The biology of Rhipicephalus bursa has not been thoroughly studied. In the present study R. bursa was bred in the laboratory and its biology worked out.Larvae, nymphs and adult Rhipicephalus bursa ticks were fed on sheep and were held at 28+/-1 degrees C and 89+/-1% relative humidity (RH) in a darkened incubator. The average weight of engorged females was 0.676g (range 0.353-1.128). The average weight of eggs laid by females was 0.344g (range 0.146-0. 554g). The average number of eggs laid was 7527 (range 6263-8860) and this was significantly correlated with the weight of the females. The index of Reproduction Efficiency (number of eggs per g weight of female) was 8469.6. The index of Conversion Efficiency (weight of eggs per weight of female) was 0.5037.Peak daily oviposition, at an average of 840 eggs per female, occurred on the 6th day of oviposition. After Day 6, the rate declined steadily. The average duration of the preoviposition period was 5.1 days (range 4-7 days). The oviposition period averaged 16.5 days (range 12-21 days), and was significantly related to both the weight of the female and the number of eggs laid. The average pre-hatching period was 31.2 days (range 24-45 days). The average feeding time of larvae and nymphs of this two host tick was 15.2 days (range 14-18 days). Female ticks fed for 5.9 days on the average.Heavier nymphs produced females and a shorter premoult period was noted for lighter male nymphs. The males/females ratio was 1.38:1. The time taken to complete the life cycle was on average 142.45 days (range 99-254 days).  相似文献

During investigations into the volatility of female pheromones of A. walkerae in a Y-shaped olfactometer, engorged conspecific male ticks responded weakly both to female-contacted filter paper discs and to intact females enclosed in nylon bags. In tests in which female-contacted filter paper discs were separated from engorged male ticks by means of a screen, no aggregation was observed. However, replete male ticks showed a vigorous response to liquefied air and hexane-trapped volatiles collected from intact females. Guanine showed a potent aggregation attraction for engorged and unfed larvae and 1st and 2nd stage nymphal ticks as well as for male and female imagines of A. walkerae over a wide concentration range. It is concluded that at least 2 pheromones, 1 volatile and 1 non-volatile, are operative in the communication system of this tick species.  相似文献

Babesia spp. infections were investigated in Bos taurus x Bos indicus dairy cows and calves and in Boophilus microplus engorged female ticks and eggs. Blood samples and engorged female ticks were collected from 25 cows and 27 calves. Babesia spp. was detected in ticks by microscopic examination of hemolymph of engorged female and by squashes of egg samples. Cattle infection was investigated in blood thin smears and by DNA amplification methods (PCR and nested PCR), using specific primers for Babesia bovis and Babesia bigemina. Merozoites of B. bovis (3 animals) and B. bigemina (12 animals) were detected exclusively in blood smears of calves. DNA amplification methods revealed that the frequency of B. bigemina infection in calves (92.6%) and in cows (84%) and of B. bovis in calves (85.2%) and in cows (100%) did not differ significantly (P > 0.05). Babesia spp. infection was more frequent in female ticks and eggs collected from calves (P < 0.01) than from cows, especially in those which had patent parasitemia. Hatching rates of B. microplus larvae were assessed according to the origin of engorged females, parasitemia of the vertebrate host, frequency and intensity of infection in engorged female tick, and frequency of egg infection. Hatching rate was lower in samples collected from calves (P < 0.01) than from cows, and in those in which Babesia spp. was detected in egg samples (P < 0.01).  相似文献

At the constant temperature of 25 degrees C and relative humidity (RH) of 84%, the average pre-oviposition period of Hyalomma lusitanicum was 47 days, the oviposition lasted an average of 26 days and the total egg production was an average of 6320 per female. At 16 degrees C the females did not lay eggs at all, but those which survived for 1 year and were transferred thereafter to 25 degrees C and 84% RH laid viable eggs. At 35 degrees C, the oviposition was identical at all levels of RH tested (25, 62 and 93%). At 25 degrees C, the pre-oviposition period was shorter at 93% RH, and the number of eggs laid was fewer at 25% RH. The eggs hatched in 32-40 days, the hatching percentage being lower in batches of eggs laid at the beginning and at the end of the oviposition period. The larval and nymphal moultings were not influenced by the type of host. As the temperature increased, the pre-moult period became shorter. The engorged larvae were more sensitive to the low RH than the engorged nymphs, whose moulting percentage was always greater than 72 in all regimes. Low temperature and high humidity had a favourable effect on the survival of unfed nymphs. The female-to-male ratio was 1:2. Hyalomma lusitanicum always behaved as a 3-host tick. The adults did not engorge on rabbits. The female ticks engorged on calves weighed an average 543 mg. Ticks maintained at 25 degrees C and 84% RH and engorged on calves completed the life cycle in 138-196 days, which does not include the period of chitinization of about 30 days. The present study evaluated the engorgement and drop-off rhythms of Anocentor nitens females feeding on horses. Drop-off rhythm was evaluated at 6h-intervals (06:00, 12:00, 18:00, and 00:00 h) on horses held in stalls or in a pasture. A new method of marking feeding female ticks (the bowknot technique) was developed to evaluate ticks on horses in pasture that attached to different parts of the horse's body. This technique was highly successful, indicating no significant interference on tick engorgement rate or final tick weight, length and reproductive capability. Horses held in the pasture during the summer produced only 28.2% of the tick detachment during the daylight period from 06:00 to 18:00 h. In contrast, 53.4% of the ticks detached during this same 12 h-period during the winter. This difference was probably related to the longer scotoperiod during the winter. Different drop-off rhythms were observed for females attached to different anatomical parts of the horse's body. For example, ticks attached to the ears, perineum, and tail showed similar drop-off patterns, but were different from ticks attached to mane, rump and other body parts. The idiosoma length of the feeding female ticks was individually measured every 6 h until the engorged female detached naturally. The engorgement rate (increase in millimeters of the body length per hour) was evaluated during the last 96 h of parasitism. The highest engorgement rates were observed during the last 24 h of parasitism (approximately 0.16 mm/h), which were four-fold higher than the engorgement rates of the previous 3 days ( approximately 0.04 mm/h), demonstrating that these lower and higher values corresponded to the slow and rapid feeding phases reported elsewhere. Based on these data, the 6 mm idiosoma length was estimated as the minimal length that would correspond to the time point (i.e. 24 h before detachment) during which ticks would undergo the rapid feeding phase and detach as fully engorged females. When this 6 mm length was tested to estimate the number of engorged females detaching from horses in a period of 24 h, the estimated accuracy varied from 58.5 to 97.7% (mean: 73.3%).  相似文献

Two isolates of Beauveria bassiana and one of Purpureocillium lilacinum (=Paecilomyces lilacinus) were found infecting Amblyomma cajennense engorged females collected on horses (0.15% infection rate from a total of 1982 specimens) and another two isolates of P. lilacinum and one Metarhizium anisopliae detected in soils (2.1% from 144 samples) collected in typical pasture habitats of this tick in Central Brazil from October 2009 to March 2011. Fungi were isolated from soils with Rhipicephalus sanguineus as surrogate baits. No fungi were found in ticks or soils during the driest months (May to August). Testing pathogenicity of fungi all R. sanguineus females were killed regardless of the isolate and fungi sporulated abundantly on the cadavers. A. cajennense was less susceptible to infection with P. lilacinum within 20 days than R. sanguineus. All three fungal species probably act as natural antagonists of A. cajennense particularly in the rainy season and have interest for integrate control of vectors of Rocky Mountain spotted fever.  相似文献

Five experiments were carried out on the movement of partially fed male Rhipicephalus appendiculatus ticks on rabbits towards other ticks of the same species. Male ticks pre-fed for 5 days were attracted towards pre-fed females, slightly attracted towards other pre-fed males, but not attracted towards nymphal ticks or recently relinquished feeding sites of adult or nymphal ticks. Female ticks fed for 5 days or longer were particularly attractive to male ticks which had also fed for 5 days or longer. Eight-week-old female ticks were most attractive to males, and 8-week-old male ticks most attracted to females. It is suggested that the male ticks may be attracted to the females by means of a sex pheromone, as has been shown in other ixodid tick species.  相似文献

The durations of the developmental periods of Amblyomma tholloni, recorded at 25°C and 85–87% RH, were longer than those of most other tick species which have been studied under similar conditions. Fully engorged females laid between 2,871 and 10,347 eggs (mean 5,805). Larvae and nymphs attached rapidly and fed successfully on rabbits and sheep. Adults had to be confined on sheep for as long as 14 days before the first attachment occurred and post-attachment mortality was frequently high. Adult females attached in the absence of males, but did not complete feeding. Prefed males were attracted, both on and off the host, by partially fed females and 2,6-dichlorophenol. Extracts of partially fed females were found to contain 2,6-dichlorophenol, which is a commonly occurring sex pheromone in ixodid ticks.  相似文献

Natural tick infestations were assessed every 14 days on horses over a 2-year period. Amblyomma cajennense adult ticks were counted individually, without detachment from the horses. Larvae and nymphs of A. cajennense were collected using a rubber scraper that scratched engorged immature ticks from the host. Adult females of Anocentor nitens larger than 4mm length were counted on the horses. Blood samples were also obtained from the horses every 14 days and macroclimatic data were obtained for the study period. Infestations of A. cajennense demonstrated distinct peaks of activity for each of the three parasitic stages over each 12-month period, showing a 1-year generation pattern. Larvae predominated from April to July and nymphs from June to October. Adults predominated from October to March with a greater number of adult males than females. Although other studies on seasonal dynamics in the states of Rio de Janeiro and Minas Gerais were performed with the free-living stages of A. cajennense on pastures, the present study in the state of S?o Paulo, performed with the parasitic stages of A. cajennense on horses, showed similar results to those observed in other states. Infestations by A. nitens demonstrated distinct peaks of activity of adult females (>4 mm), suggesting different tick generations during the year. Infestation with A. nitens was much higher in the first year than the second year which may have been related to horse nutritional status and stocking rate. Although several climatic variables showed statistical significant correlation (r) with tick counts, the determination coefficients (R(2)) were always lower than 0.40, suggesting that any single significant variable (i.e. mean temperature) would not explain the tick distribution pattern over the year. The highest peaks of A. nitens females (>4 mm) were significantly associated with decrease in horse packed cell volumes (R(2)=0.603). The ears and the perineum, tail and groin region accounted for around 70% of all A. nitens females counted on the horses.  相似文献

The infectivity rate of Babesia equi in the salivary glands of Hyalomma anatolicum anatolicum was assessed. The hungry nymphs were fed on a donkey experimentally infected with B. equi. The engorged dropped-off nymphs were collected at different levels of parasitaemia and kept in BOD incubator. After ecdysis, the hungry adults were prefed on rabbits for different time intervals, thereafter the salivary glands were dissected out and acini were examined after methyl green pyronin (MGP) staining. A total of 134 male and 139 female ticks were dissected out. Average infected acini per tick were found to be significantly higher (p<0.05) in male as compared to the female ticks. Further, maximum infected acini in both male and female ticks were found at 24h of prefeeding on rabbits and overall infected acini per tick increased with rise in parasitaemia. The release of infected ticks on susceptible donkeys resulted in development of clinical babesiosis.  相似文献

As in most parts of the world where the cattle tick Boophilus microplus is established, resistance of ticks to acaricides occurs in New Caledonia. In order to implement laboratory resistance tests on larvae, engorged females collected in suspected farms are necessary. Investigations on the detachment schedule of the engorged females were conducted to explain certain field situations such as the lack or scarcity of engorged females on highly infested cattle driven from the pasture to the pen in the morning. Three experiments on Charolais steers naturally infested on pastures showed that: (1) engorged female burdens at sunrise are similar whether the steers spend the night in pasture or in a pen; (2) compared with steers maintained in a pen, morning detachment of females increases when the steers stay on the pasture or move from the pasture to the pen; (3) detachment rhythm of engorged females on steers staying the morning in a pen, is not influenced by feeding activity or exposition of steers to sun; (4) detachment occurs earlier for females attached on anatomical sites exposed to sun, and earlier from these sites for the steers in pasture or walking than for steers in a pen.  相似文献

Partially engorged adult Hyalomma excavatum ticks infected with Theileria annulata were reared for the preparation of an infective suspension. About 6000 partially engorged infected adults were obtained from 420 non-infective adult ticks. The entire cycle required about 4 months. Six rabbits were used for the production of the fully engorged females. The larvae were fed on 180 gerbils, the nymphs on 17 infected calves and the partially fed adults on 22 goats, 7 sheep and 7 rabbits The adult ticks did not show any host preference.  相似文献

Laboratory animals exposed to feeding ticks develop resistance which is reflected by a decline in tick engorgement weight, egg-laying by adults and reduced egg viability. Serum antibodies from these hosts and their reaction with tick antigens have been detected by different methods, including precipitation techniques, immunofluorescent techniques, ELISA and Western blots. However, little is known about the effects of antibodies on ticks that engorge on resistant hosts, or which tissues of the tick body are possibly immunogenic. Some researchers, using immunohistochemistry, have detected host antibodies in the gut, salivary glands and haemolymph of ticks engorged on resistant animals. The same technique has helped considerably in determining antigenic sites or antibody targets in other arthropods. Consequently, immunohistochemistry techniques were used in this study to detect cross-reactivity between sera raised against Amblyomma cajennense (Fabricius, 1787) with Amblyomma hebraeum (Koch, 1844), and vice versa. The results show the existence of shared antigens between the 2 tick species. In general, our results point more to a 1-way cross-reactivity of A. hebraeum with A. cajennense than a reciprocal cross-reactivity, suggesting that A. hebraeum is more immunogenic than A. cajennense.  相似文献

Transmission of Hepatozoon spp. to dogs was investigated using four species of ixodid ticks: Rhipicephalus sanguineus, Amblyomma aureolatum, Amblyomma ovale and Amblyomma cajennense. We collected completely or partially engorged adult ticks of these species from dogs that were naturally infested and positive for Hepatozoon spp. We selected some of these ixodids and inoculated them orally in four negative dogs. The other ticks were dissected and examined for oocysts. Of all dogs inoculated orally with R. sanguineus, A. aureolatum, A. cajennense and A. ovale, only the animal that received the macerate of A. ovale was positive; evidence (gametocytes in peripheral blood) of infection was found 63 days after inoculation. Among all dissected ticks, we found only two oocysts; these were similar to those of Hepatozoon canis, and both were recovered from a single A. ovale specimen. We inoculated sporozoites recovered from the oocysts intraperitoneally into a Hepatozoon spp. negative dog, and circulating gametocytes were detected 84 days later. Our study demonstrated that A. ovale can be a vector of Hepatozoon spp. in Brazil.  相似文献

为了评估镰形扇头蜱肌钙蛋白I(TroponinI,TnI)作为候选疫苗分子的潜力,应用肌钙蛋白I重组蛋白进行了抗蜱免疫实验。将重组蛋白按常规方法免疫新西兰兔,三次免疫后分别用若蜱、成蜱进行攻虫实验,观察其免疫保护效果。结果发现蜱的吸血行为受到一定的影响,与对照相比,若蜱和成蜱吸附率、饱血率、饱血体重和死亡率有显著差异,而在饱血时间上差异不显著,说明TnI基因的重组表达产物具有一定的免疫保护作用。  相似文献

Some aspects of the biology of the tick Hyalomma anatolicum fed on rabbits, sheep and goats were studied. The non-feeding stages were maintained under laboratory conditions at 20-36°C and 75% relative humidity. The longest feeding periods of larvae and nymphs of H. anatolicum were observed when fed on rabbits (mean 4.58 ± 0.51 and 7 ± 1.15 days, respectively) while the longest feeding periods of females were observed on goats (9.61 ± 1.21). The pre-oviposition period (4.8 ± 0.42 days) and pre-eclosion periods (mean 21.3 ± 1.16 days) were shortest for females fed on rabbits. Engorged females reached heavier engorgement weights (482.92 ± 88.08 mg), and produced more eggs (4881.8 ± 842.71) when fed on rabbits. However, no significant differences were observed between the percentages hatchability of eggs laid by ticks fed on the three hosts studied. Most (94.31%) of the larvae fed on rabbits underwent a 2-host life cycle, while few (5.69%) of them behaved as a 3-host ticks. Few larvae were able to complete feeding as 3-host pattern on both sheep and goats, while the majority of the larvae failed to complete feeding or died on their way to molt on both sheep and goats.  相似文献

试验旨在了解铁蛋白1(ferritin 1,Fer1)基因在边缘革蜱各发育龄期、吸血期及不同器官中的表达情况。用实时定量荧光PCR法分析了Fer1基因在边缘革蜱各发育阶段、吸血期和不同器官中的相对表达情况,以饥饿雌蜱为基准(1 FC),用2-△△Ct法计算结果。结果显示,雌性蜱在开始吸血后第72 h Fer1基因的相对表达量最高,在刚刚孵化的饥饿成蜱(雌、雄)、卵及幼蜱中相对表达量较低;在若蜱中相对表达量较高,其中,幼蜱和若蜱在饥饿状态下比同时期饱血蜱的相对表达量高。在雌蜱的不同器官中,以饱血蜱唾液腺为基准(1 FC),半饱血蜱中肠的表达量最高,在卵巢和马氏管不表达;在饱血蜱唾液腺、中肠、卵巢和马氏管中均表达,其中,在卵巢和马氏管中的表达量较高,中肠次之,唾液腺中的表达较低。试验结果表明,边缘革蜱体内Fer1基因的相对表达量随着蜱吸血过程变化而变化,这与蜱体内消化细胞的完善及消化酶在该过程中的变化相关,可为该类蜱铁代谢通路中关键功能基因的研究提供参考。  相似文献

A study was made to assess the influence of host response on Haemaphysalis punctata and Ixodes ricinus biology by comparing the effects of feeding ticks on previously uninfested hosts with those fed on previously sensitised hosts. Adult H punctata were fed on sheep; larvae and nymphs of H punctata and I ricinus were fed on rabbits. The engorgement weights of all instars studied that fed on sensitised hosts fell very gradually with successive infestations. These weight reductions were greater at high than at low levels of infestation. In previously sensitised hosts, the engorgement period of all instars was prolonged: the percentage of instars which completed engorgement and engorgement weights were reduced. The host's clinical and immune response markedly affected the fecundity of adult female ticks and the viability and hatchability of the laid eggs. Eighty per cent of female H punctata successfully laid eggs following the first infestation and the hatchability of eggs was 85 per cent. The numbers successfully laying eggs decreased to 60 per cent during the fifth and sixth infestations and was less than 20 per cent following the seventh infestation; the hatchability decreased similarly. There was no marked effect on the development of immature instars when fed on sensitised rabbits.  相似文献

The objective of this study was to investigate some aspects of horse resistance to natural infestations of Anocentor nitens and Amblyomma cajennense over a 2-year period. Free-living stages of A. nitens were used to evaluate the influence of season of the year on horse resistance. Every 2 weeks, 10 selected engorged females, weighing 150-329 mg, were removed from naturally infested horses and were individually placed into glass tubes closed with a cotton stopper, and maintained in an incubator. The biological parameters evaluated varied according to the season of the year, i.e., reproductive efficiency, maximum larval survival (MLS) and egg-hatching rates were significantly lower in spring/summer than in fall/winter. At 14-day intervals, females of A. nitens and A. cajennense were counted on each side of the body of five mares. A comparison between the animal with the lowest infestation and the animal with the highest infestation showed greater differences in animals infested with A. nitens than those infested with A. cajennense. Ten animals were found to carry 64.9% of the total parasite burden of A. cajennense and 88.6% of the highest infestations of A. nitens females. A seasonal variation on intensity of horse resistance to A. nitens was observed during the study. However, this was not observed for A. cajennense. Cross-resistance was observed in animals with the highest total number of A. cajennense and the highest number of A. cajennense adults (r=0.817), larvae (r=0.756), and A. nitens females (r=0.799). These results led to the conclusion that changes in horse resistance to A. nitens occur seasonally. As far as the total population of A. cajennense and the female stage of A. nitens are concerned, horses develop cross-resistance.  相似文献

A single-step duplex polymerase chain reaction (PCR) technique and traditional microscopic examination of haemolymph smears were used to detect Babesia bigemina and/or Babesia bovis infection in engorged female ticks of Boophilus microplus recovered from calves raised in an endemic area of the State of Minas Gerais, Brazil. In the PCR amplification of tick-derived DNA, pairs of oligonucleotide primers specific for a 278-bp sequence from B. bigemina and for a 350-bp sequence from B. bovis were used conjointly. The microscopic examination of haemolymph revealed that 16.7% of the engorged ticks were infected with Babesia spp., although no significant differences (rho > 0.05) were found in the infection rate of ticks collected from calves of different age groups. PCR analysis showed that 77.8% of the engorged ticks whose haemolymph contained sporokinetes were infected with B. bigemina, 7.8% with B. bovis and 14.4% with both protozoan species. However, the PCR assay further revealed that, amongst the engorged female ticks whose haemolymph was apparently negative for the presence of sporokinetes, 15.6% were infected with B. bigemina, 2.2% with B. bovis and 10.0% with both species. The duplex PCR method is thus more efficient and sensitive than the microscopic assay and also permits facile identification of the protozoa species present in engorged female ticks.  相似文献
