The efficacy of toltrazuril (Bay Vi 9142) in preventing ovine coccidiosis due to an infection acquired immediately after turnout on pasture was evaluated by comparing the faecal consistency, weight gain, and oocyst output of treated and untreated lambs in 3 trials. The lambs were either given a single treatment with toltrazuril at 15 or 20 mg/kg, or they were given a dose of 10 mg/kg on 2 separate days. A single treatment with toltrazuril at 20 mg/kg on day 10 after turnout on pasture almost completely prevented coccidiosis in 2 trials. In a third trial the acute phase of a severe Nematodirus battus infection coincided with the outbreak of coccidiosis, and thus partly masked the clinical effect of the anticoccidial treatment. In lambs treated with toltrazuril at 15 mg/kg on day 10 after turnout, the coccidial infection caused a softening of the faeces, but the lambs were not severely affected by the coccidia. In lambs given a dose of 10 mg/kg of toltrazuril twice, either on days 10 and 11 after turnout, or on days 10 and 20, the coccidial infection caused a softening af the faeces, including some cases of diarrhoea. Oocyst production due to the initial coccidial infection on pasture was markedly reduced by all treatments with toltrazuril. The reduction was most pronounced after a dose of 20 mg/kg. Lambs treated with single doses of 15 or 20 mg/kg of toltrazuril had a better weight gain than the untreated controls in 2 of the trials. Lambs treated with toltrazuril on day 10 after turnout were partially resistant to the coccidial reinfection acquired immediately after treatment, and they had a similar level of immunity as the untreated controls to the subsequent reinfection on pasture.  相似文献

Twin lambs at pasture with their ewes, were divided into seven groups of 10 lambs. One group of 10 lambs served as a non-infected, untreated control. Five groups of 10 lambs were infected with 10,000 oocysts of Eimeria crandallis and 10,000 oocysts of Eimeria ovinoidalis when they were 3 weeks old (day 21 of the study). This produced a good level of infection with high oocysts production and diarrhoea in the lambs. Fourteen days after the primary, artificial challenge (day 35) four of these groups were treated with oral diclazuril at 0.25, 1.0, 2.0 or 4.0mg/kg. Diclazuril treatment was highly effective, dramatically reducing symptoms of diarrhoea and reducing faecal oocyst output by 79.7%, 97.3%, 99.4% and 99.5% respectively in the treated groups within four days. Two weeks post-treatment, and 28 days after the primary coccidial challenge (day 49 of the study), five groups of lambs were re-challenged with 100,000 oocysts of E. crandallis and 100,000 oocysts of E. ovinoidalis (secondary challenge). A group of lambs which had received neither the primary coccidia infection, nor drug treatment (susceptible controls) were also given the secondary challenge. All lambs given the secondary challenge produced high numbers of coccidia and exhibited varying degrees of diarrhoeic faeces. The lambs, which had previously received the higher doses of diclazuril at 2.0 and 4.0mg/kg, developed clinical signs of coccidiosis. These lambs were completely susceptible despite having received the early primary immunising infection of coccidia on day 21. The effects of the secondary challenge were more severe in the groups dosed with the two highest levels of diclazuril than in the susceptible control lambs, which had presumably been exposed to continued low levels of pasture contamination and had acquired a limited degree of immunity from this exposure. It would appear that treatment at the higher dose levels not only eliminated most of the oocysts from the primary challenge but also adventitious infection derived from the grazing paddocks. In contrast, lambs which had received the two lower drug levels of diclazuril (0.25 and 1.0mg/kg) whilst producing large numbers of oocysts, had only transient diarrhoea following secondary challenge. It was concluded that when used as a metaphylactic treatment, diclazuril works rapidly and is effective within four days of administration. Overall, a single dose of diclazuril at either 0.25-1.0mg/kg appears to be highly effective in the control of coccidiosis in young lambs at pasture whilst allowing the development of protective immunity against subsequent heavy coccidia challenge.  相似文献

The prophylactic efficacy of a single oral dose of toltrazuril against coccidiosis (mixed Eimeria infections) in naturally infected lambs was evaluated in two experiments. Toltrazuril at 20 mg kg-1, given on Day 7 or Day 10 after turnout on pasture, proved to be highly efficacious in preventing clinical coccidiosis under Norwegian conditions. Toltrazuril reduced the oocyst output to very low levels, prevented the development of diarrhoea and improved weight gain during the first 4-5 weeks after treatment. Treatment on Day 7 was superior to treatment on Day 10 with respect to improving weight gain and preventing the development of soft faeces. Lambs treated with toltrazuril on Day 7 seemed to be as immune as untreated lambs to natural reinfections with coccidia later in the grazing season. In one of the experiments, natural infections with the nematode Nematodirus battus produced a coccidiosis-like disease in some lambs simultaneously with the outbreak of coccidiosis.  相似文献

Seven groups of twin lambs, kept with their dams on pasture, were given single oral inoculations of 10,000 oocysts of Eimeria ovinoidalis and 10,000 E crandallis at one, two, four, seven, 14, 21 or 28 days after birth, respectively. All were then challenged with 100,000 of each species at 42 days of age. An eighth group was challenged without having received the earlier 'immunising' inoculum, and a ninth group was not inoculated at all. Bodyweights, faecal consistency, oocyst output, and serum coccidial antibody levels were monitored up to 12 weeks of age. No clinical response was detected to inoculation up to four days of age. Loosening of faeces and a slight setback in weight-gain occurred in lambs inoculated seven, 14 and 21 days after birth; inoculation 28 days after birth caused severe diarrhoea and weight loss. Challenge at 42 days caused severe coccidiosis with 50 per cent mortality in the 'unimmunised' group. In those that had received 'immunising' inoculations, the challenge at 42 days caused some diarrhoea and some weight loss, but much less than in the 'unimmunised' lambs. The later the 'immunisation', the less severe was the disease attributable to the challenge at 42 days. Serum antibody levels correlated fairly closely with resistance to the disease. It was concluded that very young lambs were resistant to the pathogenic effects of some coccidia, but were able to respond to them immunologically.  相似文献

Lambs born in pens with slatted floor were brought out at 2–5 weeks of age on pastures heavily grazed by sheep the previous years. About 16 days later the oocyst output of the lambs rapidly increased to high levels. Lambs on pastures which never had been grazed by sheep earlier, had moderate oocyst counts.Between 11 and 25 days after the beginning of grazing there were significantly more lambs with diarrhoea on permanent pastures compared with pastures never grazed by sheep earlier.It was found that lambs were heavily infected during the first 2 days on permanent pastures.Thirteen housed lambs were given 10–50 g of soil from a permanent pasture as a water suspension by a stomach tube. Fifteen days later there was a steep rise in the oocyst output in most of them, and 11 of the 13 lambs developed diarrhoea and 2 died. None of 10 lambs given uninfected soil and none of 12 untreated controls showed diarrhoea and the oocyst output remained on a moderate level.It is concluded that oocysts which have survived the winter in the pasture are the main source of infection with Eimeria spp. in lambs with this kind of management. Soil-eating is the most likely source of infection during the first days on pasture.  相似文献

Kinetic differences between systemic vs. intestinal and humoral vs. cellular immune responses were elucidated in chickens experimentally infected with Eimeria maxima by comparing interferon-gamma (IFN-gamma) and parasite-specific antibody levels in the intestine and serum during the course of infection. The level of serum IFN-gamma correlated significantly with fecal oocyst shedding (r2 = 0.97), thereby establishing the importance of cell-mediated immunity in coccidia infection. Moreover, intestinal IFN-gamma levels increased sooner than those in sera (4 vs. 6 days postinfection) and both were observed prior to the appearance of parasite-specific antibodies (8-10 days postinfection), again indicating the importance of intestinal cellular immunity in coccidiosis. Although immunoglobulin (Ig)G, IgA, and IgM isotypes of the antigen-specific antibody response increased significantly in both the intestine and serum after E. maxima infection, intestinal IgA-specific antibodies showed the most dramatic increase. However, the relevance of this observation in the context of primary Eimeria infection is unclear because the coccidia parasites have reached the final stages of their life cycle by this time. These results thus demonstrate the importance of T-cell immune responses against coccidia, characterized by local IFN-gamma secretion in the intestine, in mediating host protective immune response to coccidia.  相似文献

The roles of helminths and coccidia in post-weaning diarrhoea in beef calves in the dry tropics were investigated. Diarrhoea occurred approximately one month after weaning in both anthelmintic treated and untreated calves. The highest numbers of coccidial oocysts were seen 29 days after weaning and 3 necropsies confirmed Eimeria zuernii coccidiosis. In the absence of wet overcrowded conditions, disease may have been precipitated by environmental interactions leading to suppression of host immunity. Other Eimeria identified were E. bukidnonensis, E. wyomingensis, E. bovis, E. auburnensis, E. cylindrica, E. ellipsoidalis and E. subspherica. The last 5 species are believed not to have been previously documented in Australia. The presence of E. canadensis was strongly suspected.  相似文献

A 42-day broiler floor pen study was conducted comparing the anticoccidial efficacy of toltrazuril (Baycox) as a stand alone treatment and as an additional treatment to in-feed anticoccidial programs. Toltrazuril was administered on days 18 and 19 in the drinking water at 7 mg/kg of body weight. The treatments were 125 ppm nicarbazin (days 0-14) to 66 ppm salinomycin (SAL) (days 15-35) with and without toltrazuril, SAL (days 0-35) with and without toltrazuril, nonmedicated (NM) to SAL with toltrazuril, and NM with and without toltrazuril. The controls were NM noninfected and infected. The treatments were replicated in five blocks of eight pens each in a randomized complete block design. All withdrawal feed was nonmedicated. On day 14, birds, except noninfected, were exposed to coccidial oocysts (Eimeria acervulina, Eimeria maxima, and Eimeria tenella) seeded litter. On days 21, 28, 35, and 42, birds and feed were weighed, four birds per pen were coccidial lesion scored, and litter oocyst counts were performed. The coccidial infection in the NM infected treatment caused a significant (P < 0.05) coccidiosis infection. Coccidiosis was moderately controlled in the anticoccidial treatment birds without toltrazuril. Performance in the NM with toltrazuril was equal to or better (P < 0.05) than the anticoccidial programs without toltrazuril. Toltrazuril was equal to the noninfected birds in performance. Toltrazuril most completely eliminated all coccidial lesions and dramatically reduced oocyst shedding. The performance data, lesion scores, and oocyst counts showed that a 2-day treatment with toltrazuril successfully controlled the coccidiosis with no relapse of infection. Toltrazuril can thus be used for supplemental control with in-feed anticoccidials or as a primary anticoccidial with nonmedicated feed.  相似文献

The development of a naturally acquired coccidia infection in lambs in Otago is described using data derived from faecal oocyst counts and specific differentiation of the oocysts. Oocysts first appeared in faeces of lambs 19–37 days after birth. Oocyst production reached a mean peak of 250 000 oocysts per gram when lambs were 40–50 days old and slowly declined with increasing age of lambs. Coccidial infections did not appear to have any obvious effect on weight gain. The numerically most numerous species of coccidia were Eimeria ovina, E. crandallis, E. ninakohlyakimovae, E. parva and E. weybridgensis.  相似文献

利用从南宁市郊养鸡场球虫病鸡粪便中收集的球虫混合种卵囊感染小鸡,再应用单卵囊分离感染技术,从感染鸡盲肠中收集的卵囊分离纯化获得1株纯种球虫,经鸡体传代增殖,对该虫株的卵囊大小和卵形指数、潜在期、排卵高峰期、最短孢子化时间、寄生部位、致病性等指标进行观察和测定。结果测得该虫株卵囊的平均大小为(25.743±1.94126)μm×(21.4±1.85985)μm,平均卵型指数为1.2067±0.07;潜在期为140 h;其排卵囊峰期在第6~9天,最高峰在第7天;最短孢子化时间为19 h;寄生部位在盲肠;对两周龄的艾维茵鸡,当使用5×104的孢子化卵囊感染剂量时死亡率为7.5%。根据这些测定和观察到的指标综合鉴定该分离株球虫为柔嫩艾美耳球虫,并命名为柔嫩艾美耳球虫广西南宁株(Eimeria.tenella-GXNN),该研究结果为进一步研究本地区鸡球虫病的药物治疗和免疫预防等奠定了基础。  相似文献

Infections with Eimeria parasites can lead to severe diarrhoea with considerable clinical and economic consequences in first-year grazing stock. To identify and characterise the cause of diarrhoea observed during previous years, 164 animals on 14 dairy farms in northwestern Germany were included in this study. The calves were physically and parasitologically examined prior to turnout and until 21 days post turnout (d.p.t.). Mean animal weights decreased from 194.9 kg at the start to 189.3 kg bodyweight at the end of the study. In all herds, oocyst counts were very low prior to turnout and increased after the calves had been kept on pasture for at least 7 days. On Day 9 post turnout, 90% and at the end of the study (21 d.p.t.) 70% of all animals showed Eimeria-positive faecal samples. During the course of the study, 79 (48.2%) animals passed faecal samples with more than 100,000 oocysts per gram. The predominant species identified was Eimeria alabamensis, which accounted for more than 83% of the oocysts counted. These parasitological findings matched the clinical observations. Diarrhoea was found in 130 (79.3%) of the study animals. At 5d.p.t. and thus prior to the rise of faecal oocyst counts, a significant increase in diarrhoea was recorded. Calves showing diarrhoea excreted statistically significantly more often over 100,000 E. alabamensis oocysts per gram faeces (0.28; p = 0.0002) than calves without diarrhoea. Diarrhoea was also found during significantly more study days in animals with high oocyst counts (0.39; p = 0.0001). These data indicate that in endemic areas first-year grazing calves must be considered at risk to develop clinical coccidiosis due to E. alabamensis infection during the first 2-3 weeks post turnout.  相似文献

Experiments were conducted to determine whether chukar partridge (Alectoris chukar) chicks would develop protective immunity after inoculation with coccidia. Young chukar chicks in battery cages inoculated with 100 or more oocysts of Eimeria kofoidi or Eimeria legionensis had significant protection at challenge 4 wk later, as measured by greatly reduced oocyst shedding and improved weight gain as compared with unvaccinated, challenged controls. However, when birds were housed in litter pens and vaccinated by various regimens (including two species of chukar coccidia at 100/dose), coccidiosis rapidly spread through all treatments and caused significant mortality. Vaccination with Coccivac-T or with 100 oocysts of Eimeria dispersa did not prevent mortality resulting from accidental contamination, and feed treatment with a Lactobacillus competitive-exclusion product had no benefit. Most if not all of the mortality was from E. kofoidi. This study illustrated the natural fecundity of chukar coccidia in a floor-pen environment where multiplication rate and reinfection combine to produce clinical disease from a small original exposure. Further, these results cast doubt on the potential use of low doses of live oocysts as a vaccine in the chukar partridge.  相似文献

The objective of this study was to evaluate the immunotherapeutic potential of heat killed Mycobacterium phlei in broiler chicken against experimentally produced Eimeria tenella infection. The selected dose of E. tenella oocyst (5 × 103 sporulated oocysts per bird) was capable of producing a mild form of caecal coccidiosis as observed by significant difference in body weight gain, clinical findings and caecal lesion score. Heat killed M. phlei was fed orally at 10 mg per bird with sterile PBS vehicle at alternate day for four doses. Our study reveals that per day body weight gain was significantly (p < 0.01) higher for healthy control compared to coccidia infected group. The group fed M. phlei along with coccidial challenge showed significantly (p < 0.05) higher body weight gain than infected control group. Heat killed M. phlei feeding also found effective to reduce the caecal lesion score significantly (p < 0.05) in comparison to E. tenella infected untreated group. IgA concentrations in serum and bile at 7-day post challenge of coccidial oocyst was also significantly (p < 0.01) higher in M. phlei fed group when compared to coccidia infected and healthy control group. We concluded that use of heat killed M. phlei has a beneficial role as an immunostimulant against caecal coccidiosis in broiler chicken.  相似文献

We compared the therapeutic effect of three anticoccidial drugs (toltrazuril, sulphadimidine and amprolium) in buffalo (Bubalus bubalis) calves experimentally infected with Eimeria bovis (E. bovis) and E. zuernii oocysts (3 x 104oocyst/calf). Buffalo calves (1.5-4 month old, 70-kg body weight) were randomly allocated into 3 groups (9 calves each). Group T was experimentally infected with oocysts and treated with toltrazuril (20 mg/kg BW twice orally at a 1-week interval). Group S was experimentally infected with oocysts and treated with sulphadimidine (125 mg/kg injected IM followed by half dose for 4 successive days). Group A was experimentally infected with oocysts and treated with amprolium (50 mg/kg orally for 7 successive days). Each group had three subgroups (three calves/subgroup) to represent timing of the drug administration: 1st day of coccidia infection (FD), onset of clinical signs of coccidiosis (CC), and onset of oocyst shedding into the faeces (OS). Clinical signs, body-weight gain (BWG) and number of oocysts per gram feces (OPG) were monitored daily for 35 days post-infection (DPI). The OPG were reduced (but the BWG was not different) in the T calves compared to S and A calves. Within the same group, treatment from the 1st day of infection reduced the OPG and increased the BWG compared to the later treatment timings.  相似文献

The aims of the present study were to investigate the excretion of Eimeria alabamensis oocysts by young cattle during their first grazing season and during the first 16 days of their second grazing period. In trial 1, nine first-season grazing heifers were studied and found to have become infected with E. alabamensis shortly after turnout. The next grazing period they were turned out on to a permanent pasture together with two first-season grazing calves. Faecal samples were collected before turnout and then daily from day 3 to day 16. The second-season grazing heifers excreted insignificant numbers of E. alabamensis oocysts, whereas one of the two first-season grazing calves excreted up to 703,000 oocysts/g of faeces (OPG), indicating that the pasture was contaminated. In trial 2, faecal samples were collected from 12 calves before their first turnout in May, daily from day 2 to day 20 after turnout and then once a week until the end of September. The calves grazed pastures used in previous years by first-season grazing calves. Nine of the calves developed clinical E. alabamensis coccidiosis 4-7 days after turnout and excreted more than 950,000 OPG on days 9-10. By day 17 the oocyst excretion had decreased below 900 OPG and remained low throughout the rest of the grazing season. The results of the two studies indicate that reinfections with E. alabamensis are of little clinical importance in calves grazing contaminated pastures, and that young stock infected with E. alabamensis during their first grazing season may be used to cleanse contaminated pastures without risk of developing clinical coccidiosis.  相似文献

Effects of lasalocid on coccidial infection and on calf growth were examined in 16 Holstein bull calves. Calves were assigned randomly to a 2 x 2 factorial arrangement of starter ration containing 0 or 40 mg of lasalocid/kg of starter, beginning when calves were 3 days old (SE = 0.046), and single oral inoculation with 0 or 30,000 sporulated oocysts (Eimeria bovis) at 28 days. Pelleted calf starter was fed ad libitum from day 1; milk replacer was fed at a rate of 3.6 kg/d until day 28. Mean daily gain, dry-matter intake, and body weight were increased in calves fed lasalocid and decreased in those inoculated with coccidia. Addition of lasalocid to the feed improved gains by 8% in uninoculated calves and by 50% in inoculated calves. Fecal oocyst numbers were reduced when lasalocid was fed to inoculated calves. Feces were more abnormal in calves inoculated with coccidia. Respiration rates, rectal temperatures, PCV, and serum sodium and potassium concentrations were unaffected by treatment. On the basis of findings in this study, lasalocid minimized effects of coccidial challenge inoculation and increased growth of calves.  相似文献

For 15 months the anticoccidial effect of 200 ppm clopidol/methyl benzoquate and of 50 ppm robenidine, and the development of immunity against five different species of Eimeria were followed in a closed rabbit population. In unmedicated rabbits, oocyst output decreased progressively with increasing age to a very low level in animals older than four months, but none of the species present disappeared completely in adult animals. No clinical symptoms nor mortality from coccidiosis was noted in reproduction stock. In field conditions E magna and E perforans seemed to induce the weakest resistance, whereas a more marked resistance has been found for E intestinalis and E irresidua. E media appeared to have an intermediate position. Robenidine reduced oocyst output of E magna, E intestinalis, E irresidua, E media and E perforans significantly, whereas clopidol/methyl benzoquate reduced oocyst output of the latter four species only and was least active against E magna. Both drugs also reduced coccidiosis-induced mortality significantly. Medication only before weaning had no distinct influence on coccidial infection, or on mortality by coccidiosis after weaning; nor did those parameters differ significantly between continuously medicated rabbits and rabbits medicated after weaning only. As reproductive stock is protected by immunity, this makes the necessity of medicating does and bucks with anticoccidials questionable in intensive or semi-intensive reproduction systems.  相似文献
