Genetic variation in the physical properties of sweet potato starch.   总被引:10,自引:0,他引:10  
Sweet potato starch, prepared from 44 genotypes adapted to Philippine conditions, showed wide variation and distinctly different pasting profiles in Rapid Visco-Analyzer (RVA) analysis at 11% and 7% starch concentration. At 11% starch concentration, the pasting profiles were type A, characterized by high to moderate peak with a major breakdown and low cold paste viscosity. At 7%, the pasting profile was generally type C, characterized by the absence of a distinct peak with none to very slight breakdown and high cold paste viscosity. However, differentiation among genotypes was better achieved from RVA pasting profiles at 11% starch concentration. Peak viscosity (PV) and hot paste viscosity (HPV) at 11% starch paste concentration had significant negative correlation with amylose content. PV, HPV, and setback ratio were significantly correlated to adhesiveness of the starch gel. Sweet potato starch generally had high swelling volume but low solubilities at 92.5 degrees C.  相似文献

Some mutant wheat lines with low‐amylose content were grown in a field and greenhouse (15 or 20°C) to compare apparent amylose content and starch pasting properties. The apparent amylose content of flour and starch increased and starch pasting parameters as measured by a Rapid Visco Analyser (RVA) changed in the greenhouse (at cool temperatures) during seed maturation. Densitometric analysis of the protein band separated by electrophoresis suggested that the increase in amylose content by cool temperature was related to the amount of Wx‐D1 protein. This data suggests that the Wx‐D1 gene was responsible for these changes. In wheat starch from Tanikei A6099 and Tanikei A6598 at 15°C, the value of final viscosity and total setback was higher than that from the field. In wheat starch from Tanikei A6599‐4 (waxy mutant with stable hot paste viscosity), the peak viscosity temperature was higher and time maintained >80% of the peak was shorter at 15°C than that from the field. Genetic analysis using doubled‐haploid (DH) lines from a combination of Tanikei A6599‐4 and Kanto 118 (low‐amylose line) showed that apparent amylose content increased and the starch pasting curve and properties changed in waxy progenies similar to Tanikei A6599‐4.  相似文献

Changes in pH and pasting properties of instant‐noodle formula dry‐mix suspensions containing each of 12 phosphate salts were investigated. The pH values of solutions alone and then solution and flour suspensions decreased as the level of phosphate salts increased, except that of trisodium phosphate, which increased the pH value. The changes in the Rapid Visco Analyser (RVA) pasting parameters of instant‐noodle formula suspensions were not consistent with the respective changes in pH, but the change trends of the RVA parameters for the two different wheat flours (hard red winter and soft white wheat) were similar. Five of the phosphate salts gradually increased the RVA peak viscosity (PV) as phosphate concentration increased. Seven other phosphate salts increased the PV at 0.05% and then decreased PV as the phosphate concentration increased. The change in trough viscosity owing to phosphate salt and concentration was similar to that of PV. The final viscosity (FV) gradually declined, to varying degrees, as the phosphate concentration increased for some of the phosphate salts. However, seven of the phosphate salts caused slight increases in FV as their concentrations increased. The response of starch gelatinization and pasting behavior, as measured by RVA, indicated that phosphate salts exert an influence on starch during heating in water. Because RVA parameters have been linked to instant‐noodle processing and textural properties, phosphate salt identity and concentration can likely be manipulated to affect end‐product quality.  相似文献

To determine the effect of amylose content on the starch properties, the amylose content, pasting properties, swelling power, enzymatic digestibility, and thermal properties of partial and perfect waxy types along with their wild‐type parent were analyzed. As expected, amylose content decreases differently in response to the loss of each Wx gene, showing the least response to Wx‐A1a. Most of the characteristics, except the thermal properties of the amylose‐lipid complex in differential scanning calorimetry (DSC), differed significantly among the tested types. Furthermore, the breakdown, setback, and pasting temperatures from the Rapid Visco Analyser (RVA) and the enzymatic digestibility, swelling power, peak temperature, and enthalpy of starch gelatinization from DSC showed a correlation with the amylose content. The relationships between the peak viscosity from the RVA and the onset temperature of starch gelatinization determined by DSC with amylose content of the tested materials were not clear. Waxy starch, which has no amylose, showed a contrasting behavior in starch gelatinization compared with nonwaxy starches. Among the nonwaxy starches, lower setback, lower pasting temperature, higher enzyme digestibility, higher peak temperature, higher enthalpy of starch gelatinization, and higher swelling were generally associated with low amylose starches.  相似文献

The starch of wheat (Triticum aestivum L.) flour affects food product quality due to the temperature-dependent interactions of starch with water during gelatinization, pasting, and gelation. The objective of this study was to determine the fundamental basis of variation in gelatinization, pasting, and gelation of prime starch derived from seven different wheat cultivars: Kanto 107, which is a partial waxy mutant line, and six near-isogenic lines (NILs) differing in hardness. Complete pasting curves with extended 16-min hold at 93°C were obtained using the Rapid Visco Analyser (RVA). Apparent amylose content ranged from 17.5 to 23.5%; total amylose content ranged from 22.8 to 28.2%. Starches exhibited significant variation in onset of gelatinization. However, none of the parameters measured consistently correlated with onset or other RVA curve parameters that preceded peak paste viscosity. Peak paste viscosity varied from 190 to 323 RVA units (RVU). Higher peak, greater breakdown, lower final viscosity, negative setback, and less total setback were associated with lower apparent and total amylose contents. Each 1% reduction in apparent or total amylose content corresponded to an increase in peak viscosity of about 22 and 25 RVU, respectively, at 12% starch concentration. Of the seven U.S. cultivars, the lower amylose cultivars Penawawa and Klasic were missing the granule-bound starch synthase (GBSS; ADPglucose starch glycosyl transferase, EC 2.4.4.21) protein associated with the Waxy gene locus on chromosome 4A (Wx-B1 locus). Kanto 107 was confirmed as missing both the 7A and 4A waxy proteins (Wx-A1 and Wx-B1 loci). The hardness NIL also were shown to be null at the 4A locus. Apparent and total amylose contents of prime starch generally corresponded well to the number of GBSS proteins; although the hardness NIL tended to have somewhat higher amylose contents than did the other GBSS 4A nulls. We concluded that reduced quantity of starch amylose due to decreased GBSS profoundly affects starch gelatinization, pasting, and gelation properties.  相似文献

Seven early indica rice starches with different amylose contents were modified by octenyl succinic anhydride (OSA) in aqueous suspension systems to evaluate the effect of amylose contents on starch esterification. The crystalline structure and pasting properties of starches were investigated using X-ray diffraction and a Rapid Visco Analyzer (RVA). The results indicated that the amylose content had a positive impact on the OSA modification. As the amylose content increased from 0 to 39.6%, the degree of substitution increased from 0.024 to 0.030 and the reaction efficiency increased from 62.8 to 77.5%. X-ray diffraction scans confirmed that the amylose was mainly present in the amorphous domain of the granule and was highly substituted after the OSA treatment. The RVA profiles demonstrated that the OSA starches had higher viscosities than their native counterparts. Moreover, negative correlations were observed between the amylose content and the major RVA parameters (e.g., peak viscosity, hot paste viscosity, cool paste viscosity, and breakdown viscosity).  相似文献

Zero amylose starch isolated from hull-less barley (HB) showed a typical A-type diffraction pattern. The X-ray analysis suggested that granules of zero amylose (SB94794) and 5% amylose (CDC Candle) HB starches had lower crystallinity than did commercial waxy corn starch. Differential scanning calorimetry showed lower transition temperatures and endothermal enthalpies for the HB starches than for the waxy corn starch. The zero amylose HB starch showed a Brabender pasting curve similar to that of waxy corn starch, but with lower pasting and peak temperatures and a higher peak viscosity. Noteworthy characteristics of zero amylose HB starch were its low pasting temperature and high paste clarity and freezethaw stability, which make this starch useful for many food and industrial applications.  相似文献

Sweetpotato flour (SPF), prepared from 44 genotypes adapted to Philippine conditions, showed wide variation in Rapid Visco-Analyzer (RVA) pasting characteristics due to its variation in composition and endogenous amylase activity. The RVA pasting parameters of peak viscosity determined in water (PV1) and that determined in 0.05 mM AgNO(3) (used as an amylase inhibitor) (PV2) were successfully used to estimate alpha-amylase activity. The correlation of the ratio (PV2-PV1)/PV1 to alpha-amylase activity was 0.96 (p < 0.01, N = 44). Swelling volume measurements were not found to be suitable for prediction of alpha-amylase activity.  相似文献

Flours from five spelt cultivars grown over three years were evaluated as to their breadbaking quality and isolated starch properties. The starch properties included amylose contents, gelatinization temperatures (differential scanning calorimetry), granule size distributions, and pasting properties. Milled flour showed highly variable protein content and was higher than hard winter wheat, with short dough‐mix times indicating weak gluten. High protein cultivars gave good crumb scores, some of which surpassed the HRW baking control. Loaf volume was correlated to protein and all spelt cultivars were at least 9–51% lower than the HRW control. Isolated starch properties revealed an increase in amylose in the spelt starches of 2–21% over the hard red winter wheat (HRW) control. Negative correlations were observed for the large A‐type granules to bread crumb score, amylose level, and final pasting viscosity for cultivars grown in year 1999 and to pasting temperature in 1998 samples. Positive correlations were found for the small B‐ and C‐type granules relative to crumb score, loaf volume, amylose, and RVA final pasting viscosity for cultivars grown in 1999, and to RVA pasting temperature for samples grown in 1998. The environmental impact on spelt properties seemed to have a greater effect than genetic control.  相似文献

The objective of these studies was to find alternative Rapid Visco Analyser (RVA) viscoelastic parameters that are predictable by near‐infrared spectroscopy (NIRS). Currently, RVA instruments are widely used in assessing cooking and processing characteristics in rice. The ability to predict RVA parameters by NIRS would be useful in rapidly determining rice pasting qualities, but NIRS does not correlate with the traditional parameters (peak viscosity, final viscosity, breakdown, consistency, and setback). Alternative RVA parameters were sought by collecting RVA and NIRS data for a total of 86 short, medium, and long grain rice cultivars. The amylose contents were 0.41–24.90% (w/w) and protein concentrations were 8.47–11.35% (w/w). Partial least squares (PLS) regression models generated for the entire NIR spectrum against the RVA curve showed viscosity at 212–228 sec (80°C ± 1) varied linearly with NIR spectra (1,100 to ‐2,500 nm). Regression coefficient values were R = 0.961 for 212 sec and R = 0.903 for 228 sec. The PLS correlation coefficient for the prediction of amylose at 212–228 sec decreases along with the NIRS correlation to the same time frame. An opposite trend was observed for the correlation with protein at 212–228 sec. This comparison suggests the importance of amylose and protein in water absorption during this time frame.  相似文献

The influence of amylose content, cooking, and storage on starch structure, thermal behaviors, pasting properties, and rapidly digestible starch (RDS), slowly digestible starch (SDS), and resistant starch (RS) in different commercial rice cultivars was investigated. Long grain rice with high‐amylose content had a higher gelatinization temperature and a lower gelatinization enthalpy than the other rice cultivars with intermediate amylose content (Arborio and Calrose) and waxy type (glutinous). The intensity ratio of 1047/1022 cm–1 determined by Fourier Transform Infrared (FT‐IR), which indicated the ordered structure in starch granules, was the highest in glutinous and the lowest in long grain. Results from Rapid ViscoAnalyser (RVA) showed that the rice cultivar with higher amylose content had lower peak viscosity and breakdown, but higher pasting temperature, setback, and final viscosity. The RDS content was 28.1, 38.6, 41.5, and 57.5% in long grain, Arborio, Calrose, and glutinous rice, respectively, which was inversely related to amylose content. However, the SDS and RS contents were positively correlated with amylose content. During storage of cooked rice, long grain showed a continuous increase in pasting viscosity, while glutinous exhibited the sharp cold‐water swelling peak. The retrogradation rate was greater in rice cultivars with high amylose content. The ratio of 1047/1022 cm–1 was substantially decreased by cooking and then increased during storage of cooked rice due to the crystalline structure, newly formed by retrogradation. Storage of cooked rice decreased RDS content and increased SDS content in all rice cultivars. However, no increase in RS content during storage was observed. The enthalpy for retrogradation and the intensity ratio 1047/1022 cm–1 during storage were correlated negatively with RDS and positively with SDS (P ≤ 0.01).  相似文献

在大田条件下,选用3个不同类型优质小麦品种: 豫麦47(强筋品种)、山农8355(中筋品种)和豫麦50(弱筋品种),设置3个氮肥水平: 施N 0、15和30 g/m2,研究了小麦子粒淀粉的粒度分布、直支链淀粉组成、糊化特性及其对氮素水平的响应。结果表明,优质小麦子粒中淀粉粒的粒径分布范围为1~45 μm,其数目分布呈单峰或双峰曲线变化,体积和表面积分布均呈双峰曲线变化,峰谷位于10 μm处; 据此可将淀粉粒分为两种类型: A型大淀粉粒(10~45 μm)和B型小淀粉粒(1~10 μm)。优质小麦子粒淀粉粒组成存在显著的基因型差异。强筋品种豫麦47子粒中B型淀粉粒的比例较高,弱筋品种豫麦50子粒中A型淀粉粒的比例较高,中筋品种山农8355居中。施氮水平对优质小麦子粒中淀粉的粒度分布存在显著影响。在本试验条件下,随氮素水平的提高,强筋品种豫麦47子粒中A型淀粉粒的比例提高,而B型淀粉粒的比例下降; 增施氮肥后弱筋品种豫麦50和中筋品种山农8355子粒中B型淀粉粒的比例增大,而A型淀粉粒的比例降低,且前者变化的幅度较大。适量增施氮肥提高优质小麦子粒中的淀粉含量,氮肥用量进一步增大后,淀粉含量降低; 增施氮肥后优质小麦子粒中直链淀粉含量降低。增施氮肥对优质小麦子粒淀粉的糊化特性存在较大影响,且此影响的趋势因基因型和施氮量而异。其中强筋品种豫麦47表现为低谷粘度、最终粘度、反弹值、糊化温度和峰值时间提高,而高峰粘度和稀懈值降低; 当氮肥用量增大至30 g/m2时,糊化温度和峰值时间降低,而以粘度为单位的参数均提高。弱筋品种豫麦50表现为增施氮肥后,RVA参数呈下降趋势,与之相对应中筋品种山农8355的呈上升趋势。相关性分析表明,B型淀粉粒的数目、体积和表面积比例与高峰粘度和稀懈值存在显著正相关; 与低谷粘度、最终粘度和反弹值存在显著负相关。子粒中直链淀粉含量、支链淀粉含量和总淀粉含量与高峰粘度和稀懈值呈显著负相关,与低谷粘度、最终粘度、反弹值和峰值时间呈一定程度正相关; 直链淀粉相对含量与RVA特征参数之间的相关趋势与子粒中直链淀粉含量的趋势一致,但均未达显著水平。由此可以认为,氮肥通过调控小麦子粒中淀粉的直、支链组成和粒度分布而影响其糊化特性。  相似文献

Pasting profiles of selected starches were compared by using a Micro Visco‐Amylo‐Graph (MVA) and a Rapid Visco Analyser (RVA). Effects of cooking (heating/cooling) rate and stirring speed on starch pasting properties were examined. The pasting viscosity of a starch suspension (8%, w/w, dsb) was measured at a fast (6°C/min) and slow (1.5°C/min) cooking rate while being stirred at either 75 rpm or 160 rpm. The pasting temperatures (PT) of all starches were higher when measured at the fast cooking rate than those at the slow cooking rate, except for wheat measured by using the RVA. PT was also higher when measured at the slow stirring speed (75 rpm) than at the fast stirring speed (160 rpm) in both RVA and MVA. When stirring speed increased from 75 rpm to 160 rpm, peak viscosity of all starch pastes except potato decreased measured by using the RVA, but increased by using the MVA. In general, amylograms of these starches obtained by using the MVA showed less breakdown, but greater setback viscosity than did that obtained by using the RVA. Differences in starch pasting properties between MVA and RVA, measured at the same cooking and stirring rates, were attributed mainly to the difference in spindle structure.  相似文献

Brown rice was blasted with rice flour rather than sand in a sand blaster to make microperforations so that water could easily penetrate the brown rice endosperm and cook the rice in a shorter time. The flour‐blasted American Basmati brown rice, long‐grain brown rice, and parboiled long‐grain brown rice samples were stored in Ziploc storage bags under atmospheric conditions and in vacuum‐packed bags. They were periodically tested for over 10 months for changes in water absorption, free fatty acid (FFA), peroxide value (POV), viscosity changes of flour using the Rapid ViscoAnalyser (RVA), and texture of whole cooked kernel using a texture analyzer during cooking. Flour‐blasted brown rice absorbed less water but needed less cooking time than its counterpart that was not flour‐blasted. There was an increase in FFA, POV, peak viscosity (PV), final viscosity (FV), breakdown viscosity (BD), and setback viscosity (SB) during storage of flour‐blasted brown rice for 300 days, but no change was observed in texture (hardness, gumminess) and water absorption. The combined coefficient of correlation (including all types of rice) between FFA and FV is r = 0.86 and between FFA and SB is r = 0.90 at P < 0.0001.  相似文献

This study describes the effect of starch properties of Japanese wheat flours on the quality of white salted noodles (WSN). Starch was isolated from 24 flours of 17 Japanese cultivars and amylose content was determined along with pasting properties by Rapid Visco Analyser (RVA), thermal properties by differential scanning calorimetry (DSC), and the distribution of amylopectin chain length by high‐performance anion exchange chromatography (HPAEC). Twenty flours were used to prepare WSN. As expected, 5–6% lower amylose content was associated with good WSN quality (higher scores in softness, elasticity, and smoothness). RVA analysis indicated that the pasting temperature had the greatest influence on WSN quality, while breakdown and setback showed slight effects on WSN quality. DSC results showed that lower endothermal enthalpy (ΔH) in the amylose‐lipid complex was associated with good WSN quality. Chainlength distribution of amylopectin by HPAEC was not an important factor in relation to WSN quality.  相似文献

The objective of these studies was to gain a better understanding of the pasting process in rice. We chose six different medium grain rice flour samples with amylose contents of 0.41–24.9% and protein contents of 4.89–10.65%. By using the first derivative of Rapid Visco Analyser (RVA) curves, changes in the pasting rates could be obtained. We found that samples containing low amylose contents (CM101 [CA] 0.41% amylose and 7.04% protein) exhibited a single smooth transition during pasting. Pastes from all other samples, M201 (TX), Nato (LA), Koshihikari (CA), Mercury (LA), and Nanking Sel (LA) with higher amylose contents (10.65–24.9%) underwent multiple phase transitions and rate changes before the peak viscosity. Disruption of disulfide linkages using dithiothreitol (DTT) led to a decrease in the rate of the single pasting step observed for CM101 (CA). Rice containing larger concentrations of amylose showed an increase in the first, but a decrease in subsequent steps. Our data suggests that amylopectin and protein are mutually important in the initialization of pasting in rice. At later stages of pasting, amylose and its complexes seem to become important.  相似文献

以苏玉糯1号、苏玉糯5号和渝糯7号为材料,研究了拔节期追氮量(N 0、150和300 kg/hm2)对鲜食糯玉米粉糊化和热力学特性的影响。结果表明,随着拔节期追氮量的增加,峰值黏度和崩解值下降,糊化温度升高,而谷值黏度、终值黏度和回复值呈先降后升趋势; 热力学特征参数中,回生值、终值温度、糊化范围和峰值指数受拔节期追氮量影响较小。原样品热焓值和回生后样品热焓值均表现为随着拔节期追氮量的增加呈先降后升趋势。糊化和热力学特征值对拔节期追氮量的响应不同品种间存在差异。鲜食糯玉米粉的理化特性存在显著的基因型差异,峰值黏度以苏玉糯5号最高,苏玉糯1号最低; 峰值温度苏玉糯1号和苏玉糯5号无显著差异,但均高于渝糯7号; 热焓值、回生值、糊化范围和峰值指数不同品种间相对稳定。相关分析表明,峰值黏度与原样品热焓值和峰值指数呈显著正相关; 崩解值与糊化温度及转变温度,回生值与淀粉的终值黏度和回复值呈显著负相关。在本试验条件下,拔节期追氮总体上使糯玉米食用品质降低,其中以苏玉糯5号在不追氮处理下的糊化和热力学特性较优,即峰值黏度、崩解值和热焓值较高,回生值较低。  相似文献

Four rice starches were isolated from waxy and nonwaxy rice cultivars collected from different places in China. Individual rice starches were examined, along with their corresponding mixtures in different ratios, in terms of pasting and hydration properties. Analysis by micro‐viscoamylography (MVAG) showed that waxy rice starch and its blends had higher peak viscosity (PV), breakdown (BD), and setback (SB) than the remaining starches and mixtures. Apparent amylose content (AC) was 16.95–29.85% in nonwaxy individual rice starches and 13.69–25.07% in rice starch blends. Incorporating waxy rice starch (25%) significantly decreased the AC. AC correlated negatively with swelling power (SP) (r = ‐0.925, P < 0.01). SP exhibited nonlinear relationship (r2 = 0.8204) with water solubility (WS) and both increased with temperature. The correlation showed that WS is also an index of starch characteristics and the granules rigidity affected the granule swelling potential. The results show that turbidity of gelatinized starch suspensions stored at 4 ± 0.5°C generally increased during storage up to five days.  相似文献

Systematic studies were performed on the effect of the surfactant alkyl chain length (10–16 carbon atoms) and the head group charge/structure (anionic, cationic, nonionic) on the pasting properties of wheat flour and starch aqueous suspensions by means of a Rapid Visco Analyser (RVA). An excellent agreement was observed between the effect of surfactants on the onset temperature of the pasting process (PT) and the time to reach peak viscosity (tpeak) of wheat flour and wheat starch suspensions. Moreover, a correlation was found between the effect of different surfactants on these two parameters. With the exception of the cationic surfactants (alkyl trimethyl ammonium bromides), the effect of surfactants (alkyl sulfates, maltosides, monoglycerides, and sucrose esters) was found to be strongly dependent on the surfactant chain length. Shorter chain surfactants (C10–C12) induced an earlier pasting, while longer chain surfactants (C14–C16) had the opposite effect. The effect of surfactants on PT and tpeak of flour suspensions was enlarged when the surfactant concentration was increased from ≈1% to 15% (w/w) on a dry starch basis.  相似文献

The role of proteins in the pasting and cooking properties of non‐parboiled (npb) and parboiled (pb) rice was tested by means of a reducing agent dithiothreitol (DTT) and a protease (trypsin). DTT increased the swelling power and carbohydrate leaching of flour from npb rice flour but decreased its amylose leaching. Although DTT slightly increased the Rapid Visco Analyser (RVA) viscosity at the initial stages of the pasting process, it decreased RVA viscosity in the further phases of the experiment. Preincubation of flour with a trypsin decreased RVA viscosity along the whole temperature profile. Addition of DTT to the cooking water decreased water absorption and rice hardness and increased leaching of solids during cooking and stickiness of the cooked npb rice. Addition of DTT to the cooking water of flour from pb rice increased swelling power, carbohydrate leaching, and amylose leaching. Addition of DTT also increased RVA viscosity. Preincubation with trypsin had a similar effect but the changes were less pronounced. Addition of DTT increased stickiness of cooked pb rice and increased water absorption and leaching of solids during cooking. Taken together, the results provide evidence for the existence of a protein barrier affecting starch swelling, rheological, and cooking properties of both npb and pb rice.  相似文献
