The study was undertaken to determine the modulation in innate immune response of rohu (Labeo rohita) during experimental freshwater lice Argulus siamensis infection. Results showed that serum α‐2 macroglobulin (α‐2M) activity, ceruloplasmin level and alternative complement activity were significantly (P<0.05) lower in fish at different degrees of lice infection in comparison with uninfected control. No significant difference (P>0.05) in haemagglutination titre was observed in fish with low‐ and high‐degree lice infections as compared with uninfected control. The serum lysozyme level was significantly (P<0.05) lower in low degree of lice infection as compared with control fish. The total serum antiprotease, myeloperoxidase activity and total protein level were not significantly different (P>0.05) in different degrees of lice‐infected fish with respect to the control fish. The study indicated that A. siamensis infection modulated the immune system of rohu by suppressing the α‐2M, serum complement activities and ceruloplasmin level and through induction of stress response. The baseline data obtained in the present study have tremendous importance in understanding the susceptibility of rohu to different degrees of parasitosis and might be useful in controlling this dreaded ectoparasitic infection in fish.  相似文献

This study was carried out to investigate the effects of dietary pyridoxine (PN) on the antioxidant status, apoptosis, intercellular integrity and immune function of head kidney and spleen in young grass carp (Ctenopharyngodon idella). Results showed that compared with the optimal PN level, (a) PN deficiency decreased antioxidant enzyme activities and down‐regulated the mRNA levels of antioxidant enzymes, NF‐E2‐related factor 2 (Nrf2), myeloid cell leukaemia‐1 (Mcl‐1) and tight junction proteins, whereas it increased the contents of reactive oxygen species (ROS), malondialdehyde (MDA) and protein carbonyl (PC), and up‐regulated the mRNA levels of cysteinyl aspartic acid‐protease (caspase), Fas ligand (FasL), p38 mitogen‐activated protein kinase (p38MAPK) and myosin light chain kinase (MLCK) in the head kidney and spleen of fish (p < .05). (b) PN deficiency decreased lysozyme (LZ) and acid phosphatase (ACP) activities, and complement contents, and down‐regulated the mRNA levels of antibacterial peptides, anti‐inflammatory cytokines, inhibitor of κBα (IκBα) and target of rapamycin (TOR), whereas it up‐regulated the mRNA levels of pro‐inflammatory cytokines and nuclear factor kappa B p65 (NF‐κBp65) in the head kidney and spleen of fish (p < .05). (c) Dietary PN requirements of grass carp based on the malondialdehyde content and lysozyme activity were estimated as 4.97 and 4.99 mg/kg diet, respectively.  相似文献

Parasite species often show a heterogeneous, highly dispersed pattern of infestation within hosts. Varieties of factors including morphological, physiological, immunological and nutritional characteristics affect the infestation level of a specific parasite in homogenous pray. Limited attempts, however, have been made to explore such underlying drivers of infestation pattern. Here, three stages of Labeo rohita (fingerling, juvenile and pre‐adult) were challenged with ectoparasite, Argulus siamensis in same aquaria. The parasite load on individuals was determined at 5‐day interval for 1 month. The load was found to be highest in pre‐adult stage followed by juveniles and fingerlings. On day 20 post infection, the load of parasite on pre‐adult fish was high along with detectable skin damages. Skin tissues were collected for immune gene expression analysis and histopathology. Histological studies showed increased melanization in the dermis and mild inflammatory cellular reactions in pre‐adult fish whereas, massive subcutaneous myositis with engorged blood vessels were observed in fingerlings. The expression levels of various inflammation and innate immune‐related genes viz., interleukin (IL)‐8, IL‐10, IL‐11, IL‐15, natural killer enhancing factor, toll‐like receptor 4, apolipoprotein A–I and immunoglobulin Z were significantly high in skin samples of infected fingerlings as compared to other two growth stages or controls of each stage. On the other hand, the expression of immunoglobulin M was down‐regulated in all infected samples as compared to their respective controls. The results thus depict that local immuno‐inflammatory response plays a significant role in determining susceptibility of host in intra‐specific group, and has important implications for ecology and aquaculture.  相似文献

This study evaluates the effects of the dietary administration of the live yeast Debaryomyces hansenii strain L2 on the immune responses of gilthead seabream for 4 weeks. Cellular immune parameters were measured from serum and head‐kidney leucocytes respectively. The expression levels of immune‐associated genes were quantified by real‐time polymerase chain reaction. In addition, the profile of intestinal microbiota was studied by denaturing gradient gel electrophoresis. The results showed that seabream fed a diet containing D. hansenii had significantly increased cellular immune parameters. The yeast‐supplemented diet up‐regulated the expression of most seabream genes at week 2 and down‐regulated all of them at week 4, except in the head‐kidney. Finally, a reduction in the diversity of the intestinal microbiota was detected in those specimens receiving the yeast‐supplemented diet. These results support the idea that the live yeast D. hansenii strain L2 stimulates the immune system of gilthead seabream.  相似文献

The research was aimed to assess the effect of dietary carvacrol (0.025% and 0.05%) on sea bass (Dicentrarchus labrax) growth, immune response and resistance to Listonella anguillarum. Fish (69.2 ± 0.22 g) were fed the experimental diets for 9 weeks. Dietary carvacrol did not negatively affect fish survival, growth performance, feed intake and feed conversion ratio (> 0.05) nor carcass yield and viscerosomatic, hepatosomatic and mesenteric fat index (P > 0.05). Serum and head kidney leucocytes were collected after 1, 4 and 8 weeks of feeding. Carvacrol significantly reduced serum proteins, immunoglobulins and lysozyme activity (< 0.01) and moderately increased phagocytosis and pinocytosis of head kidney macrophages. The release of reactive oxygen species by leucocytes was reduced in carvacrol‐fed fish, even if significantly (< 0.05) only in those fed 0.05% carvacrol for 1 week. Dietary carvacrol did not significantly affect the aspecific immune response, although a potential antioxidant activity might be speculated. Moreover, feeding carvacrol provided an appreciable resistance to a challenge with L. anguillarum, when a bacterial dose lower than the Lethal Dose50 was used. Cumulative mortality in fish fed 0.025% carvacrol was significantly lower than that of untreated controls (75% Relative Per cent Survival).  相似文献

This study evaluated the effect of sublethal doses of antifungal drug miconazole nitrate (MCZ) on immunological responses and its role as a prophylactic drug against S. parasitica in Labeo rohita fingerlings. Fish were fed with sublethal doses of MCZ, that is, T1—6.30 mgMCZ kgBW?1, T2—12.61 mgMCZ kgBW?1 and T3—25.22 mgMCZ kgBW?1, and sampling was done at different time intervals for 240 hr. Immunological parameters viz. lysozyme activity, oxygen radical production and plasma antiprotease activity showed significant enhancement (< 0.05) in fish fed with T2 and T3 doses. Expression of immune‐relevant genes such as TLR‐22 and β2‐M showed significantly higher expression at 6 hr and 24 hr of sampling in both liver and head kidney. However, these genes showed a downregulation after 120 hr of sampling in both the tissues. Preventive efficacy study showed that single dose of MCZ provides protection against oomycetes up to the fourth day of infection. Significantly higher mortality was observed in control diet‐fed fish as compared to fish fed with MCZ medicated diet. Thus, it can be concluded that the MCZ can act as a potent antifungal agent for preventing oomycetes infection as well as to enhance the immune response.  相似文献

Toll‐like receptors (TLRs) play an indispensable role in fish immunity, being involved in pathogen recognition and the triggering of immune reactions. Here, a member of the TLR family, TLR1, from Lateolabrax japonicus was characterized and its expression pattern and intracellular localization were analysed. The full‐length LjTLR1 cDNA (2,755 bp) was found to encode a polypeptide of 827 amino acids. The deduced amino acid sequence contained three main structural domains: an extracellular leucine‐rich repeat domain, a transmembrane domain and a Toll/IL‐1 receptor domain. Tissue distribution analysis indicated that LjTLR1 was expressed in all of the examined tissues to varying degrees, with the highest levels being measured in the head kidney. In order to assess the antibacterial functions of LjTLR1 during infection, the pathogenic bacteria Vibrio harveyi and Streptococcus agalactiae were used. LjTLR1 was significantly upregulated in the three immune organs (the head kidney, spleen and liver) following bacterial stimulation, and its expression was detected 6 hr after initial exposure. In mRNA in situ hybridization experiments, positive signals were more numerous in the treatment group than the control group, verifying the expression patterns observed. Assessment of the intracellular localization of LjTLR1 revealed it to be present in the cytoplasm. These results indicate the potential role of LjTLR1 in immune responses to bacterial infection. This study enriches our knowledge of L. japonicus immune genes and provides a theoretical basis for further research concerning the antibacterial functions of fish TLRs during infection.  相似文献

A 120‐day feeding trial was conducted to assess the effects of dietary supplementation of zinc oxide (ZnO) and selenium (Se) nanoparticles (NPs) on growth, immunological and enzymatic profiles in rohu, Labeo rohita. Fishes were fed with basal diet incorporated with laboratory‐synthesized nanoparticles, viz., zinc oxide nanoparticles—10 mg/kg and Se nanoparticles—0.3 mg/kg throughout the experimental period. At an interval of every 30 days, treated and control fishes were randomly collected and subjected to estimation of growth, non‐specific immune parameters and serum enzyme assays. It was observed that there was significant (p < 0.05) increase in growth and non‐specific immune parameters like respiratory burst, lysozyme and myeloperoxidase activities as compared to the control group (only basal diet). Serum enzymatic profiles such as lactate dehydrogenase and alkaline phosphatase activities down‐regulated (p < 0.05) in treated group compared to control group, and superoxide dismutase and acetylcholine esterase activity up‐regulated in treated group. The relative percentage survival (RPS) found to be significantly higher in treated groups (60.00 ± 8.82%) as compared to control group (45.00 ± 6.17%) while challenged with virulent bacterial strain Aeromonas hydrophila. These results indicated that dietary supplementation of zinc oxide (ZnO) and selenium (Se) nanoparticles stimulates immunity and enhances resistance to bacterial infection in L. rohita.  相似文献

Using plant extracts as immunostimulants in aquaculture has prove effectiv in resisting infectious disease, in addition to being safe and inexpensive. The present study is concerned about the prospective mode of action of using lesser galangal extract (Alpinia officinarum) encapsulated into amine surface functionalized mesoporous silica nanoparticles (MSN) in order to elevate the immune status of rainbow trout (Oncorhynchus mykiss). Fish were randomly divided into 6 groups and fed for 4 weeks with commercial diets supplemented with 0% (control), 0.5% and 1.5% of lesser galangal extract, and with previous concentrations encapsulated with MSN (0.5% + MSN, 1.5% + MSN) and MSN (without extract). The effects of the feeding trial on the innate humoural immune parameters (total protein, myeloperoxidase content, lysozyme antiproteases and bactericidal activities) in rainbow trout were examined. Also, the influence of dietary supplement on some immune‐related genes in rainbow trout head kidney (TNF, IL‐8, IL‐1b, LYZ2 and CD4) after challenge with Yersinia ruckeri was examined. The results revealed enhancement in immune parameters in all treatment groups compared to the control, especially in the fish group fed with 1.5% + MSN which showed the highest significant difference (p < .05) in total protein, lysozyme and antiproteases activities. Also, feeding lesser galangal extract encapsulated into MSN led to an increase in myeloperoxidase content and bactericidal activity. An improvement in the expression of immune‐related genes has been recorded in fish groups fed doses of lesser galangal extract and lesser galangal extract encapsulated into MSN compared to the control or to the group fed with MSN only. Particularly, the group fed with 0.5% + MSN showed a significant up‐regulation in most of the immune‐related genes. The current investigation supports using lesser galangal extract encapsulated into MSN in fish diets as a supplement to enhance the immune response of rainbow trout and elevate its resistance to infectious diseases.  相似文献

The immune response in rainbow trout fry against Flavobacterium psychrophilum was elucidated using an immersion‐based challenge with or without prior exposure to hydrogen peroxide (H2O2). Samples were taken from the head kidney 4, 48, 125 and 192 h after immersion, and the regulation of several genes was examined. Bacterial load was assessed based on the presence of 16S rRNA and correlated with gene expression, and the levels of specific antibodies in the blood were measured 50 days post‐infection. Separately, both H2O2 and F. psychrophilum influenced gene expression, and pre‐treatment with H2O2 influenced the response to infection with F. psychrophilum. Pre‐treatment with H2O2 also affected correlation between gene regulation and pathogen load for several genes. A delay in antibody production in H2O2‐treated fish in the early phase of infection was indicated, but H2O2 exposure did not affect antibody levels 50 days post‐infection. An increasing amount of F. psychrophilum 16S rRNA was found in the head kidneys of infected fish pre‐treated with H2O2 relative to the F. psychrophilum group. The results show that a single pre‐treatment with H2O2 impairs the response against F. psychrophilum and may intensify infection.  相似文献

Multispecies herbs and probiotic bacteria offered to fish might enhance their immune response and increase disease resistance, but the dose effects of herbal‐probiotic application remain unclear. Therefore, the effect of herbal‐probiotic mixtures of traditional Chinese medicine (TCM) of composition Astragalus membranaceus, Angelica sinensis, Crataegus hupehensis and probiotic Bacillus species (BS) of composition Bacillus subtilis and Bacillus lincheniformis as natural immunostimulants in tilapia Oreochromis niloticus have been investigated. Fish were randomly divided into four groups: control diet (CT), TCMBS1 [TCM at 3 g/kg and BS at 7 g/kg], TCMBS2 [TCM at 5 g/kg and BS at 5 g/kg], TCMBS3 [TCM at 7 g/kg and BS at 3 g/kg]. Tilapia in the TCMBS3 group showed significant improvement in weight gain, specific growth rate, and lowered feed conversion ratio compared with other treated groups and the control. Concerning immune indexes, all treated groups significantly enhanced lysozyme, superoxide dismutase, catalase, protease and antiprotease activities, with highest values in catalase and antiprotease activities in TCMBS3 compared with control. TCMBS3 demonstrated higher expression of βeta‐defensin, lysozyme, heat shock protein 70, catalase and transforming growth factor‐βeta compared with other treated groups or the control group in both mid‐intestines and head‐kidney. After challenge with Streptococcus agalactiae, the best survival was found in TCMBS3 (97%), followed by TCMBS2 (73%), and TCMBS1 (69%) compared with the CT (35%). Collectively the present results suggest that TCMBS3 dose might potentiate a higher immune response and disease resistance in fish.  相似文献

The effects of dietary administration of inorganic zinc (zinc sulphate, ZnSO4) and nano zinc (zinc oxide nanoparticles, ZnO‐NP) were evaluated in rohu, Labeo rohita fingerlings. Fish were fed with a basal diet (Control) supplemented with ZnSO4 (T1, T2 and T3) and ZnO‐NP (T4, T5 and T6) at 10, 20 and 30 mg/kg, respectively, for a duration of 45 days. The results revealed that fish fed diet containing 20 mg ZnO‐NP per kg (T5) had the highest weight gain and specific growth rate (SGR, % per day), which was significantly different (p < .05) from the other experimental diets. Significantly (p < .05), higher activities of the digestive and metabolic enzymes were recorded in the fish fed ZnO‐NP containing diets as compared to the diets containing inorganic Zn or control diet. The maximum serum glucose and protein levels were noted in fish reared on diet T5. Both SGOT and SGPT activities were significantly increased in fish fed Zn‐supplemented diets (T1 to T6), as compared to the control group. Similarly, innate immune parameters were improved with feeding Zn incorporated diets. The highest phagocytic (40.74 ± 0.65%) and respiratory burst (0.33 ± 0.001, OD 630nm) activities were recorded in the fish fed diet containing ZnO‐NPs at 20 mg/kg (T5). The maximum superoxide production and serum peroxidase activity were detected in the fish fed T5 and T6 diets. Overall, results indicated that short‐duration feeding (≤45 days) of dietary ZnO‐NP (20 mg/kg) improved growth, enzyme activity, serum biochemical parameters and immune function in rohu fingerlings.  相似文献

Mucins are large glycoproteins that cover epithelial surfaces of the body and play important roles in prevention of inflammatory and various infectious diseases. In this study, five membrane‐bound and seven secreted mucin genes in the channel catfish were identified. All these identified mucin genes possess at least one PTS, von Willebrand D (VWD) or SEA domains. The expression of the 12 mucin genes in channel catfish was first studied with infection of Edwardsiella ictaluri and Flavobacterium columnare. Expression difference in MUC13a, MUC13, MUC2 and MUC5b was found in the intestine after E. ictaluri infection. Eight mucin gene expressions (except MUC3a, MUC2, MUC4 and MUC5f) were up‐regulated at 4 hr and down‐regulated after 24 hr in the gill with F. columnare infection. Expression level of MUC2 gene was up‐regulated in the intestine with E. ictaluri infection without no significant change in the gill under the F. columnare infection, which indicate that MUC2 is tissue‐specific gene expression and has different immune respond to two bacterial challenge. Taken together, the study showed mucin from the gill by F. columnare challenge induced an obvious response than mucin from the intestine with E. ictaluri infection.  相似文献

The aim of this study was to illustrate the effects of dietary loquat leaf extract (LLE) on the expression of some intestinal cytokines as well as serum innate immune parameters in common carp (Cyprinus carpio) fingerlings. The fish were fed with experimental diets supplemented with 0 (control), 0.25% (0.25LLE), 0.5% (0.5LLE) and 1% (1LLE) LLE over a 7‐week period. At the end of the trial, the serum lysozyme (Lys) activity, ACH50, total Ig as well as the expression of IL1B, IL8, TNF‐alpha, LYZ and TGF‐β in intestine were evaluated. The results showed that administration of 0.5% or 1% LLE significantly increased serum total Ig. However, in case of serum Lys activity significant elevation was observed just in fish fed 0.5% LLE. Also, supplementation of diet with LLE significantly increased ACH50 compared to the control group, regardless of inclusion levels. Gene expression studies revealed upregulation of TNF‐alpha, IL1B, IL8 and LYZ in intestine of fish fed LLE. However, the effects varied dependent on LLE levels and the tested immune related gene. Also, in case of TGF‐β significant downregulation was observed just in 1% LLE treatment. In conclusion, dietary LLE supplementation significantly upregulated immune related genes in intestine and improves innate immune responses. Altogether, LLE can be recommended as fish immunostimulant in early stage of carp culture.  相似文献
