An acute stress response can be provoked by abrupt social, nutritional, and environmental changes associated with weaning, and this may disrupt homeostasis and thus compromise well-being. Manipulating environmental factors, such as photoperiod, might provide a simple way to reduce the physiological consequences that piglets experience due to weaning stress. The objective of this study was to evaluate the impacts of photoperiod manipulation across various weaning ages on leukocyte populations, lymphocyte proliferation, natural killer cytotoxicity (NK), chemotaxis, phagocytosis, and immunoglobulin G, cortisol, and BW of piglets during the nursery phase. Sixty-eight crossbred piglets were obtained from sows kept on a short-day (8 h of light/d) photoperiod from d 90 of gestation until weaning. Piglets were weaned at 14, 21, or 28 d of age and kept on a short or long (16 h of light/d) photoperiod until 10 wk of age. Piglet BW and blood samples were collected at weaning and at 6, 8, and 10 wk of age. Pigs weaned at 28 d had reduced neutrophil counts (P < 0.001), phagocytosis (P < 0.001), and lymphocyte proliferation (P < 0.05) at weaning compared with those weaned at 14 and 21 d. Pigs weaned at 21 d tended to have lower (P = 0.08) lymphocyte counts than did pigs weaned at 14 or 28 d. Pigs weaned at 14 d had reduced (P < 0.01) NK relative to those weaned at 21 or 28 d. Photoperiod also influenced pig BW and immune status. Generally, those pigs on the long-day photoperiod and weaned at 28 d were heavier (P < 0.001) than their counterparts weaned at 14 or 21 d. At 6 wk of age, NK was greater (P = 0.002) in pigs kept on a long day and weaned at 14 or 21 d than in pigs weaned at 28 d. Phagocytosis was less (P = 0.005) at 6 wk of age, but was greater at 8 wk, in piglets kept on the long day and weaned at 28 d than in long-day pigs weaned at 14 or 21 d. These results suggest that photoperiod differentially influences immune responses in piglets weaned at different ages and indicate an inverse relationship between growth and immune status. Here, weaning at 28 d and a long-day photoperiod was the treatment combination that was most physiologically beneficial to piglets, whereas a 14-d weaning and short-day photoperiod was least physiologically beneficial.  相似文献

Three trials using 156 Yorkshire x Hampshire x Duroc crossbred pigs (avg initial wt, 7.9 kg) were conducted to evaluate the effects of two supplemental dietary vitamin E (11 vs 220 IU/kg of diet) and weaning age (21, 28 or 35 d) on performance and immunocompetence of pigs. Supplemental vitamin E (220 IU/kg of diet) increased (P less than .01) serum concentrations of vitamin E for all weaning ages compared with pigs fed 11 IU of vitamin E/kg of diet. However, supplemental vitamin E did not affect performance, serum cortisol concentration or the primary and secondary antibody response to sheep red blood cells. As weaning age increased, weekly ADG and avg daily feed intake increased linearly (P less than .01). Cortisol levels decreased during the 1st wk following weaning and then increased linearly (P less than .01) over time; pigs weaned at 35 d of age had higher (P less than .01) cortisol values initially and over time than pigs weaned at 21 and 28 d. Pigs weaned at 35 d had a higher (P less than .01) primary response to sheep red blood cells than pigs weaned at 21 and 28 d of age, but this effect was not observed for the secondary response. There were no interactive effects (P greater than .10) of dietary vitamin E level and weaning age. In summary, the highest level of supplemental vitamin E increased serum vitamin E concentration but did not affect performance, cortisol levels or one test of the immune response, antibody titers to red blood cells.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In a series of five 17-d replicate trials, a total of 54 cannulated and 12 noncannulated pigs were used to determine the effects of weaning age (17 d or 24 d) on pH, dry matter percentage, aerobic and anaerobic microflora, lactate, and volatile fatty acid (VFA) concentrations in the jejunum, ileum, and cecum of weanling pigs. At -14 d of age, cannulated pigs were surgically fitted with T-cannulas in the jejunum (n = 20), ileum (n = 18), or cecum (n = 16). Upon weaning, cannulated pigs were individually caged in an environmentally controlled room with ad libitum access to a phase starter diet and water. Noncannulated pigs were killed at weaning and samples were collected from the jejunum, ileum, and cecum. Digesta and fecal swabs from cannulated pigs were collected twice weekly. The pH of cecal contents was lower (P < 0.05) and dry matter percentage was greater (P < 0.05) than those ofjejunal or ileal contents. Pigs weaned at 24 d of age had increased (P < 0.05) E. coli populations 3 d postweaning compared to preweaning populations, regardless of site of collection, whereas this increase was not observed in pigs weaned at 17 d of age. Unweaned pigs maintained higher (P < 0.05) lactobacilli populations compared to weaned pigs; however, populations declined (P < 0.05) in both groups by 3 d postweaning, with pigs weaned at 24 d of age having lactobacilli populations greater than pigs weaned at 17 d of age. Fecal populations of E. coli and lactobacilli declined (P < 0.05), whereas fecal bifidobacteria populations increased (P < 0.05) postweaning, regardless of weaning age. Concentrations of total fecal anaerobes declined (P < 0.05) in pigs weaned at 17 d of age but were maintained in pigs weaned at 24 d of age. Volatile fatty acid concentrations were greater (P < 0.05) in the cecum than in the jejunum or ileum, and acetic acid concentrations decreased (P < 0.05) postweaning regardless of weaning age. A tendency for L+ lactate concentrations to be greater (P < 0.07) in the ileum and jejunum vs the cecum was observed. Results indicate that weaning and weaning age have significant effects on microbial populations and VFA concentrations.  相似文献

To examine the effects of exogenous porcine (p) ST on measures of stress and immune function in weaned pigs with or without transport, pigs (20 +/- 1 d of age) received daily injections of pST (0.5 mg/kg; n = 16) or saline (n = 16) for 5 d. On d 5, a blood sample was collected immediately before injection. At 4 h postinjection, pigs were weighed, sampled for blood, injected with di-nitrophenyl-conjugated keyhole limpet hemocyanin, and weaned. One half of the pigs in each group were transported for 3 h before placement in the nursery. Pigs were weighed, and blood was collected on 1, 7, and 14 d postweaning. Statistical significance was set at P < 0.05. Serum IGF-I concentrations were increased by pST and decreased by weaning, but not affected by transport. The free cortisol index was elevated in all pigs 1 d postweaning, although less in transported versus nontransported pigs. By 7 d postweaning, the free cortisol index returned to prewean values. Serum concentrations of immunoglobulin (Ig) G increased in all pigs by 14 d postweaning, but were not affected by pST or transport. Serum IgM concentrations were elevated at 7 and 14 d postweaning. Before weaning and again 1 d postweaning, pigs treated with pST had greater concentrations of IgM than did control animals. Circulating neutrophils increased in pST-treated pigs 4 h after the final pST injection. Improved immune function in weaned pigs by pST may lead to greater health and growth in a commercial setting.  相似文献

Phosphorylated mannans derived from the yeast cell wall of Saccharomyces cerevisiae may beneficially modulate immune function in the weanling pig, possibly providing an alternative to the use of dietary growth-promoting antibiotics. Therefore, in this study, 32 pigs averaging 19 d of age and 5.7 +/- 0.2 kg initial BW were randomly assigned to 16 pens in an environmentally controlled nursery to determine the effects of dietary supplementation with phosphorylated mannans on growth and immune function. Average daily gain and G:F ratio increased (P < 0.05) when pigs were fed diets supplemented with mannans from d 0 to 14 after weaning and in the overall experiment. Percentage of neutrophils was lower (P < 0.08) and percentage of lymphocytes was higher (P < 0.05) in blood from pigs fed mannans than when pigs were fed the basal diet. Lamina propria macrophages isolated from pigs fed diets containing mannans phagocytosed a greater (P < 0.05) number of sheep red blood cells (2.63 +/- 0.11) than did lamina propria macrophages isolated from pigs fed the basal diet (2.31 +/- 0.11). On d 19 after weaning, pigs fed diets supplemented with mannans tended to have a greater (P < 0.10) percentage of CD14+ lamina propria leukocytes than did pigs fed the basal diet. On d 21 following weaning, the percentage of CD14+MHCII+ leukocytes isolated from lamina propria tissue tended (P < 0.10) to be lower when pigs were fed mannans than when pigs were fed the basal diet. Pigs fed diets containing mannans had a lower (P < 0.05) ratio of CD3+CD4+:CD3+CD8+ T lymphocytes isolated from jejunal lamina propria tissue only on d 21 after weaning compared with pigs fed the basal diet. Supplementation of mannans in the diets of weanling pigs improved gain and efficiency, and intermittently affected selected components of the young pigs' immune function both systemically and enterically.  相似文献

试验旨在探讨牛膝多糖(ABPS)对脂多糖(LPS)刺激的断奶仔猪生长性能、炎性介质和内分泌激素的影响。选用48头(28±3)d(8.45±0.14)kg的杜洛克×长白×大白断奶仔猪,采用2×2因子设计,包括日粮处理(0或500 mg/kg ABPS)和免疫应激(注射LPS或生理盐水)。试验期28 d。在第14天和第21天,每日粮组一半的猪注射100μg/kg BW的LPS,另一半注射生理盐水作对照。注射后3 h,采血测定血浆肿瘤坏死因子-α(TNF-α)、前列腺素E2(PGE2)、皮质醇、生长激素(GH)和类胰岛素生长因子-I(IGF-I)含量。结果表明:LPS刺激显著降低了第14~21、22~28天的日增重(ADG)和日采食量(ADFI)(P<0.05或P=0.05);ABPS显著提高了22~28 d的ADG(P<0.05)。LPS刺激显著提高了14 d和21 d的TNF-α、PGE2和皮质醇的含量,降低了IGF-I的含量(P<0.01);ABPS显著缓解了LPS刺激导致的TNF-α(14 d和21 d)和皮质醇(14 d)含量的上升以及IGF-I(14 d和21 d)含量的下降(P<0.05)。说明ABPS缓解了免疫应激仔猪生长的抑制,其机制可能与其抑制了炎性介质的分泌有关。  相似文献

Postweaning growth lag in baby pigs weaned at 28 d was studied by using three weaning stress treatments. Treatments consisted of a control in which pigs continued to nurse the dam, had access to a dry feed at 14 d of age and were not weaned until after the study. Pigs were adjusted to liquid and dry feeds at 14 d of age in Treatments 2 and 3, but sows were removed from the pens at 28 d of age in Treatment 2, whereas sows were removed and room temperature lowered to 13 degrees C in Treatment 3. In Treatment 4, sows were removed but pigs were fed the dry diet only from 28 d of age. Blood and tissue were collected and evaluated every 12 h for 48 h on slaughtered pigs and blood was sampled every 12 h for 132 h from pigs catheterized in the vena cava. Pigs weaned with a dry diet in Treatment 4 were the only pigs to lose weight (P less than .01) and have typical symptoms of postweaning growth lag. These pigs had the lowest (P less than .01) mean plasma glucose, highest (P less than .01) free fatty acids and the highest (P less than .05) cortisol concentrations. Their mean duodenal pH also was higher (P less than .01), whereas pigs given both milk and dry diets and stressed by weaning in a warm or cool room (Treatments 2 and 3) had lower (P less than .01) duodenal pH values than pigs continuing to nurse the sow. In this study, pigs having access to milk and dry diets prior to weaning had no adverse symptoms when the sow was removed regardless of whether or not they were exposed to cold after weaning. However, pigs that were abruptly weaned with a dry diet had slow growth, low plasma glucose, high free fatty acids and low liver glycogen.  相似文献

The effects of weaning, week postweaning and diet composition on concentration of lipase in the pancreas and small intestinal lumen were investigated in weanling swine. In Exp. 1, lipase levels were evaluated in suckling pigs from 2 to 35 d of age and in pigs weaned at 21 or 35 d of age. Pigs weaned at 21 d of age were fed a corn-soybean meal diet with lipase levels measured from 3 to 28 d postweaning. Pancreas weights increased during the suckling period; they were lowered at 3 d postweaning and were lower at 7 d postweaning than in suckling pigs but increased linearly from 3 to 28 d postweaning. Lipase level per unit wet tissue and total pancreatic levels increased from 2 to 35 d of age in suckling pigs (P less than .01). Weaning at 21 d of age resulted in a decline (P less than .05) in lipase levels in the pancreas at 3 and 7 d postweaning, but the levels subsequently tended to increase between 7 and 28 d postweaning. Whereas relative lipase levels in the intestinal lumen increased from 2 to 35 d of age in suckling pigs, total luminal enzyme did not decline upon weaning when pigs were weaned at either 21 or 35 d of age. Total luminal lipase per unit empty body increased linearly (P less than .01) each week postweaning. To evaluate morphological characteristics and development of the immune system at different ages in neonatal pigs, 4 piglets were euthanized at 7, 14, and 18 d of age for collection of blood, bile, and intestinal tissue for morphological measurements. Blood was collected for differential cell counts, lymphocyte blastogenesis, immunoglobulin (Ig) concentrations, cytokine concentrations, and flow cytometric analysis. Bile was collected for quantification of Ig-A and Ig-M. Villus width and crypt depth from duodenum sections, as well as ileum crypt depth, were reduced (P < or = 0.08) in 18-d-old pigs compared with 7-d-old pigs. No age-related differences (P > or = 0.11) were observed in the number of goblet cells with neutral and acidic mucins, serum or enteric Ig concentrations, IL-2, IL-4, spontaneous lymphocyte proliferation, or leukocyte concentrations. When measured as counts per minute (cpm) and as a stimulation index (SI), lymphocyte proliferation responses to phytohaemagglutinin increased (P = 0.05) between 7 and 14 d of age; no changes (P = 0.10) occurred at 18 d of age. No age-related changes (P = 0.39) were observed in response to pokeweed mitogen (PWM) when measured as cpm; however, the SI from PWM-induced lymphocytes decreased (P = 0.04) 4-fold between 7 and 18 d of age. The CD4+:CD8+ and populations of lymphocytes expressing CD2+CD4+CD8- (T helper cells) and CD25+CD4+CD8- (activated T helper cells) were greater (P > or = 0.04) at 7 d of age than at 14 and 18 d. Populations of T lymphocytes, cytotoxic T cells (CD2+CD4-CD8+), activated lymphocytes (CD25+), and activated cytotoxic T cells (CD25+CD4-CD8+) were greater (P > or = 0.02) in 18-d-old pigs compared with 7-d-old pigs, whereas CD2+CD4-CD8- [double negative cells] were lower (P = 0.08) in 18-d-old pigs compared with 14-d-old pigs. The percentage of CD2+ T cells was 8.4% at 7 d of age, and by the time the pigs reached 18 d of age, the percentage of CD2+ T cells was 33.8%. Moreover, the percentage of gammadelta T cells was greater (P = 0.02) in 18-d-old pigs than in 7-d-old pigs (74.8 vs. 46.1%, respectively). Results indicate that the porcine immune system and gut are continuously changing as the young pig matures. Changes occurred in lymphocyte phenotypic expression and functional capabilities, as well as morphology and mucin production, and their role may be to further protect the neonate from antigenic challenge as protection from passive immunity declines.  相似文献

Weanling pigs (n = 176), 21 to 26 d of age (average initial weight, 7.1 kg), were used to evaluate the effects of weaning time (evening vs morning) and feeding time (immediate vs delayed) on postweaning performance. Pigs were randomly allotted within litters to four treatments in a 2 x 2 factorial arrangement as follows: 1) weaning at 2000 with immediate feeding, 2) weaning at 2000 with 12-h delayed feeding, 3) weaning at 0800 with immediate feeding, and 4) weaning at 0800 with 12-h delayed feeding. A 12:12 light:dark photoperiod was maintained. Four pigs were held per pen, and all pigs were given ad libitum access to a 20% CP corn-soybean meal diet with 15% dried whey throughout the 28-d study. The interactions between weaning and feeding times were not significant. During the 28-d period, pigs weaned at 2000 consumed 5% more feed (P less than .10) and grew 6% faster (P less than .05) than pigs weaned at 0800. Feed to gain ratios were similar for both time treatments. Variance of ADG within pens as measured by range, logarithm of variance, and CV were generally smaller for pigs weaned at 2000 than for pigs weaned at 0800 with no effect of feeding time. Pigs fed immediately after weaning and those delayed for 12 h had similar overall feed consumption, ADG, and feed to gain ratios. Weaning pigs in the evening enhanced performance, perhaps by better matching the normal diurnal eating and activity patterns and weaning time.  相似文献

Two experiments were conducted to determine the effect of nutritional status, age at weaning and room temperature on growth and immune response of pigs. In the first experiment, 72 pigs were weaned at either 2 or 3 wk and fed either a complex, simple-adequate or simple-inadequate diet for a 24-d period. Total antibody (TAb) titers in response to injections of human red blood cells (RBC) on either 0, 4, or 16 d postweaning were not affected by age at weaning or dietary treatments even though the nutritional status was compromised to such an extent that pigs fed the simple-adequate diet gained at one-half the rate (.172 vs .349 kg/d) of pigs fed the complex or simple-adequate diets (P less than .01). In Exp. 2, the immunological response of 144 pigs weaned at 3 wk were evaluated at 4 and 8 d postweaning. Pigs were weaned into rooms that averaged either 25 or 18 C, and were fed either a complex or simplex-adequate diet at either ad libitum (AL) or restricted (Res) levels for a 24-d period. Restriction of either the complex or simple diet for the first 16 d of the trial drastically reduced (P less than .01) the growth rates of Res pigs compared with AL pigs (.042 vs .236 kg/d). There were no differences in skinfold thickness (Sf) or TAb, 2-mercaptoethanol sensitive (MEs) and resistant (MEr) antibody titers when pigs were inoculated with phytohemagglutinin-P (PHA-P) or RBC at 4 d postweaning regardless of room temperature, diet source or feeding level.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This study evaluated immune cell populations in pigs following weaning and vaccination for Mycoplasma hyopneumoniae. Piglets (n = 24) were weaned (day 0) at 16 (±1) days of age, and randomly assigned to the vaccination group (n = 16) or control group (n = 8). Complete blood cell counts, flow cytometry and serology were completed for blood samples collected on days 0 (within hours of weaning), 3, 7, 14, 30 and 60. The M. hyopneumoniae S:P ratios (sample optical density: positive control optical density) were negative in the vaccination group until days 30 and 60, when the S:P ratios were 1.3 and 1.0, respectively. Control animals remained serologically negative. The percentage of CD4+ T cells was less (P < 0.01) in control pigs than vaccinated pigs at day 3. In contrast, numbers of CD8+ and CD4+CD8+ T cells were greater (P < 0.01) in control pigs than in vaccinated pigs at days 3 and 7. After day 7, few differences in immune cell types were evident between the groups. Differences in lymphocyte populations could not be solely attributed to vaccination, due at least in part, to the confounding influence of weaning. It was difficult to distinguish the influence of vaccination from the impact of weaning on peripheral immune cell populations.  相似文献

Two experiments were conducted that demonstrated that a single injection of hydrocortisone 21-acetate (HYD, 25 mg/kg BW) administered to 6-d-old nursing piglets resulted in a twofold elevation (P less than .02) of pancreatic amylase within 2 d; activity was unaffected by an injection of 15 IU adrenocorticotropic hormone (ACTH)/kg BW (P greater than .20). Intestinal sucrase and maltase activity tended to be elevated (P less than .20) 2 and 4 d postinjection with HYD but returned to normal (uninjected) levels by 14 d of age. The normal decline of intestinal lactase activity was delayed by at least 4 d in response to both hormones (P less than .10). Organ weights were not affected by either hormone. In a separate experiment, postweaning mortality was reduced (12 vs 27%) and growth rate was substantially improved by administration of HYD to piglets 4 and 2 d prior to weaning at 14 d of age. Hydrocortisone resulted in a faster rate of gain the 1st wk postweaning for pigs weaned at 21 or 28 d. Subsequent gain by control and HYD piglets weaned on d 21 was similar, but HYD subsequently impaired growth rate of piglets weaned at 28 d of age. Growth rates of control and ACTH piglets were similar at each postweaning period regardless of weaning age (weaning age [lin.] x week postweaning [quad.] x treatment, P less than .07). This differential treatment response of daily gain may be due in part to effects on feed intake (weaning age [lin.] x week postweaning [lin.] x treatment, P less than .10). We conclude that a single injection of HYD to 6-d-old piglets precociously induces pancreatic amylase and that the sensitivity of piglets to HYD is age-dependent.  相似文献

A total of 60 pigs from 15 litters were used during two experiments to assess the effects of maternal supplementation of Cr tripicolinate on performance and immune status of the offspring during the first 42 d after weaning. Gilts were raised on diets with either 0 (-Cr) or 200 (+Cr) ppb supplemental Cr from Cr tripicolinate. Their offspring were weaned at about 23 d of age. Pigs from dams fed supplemental Cr were fed Cr-supplemented diets after weaning and pigs from unsupplemented dams were not fed supplemental Cr. Pigs were housed in groups of three according to litter origin (six -Cr and nine +Cr) and BW. Feed and water were available for ad libitum consumption. Serum was collected 24 h after birth, the day after weaning (d 0), and, subsequently, every 7 d through d 28 for measurement of total immunoglobulin (Ig) G and IgM. For the 42-d nursery period, ADG (511 and 531 g/d for -Cr and +Cr, respectively), ADFI (827 and 851 g/d), and gain:feed (0.62 and 0.62) were not affected (P > 0.15) by Cr. Total IgG was not affected (P > 0.10) by Cr on d 0 (7.6 and 7.7 mg/mL for -Cr and +Cr, respectively) or on d 7 (6.3 and 6.1 mg/mL for -Cr and +Cr, respectively), when the lowest concentrations were observed. By d 28, total IgG had risen (11.7 and 8.9 mg/mL for -Cr and +Cr, respectively) and was affected by Cr (P = 0.03). Total IgM followed a similar pattern and was lowest on d 7. In each trial, IgG at 24 h after birth was inversely related to IgG from d 14 through 28 in the nursery. Additionally, the lowest Ig concentrations occurred between 4 and 5 wk of age, rather than at weaning (23 d of age); this may represent a vulnerable period for weanling pigs. Supplementation of the weanling pig diets with Cr tripicolinate did not significantly affect performance and immune status during the postweaning period.  相似文献

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.  相似文献

We conducted two experiments to evaluate the effect on growth from feeding 3,000 ppm Zn as ZnO during different weeks of the nursery period. In Exp. 1 (n = 138, 11.5 d of age, 3.8 kg BW) and Exp. 2 (n = 246, 24.5 d of age, 7.2 kg BW), pigs were fed either basal diets containing 100 ppm supplemental Zn (adequate) or the same diet with an additional 3,000 ppm Zn (high) supplied as ZnO. Pigs were fed four or two dietary phases in Exp. 1 and 2, respectively, that changed in dietary ingredients and nutrient content (lysine and crude protein) to meet the changing physiological needs of the pigs for the 28-d nursery period. Dietary Zn treatments were 1) adequate Zn fed wk 1 to 4, 2) high Zn fed wk 1, 3) high Zn fed wk 2, 4) high Zn fed wk 1 and 2, 5) high Zn fed wk 2 and 3, and 6) high Zn fed wk 1 to 4. In Exp. 1 and 2, pigs fed high Zn for wk 1 and 2 or the entire 28-d nursery period had the greatest (P < .05) ADG. During any week, pigs fed high Zn had greater concentrations of hepatic metallothionein and Zn in plasma, liver, and kidney than those pigs fed adequate Zn (P < .05). In summary, both early- and traditionally weaned pigs need to be fed pharmacological concentrations of Zn provided as ZnO for a minimum of 2 wk immediately after weaning to enhance growth.  相似文献

Syndyphalin-33 (SD-33) increases feed intake in sheep and recently weaned pigs. To assess the effects of SD-33 on hypothalamic gene expression, hypothalami were collected from unweaned pigs (n=19; 21±3 d of age) on day 0. Remaining pigs received an intramuscular injection of 0.5 μmole/kg SD-33 (SD) or saline (VEH) and weaned into individual pens. On days 1, 4, and 7 after weaning, hypothalami were collected from subsets of pigs (n=8 or 9) within each treatment group. Expression of μ-opioid receptor (MOR) was less in SD pigs than in VEH pigs on day 1 and day 4, suggesting down-regulation of the receptor by SD-33. Expression of hypothalamic melanocortin 4 receptor (MC4R) at 1 d after weaning was increased in VEH pigs (but not SD pigs) relative to levels before weaning. Expression of AGRP was not significantly altered by weaning or treatment at 1 d after weaning. At 4 d after weaning, expression of AGRP was greater in SD pigs than in VEH pigs, but at day 7 expression was less in SD pigs than in VEH pigs. A strong positive correlation was noted between expression levels of MOR and MC4R across treatment and time. Treatment with SD-33 appeared to partially abrogate the effects of weaning on expression of two key appetite-regulating genes within 24 h. Effects of SD-33 appear to be mediated at least in part by the μ-opioid receptor and include actions on the melanocortinergic pathway.  相似文献

Thirty-six 7-mo-old gilts were used to study the effects of dietary vitamin E and fat source (5% sunflower oil or animal fat) in pregnant and lactating sow diets on serum vitamin E concentration and on cell-mediated and humoral immune response in suckling and weaned piglets. Six gilts each received one of six diets throughout pregnancy and lactation. The basal diets (13 mg alpha-tocopherol/kg diet) were supplemented with dl-alpha-tocopheryl acetate to 48 and 136 mg alpha-tocopherol/kg of feed (average analyzed values). After weaning (at 4 wk of age) all pigs received identical diets (20 mg of alpha-tocopherol/kg feed). One week after weaning, pigs were immunized (i.m. with ovalbumin and tetanus toxoid) and antibody production was measured. Blood samples were taken immediately after birth, at 1 wk after birth, at weaning, and at four weekly intervals after weaning. Samples were analyzed for alpha-tocopherol concentration, total number of leukocytes, T- and B-lymphocytes, lymphocyte stimulation with concanavalin A, lysozyme activity, and immunoglobulin concentrations. It was concluded that a high vitamin E level in the sow's diet increased serum vitamin E concentration of 1-wk-old pigs (P less than .05). Immune response against ovalbumin was increased (P less than .05) at 1 wk of age after immunization for weaned pigs from sows fed the high level of vitamin E. Also, the phagocytic measures of pigs at 1 wk of age were increased by the medium vitamin E level (P less than .05). Fat sources in the sow's diet had no consistent effect on the immunological measures of pigs.  相似文献

It was hypothesized in these studies that differences would exist in baseline immune and performance measures among different breeds of pigs, and that these differences would be affected by age of the pig. Baseline immune, plasma cortisol (CORT) concentrations, and performance measures were determined among Berkshire (n = 36), Duroc (n = 18), Meishan (n = 54), Landrace x Yorkshire (White X; n = 36), and Yorkshire (n = 36) pigs at 4, 8, and 12 wk of age. All piglets were weaned at 17 to 21 d of age and moved to a common nursery environment. Total white blood cell (WBC), leukocyte differential, plasma CORT, immunoglobulin G (IgG) concentrations, natural killer cytotoxicity, neutrophil phagocytosis (PHAGO), and chemotaxis (CHTX) were evaluated. At all ages, plasma CORT was greatest in Meishan pigs, and least in Yorkshires (P < 0.05). Plasma IgG increased with age for all breeds (age: P < 0.01; breed x age: P < 0.005), except that in Meishans, IgG decreased. Natural killer cytotoxicity was greatest (P < 0.05) among Meishan pigs. There were breed x age interactions for neutrophil PHAGO (P < 0.001) and CHTX (P < 0.001). Overall, Yorkshire pigs showed the greatest (P < 0.05) percentage of PHAGO but the least (P < 0.05) CHTX. White X pigs had the greatest (P < 0.05) CHTX response. Berkshire pigs had the greatest (P < 0.001) numbers of neutrophils. At 12 wk of age, Meishan pigs had the least BW gain (P < 0.001), and Durocs had the greatest G:F (P < 0.001). There were no significant sex differences for immune (P > or = 0.15), performance (P > or = 0.20), or CORT (P = 0.70) measures. Pig breed and age influenced both baseline immune measures and plasma CORT in growing pigs, suggesting that pig breed and age are important factors influencing the response to various stressors or infectious challenges.  相似文献

Weaning in young animals is associated with an increased incidence of gastrointestinal infections. β-glucans exert numerous physiological effects, including altering immune function. The objective of this study was to determine the effects of feeding barley (Hordeum vulgare L.)-derived β-glucans on immune and intestinal function in weanling pigs (Sus scrofa). Thirty-one individually-housed Dutch Landrace pigs (21 d; initial BW, 6,298 ± 755 g) were weaned and fed a wheat-based diet (control) or a low (Lo-BG), medium (Med-BG), or high β-glucan-containing barley-based diet (Hi-BG) for 2 wk with 7 or 8 pigs/treatment. Intestinal segments were analyzed for permeability using Ussing chambers and K88 Escherichia coli adhesion to enterocytes was assessed ex vivo. Immune cells from mesenteric lymph nodes, peripheral blood, and Peyer's patches were analyzed for lymphocyte subsets by indirect immunofluorescence and the ability to respond ex vivo to mitogens by (3)H-thymidine incorporation. Hematology and neutrophil function were determined by flow cytometry. Neutrophil burst, size, and granularity, lymphocyte proliferation, and B-cell distribution in peripheral blood lymphocytes, Peyer's patches, and mesenteric lymph nodes were not affected by β-glucans content of the diet. The β-glucans content of the diet altered blood concentrations of erythrocytes and leukocytes, CD4, CD45RA, and CD8 blood cells (P < 0.05). In addition, feeding β-glucan resulted in increased (P < 0.05) percentage CD45RA positive cells in peripheral blood lymphocytes, Peyer's patches, and mesenteric lymph nodes. Mannitol permeability and tissue conductance were increased (P < 0.05) in Hi-BG fed pigs compared with control pigs. Percentage maximum K88-E.coli binding was increased in proportion to the β-glucan content of the diet (P < 0.05). Although β-glucan feeding during the weaning period increased blood lymphocytes and the proportion of na?ve T-cells, it also increased E. coli-enterocyte binding and intestinal permeability. β-glucan may alter immune and intestinal function of weaning pigs.  相似文献
