OBJECTIVE: To compare the virulence of spores of 7 fungi by tracheal inoculation of goats following exposure of goats to an aerosol of sterilized feedyard dust. Animals-54 weanling Boer-Spanish goats. PROCEDURE: A prospective randomized controlled study was conducted. There were 7 fungal treatment groups, a tent control group, and a pen control group (n = 6 goats/group). Goats in the 7 treatment and tent control groups were exposed to autoclaved aerosolized feedyard dust for 4 hours in a specially constructed tent. Goats in the 7 treatment groups were then inoculated intratracheally with 30 mL of a fungal spore preparation, whereas tent control goats were intratracheally inoculated with 30 mL of physiologic saline (0.9% NaCI) solution. These treatments were repeated each week for 6 weeks. RESULTS: Severity of pathologic changes differed significantly among the 7 fungal treatment groups as determined on the basis of gross atelectatic and consolidated lung lesions and histologic lesions of the lungs. Descending order for severity of lesions was Mucor ramosissimus, Trichoderma viride, Chaetomium globosum, Stachybotrys chartarum, Aspergillus fumigatus, Penicillium chrysogenum, and Monotospora lanuginosa. Trichoderma viride spores were the most invasive and were isolated from the bronchial lymph nodes and thoracic fluid of all 6 goats administered this organism. Spores were observed-histologically in lung tissues harvested 72 hours after inoculation from all treatment groups. CONCLUSIONS AND CLINICAL RELEVANCE: 4 of 7 fungal spore types induced significantly larger lung lesions, compared with those induced by the other 3 spore types or those evident in control goats.  相似文献

Water quality in cattle feedyard playas in winter and summer   总被引:1,自引:0,他引:1  
OBJECTIVE: To determine the impact of feedyards on endotoxin concentration, fecal coliform count, and other water quality measurements during winter and summer in feedyard playas (shallow lakes). SAMPLE POPULATION: Water samples obtained from 7 feedyard playas and 3 nonfeedyard control playas. PROCEDURE: Surface water samples were collected from each playa and at various depths from 3 feedyard playas. Endotoxin concentrations, 22 water quality variables, and fecal coliform counts were determined in samples collected in summer and winter from various combinations of playas. RESULTS: Cattle numbers per feedyard ranged from 40,000 to 175,000 head/y. Mean endotoxin concentrations were significantly lower in control playas than in feedyard playas in winter and summer. Endotoxin concentration appeared to be homogenous at various water depths. Values for 20 of 22 water quality variables were higher in the feedyard playas than in control playas in winter and summer. In winter only, mean total fecal coliform concentration in feedyard playas was significantly greater than in control playas. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that feedyards have the potential to impact water quality in playas, and cattle should not be allowed access to them. Feedyard playa water should not be used under high pressure to settle dust in pens with cattle or to cool cattle, because aerosols containing pathogens and high concentrations of endotoxin are a health hazard for humans and cattle?  相似文献

OBJECTIVE: To compare Salmonella isolates cultured from feedyard and nonfeedyard (control) playas (ie, temporary shallow lakes) of the Southern High Plains. SAMPLE POPULATION: Water and muck (sediment) samples were obtained from 7 feedyard playas and 3 nonfeedyard playas in the winter and summer. PROCEDURE: Each water and muck sample was enriched with sulfur-brilliant-green broth and incubated in a shaker at 37 degrees C for 24 hours. A sample (100 mL) of the incubated bacterial-enriched broth was then mixed with 100 mL of fresh sulfur-brilliant-green enrichment broth and incubated in a shaker at 37 degrees C for 24 hours. After the second incubation, a swab sample was streaked on differential media. Suspect Salmonella isolates were further identified by use of biochemical tests, and Salmonella isolates were confirmed and serovar determinations made. RESULTS: Salmonella isolates were not recovered from the 3 control playas. Seven Salmonella enterica serovars were isolated from 5 of 7 feedyard playas in the summer, and 13 S. enterica serovars were isolated from 7 of 7 feedyard playas in the winter. In the summer, 296 isolates were cultured, and 47 were Salmonella organisms. In the winter, 288 isolates were cultured, and 171 were Salmonella organisms. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that feedyard playas are frequently contaminated with many Salmonella serovars. These pathogens should be considered whenever feedyard managers contemplate the use of water from these playas. Water from feedyard playas should not be used to cool cattle in the summer or for dust abatement.  相似文献

OBJECTIVE: To determine effects of repeated aerosol exposures to fly ash dust on respiratory tracts of tent-confined goats. ANIMALS: 12 weanling Boer-Spanish crossbred goats. PROCEDURE: Goats were randomly assigned to 2 groups: fly ash treatment group (principal goats, n = 6) or control group (control goats, 6). Aerosolized fly ash dust was provided during a 4-hour period for each of 6 applications given over 3 months and one 2-hour application prior to necropsy. Fly ash particle diameters ranged from 0.1 to 130 microm and averaged 17.8 microm, with 1.5% of fly ash particles in the 0.1- to 5-microm-diameter range. A mean +/- SD of 748 +/- 152 g/treatment was delivered inside a tent containing principal goats; control goats were placed inside a similar tent for 4-hour treatments without dust. Following treatment, rectal temperatures were taken at 0, 4, 6, 8, 24, and 72 hours; Hcts were recorded at 0, 24, and 72 hours. RESULTS: Rectal temperatures were significantly increased at 4, 6, and 8 hours and decreased at 72 hours, compared with 0 hours. Mean +/- SEM Hct values were significantly increased for principal goats (3747 +/- 0.39%), compared with control goats (36.17 +/- 0.42%). A significant increase in the mean area of gross atelectatic lung lesions (1,410 mm2) was found in principal goats (n = 6), compared with control goats (440 mm2; 5). CONCLUSIONS AND CLINICAL RELEVANCE: An increase in atelectatic lung lesions was observed in principal goats, compared with control goats; however, overall, fly ash dust effects were nontoxic.  相似文献

OBJECTIVE: To investigate the effects of sterile fine dust aerosol inhalation on antibody responses and lung tissue changes induced by Mucor ramosissimus or Trichoderma viride spores following intratracheal inoculation in goats. ANIMALS: 36 weanling Boer-Spanish goats. PROCEDURES: 6 goats were allocated to each of 2 M ramosissimus-inoculated groups, 2 T viride-inoculated groups, and 2 control (tent or pen) groups. One of each pair of sporetreated groups and the tent control group were exposed 7 times to sterilized fine feedyard dust (mean+/-SD particle diameter, <7.72+/-0.69 microm) for 4 hours in a specially constructed tent. Goats in the 4 fungal treatment groups were inoculated intratracheally 5 times with a fungal spore preparation (30 mL), whereas tent control goats were intratracheally inoculated with physiologic saline (0.9% NaCl) solution (30 mL). Pen control goats were not inoculated or exposed to dust. Goats received an IV challenge with equine RBCs to assess antibody responses to foreign antigens. Postmortem examinations were performed at study completion (day 68) to evaluate lung tissue lesions. RESULTS: 5 of 7 deaths occurred between days 18 and 45 and were attributed to fine dust exposures prior to fungal treatments. Fine dust inhalation induced similar lung lesions and precipitating antibodies among spore-treated goats. Following spore inoculations, dust-exposed goats had significantly more spores per gram of consolidated lung tissue than did their nonexposed counterparts. CONCLUSIONS AND CLINICAL RELEVANCE: Fine dust inhalation appeared to decrease the ability of goats to successfully clear fungal spores from the lungs following intratracheal inoculation.  相似文献

Dust is an environmental stressor and can become extensive in agricultural production systems. Thirty-six female, Spanish goats (average BW 21.1 kg, SEM = 1.31; age = 4 mo) were randomly assigned to simulated dust events or no dust, with or without tilmicosin phosphate treatment in a 2 x 2 factorial arrangement of treatments to determine effects on performance, rectal temperature, and leukocyte changes. All goats were fed a standard growing diet (13.6% CP) consisting of 37% roughage and 63% concentrate (DM basis). Feed intake was measured daily, and BW (unshrunk) measured individually every 7 d. The tilmicosin-treated group received tilmicosin phosphate (10 mg/kg BW s.c.) before starting the study. Goats exposed to dust were enclosed as a group inside a canvass tent for 4 h each day and ground feed yard manure dust (mean particle size 100 microm) was aerosolized inside the tent to simulate a dust event. There was one single dust event (Phase I) followed by rectal temperature measurement, and heparinized blood collection for complete cell counts at 0 (pretrial), 4, 12, 20, 44, 68, and 210 h after dust exposure. This was followed by 21 d of chronic dust events (Phase II). The sampling procedures for Phase II were exactly the same as in Phase I, except that samples were obtained daily at 0 (before dust application), 4, 8, and 12 h after each dust event. Dust treatment had no effect (P > 0.05) on feed intake or ADG, but the gain:feed (G:F) ratio was lower (P < 0.05) in the control goats than the dust exposed group. Tilmicosin phosphate-treated goats had a higher (P < 0.05) G:F ratio than untreated goats. Dust exposure increased (P < 0.002), but tilmicosin treatment decreased (P < 0.05) rectal temperature at 4 and 8 h. Dust exposure increased (P < 0.02) blood lymphocyte counts compared with controls. These results suggest that simulated dust events altered rectal temperature and leukocyte counts of goats.  相似文献

Eighteen ponies were inoculated IV with Ehrlichia risticii-infected P388D1 mouse monocyte cells. Twenty-four hours after onset of fever (rectal temperature > 38.8 C), 9 ponies were treated with oxytetracycline (6.6 mg/kg of body weight, IV, q 24 h) for 5 days. The remaining 9 ponies served as infected nontreated controls. Mean scores of the following variables were not significantly different between groups on the day treatment was begun: rectal temperature, diarrhea, borborygmal sounds, feed intake, mental attitude, and evidence of a hyperresonant area in the abdomen. All ponies were observed for progression of clinical signs typical of ehrlichial colitis. Within 12 hours of initiation of treatment, only 1 treated pony had a rectal temperature > 38.8 C and most rectal temperatures were < 38.3 C. In contrast, only 2 control ponies had rectal temperatures < 38.8 C (mean rectal temperature values were significantly, P = 0.01, different between groups). In the treatment group, 4 ponies had no signs of depression after the first day of treatment, and only 1 had signs of depression after the second day of treatment (mean scores between groups were significantly different, P = 0.01). Feed intake remained normal in 4 treated ponies and improved in 4 of the remaining 5 after treatment began. Most of the control ponies had a progressive decrease in their feed intake during the observation period (mean scores between groups were significantly, P = 0.01, different). Bovine leukemia virus (BLV) was transmitted by rectal inoculation of BLV-infective whole blood into cattle and sheep. Two cows and 2 sheep each were given 500 ml and 50 ml of blood, respectively, by rectal infusion. Two sheep which served as positive controls each were given 1 ml of the same blood, IV. All animals became seropositive to BLV by postinoculation week 5. Although relatively large volumes of blood were used for rectal inoculation, a base line for infectivity was established for the rectal route.  相似文献

Fifteen pony mares were assigned to 1 of 3 treatment groups after foaling: Group 1, 35 ml of sterile saline solution was infused into the uterine lumen within 24 hours after parturition (6 mares); group 2, 300 mg of Escherichia coli endotoxin was infused into the uterine lumen within 24 hours after parturition (6 mares); and group 3, 300 mg of E coli endotoxin was infused into the uterine lumen between 72 and 96 hours after parturition (3 mares). Rectal temperatures were taken at -1, -0.5, 0, 0.5, 1, 1.5, 2, 3, 4, and 5 hours after treatment. Venous blood samples were also taken at these times for routine WBC counts. Data were analyzed as a repeated measurement design with linear and quadratic orthogonal contrasts performed where significant time and interaction with time occurred. Pretreatment averages of total WBC and neutrophil counts were compared with their nadir posttreatment averages by a t test when treatment-by-time interaction was significant for the parameter. Rectal temperature (37.9 +/- 0.1 C) remained stable and did not vary among treatment groups after intrauterine infusions. In contrast, total WBC and neutrophil counts did vary among treatment groups across time. However, for treatment groups 1 and 3, neither blood total WBC count nor neutrophil count after intrauterine infusions was different from pretreatment observations. In group 2, total WBC count decreased (P less than 0.10) from a pretreatment average of 11.5 +/- 0.4 X 10(3) cells/mm3 to a nadir concentration of 10.0 +/- 0.6 X 10(3) cells/mm3 by 60 minutes after infusion of endotoxin into the uterus.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Fecal samples were collected from 200 feeder-calves on farms in Tennessee, after assembly at a Tennessee auction market, and after transport to a Texas feedyard. A final fecal sample was collected from each calf after 30 days of feedyard confinement. The fecal samples were cultured for the presence of Salmonella spp. Salmonella isolates were serotyped and antimicrobial drug-resistance patterns determined. The number of calves fecal culture-positive for Salmonella spp increased from 0 on the Tennessee farms and auction market to 3/200 (1.5%) at entry into the Texas feedyard, and 16/200 (8%) after 30 days of feedyard confinement. Salmonella serotypes isolated and the number of isolates of each serotype were S reading (8), S cerro (4), S newbrunswick (3), S anatum (2), and S typhimurium (copenhagen; 2). All Salmonella isolates were resistant to 5 or more of 13 antimicrobial drugs tested. Salmonella reading isolates were resistant to 10 or 11 of 13 antimicrobial drugs. The results indicated that the calves could have been infected with Salmonella spp prior to or during the course of the study, and that marketing stress as they moved from farm through feedyard may have induced fecal excretion of salmonellae. In addition, the pattern of antimicrobial drug resistance in the Salmonella isolates was broad.  相似文献

The objective of the present study was to examine the effects of parenteral administration of iron and copper on hematological parameters, weight gain, and health of neonatal dairy calves in the period when iron and copper deficiency could be existed. Twenty-four Holstein calves were used for the experiment and randomly assigned to four different treatments. Treatments consisted of (1) control (no injections of Fe and Cu), (2) test 1 (1000 mg Fe as fe-dextran was injected to each calf at day 2 of age), (3) test 2 (160 mg Cu as methionine-copper complex was injected to each calf at day 14 of age), and (4) test 3 (Fe and Cu were injected to each calf as mentioned previously). Blood samples were collected from all of the calves within 24-48 hours after birth and at 7, 14, 21 and 28 days of age for measuring hematological parameters and within 24-48 hours after birth and at 14, 21 and 28 days of age for the determination of iron, copper, TIBC concentrations, and AST activity. Anti-coagulated blood was analyzed shortly after collection for: number of red blood cell (RBC), hemoglobin (Hb), heamatocrit (HCT), total leukocyte count (WBC), Platelet (Plt), MCH, MCV, MCHC, and differential leukocyte counts. The amounts of iron, copper, TIBC, and AST were measured in serum. Group had significant effects on the amounts of HCT, RBC, hemoglobin, MCV, neutrophil, weekly weight gain, and daily gain during each week (p < 0.05). Sampling time had significant effects on the amounts of RBC, MCV, MCH, MCHC, WBC, neutrophil, lymphocyte, monocyte, platelet, fibrinogen, copper, TIBC, AST, weight, weekly gain and, daily gain during each week (p < 0.05). significant interactions between sampling time and group were seen for HCT, RBC, hemoglobin, MCV, platelet, total protein, fibrinogen, iron, and TIBC (p < 0.05). Improved RBC parameters and MCV were seen in calves of group 4 (test 3) in comparing with control group. Total and daily gains were also significantly improved in test groups in comparing with control group (p < 0.05). No significant difference was seen for the days of treatment between groups.  相似文献

1. The effects of exogenous corticosterone on plasma corticosterone concentrations, body weight, food intake, the percentage of birds that laid an egg each day and egg weight were measured in laying Japanese quail. Data were collected from birds in the week before corticosterone treatment began, during a 21-d treatment period, and in the 22 d after treatment ended.

2. Groups of quail were treated with corticosterone at three different concentrations in their drinking water. Corticosterone intakes were calculated for each bird and birds were then assigned to 4 intake groups. The groups were 0 (control group), 0·31–0·60, 0·61–0·90, 0·91–1·50 or >1·51 mg corticosterone/bird/d.

3. Plasma corticosterone concentrations in the 4 intake groups increased from around 1 ng/ml on the day before corticosterone treatment began to maximum mean concentrations of 13–18 ng/ml on day 14 of treatment. Mean corticosterone concentrations did not change between day 14 of treatment and the day after treatment ended, and had decreased in only one of 4 intake groups one week later.

4. Mean body weight in the highest intake group remained significantly lower than in controls 22 d after corticosterone treatment ended. Whilst there was no clear effect of corticosterone on food intake during treatment, mean food intake in the three highest corticosterone intake groups was significantly lower than in controls in the week after treatment ended. The percentage of birds that laid an egg each day and egg weight were both decreased by corticosterone, and the percentage of birds that laid an egg each day remained significantly lower in the highest corticosterone intake group compared with controls in the third week after treatment ended.

5. It is suggested that elevated plasma corticosterone concentrations in quail after treatment ended were maintained by a hyperactive hypothalamo–pituitary–adrenal axis for at least one week. Although none of the negative effects of corticosterone were evident in the group of quail with the lowest corticosterone intakes, the findings of the present study show that corticosterone treatment can affect birds for up to several weeks after corticosterone intake ends.  相似文献

应用国产赛福丁对3组绵羊(每组3只)分别以10、20、40 mg/kg剂量进行了毒性试验。结果,10、20 mg/kg剂量的绵羊,药后临床未出现任何毒性反应;40 mg/kg剂量的绵羊,均于药后40~60 min出现了典型的有机磷中毒症状。全血胆碱酯酶对赛福丁非常敏感,但其抑制程度与中毒靠状的出现并无直接关系。中毒后血清GOT和GPT无变化。以50、100、200、250 ppm浓度的赛福丁溶液分别药浴患痒螨绵羊,均可迅速地治愈患羊,浴后24 h即可杀灭全部螨虫,24~72 h痒觉消失,2周后脱痂,长毛。近期疗效同于倍特和林丹.而远期疗效优于倍特和林丹。证明国产赛福丁是一种高效、低毒的杀螨剂。  相似文献

Classical hemolytic complement (C) of calves was analyzed during a protocol designed to imitate the usual market handling of feeder calves from the southeastern United States. Serum C concentrations of the calves (n = 100 x 4 years) were evaluated on their farm of origin, on arrival at an auction market, on arrival at a feedyard, and during their first 4 weeks in the feedyard. Complement concentrations (measured in CH50 units) were typically lowest at the farm of origin and highest when the calves entered the auction market 28 to 133 days later. Serum C concentrations decreased after the calves encountered the severe stresses of being in the auction market for 7 days, 24-hour truck transport (1,932 km) to the feedyard, and the first 7 days in the feedyard. The C concentrations recovered after 21 to 28 days in the feedyard. Steers had significantly (P less than or equal to 0.05) lower C concentrations than did heifers in 3 of 4 years at the farm of origin, and in 2 of 4 years at the auction market. Morbid calves had significantly (P less than or equal to 0.05) lower C values than did healthy calves on day 7 in the feedyard in 3 of 4 years. There were significant differences in C concentrations of calves from different farms of origin in each of the 4 years. There was no significant difference in C concentrations of calves that were vaccinated vs those not vaccinated with Pasteurella haemolytica.  相似文献

OBJECTIVE: To determine the prevalence of postanesthetic hyperthermia [rectal temperature >40 degrees C (104 degrees F)] in a clinical population of cats. STUDY DESIGN: Retrospective study. ANIMAL POPULATION: One hundred and twenty-five cats with an age range of 2 months to 16.1 years, and weighing 3.9 +/- 1.5 kg. MATERIALS AND METHODS: Data were obtained from the medical records of 125 cats that underwent general anesthesia. Information on perioperative rectal temperatures, breed, sex, weight, surgical procedure, anesthetic time, surgery time, anesthetic and analgesic drugs were retrieved. STATISTICAL ANALYSES: Five groups of cats were compared; group 1 (n = 15) received acepromazine and no opioids; group 2 (n = 17) received acepromazine and buprenorphine; group 3 (n = 19) received acepromazine, buprenorphine and ketoprofen; group 4 (n = 45) received acepromazine and hydromorphone and group 5 (n = 29) received acepromazine, hydromorphone and ketoprofen. Data conformed to a split-plot repeated measures analysis of variance and was analyzed using SAS PROC MIXED. Post hoc tests were by means of Bonferroni t-test; < or = 0.05 was considered significant. RESULTS: Rectal temperature was significantly decreased in all groups at the end of anesthesia. Rectal temperature was significantly elevated at 1, 1.5, 2, 3, 4 and 5 hours after the end of anesthesia in group 4, and at 2, 3 and 4 hours in group 5. Sixty-four percent of cats in group 4 and 69% in group 5 had rectal temperatures >40 degrees C (104 degrees F) at one or more times in the postanesthetic period. The highest temperature recorded was 42.5 degrees C (108.5 degrees F) in one cat in group 4. Mean rectal temperature did not exceed the preoperative temperature at any time during the postanesthetic period in group 1, 2 and 3 animals. CONCLUSIONS: This study indicates an association between hyperthermia and perioperative administration of hydromorphone in cats. CLINICAL RELEVANCE: When hydromorphone is used in cats their body temperature should be closely monitored.  相似文献

OBJECTIVE: To study effects of central- and peripheral-acting alpha2-adrenergic receptor agonists on lung parenchyma, platelets, and pulmonary intravascular macrophages (PIM) of sheep. ANIMALS: 12 healthy mature female sheep. PROCEDURE: Group-1 (control, n = 2) sheep received 5 ml of physiologic saline solution IV and were euthanatized 3 minutes later. Sheep of group 2 (n = 8) received xylazine (150 microg/kg of body weight, IV), then 2 sheep each were euthanatized 3, 10, or 60 minutes, or 12 hours later. Sheep (n = 2) of group 3 were given ST-91 (30 microg/kg, IV), then were euthanatized 3 minutes later. Immediately after euthanasia, the lungs were fixed intratracheally and tissue was obtained for light and electron microscopy after 1 hour. RESULTS: Pulmonary parenchymal damage or morphologic alterations in PIM and platelets were not evident in control sheep. Three minutes after xylazine administration, morphologic changes in PIM were appreciable. After 10 minutes, extensive damage to the capillary endothelium and alveolar type-I cells, intra-alveolar hemorrhage, and interstitial and alveolar edema were evident. Most PIM had complete internalization of the surface coat. Similar changes were seen 60 minutes after xylazine administration; however, by 12 hours, morphologic features of PIM and lung parenchyma were almost completely restored. Evidence of PIM activation, obvious damage to capillary endothelium, and extensive pulmonary edema also were evident 3 minutes after ST-91 administration. CONCLUSIONS: XYLAZINE induces severe pulmonary parenchymal damage when administered at clinical sedative doses in sheep; morphologic changes in PIM within 3 minutes after administration of these drugs are substantial; and platelet aggregation is not apparent.  相似文献

OBJECTIVE: To assess the rate and extent of ruminal degradation of warfarin, chlorophacinone, and bromadiolone in vitro and determine the oral availability and clinical and hemostatic effects of each anticoagulant rodenticide in adult sheep. ANIMALS: 3 Texel sheep. PROCEDURE: Samples of ruminal fluid were incubated with each of the anticoagulants to assess the kinetics of ruminal degradation over 24 hours. To determine the plasma kinetics of the anticoagulants, each sheep received each of the anticoagulants IV or via a rumenimplanted cannula at 2-month intervals (3 rodenticide exposures/sheep). At intervals during a 240- to 360- hour period after treatment, prothrombin time (PT) was measured, plasma anticoagulant concentration was assessed, and clinical signs of rodenticide poisoning were monitored. In plasma and rumen extracts, anticoagulant concentrations were determined via high-performance liquid chromatography. RESULTS: In the rumen extracts, anticoagulants were slightly degraded (< 15%) over 24 hours. In vivo, oral availability of warfarin, chlorophacinone, and bromadiolone was estimated at 79%, 92%, and 88%, respectively. Although maximum PT was 80 seconds after chlorophacinone and bromadiolone treatments, no clinical signs of toxicosis were detected; PT returned to baseline values within 2 weeks. CONCLUSIONS AND CLINICAL RELEVANCE: In sheep, warfarin, chlorophacinone, and bromadiolone were not degraded in the rumen but their bioavailabilities were high after oral administration; the kinetics of these compounds in sheep and other mammals are quite similar. These data suggest that the lack of susceptibility of ruminants to these anticoagulant rodenticides cannot be explained by either ruminal degradation or the specific toxicokinetics of these anticoagulants.  相似文献

Three Merino ewes were given cefotaxime IM, and 3 were given cefotaxime subcutaneously (50 mg/kg of body weight each); each dose was suspended in 6 ml of oil. Five dogs were also given an oily suspension of cefotaxime subcutaneously (SC) (50 mg/kg of body weight). The plasma concentrations (Cp) and pharmacokinetic data obtained after cefotaxime in the oily suspension was injected IM and SC were compared with data from the same animals after they were given an aqueous solution of cefotaxime by the same routes. Key pharmacokinetic values obtained after cefotaxime was administered IV to sheep and to dogs are discussed. Mean peak Cp (Cpeak) in sheep when given the oily suspension IM was approximately 53 micrograms/ml at 0.18 to 0.40 hour, and that value in sheep given the aqueous preparation was 62 micrograms/ml 0.08 to 0.18 hour. Mean Cpeak values after the oily suspension and the aqueous preparation were injected SC were 11.0 micrograms/ml (between 0.8 and 1 hour) and 51 micrograms/ml (between 0.25 and 1 hour), respectively. Bioavailabilities were approximately 70% after IM injection was done and 90% after SC injection was done. The beta-plasma half-lives were 0.7 hour after IM injection was done and 2.9 hours after SC injection was done. Mean Cpeak in dogs when given the oily suspension SC was 30 micrograms/ml at 1.0 hour, and when dogs were given the aqueous preparation SC, Cpeak was 27 micrograms/ml at 0.6 hour. OBJECTIVE: To investigate the effect of repeated daily footbathing in zinc sulphate on virulent ovine footrot associated with S1, U1 and U5 zymogram types of Dichelobacter nodosus, including the highly virulent S1 strain A198. DESIGN: A field trial with experimentally infected sheep. PROCEDURE: At week 0, 50 sheep were infected with D. nodosus strains A198 (S1), C305 (U1), BC3993 (U5) and BC3995 (U5). At weeks 1 and 47, respectively, 169 and 235 uninfected sheep were added. At week 60, sheep were allocated to control and treatment groups each containing 220 sheep. Every 2 or 4 weeks to week 113, feet were inspected, and lesions were scored and sampled. Treatment sheep were footbathed in 15 to 18% (w/v) zinc sulphate with surfactant for 5 consecutive days (10 min per day) during week 61. During week 110, the footbathing protocol was applied again, this time to all surviving treatment and control sheep. RESULTS: BC3993 and A198 were isolated from 57% (162 of 285) and 20% (58 of 285), respectively, of new lesions sampled between weeks 3 and 13, and 21% (57 of 271) and 50% (136 of 271) between weeks 49 and 59. Percentages of new lesions associated with C305 and BC3995 remained constant. During the initial 17 weeks after footbathing at week 61, 90% (75 of 83) and 19% (95 of 490) of lesions in treatment and control sheep, respectively, were score 4 or 5, and 94% (47 of 50) and 38% (33 of 87) of those were associated with A198. CONCLUSIONS: Repeated daily footbathing did not eradicate virulent ovine footrot because strain A198 produced deep, covert lesions that facilitated the survival of D. nodosus.  相似文献

OBJECTIVES: To determine the bacterial, fungal, and endotoxin concentrations in aerosolized ambient air during the winter and summer in feedyards located in the Southern High Plains, identify aerosolized microbial pathogens, and determine the size of microbial and dust components. SAMPLE POPULATION: Aerosol samples were obtained from 7 feedyards. PROCEDURE: Aerosol samples were collected upwind, on-site, and downwind from each feedyard at a point 1 m above the ground by use of biological 2- and 6-stage cascade impactors. RESULTS: Significantly more microbes were cultured from on-site and downwind samples than upwind samples. There were significantly more microbes during the summer than during the winter. However, mean endotoxin concentration was significantly higher during the winter (8.37 ng/m3) than the summer (2.63 ng/m3). Among 7 feedyards, mean +/- SE number of mesophilic bacteria (1,441 +/- 195 colony-forming units [CFUs]/m3) was significantly higher than mean number of anaerobic bacteria (751 +/- 133 CFUs/m3) or thermophilic bacteria (54 +/- 10 CFUs/m3) in feedyard air. Feedyard aerosol samples contained more mesophilic fungi (78 +/- 7 CFUs/m3) than thermophilic fungi (2 +/- 0.2 CFUs/m3). Eighteen genera of bacteria were identified by use of an automated identification system. CONCLUSIONS AND CLINICAL RELEVANCE: It appeared that gram-negative enteric pathogens offered little risk to remote calves or humans via ambient aerosols and that gram-positive pathogens of the Bacillus, Corynebacterium, and Staphylococcus spp can be spread by aerosols in and around feedyards. It was common to detect concentrations of endotoxin in the ambient air of 7 feedyards.  相似文献
