The lipid-binding properties of wheat puroindolines (PINs) make these proteins likely candidates to play an important role in the stability of lipid films in the gas cells of bread dough, an important aspect of bread making. Therefore, the interaction between PINs and water-soluble model lipids was investigated by fluorescence emission and circular dichroism (CD) spectroscopy. The secondary structure of PIN-a and PIN-b in solution, as determined by far-UV CD measurements, resembled that of plant non-specific lipid transfer protein (LTP). However, PINs contain a tryptophan-rich loop located at the exterior of the protein. It was shown by fluorescence emission spectroscopy and near-UV CD spectroscopy that this domain is involved in the lipid binding. Fluorescence titration experiments of PIN and its synthetic tryptophan-rich domains with the zwitterionic lipidn-hexadecylphosphocholine (C16PN) revealed that the binding was cooperative. For the binding of the charged lipids,n-hexadecacylphosphoglycol (C16PG) and hexadecyl-trimethylammoniumbromide (CTAB), a clear effect of ionic strength of the solution was obtained. Organisation of lipids into micelles was not a prerequisite for binding to PINs. Most likely, wheat PIN binds to monomeric lipid molecules via its tryptophan-rich domain, rendering the protein more hydrophobic. The overall secondary structure of wheat PINs did not change significantly upon binding to lipids.  相似文献

This study focuses on the effect of Aegilops longissima on wheat bread making quality. Chromosome 1Sl disomic addition line of Ae. longissima (DAL1Sl) had significantly higher dough strength, grain hardness, mixographic peak height, band width, and unextractable polymeric protein content compared with wheat. DAL1Sl also had additional glutenin and gliadin proteins contributed by Ae. longissima. The larger size of 1Sl coded HMW-GSs sequenced from DAL1Sl and their phylogenetic similarity to the D-genome-coded subunits were suspected to be one of the major reasons for the increased dough strength of DAL1Sl. To transfer the chromosome 1Sl genes responsible for the good bread-making quality to wheat, we generated a chromosome-specific disomic substitution line [DSL1Sl(1A)] by crossing DAL1Sl with nulli 1A tetra 1B genetic stock and further selection. Grain quality analysis revealed significantly lower grain hardness and significantly higher dough strength, farinograph development time, stability time, gluten index, bread loaf volume, and bread quality score in DSL1Sl(1A), compared with wheat. However, the increased bread loaf volume and quality were not proportional to the relatively higher increases in dough strength and gluten index, indicating importance of other traits influencing bread making quality. The presence of a minor hardness locus on chromosome 1A is speculated.  相似文献

1999年将 38份春小麦品种 (品系 )种植于内蒙古呼和浩特 ,对其磨粉品质和面包烘烤品质进行了评价。结果表明 ,我国春小麦品种的面包烘烤品质较差 ,不同地区品种间品质差异较大 ,辽宁和内蒙品种的磨粉品质和面包烘烤品质优于其它地区的品种。回归分析表明 ,蛋白质含量和单位蛋白质含量的面包体积决定了面包体积总变异的 99.9% ,硬度、沉淀值和吸水率对面包总分有重要作用。品种的出粉率主要取决于 1心槽路出粉率 ,2心和 1皮槽路出粉率对出粉率贡献也较大。高分子量麦谷蛋白亚基 (HMW GS)Glu A1和Glu D1位点的等位变异与品质性状密切相关。面包烘烤品质的改良应在分析HMW GS的基础上 ,将硬度作为选择指标之一 ,适当提高蛋白质含量 ,重点加强对沉淀值的选择  相似文献

为探讨Puroindolines蛋白的表达特点与籽粒硬度的关系,采用改进的SDS-PAGE凝胶分析了不同硬度小麦品种的籽粒在各个发育时期Puroindolines蛋白的表达.结果表明,不同硬度的小麦籽粒中总Puroindolines(PinA和PinB)蛋白的表达量差异不大,但与胚乳淀粉颗粒结合的Puroindolines蛋白量差异非常明显:在籽粒发育的不同时期,软质小麦籽粒淀粉粒表面结合的Puroindolines蛋白量显著高于硬质小麦;基因型同为野生型但硬度有差异的品种,籽粒较软的材料其淀粉粒表面结合的Puroindolines蛋白量也明显高于较硬的材料,说明该蛋白的结合特性是决定籽粒硬度的直接原因.结果还表明,胚乳中水溶性戊聚糖与籽粒硬度关系密切.  相似文献

Puroindolines are two small, basic cysteine-rich proteins isolated from Triticum aestivum seeds and characterized by a tryptophan-rich domain. They form the molecular basis of wheat grain hardness and display antimicrobial activity that may contribute to plant defence. Their antimicrobial activity is presumed to be due to their hydrophobic tryptophan-rich domain. However, little is known about their mode of action and there is no in vivo evidence that the binding of puroindolines to membranes is mediated by their tryptophan-rich domain. In this study, using a yeast complementation assay, we showed that puroindolines interact with the Saccharomyces cerevisiae plasma membrane. By site-directed mutagenesis of their tryptophan-rich domain, we determined that two tryptophan residues (W41 and W44) are mandatory for interaction of puroindoline-a with the yeast membrane whereas interaction of puroindoline-b depends on lysine residues. These results highlight that other residues than tryptophan play a critical role in the interaction of puroindolines with membranes, and probably their affinity for lipids and antimicrobial activities.  相似文献

Despite the great variety of physicochemical and rheological tests available for measuring wheat flour, dough and gluten quality, the US wheat marketing system still relies primarily on wheat kernel hardness and growing season to categorize cultivars. To better understand and differentiate wheat cultivars of the same class, the tensile strength, and stress relaxation behavior of gluten from 15 wheat cultivars was measured and compared to other available physicochemical parameters, including but not limited to protein content, glutenin macropolymer content (GMP) and bread loaf volume. In addition, a novel gluten compression–relaxation (Gluten CORE) instrument was used to measure the degree of elastic recovery of gluten for 15 common US wheat cultivars. Gluten strength ranged from 0.04 to 0.43 N at 500% extension, while the degree of recovery ranged from 5 to 78%. Measuring gluten strength clearly differentiated cultivars within a wheat class; nonetheless it was not a good predictor of baking quality on its own in terms of bread volume. Gluten strength was highly correlated with mixograph mixing times (r = 0.879) and degree of recovery (r = 0.855), suggesting that dough development time was influenced by gluten strength and that the CORE instrument was a suitable alternative to tensile testing, since it is less time intensive and less laborious to use.  相似文献

Puroindoline genotypes (Pina and Pinb) and their encoded proteins related to grain hardness were studied in various common wheat cultivars from Australia, China, Japan, Korea and North America. Most of the hard wheats had the Pinb-D1b genotype with a glycine to serine mutation at position 46. In addition to the known Pina and Pinb genotypes, cultivars were found with Pina and Pinb double-null mutations (Pina-D1b/Pinb-D1h (t)) and a new Pinb frameshift mutation (designated Pinb-D1i (t)) within the region encoding a tryptophan-rich domain. This new Pinb frameshift mutation was found only in Chinese cultivars. Endosperm proteins encoded by Pina and Pinb in these cultivars were analysed by 2D-gel electrophoresis (IPG×SDS-PAGE). Cultivars with Pina and Pinb double-null mutations showed no PIN-a or PIN-b protein, and cultivars with Pinb-D1i (t) had no PIN-b protein. Surprisingly, cultivars with Pinb-D1b had severely reduced amounts of PIN-b and cultivars with Pinb-D1c showed no PIN-b proteins. Grain hardness among cultivars having mutated Pinb may be explained by the amount of PIN-b protein and not by the type of amino acid substitutions.  相似文献

End-use quality of bread wheat (Triticum aestivum L.) is affected by both genetic and environmental variation. Current understanding of the genetic control of wheat quality traits is mainly based on genetic experiments conducted using grain produced in favourable conditions. The objective of this research was to extend the genetic analysis of these traits by using grain produced in water-limited environments. Grain samples harvested from a mapping population grown in field experiments at two locations in Australia were used to assess characteristics of the grain, flour, dough and bread. Quantitative trait loci (QTLs) were mapped. The parents of the population, RAC875 and Kukri, differ at several loci that are known to affect grain quality or plant phenology. Of these, a high-molecular-weight glutenin locus (Glu-B1) affected dough properties, the puroindoline-encoding Ha locus affected grain hardness, flour and loaf properties and a photoperiod response locus (Ppd-D1) affected flour extraction and protein content. Similarly, several previously reported quantitative trait loci (not associated with specific genes) also had effects in the stress environments used here. In addition, novel loci were detected for bread wheat quality traits; their effects may be specific to materials grown in water-limited environments.  相似文献

The effects of particle size of granulars (semolina and flour combined), gluten strength, protein composition and fermentation time on the breadmaking performance were compared for eleven durum wheat genotypes of diverse strength from North America and Italy grown in the same environment. All genotypes were γ-gliadin 45 types (low-molecular weight glutenin subunit 2 patterns) associated with superior pasta-making quality. Three cultivars with high-molecular weight glutenin subunit 20 exhibited relatively weak gluten, confirming that this subunit is associated with weakness in durum wheat. Gluten strength as measured by a range of technological tests was directly and strongly related to the proportion of insoluble glutenin (IG) in granulars protein as determined by a spectrophotometric procedure. Reducing the particle size of granulars by gradual reduction shortened development time in both the farinograph and mixograph. Reducing granulars also increased starch damage and, accordingly, farinograph water absorption, but remix-to-peak baking absorption was unaffected due to increased fermentation loss for finer granulars. Neither loaf volume, nor remix-to-peak mixing time were affected by the particle size of the granulars indicating that regrinding is not an asset for baking provided there is adequate gassing power. Loaf volume was directly related to gluten strength and IG content, and inversely related to residue protein, a non-gluten containing fraction. When fermentation time was reduced from the standard 165 to 90 min and 15 min, all genotypes exhibited a progressive increase in loaf volume. Therefore, regardless of strength, short fermentation time is preferred when high volume durum wheat bread is desired. Some of the stronger durum genotypes exhibited remix-to-peak bread volume comparable to that expected of good quality bread wheat, indicating that there is potential to select for genotypes with improved baking quality in conventional breeding programs by screening for high content of insoluble glutenin.  相似文献

Thirteen wheat cultivars grown in six locations were compared for kernel weight, protein content and grain texture, as determined by the Single Kernel Characterization System (SKCS). Moreover, puroindolines a (Pin-A) and b (Pin-B) bound to starch were quantified by densitometric scanning of A-PAGE fractionations. All cultivars shared allele Pina-D1a coding for wild-type Pin-A, and differed from each other in allele composition at Pinb-D1 coding for Pin-B. Cultivars with Pinb-D1a exhibited soft grain and high amounts of Pin-A and Pin-B compared to cultivars with Pinb-D1b or Pinb-D1d. Significant genetic variation for grain hardness and Pin-A level was detected in soft cultivars. The ratio between Pin-A and Pin-B levels in soft cultivars was approximately 6:5, whereas it varied between 9:5 and 10:1 in hard cultivars. Protein content was significantly correlated with Pin-B content (r=0.34) and SKCS value (r=0.36) in soft wheats. Significant correlations (0.68 and 0.73 for soft and hard wheats, respectively) were observed between Pin-A and Pin-B levels. Grain hardness was not correlated with puroindoline levels and Pin-A/Pin-B ratio in both textural classes. By contrast, kernel weight was found to act as a major environmental factor affecting grain texture in both soft and hard wheats.  相似文献

Arabinoxylans (AXs) are the major dietary fiber (DF) component in wheat and their consumption has been associated with several health benefits. Genetic improvement of the AX in refined wheat flour could be a good solution to improve the DF daily consumption while maintaining the flour desirable quality. In this study, 193 common wheat lines were analyzed for their AX content in refined flour and end-use quality. Wide variation in both the total arabinoxylan (TOT-AX) (10.8–16.5 mg/g) and water-extractable arabinoxylan (WE-AX) (3.2–7.6 mg/g) was identified and, in both cases, the genotype had the greatest impact on the observed phenotypes. Variation in the endogenous AX fractions appeared to have a moderate effect on wheat quality. The WE-AX, specifically, were positively correlated with gluten strength (r = 0.11 to 0.32) and bread loaf volume (r = 0.16), whereas the TOT-AX were negatively correlated with dough extensibility (r = −0.11) and bread making quality (r = −0.11). Overall, results of this study show that the genetic improvement of grain AX is feasible and that the AXs present in refined flour do not dramatically alter wheat quality indicating that it is possible to select varieties with high AX endosperm content end desired end-use quality.  相似文献

为明确安徽大田生产环境下软质小麦籽粒和终端产品品质表现,评价优质软麦品种的加工适用性,本研究选取该区当前推广种植的24个软质小麦品种,对其籽粒和面粉的主要品质性状及其制品南方馒头和曲奇饼干的品质进行差异性、相关性分析,并以美国软白麦近五年的主要品质性状平均值为理想指标进行灰色关联度比较。结果表明,供试材料的硬度、面粉色泽b*、湿面筋含量、面团形成时间、稳定时间等籽粒品质性状变异系数较大,而容重、面粉L*值和吸水率变异系数较小。南方馒头品质性状中,白度差异最小,比容差异最大;曲奇饼干品质性状中,感官评分变异系数较大,饼干直径均值和变异系数都较小。蛋白质含量、湿面筋含量、稳定时间均符合弱筋标准(GB/T 17320-2013)的样品数为0。相关分析表明,容重、降落值、面粉L*、b*、白度与大部分性状间相关性不显著;籽粒硬度与水SRC和乳酸SRC均呈显著正相关,与湿面筋含量和面粉a*值均呈显著负相关。蛋白质含量与面粉a*值等7个指标均呈显著正相关,与面粉b  相似文献

Wheat flour replacement from 0 to 40% by single tef flours from three Ethiopian varieties DZ-01-99 (brown grain tef), DZ-Cr-37 (white grain tef) and DZ-Cr-387 (Quncho, white grain tef) yielded a technologically viable ciabatta type composite bread with acceptable sensory properties and enhanced nutritional value, as compared to 100% refined wheat flour. Incorporation of tef flour from 30% to 40% imparted discreet negative effects in terms of decreased loaf volume and crumb resilience, and increase of crumb hardness in brown tef blended breads. Increment of crumb hardness on aging was in general much lower in tef blended breads compared to wheat bread counterparts, revealing slower firming kinetics, especially for brown tef blended breads. Blended breads with 40% white tef exhibited similar extent and variable rate of retrogradation kinetics along storage, while brown tef-blended breads retrograded slower but in higher extent than control wheat flour breads. Breads that contains 40% tef grain flour were found to contain five folds (DZ-01-99, DZ-Cr-387) to 10 folds (DZ-Cr-37) Fe, three folds Mn, twice Cu, Zn and Mg, and 1.5 times Ca, K, and P contents as compared to the contents found in 100% refined wheat grain flour breads. In addition, suitable dietary trends for lower rapidly digestible starch and starch digestion rate index were met from tef grain flour fortified breads.  相似文献

The influence of added phytate on dough properties and bread baking quality was studied to determine the role of phytate in the impaired functional properties of whole grain wheat flour for baking bread. Phytate addition to refined flour at a 1% level substantially increased mixograph mixing time, generally increased mixograph water absorption, and reduced the SDS-unextractable protein content of dough before and after fermentation as well as the loaf volume of bread. The added phytate also shifted unextractable glutenins toward a lower molecular weight form and increased the iron-chelating activity of dough. It appears that phytate negatively affects gluten development and loaf volume by chelating iron and/or binding glutenins, and consequently interfering with the oxidative cross-linking of glutenin molecules during dough mixing. Phytate could be at least partially responsible for the weak gluten network and decreased loaf volume of whole wheat flour bread as compared to refined flour bread.  相似文献

为探明协同提高强筋小麦产量、氮素利用率和品质的施氮量,以强筋小麦品种济麦20(中穗型)和洲元9369(大穗型)为材料, 研究了180、240和300 kg·hm-2三个氮肥水平(分别用N180、N240和N300表示)对强筋小麦产量、氮素利用率、品质及其相关指标的影响。结果表明,相同施氮量下,洲元9369的产量、氮素利用率、面团形成时间、面团稳定时间、面包体积和面包评分均高于济麦20。当施氮量由N180增至N240时,2个品种的产量无显著变化,但沉降值、面团形成时间、面团稳定时间、面包体积和面包评分均显著提高;施氮量增至N300后,2个品种的产量和品质又都显著下降,籽粒总蛋白含量、谷蛋白含量、SDS-不可溶性谷蛋白含量、醇溶蛋白含量和谷蛋白聚合指数均明显降低,而SDS-可溶性谷蛋白含量和谷醇比却表现为上升趋势。经相关分析,SDS-不可溶性谷蛋白含量、谷蛋白聚合指数与面团形成时间、面团稳定时间、面包体积和面包评分均呈显著正相关。以上结果表明,谷蛋白聚合程度降低是过量施氮条件下强筋小麦品质下降的主要原因。综合考虑小麦产量、氮素利用率和籽粒品质,240 kg·hm-2为本研究条件下的最佳施氮量。  相似文献

One accession of einkorn (Triticum monococcum) and five spring spelt accessions (T. aestivumsubsp.spelta) were selected and grown at five locations in 1992 and 1993 for evaluation of kernel, milling and baking properties. The high protein einkorn accession (TM23) was similar to common hard red spring (HRS) wheat in kernel size but the soft grain gave low flour yields. Einkorn flour had low sedimentation values, weak Mixograph curves and low loaf volumes with no bromate response. The two large-seeded spelt accessions, SK0021 and PGR8801, were comparable to common HRS wheat in kernel hardness, flour yield and protein content, but were very inferior in falling number, sedimentation value, Mixograph characteristics, loaf volume, crumb texture and colour. The spelt accessions, SK0505, SK0263 and RL5407, also exhibited large kernels but were soft in texture and gave high flour yields, intermediate sedimentation values, weak Mixograph characteristics, but satisfactory loaf volumes and bromate responses relative to HRS wheat. With addition of only 15 ppm bromate, the latter spelt accessions gave breads with loaf volumes similar to those of HRS wheat breads. In sensory tests, these spelt breads were intermediate in preference as compared to HRS wheat breads.  相似文献

The effects of grain texture on pastamaking and breadmaking quality were studied in three F8 soft-textured durum wheat lines (SDLs) containing wild-type alleles Pina-D1a and Pinb-D1a as compared with their hard durum sister lines (HDLs). SDLs homozygous for a small 5DS segment, less than 14.4 cM in size, accumulated puroindolines A (Pin-A) and B (Pin-B) and showed SKCS values (19.9-23.6) significantly lower than those (72.6-76.8) of their hard-textured counterparts lacking Pin-A and Pin-B. In addition, SDLs exhibited approximately 24% higher flour extraction rates compared with HDLs. Reducing the kernel hardness decreased farinograph water absorption, dough tenacity (P) and, accordingly, alveograph P/L ratio, but increased farinograph stability, mixing tolerance and dough extensibility (L). Spaghetti cooking quality, as determined by the sensory judgment of firmness, stickiness and bulkiness, was unaffected by the kernel hardness, whereas the loaf volume exhibited a 10% increase associated with kernel softening. Flour and semolina, but not spaghetti, from SDLs showed a substantial reduction in yellowness (b*) and brownness (100 − L*) likely due to their finer particle size compared with HDLs. Alleles Pina-D1a and Pinb-D1a may offer new perspectives for breeding dual purpose (pasta and bread) durum wheat varieties.  相似文献

Physico-chemical properties of bread baked by partially replacing normal wheat (Triticum aestivum L.) flour (15, 30, and 45%) with two hard waxy wheat flours were investigated. Substitution with waxy wheat flour resulted in higher loaf volume and softer loaves. However, substitution at >30% resulted in excessive post-bake shrinkage and a ‘key-hole’ shape with an open crumb structure. Bread crumb microstructure indicated a loss of starch granule rigidity and fusing of starch granules. The cells in the interior of the bread did not become gas-continuous and as a result, shrunk as the loaf cooled. Soluble starch content was significantly higher in bread crumb containing waxy wheat flour than in control bread. Debranching studies indicated that the soluble starch in bread made with 30-45% hard waxy wheat flour was mostly amylopectin. Incorporation of waxy wheat flour resulted in softer bread immediately after baking but did not retard staling upon storage.  相似文献

Two trials with a total of 75 spring bread wheat cultivars and advanced lines, were used to evaluate single kernel characterization system hardness, puroindoline alleles, milling yield, flour ash content, flour colour, and end-use qualities for Chinese noodles, steamed bread and pan bread. The results indicated that all International Maize and Wheat Improvement Center hard wheat lines surveyed were Pina-D1b genotype, whereas Pinb-D1b was the most common allele in the remaining cultivars. Genotypes with Pinb-D1b possess significantly lower flour ash content and higher milling yield than those of genotypes with Pina-D1b. For steamed bread, mean scores for loaf volume, crumb colour, width, structure and total score of Pinb-D1b genotypes were significantly higher than those of genotypes with Pina-D1b and wild type. For Chinese fresh white noodles, means for noodle a*, colour score, viscoelasticity and total score with Pinb-D1b were significantly higher than those of Pina-D1b and wild type. Means of loaf volume, texture and total score for Pinb-D1b genotypes were significantly higher than those of Pina-D1b genotypes. This study further illustrated the superiority of the Pinb-D1b allele on milling and processing qualities for various end products and provides useful information for wheat quality improvement.  相似文献

The substitution of wheat flour with barley flour (i.e. native or pretreated/extruded) reduced the loaf volume. Depending on the barley variety and flour pretreatments, the colour and firmness/texture of the bread loaves were altered. Amongst the barley breads prepared from native flours (at 15% barley flour substitution level), Phoenix had higher loaf volume and lower crumb firmness than Candle. However, amongst the barley breads prepared from extruded flours, CDC-Candle had higher loaf volume and lower crumb firmness than Phoenix. The lower loaf volume and firmer crumb texture of barley breads as compared with wheat bread may be attributed to gluten dilution. Also, the physicochemical properties of barley flour components, especially that of β-glucan, can affect bread volume and texture. β-glucan in barley flour, when added to wheat flour during bread making, could tightly bind to appreciable amounts of water in the dough, suppressing the availability of water for the development of the gluten network. An underdeveloped gluten network can lead to reduced loaf volume and increased bread firmness. Furthermore, in yeast leavened bread systems, in addition to CO2, steam is an important leavening agent. Due to its high affinity for water, β-glucan could suppress the amount of steam generated, resulting in reduced loaf volume and greater firmness. In the present study, breads made with 15% HTHM CDC-Candle flour had highly acceptable properties (loaf volume, firmness and colour) and it indicated that the use of extruded barley flours would be an effective way to increase the dietary fibre content of barley breads.  相似文献
