater than .10) between non-pregnant heifers in E1 and heifers of E3 during their puberal cycle. Pregnant heifers in E1 and E3 had higher (P less than .05) concentrations of progesterone on each day than non-pregnant heifers in their respective treatments. There were no interactions (P greater than .10) between treatment, pregnancy status and day-of-estrous cycle for concentrations of progesterone. Results of this study indicated that luteal function differed between heifers that failed to conceive at their puberal estrus and heifers that failed to conceive at third estrus. However, concentrations of progesterone did not differ between heifers that conceived at puberal or third estrus. The relationship of changes in luteal function from the puberal through the third estrous cycle and pregnancy is not clear.  相似文献

The purpose of this experiment was to determine the ovulation rate after treatment with human chorionic gonadotropin (hCG) in two groups of gilts characterized by different ovarian morphology: grape-type (GT; n = 11) and honeycomb-type (HT; n = 7). At 170 d of age (d 0), gilts were examined by laparoscopy and ovarian type was determined by the distribution of macroscopic follicles present on the ovarian surface. Five to ten minutes after surgery, each gilt received a single injection (i.m.) of 750 IU of hCG. At d 0, GT ovaries had a greater number of large follicles (greater than or equal to 6 mm) than HT ovaries (10.0 +/- .5 vs 2.6 +/- .3; P less than .05), whereas HT ovaries had more small follicles (1 to 3 mm; HT: 42.3 +/- .8 vs GT: 26.7 +/- .9; P less than .05) and total follicles (HT: 59.4 +/- 2.3 vs GT: 52.2 +/- 1.5; P less than .05), although numbers of medium follicles (4 to 5 mm) were similar (GT: 15.6 +/- .8 vs HT: 14.6 +/- 1.7; P greater than .10). Number of induced corpora lutea (CL) per ovary was greater (P less than .05) in gilts with GT ovaries (10.59 +/- 2.9 CL) than in gilts with HT ovaries (5.21 +/- .66 CL). Total weight of luteal tissue (LT) per ovary and serum progesterone concentrations 8 d after induction of ovulation were greater in GT gilts than in HT gilts (GT: 6.37 +/- 1.09 g vs HT: 3.31 +/- .49 g for LT, P less than .05; GT: 21.08 +/- 4.76 ng/ml vs HT: 13.40 +/- 2.05 ng/ml for progesterone, P less than .07).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The objective of this study was to compare performance, physiological, and reproductive responses of beef heifers consuming forages differing in nutritional quality and offered a low-starch energy supplement at 2 different frequencies. Forty-eight Brahman × British heifers (initial age = 294 ± 3 d) were allocated into 1 of 16 drylot pens (3 heifers/pen) which were randomly assigned to receive, in a 2 × 2 factorial arrangement of treatments: 1) low-quality hay [LQ; stargrass (Cynodon nlemfuensis) with 8% CP and 81% NDF, DM basis] and daily supplementation (S7); 2) LQ and supplementation 3 times weekly (S3); 3) medium-quality hay [MQ; bermudagrass (C. dactylon) with 12% CP and 74% NDF, DM basis] and S7; and 4) MQ and S3. Throughout the study (d 0 to 120), hay was offered in amounts to ensure ad libitum access, and a supplement based on soybean hulls and wheat middlings was offered at weekly rates of 15.8 and 7.9 kg/heifer (DM basis) for LQ and MQ, respectively. Forage and total DMI were evaluated daily, from d 20 to 26, d 34 to 40, and d 48 to 54. Blood samples were collected weekly for determination of plasma progesterone to evaluate puberty attainment. Blood samples were also collected daily, from d 13 to 16, d 27 to 30, d 41 to 44, and d 55 to 58 for determination of plasma urea nitrogen (PUN), glucose, insulin, IGF-I, and NEFA. On d 60, heifers were reallocated by treatment into 4 paddocks and exposed to Angus bulls (1:12 bull:heifer ratio) until d 120. Date of conception was estimated retrospectively by subtracting gestation length (286 d) from the calving date. Heifers receiving S7 had similar (P = 0.52) ADG compared with S3 heifers (0.27 vs. 0.25 kg/d). Heifers provided S7 had less daily variation in hay DMI and plasma concentrations of glucose, NEFA, and IGF-I compared with S3 cohorts (supplementation frequency × day interaction; P < 0.01). Similarly, heifers offered MQ and LQ and receiving S7 had less daily variation in total DMI, energy and protein intake, and plasma concentrations of PUN compared with heifers offered MQ and LQ and receiving S3 (hay quality × supplementation frequency × day interaction; P < 0.01). Attainment of puberty and pregnancy were hastened in S7 heifers compared with S3 heifers (supplementation frequency × week interaction; P < 0.02). Therefore, reproductive development of beef replacement heifers consuming diets based on low- and medium-quality forages are enhanced when low-starch energy supplements are offered daily instead of 3 times weekly.  相似文献

Tarentaise heifers fitted with a rumen cannula (539 +/- 7.5 and 487 +/- 15.7 kg avg initial BW in Exp. 1 and 2, respectively) were used in two Latin square metabolism experiments having 2 x 2 factorial treatment arrangements to determine the effects of supplementation with Aspergillus oryzae fermentation extract (AO) or laidlomycin propionate (LP) on intake, digestion, and digestive characteristics of Neotyphodium coenophialum-infected (IF) or uninfected (FF) tall fescue (Festuca arundinacea) hay diets consumed ad libitum. Heifers were housed in individual stanchions in a metabolism facility with ambient temperatures controlled to range between 26.7 and 32.2 degrees C daily. Total feces and urine were collected for 5 d following a 21-d dietary adaptation period. In situ DM and NDF disappearance and ruminal fermentation characteristics were also determined. In Exp. 1, DMI was 24% greater (P < 0.01) by heifers offered FF than by those offered IF (6.7 vs 5.4 kg/d). Heifers fed 2 g/d AO tended (P = 0.09) to consume 4% more DM than those fed a diet without AO. Degradable DM and NDF fractions of IF were greater (P < 0.01) than those of FF, but AO supplementation did not affect situ disappearance (P > or = 0.42). In Exp. 2, DMI was 18.9% greater (P < 0.01) by heifers offered FF than by those offered IF (6.6 vs 5.5 kg/d). Heifers fed LP (50 mg/d) consumed 10.6% less (P < 0.05) DM than those not fed LP (5.7 vs 6/5 kg/d). Digestibility of NDF tended to be greater (P = 0.08) and digestibility of ADF was greater (P < 0.05) from FF than from IF. Conversely, apparent N absorption (%) was greater (P < 0.05) from IF than from FF. Heifers fed LP had lower (P < 0.05) ADF digestibility than those not fed LP. In situ degradable DM and NDF fractions were greater (P < 0.01) from IF than from FF. Diets supplemented with LP had higher (P < 0.01) indigestible DM and NDF fractions than those without LP. Propionic acid and total VFA concentrations were greater (P < 0.05) from heifers offered FF than from those offered IF and from heifers fed LP than from those not fed LP. Therefore, it appears the major effect of N. coenophialum was a reduction in forage intake and total-tract fiber digestibility in certain situations. Response to the feed additives was similar whether heifers were offered IF or FF and no evidence was apparent that either additive would improve performance substantially by animals consuming low-quality fescue hay diets.  相似文献

The frequency of occurrence of behavioral estrus without subsequent development of functional luteal tissue (termed nonpuberal estrus, NPE), was determined in 43 Simmental X Hereford-Brahman heifers. Blood samples were collected weekly from the start of the study to first behavioral estrus and then daily from d 1 (d 0 = estrus) through d 14 following first and subsequently observed estrous behaviors. All blood samples were analyzed for serum progesterone (P4) concentrations by radioimmunoassay. More heifers (62.8%) exhibited NPE than had luteal development after their first behavioral estrus (37.2%). There was a tendency for fewer light-weight heifers (less than or equal to 240 kg at the start of the experiment) to exhibit a puberal first estrus compared with the heavy-weight (greater than 240 kg at the start of the experiment) heifers (31.2% vs 68.8%, respectively; P = .12). Heifers that had a puberal first estrus were older (376 +/- 12 d vs 334 +/- 9 d, P less than .05) compared with heifers that had NPE. Weight at first behavioral estrus was similar between heifers that had a puberal first estrus and those that had NPE (298 +/- 8 kg and 289 +/- 6 kg, respectively). More heifers that had a puberal first estrus also had an elevation in serum P4 concentrations before that first estrus (64.3% vs 20.0%, P less than .05), and the serum P4 elevation was greater (2.5 +/- .4 ng vs 1.2 +/- .1 ng, P less than .05) than heifers that had NPE. A study was done to test whether ovulatory follicles destined to form subfunctional corpora lutea differed from normal ovulatory follicles in steroidogenic function. Twenty-five ewes were treated with prostaglandin F2 alpha on d 11 of the estrous cycle, then unilaterally ovariectomized before (n = 13) or after (n = 12) the surge of luteinizing hormone (LH) at the induced estrus to collect "control" follicles, which would have produced normal corpora lutea. In 15 ewes, the second ovary was removed 63 to 84 h later to collect "treated" follicles before (n = 7) or after (n = 8) the second expected surge of LH. Five ewes (control) were allowed to ovulate from the remaining ovary at first estrus and another five (treated) at the second estrus (3 to 4 d later). Treated ewes had lower serum progesterone than control ewes during the ensuing cycle (P less than .05). Treated follicles contained less estradiol in the theca (4.4 +/- .6 vs 10.0 +/- 2.5 ng; P less than .05), less androstenedione (.1 +/- .1 vs 1.0 +/- .2 ng) and estradiol (.5 +/- .1 vs 2.9 +/- 2.2 ng) in the granulosa (P less than .05) and less progesterone in the follicular fluid (.8 +/- .4 vs 3.3 +/- .8 ng; P less than .05) than control follicles, when removed before the surge of LH. Follicles removed after the surge of LH did not differ. In conclusion, ovulatory follicles with low steroidogenic function became corpora lutea that secreted lower-than-normal quantities of progesterone.  相似文献

The objective of this study was to determine whether administration of hCG approximately 5 d after AI would increase plasma progesterone concentrations and conception rates in beef heifers. Heifers from two locations (Location 1: n = 347, BW = 367 +/- 1.72 kg; Location 2: n = 246, BW = 408 +/- 2.35 kg) received melengestrol acetate (0.5 mg.heifer(-1).d(-1)) for 14 d and an injection of PGF2alpha (25 mg i.m.) 19 d later. Heifers were observed for estrus continuously during daylight from d 0 to 4.5 after PGF2alpha and artificially inseminated approximately 12 h after the onset of estrus. Half of the heifers inseminated at Location 1 were assigned randomly to receive an injection of hCG (3,333 IU i.m.) 8 d after PGF2alpha, and a blood sample was collected from all heifers 14 d after PGF2alpha for progesterone analysis. Half of the heifers inseminated at Location 2 were administered hCG on d 9 after PGF2alpha, and a blood sample was collected from all heifers 17 d after PGF2alpha. Heifers at Location 1 had a 94% synchronization rate, exhibited estrus 2.45 +/- 0.03 d after PGF2alpha, and received hCG 5.55 +/- 0.03 d after AI. Heifers at Location 2 had an 85% synchronization rate, exhibited estrus 2.69 +/- 0.03 d after PGF2alpha, and received hCG 6.31 +/- 0.03 d after AI. Progesterone concentrations were greater (P < 0.01) for hCG-treated heifers than for controls at both locations (8.6 vs. 4.6 ng/mL for treatment vs. control at Location 1, and 11.2 vs. 5.6 ng/mL for treatment vs. control at Location 2). Pregnancy status was determined by ultrasound approximately 50 d after AI. Conception rates (65 vs. 70% for treatment vs. control, respectively) did not differ at Location 1. Conception rates tended (P = 0.10) to be increased with hCG treatment at Location 2 (61 vs. 50% for treatment vs. control, respectively). A second experiment was conducted with 180 heifers at a third location to determine the effects of hCG administration 6 d after timed insemination at approximately 60 h after PGF2alpha in heifers synchronized as in Exp. 1. Pregnancy rate to timed AI did not differ between hCG-treated (62%) and control heifers (59%). Final pregnancy rate after timed AI and bull exposure (92%) was not affected by treatment. In summary, administration of hCG 5 to 6 d after AI did not improve conception or pregnancy rates at two out of three locations evaluated, suggesting insufficient progesterone is not a major factor contributing to early pregnancy failure in beef heifers.  相似文献

In beef heifers weaned between 3 and 4 mo of age and fed a high-concentrate diet, approximately 50% reach puberty before 300 d of age (precocious puberty). The objectives of this experiment were 1) to determine whether precocious puberty could be induced experimentally by weaning heifers early and feeding a high-concentrate diet, and 2) to determine the dynamics of secretion of LH associated with precocious puberty. Crossbred Angus and Simmental heifer calves were weaned at 73 +/- 3 d of age and 115 +/- 3 kg of BW and fed a high-concentrate (60% corn; HI, n = 9) or control diet (30% corn; CONT, n = 9). Heifers were fed individually, and target BW gains were 1.50 and 0.75 kg/d for the HI and CONT treatments, respectively. Heifers were weighed every 2 wk. Blood samples were collected weekly and assayed for progesterone concentration to determine age at puberty. Serial blood samples were collected at 20-min intervals for 24 h at mean ages of 102, 130, 158, 172, 190, 203, 217, 231, and 259 d and assayed for LH concentration to evaluate the dynamics of secretion of LH. Heifers fed the HI diet exhibited greater BW gain (P < 0.01) than CONT heifers (1.27 +/- 0.05 vs. 0.85 +/- 0.05 kg/d, respectively). As a result, BW in the HI treatment was greater (P < 0.01) than in the CONT treatment by 188 d of age and remained different through the end of the experiment. Precocious puberty occurred in 8 of 9 heifers fed the HI diet and 0 of 9 heifers fed the CONT diet. Age at puberty was reduced in the HI (P < 0.01) compared with the CONT heifers (262 +/- 10 vs. 368 +/- 10 d of age, respectively). Body weight at puberty was also reduced in the HI (P < 0.05) compared with the CONT treatment (327 +/- 17 vs. 403 +/- 23 kg, respectively). Heifers attaining puberty during the experiment continued with subsequent luteal phases as evidenced by cyclic patterns of progesterone concentrations. Frequency of pulses of LH (pulses/24 h) increased with age (P < 0.01) for both treatments. Heifers in the HI treatment exhibited a greater number of pulses of LH (P < 0.01) than those in the CONT treatment by 190 d of age and in all subsequent collection periods (treatment x age, P < 0.05). Mean LH concentrations also increased with age (P < 0.01) for both treatments but did not differ between treatments. In conclusion, precocious puberty induced by early weaning and feeding of a high-concentrate diet is preceded by increasing frequency of pulses of LH.  相似文献

The objective of this study was to characterize follicular dynamics in pre-pubertal, pubertal and post-pubertal periods, as well as the effect of high-energy intake on follicular development and age at puberty in heifers. Thirty-one Nelore (Bos indicus) heifers, 6 months old, were randomly assigned to receive two different diets: one of low (GI) and other of high dietary energy intake (GII). Animals were evaluated in relation to body weight gain by being weighed every 21 days. Heifers were evaluated every other day by real-time linear ultrasonography to characterize ovarian structures development from weaning to post-pubertal period. Blood samples were collected to determine plasmatic concentrations of progesterone by RIA method. The ovulation was determined when progesterone concentrations were >1 ng/mL in three consecutive samples, and by ultrasound images of corpus luteum; and oestrous behaviour in some animals. Age at puberty differed among heifers of GII (17.00 +/- 0.46 months) compared with heifers of GI (19.87 +/- 0.47 months; p < or = 0.05). Maximum size of the dominant follicles at pre-pubertal period was greater in GII heifers than in GI (10.52 +/- 0.33 and 9.76 +/- 0.15 mm, respectively; p < or = 0.05). As heifers approached first ovulation time, size of dominant follicle increased (11.75 +/- 0.37 mm for GI and 12.52 +/- 0.91 mm for GII; p < or = 0.05). Body weight at puberty was not different in both groups (302.33 +/- 27.31 kg for GI and 326.19 +/- 27.78 kg for GII heifers; p > 0.05). We conclude that animals receiving high dietary energy intake attained the puberty earlier and the development of follicles were different than in low dietary energy intake.  相似文献

To determine influences of insulin and body condition on follicular growth, prepuberal gilts (n = 16) treated with pregnant mare's serum gonadotropin (PMSG) were used in a 2 X 2 factorial experiment with main effects of insulin (0 or .4 IU/kg every 12 h beginning at 1800 on the day before PMSG) and backfat depth (moderate, 25 +/- .8; high, 32 +/- .7 mm; P less than .0001). Body weights were similar. Blood sampling was at 6-h intervals for analyses of LH, FSH, growth hormone (GH), glucagon, cortisol, insulin, insulin-like growth factor-I (IGF-I), plasma urea nitrogen (PUN), nonesterified fatty acids (NEFA), testosterone, estradiol-17 beta, and progesterone. Ovaries were removed 75 h after PMSG treatment, and visible small (less than or equal to 3 mm), medium (4 to 6 mm), large (greater than or equal to 7 mm), and macroscopically atretic follicles were counted. Administration of insulin increased IGF-I in fluid of medium follicles (108.8 vs 60.7 ng/ml; SEM = 13.3; P less than .05). Neither insulin nor fatness affected hCG binding by granulosa cells (12.5 +/- 1.6 ng/10(6) cells) or numbers of large (16.7 +/- 2.6) and medium (10.4 +/- 2.3) follicles. However, insulin increased the number of small follicles (58.9 vs 29.9; SEM = 9.7; P less than .05) and reduced the number of atretic follicles (3.8 vs 11.3; SEM = 1.1; P less than .05). The predominant effect of insulin on reducing number of atretic follicles was in the small size class (.6 vs 6.9; SEM = .6, P less than .01). Follicular fluid estradiol and progesterone were not affected by treatments; however, testosterone concentrations in large follicles were lower in gilts with higher backfat (32.5 vs 59.9 ng/ml; SEM = 4.0; P less than .05). Systemic LH, FSH, glucagon, cortisol, PUN, NEFA, estradiol, and testosterone were not affected by insulin or level of feeding. However, GH was lower in gilts that had higher backfat (overall average of 3.2 vs 2.8 ng/ml; SEM = .1; P less than .05). Insulin reduced atresia and altered intrafollicular IGF-I independently of body condition and without sustained effects on other hormones.  相似文献

Precocious puberty (<300 d of age) can be successfully induced in a majority of heifers with early weaning and continuous feeding of a high-concentrate diet. The objective of this experiment was to determine the relative effects of timing of feeding a high-concentrate diet on age at puberty in early-weaned heifers. Sixty crossbred Angus and Simmental heifer calves were weaned at 112 +/- 2 d of age and 155 +/- 3 kg of BW and were fed a receiving diet for 2 wk. Heifers were blocked by age and BW, and assigned randomly to receive a high-concentrate (60% corn; H) or control (30% corn; C) diet during phase 1 (mean age 126 to 196 d) and H or C during phase 2 (mean age 196 to 402 d), resulting in 4 treatments (HH, n = 15; HC, n = 15; CH, n = 15; and CC, n = 15). Blood samples were collected weekly beginning at a mean age of 175 d and assayed for progesterone concentration to determine age at puberty. After 56 d on the experimental diets, BW of heifers fed the H diet during phase 1 were greater (P < 0.05) than those of heifers fed the C diet (mean age of 182 d; treatment x mean age, P < 0.01). After 70 d on the new diets (mean age of 266 d), heifers fed the H diet during phase 2 reached heavier BW (P < 0.05) than heifers fed the C diet, when compared within phase 1 diet groups (HH > HC; CH > CC). Body weights in HC and CH treatments differed from a mean age of 169 through 238 d, after which BW did not differ between these treatments. The ADG over the entire experimental period was greatest for the HH treatment (1.2 +/- 0.04 kg/d; P < 0.05), followed by the HC and CH treatments (1.0 +/- 0.03 and 1.0 +/- 0.02 kg/d, respectively), which were not different, and the CC treatment gained the least (0.7 +/- 0.04 kg/d; P < 0.05). Precocious puberty occurred in 67, 47, 47, and 20% of heifers in the HH, HC, CH, and CC treatments, respectively (HH > CC; P < 0.05). Mean age at puberty for the HH and HC treatments (271 +/- 17 and 283 +/- 17 d of age, respectively) was earlier (P < 0.05) than for the CC treatment (331 +/- 11 d of age). Age at puberty in the CH treatment (304 +/- 13 d of age) was intermediate to and not different from the other treatments. Heifers fed the H diet during phase 1 attained puberty earlier (P < 0.05) than heifers fed the C diet during phase 1. In conclusion, increasing dietary energy intake in early-weaned heifers, through feeding a high-concentrate diet from 126 to 196 d of age, decreased age at puberty regardless of the diet fed after 196 d of age.  相似文献

The objective of this study was to determine if alterations in dietary intake of heifers can influence IGF-I concentrations in plasma and(or) follicular fluid (FFL), size of follicles, and steroid concentrations in FFL (as an indicator of steroidogenic capacity). Cyclic heifers [n = 23; mean +/- SE body weight (BW) = 373 +/- 7 kg] were individually fed for 10 weeks either: a) 1.8% of BW in dry matter (DM) per d (GAIN; n = 7), b) 1.1% of BW in DM per d (MAINT; n = 8) or c) 0.7% of BW in DM per d (LOSE; n = 8). After 10 wk of treatment, heifers were ovariectomized 36-40 hr after the second injection of prostaglandin F2 alpha analog (2 injections 11 d apart), and plasma and ovaries were collected. Heifers weighed 444 +/- 13,387 +/- 8 and 349 +/- 9 kg in the GAIN, MAINT and LOSE groups, respectively, at time of ovariectomy. Mean diameter of follicles greater than or equal to 10 mm was greater (P less than .05) for GAIN (15.6 mm) than for MAINT (11.0 mm) or LOSE (12.5 mm) heifers. Numbers of follicles and concentrations of IGF-I in plasma and FFL did not differ (P greater than .20) between LOSE, MAINT and GAIN heifers. Progesterone concentrations were greater (P less than .05) in small and medium follicles of GAIN than MAINT or LOSE heifers, but were unaffected by diet in large follicles. Twelve ruminally, duodenally, and ileally cannulated (average initial BW 313 +/- 20 kg) and 27 intact Hereford heifers (average initial BW 256 +/- 17 kg) were used in two experiments to evaluate dairy food wash water solids (WWS) as a protein source in medium-quality hay diets. Heifers received a basal diet of orchardgrass hay (7.4% CP) and were assigned to one of three supplement treatments: control (C; .9% CP), WWS (18.8% CP)-, and soybean meal (SBM; 19.1% CP)-based supplements (fed at 1.5 kg of DM/d). Supplements were formulated to have similar ME concentrations. Ruminal ammonia concentrations were greater (P less than .10) for WWS- and SBM-supplemented heifers than for C heifers at most sampling times. Moreover, WWS and SBM increased (P less than .10) total VFA (mM) and acetate (mol/100 mol) and lowered propionate (mol/100 mol) at several sampling times. Ruminal fluid volume (liters) was unchanged (P greater than .10) by treatment; however, fluid dilution and flow rate (liters/h) were less (P less than .10) in C heifers than in heifers fed SBM or WWS supplements. Wash water solids and SBM supplementation increased (P less than .10) OM, NDF, and ADF digestibilities compared with C heifers. Feeding WWS and SBM supplements increased BW at 84 d (P less than .10) compared with C-supplemented heifers. Forage intake at 54 and 84 d by heifers supplemented with SBM or WWS was greater (P less than .10) than by C heifers. Control-supplemented heifers had the least, WWS intermediate, and SBM the greatest ADG at 84 d (P less than .10; .14 vs .35 vs .48 kg/d, respectively). These data indicate that WWS may be used as a protein source without serious adverse effects in heifers consuming medium-quality hay for 84 d.  相似文献

The objectives of the current study were to evaluate the correlation between reproductive status and steady-state levels of Myxovirus resistance gene (MX2) mRNA in peripheral blood leukocytes (PBL) of dairy heifers and the reliability of using change in MX2 messenger RNA (mRNA) for identification of nonpregnant heifers 18 to 19 d after AI. Holstein heifers (n = 266), 13 +/- 1 mo of age, were assigned randomly to be inseminated (BRED; n = 214) or not (NONBRED; n = 52). Estrous cycles of all heifers were synchronized with an intravaginal insert containing progesterone for 7 d. At insert removal, heifers received an injection of PGF2alpha. Heifers in the BRED group were inseminated on detection of estrus or at a fixed time, 72 h after insert removal concomitant with a GnRH treatment. Heifers in the NONBRED group received an injection of GnRH 48 h after insert removal. Blood samples collected on d 0 (d of AI or estrus) and 18 were used to determine steady-state levels of MX2 mRNA. Samples collected on d 0, 7, 14, and 21 were analyzed for progesterone concentration. Pregnancy was determined retrospectively by progesterone concentration on d 21 and was diagnosed at 30 +/- 1 and 60 +/- 3 d after AI. The fold change in levels of MX2 mRNA from d 0 to 18 was greater for heifers classified and diagnosed as pregnant on d 21 (P < 0.05) and 30 +/- 1 (P < 0.05) and 60 +/- 3 (P < 0.05) d after AI compared with nonpregnant (bred but not pregnant) and NONBRED heifers. Heifers that experienced pregnancy loss from 21 to 30 +/- 1 (P = 0.11) or 21 to 60 +/- 3 (P = 0.08) d of gestation tended to have smaller fold increases in MX2 mRNA expression than those that maintained pregnancy. The sensitivity (range 57.1 to 65.6%) and negative predictive values (range 47.9 to 57.1%) of determining reproductive status on d 18 according to the change in the level of MX2 mRNA expression in PBL were low, and the correlation between diagnosis of pregnancy by fold change in MX2 mRNA expression and other methods was small (r = 0.20 to 0.36). The current study indicates that increased expression of MX2 mRNA in PBL is related to pregnancy approximately 21, 30, and 60 d after AI in dairy heifers and that losses that occurred later in pregnancy were associated with lower fold increases in MX2 mRNA. However, using the change in MX2 mRNA expression was not a reliable method for diagnosis of pregnancy at 18 d after AI because of the low sensitivity and negative predictive value.  相似文献

To determine the influence of three levels of undegradable intake protein (UIP) supplementation on metabolic and endocrine factors that influence reproduction, 23 yearling crossbred heifers (body condition score = 4.5 +/- 0.5; initial BW = 362 +/- 12 kg) were stratified by BW and assigned randomly to one of three supplements: 1) low UIP (1,135 g x heifer(-1) x d(-1); 30% CP, 115 g UIP, n = 7); 2) mid UIP (1,135 g x heifer(-1) x d(-1); 38% CP, 216 g UIP, n = 8); or 3) high UIP (1,135 g x heifer(-1) x d(-1); 46% CP, 321 g UIP, n = 8). Heifers were estrually synchronized before initiation of supplementation. Supplement was individually fed daily for 30 to 32 d, at which time heifers were slaughtered (d 12 to 14 of the estrous cycle) and tissues collected. Heifers were fed a basal diet of sudan grass hay (6.0% CP) ad libitum. On d 28 of supplementation (d 10 of the estrous cycle), no differences were observed (P > 0.10) in serum insulin or IGF-I among treatments. At slaughter (d 10 to 12 of the estrous cycle), treatments did not influence corpus luteum weight, cerebral spinal fluid leptin, or IGFBP; serum estradiol-17beta, progesterone, leptin, IGF-I, and IGFBP; or anterior pituitary content of IGFBP (P > 0.10). Follicular fluid IGFBP-2 and IGFBP-4 were greater in high-UIP heifers than low- or mid-UIP heifers on d 12 to 14 of the estrous cycle (P < 0.05). Basal serum LH concentrations and LH area under the curve (every 15 min for 240 min) did not differ (P > 0.10) following 28 d of supplementation (d 10 of the estrous cycle); however, basal serum FSH concentrations were greater (P = 0.06) in low- and mid- vs. high-UIP heifers (5.2 and 5.2 vs. 4.6 ng/mL, respectively), and FSH area under the curve was greater (P = 0.03) in low- vs. high-UIP heifers. At slaughter (d 12 to 14 of the estrous cycle), anterior pituitary LH and FSH content and steady-state mRNA encoding alpha, LHbeta, and GnRH receptor did not differ (P > 0.10) among treatments. However, FSHbeta mRNA was increased approximately twofold (P = 0.03) in mid vs. low UIP. In summary, low and mid levels of UIP supplements fed to estrous cycling beef heifers seemed to enhance pituitary expression and/or secretion of FSH relative to high levels of UIP. Moreover, high-UIP supplementation was associated with increased low-molecular-weight IGFBP compared with supplementation of low and mid levels of UIP. These data suggest that differing levels of UIP supplementation may alter pituitary and ovarian function, thereby influencing reproductive performance in beef heifers.  相似文献

Angus and Charolais heifers (195 +/- 7 kg) were actively immunized against growth hormone-releasing factor (GRF) to evaluate the effect on concentrations of somatotropin (ST), insulin-like growth factor I (IGF-I), insulin (INS), growth, and onset of puberty. Primary immunizations were given at 184 +/- 7 d of age (d 0 of experiment) by injecting (s.c.) 1.5 mg of GRF-(1-29)-Gly-Gly-Cys-NH2 conjugated to 1.5 mg of human serum albumin (GRFi, n = 22) or 1.5 mg of human serum albumin (HSAi, n = 21). Booster immunizations of .5 mg of antigen were given on d 62, 92, 153, and 251. Antibody binding (percentage at 1:2,000 dilution) to [125I]GRF on d 69 was greater (P less than .01) in GRFi (53.7 +/- 4.5) than in HSAi (10.1 +/- .6) heifers. Serum concentration (ng/ml) and frequency (peaks/5 h) of ST release, respectively, on d 78 were lower (P less than .01) in GRFi than in HSAi heifers (3.3 +/- .1 vs 5.6 +/- .2 and .9 +/- .3 vs 2.3 +/- .2). Serum IGF-I (ng/ml) was lower (P less than .01) in GRFi than in HSAi heifers on d 69 (41 +/- 5 vs 112 +/- 4). Serum INS (microU/ml) on d 78 was lower (P less than .05) in GRFi (2.2 +/- .1) than in HSAi (3.8 +/- .2) heifers. Feed intake, ADG, and feed efficiency were lower (P less than .05) in GRFi than in HSAi heifers. Hip height was lower (P less than .01) and fat thickness was greater (P less than .05) in GRFi than in HSAi heifers by d 132 and 167, respectively. Percentage of heifers attaining puberty (progesterone greater than 1 ng/ml for two consecutive weeks) by d 209 and 379 (12.9 and 18.5 mo of age), respectively, was lower (P less than .05) in GRFi (40.9 and 45.5) than in HSAi (81.0 and 100). In conclusion, growing heifers were successively immunized against GRF. Active immunization against GRF resulted in decreased serum concentration of ST, IGF-I, and INS. In addition, GRF immunization led to lowered feed intake, ADG, and feed efficiency, increased fat depth, and delayed onset of puberty in heifers. We propose that ST and IGF-I are important metabolic mediators involved in the initiation of puberty in heifers.  相似文献

Our objectives were to determine effects of grain processing on splanchnic (gut tissues and liver) N metabolism and whole-body N balance by growing steers and to ascertain the relative contributions of ruminal and intestinal tissues to net absorption and utilization of N-containing nutrients. Seven beef steers (348 kg initial BW), surgically implanted with appropriate catheters, were fed diets containing 77% steam-flaked (SF) or dry-rolled (DR) sorghum grain. Blood flows and net output or uptake of ammonia N, urea N, and alpha-amino N (estimate of amino acids) were measured across portal-drained viscera (PDV or gut tissues) and intestinal, ruminal, hepatic, and splanchnic tissues (PDV + hepatic). The experimental design was a crossover between DR and SF diets, with six samplings of blood at 2-h intervals on 2 d for each steer. Nitrogen intake (139 +/- 3 g/d), output in urine (43 +/- 2 g/d), and retention (40 +/- 3 g/d) were similar for both processing treatments. When steers were fed SF sorghum compared to DR sorghum, N retention as a percentage of N intake was numerically greater (P < 0.12), output of fecal N was numerically lower (P < 0.13), and urinary urea N was lower (P < 0.04). For SF vs DR, net uptake of alpha-amino N by liver was higher (P < 0.04; 20 vs 9 g/d) and was numerically lower (P < 0.16) for ruminal tissues (15 vs 33 g/d). Feeding steers SF compared to DR tended to increase net transfer (cycling) of blood urea N to PDV (57 vs 41 g/d; P < 0.07), increased cycling to intestinal tissues (15 vs 6 g/d; P < 0.05), and numerically increased transfer to ruminal tissues (42 vs 32 g/d; P < 0.12) but did not alter other net output or uptake of N across splanchnic tissues. Total urea N transfer (blood + saliva) was similar for both treatments. Net uptake of alpha-amino N by ruminal tissues was about 30% of the net amount of alpha-amino N absorbed across the intestinal tissues. In summary, most of the blood urea N cycled from the liver to gut tissues was transferred to ruminal tissues for potential microbial protein synthesis, and the net ruminal utilization of alpha-amino N was about 30% of that absorbed from intestinal tissues. Feeding growing steers SF compared to DR sorghum diets numerically increased whole-body N retention (percentage of N intake) by about 15% and tended to increase transfer of blood urea N to the gut by about 40%, which could increase the supply of high-quality microbial protein for absorption.  相似文献
