Early bacterial colonization and succession within the gastrointestinal tract have been suggested to be crucial in the development of host immunity. In this study, we have investigated the changes in live weight and concentrations of selected serum parameters in relation to their fecal bacterial communities as determined by high throughput sequencing of the 16S rRNA gene over the same period in lambs. The results showed that lambs’ growth performance, the serum parameters, fecal bacterial community and fecal bacterial functions were all affected (P<0.05) by age of the lambs. Similarity within age groups of fecal microbiota was lower in the preweaning period and increased sharply (P<0.05) after weaning at 60 days. The similarity between the samples collected from birth to 90 days of age and those collected at 120 days of age, increased (P<0.05) sharply after 30 days of age. Some age-associated changes in microbial genera were correlated with the changes in concentrations of immune indicators, including negative (P<0.05) correlations between the relative abundance of Lachnospiraceae UCG-010, Eubacterium coprostanoligenes group, Ruminococcaceae UCG-005, Ruminococcaceae UCG-009, Ruminococcaceae UCG-013, Ruminiclostridium 6, Ruminococcaceae UCG-008, and Oscillibacter with serum concentrations of lipopolysaccharide (LPS), D-lactate dehydrogenase (DLA), immunoglobulin (IgA, IgM, and IgG), and cytokines (interleukin-1β (IL-1β), IL-6, IL-12, and IL-17), tumor necrosis factor-α (TNF-α), and the relative abundance of these genera increased from 45 days of age. In conclusion, these results suggested that the age-related abundances of particular genera were correlated with serum markers of immunity in lambs, and there might be a critical window in the period from birth to 45 days of age which provide an opportunity for potential manipulation of the fecal microbial ecosystems to enhance immune function.  相似文献

Early life intervention is important to shape the gut microbiome profiles of adult animals due to the tremendous alteration of diet components. Nevertheless, there is still no unified understanding about its long-term effects in lambs. In this study, sixty 20-day-old lambs were assigned into ewe-rearing (ER) and artificial-rearing (AR) treatments to evaluate the effects of AR strategy on ruminal microbiota, fermentation, and morphology of pre-weaning lambs (from 20 to 60 days of age) and its long-term effects in the fattening stage (from 61 to 180 days of age). During the pre-weaning stage, ER lambs were breastfed and supplemented starter, while AR lambs were artificially fed with milk replacer and starter. During the fattening stage, all lambs in both treatments were fed with the same fattening diets. At 60, 120 and 180 days of age, 6 lambs from each group were slaughtered to collect rumen content and tissue samples. Compared with ER lambs, the dry matter feed intakes of AR lambs increased (P<0.05) from 20 to 180 days of age, companying an increased average daily gain (ADG) from 61 to 120 days of age (P<0.05) and from 121 to 180 days of age (0.05<P<0.1). Although there was no difference in short-chain fatty acid (SCFA, including acetate, propionate, and butyrate) between treatments before weaning (P>0.05), it was higher (P<0.05) in AR lambs compared with ER lambs at the fattening stage. The rumen keratin layer of AR lambs was thinner (P<0.05) than that of ER lambs. Along with lamb growth from 60 to 180 days of age, the differences in rumen bacterial diversity between AR and ER treatments grew more distinct (P<0.05). Compared with ER lambs, AR lambs increased (P<0.05) rumen bacteria abundance, such as phylum Spirochaetes and genus Treponema at 60 days of age, phylum Actinobacteria and genus Succiniclasticum at 120 days of age, and phylum Proteobacteria at 180 days of age, but decreased genus Selenomonas from 60 to 180 days of age, and Anaerovibrio at 180 days of age. In summary, the early interventions before weaning could improve dry matter feed intake of lambs, which triggered robust rumen development and produced positive long-term effects on rumen fermentation and noticeable weight gain of fattening lambs. It suggests that the artificial rearing strategy is effective in improving rumen fermentation and microbial maturity of intensive fattening lambs.  相似文献

This study investigated the effects of dioscorea opposite waste (DOW) on the growth performance, blood parameters, rumen fermentation and rumen microbiota of weaned lambs. Sixty healthy weaned Small-Tailed Han lambs (male, (22.68±2.56) kg initially) were used as the experimental animals. Four levels of concentrate: 0 (control, CON), 10% (DOW1), 15% (DOW2) and 20% (DOW3), were replaced with DOW in the basal diet as experimental treatments. The results showed that lambs fed the DOW2 diet had a higher (P<0.05) dry matter intake (DMI) than the other groups. There was no significant difference (P>0.05) among DOW groups in average daily weight gain (ADG), and replacing concentrate with DOW linearly or quadratically increased (P<0.05) the ADG, while lambs fed the DOW2 diet showed greater (P<0.05) ADG than the CON group. The relative plasma concentration of growth hormone (GH), insulin like growth factor-1 (IGF-1) and insulin were affected by DOW, replacing concentrate with DOW linearly or quadratically (P<0.05) enhanced the plasma concentration of GH, IGF-1 and insulin, which was significantly higher (P<0.05) in the DOW2 group than in the CON, DOW1 and DOW3 groups. In addition, the DOW treatment showed a lower (P<0.05) concentration of blood urea nitrogen (BUN) than the CON group. Replacing concentrate with DOW quadratically decreased (P<0.05) the ruminal ammonia nitrogen (NH3-N) and increased (P<0.05) the total of volatile fatty acids (TVFAs) at 0 and 4 h after feeding as well as linearly decreased (P<0.05) the NH3-N at 8 h after feeding. Replacing concentrate with DOW linearly decreased (P<0.05) the propionate and increased the aceate before feeding, and linearly decreased (P<0.05) propionate and quadratically increased (P<0.05) the aceate at 4 and 8 h after feeding. Lambs fed the DOW2 diet increased the phylum Firmicutes and genera Succiniclasticum and Ruminococcus_1 groups, whereas decreased (P<0.05) the relative abundance of phylum Deferribacteres and genera intestinimonas and Ruminiclostridium. In summary, replacing the concentrate with 15% DOW was beneficial for improving the rumen fermentation and ADG by increasing the DMI and modulating the rumen microbial community.  相似文献

The objective of this study was to investigate the effect of lairage after transport on post mortem muscle glycolysis, protein phosphorylation and lamb meat quality. Two preslaughter animal treatments, transport for 3 h and lairage for 0 h (T3L0) and transport for 3 h and then lairage for 12 h (T3L12), were compared with a control treatment of 0 h transport and 0 h lairage. Data obtained showed that preslaughter transport had a significant effect on lamb meat quality. Loins from lambs of the T3L0 treatment showed higher (P=0.026) pH24 h and higher (P=0.021) pH48 h values, but lower (P<0.001) drip loss and lower (P<0.05) glycolytic potential at 0 h post mortem than those of the T3L12 and control groups. Muscle samples of the T3L0 group showed higher (P=0.046) shear force and lower (P=0.005) b* value than those of the T3L12 group. Muscle glycogen concentration at 0, 2, 4 h post mortem were lower (P<0.05) in the T3L0 group than in control. No significant difference (P>0.05) in most meat quality parameters was determined between the T3L12 group and control, showing lairage for 12 h allowed lambs to recover from the effects of transport for 3 h and resulted in similar meat quality characteristics compared to no transport. Lairage after transport did not affect most meat quality indices in comparison with control, but increased the meat drip loss and b* value of lambs possibly through decreasing glycogen concentration and glycolytic potential.  相似文献

The digestive tract of termite(Microcerotermes diversus) contains a variety of lignocellulose-degrading bacteria with exocellulases enzyme activity, not found in the rumen, which could potentially improve fiber degradation in the rumen. The objectives of the current study were to determine the effect of inoculation of rumen fluid(RF) with three species of bacteria isolated from termite digestive tract, Bacillus licheniformis, Ochrobactrum intermedium, and Microbacterium paludicola, on in vitro gas production(IVGP), fermentation parameters, nutrient disappearance, microbial populations, and hydrolytic enzyme activities with fibrous wheat straw(WS) and date leaf(DL) as incubation substrate. Inoculation of RF with either of three termite bacteria increased(P0.05) ammonia-N concentration compared with the control group(free of termite gut bacteria). Termite bacteria inoculation had no effect(P0.05) on gas production characteristics, dry matter, organic matter and neutral detergent fiber disappearance, pH, and concentration and composition of volatile fatty acids. Population of proteolytic bacteria and protozoa, but not cellulolytic bacteria, were increased(P0.05) when RF was inoculated with termite bacteria with both WS and DL substrates. Inoculation of RF with termite bacteria increased protease activity, while activities of carboxymethyl-cellulase, microcrystalline-cellulase, α-amylase and filter paper degrading activity remained unchanged(P0.05). Overall, the results of this study indicated that transferring lignocellulose-degrading bacteria, isolated from digestive tract of termite, to rumen liquid increased protozoa and proteolytic bacteria population and consequently increased protease activity and ammonia-N concentration in vitro, however, no effect on fermentation and fiber degradation parameters were detected. These results suggest that the termite bacteria might be rapidly lysed by the rumen microbes before beneficial effects on the rumen fermentation process could occur.  相似文献

The objective of this study was to observe the forage yield, silage fermentative quality, anthocyanin stability, and antioxidant activity during the storage period and in vitro rumen fermentation of anthocyanin-rich purple corn (Zea mays L.) stover (PS) and sticky corn stover (SS). Forage yield of corn stover was weighed and ensiled with two treatments: (1) hybrid sticky waxy corn stover (control), and (2) hybrid purple waxy corn stover (treatment). Samples were stored in mini-silos for periods of 0, 7, 14, 21, 42, 63, 84, and 105 d. The results showed that PS had significantly higher (P<0.05) yields of dry matter (DM), organic matter (OM), gross energy (GE), crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), and total anthocyanins than that of the SS. Anthocyanin-rich purple corn stover silage (PSS) showed higher (P<0.05) levels of DM and CP relative to the sticky corn stover silage (SSS). Although anthocyanin-rich PSS displayed a lower (P<0.05) level of pelargonidin-3-glucoside (P3G), it had higher (P<0.05) levels of peonidin (Peo) and pelargonidin (Pel) compared to the control. Delphinidin (Del) and malvidin (Mal) were not detected in SSS during the ensilage period; in PSS, Del was no longer detected after 7 d of ensilage. Specifically, total anthocyanins in anthocyanin-rich PSS decreased rapidly (P<0.05) prior to 7 d of ensilage, and then remained at relatively stable (P>0.05) constants. Compared to the anthocyanin-rich PSS, SSS displayed significantly higher (P<0.05) pH value and ammonia nitrogen (NH3-N) content. Propionic acid (PA) at 0 d and butyric acid (BA) during the entire study period were not detected, whereas anthocyanin-rich PSS showed a higher (P<0.05) level of lactic acid (LA) than that of the SSS. Compared with the SSS extract, anthocyanin-rich PSS extract showed a higher (P<0.05) level of 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and displayed a lower (P<0.05) half maximal inhibitory concentration (IC50) value. Moreover, anthocyanin-rich PSS reduced (P<0.05) gas production (GP), and displayed lower levels of immediately soluble fraction and ratio of acetic acid (AA) to PA at 12 h, but the other parameters were unaffected (P>0.05) relative to the control. Taken together, the results indicated that: (1) anthocyanins could be stable in silage; (2) anthocyanin-rich PSS showed better silage fermentative quality and stronger antioxidant activity; and (3) anthocyanin-rich PSS had no negative effect on rumen fermentation parameters.  相似文献

Grain-induced subacute ruminal acidosis (SARA) impairs rumen epithelial barrier function, but it is yet to be determined if SARA can cause persistent damage to the morphology and function of the rumen epithelial barrier. The objective of the present study was to investigate if SARA has persistent effects on the morphological structure and permeability of ruminal epithelium and the expression of the genes involved in epithelial barrier function using a lactating goat model. Twelve mid-lactating Saanen goats with rumen cannulas were randomly assigned to 1 of 2 groups: control group (Ctrl, n=4) fed a basal diet with a non-fiber carbohydrate (NFC) to neutral detergent fiber (NDF) ratio of 1.40, and SARA group (SARA, n=8) fed the same basal diet but with increasing NFC to NDF ratio from 1.4 to 1.79, 2.31, and 3.23 overtime to induce SARA. At the end of the SARA challenge (post-SARA), 4 goats were randomly selected from the SARA group and fed only hay mixture adlibitum for another 4 weeks to allow for restitution (post-SARA). Ruminal pH was continuously recorded to monitor the severity of SARA. Samples of the ventral ruminal epithelium were collected after slaughter to examine the structural and functional changes of the ruminal epithelium using transmission electron microscopy (TEM), Ussing chambers, qRT-PCR, and Western bolt analyses. Compared with the Ctrl group, ruminal papilla length, width, surface area and thickness of stratum corneum increased (P<0.05), while stratum spinosum and basale thickness, and total depth of the epithelium decreased (P<0.05) in the SARA group. These changes diminished or tended to return to the levels of the Ctrl group in the post-SARA group (P>0.05). The SARA challenge also decreased cellular junction and widened the intercellular space between epithelial cells. Rumen transepithelial short-circuit current (Isc), tissue conductance (Gt), and mucosa-to-serosa flux of paracellular horseradish peroxidase (HRP) all increased (P<0.05) both in the SARA and post-SARA groups, which indicates that SARA can induce a sustained increase in epithelial permeability and barrier dysfunction. Moreover, the mRNA and protein expressions of CLDN1, OCLN and ZO-1 were down-regulated (P<0.01) in both the SARA and post-SARA groups. The results of this study showed that SARA could result in sustained epithelial barrier dysfunction, at both structural and functional levels, which is associated with decreased expression of rumen epithelial tight junction proteins, and the restitution of rumen epithelial barrier function is slower than that of its morphology.  相似文献

为了解巴音布鲁克高寒湿地土壤真菌群落对退化的响应,探索湿地退化机制,本研究以巴音布鲁克高寒湿地未退化(ND)、轻度退化(SD)及重度退化(HD)区域的土壤为研究对象,采用高通量测序技术分析不同程度退化高寒湿地土壤真菌群落的多样性及结构差异,并结合理化指标进一步分析影响真菌群落的环境因子。结果表明:真菌群落Simpson和Shannon多样性指数在ND和HD区土壤中无显著差异(P>0.05);SD区土壤中的真菌群落Simpson指数显著高于ND和HD区,而Shannon指数显著低于ND和HD区(P<0.05)。在不同程度退化区土壤中,真菌群落ACE和Chao1丰富度指数均随退化的加剧呈降低趋势。在3个区域土壤中,门水平上,子囊菌门(Ascomycota)的相对丰度最高;纲水平上,粪壳菌纲(Sordariomycetes)为主导类群;肉座菌目(Hypocreales)、Pleosporales菌目、被孢霉目(Mortierellales)的相对丰度均随退化程度的增加而增加;属水平上,珊瑚菌属(Clavaria)、稻瘟病菌属(Magnaporthe)为优势菌属。LEfSe分析发现了11种可作为判断退化的潜在生物标志物。RDA分析及蒙特卡罗检验结果显示,在门水平上真菌群落结构变化不受环境因子的显著影响;在属水平上,真菌群落结构变化仅与土壤总有机碳存在显著相关性(P<0.05)。研究表明,高寒湿地退化显著改变了土壤pH、土壤含水量、总有机碳、容重等理化指标,进而影响真菌群落多样性和结构。  相似文献

Isolated ruminal epithelia from caudal blind sacs of dairy goats were incubated with butyrate and insulin-like growth factor-1 (IGF-1) at different concentrations. Proportions of ruminal epithelium in different phases of the cell division cycle were determined by flow cytometric analysis. The proportion of epithelial cells in S phase and G2-M phase (PS&G2-M) increased significantly (P<0.01) whereas the proportion of epithelial cells in G0-G1 phase (PG0-G1) decreased after incubation with IGF-1. PS&G2-M decreased whereas PG0-G1 increased markedly (P<0.01) after incubation with sodium butyrate. PS&G2-M incubated with IGF-1 and butyrate sodium together increased more than that incubated with IGF-1 alone; PG0-G1, however, decreased significantly (P<0.01). Our results indicate that IGF-1 enhances whereas sodium butyrate inhibits the proliferation of rumen epithelial cells. Furthermore, butyrate and IGF-1, together, have a synergic effect on the proliferation of rumen epithelium.  相似文献

为了研究冬季补饲对妊娠期藏母羊瘤胃挥发性脂肪酸(volatile fatty acid,VFA)、微生物区系及羔羊生长发育影响.采用单因子试验设计,随机选择健康藏母羊440只,分为2组.对照组(control check,CK)为放牧饲喂,处理组(test,T)在放牧基础上,每只母羊补饲0.1 kg·d-1精料补充料,试验期为45 d(配种后106~150 d).试验结束后,从处理组和对照组中随机选择5只和6只母羊屠宰后分析瘤胃挥发性脂肪酸和细菌菌群组成.同时测定新生羔羊的生长发育指标.结果 表明:(1)处理组羔羊的体重、体高和胸围显著高于对照组(P<0.05);(2)处理组瘤胃液总挥发性脂肪酸(Total volatile fatty acids,TVFA)、丙酸和丁酸含量显著高于对照组(P<0.05),同时乙酸和异丁酸显著低于对照组(P<0.05);(3)两组母羊瘤胃微生物在门和属水平上组成相似,拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)以及疣微菌门(Verrucomicrobia)为优势菌门;而普雷沃菌属-1(Prevotella l)、理研菌科RC9肠道群(Rikenellaceae-RC9-gut-group)、普雷沃氏菌科-UCG-001(Prevotellaceae-UCG-001)为优势菌属;(4) Alpha多样性指数中,对照组的Shannon指数显著高于处理组(P<0.05),说明对照组瘤胃微生物多样性优于处理组;(5)瘤胃微生物功能预测发现,两组母羊瘤胃微生物主要富集于复制和修复(Replication and Repair)、碳水化合物代谢(Carbohydrate Metabolism)和信号转导(Signal Transduction).综上,妊娠后期藏母羊的冬季补饲能够为胎儿的生长发育提供必要的营养需求,同时提高母羊瘤胃液中TVFA、丙酸和丁酸含量,但在一定程度上影响了瘤胃细菌的多样性.  相似文献

Five lactating Holstein cows in a 5×5 Latin square experiment were fed five high-concentrate total mixed rations(TMRs) to investigate the effects of step-wise improvement of forage combination on ruminal and milk fatty acid profiles. The ratio of concentrate to forage was fixed as 61:39, and the step-wise improvement of forage combination was applied as: TMR1, a ration containing corn stover; TMR2, a ration containing corn stover and ensiled corn stover; TMR3, a ration containing ensiled corn stover and Chinese wild ryegrass hay(Leymus chinensis); TMR4, a ration containing the ryegrass hay and whole corn silage; TMR5, a ration containing the ryegrass hay, whole corn silage and alfalfa hay. The TMRs were offered to the cows twice daily at 0700 and 1900 h. The entire experiment was completed in five periods, and each period lasted for 18 days. Diurnal samples of rumen fluids were collected at 0100, 0700, 1300 and 1900 h(day 16); 0300, 0900, 1500 and 2100 h(day 17); and 0500, 1100, 1700 and 2300 h(day 18). The step-wise improvement of forage combination increased energy and crude protein contents and decreased fibre content. As a result, the step-wise improvement of forage combination increased dry matter intake and milk yield(P0.05). The step-wise improvement increased dietary content of linolenic acid(C18:3 n-3), but did not alter dietary proportions of palmitic acid(C16:0), stearic acid(C18:0), oleic acid(C18:1 cis-9), linoleic acid(C18:2 n-6) and arachidic acid(C20:0). In response to the forage combination, ruminal concentration of C16:0, C18:2 n-6 and C18:3 n-3 linearly increased against their dietary intakes(P0.10). The step-wise improvement increased milk contents of C10:0, C12:0, C14:0, C16:0, C18:2 n-6 and C18:3 n-3(P0.10) and decreased milk contents of C18:0 and C18:1 cis-9(P0.05). Milk yields of C16:0, C18:1 cis-9, C18:2 n-6 and C18:3 n-3 were linearly increased by the increase of these fatty acids in the rumen(R2≥0.79, P0.05), and milk yields of C18:2 n-6 and C18:3 n-3 were also positively correlated with dietary intake of these fatty acids(R2≥0.85, P0.05). The step-wise improvement increased the transfer efficiencies from feed to milk for C18:2 n-6 from 11.8 to 14.2% and for C18:3 n-3 from 19.1 to 22.3%. In a brief, along with the step-wise improvement of forage combination, more dietary linoleic and linolenic acids might escape microbial hydrogenation in the rumen and consequently accumulated in milk fat though these fatty acids were present in low concentrations in ruminal fluids. The step-wise improvement of forage combinations could be recommended as a dietary strategy to increase the transfer efficiency of linoleic and linolenic acids from feed to milk.  相似文献

为研究集约化饲养模式下巴美肉羊与小尾寒羊的生长性能、屠宰性能及肉品质指标,试验选择4月龄体况良好、体重相近的巴美肉羊和小尾寒羊公羔各10只,按品种分为2组,饲喂以精粗质量比为50∶50(干物质基础)的全混合日粮(TMR)。测定各试验羊饲养120d期间的相关生长性能指标和相关屠宰性能及肉品质指标。结果表明:1)巴美肉羊的干物质采食量、试验末体重、总增重和日增重均极显著高于小尾寒羊(P0.01);2)巴美肉羊宰前活重(P0.01)、热胴体重(P0.05)和眼肌面积(P0.05)均显著高于小尾寒羊,而屠宰率则显著低于小尾寒羊(P0.01);3)巴美肉羊背最长肌宰后45min的pH(P0.05)、宰后1h的红色度(a*)和黄色度(b*)(P0.01)以及宰后24h的红色度(a*)(P0.05)等指标均显著高于小尾寒羊,而巴美肉羊背最长肌的剪切力、滴水损失和蒸煮损失等指标显著低于小尾寒羊(P0.05)。由此可见,在以TMR日粮为主要饲喂方式的集约化饲养模式下,巴美肉羊比小尾寒羊能发挥更高的生产性能并生产出品质更佳的羊肉。  相似文献

The bone morphogenetic protein (BMP) and mitogen-activated protein kinase (MAPK) signaling pathways play an important role in regulation of bone formation and development, however, it remains unclear that the effect of dietary different levels of non-phytate phosphorus (NPP) on these signaling pathways and their correlations with bone phosphorus (P) retention and bone development in broilers. Therefore, this experiment was conducted to investigate the effect of dietary P supplementation on BMP and MAPK signaling pathways and their correlations with bone P retention and bone development in broilers. A total of 800 one-day-old Arbor Acres male broilers were randomly allotted to 1 of 5 treatments with 8 replicates in a completely randomized design. The 5 treatments of dietary NPP levels were 0.15, 0.25, 0.35, 0.45 and 0.55% or 0.15, 0.22, 0.29, 0.36 and 0.43% for broilers from 1 to 21 days of age or 22 to 42 days of age, respectively. The results showed that extracellular signal-regulated kinase 1 (ERK1) mRNA expression in the tibia of broilers on days 14 and 28, phosphorylated-ERK1 (p-ERK1) on day 14, and BMP2 protein expression on days 28 and 42 decreased linearly (P<0.04), while c-Jun N-terminal kinase 1 (JNK1) mRNA expression on day 42 increased linearly (P<0.02) with the increase of dietary NPP level. At 14 days of age, total P accumulation in tibia ash (TPTA), bone mineral concentration (BMC), bone mineral density (BMD), bone breaking strength (BBS) and tibia ash were negatively correlated (r=–0.726 to –0.359, P<0.05) with ERK1 and JNK1 mRNA as well as p-ERK1; tibia alkaline phosphatase (ALP) and bone gal protein (BGP) were positively correlated (r=0.405 to 0.665, P<0.01) with ERK1 mRNA and p-ERK1. At 28 days of age, TPTA, BMC, BMD, BBS and tibia ash were negatively correlated (r=–0.518 to –0.370, P<0.05) with ERK1 mRNA and BMP2 protein, while tibia ALP was positively correlated (r=0.382 to 0.648, P<0.05) with them. Twenty-eight male, weaned Chinese Holstein calves((156.8±33.4) kg) were used to investigate the effects of dietary forage to concentrate ratio(F:C) and forage length on nutrient digestibility, plasma metabolites, ruminal fermentation, and fecal microflora. Animals were randomly allocated to four treatments in a 2×2 factorial arrangement: whole-length forage(WL) with low F:C(50:50); WL with high F:C(65:35); short-length forage(SL) with high F:C(65:35); and SL with low F:C(50:50). Chinese wildrye was used as the only forage source in this trial. The grass in the SL treatments was chopped using a chaff cutter to achieve small particle size(~50% particles 19 mm). Dry matter intake(DMI) and organic matter(OM) intake was increased by increasing both F:C(P0.01) and forage length(FL)(P0.05), while acid detergent fiber(ADF) and neutral detergent fiber(NDF) intakes were only increased by increasing the F:C(P0.01). The digestibility of NDF was increased as the FL increased(P0.01), and it was also affected by interaction between F:C and FL(P0.05). Cholesterol(CHO)(P0.01), leptin(LP)(P0.05), and growth hormone(GH)(P0.01) concentrations in plasma were increased as dietary F:C increased. A significant increase in plasma triglyceride(TG)(P0.01), insulin(INS)(P0.05), and GH(P0.01) levels was observed with decreasing dietary FL. Ruminal p H values of calves fed with low F:C diets were significantly lower than those in high F:C treatment(P0.05). Increasing the F:C enhanced ruminal acetic acid(P0.05) and acetic acid/propionic acid(P0.01). Fecal Lactobacillus content was significantly higher, while Escherichia coli and Salmonella contents were significantly lower in WL and high F:C groups(P0.05). Lower fecal scores(higher diarrhea rate) were observed in calves fed with SL hay compared to WL hay(P0.05). Denatured gradient gel electrophoresis(DGGE) bands and richness index(S) were significantly affected by the interaction between F:C and FL(P0.05), under high F:C, band numbers and richness index from WL group were higher than that from SL group(P0.05), whereas there were no differences between WL andSL groups under low F:C(P0.05). Microflora similarity was 50–73% among the different treatments. It is concluded that the WL with high F:C(65:35) diet is suitable for weaned calves.  相似文献

Eight Chinese Holstein cows were used in a 4 × 4 Latin square design to determine the effects of rumen protected methionine (RPMet) and Niacin on milk yield and milk composition of lactating cows with 14 d adaptation and 6-d sampling periods. The cows were fed the control diet or the control diet plus RPMet (25 g·−1), niacin (6 g·−1), or RPMet (25 g·−1) + niacin (6 g·−1). The results showed that RPMet supplementation in the diet increased 4% fat corrected milk (FCM) yield (P < 0.05) and milk fat percentage (P < 0.05). However, supplemental RPMet had no effect on the solid non fat (SNF) (P > 0.05) and lactose percentage (P > 0.05). Dietary niacin supplementation increased milk yield (P < 0.05), milk protein percentage (P < 0.05) and lactose percentage (P < 0.05), but had no effect on milk fat percentage (P > 0.05) and SNF (P > 0.05). RPMet supplementation in the diet of lactating cows significantly decreased serum urea nitrogen (P < 0.01) and glucose contents (P < 0.05), but had no effects on nonesterified fatty acids (NEFA), total cholesterol and triglyceride (P > 0.05). Niacin supplementation influenced the contents of glucose and NEFA in serum, but had no effects on the urea nitrogen, total cholesterol and triglyceride (P > 0.05).  相似文献

This experiment was conducted to evaluate the effects of Bupleurum extract(BE) on blood metabolites, antioxidant status, and immune function in dairy cows under heat stress. Forty lactating Holstein cows were randomly assigned to 1 of 4 treatments. The treatments consisted of 0, 0.25, 0.5, and 1.0 g of BE kg–1 dry matter. Supplementation with BE decreased(P0.05) blood urea nitrogen(BUN) contents and increased blood total protein(TP) and albumin(ALB) levels compared with control cows, but it had no effects(P0.05) on blood glucose(GLU), nonesterified fatty acid(NEFA), total triglyceride(TG), low-density lipoprotein cholesterol(LDL-C), and high density lipoprotein cholesterol(HDL-C). Compared with control cows, cows fed BE had higher(P0.05) superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) activity. However, supplementation with BE had no effect(P0.05) on total antioxidant capacity(T-AOC) or malondialdehyde(MDA) levels. The immunoglobulin(Ig) A and G contents increased(P0.05) in cows fed 0.25 or 0.5 g of BE kg–1. Interleukin(IL)-2 and IL-4 levels were higher(P0.05) in cows fed 0.5 and 1.0 g of BE kg–1, and IL-6 was significantly elevated(P0.05) in cows fed 0.5 g of BE kg–1. There were no treatment effects(P0.05) on the CD4+ and CD8+ T lymphocyte ratios, CD4+/CD8+ ratio, or tumor necrosis factor-α(TNF-α) level among the groups. These findings suggest that BE supplementation may improve protein metabolism, in addition to enhancing antioxidant activity and immune function in heat-stressed dairy cows.  相似文献

在不同季节分别随机选取3只藏系绵羊,采用PCR-DGGE技术对12只藏系绵羊瘤胃内容物中的细菌区系进行分子生态学分析.对古菌16SrDNA的V2~V4可变区的扩增产物进行DGGE图谱分析、相似性分析和Shannon多样指数分析.结果表明:瘤胃内容物中古菌群落多样性指数冬季与春季间差异不显著(P>0.05),但冬季显著高于夏季(P<0.05),并极显著高于秋季(P<0.01);春季与夏季差异不显著(P>0.05),春季显著高于秋季(P<0.05);夏、秋季之间差异不显著(P>0.05).各季间瘤胃古菌区系组成结构相似性高于94%.冬、春季节,由于牧草中的纤维性物质含量增高,导致瘤胃古菌群落多样性增加,不同季节牧草中粗蛋白和酸性洗涤纤维含量的变化可以引起特异性的古菌群落发育,可能导致不同的优势种群.  相似文献

The study was conducted to evaluate the effects on subsequent performance, carcass traits, jejunal histomorphology for broilers subjected to various time of feeding prestarter diet. For this purpose three hundreds day old Cobb 500 chickens were used. Chickens were grouped as a completely randomized design with 4 treatments and 4 replications. Prestarter diet was fed to 4, 6, 8 or 10 days of age. Starter diet was fed for variable times depending upon termination of feeding prestarter. Finisher diet was fed beginning at 22 days of age. Diets were identical in terms of all the nutrients levels. Morphometric indices of jejunum were measured at 10 days of age and the end of the production period. Increasing the duration of feeding prestarter diet significantly increased body weight and decreased FCR at 10 days of age (P < 0.05). However, treatments' effect on final BW was not significant (P > 0.05). The FCR of birds fed the prestarter for 10 days significantly lower from that of birds fed the prestarter for 4 days at the end of experiment (P < 0.05). The time of feeding prestarter diet had no significant effect on feed intake (P > 0.05). There was no significant effect of time of prestarter diet on dressing percentage, breast meat, legs or wings weight (P > 0.05). Different feeding strategies had significant effect on intestinal histomorphology (P < 0.05). Birds fed prestarter diet for the first 10 days had higher villi width and villus surface area (VSA) compared with other treatments at 10 days of age. Based on the data under the experimental conditions, the optimum time of feeding prestarter diet for birds grown to a target weight of approximately 2.2 kg appears to be 10 days.  相似文献

Two experiments were conducted to investigate the effect of in ovo zinc(Zn) injection on the embryonic development, tissue Zn contents, antioxidation and related gene expressions of fertilized eggs of Arbor Acres broiler breeders. Experiment 1 was conducted to determine an optimal embryonic age for early in ovo injection. A total of 720 fertilized eggs with similar weights were randomly allotted to 4 treatments with 6 replicates per treatment and 30 eggs per replicate in a completely randomized design. The eggs were injected with 0.1 m L sterilized water at 3, 6 and 9 embryonic days of incubation(E3, E6 and E9) or non-injection(the control), respectively. The results from experiment 1 showed that E3 and E6 injections increased(P0.05) the embryonic mortalities, and decreased(P0.05) hatchabilities compared to the non-injected control, but no differences(P0.05) between E9 injection and the non-injected control were observed in either embryonic mortality or hatchability. The findings suggest that the E9 is the optimal embryonic age for early in ovo injection. In experiment 2, a total of 672 fertilized eggs with similar weights were randomly allocated to 7 treatments with 6 replicates per treatment and 16 eggs per replicate in a completely randomized design. The eggs were injected with 0(the negative control), 50, 100, 150, 200, or 250 μg Zn/egg as reagent grade Zn SO4·7 H2 O in a 0.1-m L solution, or non-injection(the positive control), respectively at E9–10. The results from the experiment 2 demonstrated that no differences(P0.05) among 50 and 100 μg Zn/egg groups and the negative control were observed in the embryonic mortality and hatchability, however, the injection of 200 μg Zn/egg increased(P0.05) the embryonic mortality, and injections of 150 and 200 μg Zn/egg decreased(P0.05) hatchabilities compared with the controls. The embryonic tibia Zn contents at E20 were increased(P0.05) by injections of 150, 200 and 250 μg Zn/egg. Zinc injection did not affect(P0.05) malonaldehyde(MDA) contents, copper-and Zncontaining superoxide dismutase(Cu Zn SOD) activities and m RNA expression levels in the liver and heart of chick embryos at E15 and E20. Compared with the negative control, injections of 50, 150 and 200 μg Zn/egg up-regulated(P0.05) the metallothionein(MT) mR NA expression levels in the embryonic liver at E20. These results indicated that in ovo Zn injections increased Zn contents in the embryonic tibia and MT m RNA expression levels in the embryonic liver at E20, however, injections of 150–200 μg Zn/egg were harmful to the embryonic development.  相似文献
