Infectious salmon anemia (ISA) has caused severe morbidity and mortality in farmed Atlantic salmon in North America, Norway, Scotland and the Faroe Islands. The Quoddy region of Maine, United States of America (USA), and New Brunswick (NB), Canada is characterized by extensive tidal mixing and close proximity between farms. This region is also prone to recurrent appearances of ISA, though control measures limit disease spread and severity on infected farms. We conducted a retrospective longitudinal analysis of the apparent impact of hydrographics on the incidence and timing of ISA outbreaks on Atlantic salmon (Salmo salar L.) farms in the Quoddy region from May 2002 to August 2004. A time-series cross-sectional regression of 32 farms over 28 months demonstrated a limited, but statistically significant, spatio-temporal clustering of ISA outbreaks linked hydrographically. New outbreaks correlated temporally with those occurring on-site 1 and 3 months prior, and those occurring within one tidal-excursion upstream the same month. Other risk factors included holdover of previous year-class fish, wharf sharing, and possibly harvests of cages infected in previous months. Conclusions suggest that tidal dispersion does play a role in ISAV transmission in the Quoddy region. Dispersal of free virus and/or tidal distribution of lice or other hydrographically influenced vectors or fomites could all contribute to the spatio-temporal patterns described.  相似文献

A case-control study is reported of 57 holdings (holdings in which salmon are raised in sea-water) with infectious salmon anaemia (ISA) and 61 holdings without ISA, together with a follow-up study of the control holdings, during 1988–1990. Generally the identified risk factors implied poor biosecurity since passive transmission (proximity to other ISA holdings) and active transmission (management practices which increased exposure to foreign biological material) were found to be major risk factors. The findings indicated that ISA was spread by infected live salmon or infected biological material i.e. animal waste or discharge from normal operations and slaughter. The findings also indicated that decreased host resistance could be important, but these results could be also be explained by a misclassification bias. Moreover, the findings indicate that a prompt eradication policy and improved biosecurity would be important means of controlling ISA.  相似文献

Abstract

Largemouth bass virus (LMBV; family Iridoviridae) has recently emerged as a causative agent in fish kills of largemouth bass Micropterus salmoides. Little is known about how the virus is transmitted or what factors predispose fish to mortality subsequent to infection. Concern has nevertheless arisen that activities related to recreational angling may affect transmission dynamics and may alter the susceptibility of infected fish to clinical disease. This study examined the separate effects of two angling-related factors on the susceptibility of juvenile largemouth bass to mortality from LMBV infection and on the transmission of LMBV from infected to uninfected fish. The first factor was hook-and-line angling. Infected fish that underwent a simulated angling treatment did not experience higher mortality or have higher viral loads in their tissues than those that were not angled. The second factor was direct contact between infected and uninfected fish, as would occur in live wells and holding tanks. The LMBV was transmitted from infected to uninfected fish through water, even when direct contact was prevented. Transmission of LMBV between infected and uninfected fish separated by a fenestrated barrier was nearly as efficient as LMBV transmission between infected and uninfected fish that were allowed direct contact. These results imply that angling itself may have only minimal effects on the survival of largemouth bass infected with LMBV but that angling-related practices that place infected and uninfected fish together in a limited water volume may facilitate viral transmission. Partitioning or cooling of live wells and holding tanks, as well as limiting their use, could reduce LMBV-associated mortality and viral transmission.  相似文献

Infectious salmon anemia (ISA) virus (ISAV), an economically important new pathogen in marine aquaculture, is classified in the family Orthomyxoviridae, genus Isavirus. The main structural properties of this genus include enveloped virions 90-140 nm in diameter with surface projections of a combined receptor-binding hemagglutinin and receptor-destroying enzyme activity demonstrated to be an esterase, hence recently designated HE, and a genome composed of eight segments of linear, single-stranded, negative sense RNA ranging in length from 1.0 to 2.4 kb, with a total size of approximately 14.3 kb. The viral genome encodes at least ten proteins, of which nine are structural and one is non-structural. Examination of more than 160 ISAV isolates has led to the identification of two hemagglutinin subtypes of ISAV, one North American and one European. The immune response against ISAV after infection or vaccination does not provide full protection against the infection. The recent discovery of antibody-mediated uptake and replication of ISAV in macrophage-like fish cell lines suggests that Fc receptor-mediated antibody-dependent enhancement of the ISA virus infection might also occur in vivo, as the virus in Atlantic salmon (Salmo salar) targets endothelial cells lining blood vessels and macrophage-like cells. Cumulative mortalities in Atlantic salmon during natural ISA outbreaks and experimental infections range from 0 to 100%. ISAV causes fatal systemic infections in marine-farmed Atlantic salmon and asymptomatic infections in feral fish. Experimentally induced fatal clinical disease in rainbow trout (Oncorhynchus mykiss) has identified a correlate of virulence of ISAV that may explain its emergence as a fish pathogen.  相似文献

A stochastic model describing disease transmission dynamics for a microparasitic infection in a structured domestic animal population is developed and applied to hypothetical epidemics on a pig farm. Rational decision making regarding appropriate control strategies for infectious diseases in domestic livestock requires an understanding of the disease dynamics and risk profiles for different groups of animals. This is best achieved by means of stochastic epidemic models. Methodologies are presented for 1) estimating the probability of an epidemic, given the presence of an infected animal, whether this epidemic is major (requires intervention) or minor (dies out without intervention), and how the location of the infected animal on the farm influences the epidemic probabilities; 2) estimating the basic reproductive ratio, R0 (i.e., the expected number of secondary cases on the introduction of a single infected animal) and the variability of the estimate of this parameter; and 3) estimating the total proportion of animals infected during an epidemic and the total proportion infected at any point in time. The model can be used for assessing impact of altering farm structure on disease dynamics, as well as disease control strategies, including altering farm structure, vaccination, culling, and genetic selection.  相似文献

The purpose of this investigation was to identify factors associated with the clinical diagnosis of foot and mouth disease during the 2001 epidemic in the United Kingdom. Using logistic regression, we compared: (1) reports of suspect disease that resulted in the declaration of FMD to reports that did not, and (2) laboratory-positive cases to laboratory-negative cases. From 6,801 reports of suspect disease, 2,026 cases of FMD were identified. Suspect cases were more likely to become clinical cases if: (1) the report originated from the disease control authorities ('active surveillance') rather than the public, usually farmers ('passive surveillance'); (2) cattle were the species suspected of disease, as opposed to sheep; (3) the report was filed during the peak of the epidemic; (4) the reporting premises was within 3 km of an FMD case detected within the previous 2 weeks; or (5) the report originated from certain local disease control centres. There were significant two-way interactions between: type of surveillance and species suspected of disease, type of surveillance and proximity of other infected premises, species suspected and time in the epidemic, and time in the epidemic and proximity of other infected premises. Clinical cases were more likely to be laboratory positive if: (1) they were found by passive versus active surveillance, (2) cattle were suspected of disease (versus sheep), (3) oldest lesions were less than 3 days, (4) the report was filed at any time other than the peak of the epidemic, or (5) the report originated from certain local disease control centres. Significant two-way interactions were found between: type of surveillance and species suspected of disease, and type of surveillance and time in the epidemic.  相似文献

In the course of the 1997-1998 CSF epidemic in the Netherlands, two semen collection centres (SCC) became infected. As an eradication strategy for an acute crisis situation, it was concluded that all semen of the boars at the SCCs collected and distributed in the risk period of 28 January to 7 March 1997 was potentially contaminated (suspect semen). As a consequence, a total of 1,680 pig herds, mainly located in the southern part of the Netherlands, were officially declared CSF suspect. The purpose of this study was to investigate whether infection of farms through contaminated semen played a significant role in the CSF epidemic. A total of 123 CSFV infected herds were identified, that had received suspect semen from one or both of the infected SCCs. In 87 out of these 123 infected herds, infection by way of artificial insemination (AI) could be excluded either according to the insemination information or the infection pattern observed. In only 21 herds, infection by way of AI was regarded as possible according to the insemination information and infection pattern. Owing to missing information, no conclusion could be drawn about the possibility of infection of 15 farms by way of AI. Thus, we conclude that at most 36 farms may have been infected through AI during the CSF epidemic in the Netherlands.  相似文献

A comparison was made of the susceptibility of buffaloes, cattle and goats to infection with Trypanosoma vivax transmitted either by Glossina morsitans centralis or by syringe inoculation. Three different isolates of T vivax (two from East Africa, one from West Africa) were used to compare skin reactions, parasitaemia, anaemia and the development of trypanosome-specific antibodies in buffaloes, cattle and goats. African buffaloes reared in captivity in an area free from trypanosomiasis proved to be highly resistant to infection with the three stocks of T vivax tested, irrespective of whether infection was by tsetse transmitted metacyclic forms or by intradermal or intravenous inoculation of bloodstream forms of the parasite. The bites of 19 tsetse infected with a West African T vivax stock did not cause local skin reactions, detectable bloodstream infections or antibody responses in two buffaloes. Following the bites of 120 tsetse flies infected with the same stock, two different buffaloes showed no local skin reactions, but had detectable bloodstream infections without showing signs of anaemia. Cattle and goats infected in a similar way showed severe local inflammatory skin reactions, high levels of parasitaemia and severe anaemia. The two East African stocks of T vivax caused no local skin reactions and only a transient parasitaemia in buffaloes following tsetse-transmitted infection or intradermal inoculation of bloodstream forms. Despite the ubiquitous presence of atypical mycobacteria in the environment and the potential risk of infection in humans and animals, the pathogenesis of diseases caused by infection with atypical mycobacteria has been poorly characterized. In this study, goldfish, Carassius auratus were infected either with the rapidly growing fish pathogen, Mycobacterium fortuitum or with another rapidly growing mycobacteria, Mycobacterium smegmatis. Bacterial persistence and pathological host response to mycobacterial infection in the goldfish are described. Mycobacteria were recovered from a high percentage of inoculated fish that developed a characteristic chronic granulomatous response similar to that associated with natural mycobacterial infection. Both M. fortuitum and M. smegmatis were pathogenic to fish. Fish infected with M. smegmatis ATCC 19420 showed the highest level of giant cell recruitment compared to fish inoculated with M. smegmatis mc(2)155 and M. fortuitum. Of the three strains of mycobacteria examined, M. smegmatis ATCC 19420 was the most virulent strain to goldfish followed by M. fortuitum and M. smegmatis mc(2)155, respectively.  相似文献

Highly pathogenic avian influenza (HPAI) virus subtype H5N1 threatens poultry production and human health. Understanding the role that migratory waterfowl play in introducing and maintaining this infection is critical to control the outbreaks. A study was conducted to determine if the occurrence of HPAI subtype H5N1 outbreaks in village poultry in Romania, 2005-2006, was associated with proximity to populations of migratory waterfowl. Reported outbreaks--which could be grouped into three epidemic phases--and migratory waterfowl sites were mapped. The migratory waterfowl site closest to each outbreak was identified. The distances between outbreaks occurring in phase 1 and 2 of the epidemic and the closest migratory waterfowl site were significantly (P<0.001) less than in phase 3, but these distances were only useful in predicting when outbreaks occurred during phase 1 (October-December, 2005) of the epidemic. A spatial lag (rho=0.408, P=0.041) model best fit the data, using distance and [distance]*[distance] as predictors (R2=0.425). The correlation between when outbreaks were predicted to occur and when they were observed to occur was 0.55 (P=0.006). Results support the hypothesis that HPAI virus subtype H5N1 infections of village poultry in Romania during the autumn of 2005 might have occurred via exposure to migratory populations of waterfowl.  相似文献

We summarised the challenges faced in an ex ante cost-benefit appraisal of United Kingdom government spending on disease surveillance for three notifiable fish diseases: infectious salmon anaemia (ISA), viral haemorrhagic septicaemia (VHS) and infectious haemorrhagic necrosis (IHN). We used a social cost-benefit analysis and adopted a national perspective. We compared costs of current public and private surveillance effort with the benefits stated in terms of the avoided private and social costs of potential disease outbreaks. Spending on ISA and VHS were predicted to be efficient; the benefit-cost ratios were always >or=3.2 for ISA and >or=5.8 for VHS for all nine scenarios examined for each infection. However, the benefit-cost ratio for IHN was predicted never to exceed 1.6, and was <1.0 in five of the nine scenarios-so spending on IHN would be harder to justify.  相似文献

To provide a basis for effective foot-and-mouth disease (FMD) prevention measures, factors associated with local spread were investigated in this study using data of the 2010 FMD epidemic in Japan. Thirty-eight local clusters within a 500-m radius from source farms were selected. In the clusters with pig source farms, more neighboring farms were infected in a short time compared with the clusters with cattle source farms. The influence of distance and wind upon local spread did not show a significant difference between infected and noninfected neighboring farms. Large-size pig farms posed a greater risk of inducing local spread; the odds ratio with reference to small-size cattle farms was 16.73. Middle-size and large-size cattle farms had a greater risk of infection; odds ratios with reference to small-size cattle farms were 15.65 and 25.52, respectively. The present results are useful for understanding features of local spread and prioritizing farms for control measures.  相似文献

Abstract

Laboratory exposures to the infectious stages (triactinomyxons) of Myxobolus cerebralis demonstrated a range of susceptibility to whirling disease among four species of inland salmonids. Replicate groups of each species were exposed to two concentrations of triactinomyxons, a low dose (100–200 per fish) and a high dose (1,000–2,000 per fish). Exposed fish were evaluated for clinical signs, for severity of microscopic lesions at 35 d, 2 and 5 months, and for spore concentrations in the head cartilage at 5 months. A standard strain of rainbow trout Oncorhynchus mykiss matched for age served as a susceptible species control. Rainbow trout, westslope cutthroat trout O. clarki lewisi, Yellowstone cutthroat trout O. clarki bouvieri, and bull trout Salvelinus confluentus were susceptible to M. cerebralis infections. Clinical signs, including radical swimming (“whirling”) and black tails, were observed at 7 weeks postexposure among rainbow and cutthroat trout challenged at 3 weeks of age. Clinical signs were rare among bull trout exposed at an age of 4 weeks and absent among rainbow and cutthroat trout exposed at 3 months posthatch. Most rainbow, cutthroat, and bull trout were found to be infected when examined at 5 months postexposure. The most severe microscopic lesions among infected fish at 5 months postexposure were found among rainbow trout. Cutthroat trout had less severe lesions, bull trout had mild infections, and no evidence of infection was found among Arctic grayling Thymallus arcticus. Mean spore concentrations among infected fish correlated with the severity of microscopic lesion scores. Rainbow trout had mean concentrations of spores in head cartilage reaching 106, whereas more resistant species such as bull trout had 104 spores; no spores were found among Arctic grayling at 5 months postexposure.  相似文献

Abstract

The intensity and prevalence of whirling disease was tested by exposure of 2-monthold fry and 1-, 2-, and 3.5-year-old adults of rainbow trout Oncorhynchus mykiss to a known number of laboratory-produced Myxobolus cerebralis at the actinosporean triactinomyxon stage. Fry exposed to graded concentrations of infectivity (triactinomyxons) for 3 h were individually examined for spores of Myxobolus cerebralis 5 and 6 months later. Exposure of fish to the lowest doses, 1 and 10 triactinomyxons per fish, did not result in detectable myxosporean spores. Fish that became lightly infected by a dose of 100 triactinomyxons per fish experienced a decrease in the incidence of infection between 5 and 6 months after exposure. A linear relationship was found between the numbers of recovered myxosporean spores and doses of 100–10,000 triactinomyxons per fish, and the spore burden appeared to plateau at doses of 10,000–100,000 triactinomyxons per fish. Adult fish continuously exposed to the highest dose of triactinomyxons for 3.5 months were infected and asymptomatic, however, the severity of myxosporean infection decreased with increased age of fish. This information may help in controlling whirling disease in salmonids.  相似文献

Infectious salmon anemia (ISA) is a viral disease occurring in farmed Atlantic salmon (Salmo salar) that is characterized by lethargy, anorexia, anemia and death. To control the disease in New Brunswick, Canada, 7.5 million fish from outbreak cages have been destroyed since 1997. Despite changes made by farmers, 2002 was the worst year ever for ISA losses in the region.

We evaluated the associations between potential risk factors and ISA outbreaks in the Atlantic-salmon sites in New Brunswick. This was a multilevel study in which the site-level design was a retrospective cohort study while the cage-level design was a modified case-cohort study. The questionnaire was divided into site-level questions, cage-level questions and hatchery information.

The important factors identified by this study can be categorized as environmental, farmer controlled or industry controlled according to the capacity to change or eliminate them. Environmental risk factors such as increasing the depth of the net (if nets were ≤9 m, odds ratio (OR) = 3.34) and decreasing the depth of water underneath the net (if depth of water underneath the net >3 m, OR = 3.34) are for the most part dictated by site location. Wild pollock (Pollachius virens) in the cage reflects the number of wild pollock that live in the site location. If there were ≥1000 pollock in the cage, the odds of disease in the cage increased 4.43-fold. Risk factors that are under farm control include increasing the number of times that the salmon are treated for sea lice (OR = 3.31 if lice treatments are ≤2 times), transferring small smolts into seawater (OR = 2.40 if smolts weighed >99 g) and improving on the adaptation of smolts to seawater to reduce post-transfer mortalities (OR = 4.52 if there was at least one cage with post-transfer mortalities >5%). The industry-controlled factors need to be addressed by the industry as a whole. Organizing boat travel to minimize the time and frequency of boats travelling to or by sites currently is being reviewed. This will be extremely important because the OR = 9.43 if processing boats travel within 1 km of the site and the OR = 4.03 if the site has dry feed delivered by the feed company. Because the hazard ratio increased stepwise from 1 if the nearest neighbor with ISA was ≥5 km up to 5.5 if the nearest site with ISA was within 0.5 km, increasing the distance between sites might be necessary for effective control.  相似文献

This study investigated protection against Eimeria acervulina (E. acervulina) following vaccination of chickens with an Eimeria recombinant profilin in conjunction with different adjuvants, or by changing the route of administration of the adjuvants. Day-old broilers were immunized twice with profilin emulsified in Montanide IMS 1313 N VG PR adjuvant (oral, nasal, or ocular routes), Montanide ISA 71 VG adjuvant (subcutaneous route), or Freund's adjuvant (subcutaneous route) and orally challenged with virulent E. acervulina parasites. Birds orally immunized with profilin plus IMS 1313 N VG PR, or subcutaneously immunized with profilin plus ISA 71 VG, had increased body weight gains compared with animals nasally or ocularly immunized with profilin plus IMS 1313 N VG PR, or subcutaneously immunized with profilin plus Freund's adjuvant. All adjuvant formulations, except for IMS 1313 N VG PR given by the nasal or ocular routes, decreased fecal parasite excretion and/or reduced intestinal lesions, compared with non-vaccinated and infected controls. Compared with animals vaccinated with profilin plus Freund's adjuvant, chickens immunized with profilin plus IMS 1313 N VG PR or ISA 71 VG showed higher post-infection intestinal levels of profilin-reactive IgY and secretary IgA antibodies. Finally, immunization with profilin in combination with ISA 71 VG was consistently better than profilin plus IMS 1313 N VG PR or Freund's adjuvant for increasing the percentages of CD4(+), CD8(+), BU1(+), TCR1(+), and TCR2(+) intestinal lymphocytes. These results indicate that experimental immunization of chickens with the recombinant profilin subunit vaccine in conjunction with IMS 1313 or ISA 71 VG adjuvants increases protective mucosal immunity against E. acervulina infection.  相似文献

This review investigates how recreational fishing affects the physiology, behaviour, and welfare of fish. Sentience and the capacity of fish to experience pain, suffering and fear are discussed, and practical recommendations for improving the treatment of fish during recreational fishing are provided. Handling procedures used in recreational fishing should match the environment where the fish is caught and the size and strength of the fish. Minimising the number of hooks on lures and baits, and using barbless hooks and circle hooks generally reduce rates of injury and the severity of tissue trauma. Capture time, handling time, and exposure to air play significant roles in the stress responses of fish and should be minimised by anglers. Keep-nets, gaffs, landing-nets, live-wells and other restraining devices should only be used when necessary because each device can prolong or intensify the negative influences of catching fish by hook-and-line. The use of fish as bait is discouraged unless they have been euthanised. Euthanasia of fish used as bait or fish captured recreationally should include a stunning blow to the head, followed by bleeding-out, or pithing the brain and spinal cord. Fish should be euthanised if they are bleeding, injured, deeply-hooked, foul-hooked, or severely exhausted, but local fisheries' regulations must be obeyed. Fish that are released after being caught may be subjected to additional factors that influence their welfare, such as elevated stress, barotrauma, suppressed feeding and growth, impaired reproductive function, increased potential for disease, infection, and delayed mortality.  相似文献
