Thyroid glands from 64 bulls with hyperplastic and/or neoplastic changes in ultimobranchial remnants and in the parafollicular (C) cell system were studied structurally and with immunohistochemical methods. Antibodies against thyroglobulin, calcitonin, somatostatin, and neurotensin were used to detect these substances. Two different types of changes were observed. One change was hyperplasia and neoplasia of the ultimobranchial remnants that affected all their epithelial constituents. These included ultimobranchial follicles, cysts and tubules, as well as solid nests formed by basophilic immature cells which were functionally undifferentiated and unreactive with all the antisera used. Differentiated follicular cells that formed thyroid follicles and cribriform structures with immunohistochemical evidence of thyroglobulin production were also found. In addition, differentiated light and cytoplasm-rich cells were scattered in the walls of the thyroid follicles, ultimobranchial follicles, cysts and tubules as well as in the solid component. They were argyrophilic and reacted with antibodies against calcitonin and somatostatin. The other change was a diffuse or multifocal hyperplasia of the parafollicular (C) cells that was present in other parts of the thyroid parenchyma--sometimes with gradual development of sclerotic tumors that had been exclusively formed by these cells. They corresponded to light cytoplasm-rich cells seen in the ultimobranchial lesions that were argyrophilic and harbored material reactive with antibodies against calcitonin and/or somatostatin. The changes observed in the parafollicular cell system resembled lesions seen in human thyroid glands with the familial variant of medullary carcinoma as well as those reported in thyroid glands of patients with longstanding hypercalcemia.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This remnant appeared in the form of irregular and elongated follicles of various size and shape with folded lumen. These structures occupied a peripheral location beneath the capsule, as well as being embedded mostly in the vascular connective tissue of the thyroid. Calcitonin cells were predominantly localized in the UB follicles. Scanning electron microscopy revealed that surface of most UB follicular cells was almost flat and polyhedral or hexagonal in shape. Other follicles possessed cells with dome shaped apical surface demarcated by a shallow intercellular depression. The luminal surface of the follicular cell lining was studded with microvilli that appeared with various density and length and were numerous at the cellular borders. Interestingly, most of UB follicular cells were provided with single cilium, projecting over the cell surface. Notably some UB follicles presented various stages of apocrine activities.  相似文献

Surface ultrastructural observation of the thyroid gland in donkey revealed the coexistence of certain unusual follicles or cysts, the ultimobranchial (UB) follicles, which exhibited large diameters and less regular outlines. In addition, several small follicles, cell nests or solid cellular masses were also demonstrated in association with these structures. Surface of most UB follicular cells was almost hexagonal in shape, studded with sparse or dense pleomorphic microvilli, which were much denser at the cellular borders. Each follicular cell bore single cilium that projecting over the cell surface. Interestingly, bleb-like apocrine protrusion of the apical cytoplasm of some follicular cells was frequently demonstrated. These apocrine blebs (or aposomes) were varied in shape, size and showed smooth or irregular surface. In addition to such aposomes, deep to shallow circumscribed or eroded areas of the apical cell surface was frequently demonstrated. These findings indicate that the UB follicular cells in donkeys, similar to those of camels, exhibit an apocrine activity alongside with the usual merocrine mode of secretion.  相似文献

Eight thyroid gland epithelial tumors were found in 7 cows and 1 bull in a retrospective study of thyroid gland lesions in slaughtered cattle. All tumors were classified as ultimobranchial thyroid carcinomas based on morphology and immunohistochemistry. All tumors consisted of solid sheets and nests of polygonal to oval epithelial cells, with more sparsely dispersed colloid-filled follicles. Connective tissue separating nests of epithelial cells varied from delicate fibrovascular stroma to dense collagenous stroma. Fusiform epithelial cells with rare neural fibers and ganglion cells were present in 1 tumor. Cells within solid areas of these tumors were immunoreactive for calcitonin, calcitonin gene-related peptide, neuron-specific enolase, and synaptophysin. Colloid and follicle cells were immunoreactive for thyroglobulin. Few follicle cells also were reactive for calcitonin gene-related peptide. Neoplastic cells invaded the fibrous capsules in all 8 cattle. These tumors represented proliferation of a mixed population of undifferentiated cells, C cells, and thyroid follicular epithelial cells, presumably derived from the thyroid ultimobranchial bodies. These ultimobranchial carcinomas in slaughtered cattle are comparable to ultimobranchial tumors described in dairy bulls and the intermediate type of thyroid gland carcinomas (mixed thyroid medullary carcinomas) described in human beings.  相似文献

The development of endocrine cells in the thyroid and parathyroid glands in the golden hamster was studied immunohistochemically in relation to the formation of these glands. The thyroid was formed on day 9 of gestation by the ventral outpocketing of the foregut between the first and second branchial pouches. The thyroid epithelial cells were faintly thyroglobulin-immunoreactive on day 10.5 of gestation. This immunoreaction became intense thereafter, but was almost confined to the cytoplasm of epithelial cells until birth. It appeared in the follicular lumen in newborn animals. The ultimobranchial body was derived from the fifth pouch and fused with the thyroid on day 12 of gestation. Calcitonin-immunoreactive cells first appeared on day 14 of gestation in the dorsomedial part of the thyroid derived from the ultimobranchial body and increased in number and intensity thereafter. Somatostatin-immunoreactive cells also appeared in the dorsomedial part of the thyroid derived from the ultimobranchial body on day 13 of gestation, and increased in number in newborn animals, but decreased thereafter. The parathyroid was derived from the third pouch, situated on day 13 of gestation on the dorsolateral side of the thyroid, and surrounded by a common capsule with the thyroid. Parathormone-immunoreactive cells first appeared on day 15 of gestation in the parathyroid and increased in number and intensity after birth.  相似文献

The UB population was observed as clear cells, either singly distributed or as groups of cells between the component cellular elements lining the UB structures. They displayed various shapes: oval, pear-like, rounded, cubical, columnar and or ellipsoid. Those populating the cell nests or solid cellular masses were almost polygonal or ovoid in shape. The cytoplasm contained well-developed golgi complexes, several mitochondria and characteristically presented rounded or oval membrane-bound electron dense granules of different sizes. The latter granules were irregularly distributed within the cytoplasm especially at the vascular pole. The nucleus of C cells was relatively large, indented, occupying a more or less central position in the cytoplasm. It presented various chromatin contents; the heterochromatin variety was predominantly demonstrated at the marginal zone. However, the euchromatin occupied a more central position within the nucleus.  相似文献

A systematic immunohistochemical study of the ultimobranchial tubule (UBT) has been carried out in 45 Wistar rats of different ages (0, 5, 10, 15, 20, 25, 30, 60 and 120 days). The existence of calcitonin immunoreactive cells in the UBT wall has been demonstrated in a 5-days old rat. In addition, immunohistochemical studies for thyroglobulin revealed positive staining in follicular cells connected to the UBT and, occasionally, in isolated cells lying within solid clusters from the UBT. These last results together with the continued and repeated existence of numerous mitosis and PAS (+) microfollicles, apparently rising from the UBT, support the hypothesis that the ultimobranchial body (UBB) may contribute partially to the formation of a part of the follicular component.  相似文献

There is good evidence for the interaction of neuroendocrine and immune systems. Endotoxin (LPS)-induced mediators (e.g., cytokines, prostaglandins etc) set on endocrine organs (e.g., the hypothalamo-pituitry-adrenal axis; thyroid glands etc). Endotoxin-treated, intestinal ischemic, or irradiated rats show decreased T4 levels of blood. These animals cannot respond to TSH because the TSH-receptors of follicular membranes are disturbed by LPS in the thyroid glands. Radiodetoxified endotoxin is an effective immunstimulator and does not disturb the follicular membrane of thyroid gland. Thus, the T4 production remains normal. The bile acids—as the end-product of cholesterol metabolism—play an important role in the physiological defense of macroorganisms against endotoxin and other lipid-like agents (Physico-chemical defense) and in the regulation of endocrine system, including the reproduction.  相似文献

Freshwater mud eel, Amphipnous cuchia, were injected intraperitoneally daily with 100 ng of vitamin D3/100 g body weight and maintained in media containing either no calcium or different calcium concentrations. The eels were killed after 1, 3, 5, 10 and 15 days following the treatment and their serum calcium levels were measured. The ultimobranchial glands were fixed and processed using the routine paraffin method for histological studies. The results of the present study indicate that vitamin D3 can induce hypercalcaemia in eels kept in different calcium environments. Also, the ultimobranchial glands became hyperactive following vitamin D3 treatment. It is concluded that in mud eels, the gland has a calcium-regulating function.  相似文献

The complex carbohydrates in the camel duodenal glands were examined histochemically at light and electron microscopic levels. The duodenal glands of the camel were distributed in the submucosa 2 m caudal from the pylorus. These were branched tubuloalveolar glands. The terminal portion of each lobule was formed by only one type of mucous cell. The duodenal gland cells contained acidic and neutral carbohydrates. The mucous cells mainly contained sulfate and carboxyl carbohydrate with sialic acid, and they also contained a few neutral carbohydrates with different saccharide residues such as mannose, glucose, galactose, N-acetyl glucosamine and N-acetyl galactosamine. The results showed that the secretary granules of the duodenal glands in the camel contain mainly acidic carbohydrates. These findings seem to be the morphological characteristics of the duodenal glands in the camel.  相似文献

We investigated the morphology and the distribution of the nerve fibres in golden hamster (Mesocricetus auratus) parathyroid glands using antibodies to calcitonin gene-related peptide (CGRP) and substance P, and electron microscopy. CGRP-immunoreactive nerve fibres were densely distributed in the interstitial tissues and the capsules of the hamster parathyroid glands. Some nerve fibres were detected in close proximity of the parathyroid chief cells. The distribution pattern for substance P-immunoreactive nerve fibres was roughly the same as for CGRP-immunoreactive fibres. Ultrastructurally, we found numerous nerve fibres joining the blood vessels. Axon bundles were located adjacent to the smooth muscle cells of the arterioles. The axons formed structurally specialized neuromuscular junctions with the vascular smooth muscle cells. Some axons were in close vicinity to the parathyroid chief cells. These findings indicate that the hamster parathyroid gland contain CGRP and substance P, which may regulate the blood flow and the secretory activity of the gland.  相似文献

Apoptosis has been shown to be an important regulator of endometrium function. To clarify the regulation of apoptosis in the cat endometrium during the normal oestrus cycle, the expressions of the apoptosis‐related proteins (Bcl‐2 and Bax) and their correlation to the inhibitor of apoptosis protein Survivin were analysed using immunohistochemistry. The TUNEL technique (TdT‐mediated dUTP nick end labelling) was also used to detect DNA fragmentation characteristic of apoptotic cells. The results demonstrated that TUNEL labelling is not effective for the detection of apoptosis in cat endometrium. Survivin was expressed in the luminal and glandular epithelial cells of cat endometrium during all phases of the oestrus cycle. Survivin was localized in both the cytoplasm and nuclei of superficial and deep uterine gland cells during the luteal phase, while only cytoplasmic staining was observed during the follicular and anoestrus phases. Bax immunoreactivity in the cytoplasm of luminal and glandular epithelial cells as well as the smooth muscle cells of blood vessels was weak in the anoestrus phase. Compared with anoestrus, the intensity of Bax immunostaining was moderate in the follicular phase and increased dramatically in the luteal phase. Bcl‐2 immunostaining in the cytoplasm of luminal and glandular epithelial cells was moderate in the anoestrus phase. During the early follicular phase, cytoplasmic Bcl‐2 immunostaining was detected mostly in glandular epithelial cells. In the mid‐follicular phase, in glands, the amount of Bcl‐2 protein increased progressively from the superficial to the deep layer. In contrast, the expression of Bcl‐2 decreased in the secretory phase, being very low or absent in the mid‐ and late luteal phases. The overall results suggest that Survivin, Bax and Bcl‐2 proteins may cooperatively contribute to cell apoptosis and cell proliferation in the cat uterus during the oestrus cycle.  相似文献

The thyroid gland of sexually immature dromedary camels was studied using both light and electron microscopy. The thyroid gland contained follicles of different sizes in both summer and winter. However, most of the follicles were large in summer and small in winter. The large follicles were lined by very low cuboidal or semi-squamous follicular cells whereas the small ones were lined by high cuboidal or low columnar follicular cells. Electron microscopy showed that the very low cuboidal follicular cells were poor in organelles and were considered hypoactive. High cuboidal follicular cells on the other hand, were rich in organelles that included mitochondria, cisternae of rough endoplasmic reticulum, secretory vesicles, colloid droplets, heterosomes and autophagic vacuoles; they were considered to be very active. The possible role played by these organelles is synthesis of thyroglobulin and liberation of tri- and tetraiodothyronine is discussed. A few degenerate follicular cells were infrequently encountered in the camel thyroid. Parafollicular (C) cells were not seen in this study either with light or electron microscopy.  相似文献

Syringocystadenoma papilliferum, a hamartoma with mostly sweat gland, but also follicular infundibular elements, is described on the flank and head of two young cats. Clinically, lesions were cutaneous plaques characterized by irregular but sharply demarcated borders and a roughened, hyperpigmented surface. Complete surgical excision in one case was curative. Histologically, the lesion was limited to the superficial dermis and consisted of coalescing units of proliferating sweat glands. The proliferation was tubular or papillary, and may have been epitrichial, opening within dilated hypertrophied follicular infundibuli, or atrichial. Three types of epithelium were observed, recapitulating the formation of the follicular-sweat gland unit with infundibular, ductal and secretory epithelia. The glands reacted positively for alpha cytokeratin 8 and were supported by fibrous tissue with a plasmacytic, lymphocytic and neutrophilic infiltrate. As in humans, this lesion may be classified within the hamartoma-nevus-type category.  相似文献

The distribution of various sugars, that contribute to the composition of the carbohydrate components of the glycoprotein in the ultimobranchial tubule and the thyroid C-cells has been studied, using eight peroxidase-labelled lectins (Dolichos biblorus, Glycine max, Bauhinia purpurea, Helix pomatia, Arachis hipogaea, Ulex europaeus I, Limax flavus and Sophora japonica). The results obtained seem to show that there are receptors for Dolichos biflorus, Glycine max, Bauhinia purpurea, Helix pomatia, Arachis hipogaea, Ulex europaeus I and Limax flavus in the ultimobranchial tubule, suggesting the existence of N-acetylgalactosamine, galactose, L-fucose and sialic acid. However, the C-cells show only a positive reaction with Ulex europaeus I indicating the existence of fucosyl residues in those cells. All these findings support the contention that the ultimobranchial body might contribute decisively not only to the formation of C-cells but also, partially, to the constitution of certain follicular cells.  相似文献

Four domestic rabbits were presented with a history of nonpruritic scale. Multiple skin biopsies revealed findings compatible with sebaceous adenitis as reported in other species: inflammation directed at the sebaceous gland, and/or an absence of sebaceous glands, a perifollicular lymphocytic infiltrate at the level of the absent sebaceous glands, hyperkeratosis, follicular keratosis, follicular dystrophy, perifollicular fibrosis, and a mural infiltrative lymphocytic folliculitis. Histological changes not consistent with sebaceous adenitis in other species were interface dermatitis and interface folliculitis with single cell necrosis and basal cell hydropic degeneration.  相似文献

The thyroid glands of 31 chickens at the age of 17 to 24 months were investigated. Different methods of anatomical preparation, casts of vessels and scanning electron microscopy were used. The thyroid gland of birds is a paired organ. It is located on the ventral surface of the base of the neck within the thoracic inlet. The left thyroid gland is placed more cranially than the right one. Each thyroid gland is closely connected to the common carotid artery on the medial side, from which it is supplied and to the jugular vein on the lateral side. It is a reddish-brown organ and of lenticular profile. The gland measures on average 10 mm in length, 6 mm in width and 2 mm in thickness, and is covered by a thin connective tissue capsule which holds adipose cells. It seems that each thyroid follicle is surrounded by a net of capillaries. The investigation by scanning electron microscopy proved that the follicles are oval with a pyramidal top on each end. The cuboidal epithelium cells leave impressions in the colloid. Epithelium cells carry microvilli on the follicle side surface. Described seasonal changes of the thyroid gland in size and activity were able to be confirmed by the examination of the organ in July and December. In winter the follicular cells were higher and the follicles had a greater volume.  相似文献

On each day of feeding on susceptible calves, salivary glands obtained from groups of adult ticks that transmitted Anaplasma marginale were examined for A marginale colonies by use of light microscopy and transmission electron microscopy. On day 8 of feeding, salivary glands were examined, using fluorescein-labeled antibody and methyl green-pyronine stain. Use of fluorescein-labeled antibody consistently revealed small numbers of fluorescent foci in salivary gland acinar cells obtained from ticks that had fed for 8 days. Colonies of A marginale were seen by transmission electron microscopy only in salivary gland acini of male ticks; these colonies could not be identified, using light microscopy, in companion 1-micron plastic sections stained with Mallory stain. Methyl green-pyronine stain, used commonly to detect theilerial parasites in tick salivary glands, did not differentiate A marginale from cytoplasmic inclusions normally found in salivary gland acinar cells.  相似文献
