The effects of temperature on anatomical and chemical characteristics of Quercus cerris cork were examined. Cork samples were subjected to isothermal air heating between 150 °C and 400 °C and analyzed for mass loss, cellular structure and chemical composition.The thermal decomposition of Q. cerris cork is similar to that of Q. suber cork. Cork is thermally stable below 200 °C and after that degradation depended on temperature and heating time with increasing mass loss, i.e. 3% at 200 °C 10 min and 46% at 350 °C 60 min. With temperature and starting at 200 °C, cells expanded, cell wall thickness was reduced and corrugations were lost.Extractives degraded at lower temperatures, although aliphatic extractives were found to be more stable. Suberin from Q. cerris was more heat resistant than Q. suber suberin, while lignin showed similar resistance.These results provide a basis for studies on the production of Q. cerris bark expanded cork agglomerates for insulation purposes.  相似文献

A comparative study on the chemical composition of oak cork (Quercus suber L.) and corresponding industrial residues and birch (Betula pendula L.) outer bark is reported. Cork oak samples have lower extractives contents (6–9%) and higher contents of carbohydrates and lignin (23–27 and 33–38%, respectively) than those found for birch outer bark (40, 6 and 9%, respectively); suberin contents accounted for around 30% of cork, 11% of industrial cork powder and 45% of birch outer bark. Analysis of the suberin monomeric composition revealed that C18 and C22 ω-hydroxyfatty acids (including mid-chain epoxy- and dihydroxy-derivatives), followed by α,ω-dicarboxylic acids, are the main components in both suberins, with 9,10-epoxy-18-hydroxyoctadecanoic, 18-hydroxyoctadec-9-enoic, 9,10,18-trihydroxyoctadecanoic and octadec-9-enoic acids as the major components. The differences in the relative amounts of these acids in the suberin samples and the impact on the potential exploitation of the different industrial by-products are discussed.  相似文献

Benthic diatoms of the genus Cocconeis contain a specific apoptogenic activity. It triggers a fast destruction of the androgenic gland in the early post-larval life of the marine shrimp Hippolyte inermis, leading to the generation of small females. Previous in vitro investigations demonstrated that crude extracts of these diatoms specifically activate a dose-dependent apoptotic process in human cancer cells (BT20 breast carcinoma) but not in human normal lymphocytes. Here, a bioassay-guided fractionation has been performed to detect the apoptogenic compound(s). Various HPLC separation systems were needed to isolate the active fractions, since the apoptogenic metabolite is highly active, present in low amounts and is masked by abundant but non-active cellular compounds. The activity is due to at least two compounds characterized by different polarities, a hydrophilic and a lipophilic fraction. We purified the lipophilic fraction, which led to the characterization of an active sub-fraction containing a highly lipophilic compound, whose molecular structure has not yet been identified, but is under investigation. The results point to the possible medical uses of the active compound. Once the molecular structure has been identified, the study and modulation of apoptotic processes in various types of cells will be possible.  相似文献

In order to apply state-of-the-art molecular breeding techniques in fibre crop it is necessary to have a good knowledge of major polymer biosynthesis gene sequences and their expression pattern. Polymerase chain reaction was employed to isolate sequences of the major genes for lignin and cellulose biosynthesis in a kenaf cultivar. CeSA, 4cl, c4h, cad, and ccr gene primers were designed according to their conservative regions; partial sequences of lignin and cellulose biosynthesis genes were obtained. One actin II gene sequence was also isolated from the kenaf genome as a housekeeping gene to be employed in qPCR analysis. Expression levels of genes c4h, cad and CeSA in bark and core from plants harvested at three different growth stages were evaluated. Using qPCR analyses it was found that the expression levels of the two biosynthesis lignin genes in bark tissues increased during plant growth, while a negative trend was recorded in core tissues. In both bark and core, the quantity of lignin was positively correlated to plant growth while cellulose content decreased.  相似文献

Suberin oligomers, isolated from cork (Quercus suber L.), were used as additives in ‘Waterless’ and vegetable-oil ink formulations, in the range of 2–10% w/w. The rheological behaviour of the suberin oligomers as well as of the inks, with and without suberin, were investigated as a function of temperature. It was shown that the addition of suberin induces a decrease of viscosity of both inks. The tack of pristine inks, suberin oligomers and their mixtures were determined at different temperatures: the variation of this parameter as a function of time provided information about the drying kinetics of these formulations. The tack of the ‘Waterless’ ink was found to increase with the introduction of suberin, whereas that of vegetable-oil based counterparts decreased. All the trends observed were interpreted in terms of the differences in composition between the two types of inks. Preliminary printing tests were carried out with the various suberin-containing inks.  相似文献

To extract essential oil for industrial use, the yields and compositions of bark oil during Cinnamomum cassia growth (1-3 years old for the branch bark; 5-12 years old for the stem bark) were determined. The branch bark fraction had a higher essential oil yield than the whole branch. The essential oil yield from branch bark varied within 2.70-3.11% (w/w), while that from stem bark was 0.41-2.61% (w/w) due to differences in age and segment (top, center and lower) of the tree. There were 41 volatile compounds identified in bark oil, among which the majority presented high fluctuations in percentage of composition both in different growth stages and segments. Variations in oil yields did not present the same pattern as the percentages of trans-cinnamaldehyde. The results suggest that choosing bark according to trees’ growth stages and separating stem barks into top, center and lower sections within a tree should significantly improve the extraction efficiency.  相似文献

The composition of Norway spruce (Picea abies (L.) Karst.) and Scots pine (Pinus sylvestris L.) barks was studied after grinding and fractioning into different particles sizes.Both barks fractionated well and with similar fraction yield profile. The yield of fines was low and the major fractions were larger particles, i.e. 2.4% and 3.1% of particles under 0.425 mm and 66.0% and 50.3% of particles over 2 mm, respectively for spruce and pine bark.The chemical composition of spruce and pine barks, as a mass weighed average of all granulometric fractions was, respectively: ash 3.3 and 4.6%; total extractives 21.6 and 18.8% (hydrophilic extractives were dominant), lignin 27.9 and 33.7% and holocellulose 42.7 and 37.6%. Suberin accounted for 1.3% and 1.6% of spruce and pine bark, respectively. The non-cellulosic monosaccharides showed in both barks predominance of arabinose followed by xylose and mannose.Ash elemental composition showed that N represented about 35% of the total inorganics, Ca 35% and K 17%. Zn, Cu, Ni, Cr and Pb were present in both barks at levels under 1% of the total inorganics. Spruce bark had in average higher contents compared to pine bark, except for Pb and Cr.Size reduction of spruce and pine bark did not apply randomly to the different components and instead resulted into partial separation of the inorganic and organic matter into different size particles. Many biomedical products have already been obtained from marine organisms. In order to search more therapeutic drugs against cancer, this study demonstrates the cytotoxicity effects of Cladiella australis, Clavularia viridis and Klyxum simplex extracts on human oral squamous cell carcinoma (SCC4, SCC9 and SCC25) cells using cell adhesion and cell viability assay. The morphological alterations in SCCs cells after treatment with three extracts, such as typical nuclear condensation, nuclear fragmentation and apoptotic bodies of cells were demonstrated by Hoechst stain. Flow cytometry indicated that three extracts sensitized SCC25 cells in the G0/G1 and S-G2/M phases with a concomitant significantly increased sub-G1 fraction, indicating cell death by apoptosis. This apoptosis process was accompanied by activation of caspase-3 expression after SCC25 cells were treated with three extracts. Thereby, it is possible that extracts of C. australis, C. viridis and K. simplex cause apoptosis of SCCs and warrant further research investigating the possible anti-oral cancer compounds in these soft corals.  相似文献

The chemical composition of the lipophilic extracts of the inner and outer bark fractions of Eucalyptus grandis and Eucalyptus urograndis (E. grandis × Eucalyptus urophylla) cultivated in Brazil and Eucalyptus maidenii, cultivated in Portugal was studied by gas chromatography-mass spectrometry. The extracts were shown to be mainly composed of triterpenic compounds (along with mono and sesquiterpenes in E. maidenii) followed smaller amounts of fatty acids, fatty alcohols, and aromatic compounds.Triterpenic acids (mainly ursolic, betulinic and oleanolic acids), are particularly abundant in outer barks representing 5.2 g/kg, 5.7 g/kg and 9.3 g/kg in E. urograndis, E. grandis and E. maidenii outer barks, respectively. Although these compounds were found in considerably smaller amounts than those previously reported for Eucalyptus globulus, the total amounts of bark generated every year in South American pulp mills using E. urograndis and E. grandis, as well as the growth potential of E. maidenii plantations, the bark residues from these species are obvious candidates for the extraction of valuable triterpenic compounds.  相似文献

Five new 7α-hydroxyeunicellin-based diterpenoids, designated as cladieunicellins M–Q (1–5), were isolated from a Formosan octocoral Cladiella sp. The structures of 1–5 were elucidated on the basis of spectroscopic methods and by comparison of the data with those of the related metabolites. Cytotoxicity of metabolites 1–5 against the human leukemia Molt 4 and HL 60 is also described. Among them, compounds 1, 3 and 5 exhibited moderate cytotoxicity toward Molt 4 cells with IC50 values 16.43, 14.17 and 15.55 μM, respectively. Preliminary SAR (structure activity relationship) information was obtained from these compounds and their analogues.  相似文献

Tomicus brevipilosus (Eggers) (Coleoptera: Curculionidae, Scolytinae) was recently discovered as a new pest of Yunnan pine (Pinus yunnanensis Franchet) in Yunnan Province in southwestern China. However, little was known on its reproductive biology and pattern of trunk attack on Yunnan pine. The objectives of this study were to better understand the reproductive biology of T. brevipilosus by investigating the seasonality of trunk attacks by parent adults for the purpose of reproduction (i.e., breeding attacks) and the within-tree pattern of these attacks. Our results showed that T. brevipilosus breeding attacks in P. yunnanensis generally started in early March and ended in early June in Anning County, Yunnan. T. brevipilosus exhibited two general patterns of infestation. From early March to mid-April, T. brevipilosus bred preferentially in the trunks of Yunnan pine trees that were already infested by Tomicus yunnanensis Kirkendall and Faccoli and Tomicus minor (Hartig), colonizing spaces along the trunk (mostly in the mid- and lower trunk) that were not already occupied by the other two Tomicus species. Later, from about mid-April to early June, when there were no Yunnan pine trees newly infested by T. yunnanensis and T. minor, T. brevipilosus attacked Yunnan pine by itself, infesting the lower parts of the trunk first and then infesting progressively upward along the trunk into the crown. Infestation by T. brevipilosus extends the total period that P. yunnanensis trees are under attack by Tomicus beetles in southwestern China, which helps explain why Yunnan pine has suffered high levels of tree mortality in recent decades.  相似文献

During our search for new natural products from the marine environment, we discovered a wide range of cyclic peptides from a marine Photobacterium, closely related to P. halotolerans. The chemical fingerprint of the bacterium showed primarily non-ribosomal peptide synthetase (NRPS)-like compounds, including the known pyrrothine antibiotic holomycin and a wide range of peptides, from diketopiperazines to cyclodepsipeptides of 500–900 Da. Purification of components from the pellet fraction led to the isolation and structure elucidation of four new cyclodepsipeptides, ngercheumicin F, G, H, and I. The ngercheumicins interfered with expression of virulence genes known to be controlled by the agr quorum sensing system of Staphylococcus aureus, although to a lesser extent than the previously described solonamides from the same strain of Photobacterium.  相似文献

As part of on-going efforts to use eco-friendly alternatives to chemical pesticides, methanol crude extracts of Plectranthus glandulosus and Callistemon rigidus leaves were sequentially fractionated in hexane, chloroform, ethyl acetate, and methanol to establish the most active fraction(s) against Callosobruchus maculatus in cowpea. Cowpea seeds (25 g) were treated with 0.5, 1, 2, and 4 g/kg of extract to evaluate the contact toxicity and F1 progeny production of the beetles in the laboratory. Mortality was recorded 1, 3, and 7 d postexposure. P. glandulosus hexane fraction was more toxic than the other fractions recording 100% mortality at 4 g/kg, within 7 d with LC50 of 0.39 g/kg. Hexane fraction of C. rigidus showed superior toxicity, causing 100% mortality at 4 g/kg within only 1 d of exposure with LC50 of 1.02 g/kg. All the fractions greatly reduced progeny emergence, with C. rigidus hexane fraction being the best progeny inhibitor. Fractions of P. glandulosus and C. rigidus leaves had sufficient efficacy to be a component of storage pest management package for C. maculatus.  相似文献

The Asiatic oak weevil, Cyrtepistomus castaneus Roelofs (Coleoptera: Curculionidae), is a nonnative defoliator of trees in the Fagaceae family in the United States but has not been studied on Castanea species in the southern Appalachian Mountains. Planted trees of Castanea dentata (Marsh.) Borkh. (Fagales: Fagaceae), Castanea mollissima Blume (Fagales: Fagaceae), and four hybrid breeding generations were evaluated in 2012 for insect defoliation and C. castaneus abundance and frequency. Defoliation was visually assessed throughout the growing season at two sites in the southern Appalachian Mountains (western North Carolina and eastern Tennessee). C. castaneus abundance and frequency were monitored on trees using beat sheets and emergence was recorded from ground traps. Asiatic oak weevils were more abundant and more frequently collected on American chestnut (Ca. dentata) and its most closely related BC3F3 hybrid generation than on the Asian species Ca. mollissima. In most months, C. castaneus colonization of hybrid generations was not significantly different than colonization of parental species. Frequency data for C. castaneus suggested that adults were distributed relatively evenly throughout the study sites rather than in dense clusters. Emergence of C. castaneus was significantly higher under a canopy dominated by Quercus species than under non-Quercus species or open sky. C. castaneus emergence began in May and peaked in late June and early July. These results may be useful for resource managers trying to restore blight-resistant chestnut to the Southern Appalachians while minimizing herbivory by insect pests.  相似文献

Background

Stemflow is an essential hydrologic process shaping the soil of forests by providing a concentrated input of rainwater and solutions. However, the transport of metazoans by stemflow has yet to be investigated. This 8-week study documented the organisms (<?2 mm) present in the stemflow of different tree species. Because the texture of the tree bark is a crucial determination of stemflow, trees with smooth bark (Carpinus betulus and Fagus sylvatica) and rough bark (Quercus robur) were examined.

Results

Up to 1170 individuals per liter of stemflow were collected. For rotifers and nematodes, a highly positive correlation between abundance and stemflow yield was determined. Both taxa were predominant (rotifers: up to 70%, nematodes: up to 13.5%) in the stemflow of smooth-barked trees whereas in that of the oak trees collembolans were the most abundant organisms (77.3%). The mean number of organisms collected per liter of stemflow from the two species of smooth-barked trees was very similar. A higher number of nematode species was found in the stemflow of these trees than in the stemflow of rough-barked oak and all were typical colonizers of soil- and bark-associated habitats.

Conclusion

This pilot study showed for the first time that stemflow is a transport vector for numerous small metazoans. By connecting tree habitats (e.g., bark, moss, lichens or water-filled tree holes) with soil, stemflow may influence the composition of soil fauna by mediating intensive organismal dispersal.

Escherichia coli BL21 (DE3) and its derivatives are widely used for the production of recombinant proteins, but these purified proteins are always contaminated with lipopolysaccharide (LPS). LPS is recognized by the toll-like receptor 4 and myeloid differentiation factor 2 complex of mammalian immune cells and leads to release of pro-inflammatory cytokines. It is a vital step to remove LPS from the proteins before use for therapeutic purpose. In this study, we constructed BL21 (DE3) ∆msbB28pagP38 mutant, which produces a penta-acylated LPS with reduced endotoxicity. The plasmids harboring pagL and/or lpxE were then introduced into this mutant to further modify the LPS. The new strain (S004) carrying plasmid pQK004 (pagL and lpxE) produced mono-phosphoryated tetra-acylated lipid A, which induces markedly less production of tumor necrosis factor-α in the RAW264.7 and IL-12 in the THP1, but still retains ability to produce recombinant proteins. This study provides a strategy to decrease endotoxic activity of recombinant proteins purified from E. coli BL21 backgrounds and a feasible approach to modify lipid A structure for alternative purposes such as mono-phosphoryl lipid A (MPL) as vaccine adjuvants.  相似文献

Iron, as inorganic ion or as oxide, is widely used by biological systems in a myriad of biological functions (e.g., enzymatic, gene activation and/or regulation). In particular, marine organisms containing silica structures—diatoms and sponges—grow preferentially in the presence of iron. Using primary sponge cell culture from S. domuncula–primmorphs—as an in vitro model to study the Demospongiae spiculogenesis, we found the presence of agglomerates 50 nm in diameter exclusively inside sponge specialized cells called sclerocytes. A clear phase/material separation is observed between the agglomerates and the initial stages of intracellular spicule formation. STEM-HRTEM-EDX analysis of the agglomerates (30–100 nm) showed that they are composed of pseudohexagonal nanoparticles between 5 and 15 nm in size, displaying lattice parameters corresponding to hematite (Fe2O3) and mixed iron oxide phases typically attributed to ferritin. Further analysis, using western blotting, inductively coupled plasma mass spectrometry (ICP-MS), sequence alignment analysis, immunostaining and magnetic resonance imaging (MRI), of mature spicule filaments confirm the presence of ferritin within these organic structures. We suggest that S. domuncula can be classified as a dual biomineralizating organism, i.e., within the same cellular structure two distinct biomineralizing processes can occur as a result of the same cellular/metabolic function, spiculogenesis.  相似文献

Bacteria are known to influence domoic acid (DA) production by Pseudo-nitzschia spp., but the link between DA production and physiology of diatoms requires more investigation. We compared a toxic P. multiseries to a non-toxic P. delicatissima, investigating links between DA production, physiological parameters, and co-occurring bacteria. Bacterial communities in cultures of both species were reduced by antibiotic treatment, and each of the diatoms was inoculated with the bacterial community of the other species. The physiology of P. delicatissima was minimally affected by the absence of bacteria or the presence of alien bacteria, and no DA was detected. P. multiseries grew faster without bacteria, did not produce a significant amount of DA, and exhibited physiological characteristics of healthy cells. When grown with alien bacteria, P. multiseries did not grow and produced more DA; the physiology of these cells was affected, with decreases in chlorophyll content and photosynthetic efficiency, an increase in esterase activity, and almost 50% mortality of the cells. The alien bacterial community had morphological and cellular characteristics very different from the original bacteria, and the number of free-living bacteria per algal cell was much higher, suggesting the involvement of bacteria in DA production.  相似文献

In commercial apple orchards in southern Brazil, it is common to avoid hail damage by covering the trees with nets. This study was carried out to assess the effects of both black and white hail netting on the temporal dynamics of Glomerella Leaf Spot disease (GLS) in a commercial ‘Royal Gala’ apple orchard in a high-altitude region of southern Brazil during two growing seasons (2003/2004 and 2004/2005). Apple trees on M9 rootstock, trained to a slender spindle and with a spacing of 5.5 m × 3 m, were left uncovered (control) or covered with black or white hail netting. The incidence and severity of GLS were quantified biweekly from October to February by evaluating 100 leaves distributed randomly on four medium-height branches per plant on each 12 replications per treatment. Disease progress curves were constructed from the data and the epidemics were compared by using four epidemiological measures: the beginning of symptom appearance (BSA), the time to reach the maximum disease intensity (TRMDI), the maximum value of disease severity (Ymax), and the area under the disease progress curve (AUDPC). The highest intensity of GLS was observed in the plants under the black hail netting. Significant reductions in BSA and TRMDI and significant increases in Ymax and AUDPC were found among treatments in both growing seasons. However, no significant differences of GLS were found between the white hail netting and the control in either growing season.  相似文献

The effects of extracts of different parts of the perennial tropical plant Balanites aegyptiaca (L) Del., including various solvent extracts of roots, methanol extracts from leaves, fruits, flowers and roots, partially purified saponins obtained from its roots and a standard saponin were studied on the life cycle (adult longevity, number of eggs, crawlers, adults, weight of adults and % wax content) of a laboratory-reared parthenogenic line of the mealy bug, Maconellicoccus hirsutus (Homoptera: Pseudococcidae). Extracts derived from various parts of B. aegyptiaca (leaves, fruits, flowers, and roots in methanol) affected the life cycle of M. hirsutus with a methanol root extract being the most effective at a concentration of 500 μg ml−1. Partially purified saponin of B. aegyptiaca and the commercial bark saponin extract (Sigma) from Quillaja saponaria at a concentration of 500 μg ml−1 were effective in reducing the longevity of M. hirsutus. Significant reductions in oviposition by M. hirsutus were found for all the extracts at a concentration of 500 μg ml−1. Extracts also affected the number of emerging crawlers, number of adults as well as the weight and wax content of emerging adults. These studies suggest that B. aegyptiaca plant extracts and saponins can be useful botanical insecticides for the protection of crops from mealy bugs.  相似文献
