The haemagglutination activity of the causal agent of canine parvovirosis is described. Out of the tested erythrocytes of pig, monkey, cat, horse, cattle, sheep, rabbit and guinea-pig, a positive reaction was recorded only in the erythrocytes of pig and monkey at the temperature of 4 degrees C. As a result of the examination of 20 faeces samples of hospitalized dogs by the method of haemagglutination reaction, a positive reaction typical of canine parvovirosis was obtained in 17 cases. The specificity of the reaction was confirmed by antiserum against the virus of panleucopenia of cat in the haemagglutination-inhibition test.  相似文献

Previous findings, viz. that mice can be successfully immunized against infection with Salmonella dublin with either live or inactivated vaccine, were confirmed. Immunity lasted for at least 12 weeks in mice which had been immunized with inactivated alum-precipitated vaccine. The immunogenicity of inactivated vaccine gradually decreased on storage at 4 degrees C, but this was only detectable if a single injection was used for immunization: 2 injections virtually eliminated this phenomenon. The immunogenicity of live vaccine in mice was not enhanced by levamizole or the simultaneous injection of inactivated organisms. Both live and inactivated vaccines provided immunity in calves. A single injection of lyophilized vaccine, prepared from live rough Salmonella dublin strain (HB 1/17),protected 3 out of 6 calves, while 2 injections of a formalin-inactivated, alum-precipated vaccine, containing 1% packed cells of S. dublin strain 2652 V, protected 5 out of 6 calves against intraduodenal challenge with 2 x 10(9), S. dublin strain 2652 V. Two calves which had been immunized with an inactivated oil adjuvant vaccine were also solidly immune to this challenge. Serum antibody response in calves was poor when measured by the tube agglutination and the haemagglutination tests. Similarly, the sera had only marginal protective values when tested by means of a passive protection test in mice. Antibody titres alone are not a valid measure therefore, for the immune status of immunized animals.  相似文献

Fecal and nasal samples were collected from 180 calves with diarrhea and 36 clinically normal co-habitants, and tested for virus using HRT-18 cell cultures derived from human rectal adenocarcinoma. A cytopathic virus was isolated from 5 fecal and 56 nasal samples obtained from diarrheic calves. All calves in which the virus was isolated from diarrheic feces were positive for virus isolation from nasal swabs. The virus was also isolated from the nasal swabs of 10 clinically normal calves that were co-habitants with diarrheic calves. Because they were morphologically similar to coronavirus, agglutinated mouse erythrocytes and serologically identical with the Nebraska calf diarrhea coronavirus, new isolates were identified as bovine coronavirus. The demonstration of viral antigens in nasal epithelial cells by a direct immunofluorescence was in close agreement with the virus isolation in HRT-18 cell cultures. This is the first report on the isolation of bovine coronavirus from newborn calves with diarrhea in Japan. The evidence that the virus was frequently isolated from nasal swabs is of great interest for understanding the pathogenesis of bovine coronavirus infection.  相似文献

Basic variables of the haemagglutination inhibition (HI) test for porcine parvovirus antibody were investigated. Nonspecific serum inhibitors were satisfactorily removed without loss of specific antibody when undiluted serum was adsorbed with 25 percent kaolin in borate saline at pH 9.0. Natural haemagglutinins in test serums could be completely removed using 0.1 ml of packed erythrocytes to 0.6 ml of kaolin treated serums. Adsorption of prediluted serum resulted in a depression of specific antibody titres. Highest HI titres were obtained using guinea pig erythrocytes, following incubation of virus-serum mixtures for 18 hours at 4 degrees C, 3 hours at 25 degrees C or 2 hours at 37 degrees C. Micro- and macro-tests gave comparable HI titres.  相似文献

The purpose of this study was to determine aetiological agents of diarrhoea in neonatal calves and to investigate virulence gene markers of Escherichia coli strains isolated from calves by multiplex polymerase chain reaction (PCR). Eighty-two diarrhoeic calves and 18 healthy calves were used as subjects. Faeces were taken from the rectums of all the calves and were subjected to bacterial culture. Antigen enzyme-linked immunosorbent assay (ELISA) was performed to detect rotavirus, coronavirus and E. coli K99 in faeces of all the calves. A multiplex PCR was used to characterize E. coli strains in all the calves. Escherichia coli was isolated from 37 faeces samples, Enterococcus ssp. was isolated from 22 faeces samples and Salmonella was isolated from one faeces sample in diarrhoeic calves. Furthermore, only E. coli was isolated from all 18 faeces samples of healthy calves. Of the 37 E. coli isolated from diarrhoeic calves, K99 (18.9%), F41 (18.9%), heat-stable enterotoxin a (STa) (18.9%), Shiga toxin 1 (Stx1; 13.5%) and Shiga toxin 2 (Stx2; 5.4%) and intimin (8.1%) genes were identified by multiplex PCR. Of the 18 E. coli isolated from healthy calves, K99 (16.6%) and intimin (55.5%) genes were identified by PCR. A total of 15 rotavirus, 11 coronavirus and 11 E. coli K99 were detected in diarrhoeic calves by the antigen ELISA. As a result, this study shows that rotavirus, coronavirus, E. coli and Enterococcus ssp. were determined to play a role in the aetiology of diarrhoea in the neonatal calves. K99, F41, STa, Stx1 and Stx2 were found as the most common virulence gene markers of E. coli strains isolated from calves with diarrhoea. Multiplex PCR may be useful for characterization of E. coli isolated from calves.  相似文献

Standardised procedure for obtaining reproducible haemagglutination-inhibition results for FPV antibody which correlate with serum-neutralization titres was described. Optimal conditions were found to be Alsevers anticoagulant, PBS/0.05% BSA (pH 6.8) as buffer, especially washed round bottom microplates, determination of maximally sensitive porcine erythrocytes, use of reproducible erythrocyte concentrations, inactivation of serum samples at 56 degrees C for 30 min and serum treatment with koalin pH 9.0. The concentration of erythrocyte used for estimation of haemagglutination units in H1 test should not differ from that used as indicator in the test. Predilution of serum beyond 1:4 associated with false results. Reproducible method for removing natural agglutinins in serum by adsorption with erythrocytes was described.  相似文献

Coronaviruses were isolated from neonatal calves with diarrhoea in Great Britain and Denmark. They were serially passed in gnotobiotic calves which developed acute diarrhoea. Pathological lesions were found in the small and large intestines. Coronaviruses were demonstrated by electron microscopic examination of the faeces and intestinal contents, immunofluorescent staining of sections of small and large intestine and by isolation in tracheal organ cultures. In early passages of the British coronavirus, particles of about 30 nm in diameter were observed in the faeces by electron microscopy. These particles were removed from the coronavirus preparations by cross-protection experiments in calves. The coronaviruses were morphologically and antigenically similar to the bovine coronavirus isolated in the United States and the British virus was adapted to replicate in calf kidney cell cultures.  相似文献

Faeces were collected per rectum from calves infected with Dictyocaulus viviparus (D.v.), from lambs infected with Dictyocaulus filaria (D.f.) and donkeys infected with Dictyocaulus arnfieldi (D.a.). In one experiment, the influence of storage temperature before Baermannization was investigated. Recovery rate for D.v. was approximately 80% after 24 h at 4 degrees C or 16 degrees C but only 40% at 20 degrees C. After two days at 20 degrees C the rate had fallen to 20%. Recovery rates for D.f. decreased so markedly during the first 12 h at 4, 16 and 20 degrees C that storage can not be recommended. Losses in the recovery rates of D.a. appeared insignificant after 48 h at 4 degrees C but not at 16 degrees C and 20 degrees C. In experiment II the time taken for larvae to emerge from a 10-g sample as well as the sedimentation time in Baermann tubes was investigated. The bulk of the D.v. larvae remained in the faeces for about 10 h whereas D.f. larvae emerged during the first few hours. D.a. larvae were intermediate in this respect. Sedimentation of the bulk of larvae from all three species took place within a few hours with the modified Baermann technique used.  相似文献

Thirteen Escherichia coli strains isolated from calves with diarrhoea, supposed to carry a common antigen were examined for their hemagglutinating activity and compared by bacterial agglutination, double diffusion in two dimensions and by crossed immunoelectrophoresis (CIE). Two of the strains were examined also in the electron microscope. Most of the strains agglutinated red blood cells of horse, ox, guinea pig and chicken, of which the agglutination of ox erythrocytes was mannose-resistant (MRHA). None of the strains agglutinated human erythrocytes. All strains with MRHA of ox red blood cells, regardless to their O:K:H antigens could be agglutinated in unabsorbed or absorbed antisera produced against cultures C1209 (020:K-:H9) and C1213 (09:K36:H-) when live cells as antigens were used. None of these sera agglutinated reference strains carrying K88, K99, 987P, F41 or FY (Att25) antigen respectively. By the double gel diffusion test and by CIE in extracts (60 degrees C) of the strains a common heat labile antigen, responsible for the MRHA of ox red blood cells was identified. Electron microscopy revealed that this common antigen was represented by thin, long, hair-like fimbriae on cells of E. coli C1213, and that specific homologous antibodies attached to these fimbriae.  相似文献

Eight-month-old calves, housed under maximum isolation, were exposed to pathogenic Leptospira interrogans serovar hardjo by the conjunctival route or IV. One calf served as an unexposed control. Infection was monitored serologically (microscopic agglutination test and enzyme-linked immunosorbent assay; ELISA) and by leptospiral culture isolation from periodic urine samples and from the kidneys, epididymides, and aqueous humor collected at slaughter. Microscopic agglutination test titers of greater than or equal to 1:40 were detected among all IV exposed calves at postinoculation day (PID) 7 and among conjunctival exposed calves at PID 14. By ELISA, all IV exposed calves were positive by PID 3, whereas conjunctival exposed calves were positive at PID 14. The ELISA was more sensitive for the detection of antibodies against leptospires in cattle. Leptospires were isolated from the urine of 4 calves and from the kidney of 3 calves exposed by the conjunctival route, but not from IV exposed calves. The results indicated that the conjunctival route of exposure was a more natural and successful route for experimental infection of cattle with serovar hardjo.  相似文献

Antibodies against Fasciola hepatica were detected in sera from experimentally infected sheep and calves. Titres in sera from field-infected sheep were related to fluke burdens. Indirect haemagglutination, counter-immunoelectrophoresis and double immunodiffusion were compared. Indirect haemagglutination and counter-immunoelectrophoresis were superior to double immunodiffusion and positive reactions in these techniques preceded the presence of parasite eggs in the faeces by at least 50 days. The relative advantages of the immunological techniques are discussed.  相似文献

A study was undertaken to investigate the effects of the immune response induced by combined aerosol and parenteral vaccination on the lung lesions induced in calves by Pasteurella haemolytica AI. Twenty-four calves, twelve of which had been vaccinated with killed P. haemolytica by aerosol and subcutaneous injection in Freund's complete and incomplete adjuvant were challenged by intratracheal inoculation of live P. haemolytica. Serological response to vaccination was not marked but was best measured by the whole cell agglutination test or by indirect bacterial agglutination rather than by the passive haemagglutination test. Titres of vaccinates were positively correlated with the degree of pneumonic change following challenge while in nonvaccinated controls, titres were negatively correlated with lung lesions. These findings suggest the occurrence of an immunologically mediated hypersensitivity pneumonitis in the lungs of vaccinates and point to the potential efficacy of live bacterial aerosols for stimulation of protective immunity in pneumonic pasteurellosis.  相似文献

Faecal pellets from a sheep that was artificially infected with a monoculture of Haemonchus contortus were collected over a 2-h period in the morning. In the laboratory the faeces were thoroughly mixed by hand and 48 by 1 g aliquots of the pellets were sealed in plastic bags, from which the air had gently been expressed. The faecal worm egg count of the sheep was about 14,000 g(-1). Varying numbers of the bags were either processed for faecal worm egg counting (FEC) by the McMaster technique on day 0, or were stored at one of the following temperatures: about 4 degrees C, -10 degrees C or -170 degrees C before processing. The faecal aliquots that were frozen were thawed at room temperature after having been frozen for either 2 h or 7 days, and processing of aliquots maintained at 4 degrees C proceeded shortly after the samples had been removed from the refrigerator. A dramatic reduction in egg numbers was found in all the aliquots that were frozen at -170 degrees C before faecal worm egg counts were done, as well as in those frozen for 7 days at about -10 degrees C. Numerous empty, or partially empty, egg shells were observed when performing the counts in faeces that had been frozen. In contrast, there was no significant reduction in the numbers of eggs in aliquots maintained for 7 days in a refrigerator at +/- 4 degrees C before examination, when compared with others examined shortly after collection of the faeces. Since H. contortus eggs in faeces are damaged by freezing, some methods that can be used for short term preservation are outlined. It is concluded that all nematode egg counts from cryopreserved faeces (whether in a freezer at -10 degrees C or in liquid nitrogen) should possibly be regarded as being inaccurate, unless the contrary can be demonstrated for different worm genera. However, exceptions are expected for the more rugged ova, such as those of the ascarids and Trichuris spp.  相似文献

A virological survey was carried out on 150 calf faeces from 30 central North Island properties, utilising electron miscroscopic examination of ultracentrifuged faecal pellets. Rotaviruses were detected on three farms, and coronavirus-like particles on two farms.

In the same area, three other properties with past histories of rotavirus infection were also studied. Weekly examination of faeces from 10 calves from each property over a period of 5 weeks demonstrated that rotavirus excretion commenced at the same time as the onset of outbreaks of diarrhoea. On two of these rotavirus-positive farms, coronaviruses were also detected. At one farm, this coincided with rotavirus excretion and diarrhoea, and was associated with a more severe clinical disease but, on the other property, the coronavirus was unassociated with clinical disease. A similar study, carried out on a further property with a past history of coronavirus infection, showed that coronavirus excretion commenced simultaneously with the onset of an outbreak of diarrhoea. The virus was, however, detected in small numbers in 1 of 10 calves only.

The results of the survey, coupled with previously gathered data, indicate that both viruses may be widespread in the cattle population. On some properties, rotavirus had a clear association with diarrhoea, but the relationship of coronavirus to diarrhoea was less well-established.  相似文献

Faecal samples of different consistency from 15 calves were subjected to two methods to separate faecal fluid. Osmolality as well as the concentrations of electrolytes and short chain fatty acids were determined for each of the fluid samples obtained and the results compared. For dialysis the following procedure was used: small dialysis bags filled with dextran 10% were placed in the cooled (4 degrees C) faecal sample to which gentamycin was added to inhibit fermentation. Equilibrium between dialysate and surrounding fluid was reached after 24 hours. However, depending on the consistency of the faeces, after 6 hours 90 to 95% of the final osmolality at 24 hours was reached. In regard to osmolality, sodium, potassium and chloride concentrations as well as to concentrations of short chain fatty acids no significant differences could be observed between the faecal fluid obtained by dialysis or by centrifugation (3500 g, 20 min, 4 degrees C) and subsequent filtration. Distinctly higher concentrations of calcium and inorganic phosphate were measured in the filtrates; the probable explanation is that these ions in faeces are largely present as poorly diffusible compounds (in particular with faeces of a more solid consistency) or bound to the cell membranes.  相似文献

A simple and rapid procedure has been developed for the detection of bovine coronaviruses in faecal specimens. The method consists of adsorption of the virus onto mouse erythrocytes at 4°C, removal of unadsorbed material and elution of adsorbed viral material at 37°C. The eluate is then used in a haemagglutination test. Specificity of the reaction is checked by a blocking assay. No non-specific reactions have been observed. The sensitivity of the test appeared to be better than that of the electron microscope, at least when crude faecal extracts are used. By ultracentrifugation of the eluates the sensitivity of the assay can be further improved.  相似文献

In a series of five laboratory experiments observations were made on the role of Pilobolus in the translation of infective lungworm larvae from faeces of cattle. The results indicated that a substantial proportion of the lungworm larvae present in the faeces may be translated from the faeces by this fungus within eight days at a temperature of 15 degrees C. No clear relationship was observed between the numbers of Pilobolus and the translation of lungworm larvae. Further a longitudinal study on the occurrence of Pilobolus on faecal pats of grazing calves showed that between the beginning of July and the middle of September peak emergence of sporangia generally occurred within one week and most sporangia emerged within three weeks. From faecal pats which had been deposited at the end of September and the middle of October emergence of sporangia was lower and mainly occurred after two to four weeks.  相似文献

Between late December 1999 and late April 2000, three locally bred Friesian calves (ageing 25, 28 and 35 days) in a dairy farm, at Al-Ahsa locality of the eastern region of Saudi Arabia showed dullness and inappetence. Their rectal temperatures ranged between 41 and 41.5 degrees C. One to 2 days later and onwards, the calves showed lacrimation, nasal discharge, salivation, oedema of the head, conjunctivitis, exo-ophthalmia and corneal opacity. One calf showed diarrhoea. The superficial lymph nodes were oedematous and swollen. The calves died after 7, 5 and 8 days, respectively, following the onset of the disease. Calves and rabbits were experimentally infected with materials from the naturally infected calves. The rabbits showed fever, mild conjunctivitis and one rabbit showed wet faeces. The experimentally inoculated calves showed rise in temperature and mild symptoms but none of them died. The virus from the naturally infected calves and from the experimentally infected rabbits was identified as malignant catarrhal fever (MCF) virus using both the complement fixation test and the fluorescent antibody test, employing a reference anti-serum against the WC 11 strain of MCF virus. Serological survey for MCF antibodies in cattle, sheep and goats from the affected farm revealed that 54% of the examined animals were positive. The situation of MCF in Saudi Arabia was discussed in relation to sheep and wild game. This paper constitutes the first report of MCF in Saudi Arabia and the Gulf region.  相似文献

Forty-five calves on four veal calf units were monitored during the first four weeks after their arrival. Faecal samples were collected on alternate days and screened for the presence of rotaviruses, bovine coronavirus, Cryptosporidium oocysts, K99 positive strains of E. coli and Salmonella spp. Rotaviruses and Cryptosporidium were the most commonly detected agents (78% and 60% respectively of the calves). Bovine coronavirus was detected in the faeces of 18% of the calves, whilst K99 positive E. coli was only found in 2 samples from one calf. Salmonella spp. were not isolated from any of the 646 faecal samples examined. Shedding of rotaviruses occurred in a bimodal pattern beginning in the first week of the survey. Cryptosporidium oocysts were detected most frequently in the interval between the two peaks of rotavirus shedding. The presence of rotaviruses or Cryptosporidium oocysts in faeces was not strongly associated with scour, nor were combined infections with these agents or the cases of bovine coronavirus infection. The condition of the calves throughout the survey was generally satisfactory.  相似文献

Previous reports on the spread of bovine virus diarrhoea virus (BVDV) from animals primarily infected with the agent are contradictory. In this study, the possibility of transmission of BVDV from calves simultaneously subjected to acute BVDV and bovine coronavirus (BCV) infection was investigated. Ten calves were inoculated intranasally with BVDV Type 1. Each of the 10 calves was then randomly allocated to one of two groups. In each group there were four additional calves, resulting in five infected and four susceptible calves per group. Virulent BCV was actively introduced in one of the groups by means of a transmitter calf. Two calves, susceptible to both BVDV and BCV, were kept in a separate group, as controls. All ten calves actively inoculated with BVDV became infected as shown by seroconversions, and six of them also shed the virus in nasal secretions. However, none of the other eight calves in the two groups (four in each) seroconverted to this agent. In contrast, it proved impossible to prevent the spread of BCV infection between the experimental groups and consequently all 20 study calves became infected with the virus. Following infection, BCV was detected in nasal secretions and in faeces of the calves and, after three weeks in the study, all had seroconverted to this virus. All calves, including the controls, showed at least one of the following clinical signs during days 3-15 after the trial started: fever (> or =40 degrees C), depressed general condition, diarrhoea, and cough. The study showed that BVDV primarily infected cattle, even when co-infected with an enteric and respiratory pathogen, are inefficient transmitters of BVDV. This finding supports the principle of the Scandinavian BVDV control programmes that elimination of BVDV infection from cattle populations can be achieved by identifying and removing persistently infected (PI) animals, i.e. that long-term circulation of the virus without the presence of PI animals is highly unlikely.  相似文献
