It has been difficult to establish reliable indices of exposure to endocrine disrupting chemicals (EDCs) appropriate for a variety of avian species because of a vast array of reproductive strategies. Data from mammals, reptiles and fish provide insight on likely mechanisms of action for EDCs. However, many of the effects of EDCs are weaker than the actions of the native hormones, making it difficult to assess adverse effects in domestic and wild birds. It is clear that differential sensitivity to EDCs exists across species, due to the timing and mode of exposure, compound toxicity and age of the individual. Our studies on EDCs are conducted in the quail model system, with focus on reproductive endocrine, neuroendocrine and behavioral responses. Studies have included EDC exposure, either by egg injection or via diet. Results from egg injection studies showed the following: (1) estradiol administered by embryonic day 12 demasculinized male sexual behavior, altered hypothalamic neurotransmitters and reduced hen day production and fertility in a dose dependent fashion, (2) methoxychlor (MXC) or vinclozolin impaired male sexual behavior in adult quail and (3) DDE exposure impaired reproductive and immune related end points. Two-generation studies were conducted on Japanese and northern bobwhite quail with dietary methoxychlor (MXC) exposure (0, 5 and 10 ppm) beginning in adults (P1), continuing in their offspring (F1), with F2 offspring raised on control diet. MXC exposure impaired male sexual behavior, hypothalamic catecholamines and plasma steroid hormones. Moreover, MXC exposure had reproductive consequences observable at both the lower and higher doses of MXC in F1 and F2 generations. These data demonstrate that embryonic EDC exposure interferes with sexual differentiation of neural systems that direct reproduction.  相似文献

The copulatory behavior and the parvocellular vasotocin (VT) system of the nucleus of the stria terminalis (BST) are sexually dimorphic in the Japanese quail. Embryonic administration of estradiol benzoate (EB) induces an organizational effect determining the disappearance of such a dimorphism (male shows behavior and cerebral phenotype of the female). The VT parvocellular system can therefore be considered an accurate marker of the sexual differentiation of brain circuits and a very sensitive indicator of the activity of estrogen-like substances on neural circuits. To test this hypothesis we administered diethylstilbestrol (DES), a powerful synthetic xenoestrogen, genistein (GEN), a phytoestrogen produced by soy, and bisphenol A (BPA). After 3 days of incubation, quail eggs were injected with vehicle, EB, DES, GEN or BPA. Administration of BPA caused an early blockage of development and no further analyses were done on the BPA groups. At puberty, the copulatory behavior of EB- or DES-treated male quail was totally abolished, whereas only the highest doses of GEN determined a significant decrease of the behavior. After the tests, the animals were sacrificed and perfused. The fractional area (FA) covered by VT immunoreactivity was analyzed in BST, medial preoptic nucleus, and lateral septum by computerized image analysis. The FA was significantly reduced after treatment with EB, DES and GEN at high doses. These results confirm that the sexually dimorphic VT system of the Japanese quail is a sensible indicator of the effects of xenoestrogens at the level of the central nervous system.  相似文献

In a previous study, we found that 3-methyl-4-nitrophenol (PNMC), a component of diesel exhaust particles and also a degradation product of the insecticide fenitrothion, exhibits reproductive toxicity in the adult male Japanese quail. The present study investigated the toxicity of PNMC in the female Japanese quail and its ability to influence reproduction in immature females. The quail (21-day-old) were injected intramuscularly (im) with PNMC at doses 0.1, 1 or 10 mg/kg body weight daily for 3 days. There was no significant difference in body growth between the PNMC-administered and control birds. However, the weights of the oviducts were significantly lower in the PNMC-treated birds at all doses. Furthermore, the plasma concentrations of luteinizing hormone (LH) and estradiol-17 beta were significantly decreased with 1 and 10 mg/kg of PNMC. These findings suggest that PNMC might influence the hypothalamo-pituitary-gonadal axis with decreasing in secretion of GnRH, LH and ovarian steroid hormones and subsequently disturb growth of the reproductive organs of immature female quail. This study indicates that PNMC induces reproductive toxicity at the central level and disrupts reproductive function in the immature female quail.  相似文献

为研究雌激素在鸡性腺和性行为分化过程中的作用,本试验在受精蛋孵化第3天气室注入100μL生理盐水或芳香化酶抑制剂(AI),出雏后常规饲养到8月龄性成熟,观察其性行为的表现,检测性腺结构和血清性激素水平.结果发现,AI处理获得的性反转母鸡出现雄性第二性征和雄性交配行为,性腺形态和组织学检查结果表明其性腺得到了不同程度的反转(1)完全反转母鸡右侧性腺发育成为睾丸,而左侧性腺出现不同程度的反转.两侧性腺均出现精细管结构,管径较正常窄,发育不完整,精细管内大量分布生殖细胞,右侧性腺的间质较左侧发达;(2)不完全反转母鸡的右侧性腺退化,左侧仍为完整卵巢.血清中雌二醇(E2)含量为不完全反转母鸡>正常母鸡>完全反转母鸡>正常公鸡;睾酮(T)含量为完全反转母鸡>不完全反转母鸡>正常公鸡>正常母鸡,雄性交配频率与血清T/E2的比值呈正相关.以上结果表明,在性分化前用AI阻断雌激素的合成可引起雌性性腺结构和激素分泌功能及性行为雄性化,而血清T和E2含量及其比值决定了性反转母鸡雄性交配行为的频率和强度.  相似文献

The blue‐breasted quail (Coturnix chinensis), the smallest species of quail with short generation interval and excellent reproductive performance, is a potential avian research model. A normal series of skeletal development of avian embryos could be served as a reference standard in the fields of developmental biology and teratological testing as well as in the investigation of mutation with skeletal abnormalities and in the study of the molecular mechanisms of skeletal development through genome manipulation. Furthermore, ossification sequence shows a species‐specific pattern and has potential utility in phylogeny. However, data on the skeletal development of blue‐breasted quail embryos are scarce. Here, we established a series of normal stages for the skeletal development of blue‐breasted quail embryos. Cartilage and ossified bones of blue‐breasted quail embryos were stained blue and red with Alcian blue 8GX and Alizarin red S, respectively. The time and order of chondrification and calcification of their skeletons were documented every 24 hr from 3 to 17 days of incubation, and a 15‐stage series of skeletal development was created. Moreover, a comparative study with the Japanese quail (Coturnix japonica) demonstrated that ossification sequence differed significantly between these two species.  相似文献

A sensitive bioassay for detection of dietary estrogens in animal feeds   总被引:3,自引:0,他引:3  
Estrogen-responsive proliferation in the MCF-7 cell line was used as a bioassay for detection of dietary estrogens. The bioassay procedure was adapted to screen for estrogenic activity in feedstuffs that have been associated with hyperestrogenism in livestock. Methanolic feed extracts were added to the cell culture medium at microliter/ml concentrations for 4 days, after which the cell proliferation response was measured as DNA content. The half-maximal response for estradiol occurred at 2 pM, or 0.54 pg/ml. For zearalenone, a weaker estrogen, the half-maximal response occurred at approximately 200 pM, or 64 pg/ml. The bioassay was calibrated against a number of known estrogens (estradiol, diethylstilbestrol, zearalenone, zearalanol [cattle implant], beta-zearalenol, zearalane), including the naturally occurring phytoestrogens (formononetin, genistein, daidzein, biochanin A, and coumestrol). The estrogenic activity of feed samples was expressed as equivalents of zearalenone (ppm zearalenone) that would have to be present to equally stimulate proliferation of the MCF-7 cells. The sensitivity of the bioassay was 0.05-0.1 ppm equivalents of zearalenone in feed, well below the threshold level associated with reproductive problems. The feed additive melengestrol acetate (MGA) showed no estrogenic activity in this assay. Estrogenic activity of feed extracts was confirmed by competitive inhibition with the antiestrogens tamoxifen or LY156758 (keoxifene) to show that stimulation of growth by feed extracts was through an estrogenic mechanism. Confirmation of known estrogens was by tandem mass spectroscopy. The assay is a sensitive and reliable screening procedure for detecting estrogenic activity in feedstuffs.  相似文献

性别分化是生物界普遍存在的一个自然现象。单性花由于仅含一种有功能的性器官,通过对其研究有助于解释植物性别分化和性别决定调控机制。植物性别决定方式多样且复杂,既有通过性别决定基因决定性别,又有性染色体,通过阻止性别决定基因重组确保稳定的性别分离;同时表观遗传由于影响基因表达活性,对性别分化也发挥重要作用。此外,植物激素、遗传因子、表观遗传修饰等之间存在相互作用,共同决定单性花性别。本文从单性花分类、雌雄花表型性状差异、遗传基础、表观遗传修饰、激素调控等方面综述单性花性别分化和决定调控机理,并提出未来研究中将面临的挑战和应对策略,为揭示植物性别分化和决定的机理提供有效参考。  相似文献

An ontogenetic study was designed on developing rats in uterus of mothers tryptophan deprived at day 1 (exp. 1) and day 14.5 (exp. 2) of conception to verify the supposed determining role of the serotoninergic system (SS) in sexual differentiation in mammals. Tryptophan-free feeding was pursued uninterruptedly in the litter after birth, during lactation and post-natal development. Tryptophan-free pregnant rats were obtained by exclusion of tryptophan sources from chow. In both exp. 1 and exp. 2, the litter showed at birth a significant physical under evolution that worsened, during post-natal development, to a much more marked dwarfism in exp. 1 pups. Growth hormone concentrations in both sexes of dwarf rats were lower than that in the control rats. At 30 days post-natal age, whereas exp. 1 female rats showed a right-timed onset of puberty, no descensus of testes could be observed in male rats of same experiment. Dwarf male rats showed an evident hypotrophy of the whole reproductive apparatus. In histological examination of testes, neither spermatogenesis nor Leydig cells have been observed. Moreover, dwarf female rats showed a pronounced hypotrophy of reproductive organs, but a normal puberal status pattern was evident. In exp. 2, litters showed a less pronounced dwarfism, but a normal right-timed onset of puberty in both male and female rats. Data indicate that role of tryptophan in physical and sexual maturation in both male and female rats is essential.  相似文献

采用RT-PCR方法,用Wpkci和β-actin引物进行多重PCR鉴定66~120 h鹌鹑胚胎样本性别后,选取不同时间点雌、雄胚胎各4枚,以β-actin为内标,测定bcl-2 mRNA的相对丰度;探讨bcl-2对鹌鹑早期胚胎发育的影响。结果表明:①雄性胚胎在66~90 h bcl-2 mRNA表达量一直维持在较低水平,96~102 h bcl-2 mRNA有所增加,但96与102 h间差异不显著(P>0.05),在108 h显著降低(P<0.05);与108 h相比,114 h显著升高,120 h维持在较高水平。雄性胚胎在66~108 h(72 h除外)bcl-2 mRNA表达均比雌性胚胎的表达量低,在114~120 h bcl-2 mRNA表达水平均比雌性胚胎的高。②雌性胚胎在66~90 h bcl-2 mRNA表达量一直维持在较低水平;与90 h相比,96 h显著升高,96~120 h逐渐降低,其中96~108 h下降,但三者间差异不显著(P>0.05);与108 h相比,114~120 h显著下降(P<0.05)。③相同时间点雌、雄胚胎间的比较结果表明,84 h雌性胚胎bcl-2 mRNA表达显著高于雄性胚胎(P<0.05),90、108 h雌性胚胎bcl-2 mRNA表达极显著高于雄性胚胎(P<0.01);而在120 h雄性胚胎bcl-2 mRNA表达极显著高于雌性胚胎(P<0.01)。鹌鹑胚胎发育早期,雌、雄样本bcl-2 mRNA表达存在一定的时序性,说明bcl-2对雌、雄鹌鹑早期分化及早期胚胎发育有重要影响。  相似文献

The goal of this study was to determine whether in the Japanese quail the male genital tract contains receptors for progesterone, androgen and estrogen (PR, AR and ER, respectively), which have significant roles in reproductive functions, and whether their localization changes during sexual maturation. The epididymis and ductus deferens (middle and ampulla regions) of immature (approximately 30-day-old) and mature male Japanese quail were collected and frozen sections of them were immunostained for PR, AR and ER. The immunoreaction products for AR and PR were found in the nuclei of epithelial cells in the efferent ductules, epididymal duct, and the middle and ampulla regions of the ductus deferens of mature and immature birds. In the mature birds, the epithelial cells of the efferent ductules, epididymal duct, and the middle and ampulla regions of the ductus deferens were positive for ER, although some of the cells in the ductus deferens were negative. The epithelial cells of the ductules in the epididymis stained positive for ER, but the immunoreactions were negligible in the ductus deferens of immature birds. These results suggest that the epididymis and ductus deferens in quail possesses PR, AR and ER receptors. Each receptor is expressed before sexual maturation, although enhancement of ER expression may occur during maturation.  相似文献

Up to the present natural and synthetic steroids have only rarely been considered a cause of disturbances of sexual behavior and anomalies of sex organs. The increasing environmental contamination with chemicals showing estrogenic effects underlines the importance of further investigations in that matter. Therefore, the concentrations of estrogens and progesterone in the faeces of cattle were examined over a 13 weeks period. The values obtained are presented as equivalents of the standard substances estrone and 4-pregnene-20 beta-ol-3-one. The samples were stored in a refrigerator or incubator at temperatures of 5 degrees C and 30 degrees C, respectively, and the investigations were carried out by using an enzyme-immunoassay. Estrogen and progesterone concentrations decreased more rapidly when incubation was performed at 30 degrees C as compared to storage at 5 degrees C, and estrogen values declined more slowly than those of progesterone. When keeping the samples at 5 degrees C the estrogen concentrations decreased to below the basal values only beyond the eleventh week of storage. In contrast, comparable concentrations were already obtained during the third week of the experiment if the samples were stored at 30 degrees C. Progesterone levels decreased to below the basal values during the fourth week of storage at 5 degrees C and during the second week at 30 degrees C.  相似文献

1. This study examines the time courses of male mating behaviours using quail that were categorised at 2 d of age as high performance (HP) or low performance (LP) individuals in a T-maze. 2. Individually caged males from each T-maze category were observed for 5 min after a female of the same category (HP or LP) was introduced into their home cages as a sexual partner at 4, 5, 6, 8, 10 and 12 weeks of age. The number of grabs, mounts and cloacal contacts performed by each male and his copulatory efficiency (number of cloacal contacts/number of grabs; CE) were determined at each time interval. 3. No differences were observed in the proportion of HP and LP males performing grabs at any of the ages evaluated. However, a greater proportion of HP males showed mounting and cloacal contact behaviour, coupled with a higher CE than did their LP counterparts at 5 and 6, 6 and 8, and 8 weeks of age, respectively. At those ages, a higher mean number of mounts and cloacal contacts were also observed. No significant sexual behaviour differences between HP and LP quail were observed at 10 and 12 weeks of age. 4. The results suggest that rapid negotiation of the T-maze in hatchlings is associated with accelerated puberty in male quail. The differential reproductive behaviour of HP males during the development of puberty does not remain extant in fully developed quail. 5. The finding of accelerated male developmental sexual activity responses in HP quail indicates that selection for T-maze HP behaviour may allow males to be successfully bred at an earlier age.  相似文献

For the purpose of investigation of working mechanisms in endocrine disruptors, we evaluated the dose-related effects of fetal and/or neonatal exposure to an estrogenic compound on the male reproductive organs in adult mice, particularly with respect to gene expression of steroidogenic acute regulatory protein (StAR). The pregnant ICR mice were given subcutaneous injections of 10 micro g/day/animal of diethylstilbestrol (DES) to subject the fetal mice to in utero exposure (IUE). Subsequently, the newborn male mice were subjected to neonatal exposure (NE) by treatment with vehicle or 0.1-10 micro g/day/animal of DES. Fertility rates of each group were as follows: control, 100%; IUE only, 60%; IUE+NE 0.1 micro g, 25%; IUE+NE 1 micro g, 0%; IUE+NE 10 micro g, 0%. In general histology, germ cell layers in the seminiferous tubules were thinned in the group of IUE+NE 10 micro g. Hypoplasia of the Leydig cells, in which the staining intensity of eosin was diminished, was also observed in the groups of IUE+NE 0.1-10 micro g. The androgen receptor (AR) and estrogen receptor alpha (ERalpha) immunoexpression in the Leydig cells of IUE+NE 1-10 micro g was slightly lower than that in the controls. Long-term dysfunction of the hypothalamo-pituitary-testicular axis, including sustained hypoproduction of gonadotropin and testosterone, and altered expressions of steroid hormone receptors and StAR genes were observed. The hypothalamo-pituitary control of gonadotropin secretion may be affected by the smaller doses of estrogenic agents than the reproductive organs. Furthermore, the fertility rate in the male mice exposed to this estrogenic agent was closely correlated with the testosterone levels, and even more so with the rate-limiting factor of steroidogenesis, StAR. This finding suggests that endocrine disruptors have an important pronounced effect on StAR gene expression.  相似文献

Effect of coccidiosis on reproductive maturation of male Japanese quail   总被引:1,自引:0,他引:1  
The effects of coccidiosis on reproductive development of male Japanese quail were examined. Male Japanese quail were exposed to high (5 x 10(5) sporulated oocysts/quail) or low (5 x 10(3) sporulated oocysts/quail) doses of Eimeria uzura at 16 or 30 days of age and sampled at 37 days. Quail given high doses of coccidia had reduced testes weight and lowered circulating concentrations of androgen compared with control males. Low doses of coccidia did not affect testes weight but did result in elevated plasma androgen levels. There were no differences in average testes weights by 51 days; however, plasma androgen was still reduced in most groups. To study the effects of coccidiosis on egg production, males exposed to high doses of coccidia at 16 (16H) or 30 (30H) days of age were mated with control females, and control males were mated with control or 16H females. The onset of laying was delayed for 5 days in the control male: 16H female group. During the first week of production, eggs from females bred to 30H males had lower fertility and hatchability than those bred to control or 16H males. By the third week of production, levels of fertility were similar. Apparently, exposure of quails to coccidiosis before sexual maturation might result in long-term effects on later reproductive capability.  相似文献

Xenotoca eiseni is a viviparous teleost belonging to the family Goodeidae. Here, we report histological observations of the reproductive organs in an adult male, an adult female, a pregnant female with intraovarian embryo and an extracted embryo of X. eiseni. High-resolution images of haematoxylin–eosin-stained sagittal sections revealed the detailed structure of gonads, gametes and reproductive components of the mother–embryo relationship. In the male, mature spermatozoa in the epididymis formed sperm packages. In the female, oogenesis proceeded asynchronously in the ovarian wall, and various stages of oocytes were observed in single ovary. In both sexes, genital openings were located between the anus and anal fin. Developing embryos were observed in an ovary of the pregnant female. Fine structures of components of the mother-to-embryo nutrient supply, ovarian septum and trophotaenia were observed in the pregnant ovary. An immature gonad prior to gamete formation was identified in the extracted embryo. With the aim of supporting the development and extension of studies on this viviparous teleost, we have shared our histological images as raw data in an open online archive, the 'NAGOYA repository ( http://hdl.handle.net/2237/00032456 )'. Our goal is a comprehensive understanding of the viviparous system in fish using both histological observation and molecular biology methods including genomics and proteomics.  相似文献

In rodents and swine, individual differences in a broad range of characteristics correlate with intrauterine position during fetal life. By identifying the intrauterine position of mice at cesarean delivery, we can predict reliably postnatal reproductive traits such as genital morphology, timing of puberty, length of estrous cycles, timing of reproductive senescence, sexual attractiveness, sexual behavior, aggressiveness, daily activity level, body weight and tissue enzyme activity in females; in males we can predict genital and brain morphology, sexual behavior, aggressiveness, daily activity level, body weight, and tissue enzyme activity. In mice, as in all mammals, male fetuses have greater concentrations of testosterone than do females. In addition, female mouse fetuses have greater circulating concentrations of estradiol than do male fetuses, a condition not found in all mammals. A mouse fetus positioned between males has greater concentrations of testosterone than does a fetus of the same sex positioned between females, and a fetus positioned between females has greater concentrations of estradiol than does a fetus of the same sex positioned between males. Gonadal steroids regulate differentiation of secondary sexual characteristics. Studies in which the effects of intrauterine position have been eliminated by exposing fetuses to steroid receptor blockers reveal the critical role of steroids in mediating this phenomenon. The intrauterine position phenomenon provides the only mammalian model for relating postnatal traits to concentrations of endogenous hormones to which individuals are exposed during fetal life. Results from studies using this naturally occurring experimental system in litter-bearing species have given insights concerning the consequences of individual differences in steroid concentrations during sexual differentiation that likely apply to all mammals. One specific hypothesis is that circulating estradiol may interact with testosterone in mediating some aspects of sexual differentiation in rodents and, thus, possibly in other mammals.  相似文献

Chickens and Japanese quail (Coturnix japonica) have traditionally been the primary avian models in developmental biology research. Recently, the blue‐breasted quail (Coturnix chinesis), the smallest species in the order Galliformes, has been proposed as an excellent candidate model in avian developmental studies owing to its precocious and prolific properties. However, data on the embryonic development of blue‐breasted quail are scarce. Here, we developed a normal developmental series for the blue‐breasted quail based on developmental features. The blue‐breasted quail embryos take 17 days to reach the hatching period at 37.7°C. We documented specific periods of incubation in which significant development occurred, and created a 39‐stage developmental series. The developmental series for the blue‐breasted quail was almost identical to that for chickens and Japanese quail in the earlier stages of development (stages 1–16). Our staging series is especially useful at later stages of development (stages 34–39) of blue‐breasted quail embryos as a major criterion of staging in this phase of development was the weight of embryos and the length of third toes.  相似文献
