A total of 60 isolates of Haemophilus spp. from chickens, including four reference strains of H. paragallinarum and one of H. avium, were examined for their physiological and biochemical properties. The isolates could be placed into two groups. One group was identified as H. paragallinarum and consisted of 43 isolates including the four reference strains of H. paragallinarum. The other group was identified as H. avium and consisted of 17 isolates including the reference strain of H. avium. H. avium can be differentiated from H. paragallinarum by its possession of the enzymes catalase and alpha-glucosidase, capacity to grow in air, production of acid from galactose, and by the fact that its growth is not improved by the addition of chicken serum. In addition, the majority of H. avium isolates, unlike H. paragallinarum, possess a yellow pigment and produce acid from trehalose.  相似文献

Bordetella avium heat-labile toxin (HLT) was lethal for poults, mice, and embryonating chicken eggs. It produced hemorrhagic lesions in turkey and guinea pig skin. Antiserum made in turkeys neutralized the lethality of the toxin and its ability to produce hemorrhagic skin lesions. Further, antiserum against HLT of an Ohio strain neutralized lethality of HLT of strains from Iowa, North Carolina, and West Germany. The antiserum did not neutralize lethality of HLT from B. bronchiseptica. Bordetella avium HLT was not ciliostatic for turkey tracheal-ring cultures and did not stimulate adenyl cyclase activity using mouse adrenal cell cultures.  相似文献

Histophilus somni is an important pathogen of cattle and sheep. H. somni requires iron and can use ruminant transferrins as iron sources for growth. Here, we investigated the abilities of bovine (strains 649 and 2,336) and ovine (strains 9L and 3384Y) isolates of H. somni to acquire iron from haemoglobins. Using growth assays, the bovine isolates were shown to acquire iron from bovine haemoglobin, but not from ovine, porcine or human haemoglobins; the ovine isolates, however, failed to use any of these haemoglobins as iron sources for growth. In solid phase binding assays, the bovine isolates, grown under iron-restricted conditions in the presence of bovine haemoglobin, bound not only bovine but also ovine and human haemoglobins. Competition binding assays indicated that all three haemoglobins were bound by the same receptor(s) and SDS-PAGE of membrane fractions revealed that expression of haemoglobin-binding activity was associated with the production of an approximately 120-kDa outer membrane protein. PCR approaches allowed the amplification and sequencing of hgbA, and also hugX and hugZ homologues from strains 649, 9L and 3384Y. While hgbA of strain 649 was predicted to encode an HgbA precursor that is processed to yield a mature, 123.9-kDa haemoglobin-binding protein, the hgbA genes of strains 9L and 3384Y were predicted to give rise to truncated products. RT-PCR experiments revealed that in strain 649, hugX, hugZ and hgbA are co-transcribed and iron-regulated and additional sequencing suggested that in strain 2336, expression of HgbA is subject to phase variation involving a poly C tract within hgbA.  相似文献

Extraintestinal pathogenic E. coli (ExPEC) are responsible for many infectious diseases in livestock, such as airsacculitis in poultry, acute mastitis in dairy animals and neonatal septicaemia and urinary tract infections (UTI) in pigs and cattle. In their animal hosts, ExPEC have to cope with low iron availability. By using different strategies, ExPEC strains are able to retrieve iron sequestered by host proteins. One of these strategies is the use of siderophores, which are small secreted molecules with high affinity for iron. ExPEC are known to synthesize up to four different types of siderophores: enterobactin, salmochelins, yersiniabactin and aerobactin. Steps required for iron acquisition by siderophores include (1) siderophore synthesis in the cytoplasm, (2) siderophore secretion, (3) ferri-siderophore reception, (4) ferri-siderophore internalization and (5) iron release in the cytoplasm. Each siderophore has specific properties and may be differentially regulated to provide different advantages, potentially allowing ExPEC to adapt to different environmental conditions or to overcome host innate immunity. Iron acquisition by siderophores plays a significant role in ExPEC virulence and, as it requires outer membrane receptors, it constitutes an interesting target for the development of vaccines that could be used to limit the number of infectious diseases due to ExPEC in livestock.  相似文献

Four field isolates (S4, S10, S15, and S17) of Haemophilus paragallinarum were recovered from chickens affected with infectious coryza in widely separated regions of Japan. Their hemagglutinating (HA) activity and immunological properties were compared with those of strain 221 of serovar A/1 and strains Modesto and S1 of serovar C/2. When treated with potassium thiocyanate or hyaluronidase, all the isolates showed HA activity against formaldehyde-fixed chicken erythrocytes but not against fresh chicken erythrocytes. In the hemagglutination-inhibition (HI) test, the isolates cross-reacted with strains Modesto and S1 but not with strain 221. The immunological properties of these isolates, as determined by cross-protection tests, were similar to those of strain S1 and, to a lesser degree, strain Modesto, but not to strain 221. Our results indicated that the four field isolates belong to serovar C/2 and that the HI test is a suitable method for serotyping H. paragallinarum.  相似文献

The virulence of the reference strains of the nine currently recognized Kume serovars of Haemophilus paragallinarum was investigated. The capacity of the H. paragallinarum strains to cause the typical clinical signs of upper respiratory tract disease associated with infectious coryza in unvaccinated, nasal-challenged chickens was assessed. Differences in virulence were assessed by means of a standardized scoring system for clinical signs. All nine strains were pathogenic to chickens, producing typical clinical signs of infectious coryza. The highest clinical signs score was obtained for serovar C-1 (1.72), while the lowest clinical signs score was obtained for serovar C-4 (0.32). Our results indicate that virulence differences exist among the serovars of H. paragallinarum.  相似文献

Ornithobacterium rhinotracheale (ORT) is an emerging respiratory pathogen of poultry in North America that is causing millions of dollars in economic losses to the poultry industry. Ornithobacterium rhinotracheale is associated with airsacculitis, pleuritis, pneumonia, and consolidation of lungs. Little is known about the molecular mechanisms of infection. In this study, the mechanism of iron acquisition by O. rhinotracheale was explored. O. rhinotracheale strains grown under iron deprivation in media containing 200 microM 2,2'-dipyridyl did not secrete siderophores as measured by the chrome azurol S (CAS) agar and CAS solution assays. Filter disks impregnated with various protein-bound iron compounds and inorganic iron salts of Fe(III) and Fe(II) placed on iron-restricted agar inoculated with a lawn of O. rhinotracheale supported growth from sheep and porcine hemoglobins, ovotransferrin, Fe(III), and Fe(II), but they did not support growth from bovine transferrin, bovine apo-transferrin, bovine lactoferrin, and hemin. However, both bovine hemoglobin and transferrin supported growth of O. rhinotracheale serotype C. Four immunoreactive proteins involved in iron acquisition were identified in an O. rhinotracheale membrane extract by using mass spectrometry. Furthermore, O. rhinotracheale field strains showed differential sensitivity to 2,2'-dipyridyl. Of the 72 field strains tested, 22 strains were resistant to the iron chelator at concentrations of 50 microM and 100 microM, suggesting this attribute may be related to disease-producing potential of these strains. This is the first report on the identification of the iron acquisition mechanism of O. rhinotracheale.  相似文献

Two isolates of Haemophilus paragallinarum were obtained from a layer chicken in Mexico. The isolates were confirmed as H. paragallinarum by polymerase chain reaction and conventional biochemical identification. The isolates were nicotinamide adenine dinucleotide (NAD) independent-growing on blood agar without the need of a nurse colony as well as on a complex medium that lacked both NAD and chicken serum. Both isolates were pathogenic, causing the typical clinical signs of infectious coryza in susceptible chickens. One isolate was Page serovar B/Kume serovar B-1 and the other isolate was Page serovar C/Kume serovar C-2. The isolates were associated with a field outbreak that involved an egg drop of 20% over a 3-wk period and a doubling of weekly mortality (from 0.1% to 0.2%). This is the first report of NAD-independent H. paragallinarum outside South Africa and is the first time that NAD-independent H. paragallinarum of serovar B has been reported.  相似文献

Two serovar-specific monoclonal antibodies (MAbs) to Haemophilus paragallinarum serovars A/1 and C/2 strains, respectively, were developed and characterized by hemagglutination-inhibition (HI) and dot-blotting tests using representative H. paragallinarum serovars A/1, B, and C/2 strains. In both the HI and dot-blotting tests, one MAb (E5C12D10), raised against strain 221, serovar A/1, reacted only with serovar A/1 strains, while the other MAb (F2E6), raised against strain S1 of serovar C/2, reacted with only serovar C/2 strains examined. In both tests, the two MAbs did not react with two serovar B strains. These results indicated that the two MAbs recognize serovar-specific hemagglutinating (HA) antigens of H. paragallinarum serovars A/1 and C/2 strains, respectively, and that a dot-blotting test using these MAbs is a practical alternative to the HI test for serotyping H. paragallinarum. Strains 0222 and Spross of serovar B, which did not react with these two MAbs, were found to possess serovar-specific HA antigen in cross-HI tests.  相似文献

An oil-based bacterin, containing strains 083 and 0222 of Haemophilus paragallinarum, is commonly used in South Africa to vaccinate laying flocks against infectious coryza. Two strains of H. paragallinarum, designated M85 and SB86, were isolated from infected but vaccinated commercial laying flocks in two incidental outbreaks of coryza in 1985 and 1986. A panel of five monoclonal antibodies was established which clearly distinguished the vaccine strains from the field isolates. One of these reacted with only vaccine strains A and B, another reacted with only field strains M85 and SB86, and the remaining three cross-reacted to various degrees with all four strains or isolates. Immunoassays were performed by enzyme-linked immunosorbent assay using whole bacteria as solid-phase antigen. These monoclonal antibodies may aid in serotyping new field isolates of H. paragallinarum and in improved standardization of vaccine strains.  相似文献

ABSTRACT: This study assessed the presence of sialic acid α-2,3 and α-2,6 linked glycan receptors in seven avian species. The respiratory and intestinal tracts of the chicken, common quail, red-legged partridge, turkey, golden pheasant, ostrich, and mallard were tested by means of lectin histochemistry, using the lectins Maackia amurensis agglutinin II and Sambucus nigra agglutinin, which show affinity for α-2,3 and α-2,6 receptors, respectively. Additionally, the pattern of virus attachment (PVA) was evaluated with virus histochemistry, using an avian-origin H4N5 virus and a human-origin seasonal H1N1 virus. There was a great variation of receptor distribution among the tissues and avian species studied. Both α-2,3 and α-2,6 receptors were present in the respiratory and intestinal tracts of the chicken, common quail, red-legged partridge, turkey, and golden pheasant. In ostriches, the expression of the receptor was basically restricted to α-2,3 in both the respiratory and intestinal tracts and in mallards the α-2,6 receptors were absent from the intestinal tract. The results obtained with the lectin histochemistry were, in general, in agreement with the PVA. The differential expression and distribution of α-2,3 and α-2,6 receptors among various avian species might reflect a potentially decisive factor in the emergence of new viral strains.  相似文献

The antigenic interrelatedness of 3 clone-purified turkey reoviruses (NG-Turkey, 82-88, and NC-TEV) to each other and to 4 clone-purified chicken reoviruses (S1133, Co8, Fahey-Crawley, and avian type 2) was determined in reciprocal cross-neutralization tests, using polyclonal antisera and the plaque-reduction technique. The morphologic features of plaques formed under agar were studied for all 7 reoviruses, and size comparisons for turkey vs chicken isolates were made. All 3 turkey reoviruses (with the exception of NG-Turkey vs Fahey-Crawley chicken reovirus) formed plaques significantly (P less than 0.05) smaller than plaques produced by their chicken counterparts. The 3 turkey reoviruses were closely related to each other and to chicken reovirus CO8. The antigenic differences between turkey reoviruses 82-88 and NC-TEV and chicken reovirus S1133 were slight (minor subtype); however, the latter and NG-Turkey were serotypically distinct. The NG-Turkey and 82-88 turkey reoviruses were more related (minor subtype) to the Fahey-Crawley and avian type 2 chicken reoviruses, than was NC-TEV turkey reovirus (major subtype).  相似文献

Both the native intestinal microflora of chickens that protected chicks against salmonellae and Escherichia coli and native turkey intestinal microflora were evaluated for their reciprocal protective capacity in both species against Salmonella typhimurium and a pathogenic strain of E. coli. Nalidixic-acid-resistant forms of the S. typhimurium and E. coli strains were used in seeder-bird and individual-bird challenge tests. Reciprocal protection was provided by native chicken and turkey intestinal microflora in chicks and poults against S. typhimurium and the pathogenic strain of E. coli. The chicken and turkey microflora appeared to be equally effective in protecting the two species from S. typhimurium, but protection against E. coli was somewhat greater in the chicken than in the turkey.  相似文献

Acinetobacter baumannii (A. baumannii) is a miscellaneous bacterium with ability of extensive antibiotic resistance. A. baumannii strains have also been isolated from animal origins. The objective of our atudy was characterization of A. baumannii antibiotic resistance and virulence traits from turkey and chicken raw meat. Of 576 turkey and 424 chicken specimens during 2017–2019, 200 (120 from turkey and 80 from chicken) isolates were identified as A. baumannii. Virulence factors and antibiotic resistance patterns of A. baumannii were determined using polymerase chain reaction (PCR) technique and Kirby-Bauer test. All the isolates were resistant to tetracycline and cefoxitin and 81 % and 56 % of them produced ESBLs and carbapenemases. Also 74 % of them (34 % from chicken and 40 % from turkey) were multidrug-resistant (MDR) A. baumannii. Colistin and fosfomycin non-susceptibility was detected among 12 % and 10 % of them, respectively. The existence of tetA, dfrA, tetB, blaoxa-51-like, blaoxa-23-like, sul1, blaoxa-24-like, blaoxa-58-like, fosA3 and mcr-1 genes accounted for 80 %, 71 %, 70.5 %, 66 %, 62 %, 43 %, 34 %, 22 %, 11 % and 13 % of them, repectively. Additionally, predominant virulence factors included the fimH, afa/draBC, sfa/foc DE, cnfI and cnf2 genes. The rate of antibiotic resistance genes and virulence factors was not significantly different between turkey and chicken (p > 0.05). High rate of antibiotic non-susceptibility even against last-line resorts in poultry products is a concern and suggest that animals play a potential role as reservoirs of transmission of MDR A. baumannii.  相似文献

OBJECTIVE: To determine the level of adoption within the Australian poultry industries of a range of standard biosecurity practices. DESIGN: A questionnaire survey of owners and managers of 1753 commercial poultry farms. Procedure Information was collected by telephone or in person from the owners and managers of poultry farms. Data were entered onto an Excel spreadsheet and analysed using the statistical tools within Excel. RESULTS: The response rates to the questionnaire for the chicken meat, egg, duck and turkey sectors were 94%, 37%, 92% and 96% respectively. Within the chicken meat sector, the adoption rate exceeded 98% for 11 practices with only the rates for appropriate disposal of spent litter/manure (94%) and development of farm level biosecurity plans (88%) falling below this level. Adoption rates for all practices had increased or remained at 100% since the last survey conducted in 2001. Integrated companies in the turkey and duck sector also reported high levels of adoption for most biosecurity practices although rates for the use of fresh litter, cleaning of sheds between batches of birds and maintenance of single age flocks were lower than in the chicken meat sector. Adoption rates for biosecurity practices on independently owned farms in the turkey and duck sectors were lower than for farms operated by integrated companies. CONCLUSIONS: Farms in the chicken meat, turkey and duck sectors of the Australian poultry industries have a high level of adoption of biosecurity practices with only a small proportion of farms needing to further enhance their biosecurity.  相似文献

Adherence to turkey tracheal mucosa and agglutination of guinea pig erythrocytes were determined for 20 strains of Bordetella avium. Ten type-I strains, 7 type-II strains, 2 transposon-induced mutants, and 1 revertant were evaluated. All type-I strains adhered readily to tracheal mucosa and agglutinated erythrocytes, whereas no type-II strains adhered to trachea or caused hemagglutination (HA). Two mutants selected for loss of HA activity were less adherent to tracheal mucosa, compared with the parent strain. Reversion of one mutant to HA-positive status was accompanied by reconstitution of much of its adherence capacity. Results of this study provide preliminary evidence that tracheal adherence and HA of B avium are closely related.  相似文献

A cross-sectional study determined the prevalence of ecto-, endo- and haemoparasites in free-range chickens from the Goromonzi District, Zimbabwe. Fifty young and 50 adult birds were selected randomly. All chickens harboured ecto- and endoparasites, and 32% were infected with haemoparasites. Eight different ectoparasites were identified; the more prevalent ones had the following prevalences (young, %; adult, %): Argas persicus (6; 14), Cnemidocoptes mutans (6; 32), Echidnophaga gallinacea (72; 74), Goniocotes gallinae (0; 22), Menacanthus stramenius (90; 88) and Menopon gallinea (24; 66). The prevalences of C. mutans, G. gallinae and M. gallinae were higher in adults compared to young chickens. The mean (+/-S.D.) number of helminth species per chicken was 6.7+/-2.0 for young chickens and 6.4+/-2.0 for adult chickens with a range of 1-10 for young chickens and a range of 1-11 for adult chickens. The most prevalent nematodes identified were (with prevalence in % for young/adult birds): Allodapa suctoria (76; 72), Ascaridia galli (48; 24), Gongylonema ingluvicola (28; 56), Heterakis gallinarum (64; 62) and Tetrameres americana (70; 62). For cestodes the prevalences were: Amoebotaenia cuneata (60; 68), Hymenolepis spp. (62; 80), Raillietina echinobothrida (66; 34), Raillietina tetragona (94; 100) and Skrjabinia cesticillus (50; 76). The young chickens had higher prevalences of A. galli and R. echinobothrida compared to adults, but lower prevalence of G. ingluvicola and S. cesticillus. Eimeria spp. oocysts were isolated in 36% of 47 investigated samples. The prevalence was 47% for young chickens and 18% for adult chickens. Prevalences (in %) of haemoparasites in young and adult chickens were: Aegyptinella pullorum (7; 6), Leucocytozoon sabrazesi (3; 1), Plasmodium gallinaceum (8; 6) and Trypanosoma avium (2; 3).  相似文献
