The immature embryos (IEs) of wheat are the most widely used tissues for in vitro culture and genetic transformation due to its high regeneration competency. However, this explant can only be maintained in 4℃ daily cooler for a short period time for its use in plant tissue culture or transformation experiments. This study aimed to investigate the effects of environmental temperature, cryopreservation storage temperature, and heat shock culture (HSC) temperature on the regeneration frequency of wheat IEs. Results indicated that environmental temperature significantly affected the induction of embryonic calli. The optimum total accumulated temperature (TAT) during the time of anthesis and sampling for regeneration of these tissues was around 280℃ for spring wheat type cv. CB037 and approximately 300℃ for winter wheat type cv. Kenong 199. Regeneration ability obviously declined when the highest environmental temperature was over 35℃ for 1 d or a high temperature between 30 and 33℃ lasted for 5 d during anthesis and sampling. This finding was verified by culturing the freshly isolated IEs under different temperatures from 29 to 37℃ in different controlled growth incubators for 5 d; the IEs almost completely lost regeneration ability when the temperature rose to 37℃. Cryopreservation of-20℃ caused the wheat samples lost ability of producing callus or embryonic callus in a few days, and cryopreservation of-10℃ more than 10 d made the regeneration potential of the tissues dramatically declined. Comparatively, the temperature that best maintained high regeneration ability was -5℃, at which the materials can be maintained for around 1 mon. In addition, the preservation of the immature samples at -5 or -10℃ inhibited the direct germination of the IEs, avoiding the embryo axis removing process. Our results are useful for ensuring that field collection and cryopreservation of the wheat IEs are done correctly to enable tissue culture and genetic transformation.  相似文献

草地贪夜蛾是一种原于美洲的杂食性害虫,此前所未有的外来物种今年初首次在我国云南被发现。5月19日,南充市顺庆区潆溪镇首次发现有草地贪夜蛾为害,该研究经过对南充市周边县(市、区)草地贪夜蛾迁飞轨迹、降落地形和为害玉米生育期展开调查。草地贪夜蛾前期在南充主要沿嘉陵江从下游往上游迁飞,而周边远离江边的乡镇为害较轻;草地贪夜蛾在同个乡镇的不同地势的地块分布各有差异,平坦、通风口地形有利于草地贪夜蛾降落为害;在不同生育期中草地贪夜蛾更倾向于在玉米小喇叭口期取食为害。  相似文献

A study was conducted in attempting to identify the cold-resistant apple rootstocks and to establish a comprehensive evaluation system. In this study, 10 elite apple dwarfing rootstocks(GM256, JM7, M26, M7, SC1, SH1, SH38, SH6, M9, and T337) were employed for the experiment and the following parameters were investigated under different low temperature stress conditions(0, –15, –20, –25, –30, and –35°C): the changes of the relative electrical conductivity(REC), anthocyanin content, protein content, soluble sugar content, soluble starch content, proline content, malondialdehyde(MDA) content, superoxide dismutase(SOD) activity, and peroxidase(POD) activity of the dormant branches. The inflection temperature that could represent the plant tissue semi-lethal temperature(LT_(50)) was obtained by the measurements of REC. The LT_(50) was used to evaluate eight other indices. The results showed that there was no significant correlation between LT_(50) and POD activity as well as between the soluble sugar, protein and proline contents at 0 and –15°C. Soluble starch content at 0 and –15°C and anthocyanin content at –15–(–30)°C were significantly but negatively correlated to the LT50 and the MDA content at 0–(–20)°C was significantly positively correlated to the LT_(50). Statistical analysis based on principal component analysis and LT50 showed that cold resistant apple rootstocks in the decreasing order from high to low as GM256, SH6, SH38, SH1, SC1, M26, M7, JM7, T337, and M9.  相似文献

The presence of adequate nectar sources along the migratory pathway is a prerequisite for moths of oriental armyworm, Mythimna separata, to reach target habitats and lay eggs. Identification of plant species and assessment of their suitability as wild nectar sources along the summer migration pathway of adult M. separata are critical steps in making effective forecasts of second-and third-generation armyworm outbreaks in North China. We explored five aspects of moth-nectar source relationships:(1) Field investigations in 2014 and 2015 showed that the florescence of chaste tree, Vitex negundo var. heterophylla, a primary nectar plant in apiculture, coincided with the summer migration of oriental armyworm moths. The flowers of chaste tree opened day and night and M. separata moths were found actively flying around and resting on tree.(2) Identification of moth-borne pollen by scanning electronic microscopy(SEM) micrographs in 2015 and 2016 revealed that 15.4% of 369 moths trapped by blacklight traps, 17.1% of 557 moths captured by vertical-pointing searchlight traps and 24.4% of 311 moths caught by pheromone traps were found with pollen attached to their proboscis. A total of 228 pollen-carrying individuals out of 1237 armyworm moths were captured by the three types of traps. The three highest percentages of pollen-bearing armyworm moths were carrying pollen of Toona sp.(59.65%), Ligustrum sp.(27.19%) and Syringa sp.(4.39%). In total, 14 types of pollen were detected where three were identified to species, 10 to genus and one as an unknown species.(3) Analysis of geographical distribution, flowering periods and relative abundances of candidate nectar plants suggested that Chinese toon, Toona sinensis, and privet, Ligustrum lucidum, were two important nectar plant species.(4) Analysis on relationship between pollen and ovarian development showed that 14, 15 and 10 females carrying pollen of T. sinensis had ovaries at developmental stages Ⅰ, Ⅱ and Ⅲ, respectively. Many T. sinensis pollen grains were present on the proboscises of female moths.(5) Comparison of trap effectiveness between blacklight and vertical-pointing searchlight traps indicated blacklight traps caught the highest proportion(35.2%) of female in ovarian stage Ⅴ, whereas vertical-pointing searchlight traps caught the highest proportion(43.5%) of female in ovarian stage I with additional proportions of decreasing percentages for stages Ⅱ, Ⅲ and Ⅳ. Integrated analysis on these five aspects suggests that T. sinensis is an important, suitable summer nectar source that potentially influences migration and reproduction of M. separata moths andthus contributes to outbreaks of this pest. The vertical-pointing searchlight trap was the most effective tool for monitoring the migratory flight of adult M. separata. Privet, Ligustrum lucidum, and clove, Syringa sp., are possible suitable summer nectar plants that need further study to elucidate their importance. The reason why field investigation results on chaste tree differed from the results of moth-borne pollen analyses was discussed.  相似文献

The fall armyworm(FAW), Spodoptera frugiperda is one of the most damaging crop pests, and it has become major threat to the food security of many countries. In order to monitor possible invasion of this pest into China, a searchlight trap was established in March 2018 in western Yunnan Province, China, where it has served as the "first station" for many pests that have migrated from Myanmar to China. A number of suspected FAW moths were captured and identified by DNA sequencing. The results showed that the FAW moth was first captured on December 11 and formed its first immigration peak in mid-December 2018. DNA detection revealed that the early invading FAW population was the "corn-strain". The field survey indicated that the pest mainly colonized corn in Pu'er, Dehong and Baoshan areas. Migration trajectory simulation implied that the moths might have mainly come from the eastern area in the mid-latitude region of Myanmar(20–25°N, 94–100°E). This case study confirmed the first immigration of FAW into China, and will be helpful for guiding monitoring and management work to control this pest.  相似文献

[目的]为在江西省对桔小实蝇进行有效的监测和预警提供科学依据。[方法]利用桔小实蝇的分布点数据和环境因子数据,通过GARP生态位模型预测桔小实蝇在江西省的潜在分布区域。[结果]桔小实蝇在江西省的适生面积占全省面积的2/3以上。在24°~26°N赣南盆地的大部分区域,年平均气温在19~20℃,是桔小实蝇的高适生区。在26°~27°N的赣州北部的部分地区以及吉安西南部、抚州、鹰潭、上饶东南部的少部分地区,年平均气温在18~19℃,是桔小实蝇的中度适生区。在26°~30°N的滨湖、赣江中下游、抚河、袁水区域和赣西南山区,年平均气温在17~18℃,是桔小实蝇的低适生区。赣东北、赣西北和长江沿岸地区,年平均气温为16~17℃,是桔小实蝇的非适生区。[结论]该研究结果表明,桔小实蝇的实际分布与GARP的预测分布基本相符。  相似文献

为探索草莓炭疽病潜伏侵染条件提供理论依据。本研究在离体条件下通过调控室内温湿度对草莓炭疽病的发病情况及侵染特性差异性进行研究。结果表明:温湿度组合处理25℃/85%、20℃/80%、15℃/80%、10℃/80%、15℃/75%、10℃/75%在接种7天后均能发病,处理10℃/70%接种后28天才轻微发病。侵染特性差异性研究表明,处理25℃/85%,接种6 h,25%的分生孢子开始萌发;接种12 h,附着孢开始形成,形成率为16%,接种48 h,叶片开始出现零星病斑;接种7天,叶片组织细孢大量腐烂崩解,病指为39.07。处理10℃/70%,接种1天,2.88%分生孢子开始萌发;直至接种24天,分生孢子萌发率达到100%,附着孢开始形成,形成率为8.83%;接种28天,叶片开始出现零星病斑,病指仅为1.85;接种42天,叶片组织大面积腐烂,形成典型的炭疽病斑。得出处理10℃/70%接近该病害发病的底线温湿度条件,此条件下的潜伏期能够达到28天。  相似文献

Rice grain yield and quality declines are due to unsuitable temperatures from wide regions and various sowing dates. This study aimed to evaluate the effects of temperature on rice yield and quality at different phenological periods and obtain suitable temperatures for phenological periods in the Yangtze River Basin, China. This study conducted experiments on different sowing dates under different areas in the Yangtze River Basin to observe and compare the differences in rice growth, yield, and quality, controlling for regional varieties. The results showed significant differences in rice growth, yield, and quality among sowing dates and areas, which were related to the average daily temperature during the vegetative period (VT) and the first 20 days of the grain-filling period (GT20). In addition, there was a smaller variation in the average daily temperature in the reproductive period (RT) than in the two phenological periods. Therefore, according to the VT and GT20 thresholds of different yields and qualities, the experimental results were divided into four scenarios (I, II, III, and IV) in this study. In Scenario I, high head rice production (rice grain yield multiplied by head rice rate) and rice quality could be obtained. The head rice production of Scenarios III and IV was lower than that of Scenario I, by 30.1 and 27.6%, respectively. In Scenario II, the head rice production increased insignificantly while the chalky grain rate and chalkiness were 50.6 and 56.3% higher than those of Scenario I. In conclusion, the Scenario I combination with VT ranges of 22.8–23.9°C and GT20 ranges of 24.2–27.0°C or the combination with VT ranges of 23.9–25.3°C and GT20 ranges of 24.2–24.9°C, which can be obtained by adjusting sowing date and selecting rice varieties with suitable growth periods, is recommended to achieve high levels of rice grain yield and quality in the Yangtze River Basin.  相似文献

Many proteins require assistance from molecular chaperones at various stages to attain correctly folded states and functional conformations during protein synthesis. In this study, the gene encoding T-complex polypeptide 1(TCP-1), which belongs to the heat shock protein 60(HSP60) family, was isolated and characterized from the rice stem borer, Chilo suppressalis, by RACE and q PCR, respectively. The full-length c DNA of Tcp-1 was 2 144 bp and encoded a 1 635-bp ORF; the deduced translational product contained 545 amino acids with 5′-and 3′-UTRs and an isoelectric point of 5.29. Cluster analysis confirmed that the deduced amino acid sequence shared high identity(60–99%) with TCP-1 from other insects. To investigate Tcp-1 expression in response to abiotic stress, q PCR was used to analyze expression levels of Tcp-1 m RNA in C. suppressalis larvae exposed to temperatures ranging from –11 to 43°C. With respect to heat shock, Tcp-1 expression was higher than the control after a 2-h exposure to 30 and 36°C and declined at 39 and 43°C. Difference in Tcp-1 expression was observed at temperatures ranging from –11 to 27°C. q PCR analyses revealed that Tcp-1 expression was the highest in hindgut tissue as compared to heads, epidermis, fat body, foregut, midgut, and malpighian tubules. Our results indicated that Tcp-1 expression was differentially expressed in C. suppressalis tissues, and was impacted by temperature stress.  相似文献

The armyworm Mythimna roseilinea(Walker) is a major pest of grain crops in South China. So far little is known about its basic biology and ecology, making prediction of population dynamics difficult. This study examined the relationships of individual development and population growth with temperature based on an age-stage, two-sex life table of M. roseilinea reared on maize in the laboratory at 18, 21, 24, 27 and 30°C. The highest values of net reproductive rate(R_0) and fecundity were observed at 21 and 24°C, respectively. Both the intrinsic rate of increase(r) and finite rate of increase(λ) increased significantly and mean generation time(T) decreased significantly with increasing temperature. M. roseilinea was able to develop, survive and lay eggs at all temperature regimes tested. Development rates of the egg, larval, pupal, as well as the whole pre-oviposition stages had a positive linear relationship with temperature. The calculated development threshold temperatures of egg, larval, pupal, pre-oviposition and total pre-oviposition stages were 13.29, 8.39, 14.35, 7.42, and 12.24°C, respectively, and their effective accumulated temperatures were 63.59, 445.00, 211.11, 89.02, and 698.95 degree-days, respectively. These results provide insight into temperature-based phenology and population ecology of this insect pest and will allow population prediction and management available in the field.  相似文献

The present study aimed to investigate the effect of relative humidity(RH) at either acute or chronic moderate ambient temperature(Ta) on growth performance and droppings' corticosterone metabolites of broilers.Two experiments were conducted: effect of RH(35,60 or 85%) on average daily feed intake(ADFI) and droppings' corticosterone metabolites at acute(1 d: 20–26 or 31–20°C,26 or 31°C for 6 h d–1 at 10:00–16:00) moderate Ta(experiment 1) and effect of RH(35,60 or 85%) on growth performance and droppings' corticosterone metabolites at chronic(step-wisely increasing temperature by 3°C every 3 d from 20 to 32°C within 15 d: 20–23–26–29–32°C) moderate Ta(experiment 2).Droppings were collected at the 2,4,6,8,and 22 h after Ta-RH controlled in experiment 1 and at the 2,4,6,and 22 h after Ta controlled to 32°C in experiment 2.The results showed that: 1) In experiment 1,85% RH increased(P0.05) the droppings' corticosterone metabolites at the 2,6,8,and 22 h and 35% RH increased(P0.05) it at the 2 and 22 h compared to the 60% RH.Moreover,85% RH further increased(P0.05) it compared to the 35% RH,however,no difference(P0.05) was found in ADFI among the three RH groups at acute moderate 26°C; 35 and 85% RH increased(P0.05) droppings' corticosterone metabolites at the 2,6,8 and 22 h and decreased(P0.05) ADFI compared to the 60% RH,moreover,85% RH further increased(P0.05) droppings' corticosterone metabolites and further decreased(P0.05) ADFI compared to the 35% RH at acute moderate 31°C; and the average of droppings' corticosterone metabolites in the whole period had a negative correlation(P0.02) with the ADFI.2) In experiment 2,85% RH increased(P0.01) droppings' corticosterone metabolites only at the 2 h and decreased(P0.02) ADFI and average daily gain(ADG) compared to the 60% RH,no difference(P0.05) in droppings' corticosterone metabolites was found between the 35 and 60% RH,however,35% RH decreased(P0.01) ADG compared to the 60% RH,and the average of droppings' corticosterone metabolites in the whole period also had a negative correlation(P0.02) with ADFI and ADG.In conclusion,droppings' corticosterone metabolites could be used as a RH stress index and low and high RH,especially high RH,reduced growth performance possibly through inducing RH stress at moderate temperature.  相似文献

Factors influencing vegetative growth and spore germination of strain F603 of Epicoccom sp., an antagonistic fungus for Phytophthora infestans (Mont) de Bary, were studied. Among the different growth media tested, Rye agar was the best medium for its vegetative growth. The range of temperature and pH value for mycelial growth was 5–35°C and 2–12, respectively, with the optimum 25°C and 6–9, respectively. The fungus grew better in Czapek medium with maltose and dextrose as carbon sources and peptone, KNO3, and NaNO3 as nitrogen sources. The range of temperature for spore germination of strain F603 was 5–35°C, the optimum was 20°C. The range of temperature for sporulation was 10–30°C, and the optimum was 15–18°C.  相似文献

采用不同密度性诱盆处理与常规化学防治处理的对比试验方法,综合评估性信息素防治甜菜夜蛾的作用和效应.结果显示:性诱盆放置密度直接影响诱获雄蛾的频次与平均日单盆诱蛾量,低密度处理(1 hm2放置3个性诱盆)与高密度处理(1 hm2放置15个性诱盆)诱获雄蛾频次分别为71.22%与48.50%;平均日单盆诱蛾量在3头以上和3头以下的天数,低密度处理分别为29 d和38 d,分别占总诱蛾日的43.30%和56.70%,而高密度处理平均分别为7 d和60 d,分别占总诱蛾日的10.50%与89.50%;按单位面积诱捕雄蛾量计算,高密度处理雄蛾诱杀效果较低密度处理提高93.90%;在甜菜夜蛾主害代以1 hm2放置15个性诱盆的密度进行性诱防治,性诱防治区累计诱捕雄蛾874头,对幼虫的控制效果与常规化学防治相当.表明,应用昆虫性信息素诱杀防治主害代甜菜夜蛾可作为减少化学农药的无公害防治手段列入甜菜夜蛾综合防治体系中.  相似文献

In order to improve beef color and color stability,step-chilling(SC)was applied on excised bovine longissimus lumborum muscle,with chilling starting at 0–4℃ for 5 h,then holding the temperature at 12–18℃ for 6 h,followed by 0–4℃ again until 24 h post-mortem.pH and temperature were measured during rigor on SC loins as well as those subjected to routine chilling(RC,0–4℃,till 24 h post-mortem).Color L*,a*,b*values,metmyoglobin(MetM b)content,MetM b reducing ability(MRA)and NADH content were determined on samples aged for 1,7,and 14 d.Sarcoplasmic proteome analysis was only conducted on d 1 samples.The results showed muscles subjected to SC maintained a temperature at around 15℃ for 5 to 10 h post-mortem,and exhibited a slow temperature decline,but rapid pH decline.Beef steaks treated with SC had higher L*,a*,b* and chroma values than those of RC samples at 1 and 7 d chilled storage(0–4℃),while showing no significant difference for a*,b* and chroma values at d 14.The SC samples also exhibited a lower relative content of surface MetM b,higher MRA and NADH content,compared with RC beef steaks during storage,indicating the SC-treated beef showed an improved color stability.Eleven differential protein spots/nine proteins were identified by two-dimensional gel electrophoresis and mass spectrometry,and those proteins were mainly involved in redox,chaperone binding,metabolic and peroxidase activity.Oxidoreductases play a role in decreasing the oxidation-induced myoglobin oxidation and benefiting the production of NADH,and finally improving the colour of beef.Of these,pyruvate dehydrogenase E1 component subunit beta showed a positive correlation with color L*,a*,b*values and accounted for more than 60% of the variation in color values;this protein can be considered as a potential beef color biomarker.The present study provided valuable information for studies on the molecular mechanism of color improvement from step-chilling,as well as for identifying markers associated with beef color.  相似文献

The fall armyworm(FAW), Spodoptera frugiperda, is a migratory noctuid pest that has recently invaded eastern Asia. From 2017 up till 2020, searchlight trapping was used to assess the extent of FAW migration across the South China Sea. Molecular and morphology-based identification confirmed that FAW made its first appearance on Yongxing Island on 11 April 2019, with most trapped individuals belonging to the S. frugiperda "corn-strain". Carbon isotope analysis further showed that FAW moths originated from C4 host plants, while trajectory analyses revealed that migratory S. frugiperda adults are able to cross the South China Sea and enter mainland China. This long-distance migration process plausibly results in frequent genetic mixing between domestic FAW populations and those of neighboring Southeast Asian countries. Overall, this work provides unique insights into FAW migration across eastern Asia and ultimately can help advance pest forecasting, risk assessment and area-wide pest management.  相似文献

[目的]合理利用气象条件,提高香榧产量。[方法]通过了解枫桥香榧(细榧)产量的年际变化及其生物学特性,分析气象因子与香榧产量的关系。[结果]枫桥香榧(细榧)产量年际波动大,关键是主要生育物候期的气象因子,本真阶段:2~3月寒潮及临界低温-7℃,盛夏临界高温天数20d和临界雨日12d;假真阶段:临界日平均气温15℃,中下旬低温和连阴雨临界累计日数8d,中旬低温和连阴雨临界累计日数5d;实真阶段:5~6月临界日照时数300h和临界降水日30d。[结论]开花前(本真阶段)雨水调匀、温度适宜,花期(假真阶段)和落花落果期(实真阶段)少雨多照和多雨凉夏的气候有利于香榧丰产。  相似文献

为了评价引进文冠果种质的抗寒性,为其进一步栽培和育种利用提供参考,以引进种质6份文冠果种质((G27、BD12、BD特异、建2、株系2、G13))为试材,设置-10 °C、-20 °C、-30 °C低温胁迫梯度,测定相对电导率,并拟合Logistic方程计算半致死温度(LT50),评价供试种质抗寒性强弱。结果显示供试种质半致死温度分别是-13.272、-12.244、-11.543、-8.246、-5.643、-2.642 ℃,依据半致死温度评价抗寒性强弱为BD特异>G27>BD12>建2>G13>株系2。相对电导率结合Logistic方程评价文冠果种质抗寒性结果与前期研究结果基本一致,可作为评价种质抗寒性的简要方法之一,同时为引进种质进一步在新疆地区栽培与育种利用提供参考。  相似文献

Three transgenic maize events(IE09 S034, Shuangkang 12–5 and C0030.3.5) produced Cry1 Ie, Cry1 Ab/Cry2 Aj and G10-EPSPS, Cry1 Ab and EPSPS, respectively, all of which target the Asian corn borer. The oriental armyworm Mythimna separata(Walker) is the secondary target. In this study, the effects of the three Bt maizes on the development and survival of armyworm were studied. The results showed that IE09 S034 had insecticidal activity against 1 st instar larvae, and the survival rate of armyworm fed with Bt maize for 10 days was 46.2%, significantly lower than that of the control. The larvae at 3 rd–6 th instar were more tolerant of the Bt toxin than the early instar larvae. However, Shuangkang 12-5 had good insecticidal activity against 1 st–5 th instar larvae. The mortality was nearly 100% when the larvae were fed with Shuangkang 12-5 before 3 rd instar, and the toxin had quick-acting efficacy. This event significantly inhibited the development of armyworm; that is, the larval duration of the 3 rd and 4 th instar larvae fed with Shuangkang 12-5 was prolonged by 4.5 and 3.0 days, respectively. The pupal weight and egg number were also significantly lower than those of the control. For C0030.3.5, it could control 1 st–5 th instar larvae effectively. The mortality rates were all over 50% if 1 st–3 rd larvae were fed with this event. The pupal weight of 4 th–6 th instar larvae fed with Bt maize were only 53.9, 56.8 and 54.6%, respectively, compared to that of the control. The number of eggs laid was significantly less than the control. The results indicate that all three transgenic maize events exhibit the potential to provide effective control of early instar larvae of armyworm, which can be commercialized in future to control lepidoptera pests such as Asian corn borer and armyworm.  相似文献

The fall armyworm(FAW, Lepidoptera: Noctuidae), Spodoptera frugiperda(J. E. Smith), invaded China in mid-December 2018; since then, it has become a great threat to Chinese agricultural production. Qinling Mountains–Huaihe River region(QM–HRR) is the transitional zone between northern and southern China, an important region for both corn and wheat production. Based on the actual occurrence of QM–HRR invaded by FAW in 2019, daily mean surface air temperature and nocturnal wind conditions at 925 hPa were examined, and migratory routes of FAW moths originated in QM–HRR were modeled by a forward-trajectory-analysis approach. The results indicated that migratory activities of FAW adults emerged in QM–HRR were initiated from late June. The moths from western QM–HRR, where has complex topographic terrain, mainly flied to Ningxia and Inner Mongolia before mid September. However, FAW moths from the eastern QM–HRR primarily engaged in high-altitude northward transport assisted by the prevailing southerly winds before mid August, and the North China Plain was identified as the main destination of FAW. Meanwhile, the migration trajectories of FAW moths had a possibility to reach the Northeast China Plain. From mid August, FAW moths in eastern QM–HRR largely migrated southward and returned to the Yangtze River Valley. This study provides detailed information on the occurrence and migration routes of FAW moths from QM–HRR and will be helpful for early warning and development of integrated pest management strategies for the control of this exotic insect pest.  相似文献
