SUMMARY An investigation of sheep flocks with unknown histories of fleece derangement, selected at random from the main sheep raising areas of New South Wales, showed that Psorergates ovis was present in 53 of the 177 flocks surveyed and In 162 of the 3495 sheep examined. Of the single causes of fleece derangement, the most prevalent was grass seed contamination (15.7%) followed by Infestation with sheep body lice, Bovicola ovis, (14.7%) and Infestation with Itch mite (7.8%). The prevalence of fleece derangement was significantly higher on sheep In which lice were the only ectoparasites found than on those In which Itch mites were the only ectoparasites present. The greatest prevalence of P ovis was In flocks In the Young, Yass, Mudgee and Tenterfield districts. In most affected flocks, more than one potential cause of fleece derangement was present. Itch mite Infestation was more prevalent in flocks that received no treatment for ectoparasites than in those that had been treated in the year preceding the survey. Our results did not support a commonly held belief among farmers that arsenical compounds control Infestations of Itch mites more effectively than other types of pesticides. There was no significant difference in prevalence of fleece derangement between flocks treated with different pesticide groups and those that received no treatment, but mean fleece derangement score was significantly lower in sheep treated with organophosphates than in untreated sheep. Infestations of lice were significantly more prevalent in sheep from flocks treated with synthetic pyrethroids than from those treated with organophosphates or those that received no treatment.  相似文献

The sampling distribution of Bovicola ovis (Schrank) on sheep was examined in two flocks, one with a light and one with a heavy infestation of lice. The derived distributions were used to calculate the sensitivity of detecting lice on individual sheep and in flocks by fleece parting regimes that varied in number of parts per animal and number of sheep per flock, different scenarios of flock sizes, proportion infested and louse density were examined. Lice were aggregated among fleece partings in the heavily infested flock and described by a negative binomial distribution with k values between 0.3 and 1.92. The distribution was indistinguishable from Poisson in the lightly infested flock. The assumed distribution had little effect on sensitivity, except when only one fleece part per animal was examined. On individual sheep where louse density was 0.5 per 10 cm part or greater, there were only marginal gains from inspecting more than 10 parts per animal. Increasing the number of sheep inspected always increased sensitivity more than increasing number of parts per sheep by an equivalent amount. This advantage was greatest in situations where a low proportion of sheep in the flock were infested with a high density of lice, and less where a low proportion of sheep were infested with a low density of lice, or a high proportion of sheep were infested with a high density of lice.  相似文献

Knowledge of the presence or absence of lice in a flock of sheep enables wool growers to make informed decisions as to the need for insecticidal treatments. However, with inapparent infestations, traditional methods of detection are not sufficiently sensitive and, as a consequence, flocks may be left untreated. Conversely, the routine application of insecticide to sheep with no sign of infestation is an unnecessary cost. The sensitivity of 3 procedures for detecting lice was evaluated in 68 mobs of sheep from 50 farms. In 24 mobs of sheep known to be lightly infested, lice were detected in 17% by either parting the fleece of 10 sheep or by the lamp test in which 8 g samples of shorn wool from 30 randomly selected fleeces were placed under lamps for 10 min to repel the lice. Twenty of 23 mobs (87%) were found to be infested by the table locks test in which a 30 g sample of locks wool was dissolved in 10% sodium hydroxide and the filtered residue examined with x 40 magnification. A screening test, in which either fleeces on 5 sheep were examined by fleece parting or lice were repelled from 30 shorn fleeces for 5 minutes, detected about 60% of lightly infested mobs. When this was followed by the table locks test 91% of lightly infested mobs were detected. Conducting any one of the tests on more than one mob, and in large mobs testing more frequently, increases the sensitivity of detection of lice within the whole flock.  相似文献

Pruritic behaviour and deranged fleece are often used as indicators of sheep louse infestation but the exact relationship between infestation and the observation of signs of pruritis was unclear. Two studies were conducted to examine this association. In the first, 24 castrate Merino sheep were randomly assigned to six pens in groups of four and the sheep in three pens infested with 10 lice each on the right mid-side. Louse numbers were counted, fleece derangement scored and pruritic behaviour assessed periodically on each sheep until 38 weeks after infestation. In the second study a single moderately infested sheep was paddocked for 15 weeks with 32 uninfested sheep and louse numbers and fleece derangement monitored for 41 weeks. In the pen studies, differences between infested and non-infested sheep in fleece derangement and pruritic behaviour first became significant (p<0.05) at 8 and 14 weeks, respectively and at louse densities of 0.06 and 0.27 per 10 cm wool part. Some sheep showed definite signs of deranged fleece as early as 5 weeks after initial infestation. In the paddock studies, it took 37 weeks until lice were detected on all sheep in the flock. The correlation between louse numbers and fleece derangement score first became significant (r=0.44 and p<0.05) at 9 weeks after introduction of the lousy sheep, reached a maximum of r=0.79 (p<0.001) at 22 weeks when 84% of sheep had lice detected and the mean louse density was 0.29 per part, and then declined to r=0.12 (n.s.) at 41 weeks when all sheep were infested and the mean louse density was 3.04 per part. It is concluded that fleece derangement is a powerful early indicator of the presence of lice and that sheep may exhibit signs of pruritis well before lice can be readily found by direct inspection. Fleece derangement may be useful as a basis for establishing economic thresholds for the application of long wool treatments in developing louse infestations but appears to be a poor indicator of louse numbers once the infestation is advanced.  相似文献

Information provided by wool growers in Queensland, Australia between 1995 and 1997 was used to assess the prevalence and spatial distribution of louse (Bovicola ovis) infestation in sheep flocks. The estimated prevalence of louse-infested flocks was 40% (95% confidence interval, 35-46%). Although the prevalence of infestation was higher in western regions (41-50%) compared to the south region of Queensland (31%), the difference was not statistically significant (P > 0.05). Significant (P = 0.02) clustering of infested flocks was detected in the south region where two foci were apparent. We conclude that Queensland sheep flocks have a moderate prevalence of louse infestation, and that clustering of infestation is not strong. The control of lice is an industry-wide issue that needs to be addressed by most wool growers in Queensland.  相似文献

Oestrosis is a nasal myiasis of sheep and goats caused by larvae of the fly Oestrus ovis and can lead to severe clinical signs, which together with the disturbance caused by the adult fly may result into serious economic losses. Infection rates and larval burdens are always higher in sheep than in goats after either natural or artificial infestation. The aim of this study was to compare the host preference of the adult fly O. ovis between sheep and goats in mixed flocks, where they are kept together under the same husbandry conditions and hence, are very similarly exposed to the fly preference. Blood sera samples were collected from a total of 397 sheep and 335 goats, from 43 mixed flocks located at different regions of Greece. Antibodies specific to O. ovis IgG were measured by ELISA. A flock was considered positive when at least one individual was positive, i.e. showed a seropositivity of >or=20% in relation to positive control sera. A total of 193 (48.6%) sheep and 58 (17.9%) goats were found to be seropositive against O. ovis. Thirty-eight (88.4%) out of 43 flocks had at least one seropositive animal. The mean seroconversion against O. ovis in animals from the different flocks was 38.6% and 13.6% for sheep and goats, respectively, whereas the variance of infection within each flock was 0-100%. The mean seropositivity between sheep that were found to be positive or negative was 60.6% and 5.4%, respectively, whereas the corresponding values between goats were 35.2% and 5.2%, respectively. No significant difference in the seroconversion values was noted between flocks from the different areas (P=0.817), whereas a very significant difference was observed between animal species (P=0.001). However, there was no significant difference when seroconversion comparisons were made within samples of the same animals species, sheep or goats from different flocks of all the regions included in the study (P=0.695). The results of this study clearly demonstrate that O. ovis has a widespread distribution in Greece, and the seroprevalence is significantly higher in sheep than goats (P=0.001).  相似文献

An enzyme-linked immunosorbent assay (ELISA) was developed for the detection of specific antibodies against crude Psoroptes antigen. The diagnostic sensitivity was 93.7% in 191 sheep with clinical signs associated with mange. These animals originated from 29 flocks in which psoroptic mites were detected. All of 59 sheep infested with Psoroptes ovis were seropositive. Additionally, in 49% of 70 clinically unaffected sheep originating from P. ovis-infested flocks, specific antibodies could be detected, suggesting that asymptomatic infestations can be diagnosed by serology. The specificity of the ELISA was 96.5% as determined with 254 sheep originating from 44 flocks without clinical mange. Cross-reactivity in a low range was detected with selected sera of sheep with clinical chorioptic or forage mite infestations. Four sheep seroconverted 2 weeks after experimental P. ovis infestation, i.e. 2 weeks before clinical signs became obvious. After successful doramectin treatment of 14 sheep with naturally acquired P. ovis infestation, the ELISA values declined slowly but remained positive in seven cases beyond 17 weeks.  相似文献

Oestrosis, the nasal myiasis of sheep and goats, is caused by the larvae of Oestrus ovis L. 1758 (Diptera, Oestridae) that develop from the first to the third stage larva in the nasal cavities and frontal sinuses of affected animals. The authors report the results of an epidemiological study of oestrosis of sheep in Sardinia, Italy. Heads of 6-month to 10-year-old Sardinian sheep (n=566) from 124 free-ranging flocks were examined for the presence and location O. ovis larvae from December 1996 to November 1997. Larvae were collected, counted, and larval stages were identified. O. ovis larvae were found in 100% of examined flocks and in 91% (514/566) of examined sheep. The monthly prevalence ranged from 69% in May to 100% in July. First stage larvae were found in 82% (463) of all heads examined, second stage larvae in 65% (367) and third stage larvae in 10% (56). The majority of sheep harboured first stage larvae, with prevalences of over 80% throughout most of the study period. The prevalence of O. ovis found in this study of Sardinian sheep is the highest reported in the Mediterranean area. The high percentage of first stage larvae found throughout the entire study period may be due to a brief period of decreased rate of larval maturation, in particular in December 1996 (96%) and January-October 1997 (94%). Third stage larvae were consistently present, often however, with extremely low prevalences compared to total larval burden.  相似文献

Sheep scab, caused by Psoroptes ovis, is a severe and debilitating disease that can be treated and controlled by the use of acaricidal dips or the use of broad-spectrum avermectins. In Switzerland, control measures are state regulated. In particular, sheep should be routinely treated with ectocide dips or avermectin injections before they are moved onto common alpine pasture in late spring. However, a substantial part of the sheep population remains untreated and represents a potential reservoir for the mite population. Untreated sheep that are not moved to alpine pasture may infest treated sheep when flocks are reassembled in autumn. In an attempt to identify infested sheep, all flocks in the Canton of Schwyz (Switzerland) were serologically tested in 2001 and in 2002 (587 and 565 flocks, respectively). In 2003, a representative number (182 of the 531 flocks) was again investigated. Seropositive flocks were treated with doramectin (0.3 mg kg(-1) body weight, intramuscularly) from 2001 to 2003. In spring 2002, no chemo-methaphylaxis was given to seronegative flocks before movement onto common alpine pastures. Of the 587 flocks surveyed in spring 2001, 34 were seropositive (5.8%). These consisted of 21 infested with P. ovis, 1 with P. cuniculi, 4 with Chorioptes spp. and 8 of seropositivity of unknown origin; there was a decrease of seropositive flocks in spring 2002 (4.4%) with 15, 0, 2 and 8, respectively. Of the 182 flocks surveyed in spring 2003, just 4 flocks (2.2%) were seropositive. All the seropositive reactions in these flocks were the result of Chorioptes spp. infestations. There was a corresponding decrease in the proportion of seropositive animals from 6.3% in spring 2001 to 2.1% in spring 2003. These results corroborate the concept that it may be possible to target chemo-metaphylaxis and hence decrease the use of endectocides as well as of ectocides to control sheep scab. This would be of great benefit in reducing the likelihood of development of anthelmintic resistance against avermectins, decreasing the extent of environmental and human contamination with potentially toxic products and diminishing potential drug residues in meat and milk.  相似文献

Skin biopsies from two Merino sheep heavily infested with Psorergates ovis were immersed in liquid nitrogen and cut into vertical frozen sections stained with lipophilic Sudan IV or Oil Red O and haematoxylin. A survey of the lateral distribution of mite sections showed a majority (ca 80%) were in or within 0.2 mm of the follicle mouth. A survey of vertical distribution showed no mite penetration deeper than inner stratum corneum where 57% of mite sections were seen; 30% were within outer stratum corneum or scurf; 13% were on the outer surface and less than 1% were detached. Lipid was the only material seen within stained mites at a location considered to be gut. This was supported by dosing sheep with quinacrine, taking biopsies at Day 6 and Day 14 and examining frozen sections under blue light. Fluorescent lipid was seen at a location considered to be mite gut. From these results it was recommended that acaricide treatments against P. ovis be lipophilic and administered transepidermally because less than 15% of mites were at a superficial location likely to be reached by topical application.  相似文献

A cross-sectional epidemiologic study was conducted in order to detect the presence of and to estimate the seroprevalence of Oestrus ovis L. infection in flocks of sheep from the central region of the state of Yucatan, Mexico. The risk factors associated with disease were also identified. A sample size of 10 animals per farm was used to detect seropositive animals, considering a 30% prevalence and 95% confidence level. Blood samples of 689 sheep from 88 flocks were collected and a questionnaire with questions about the flock and the host was applied. The thin layer immune assay test was used. The risk factors were screened using logistic regression procedures. 77% of the flocks had at least one-positive animal with antibodies against O. ovis. The overall seroprevalence and standard error was 30.6 +/- 3.5%. Only flock size and sheep nose color showed association (P < 0.05) with the disease. The odds ratios for flocks with less than 11 and with 11 to 25 sheep, as related to herds with 25 or more sheep, were 0.74 and 1.73, respectively. Sheep with dark noses had a higher risk (OR = 1.46) compared with sheep having light noses (P < 0.05).  相似文献

Four trials including 11,266 sheep were conducted in South Africa to evaluate the efficacy of the systemic parasiticide ivermectin against field outbreaks of sheep scab (Psoroptes ovis) when two doses of approximately 200 micrograms/kg were administered subcutaneously seven days apart (days 0 and 7). As sheep scab is a notifiable disease in South Africa, it was not possible to include an untreated control group. The prevalence of clinically affected animals in the four treated flocks varied from 0.4 per cent to 99 per cent before the two treatments. After the treatments, there were no signs of active clinical infection in any of the sheep between days 28 and 30, or at subsequent examinations. P ovis mites were recovered from scrapings from 114 of 127 indicator sheep before the treatment but no mites were recovered from them between days 28 and 30 or 42 and 58 after the treatments.  相似文献

Psoroptes ovis of sheep origin, and Psoroptes cuniculi of rabbit origin were used in experimental infestations. In experiment I, groups of four rabbits and four sheep were infested with 50-100 mites of each isolate on the skin of the back (skin infestation, SI) or in the external auditory canal (aural infestation, AI). In rabbits, SI and AI with P. cuniculi and AI with P. ovis induced in all animals typical ear lesions and pronounced antibody reactions to P. cuniculi antigens in ELISA. After SI of rabbits with P. ovis no clinical signs were detected, no mites could be reisolated and no specific antibodies were detected. In sheep, P. ovis SI induced mange whereas AI did not induce typical clinical signs and mites could not be reisolated. In both these animal groups, ELISA revealed pronounced and comparable specific antibody reactions. After SI and AI with P. cuniculi no clinical symptoms were observed and no mites could be reisolated. Nevertheless, low levels of specific antibody were detected. In experiment II, clinical progression and antibody reactions to P. ovis SI in naive sheep were compared with sheep previously exposed to P. ovis or P. cuniculi. In both pre-exposed groups of animals, clinical signs appeared within 2 days after challenge infestation and three days earlier than in primarily infested sheep. Subsequently, no obvious difference in the clinical progression was observed between the three groups of animals. The results of this study document antigenetic crossreactivity of the two morphologically and genetically distinguishable Psoroptes species but differences in their biological behaviour and virulence which both are of epidemiological and taxonomic relevance.  相似文献

The results of an epidemiological and clinical study of flea infestations of farm animals in northern Libya is reported. Of 12,130 sheep examined from 124 flocks, 150 sheep were found to be infested with fleas from 50 different flocks. Likewise 23 goats from 2981 examined, and 11 calves from 1124 cattle examined were infested No fleas were recovered from camels or horses. Of 1861 fleas recovered from farm livestock, 1857 were Ctenocephalides felis strongylus and 4 were Pulex irritans. Dogs from farms and local clinics were also examined. Eight farms dogs were found to be infested with P. irritans. Of 79 infested dogs examined in veterinary clinics, 53 were found infested with P. irritans, 11 with Ctenocephalides felis felis, 12 had a mixed infestation of P. irritans and C. felis felis. Single dogs had mixed infestation of P. irritans and C. canis; C. felis felis and C. canis; and P. irritans, C. felis felis and Echidnophaga gallinacia. C. felis felis was also found on 15 infested cats. C. felis felis was never found on large farm animals despite frequently sharing their environment with dogs or cats. Likewise C. felis stongylus was never isolated from dogs or cats. This is consistent with the hypothesis that C. felis strongylus has become adapted to large farm animals, whilst C. felis felis is better adapted to dogs and cats. However, four stockmen were found infested with a total of 176 C. felis strongylus, which suggests that this subspecies is also a potential zoonosis. A significantly higher proportion of intensive farms had animals with flea infestation compared to semi-intensive farms. Fleas were not found in nomadic herds. Infested farm animals often presented with excoriation, alopecia, pruritis and hyperkeratitis particularly on the lower limbs. These signs are consistent with the generation of flea-bite hypersensitivity.  相似文献

Sheep scab, which is caused by the mite Psoroptes ovis, is a notifiable disease in Switzerland. In the framework of an epidemiological study in the canton of Schwyz, a sheep scab outbreak on 2 adjacent alpine pastures with 62 flocks (1770 sheep) was followed up clinically, parasitologically and serologically. No mass treatment of the flocks with organophosphates or avermectins had been undertaken before the flocks were taken up to the alpine pastures, but they were treated according to their serological status. Supervised treatment of seropositive judged flocks (at least one seropositive or at least 2 serologically equivocal animals per flock) with Doramectin was undertaken, whilst seronegative judged flocks remained untreated. Sheep returned from the summer pastures in early September 2001 and within three months scab infestation was diagnosed in 53 flocks (85.5%). These infested flocks were scattered all over the canton. Furthermore, the transfer of sheep from 6 infested flocks resulted in the transmission of the disease to 10 new flocks. Infested flocks were kept in quarantine and sanitized. The complexity of the traditional use of shared alpine pastures and the intense, uncontrolled trading with animals render the control of sheep scab difficult.  相似文献

The possibility that Damalinia ovis ingests intact epidermis could explain economically important irritation behaviour by infested sheep. To test this, feeding lice were observed on sheep by hand lens and macrophotography and on light and scanning electron microscope preparations. Aspects of feeding behaviour were described, particularly an epidermal ingestion posture. Louse ingesta were examined in paraffin and frozen sections. Lipid-covered stratum corneum squames were identified in crop, midgut, rectum and faeces. Nucleated keratinocytes from inner epidermal strata were not seen, neither were wool fibres. Vertical frozen sheep skin sections, with feeding lice cryofixed by liquid nitrogen, showed mandibles engaged in the outer stratum corneum of the epidermis. The epidermal origin of some ingesta was confirmed by confirming lice over sheep whose skin was stained with Oil Red O, whose colour was seen in crop, midgut, rectum and faeces of harvested lice. Lipase was found in louse midgut. It was concluded that sebaceous secretions may form an important component of the D. ovis diet. The relative contribution from loose scurf or from stratum corneum squames was not determined. Mechanical stimuli by feeding lice did not seem to be the source of host irritation. The role of sensitising agents in louse brei filtrates was investigated with inconclusive results. Intradermal injections of louse faeces did not elicit a demonstrable response.  相似文献

A survey for oestrosis was carried out in the slaughterhouse at Messine (Sicily) from May 1996 to April 1998. The heads of sheep from local flocks were collected each month. A total of 841 heads were examined according to a validated procedure. 469 (55.8%) were infested by OEstrus ovis larvae. 4423 larvae were harvested: 1829 first instar larvae or L1 (41.4%), 1286 second instar larvae or L2 (29.1%) and 1308 third instar larvae or L3 (29.6%). The mean larval burden for infected sheep was 9.4 larvae with an average of 3.9 L1, 2.7 L2 and 2.8 L3. No period of hypobiosis was recorded. The proportions of larvae in each of the different larval stages was similar from January to September, however, from October to December the percentage of L1 was higher, indicating a period of slowed development. In order to lower the overall prevalence of this disease, it would be necessary to use a parasiticide effective against O. ovis, for all routine parasite control treatments.  相似文献

After several thousand sheep had been imported from Australia and New Zealand to Croatia during 1995, many native sheep that had been in contact with the imported animals acquired a severe ocular disease closely resembling infectious keratoconjunctivitis. In affected flocks glucose-fermenting mycoplasma were isolated from 48 per cent of conjunctival swabs and Branhamella ovis from 58 per cent. Twelve of 42 culturally and biochemically identical isolates were identified as Mycoplasma conjunctivae by polymerase chain reaction. From the conjunctivae of two animals M conjunctivae and M arginini were isolated in mixed culture. For many reasons most farmers removed the imported animals from their flocks and only sporadic cases of the disease were recognised in 1996. At the end of 1997, six flocks which were clinically free of the disease but had been affected during 1995, and five flocks with no history of the severe ocular disease were examined clinically and microbiologically, and were found to be free of M conjunctivae infection. At the time, B ovis was cultured almost exclusively from sheep originating from flocks which had been affected during 1995 and/or 1996. It was usually isolated in pure culture or as the predominant bacterial species, and was often accompanied by mild conjunctivitis. There were no microbiologically confirmed new cases of infectious keratoconjunctivitis during 1998 and 1999.  相似文献

Lipid ingestion from sheep epidermis by Psoroptes ovis (Acari: Psoroptidae)   总被引:1,自引:0,他引:1  
Skin biopsies from three groups of sheep infested with Psoroptes ovis were cryofixed in liquid nitrogen to preserve outer epidermis with its lipid. From one group of five sheep (Group A), biopsies were taken from relatively healthy skin near the edge of scab lesions where mites had congregated. Frozen vertical sections from these biopsies were stained with Oil Red O and haematoxylin before mounting. Red lipid globules were plentiful within the body cavity of sectioned mites, but ingested lipid could not be distinguished from endogenous mite body stores by this technique. In a second group of five sheep (Group B), enclosed lumbar skin areas were coloured red with the lipophilic stains Oil Red O or Sudan IV. These enclosed coloured skin areas were inoculated with P. ovis and sampled by skin biopsy 1 or 4 days later. Cryofixed biopsies were cut into vertical frozen sections and mounted without staining for examination. Red-coloured lipid within mites, matching red-coloured lipid on outer epidermis, was evidence for the epidermal origin of P. ovis ingesta in the early stages of an infestation. From two other sheep (Group C), cryofixed biopsies were examined by scanning electron microscope and mites were seen with mouthparts embedded in abraded outer epidermis, but the precise depth of epidermal penetration was not determined. A light microscope survey of 3198 frozen vertical skin sections from the first 10 sheep (Groups A and B) showed that inner stratum corneum of the epidermis was the deepest penetration recorded for gnathosomes of P. ovis cryofixed in situ by liquid nitrogen. No structure of P. ovis was identified in dermal tissues.  相似文献

The astigmatid mite Psoroptes ovis is the causative agent of sheep scab, a highly contagious parasitic disease of sheep. Infection causes severe allergic dermatitis, resulting in damage to the fleece and hide, loss of condition and occasional mortality. Interest in the P. ovis allergens led us to characterise a glutathione S-transferase (GST) which displays homology to GST allergens isolated from the house dust mite, Dermatophagoides pteronyssinus and the cockroach, Blatella germanica. A cDNA encoding a mu-class GST from P. ovis was expressed in Escherichia coli and the recombinant protein purified for biochemical analysis. SDS-PAGE analysis indicated that the purified product was homogeneous and had an apparent molecular weight of 30 kDa. The recombinant GST (rGST) is active towards the substrate 1-chloro-2,4-dinitrobenzene (CDNB), whereas 1,2-dichloro-4-nitrobenzene (DCNB) is a poor substrate. The recombinant protein was also tested for recognition by IgE and IgG antibodies in serum from P. ovis na?ve and P. ovis infested sheep. Neither IgE nor IgG antibodies were detected to the rGST. Prausnitz--Küstner testing with rGST did not provoke a characteristic weal and flare response. Biopsies collected at the PK test sites were stained for eosinophils, neutrophils, mast cells and basophils. Neutrophil, mast cell and basophil counts were not significantly different to the controls. Eosinophil numbers were significantly higher than controls, but were not due to an IgE response.  相似文献
