In this review, possible comparative advantages of studying endocrine disruption in farm animals vs laboratory rodents are discussed. First, using farm animals, the generality of findings in laboratory rodents are challenged. Farm animals may in certain aspects be better models for humans than laboratory rodents, and sometimes there might be methodological advantages in using farm animals. Second, there are several in vitro studies based on cell-culture systems from sows and cows where the effects of chemicals on sex steroid secretion can be measured and maturation and fertilization of oocytes may be assessed. These in vitro systems are powerful tools for dissecting the mechanisms of action for endocrine-disrupting chemicals. Third, in a set of recent in vivo studies using sheep, goats and pigs, in which very different exposure regimens to endocrine-disrupting chemicals have been used, a full panel of reproductive parameters pertinent to farm animals were assessed. Clinically, it is suggested that endocrine disruption in farm animals should be considered when impaired reproduction could be linked to change in source of feed or pasture. Finally, epigenetic and toxicogenomic approaches can be particularly rewarding in elucidating endocrine disruption in future farm animal studies.  相似文献

Contents The sources and characteristics of endocrine disrupting compounds (EDCs) are reviewed and discussed with respect to their potential effects on farm animal health, welfare and productivity. The importance of certain properties of these compounds in relation to the expression of their biological effects is addressed together with potential routes of exposure. It is concluded that little is known of factors affecting the tissue concentrations of EDCs in farm animals, the concentrations that are required to perturb physiological function in these species, the effects of prolonged exposure to low doses, the effect of cocktails of EDCs and other pollutants or the responses of specific organs and physiological systems that are affected by EDCs. Much of the available information pertaining to EDCs is derived from epidemiological studies of wildlife species and from laboratory animal studies and while these studies have significant limitations, they are considered to be valuable indicators of potential effects in farm animal species. The results of such studies, together with the small amounts of data from studies of ruminants, indicate that there may be significant effects of exposure to environmental levels of EDCs on farm animal health, even although effects are not generally apparent in practice, at this time.  相似文献

Polyphenols are secondary plant metabolites which have been shown to exert antioxidative and antiinflamma tory effects in cell culture, rodent and human studies. Based on the fact that conditions of oxidative stress and inflammation are highly relevant in farm animals, polyphenols are considered as promising feed additives in the nutrition of farm animals. However, in contrast to many studies existing with model animals and humans, potential antioxidative and antiinflammatory effects of polyphenols have been less investigated in farm animals so far. This review aims to give an overview about potential antioxidative and antiinflammatory effects in farm animals. The first part of the review highlights the occurrence and the consequences of oxidative stress and inflammation on animal health and performance. The second part of the review deals with bioavailability and metabolism of polyphenols in farm animals. The third and main part of the review presents an overview of the findings from studies which investigated the effects of polyphenols of various plant sources in pigs, poultry and cattle, with particular consideration of effects on the antioxidant system and inflammation.  相似文献

The objective of this review is to consider the ethics of stockmanship, particularly from the perspective of the nature and extent of the duties of stockpeople to their farm animals. It will consider what science tells us about the impact of stockmanship on the animal, particularly the welfare of the farm animal. The effects of human-animal interactions on the stockperson will also be considered, since these interactions affect the work performance and job satisfaction of the stockperson and thus indirectly affect animal welfare. Animal ethics is broader than animal welfare and includes economic as well as philosophical, social, cultural and religious aspects. This paper is predicated on the view that farm animals can suffer, and that animal suffering is a key consideration in our moral obligations to animals. Housing and husbandry practices affect farm animal welfare and thus farmers and stockpeople have a responsibility to provide, at minimum, community-acceptable animal housing and husbandry standards for their animals. The farmer's or stockperson's attitudes and behaviour can directly affect the animal's welfare and thus they also have a responsibility to provide specific standards of stockmanship for these animals. However, research suggests that the behaviour of some stockpeople is not as correct as it might be. Such situations exemplify the inevitably unequal human - domestic animal relationship, and this inequality should be considered in analysing the boundary between right and wrong behaviour of humans. Thus ethical discussion, using science and other considerations and involving stockpeople, livestock industries, government and the general public, should be used to establish and assure acceptable stockperson competencies across the livestock industries. Training programs targeting the key attitudes and behaviour of stockpeople presently offer the livestock industries good opportunities to improve human-animal interactions.  相似文献

Theileria parva causes widespread morbidity and mortality in cattle in endemic regions. An outbreak of theileriosis occurred on a farm near Ladysmith in KwaZulu-Natal, South Africa, which is not a declared Corridor disease-infected area. A survey of Red Brangus cattle from all age groups and areas of the farm was performed. Transmission of the parasite from infected animals on the farm to susceptible animals by tick transmission and tick-stabilate injection, was attempted. The survey indicated high numbers of animals with antibody titres to T. parva but only 6 infected animals, based on real-time PCR and RLB analysis. The transmission experiments failed to transmit the parasite. The study shows the difficulty in elucidating a source of infection and determining the dynamics of new infections in a herd where multiple possible sources are present and treatment with tetracyclines has taken place.  相似文献

Hypodermosis is a myiasis of cattle mainly found in the Northern Hemisphere. In Italy it has been reported primarily in southern regions with a farm prevalence reaching a peak of 85%, while few data are available in the north of the country.This paper reports the results of a serological survey carried out on hypodermosis in Vicenza province (northeastern Italy). During January and February 1999, bulk milk samples were collected from 224 dairy farms and tested, using an hypodermosis ELISA kit. Epidemiological data, such as farm location, herd size, number of cows, grazing management, treatments against Hypoderma larvae, presence of imported animals, were gathered and statistically analysed in relation to the serological results. In 15 farms the presence of warbles was monitored by manual palpation and larvae collected for identification.Ninety-seven (43.3%) samples were detected as positive, whereas 12 (5.4%) and 115 (51.3%) were inconclusive and negative, respectively.Thirteen farms were positive upon parasitological examination and the larvae collected were identified as Hypoderma bovis.Seroprevalence was highly correlated with the practice of free grazing and slightly, but significantly, correlated with the number of cows on the farm. The mean antibody level of positive herds was significantly higher on farms without imported animals. On farms with warbled animals there was a high inverse correlation between antibody level and quantity of milk in the tanks and the number of lactating cows.The ELISA test on bulk milk samples has been shown, under field conditions, to be a useful, cost-effective and easy test for the diagnosis of bovine hypodermosis.  相似文献

Neospora caninum is known to cause abortion in cattle. This study demonstrated the presence of specific IgG to Neospora in milk and serum samples obtained from three dairy farms in Georgia and two in Texas. Samples from four hundred fourteen dairy cows were examined using a western blot assay of which 362 were milk and 87 were serum. Samples with antibodies to Neospora were identified in 32.1% (105/327) of the examined animals in Georgia, whereas in Texas it was identified in 10.3% (9/87). Positive Georgia samples were found in 24.4% from farm A (28/115), 21.6% from farm B (30/139), and 64.4% from farm C (47/73). In Texas, 13.5% (7/52) of animals in farm D and 5.71% (2/35) from farm E also had specific antibodies to Neospora. The number of animals from Georgia dairy farms with antibodies to Neospora was significantly higher than the Texas dairy farms. This may be related to the age of the animals examined in this study (more than 2 years old). Antibodies present in sera had excellent agreement with the antibodies present in milk. Collection of milk samples for serological testing is easier and less invasive than obtaining bovine sera, therefore offering an alternative for animal testing.  相似文献

The effects of passive immunization against somatostatin on retention time of marker have been investigated in sheep using Cr2O3 as a marker. The mean transit time for clearance of 50% of the marker was significantly retarded in all the anti-somatostatin-treated animals by approximately 19.0 h, as compared with control lambs given a nonspecific antiserum. The findings suggest an important role for circulating somatostatin in the regulation of the rate of flow of digesta through the gastrointestinal tract in lambs. Moreover, it enhances the claim of the immunological approach as a new technique to investigate factors controlling food utilization in farm livestock.  相似文献

Verocytotoxin-producing Escherichia coli (VTEC) O157 phage type 2 (PT2) was isolated from three calves and two goats on a farm open to the public. Phenotypic and DNA-based typing showed that the strains were identical or very closely related to those obtained from an outbreak of VTEC O157 infection in two separate family groups who visited the farm. No VTEC O157 PT2 was isolated again from the farm during a 12-month longitudinal bacteriological study undertaken after the infected animals had been removed. However, phenotypically and genotypically indistinguishable VTEC O157 PT2/28 strains were detected in two of 474 faecal samples collected at monthly visits from 15 species of animals of various ages. The two isolates were obtained from calves from different sources sampled 146 days apart, suggesting that the infection had persisted on the farm although it was not detected in the other species. The same strain was subsequently isolated from another calf housed in the same pen as one of the infected calves. The longest period during which the organism was excreted was seven days. No VTEC O157 was isolated either from 204 replacement animals (including 138 orphan lambs and 10 calves) brought in from various sources, and sampled while they were kept in isolation for two weeks before being introduced to the farm, or from environmental samples. During the study a visitor became ill with VTEC O157 PT2. However, the isolate was distinct from those recovered from the farm and there was no evidence to suggest that the visit was the source of the infection.  相似文献

The aim of the study was to determine the burden of Toxoplasma gondii-infections in sheep in Sicily, southern Italy and the risk factors for infection. Sera from 1961 sheep were collected just before slaughtering from 62 farms located in 8 out of 9 Sicilian administrative districts. The sera were analysed for Toxoplasma-specific IgG antibodies using commercially available enzyme-linked immunosorbent assay. Sheep less than 4 weeks old were further analysed by ELISA for Toxoplasma-specific IgM-antibodies. Data on farm size and location were obtained from slaughterhouse sanitary reports and through structured telephone interviews of the veterinary officers from public health districts. The overall seroprevalence of Toxoplasma-specific IgG-antibodies were 49.9% (937/1876) by ELISA. Eighty-seven (54/62) percent of the farms had at least one Toxoplasma-positive animal. All the farms fed the animals outdoor on pasture and only one was claiming organic farming. Having cats on the farm, age of the animals, farm size and the use of surface water sources for drinking were all significantly associated with T. gondii-infected animals on the farm. T. gondii infection in mutton used for human consumption is very prevalent, and eating unprocessed sheep and lamb meat has a high risk of transmitting infections to humans. The presence of cats on the farm, farm size and using surface water as drinking water for the animals were risk factors for infection in sheep, with age as a significant confounder.  相似文献

In recent foot and mouth disease outbreaks, many healthy animals have been culled to prevent disease transmission. Emergency vaccination is discussed as an alternative to culling of unaffected animals. A spatial and temporal Monte–Carlo simulation model was used to compare preventive culling and emergency vaccination. Different outbreaks are described using additional influence factors such as airborne spread, farm density, type of index-case farm and delay until establishment of the control strategies. The fewest farms were infected establishing a combined strategy including a 1 km preventive culling and 1–10 km emergency vaccination zone around each outbreak farm. Taking the number of culled and vaccinated farms into account, vaccination around the first diagnosed farm combined with the baseline strategy (culling of outbreak farms, protection and surveillance zone, contact tracing) is to be preferred. In the present study, emergency vaccination was an effective control strategy especially in densely populated regions.  相似文献

To avoid health risks in farm animals it is necessary to limit the intake of unwanted chemical compounds via air, feed or drinking water. The basis for this procedure are experimental results of Veterinary Toxicology, after which acceptable daily intakes can be estimated. A health risk is not present, if those limits are not exceeded. Risks may not only occur as an impairment of health itself but as well as a reduction of yield of food producing animals (body growth, production of meat and fat, fertility, milk and egg production). In addition the cumulation of unwanted compounds in tissues, which are used for the production of food of animal origin, is of great importance for the limitation of daily intakes of those environmental pollutants. Following the results of toxicological experiments in the target animals "Toxicological Drinking Water Standards for Animals" can be established.  相似文献

This paper considers (potentially) harmful consequences of transgenesis for farm animal welfare and examines the strategy of studying health and welfare of transgenic farm animals. Evidence is discussed showing that treatments imposed in the context of farm animal transgenesis are by no means biologically neutral and may compromise animal health and welfare. Factors posing a risk for the welfare of transgenic farm animals include integration of a transgene within an endogenous gene with possible loss of host gene function (insertional mutations), inappropriate transgene expression and exposure of the host to biologically active transgene-derived proteins, and in vitro reproductive technologies employed in the process of generating transgenic farm animals that may result in an increased incidence of difficult parturition and fetal and neonatal losses and the development of unusually large or otherwise abnormal offspring (large offspring syndrome). Critical components of a scheme for evaluating welfare of transgenic farm animals are identified, related to specific characteristics of transgenic animals and to factors that may interact with the effects of transgenesis. The feasibility of an evaluation of welfare of transgenic farm animals in practice is addressed against the background of the objectives and conditions of three successive stages in a long-term transgenic program. Concrete steps with regard to breeding and testing of transgenic farm animals are presented, considering three technologies to generate transgenic founders: microinjection, electroporation and nuclear transfer, and gene targeting including gene knockout. The proposed steps allow for unbiased estimations of the essential treatment effects, including hemi- and homozygous transgene effects as well as effects of in vitro reproductive technologies. It is suggested that the implementation of appropriate breeding and testing procedures should be accompanied by the use of a comprehensive welfare protocol, specifying which parameters to monitor, at which stages of the life of a farm animal, and in how many animals. Some prerequisites and ideas for such a protocol are given. It is anticipated that systematic research into the welfare of farm animals involved in transgenesis will facilitate the use of the safest experimental protocols as well as the selection and propagation of the healthiest animals and, thereby, enable technological progress that could be ethically justified.  相似文献

The phenomenon of endocrine disruption can be regarded as part of the disciplines of toxicology and environmental toxicology. These two disciplines have generated guideline protocols on how various effects of chemicals should be tested as a basis for regulatory decisions. These protocols almost exclusively involve laboratory rodents and the data obtained are then used for human risk assessment. Would it be justifiable, then, to introduce or promote the use of other species in these test protocols? There are, at any rate rationales for studying effects in species other than laboratory rodents: (1) other species may better mimic the human system; (2) they may in some cases be more useful for studying a certain mechanism or phenomenon; (3) they may highlight the diversity of effects or sensitivity between species. However, there are at least two basic criteria that must be met for a species before it can be introduced in this context: (a) we must have a good understanding of the physiological system to be studied; and (b) we must have a number of tools to study effects on this system. When it comes to the reproductive system – regarding which most endocrine disruption has been reported – farm animals are second only, or in some respects superior, to laboratory rodents with respect to these criteria. This review gives examples of how farm animals can be of use in the study of endocrine disruption with a focus on the author's own data from studies in the pig.  相似文献

A study was performed on two horse farms to evaluate the use of age-clustered pooled faecal samples for monitoring worm control in horses. In total 109 horses, 57 on farm A and 52 on farm B, were monitored at weekly intervals between 6 and 14 weeks after ivermectin treatment. This was performed through pooled faecal samples of pools of up to 10 horses of the groups 'yearlings' (both farms), '2-year-old' (two pools in farm A), '3-year-old' (farm A) and adult horses (four pools on farm A and five pools on farm B), which were compared with the mean individual faecal egg counts of the same pools. A very high correlation between the faecal egg counts in pooled samples and the mean faecal egg counts was seen and also between the faecal egg counts in pooled samples and larval counts from pooled faecal larval cultures. Faecal egg counts increased more rapidly in yearlings and 2-year-old horses than in older horses. This implied that in these groups of young animals faecal egg counts of more than 200 EPG were reached at or just after the egg reappearance period (ERP) of 8 weeks that is usually indicated for ivermectin. This probably means that, certainly under intensive conditions, repeated treatment at this ERP is warranted in these young animals, with or without monitoring through faecal examination. A different situation is seen in adult animals. Based on the mean faecal egg counts on both farms and on the results of pooled samples in farm A, using 100 EPG as threshold, no justification for treatment was seen throughout the experimental period. However, on farm B values of 100 EPG were seen at 9 and 11, 13 and 14 and 14 weeks after ivermectin treatment in pools 10, 12 and 13, respectively. This coincided with the presence of one or two horses with egg counts above 200 EPG. The conclusion is that random pooled faecal samples of 10 adult horses from a larger herd, starting at the ERP and repeating it at, for instance, 4-week intervals, could be used for decisions on worm control. However, there would be a certain risk for underestimating pasture contamination through missing high-egg excreters. An alternative use of pooled samples would be as a cheap first screening to detect which adult horses really contribute to pasture contamination with worm eggs on a farm. All horses should be sampled and subsequently animals from 'positive' pools can be reexamined individually.  相似文献

A total of 120 pigs from farm I and 109 from farm II were examined for Dde I polymorphism in myf3 and for Msp I polymorphism in myogenin genes. Meatness, weight and ratio of ham and loin meat, and loin eye area of animals with different genotypes for myf3 and for myogenin were compared. In myf3 polymorphism pigs A/A from farm I displayed significantly lower values for these characteristics. In farm II there was a reverse tendency. Present results indicate that the Dde I polymorphic region in porcine myf3 gene does not influence functionality of this gene but it may point at another polymorphism located nearby and being of functional importance. Two regions of the porcine myogenin gene were analysed for Msp I polymorphism. The region encompassing coding sequences ( MYOG1 ) was polymorphic only in three Pietrain pigs (out of 150 individuals tested) making it useless for assessment of meat productivity in pigs. The 3' untranslated region was polymorphic and the frequency of 4.2 and 4.9 alleles varied among breeds. The 4.2 variant was predominant in Pietrain pigs. Statistical analysis indicated that meat, ham and loin meat percentages as well as loin eye area were significantly higher in 4.2/4.2 homozygotes. Further study will be undertaken to explain the relations between meat productivity and myf3 and MYOG2 alleles within breed.  相似文献

The objective of this field study was to evaluate in an endemically porcine reproductive and respiratory syndrome (PRRS) virus-infected farm the reproductive performance of sows after their vaccination with a PRRS attenuated vaccine. In a farrow-to-finish pig farm with history of endemic PRRS virus infection, a total of 200 gilts and sows were used. They were divided in 2 groups of 100 animals. The first group was used as untreated controls, while the animals of the second group were vaccinated against PRRS virus using the attenuated Porcilis PRRS vaccine (Intervet International, The Netherlands) based on European strain. All health and reproductive parameters were recorded from the time of vaccination up to next weaning. No adverse systemic or local reactions or side effects relative to vaccination were noted. Compared to controls, vaccinated sows showed significantly improved farrowing rate (89% versus 78%) and a tendency for fewer returns to oestrus, particularly those at irregular intervals. Fewer sows farrowed prematurely and showed post-partum dysgalactia syndrome, but more live pigs were born and weaned in each litter after vaccination. It was concluded that vaccination of sows with Porcilis PRRS attenuated vaccine in farms with endemic PRRSV infection has beneficial effects on their health and fertility.  相似文献

Epigenetics provides a molecular mechanism of inheritance that is not solely dependent on DNA sequence and that can account for non-Mendelian inheritance patterns. Epigenetic changes underlie many normal developmental processes, and can lead to disease development as well. While epigenetic effects have been studied in well-characterized rodent models, less research has been done using agriculturally important domestic animal species. This review will present the results of current epigenetic research using farm animal models (cattle, pigs, sheep and chickens). Much of the work has focused on the epigenetic effects that environmental exposures to toxicants, nutrients and infectious agents has on either the exposed animals themselves or on their direct offspring. Only one porcine study examined epigenetic transgenerational effects; namely the effect diet micronutrients fed to male pigs has on liver DNA methylation and muscle mass in grand-offspring (F2 generation). Healthy viable offspring are very important in the farm and husbandry industry and epigenetic differences can be associated with production traits. Therefore further epigenetic research into domestic animal health and how exposure to toxicants or nutritional changes affects future generations is imperative.  相似文献
