Young leaves of 62 citrus cultivars were inoculated with conidia of three Spanish isolates of Alternaria alternata pv. citri, the causal agent of brown spot of citrus. Hybrids with Dancy mandarin, King mandarin or their derivates as a parent, grapefruit cultivars and the mandarin cultivars Guillermina, Emperor, Clemenpons and Esbal were highly susceptible to the pathogen. Satsuma cultivar Clausellina and orange cultivars, with the exception of Sanguinelli, were slightly susceptible. Lemon and lime cultivars were not susceptible, with the exception of Mexican lime (Citrus aurantifolia), which was slightly susceptible. Although this study shows a range of potential hosts for this pathogen, to date the only affected cultivars in Spain are Fortune and Nova mandarins, and Minneola tangelo. The susceptibility of Fortune fruits decreased as diameter increased, being susceptible through the whole season. This was confirmed with field observations in autumn where fruit infections have been detected when the diameter reaches 6–7 cm.

Leaf blight disease was found on Gloriosa superba L. (Liliaceae), an endangered, herbaceous, perennial, climbing lily that produces colchicine, in West Bengal, India in 2004. Small brownish spots on leaves developed into concentric rings, which eventually darkened and coalesced to blight the entire leaf. The causal fungus was morphologically identified as Alternaria alternata (Fr.) Keissler. This is the first record of A. alternata on G. superba.

A new disease of peach (Prunus persica Batsch var. vulgaris Maxim.), causing brown, sunken lesions and brownish to blackish brown spots with cracks on peach fruits, was found in Okayama prefecture, Japan, in 1995. The disease was observed not only on peach fruits but also on twigs and leaves. An Alternaria sp. was consistently isolated from these diseased fruits, twigs and leaves. The isolates were pathogenic to peach fruits and leaves. Based on the morphological characteristics, the causal fungus was identified as Alternaria alternata (Fr.) Keissler. After cross-inoculation with isolates from peach, Japanese pear and apple, the isolates were found to be pathogenic only to their original host. This is the first report on a peach disease caused by a host-specific A. alternata; therefore, the common name of black spot (`Kokuhanbyo' in Japanese) was proposed. Branch cankers on citrus trees in Spain caused by Phytophthora citrophthora
Considerable losses of citrus trees have been observed in the major citrus-growing areas of Spain. Samples were collected from 132 orchards, and isolations and pathogencity tests were conducted to determine the aetiology of a serious canker disease. Affected trees showed cankers on the scion that frequently began on the branches. Three Phytophthora species were identified based on their morphological, cultural, physiological and molecular profiles. Phytophthora citrophthora was the main species associated with this new syndrome in 114 orchards. Phytophthora nicotianae (syn. P. parasitica ) was isolated from nine orchards as the sole Phytophthora species and in coinfection with P. citrophthora from another nine orchards. Phytophthora citricola was isolated only from one orchard. In stem-inoculation studies conducted under greenhouse conditions, clementine mandarin cv. Hernandina and sweet orange cv. Navel Late were more susceptible to P. citrophthora than sour orange and Carrizo citrange rootstocks. Clementine cv. Hernandina was also highly susceptible in field inoculation experiments. In agreement with field surveys, clementine mandarin cultivars were the most affected, their rootstocks remaining healthy. Phytophthora citrophthora was found to be the predominant species in orchard soils; however, P. nicotianae was also isolated. This information changes the scenario of diseases caused by Phytophthora spp. in Spain and consequently, the present knowledge of epidemiology and the effectiveness of the current control measures should be reassessed.

The sterol biosynthesis inhibitors bromuconazole and difenoconazole and tank mixes of each fungicide with captan were applied to apples and evaluated as controls for moldy-core and fruit decay caused by Alternaria alternata. Effectiveness of a mixture of bromuconazole and captan in controlling colonization by the fungus was also evaluated. Decay formation by A. alternata on mature detached fruits was partially inhibited by bromuconazole at 0.5 μg ml−1 and was completely inhibited at 50 μg ml−1; it was significantly affected by either bromoconazole at 5 μg ml−1 or captan at 1,250 μg ml−1, and was completely inhibited by their mixture. In general, three foliar applications of bromuconazole or difenoconazole in the field, during the bloom period, reduced the numbers of infected fruits by 40–60% compared with untreated control trees. However, tank mixes of either fungicide with captan improved control of moldy-core in fruits at harvest. Tank mixtures of bromuconazole and captan also significantly reduced the percentage of fruits colonized by A. alternata when sampled at various days after full bloom. Artificial inoculations in the orchard at full bloom did not change the inhibitory effects of the tank mixtures. Large-scale demonstration trials in commercial orchards supported these findings. The inhibitory effects of tank mixes on decay development in detached fruits, and on moldy-core in the field indicate that a control programme based on mixtures of either bromuconazole or difenoconazole with captan during the bloom period can effectively reduce moldy-core on Red Delicious apples.

The susceptibility of Fortune (Citrus clementina × Citrus reticulata), Citrus paradisi and Citrus limon fruits to Alternaria alternata pv. citri was investigated using different artificial inoculation methods. The results obtained reveal that the C. paradisi and C. limon fruits are less susceptible to A. alternata pv. citri than Fortune fruits, although all showed symptoms of Alternaria brown spot when the cuticle was broken and the flavedo or flavedo + albedo was removed. Furthermore, it was seen that susceptibility to the fungus decreased as the age of the fruit increased. There was a positive correlation between the susceptibility of the different Citrus fruits to A. alternata pv. citri and their “in vivo” ethylene levels, the most susceptible fruit (Fortune) producing more ethylene during growth than the less susceptible C. limon and C. paradisi. This suggests that ethylene may well be considered as a possible marker of Citrus fruit susceptibility to A. alternata pv. citri. Disease development increased when the Fortune fruits were treated with 1 mM ACC (a precursor of ethylene biosynthesis) or 1 mM Ethephon (an ethylene-releasing compound) prior to inoculation with A. alternata pv. citri. The role of ethylene as a factor involved in disease development is discussed.

The fungus Alternaria alternata, is considered to be the predominant fungus involved in moldy-core of Red Delicious strains of apple. In this paper, we report on the sensitivity of various phenologic stages to infection by A. alternata, and on the efficacy of various fungicides in controlling moldy-core disease in apple orchards. Artificial inoculations conducted in the orchard during 1999 and 2000 seasons revealed that the beginning of bloom (10–30%) and full bloom were the most susceptible developmental stages for infection. Natural infection with A. alternata in fruits was relatively high, reaching 44% and 46% of the fruits on control non-treated trees in 1999 and 2000. Four foliar applications of polyoxin B, difenoconazole and azoxystrobin, starting from the beginning of bloom until fruit set, reduced the number of infected fruits by 54–70%, 61–70% and 50–65%, respectively, compared with non-treated trees. Four or eight foliar applications of each fungicide provided similar levels of control. There were no significant differences between two, four or six foliar applications of difenoconazole, neither between two or four applications of polyoxin B. Adding CaCl2, as a tank mixture with difenoconazole at full rate, did not improve efficacy. Alternaria was recovered from the inner part of the core region of 71–88% of the fruits of the non-treated control, but was recovered less frequently from the outside part of the core region. Fruits of difenoconazole and polyoxin B treated trees were less colonized with A. alternata at both the inner and outside parts of the core region, as compared with controls. Results indicate that a control programme based on spray applications of difenoconazole or polyoxin B, during bloom period, can effectively reduce Alternaria on Red Delicious.

An outbreak of black mottle and dieback on basil (Ocimum basilicum L.; Lamiaceae) was recorded in a greenhouse in Okinawa Prefecture, Japan during 2004. The causal agent was identified as Alternaria alternata (Fr.) Keissler based on morphological characters and growth temperature. This report is the first of Alternaria leaf spot of basil caused by A. alternata.

In tomato plants, α-tomatine, a steroidal glycoalkaloid saponin, inhibits fungal growth. Tomato pathogens that produce host-specific toxins, Alternaria alternata tomato pathotype causing Alternaria stem canker and Corynespora cassiicola causing Corynespora target spot, were investigated for sensitivity to α-tomatine. Although spore germination of A. alternata pathogenic and nonpathogenic to tomato and of C. cassiicola pathogenic to tomato was not affected by 0.1 mM α-tomatine, spore germination of C. cassiicola nonpathogenic to tomato was significantly inhibited. This result showed that A. alternata, regardless of its pathogenicity, and only the C. cassiicola pathogenic to tomato are resistant to α-tomatine. Germinating spores of A. alternata and C. cassiicola resistant to α-tomatine detoxified α-tomatine by degrading it to a less polar product. After inoculation of tomato leaves, spores of A. alternata and C. cassiicola nonpathogenic to tomato germinated and formed appressoria, but did not form infection hyphae in host tissues. When a host-specific toxin (CCT-toxin) produced by C. cassiicola pathogenic to tomato was added to nonpathogenic spores, colonization within leaves was observed in A. alternata, but not in C. cassiicola. On the other hand, when spores of C. cassiicola nonpathogenic to tomato were suspended in spore germination fluid of nonpathogenic A. alternata with α-tomatine detoxification activity, the fungus could be induced to colonize leaves in the presence of CCT-toxin. These results indicate that A. alternata tomato pathotype and C. cassiicola pathogenic to tomato detoxify α-tomatine during infection and that this detoxification is essential for host colonization by pathogens that produce host-specific toxins.

Stemphylium vesicarium (teleomorph: Pleospora herbarum) is the causal agent of brown spot disease in pear. The species is also able to cause disease in asparagus, onion and other crops. Saprophytic growth of the fungus on plant debris is common. The objective of this study was to investigate whether isolates of S. vesicarium from different hosts can be pathogenic to pear. More than hundred isolates of Stemphylium spp. were obtained from infected pear fruits, dead pear leaves, dead grass leaves present in pear orchard lawns as well as from necrotic leaf parts of asparagus and onion. Only isolates originating from pear orchards, including isolates from dead grass leaves, were pathogenic on pear leaves or fruits in bioassays. Non-pathogenic isolates were also present in pear orchards. Stemphylium vesicarium from asparagus or onion, with one exception, were not pathogenic to pear. Analysis of the genetic variation between isolates using Amplified Fragment Length Polymorphism (AFLP) showed significant concordance with host plants. Isolates from asparagus or onion belonged to clusters separate from the cluster with isolates from pear or grass leaves collected in pear orchards. Multilocus sequencing of a subset of isolates showed that such isolates were similar to S. vesicarium.

Host-specific AAL-toxins and mycotoxin fumonisins are structurally related and were originally isolated from the tomato pathotype of Alternaria alternata and from Fusarium verticillioides, respectively. Previous reports on the production of fumonisin derivatives by the tomato pathotype suggested a possible involvement in the pathogenicity of the pathogen. Here, we have evaluated the role of fumonisin in A. alternata–tomato interactions. The results indicate that highly pathogenic isolates of A. alternata tomato pathotype produce AAL-toxin as the sole toxin, strongly implicating it as a pathogenicity factor. The related compound, fumonisin, is also toxigenic and has infection-inducing activity on susceptible tomato plants.

The Japanese pear pathotype of Alternaria alternata, a toxin-dependent necrotrophic pathogen, causes black spot of Japanese pear by producing the host-specific AK-toxin. Pre-inoculation with nonpathogenic A. alternata or pretreatment with an elicitor prepared from A. alternata reduced disease symptoms caused by the pathogen. Salicylic acid- and jasmonic acid-dependent signaling pathways are not involved in the induced resistance to infection by the pathogen. The expression of multiple defense-related genes in Japanese pear leaves inoculated with nonpathogenic A. alternata was examined using suppression subtractive hybridization. A disease caused by Alternaria alternata occurred on the leaves of European pear cultivar Le Lectier in Niigata Prefecture, Japan, and was named black spot of European pear. In conidial inoculation tests, the causal pathogen induced not only small black lesions on the leaves of European pear cultivar Le Lectier, but severe lesions on the leaves of apple cultivar Red Gold, which is susceptible to the A. alternata apple pathotype (previously called A. mali) causing Alternaria blotch of apple. Interestingly, the apple pathotype isolate showed the same pathogenicity as the European pear pathogen. HPLC analysis of the culture filtrates revealed that A. alternata causing black spot of European pear produced AM-toxin I, known as a host-specific toxin of the A. alternata apple pathotype. AM-toxin I induced veinal necrosis on leaves of Le Lectier and General Leclerc cultivars, both susceptible to the European pear pathogen, at 5?×?10?7 M and 10?6 M respectively, but did not affect leaves of resistant cultivars at 10?4 M. PCR analysis with primers that specifically amplify the AM-toxin synthetase gene detected the product of expected size in the pathogen. These results indicate that A. alternata causing black spot of European pear is identical to that causing Alternaria blotch of apple. This is the first report of European pear disease caused by the A. alternata apple pathotype. This study provides a multiplex PCR protocol, which could serve as a useful tool, for the epidemiological survey of these two diseases in European pear and apple orchards.

Fistupyrone (FP), a metabolite from Streptomyces sp. TP-A0569, inhibited the in vivo infection of Chinese cabbage seedlings by Alternaria brassicicola. To detect the possible action sites of FP, the effect of FP on the infection behavior of A. brassicicola and A. alternata was investigated. When spores of A. brassicicola were suspended in FP solution and inoculated on host leaves, FP at 0.1ppm significantly inhibited spore germination, appressorial formation, and infection hypha formation of A. brassicicola. Host-specific AB-toxin production and lesion formation by A. brassicicola spores were also reduced significantly by treatment with FP 1ppm. The effect of FP seemed to be irreversible because significant washing of FP-treated spores with distilled water (DW) did not change the inhibitory effects. In contrast, A. alternata isolates such as Japanese pear pathotype, apple pathotype, and saprophyte behaved almost equally in both FP- and DW-treated spores. Mycelial dry weight in potato dextrose broth and mycelial diameters on potato dextrose agar, gelatin glucose agar, and Czapek solution agar of both A. brassicicola and A. alternata were not different with or without addition of FP. These results indicate that FP at low concentrations has a fungicidal effect on spores of A. brassicicola but not on spores of A. alternata; FP also does not affect the vegetative phase of these fungi.

When detached broad bean leaves were preinoculated with virulent strain B304 of Botrytis cinerea 24 h before a challenge inoculation with strain B304, lesion formation by B304 was significantly inhibited in red light but not in the dark. In leaves that were preinoculated with avirulent strain 021 and then challenged by B304, however, lesion formation was not inhibited even under red light. Such differences in lesion formation after the challenge inoculation with an avirulent strain were also observed with lesions caused by Alternaria alternata, a nonpathogen of broad bean and by avirulent strain 021R in the presence of germination fluid from spores of strains B304 and 021R. These results suggest the possibility that virulent B. cinerea produced a suppressor involved in induced susceptibility and an elicitor involved in resistance induced by red light during spore germination.
