Vibrio anguillarum is the causative agent of vibriosis, a deadly haemorrhagic septicaemic disease affecting various marine and fresh/brackish water fish, bivalves and crustaceans. However, the diversity and virulence mechanisms of this pathogen are still insufficiently known. In this study, we aimed to increase our understanding of V. anguillarum diversity and virulence through comparative genome analysis of 15 V. anguillarum strains, obtained from different hosts or non‐host niches and geographical regions, among which 10 and 5 strains were found to be virulent and avirulent, respectively, against sea bass larvae. First, the 15 draft genomes were annotated and screened for putative virulence factors, including genes encoding iron uptake systems, transport systems and non‐ribosomal peptide synthetases. Second, comparative genome analysis was performed, focusing on single nucleotide polymorphisms (SNPs) and small insertions and deletions (InDels). Five V. anguillarum strains showed a remarkably high nucleotide identity. However, these strains comprise both virulent and avirulent strains towards sea bass larvae, suggesting that differences in virulence may be caused by subtle nucleotide variations. Clearly, the draft genome sequence of these 15 strains represents a starting point for further genetic research of this economically important fish pathogen.  相似文献

Orange‐spotted grouper (Epinephelus coioides) is one of the most important marine food species in Southeast Asia and China and has been cultured for decades. In this study, we fully utilized the limited capacity of semiconductor sequencing, the high efficiency of long‐range PCR for target enrichment and a non‐indexed pooling strategy to screen single‐nucleotide polymorphisms (SNPs) in a breeding population of orange‐spotted grouper. Forty‐one genomic DNA fragments, with a total length of approximately 180 kb, including 22 candidate genes that control growth, and from a DNA pool of 20 heaviest and 22 lightest individuals of the sampled population were successfully sequenced using an Ion Torrent Personal Genome Machine. 3 503 466 clear reads were produced with a length of 192 ± 56 bp, 86.8% of which were mapped to the reference with an average coverage depth of 2567‐fold and physical coverage of 98.8%. Finally, 1623 high‐quality SNPs were adopted. Compared with Sanger sequencing of three random common regions, the sensitivity and specificity of our approach were 39.4% and 100.0% respectively. A mutation located at the third position of the previously labelled start codon of growth hormone receptor type 1 invalidated the start codon. Furthermore, comparison of the frequencies of genotypes and alleles of this site between the two extreme groups, prediction of signal peptide and identification of conservative mRNA sequences suggested that the functional start codon is likely located at the position of another downstream in‐frame ATG in the mutant. These detected SNP markers will provide important tools for the selective breeding of orange‐spotted grouper.  相似文献

The effects of fulvic acid (FA) on survival and immune‐related gene expression were investigated in Litopenaeus vannamei challenged with Vibrio parahaemolyticus by immersion. Shrimp were fed with different dietary FA concentrations (1, 2, 4 and 6 g/kg feed) for 20 days (first bioassay) or 8 days (second bioassay, 2 g/kg feed of FA added every 2 days) and then challenged with V. parahaemolyticus. In a third bioassay, the expression of three immune‐related genes (translationally controlled tumour protein [TCTP], superoxide dismutase [SOD] and heat‐shock protein 70 [HSP70]) in haemocytes or hepatopancreas of experimental shrimp was measured by real‐time quantitative PCR at 0, 6, 12, 24, 48, 72 and 96 hr after FA (2 g/kg feed) administration. Fulvic acid increased survival at a concentration of 2 g/kg feed supplied every two days. Interestingly, TCTP gene expression was upregulated, whereas gene expression of SOD and HSP70 was downregulated. In conclusion, dietary fulvic acid improves survival in white shrimp challenged with V. parahaemolyticus and modulates the immune response. Therefore, FA merits further evaluation as prophylactic treatment in commercial shrimp farms.  相似文献

Endocrine disrupting chemicals (EDCs) are of substantial concern when contaminating waters. We studied gene expression of larvae from river pufferfish, Takifugu obscurus exposed to EDCs. We cloned the full‐ or partial‐length cDNA sequence of genes related to the cortisol pathway, such as the glucocorticoid receptor (GR), mineralocorticoid receptor (MR), heat shock protein (HSP) 70 and 90, phosphoenolpyruvate carboxykinase (PEPCK) and the FK506‐binding protein 4 (FKBP52). The tissue and time‐dependent expression of mRNA was studied by real‐time PCR in T. obscurus exposed to two representative EDCs (bisphenol‐A and 4‐tert‐octylphenol). HSP70, HSP90, MR and FKBP52 mRNA expression level was much higher than GR and PEPCK expression. GR mRNA expression level was significantly upregulated after 48 h in both EDC‐treated fish groups compared with the control. Both groups also showed down‐regulation patterns after 48 h. The initial expression of PEPCK in both groups was down‐regulated after 24 h. In the bisphenol‐A treated group, the expression of FKBP52 showed substantially high up‐regulation from 6 to 48 h after exposure, and then strikingly reduced its expression level at 72 h. Our results provide insights into the EDCs metabolizing system of T. obscurus and offers baseline information for further research related to the possible use as a bio‐indicator of this commercially important aquaculture fish species.  相似文献

Harmful algal blooms (HABs) are natural phenomena with different effects on the aquatic environment that affect both human economy and health. Several genomic studies have been done to characterize the effects of contaminants on Crassostrea gigas. However, oysters’ molecular response to HABs exposure needs to be studied more. In this study, we challenged C. gigas with Prorocentrum lima, a diarrhoetic toxin producer, under controlled experimental conditions considering dinoflagellate density and exposure time (acute and sub‐chronic) as variables. The expression profile of six stress response genes was analysed by semiquantitative RT‐PCR: Glutamine synthetase (GS), Glutathione S‐transferase (GST), heat shock protein 70 (HSP70), and heat shock protein 90 (HSP90), CuZn superoxide dismutase (SOD) and melanogenic peroxidase (POX). The results revealed that these gene expressions depend on exposure time and cell concentration. In general, an increased expression was observed in all tested cell densities as immediate response to exposure (0–3 h); whereas lower expression levels than control were observed after 6 h. Treatment with 3,000 cells mL‐1 promoted higher expression than control on most genes after 336 h of exposure. This is the first report providing information at molecular level on C. gigas response to dinoflagellate blooms.  相似文献

In this study, we evaluated the growth, osmoregulation and energy metabolism of the oriental river prawn, Macrobrachium nipponense, reared during 6 weeks with different salinities (0, 8, 14 and 22 g/L). The results showed that the haemolymph osmolality of M. nipponense increased with an increase in ambient osmotic pressure; the isosmotic point was 490 mOs/kg H2O. The prawns showed a higher survival rate, weight gain rate and hepatopancreas index in salinity 14 g/L. Digestive enzymes were all affected by salinity, and the highest activities were observed in the salinity 14 g/L. The mRNA expression of Na+‐K+‐ATPase in gills and p53 in hepatopancreas were the highest in salinity 22 g/L. The expressions of heat shock protein 90 and glutathione S‐transferase genes in hepatopancreas were significantly higher in the salinity 8 g/L. Lipid metabolism‐related genes in hepatopancreas were significantly expressed in the salinity 14 g/L. The glucose‐6‐phosphatase gene in hepatopancreas was highly expressed in the salinity 8 and 22 g/L, and the expression of the ecdysone receptor gene in hepatopancreas was significantly higher in the salinity 14 g/L. The results showed that salinity 14 g/L could promote the growth of M. nipponense. However, higher salinity conditions may cause physiological damage, which provides a theoretical basis for brackish water culture of M. nipponense.  相似文献

Elucidation, through molecular analyses, of bacterial afflictions in commercially important aquaculture‐reared shrimps is pivotal for the prevention and/or control of disease outbreaks. In this study, we examined the phylogenetic relatedness and compared the possible immune‐related functional roles of both translationally controlled tumour protein (TCTP) and ferritin genes with previous studies. Both TCTP and ferritin genes were substantially upregulated in the Indian white prawn, Fenneropenaeus indicus (H. Milne Edwards), post‐larvae following bath challenge with the virulent strain of bacteria, Vibrio harveyi D3. Full‐length cloning of these genes by rapid amplification of complementary DNA ends ‐polymerase chain reaction (RACE‐PCR) yielded 727‐base pair (bp)‐long TCTP and 1212‐bp‐long ferritin gene sequences. Their open reading frames (ORFs) were 507 and 510 bp, respectively. The TCTP‐ORF coded for 168 amino acids with three substitutions at positions 37, 141, 155, and the ferritin ORF coded for 170 amino acids with no species‐specific substitutions. Phylogenetic analysis suggested the closest relatedness of both TCTP and ferritin from F. indicus to Chinese white prawn, Fenneropenaeus chinensis (Osbeck). In addition to reporting the full‐length sequences of these immune‐relevant genes, this study highlighted their conserved natures, which perhaps make them important defence‐related proteins in the innate immune system of F. indicus.  相似文献

Bioflocs are rich in a variety of probiotics and bioactive compounds, which have been documented to promote growth, regulate antioxidant status and enhance the immune system. A 60‐day experiment was conducted to investigate the effects on growth, antioxidant and immune status, NF‐κB/Nrf2 signalling molecules and stress resistance in juvenile Rhynchocypris lagowskii Dybowski fed different dietary biofloc supplementations. Four hundred fifty R. lagowskii were fed five experimental diets containing graded levels of biofloc from 0 to 160 g/kg (referred to as B0, B4, B8, B12 and B16, respectively). The results showed that biofloc supplementation markedly promoted growth performance (weight gain rate, specific growth rate); improved amylase, lipase and protease activity; and enhanced the activities of glutathione peroxidase, catalase, total superoxide dismutase, total antioxidant capacity, lysozyme, complement C3, complement C4 and immunoglobulin M in the gill, hepatopancreas, kidney, gut and serum. The malondialdehyde content was significantly reduced with treatment B12. In addition, the levels of heat shock protein 70, heat shock protein 90, interferon‐γ and insulin‐like growth factor I were substantially increased with treatment B12 compared to those with the control treatment. Furthermore, biofloc assisted in regulating the expression of NF‐κB/Nrf2 signalling molecule genes, including NF‐κB, TNF‐α, IL‐1β, IL‐8, IL‐10, TGF‐β, Nrf2, Keap1, Maf, HO‐1, CAT, GPX, GCLC and CuZn‐SOD. Overall, our results suggest that dietary supplementation with biofloc can promote growth, improve immune and antioxidant status, and enhance NF‐κB/Nrf2 signalling molecule expression and stress resistance in juvenile R. lagowskii. A suitable supplementation level of approximately 120 g/kg biofloc is recommended in the present study.  相似文献

Major histocompatibility complex (MHC) loci encode glycoproteins that bind to foreign peptides and initiate immune responses through their interaction with T cells. MHC class II molecules are heterodimers consisting of α and β chains encoded by extremely variable genes; variation in exon 2 is responsible for the majority of observed polymorphisms, mostly concentrated in the codons specifying the peptide‐binding region. Lactococcus garvieae is the causative agent of lactococcosis, a warm‐water bacterial infection pathogenic for cultured freshwater and marine fish. It causes considerable economic losses, limiting the profitability and development of fish industries in general and the intensive production of rainbow trout, Oncorhynchus mykiss (Walbaum), in particular. The disease is currently controlled with vaccines and antibiotics; however, vaccines have short‐term efficacy, and increasing concerns regarding antibiotic residues have called for alternative strategies. To explore the involvement of the MHC class II β‐1 domain as a candidate gene for resistance to lactococcosis, we exposed 400 rainbow trout to naturally contaminated water. One single nucleotide polymorphism (SNP) and one haplotype were associated with resistance (P < 0.01). These results are promising for using MHC class IIβ as a molecular marker in breeding rainbow trout resistant to lactococcosis.  相似文献

Using plant extracts as immunostimulants in aquaculture has prove effectiv in resisting infectious disease, in addition to being safe and inexpensive. The present study is concerned about the prospective mode of action of using lesser galangal extract (Alpinia officinarum) encapsulated into amine surface functionalized mesoporous silica nanoparticles (MSN) in order to elevate the immune status of rainbow trout (Oncorhynchus mykiss). Fish were randomly divided into 6 groups and fed for 4 weeks with commercial diets supplemented with 0% (control), 0.5% and 1.5% of lesser galangal extract, and with previous concentrations encapsulated with MSN (0.5% + MSN, 1.5% + MSN) and MSN (without extract). The effects of the feeding trial on the innate humoural immune parameters (total protein, myeloperoxidase content, lysozyme antiproteases and bactericidal activities) in rainbow trout were examined. Also, the influence of dietary supplement on some immune‐related genes in rainbow trout head kidney (TNF, IL‐8, IL‐1b, LYZ2 and CD4) after challenge with Yersinia ruckeri was examined. The results revealed enhancement in immune parameters in all treatment groups compared to the control, especially in the fish group fed with 1.5% + MSN which showed the highest significant difference (p < .05) in total protein, lysozyme and antiproteases activities. Also, feeding lesser galangal extract encapsulated into MSN led to an increase in myeloperoxidase content and bactericidal activity. An improvement in the expression of immune‐related genes has been recorded in fish groups fed doses of lesser galangal extract and lesser galangal extract encapsulated into MSN compared to the control or to the group fed with MSN only. Particularly, the group fed with 0.5% + MSN showed a significant up‐regulation in most of the immune‐related genes. The current investigation supports using lesser galangal extract encapsulated into MSN in fish diets as a supplement to enhance the immune response of rainbow trout and elevate its resistance to infectious diseases.  相似文献

We examined the effects of salinity stress and ammonia stress on alteration of the haemocyte count, phenoloxidase activity, expressions of immune‐related genes including prophenoloxidase (proPO), crustin, heat shock protein 70 (HSP70), and expressions of stress‐responsive neurohormone (Bur‐α and Bur‐β) in the thoracic and abdominal ganglia of giant river prawn Macrobrachium. These parameters of prawn that subjected to salinity stress (transferred from 0‰ to 5‰ and 10‰), and subjected to ammonia‐nitrogen (ammonia‐N) stress (transferred from 0 to 0.262 mg/L and 0.786 mg/L) were examined after 0, 3, 6, 24, 72 and 168 hr respectively. During the initial period of 3, 6 and 24 hr, granulocyte haemocyte (granular and semi‐granular hemocyte) count and PO activity significantly decreased, while expressions of Bur‐α and Bur‐β significantly increased. After 24 hr, granulocyte haemocyte count and PO activity significantly increased, whereas expressions of Bur‐α and Bur‐β significantly decreased. The expressions of proPO, crustin and HSP70 were significantly downregulated in the prawn that subjected to salinity stress and ammonia‐N stress at all time periods of 3–168 hr. In conclusion, changes in the granulocyte haemocyte count of M. rosenbergii following salinity stress and ammonia‐N stress are closely associated with the changes of Bur‐α and Bur‐β expressions.  相似文献

Resistant starch (RS) is considered to be a good alternative to antibiotics. A 56‐day feeding trial was followed by induction of acute ammonia stress to evaluate the effects of dietary RS on growth, digestion, metabolism, immunity and resistance to ammonia stress in Litopenaeus vannamei. The four diets used differed only in RS content: 0 g/kg (Control), 10 g/kg (RS1), 30 g/kg (RS2) and 50 g/kg (RS3). The results showed that dietary RS improved the growth and the survival rates of shrimp exposed to ammonia stress. At 56 days, the activity of digestive and metabolic enzymes (amylase, lipase, pepsin, hexokinase and pyruvate kinase) was increased in the three RS groups, while trypsin activity only increased in the RS1 and RS2 groups; immune‐related parameters (the total antioxidant capacity; the activity of superoxide dismutase, total nitric oxide synthase and phenoloxidase; and the expression of heat‐shock protein 70, thioredoxin, prophenoloxidase, lysozyme and toll gene) were increased in the three RS groups. After exposure to ammonia stress, all the selected immune parameters of the three RS groups were higher than those of the control group at 24 hr. Therefore, dietary RS improved the growth, digestion, metabolism, immunity and resistance to ammonia stress in L. vannamei.  相似文献

In recent years, more and more attentions have been paid to the development and application of probiotics in aquaculture, and viable probiotics have been extensively studied, while rare information was available about inactivated probiotics in aquaculture. Therefore, in this study, a feeding trial was designed to investigate the effect of dietary supplementation of heat‐inactivated probiotic Bacillus clausii DE5 on growth performance, immune response and key immune genes expression in head kidney and intestine in grouper Epinephelus coioides. Fish were fed for 60 days with control diet (C) and two experimental diets containing 1.0 × 108 CFU/g live (T1) and heat‐inactivated (T2) B. clausii DE5, respectively. The probiotic treatments did not affect the final weight, weight gain (WG) and specific growth rate (SGR) of E. coioides at days 30 and 60 (> .05), while both heat‐inactivated and live B. clausii DE5 significantly decreased the feed intake and feed conversion ratio (FCR) at day 60 (< .05). Serum lysozyme activity and complement C3 level in the two probiotic treatments were significantly higher than those in the control (< .05). The lysozyme activity and complement C3 level at day 60 were significantly higher than those at day 30 (< .05), while no significant interaction effect between diet and administration date was observed. Moreover, the heat‐inactivated B. clausii DE5 significantly improved the expression of TLR5, pro‐inflammatory cytokines (IL‐8 and IL‐1β) and TGF‐β1 in head kidney and intestine (< .05), while the live probiotic did not show any significant effect on the expression of these key immune‐related genes in head kidney and intestine. These results indicate that dietary supplementation of heat‐inactivated B. clausii DE5 effectively improved feed utilization and both the local and systemic immune responses of E. coioides.  相似文献
